Aqueous-based electrospun P(NIPAAm-co-AAc)/RSF medicated fibrous mats for dual temperature- and pH-responsive drug controlled release

Juan Li,Jingxin Zhu,Lan Jia,Yanlong Ma,Haijuan Wu

This paper presents a green method for fabricating dual temperature- and pH-responsive electrospun fibrous mats from an aqueous-based blend poly(N-isopropylacrylamide-co-acrylic acid) (P(NIPAAm-co-AAc)) and regenerated silk fibroin (RSF) by employing electrospinning technique. P(NIPAAm-co-AAc) was synthesized by free radical solution polymerization and its low critical solution temperature (LCST) was in the physiological range (38.8 °C). The P(NIPAAm-co-AAc)/RSF fibers were prepared by electrospinning technology in the presence of the crosslinking agents (EDC·HCl and NHS) with water as solvent. After in situ crosslinking and water-annealing process, the water-stable composite fibrous mats were obtained. Scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR) were used to analyze the crosslinking process. Temperature and pH dual stimuli-responsive swelling-shrinking behavior of the fibrous mats were observed when the temperature was below and above the LCST of the copolymer at different pHs. In addition, rhodamine B-loaded the fibrous mats also showed dual temperature and pH controlled release behavior, demonstrating the potential use of the fibrous mats for “smart” controlled drug delivery applications.

A Double-Switch Temperature-Sensitive Controlled Release Antioxidant Film Embedded with Lyophilized Nanoliposomes Encapsulating Rosemary Essential Oils for Solid Food

Xi Chen,Qing Long,Lei Zhu,Li-Xin Lu,Li-Nan Sun,Liao Pan,Li-Jing Lu,Wei-Rong Yao

In order to match the solid food oxidation during logistics and storage process under severe high temperature, a double-switch temperature-sensitive controlled release antioxidant film embedded with lyophilized nanoliposomes encapsulating rosemary essential oils (REOs) was prepared. The double switch temperature at 35.26 and 56.98 °C was achieved by development of a temperature sensitive polyurethane (TSPU) film. With biaxially oriented polyethylene terephthalate (BOPET) as a barrier layer, the intelligent complex film was prepared via coating the TSPU embedded with lyophilized nanoliposomes encapsulating REOs on BOPET. The results indicate that the REO is well encapsulated in nanoliposomes with encapsulation efficiency (EE) of 67.3%, high stability and lasting antioxidant effect during 60 days. The incorporation of lyophilized nanoliposomes containing REOs into TSPU remains the double-switch temperature-sensitive characteristic of the prepared TSPU. In agreement with porosity andWVTRresults, the diffusion coefficient (D) of the antioxidant complex film sharply increases respectively at two switching temperatures, indicating that the intelligent double-switch temperature-sensitive controlled release property is functioning. Furthermore, compared with films directly added with REO, the lowerDs of films added with lyophilized nanoliposomes encapsulating REOs provides a longer-lasting antioxidant activity. Thus, the acquired controlled release antioxidant film sensitive to temperature at 39.56 and 56.00 °C can be potentially applied for protection of solid food during distribution and storage process under severe high temperatures.Keywords:temperature-sensitive;double-switch;controlled release;antioxidant;nanoliposome;rosemary essential oils

Stimulus-Detonated Biomimetic “Nanobomb” with Controlled Release of HSP90 Inhibitor to Disrupt Mitochondrial Function for Synergistic Gas and Photothermal Therapy

Geng Yang,Ting Song,Hanxi Zhang,Mengyue Li,Xiaodan Wei,Wanyi Zhou,Chunhui Wu,Yiyao Liu,Hong Yang

Photothermal therapy (PTT) is considered a promising treatment for tumors; however, its efficacy is restricted by heat shock proteins (HSPs). Herein, a stimuli-responsive theranostic nanoplatform (M/D@P/E-P) is designed for synergistic gas therapy and PTT. This nanoplatform is fabricated by a load of manganese carbonyl (MnCO, CO donor) in dendritic mesoporous silicon (DMS), followed by the coating with polydopamine (PDA) and loading of epigallocatechin gallate (EGCG, HSP90 inhibitor). Upon near-infrared (NIR) irradiation, the photothermal effect of PDA can kill tumor cells and allow for the controlled drug release of MnCO and EGCG. Moreover, the acidity and H 2 O 2 -rich tumor microenvironment enable the decomposition of the released MnCO, accompanied by the production of CO. CO-initiated gas therapy can realize to disrupt the mitochondrial function, which will accelerate cell apoptosis and down-regulate HSP90 expression by decreasing intracellular ATP. The combination of EGCG and MnCO can significantly minimize the thermo-resistance of tumors and improve PTT sensitivity. In addition, the released Mn 2+ enables T 1 -weighted magnetic imaging of tumors. The therapeutic efficacy of the nanoplatform is methodically appraised and validated both in vitro and in vivo. Taken together, this study affords a prime paradigm for applying this strategy for enhanced PTT via mitochondrial dysfunction.

Amphiphilic block copolymer NPs obtained by coupling ricinoleic acid/sebacic acids and mPEG: Synthesis, characterization, and controlled release of paclitaxel

Shiya Zhou,Wei Sun,Yinglei Zhai

Currently, nanoparticles (NPs) made of amphiphilic block copolymer are still an essential part of drug delivery system. Here, we report a novel amphiphilic block copolymer and paclitaxel (PTX)-loaded copolymer NPs for the controlled delivery of PTX. The block copolymer was synthesized via melt polycondensation method of methoxy poly(ethylene glycol) (mPEG), sebacic acid (SA) and ricinoleic acid (RA). A series of characterization approaches such as Fourier Transform Infrared Spectroscopy (FTIR), 1Hydrogen-Nuclear Magnetic Resonance (1H-NMR), Differential Scanning Calorimetry (DSC), X-Ray Diffraction (XRD) and Gel Permeation Chromatography (GPC) applied have shown that the amphiphilic block copolymer was prepared as designed. NPs prepared by nanoprecipitation method consist of mPEG segments as the hydrophilic shell and RA-SA segments as the hydrophobic core, hydrophobic PTX was encapsulated as model drug. Subsequently, Transmission Electron Microscopy (TEM) analysis indicated that the spherical NPs have effective mean diameters ranging from 100 to 400 nm. Dynamic Light Scattering (DLS) analysis also revealed the controllable NPs diameter by modulating the mass ratio of RA to SA and drug loading amount (DLA). Besides, biphasic profile with zero order drug release was observed in general in vitro release behaviors of PTX from NPs. Further investigation confirmed that the release behaviors depend on the crystallinity of hydrophobic RA-SA segments. Results above suggest that NPs with amphiphlic block copolymer mPEG-b-P(RA-SA)-b-mPEG have a remarkable potential as a carrier for hydrophobic drug delivery in cancer therapy.

Dual responsive aerogel made from thermo/pH sensitive graft copolymer alginate-g-P(NIPAM-co-NHMAM) for drug controlled release

Lin Shao,Yang Cao,Zhanying Li,Wenbin Hu,Shize Li,Lingbin Lu

Alginate was grafted with NIPAM and NHMAM successfully, and a new responsive copolymer, alginate-g-P(NIPAM-co-NHMAM), was obtained. A novel dual responsive polysaccharide-based aerogel with thermo/pH sensitive properties was designed from the copolymer as drug controlled release system. The chemical structure of the copolymer was characterized by FT-IR and 1 H NMR. Lower critical solution temperature (LCST) of the copolymer covered a wide temperature range from 27.6 °C to 42.2 °C, which could be adjusted with changing the ratio between NIPAM and NHMAM. The dual responsive aerogel had a three-dimensional network structure. As a drug controlled release system, the aerogel was high responsive to both temperature and pH with drug loading efficiency up to 13.24%. Above LCST, the aerogel had a faster drug release, and drug was completely released in neutral environment, while the drug release was obstructed in acid environment. Furthermore, the drug release mechanism of the aerogel was illuminated. These results indicated that the dual responsive aerogel was a promising candidate for drug carriers.

Design and self-assembly of albumin nanoclusters as a dynamic-covalent targeting co-delivery and stimuli-responsive controlled release platform

Wen Liu,Jian Dai,Wei Xue

Stimuli-responsive nanomaterial-based drug delivery systems that are able to actively target the tumor microenvironment, enhance intratumoral accumulation and release drugs at target sites are attractive therapeutic platforms at present. Here, the first approach that uses proteins to construct dynamic-covalent targeting stimuli-responsive cationic albumin nanoclusters for the highly efficient co-delivery of siRNA and DOX in vitro and in vivo has been developed and achieved considerable efficacy in combination therapy for the growth, migration and invasion of cancer. In this study, the critical strategy of ‘dynamic-covalent targeting’, as previously mentioned, was developed for the active accumulation of drugs at target sites by fabricating a reversible PEG corona using a pH-sensitive borate ester so as to facilitate the formation of a compact complex and the efficient release of siRNA with or from the nanocluster. In addition, after internalization, the nanocluster can dissociate into cationic albumin via intracellular reduction in an acid environment to achieve stimuli-responsive triggered drug release. As a result, the nanocluster for the co-delivery of DOX and VEGF-siRNA exhibits highly efficient capacity for gene silencing and apoptosis-inducing ability and also markedly suppresses the migration and invasion of cancer cells. Most importantly, it displayed significant combination effects against tumor growth with minimal damage to major organs and provides an effective approach for the safe and efficient delivery of multiple cargoes at target sites.

Fabrication of pH-responsive system based on cationic gemini surfactant/sodium octanedioate and its application on controlled release of paclitaxel

Jie Yang,Le Yun,Guowei Zhao,Feng Zhang,Yashao Chen,Changhao Wang

A pH-responsive surfactant system has been self-assembled between a cationic gemini surfactant hexamethylene-1,6-bis(dodecyldimethylammonium bromide) (12-6-12) and sodium octanedioate (C 8 Na 2 ), which was characterized by surface tension, proton nuclear magnetic resonance ( 1 H NMR), dynamic light scattering (DLS), transmission electronic microscopy (TEM) and fluorescence probe spectroscopy. The interaction between 12 and 6–12 and C 8 Na 2 are based on the electrostatic and hydrophobic interaction as evidenced by surface tension and 1 H NMR. The self-assembled 12-6-12/C 8 Na 2 system has been demonstrated to be concentration dependence and pH dependence. With increasing the concentration of C 8 Na 2 , the vesicle form of 12-6-12 can transform to micelles. Vesicle-to-micelle transition of the 12-6-12/C 8 Na 2 aggregates is reversible with tuning the pH values. The micropolarity inside the 12-6-12/C 8 Na 2 aggregates was characterized by fluorescence spectroscopy using pyrene as probe, indicating that the micropolarity is enhanced with the increased pH. Furthermore, the release assay of an oil-soluble drug paclitaxel performs well in this pH-responsive surfactant system and the drug releasing rate could be controlled by pH values.

Fibrous guided tissue regeneration membrane loaded with anti-inflammatory agent prepared by coaxial electrospinning for the purpose of controlled release

Min He,Jiajia Xue,Huan Geng,Hao Gu,Dafu Chen,Rui Shi,Liqun Zhang

Here, with the aim of inhibiting inflammation during guided tissue regeneration membrane (GTRM) implant surgery, coaxial electrospinning was used to fabricate drug-loaded core/sheath nanofiber GTRMs capable of controlled drug release. Various amounts of the anti-inflammatory agent metronidazole (MNA) were encapsulated into the core/sheath nanofibers (where PCL was the core, gelatin the sheath, and the gelatin shell was crosslinked with genipin) in order to establish the minimal drug content necessary to achieve the appropriate anti-inflammatory effect. By using TEM and SEM, the core/sheath structure was confirmed. In vitro drug disolution results showed that the core/sheath nanofibers exhibited sustained release profiles that were superior to those nanofibers produced by blending electrospinning. Additionally, the membrane significantly inhibited the colonization of anaerobic bacteria. Furthermore, with gelatin as a shell, the core/shell nanofiber membranes showed improved hydrophilicity, which resulted in better cell adhesion and proliferation without cytotoxicity. Therefore, in this study, a simple and effective coaxial electrospinning approach was demonstrated for the fabrication of anti-inflammatory GTRMs capable of providing controlled drug release.

Core-shell structured polypyrrole/mesoporous SiO2 nanocomposite capped with graphene quantum dots as gatekeeper for irradiation-controlled release of methotrexate

Xiaolin Liu,Dan Shou,Chuanxiang Chen,Huihui Mao,Yong Kong,Yongxin Tao

A core-shell structured nanocomposite of polypyrrole/mesoporous SiO 2 (PPy/mSiO 2 ) is rationally designed as the nanocarrier for methotrexate (MTX), a chemotherapeutic drug for cancer treatment. Graphene quantum dots (GQDs) are introduced to the outer surface of PPy/mSiO 2 , and it functions as a gatekeeper for the loaded MTX through the formation of H-bonds with the functionalized mSiO 2 . In the proposed nanocarrier for MTX, the mesopores in mSiO 2 are beneficial for the accommodation of MTX, resulting in enhanced encapsulation capacity of the nanocarrier; on the other hand, PPy can effectively convert the near-infrared (NIR) light to heat. Under the irradiation of NIR light, the H-bonds between GQDs and mSiO 2 are broken due to the gradually increased temperature, and therefore the GQDs cap is removed and consequently the encapsulated MTX is released from the nanocarrier. In this study, NIR irradiation-controlled drug delivery is achieved successfully owing to the synergistic effects of PPy, mSiO 2 and GQDs, which opens a new window for the construction of smart drug delivery systems.

A pH-responsive emulsion stabilized by alginate-grafted anisotropic silica and its application in the controlled release of λ-cyhalothrin

Kai Chen,Gaobo Yu,Furui He,Qingfeng Zhou,Dunchao Xiao,Jiacheng Li,Yuhong Feng

Alginate (Alg) was grafted on the surface of anisotropic silica (SiO 2 -x) via the Ugi reaction (Alg-SiO 2 -1, Alg-SiO 2 -2, and Alg-SiO 2 -4). Compared with pristine SiO 2 -x, modified SiO 2 -x is more sensitive to pH. Three stable liquid paraffin-in-water emulsions were prepared with Alg-SiO2-1, Alg-SiO2-2, and Alg-SiO2-4. Alg-SiO 2 -2 exhibited satisfactory emulsification ability. The emulsions became more stable as emulsion pH varied from 2.0 to 6.2 because of polymer chain interactions that led to the formation of a three-dimensional network. When the emulsion pH varied from 6.2 to 8.0, the particle charge increased, in turn increasing interparticle the electrostatic interactions that increased emulsion stability. When the emulsion pH was 9.0, the subsequent decrease in particle charge, decreased the emulsion stability. The model drug λ-cyhalothrin was embedded in the emulsions. A sustained-release assay demonstrated that increasing emulsion pH from 3.0 to 8.0 decreased cumulative drug release from the emulsion from 99.7% to 13.5%. This result indicated that the emulsion is a pH triggered drug delivery system. The sustained-release curves of λ-cyhalothrin are describable by the Weibull model.

Controlled release of 1,2-benzisothiazolin-3-one-based anion encapsulated in MgAl layered double hydroxides for antibacterial and antifouling applications

Xiaoxian Hao,Xin Zhang,Wenjing Wang,Wenjing Liu,Guangming Lu,Wenru Zheng,Dirong Gong,Haichao Zhao

The controlled release of antifouling agents from organic coatings is an effective measure against marine biofouling. In this study, Mg Al layered double hydroxides (LDH), a two-dimensional nanoparticle , were intercalated with the 1,2-benzisothiazolin-3-one-based anions (BIT-COO − ) via an ion-exchange process for achieving controlled release of BIT-based antibacterial agent. Multifunctional X-ray photoelectron spectroscopy, thermogravimetric analyzer, and transmission electron microscopy confirmed the successful loading of BIT intercalated into LDH interlayers. The antibacterial performance of LDH@BIT-COOH was tested by the zone of inhibition method, the plate colony counting method, the turbidity method, and the biofilm method, and results proved that 0.4 mg/mL LDH@BIT-COOH exhibited complete inactivation of E. coli (6 h) and S. aureus (12 h ). The release study of the LDH@BIT-COOH proved that the release time prolonged to 4500 min after loading. The 5 wt% LDH@BIT-COOH coating exhibited outstanding antibacterial and antifouling properties by using bio-laser confocal microscope , and without obvious toxicity as observed by L929 cell test using enzyme marker, showing potential applications in marine antifouling.

Controlled release mechanism of off-flavor compounds in Bacillus subtilis BSNK-5 fermented soymilk and flavor improvement by phenolic compounds

Miao Hu,Yaxin Gao,Weimin Meng,Pengfei Zhang,Jiao Wang,Zifan Yuan,Bei Fan,Fengzhong Wang,Shuying Li

Bacillus subtilis is the primary strain used in fermented soy products. The applicant fermented soymilk with the laboratory strain B. subtilis BSNK-5 to enhance nutrient density. However, prolonged fermentation caused rapid deterioration in flavor, resulting in an off-flavor poorly accepted by consumers, limiting its industrial application. Studies identified isovaleric acid (IVA), isobutyric acid (IBA), and 2-methylbutyric acid (2-MBA) as the main off-flavor compounds. The mechanism controlling off-flavor release remains unclear. Therefore, soybean protein was used as a model to investigate off-flavor release by analyzing binding percentage, physicochemical properties, conformation, and interaction forces. The modification of off-flavor by phenolic compounds was also examined. Results showed that soybean protein bound over 90 % of the flavor compounds, including a 98.6 % binding rate for IVA. Binding between soybean protein and off-flavor compounds was confirmed by the formation of large aggregates, decreased surface hydrophobicity, and a structural transformation from α-helix to β-sheet. Hydrogen bonds and hydrophobic interactions were identified as the primary interaction forces. Adding phenolic compounds significantly reduced soybean protein binding to flavor compounds. Phenolic compounds had a stronger binding affinity to soybean protein compared to flavor compounds and occupied binding sites on soybean protein, preventing flavor compound binding. L-epicatechin, (−)-epicatechin gallate, and epigallocatechin-3-gallate primarily occupied binding sites for 2-MBA, while ferulic acid, chlorogenic acid, and caffeic acid occupied sites for IVA, IBA, and 2-MBA. This research aids in controlling flavor release by soybean protein in food systems, supporting the development of nutrient-rich fermented foods.
