Fabricating an Acetylcholinesterase Modulated UCNPs-Cu2+ Fluorescence Biosensor for Ultrasensitive Detection of Organophosphorus Pesticides-Diazinon in Food

Pingyue Wang,Huanhuan Li,Md Mehedi Hassan,Zhiming Guo,Zheng-Zhu Zhang,Quansheng Chen

In this study, a highly sensitive upconversion fluorescence (FL) biosensor was developed for the detection of organophosphorus pesticides (OPs) based on an acetylcholinesterase (AChE) modulated FL “off–on–off” strategy. The luminescence of synthesized UCNPs could be quenched strongly by Cu2+ due to an energy transfer effect. Upon addition of AChE and acetylthiocholine (ATCh), the enzymatic hydrolysate (thiocholine) could seize Cu2+ from UCNPs-Cu2+ mixture, resulting in the quenched FL triggered on. OPs could irreversibly impede the activity of AChE, which caused the formation of thiocholine to decrease, thus, reduced the recovery of FL. Under the optimum conditions, a linear detection range from 0.1 to 50 ng/mL was achieved for the representative OPs (diazinon) with LOD of 0.05 ng/mL. Furthermore, the ability of the biosensor to detect OPs was also confirmed in adulterated environmental and agricultural samples. In validation analysis, the proposed sensor showed satisfactory results (p > 0.05) with GC–MS.

Luminescent NaYF4:Yb,Er@UiO-66 composite based probe for the selective and sensitive detection of Cu2+

Haoran Zhang,Tianxi Feng,Song Ye,Deping Wang

In this research, the modified upconversion nanocrystals NaYF 4 :Yb,Er were adopted as the precursors for the synthesis of the NaYF 4 :Yb,Er@UiO-66 composite. The XRD, XPS and TEM characterizations indicated that the composite has good crystallinity and uniform morphology. Under 980 nm excitation, the NaYF 4 :Yb,Er@UiO-66 composite exhibits bright upconversion emission that characterized of Er 3+ : 2 H 11/2 → 4 I 15/2 , 4 S 3/2 → 4 I 15/2 , and 4 F 9/2 → 4 I 15/2 radiative transitions. After modified with BPEI (branched polyethyleneimine), the NaYF 4 :Yb,Er@UiO-66/BPEI fluorescence probe was constructed and was used for the detection of Cu 2+ . The results revealed that the integrated emission intensity of Er 3+ displayed a linear relationship with the concentration of Cu 2+ within the range of 5 to 45 μM with a LOD of 0.9615 μM. This study offered a feasible method for constructing UCNC@UiO-66 composites with regular morphology, which may have a potential application as fluorescent probes for the detection of various metal ions by adjusting the doping luminescent center.

Catalytic Activation Coupled Regulation of Carbon Structural Domains Enhances the Performance of Capacitive Deionization Purification of Cu2+

Yang Zhao,Xinru Gong,Rui Dai,Shuangfeng Fang,Baihang Cai,Fei Wang,Yufeng Yan,Puyuan Liu,Yi Luo,Junheng Fu,Yong Wang,Liang Qiao,Minglei Yan

The efficient purification of Cu 2+ -contaminated wastewater poses a formidable challenge. Capacitive deionization (CDI) technology, which operates on the principle of electrochemical capacitance, has emerged as a promising candidate for serving as a key technology to address this problem. In this work, chitosan is employed as a carbon precursor for the fabrication of a CDI carbon electrode (CoZn-C). This electrode features a high specific surface area and short-range ordered carbon domains, which are achieved through the strategic utilization of basic carbonate templates to induce site-occupying effects, coupled with the synergistic catalysis of metal. Importantly, benefiting from sufficient electrochemical adsorption active sites and the carbon structural channels that facilitate rapid Cu 2+ transport, the constructed symmetric CoZn-C//CoZn-C CDI device demonstrates excellent purification capability for Cu 2+ . The adsorption capacity and removal efficiency of the CDI electrode for Cu 2+ are as high as 124.3 mg g −1 and 94.5%, respectively (at 1.2 V, 50 mg L −1 ). The electrochemical capture of Cu 2+ by CoZn-C CDI electrodes is achieved through the coupling effect of the electrical double layer and electro-reduction deposition. Accordingly, the carbon domain regulation method offers a new strategy for designing and constructing high-efficiency CDI carbon electrodes for the heavy metal ion wastewater purification.

Biomimetic Cu2+/Zn2+ bimetallic surface with glycocalyx-mimicking polymer for selective vascular cell regulation and enhanced hemocompatibility

Shuaihang Guo,Shengen Gu,Qiulian Wu,Mengying Zhan,Lisha Pan,Dan Li,Yiping Chen,Xiaoli Liu

The vascular endothelium represents the ideal hemocompatible interface, exhibiting unique capabilities to prevent thrombus formation while maintaining vascular homeostasis. To emulate these properties, we have developed a biomimetic surface combining two critical endothelial components: biologically active metal ions that regulate vascular function, and synthetic glycosaminoglycan analogs that replicate the endothelial glycocalyx's antithrombogenic properties. Here, a bimetallic Cu 2+ /Zn 2+ composite coating was developed on polyvinyl chloride (PVC) substrates through dopamine-assisted immobilization, with the surface metal composition being controlled by adjustment of the Cu 2+ /Zn 2+ feeding ratio. The Cu 2+ -rich surface (PC 5 Z 1 ) demonstrated enhanced nitric oxide (NO) catalytic release, effectively suppressing human umbilical vein smooth muscle cell (HUVSMC) proliferation, while the Zn 2+ -enriched surface (PC 1 Z 4 ) promoted human umbilical vein endothelial cell (HUVEC) growth. The optimal Cu 2+ /Zn 2+ (1:1, PC 1 Z 1 ) surface was further functionalized with glycosaminoglycan analog poly(SS- co -OEGMA) (PSO), creating a multifunctional coating that synergistically combines metal ion regulation with glycocalyx-mimicking chemistry. The resulting PC 1 Z 1 -PSO surface achieved a HUVEC/HUVSMC ratio of 2.4 in co-culture, demonstrating its dual capability in promoting endothelialization while inhibiting smooth muscle cell proliferation. Furthermore, platelet adhesion and clotting time assays showed the excellent hemocompatibility of the PC 1 Z 1 -PSO surface. These results demonstrate that strategic integration of bioactive metal ions with glycocalyx-mimicking polymers can effectively mimic key endothelial functions, offering a promising strategy for optimizing the hemocompatibility of blood-contacting materials.

Haematococcus pluvialis residues derived deep red emissive carbon dots for ultrasensitive and selective Cu2+ detection in seawater

Shuju Guo,Na Yang,Wenjie Yu,Litao Zhang,Jianguo Liu

Accurate and reliable detection of trace copper ions (Cu 2+ ) is of great importance due to their dual role as essential micronutrients and potential environmental toxins. Herein, we report the first synthesis of deep red emissive CDs (CD-HP) derived from Haematococcus pluvialis residues, followed by nitrogen-doping modification (CD-HPE series) to enhance ion selectivity. Among them, CD-HPE2 exhibits excitation-independent emission at 674 nm with a narrow full width at half maximum (FWHM) of 34 nm, rivaling heavy metal-based quantum dots while retaining biocompatibility. The sensor demonstrates outstanding sensing performance, achieving an ultralow detection limit of 3.03 nM and a linear response range of 10–100 nM. The sensor shows excellent selectivity for Cu 2+ against potentially interfering ions and maintains stable fluorescence across pH 6–9 and salinity levels up to 50 ‰. Application to real seawater samples yielded recoveries of 94.10–106.45 %, confirming its reliability in complex matrices. Compared with recently reported biomass-derived CDs, CD-HPE2 offers unique optical properties, superior sensitivity, and strong anti-interference capability. This work highlights an eco-friendly strategy to upcycle microalgal residues into high-value nanosensors and establishes a robust platform for ultrasensitive Cu 2+ detection in environmental analysis.

Ultrasmall Cu2−xSe nanoparticles alleviate vascular calcification through inhibiting oxidative stress and NF-κB/NLRP3-mediated inflammation

Ding Liu,Yuanzhi Ye,Zirong Lan,An Chen,Xingchen Zhou,Zhenhao Li,Xin Wen,Jisheng Xiao,Fan Ouyang,Jianyun Yan

Vascular calcification, prevalent in patients with chronic kidney disease, atherosclerosis, and diabetes, is strongly associated with elevated cardiovascular morbidity and mortality, highlighting the urgent need for effective treatments. Oxidative stress is a key contributor to the progression of vascular calcification. Nanozymes, nanomaterials with enzyme-like catalytic properties, exhibit strong reactive oxygen species (ROS) scavenging abilities and good biocompatibility, making them promising therapeutic candidates. This study aims to investigate whether polyvinylpyrrolidone (PVP)-functionalized ultrasmall Cu 2-x Se nanoparticles (CSP NPs) act as nanozymes for treating vascular calcification. In vitro , CSP NPs significantly inhibit calcification of rat and human vascular smooth muscle cells (VSMCs) and reduce the expression of osteogenic markers Runx2 and BMP2. Moreover, CSP NPs alleviate calcification of rat and human arterial rings. In a mouse model, CSP NPs localize to certain areas, such as the aortic arch and abdominal aortas, and are safely metabolized by the liver and kidneys without organ toxicity. Further analyses confirm that CSP NPs inhibit mouse and chronic kidney disease (CKD) rat aortic calcification. Mechanistically, CSP NPs inhibit oxidative stress and mitochondrial dysfunction. Additionally, CSP NPs decrease the expression of NF-κB and NLRP3, thus reducing the levels of inflammatory cytokines IL-1β and IL-6. CSP NPs suppress NLRP3 activator-induced calcification in VSMCs and arterial rings. This study provides the first evidence that CSP NPs alleviate vascular calcification by inhibiting oxidative stress and NF-κB/NLRP3-mediated inflammation, suggesting a promising therapeutic approach.

Turning Waste into Treasure: A Magnetic Sludge-Based Biochar for Efficient Co-Removal of Tetracycline and Cu2+

Leyao Shi,Jiexiong Zhong,Miao Wu,Qun Liu,Liu Yang,Xiaohui Xu,Weixue Wang,Xiaohui Guan,Min Lu

The treatment and disposal of excess sludge, a major solid byproduct of wastewater treatment processes, pose significant environmental and public health challenges, making its rational utilization crucial for promoting resource recycling. This study presents a one-step synthesis of a magnetically responsive bifunctional biochar composite (Fe-SDB600) derived from excess sludge for the simultaneous and enhanced removal of tetracycline (TC) and copper ions (Cu2+) from aqueous solutions. Characterization confirmed the successful loading of nanoscale iron oxide particles onto the biochar surface, resulting in a hierarchical porous structure. Under optimized conditions, Fe-SDB600 exhibited significantly improved adsorption capacities for TC (269.39 mg/g) and Cu2+ (271.65 mg/g) compared to the pristine biochar. The composite demonstrated excellent resistance to interference from common coexisting ions and maintained a high TC removal efficiency of 83.28% after four adsorption–desorption cycles, with minimal iron leaching. Adsorption kinetics were best described by the pseudo-second-order model, and isotherm data fitted the Langmuir model, indicating a chemisorption-dominated process. Density functional theory (DFT) calculations provided atomic-level mechanistic validation, confirming the spontaneous coordination of Cu2+ to surface Fe–OH sites with a high binding energy of −2.05 eV and identifying the A-ring of TC as the dominant moiety for π–π electron donor–acceptor interactions with an adsorption energy of −1.85 eV. Furthermore, in the binary system, a metal-bridging effect was identified, where preadsorbed Cu2+ enhanced TC uptake by forming ternary complexes. This work provides a sustainable and efficient strategy for sludge valorization and the coremoval of organic and inorganic pollutants, highlighting the potential of sludge-derived adsorbents in practical wastewater treatment.

Glucose oxidase loaded Cu2+ based metal-organic framework for glutathione depletion/reactive oxygen species elevation enhanced chemotherapy

Jinzhi Xu,Yan Xu,Lixin Sun,Bei Lu,Xi Yan,Zhonghao Wang,Tong Zhang

Introduction The development of multidrug resistance (MDR) is a major cause for the failure of chemotherapy, which requires the aid of nanomedicine. Methods Here in our study, a Cu 2+ based metal-organic framework (COF) was firstly developed and employed as a carrier for the delivery of glucose oxidase (GOx) and doxorubicin (Dox) (COF/GOx/Dox) for the therapy of MDR lung cancers. Results Our results showed that the GOx can catalyze glucose and produce H 2 O 2 . In the mean time, the Cu 2+ can react with GSH and then transform into Cu + , which resulted in GSH depletion. Afterwards, the produced Cu + and H 2 O 2 trigger Fenton reaction to generate ROS to damage the redox equilibrium of cancer cells. Both effects contributed to the reverse of MDR in A549/Dox cells and finally resulted in significantly enhanced in vitro/in vivo anticancer performance. Discussion The combination of glutathione depletion/reactive oxygen species elevation might be a promising strategy to enhance the efficacy of chemotherapy and reverse MDR in cancers.

“Four-in-One” Nanozyme and Natural Enzyme Symbiotic System of Cu2-xSe−GOx for Cervical Cancer Therapy

Yubo Hu,Ke Wang,Cong Ye

Graphical The nanozyme@natural enzyme symbiotic system of Cu 2- x Se−GOx exhibited prominent “four-in-one” cascaded catalytic enzyme activities including catalase, peroxidase, glutathione oxidase and glucose oxidase. The ingenious Cu 2- x Se−GOx could significantly induce the accumulation of reactive oxygen species (ROS) at tumor regions; this results in a tumor microenvironment (TME)-activated “four-in-one” enzyme cascade that is lethal to cervical cancer. All in all, Cu 2- x Se−GOx can act as a high-performance and safe bioactive material for cancer treatment. Cervical cancer, as a common malignant tumor of the reproductive system, seriously threatens women‘s life and health, and is difficult to be cured by traditional treatments, such as surgery, chemotherapy and radiotherapy. Fortunately, tumor microenvironment (TME)-activated catalytic therapy with high efficiency and reduced off-target toxicity has emerged as a novel treatment model. Herein, we designed a “four-in-one” nanozyme and natural enzyme symbiotic system of Cu 2- x Se−GOx for TME-triggered cascaded catalytic enhanced cancer treatment. In response to unique TME, Cu 2- x Se with catalase activity could effectively catalyze over-expressed H 2 O 2 in cancer cells into O 2 . Subsequently, the glucose oxidase (GOx) could deplete intracellular glucose with the assistance of O 2 ; this not only achieves starvation therapy, but also regenerates H 2 O 2 to boost the generation of highly cytotoxic . OH due to the peroxidase activity of Cu 2- x Se. Moreover, although the free-radical scavenger glutathione (GSH) is overexpressed in tumor cells, Cu 2- x Se with glutathione oxidase activity could effectively consume GSH for enhanced ROS production. Thus, the “four-in-one” nanozyme@natural enzyme symbiotic system of Cu 2- x Se−GOx could induce significant ROS accumulation at the tumor regions, thus providing a potential approach for the treatment of cervical cancer.

Rapid determination of trace Cu2+ by an in-syringe membrane SPE and membrane solid-phase spectral technique

Long Li,Kehan Xu,Zuohua Huang,Xinxin Xu,Jibran Iqbal,Liang Zhao,Yiping Du

A new in-syringe membrane SPE and solid-phase visible spectral method was proposed for the rapid extraction and visible spectral determination of trace Cu2+. The chelation and membrane SPE can be accomplished in a syringe. The yellow Cu(DDTC)2 complex was separated using a polyethersulfone membrane from the sample solution. Then, the complex can be detected directly on the polyethersulfone membrane utilizing solid-phase visible absorbance spectra without elution. The proposed method simplified the experimental procedure and improved the sensitivity to the μg L−1 level. Furthermore, this method is environmentally friendly since it avoids the use of organic solvents. After the investigation of the influence of different variables on the membrane SPE procedure, water and blood plasma were analyzed to validate the proposed method. A LOD of 0.04 μg L−1 and recoveries of 96.0–103.7% confirmed that the present work can be applied for the determination of trace Cu2+ in water and blood plasma samples.

Silk fibroin-based coating with pH-dependent controlled release of Cu2+ for removal of implant bacterial infections

Tongyao Lin,Jinglin Zhang,Dongliang Huo,Fengjuan Yang,Jingxian Zhang,Langhuan Huang,Sui-Ping Deng,Shaozao Tan,Huifang Chen

The implantation of medical devices is frequently accompanied by the invasion of bacteria, which may lead to implant failure. Therefore, an intelligent and responsive coating seems particularly essential in hindering implant-associated infections. Herein, a self-defensive antimicrobial coating, accompanied by silk fibroin as a valve, was successfully prepared on the titanium ( [email protected] ) for pH-controlled release of Cu 2+ . The results showed that the layer could set free massive Cu 2+ to strive against E. coli and S. aureus for self-defense when exposed to a slightly acidic condition. By contrary, a little Cu 2+ was released in the physiological situation, which could avoid damage to the normal cells and showed excellent in vitro pH-dependent antibiosis. Besides, in vivo experiment confirmed that [email protected] could work as an antibacterial material to kill S. aureus keenly and display negligible toxicity in vivo . Consequently, the design provided support for endowing the layer with outstanding biocompatibility and addressing the issue of bacterial infection during the implantation of Ti substrates.
