The exploration of efficient hazardous chemical sensing technology is of great importance to protect the environment and maintain public security. Here, a design strategy of a trimode upconversion nanoprobe was developed for H2O2 detection through establishing the inner filter effect (IFE) between the anthracene-based probes and the NaYF4:Yb/Tm/Er upconversion nanoparticles (UCNPs). The trimode nanoprobe exhibits superior H2O2 detection performance, including a low limit of detection (LOD) of 152 nM, fast response (<3 s), outstanding selectivity, and robust anti-interference ability even in the presence of 15 common interferents. Furthermore, the practical utility of this design is conclusively demonstrated through a sensing chip capable of accurately and selectively identifying H2O2 even in the presence of more than 7 types of coexisting multicolor interferents. This sensing strategy, characterized by its strong anti-interference capability even when facing extremely complex real-world samples, is expected to establish a promising framework for ultra-accurate detection of hazardous substances.

Advanced data storage and anti-counterfeiting technologies are critical in addressing modern information safety. In this work, novel nanocomposites of Eu3+, Tb3+, Zn2+ and -hyaluronan for down-conversion and NaGdF4:Yb,Er nanoparticles for up-conversion have been synthesized and applied for innovative anticounterfeiting luminescent inks. These inks exhibit excellent fluorescence under dual UV or NIR excitation, and sensitive quenching in response to some ions or compounds, enabling multi-level data encoding, reversible Cu2+ detection, and dynamic information concealment. Utilizing electrohydrodynamic (EHD) printing, we achieved precise patterns with line widths of approximately 20–50 μm, facilitating dynamic information concealment and secure data storage. The printed patterns, invisible under natural light, displayed distinct and bright fluorescence when stimulated by UV and NIR light, offering orthogonal readout capabilities for secure information retrieval. Additionally, the luminescent inks demonstrated remarkable sensitivity to Cu2+ ions, with fluorescence quenching efficiency reaching 85.3% at a Cu2+ concentration of 100 μg mL−1. The quenching process was reversible upon introducing ethylenediaminetetraacetic acid (EDTA), enabling up to three cycles of fluorescence recovery and suppression with minor degradation (<15%) in intensity after repeated use. The high-resolution EHD printing technology combined with excellent sensing performance, such as a detection limit of 1.2 μg mL−1 for Cu2+, enables a promising strategy for advancing intelligent information systems and hazardous ion detection. This work offers a versatile solution for secure information coding, anti-counterfeiting, and environmental sensing, providing a significant advancement in multifunctional luminescent materials, especially for smart quadruplicate information coding and hazard sensing, with dual UV/IR excitation.

A-type dimeric epigallocatechin-3-gallate (A-type-EGCG-dimer, AEd), a new proanthocyanidins dimer from persimmon fruits, has been shown to have health benefit effects. However, A-type-EGCG-dimer affects gluose metabolism in the liver and the underlying mechanism is not clarified. The present study aims to examine the protective effects of A-type-EGCG-dimer on Pb-induced hepatic insulin resistance, endoplasmic reticulum (ER) stress and apoptosis in rats. Male wistar rats exposed to 0.05% w/v Pb acetate in the drinking water with or without A-type-EGCG-dimer coadministration (200 mg/kg body weight/day, intragastrically) for three months. We found that A-type-EGCG-dimer and pioglitazone supplementation significantly deceased glucose and insulin levels in plasma as compared with the Pb group. A-type-EGCG-dimer markedly prevents Pb-induced oxidative stress , ER stress and apoptosis in livers. A-type-EGCG-dimer and pioglitazone reduced the expression levels of the GRP78, PEPCK , G6Pase , p-PERK, p-IRE1, p -JNK, ATF4, CHOP and increased p -AKT in livers of the Pb group. Moreover, A-type-EGCG-dimer reduced ROS production and restored the activities of SOD and GPx in livers. A-type-EGCG-dimer decreased Bax, cytosolic cytochrome c and cleaved caspase-3 and increased Bcl-2 in livers of Pb-exposed rats. Our results suggest that A-type-EGCG-dimer might be a potential natural candidate for the prevention of hepatic insulin resistance and apoptosis induced by Pb.

Background Sorafenib (Sora), a multi-target tyrosine kinase inhibitor, is widely recognized as a standard chemotherapy treatment for advanced hepatocellular carcinoma (HCC). However, drug resistance mechanisms hinder its anticancer efficacy. Derived from Withania somnifera , Withaferin A (WA) exhibits remarkable anti-tumor properties as a natural bioactive compound. This study aimed to examine the mechanisms that underlie the impacts of Sora and WA co-treatment on HCC. Methods Cell proliferation was evaluated through colony formation and MTT assays. Flow cytometry was employed to determine cellular apoptosis and reactive oxygen species (ROS) levels. The evaluation of apoptosis-related protein levels, DNA damage, and endoplasmic reticulum stress was conducte utilizing IHC staining and western blotting. Moreover, the caspase inhibitor Z-VAD-FMK, ATF4 siRNA, ROS scavenger N-acetyl cysteine (NAC), and TrxR1 shRNA were used to elucidate the underlying signaling pathways. To validate the antitumor effects of Sora/WA co-treatment, in vivo experiments were ultimately executed using Huh7 xenografts. Results Sora/WA co-treatment demonstrated significant synergistic antitumor impacts both in vivo and in vitro . Mechanistically, the enhanced antitumor impact of Sora by WA was achieved through the inhibition of TrxR1 activity, resulting in ROS accumulation. Moreover, ROS generation induced the activation of DNA damage and endoplasmic reticulum (ER) stress pathways, eventually triggering cellular apoptosis. Pre-treatment with the antioxidant NAC significantly inhibited ROS generation, ER stress, DNA damage, and apoptosis induced by Sora/WA co-treatment. Additionally, the inhibition of ATF4 by small interfering RNA (siRNA) attenuated Sora/WA co-treatment-induced apoptosis. In vivo , Sora/WA co-treatment significantly suppressed tumor growth in HCC xenograft models and decreased TrxR1 activity in tumor tissues. Conclusion Our study suggests that WA synergistically enhances the antitumor effect of Sora, offering promising implications for evolving treatment approaches for HCC.

Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer with limited targeted therapeutic options, underscoring the urgent need for novel treatment strategies. Feilike (FLK), a Traditional Chinese Medicine formula with heat-clearing and detoxifying properties, aligns with key pathological features implicated in breast cancer progression. In addition, several of its components have demonstrated anti-tumor activity, positioning FLK as a potential therapeutic candidate for TNBC. In this study, we employed an integrated approach combining network pharmacology, transcriptomic analysis, and experimental validation to investigate the anti-TNBC effects of FLK. Our results demonstrate that FLK significantly inhibits the proliferation of TNBC cell lines and patient-derived organoids and induces typical pyroptotic features, including cell swelling and increased lactate dehydrogenase (LDH) release. Mechanistically, FLK triggers a mutant p53 signaling cascade involving calcium dysregulation, endoplasmic reticulum stress (ERS) activation, mitochondrial dysfunction, and reactive oxygen species (ROS) accumulation, which collectively activate the P38/JNK–Caspase-3/GSDME pathway to induce pyroptosis. In vivo, FLK markedly suppresses tumor growth in a 4T1 orthotopic mouse model and enhances the anti-tumor efficacy of Cyclophosphamide. Furthermore, Licochalcone B (LCB) is identified as a key bioactive constituent that recapitulates the pyroptosis-inducing effects of FLK. Collectively, our findings uncover a previously unrecognized mutant p53–ERS–ROS–MAPK signaling axis underlying FLK-induced pyroptosis and provide mechanistic insight and experimental evidence supporting the repurposing of FLK as a potential therapeutic strategy for TNBC.

Accurate detection of temperature and oxygen concentration at the cellular level is important in tumor diagnosis. Multifunctional nanocomposites are described that consist of upconversion luminescent nanoparticles capped with mesoporous silica and loaded with an oxygen-sensitive luminescent ruthenium complex. The nanocomposites of type NaYF 4 :Yb,Er@NaYF 4 @mSiO 2 -Ru have two modes of operation: Its red downconversion luminescence (at excitation/emission peaks of 455/606 nm) is quenched by oxygen (O 2 ), and this is used to sense and image O 2 . The green upconversion luminescence (typically acquired at excitation/emission wavelengths of 980/525 and 544 nm), in turn, is used to measure temperature. The nanocomposites were then applied to dual mode in-vitro imaging of temperature and O 2 in hepatocellular carcinoma cells (HepG-2).
A new multifunctional nanocomposite of mesoporous silica coating NaYF 4 :Yb,Er@NaYF 4 upconversion nanopaticless loaded with ruthenium (II) complex has been designed with both green upconversion and red downconversion luminescence for temperature detection, oxygen sensing and dual mode cell imaging.

Owing to their excellent photoelectric and optical properties, lead halide perovskite quantum dots (PeQDs) have attracted great attention and have broad applications in various fields. However, the shortcomings of poor stability and short-wavelength (ultraviolet/visible) excitation restrict their practical applications. Herein, to address the above issues, we report a polyacrylonitrile-based composite film containing CsPbX3 (X = Br and I) PeQDs and β-NaYF4:Yb/Ln (Ln = Tm and Er) upconversion nanoparticles (UCNPs) via an electrospinning strategy. Interestingly, all-inorganic CsPbX3 PeQDs are synthesized through an in situ growth process in polyacrylonitrile nanofibers, which provide a polymer matrix for the protection of PeQDs against decomposition under the effect of an external environment. Upon 980 nm laser irradiation, lanthanide-sensitized UC luminescence of stable CsPbX3 can be realized through radiative energy transfer from Tm3+/Er3+ ions to PeQDs. Consequently, dual-mode luminescence, including downshifting and UC with color-tunable outputs, can be observed from PeQDs under dual-wavelength excitation (UV and NIR). Benefiting from the special luminescent feathers (temperature-sensitive UC and dual-mode luminescence), the as-obtained composite nanofibers exhibit great potential in high-level anticounterfeiting and accurate noncontact thermometry.

In this study, (Ce 0.2 Zr 0.2 La 0.2 Yb 0.2 Ln 0.2 )O 2- δ (Ln = Er, Y, Sm; labelled as H-Er, H-Y, H-Sm) high-entropy fluorite oxides (HEFOs) are designed by standard deviation and first-principles calculation. These HEFOs are subsequently synthesized via solid-phase reaction method, and the phase formation, microstructure, mechanical and thermal properties of the three samples have been systematically investigated. The results show that the three samples, exhibiting a single-phase fluorite structure, are successfully prepared after sintering at 1600 °C, and all the elements are uniformly distributed with no elemental segregation and agglomeration. The calculated δ of H-Er, H-Y and H-Sm are 0.3399, 0.3668 and 0.3458, respectively, so the chemical formula of three HEFOs are (Ce 0.2 Zr 0.2 La 0.2 Yb 0.2 Er 0.2 )O 1.6601 , (Ce 0.2 Zr 0.2 La 0.2 Yb 0.2 Y 0.2 )O 1.6332 and (Ce 0.2 Zr 0.2 La 0.2 Yb 0.2 Sm 0.2 )O 1.6542 , respectively. Additionally, the effects of rare earth ions (Er, Y and Sm) on the hardness, Young's modulus and thermal conductivity of HEFOs are analyzed. H-Y exhibits the highest hardness value and largest Young's modulus among the three HEFOs, primarily due to the largest mean acoustic velocity and cohesion energy in absolute value, smallest AGS and average bond lengths. The thermal conductivities ( κ ) of the three samples are only 1.465–1.598 W m −1 K −1 at 1000 °C. Furthermore, the thermal conductivities of the three samples are significantly lower than that of 8YSZ within the temperature range from 25 °C to 1000 °C, and H-Sm exhibits the lowest thermal conductivity in this temperature range. Consequently, they are anticipated to have promising applications in the field of thermal barrier coatings (TBCs).

KSrGd(PO 4 ) 2 :Ln 3+ (Ln = Ce, Sm, Eu, Tb, Dy, Dy/Eu, Yb/Ho, Yb/Er, Yb/Tm) phosphors were synthesized by high temperature solid state method. Doping the KSrGd(PO 4 ) 2 matrix with 5%Dy 3+ ions resulted in white light emission under 345 nm excitation. The calculated color temperature (CCT) of this white light is 5523 K, indicating as positive white light. It was found that the luminous intensity of the obtained KSrGd(PO 4 ) 2 :5%Dy 3+ at 200 °C was 76.80 % of that at room temperature, showing good thermal stability as well as possibility of application in white LED. Under the excitation at 392 nm, the fluorescence quantum yield of KSrGd(PO 4 ) 2 :10%Eu 3+ is 79.92 %. Energy transfer between Dy 3+ and Eu 3+ is observed in KSrGd(PO 4 ) 2 :Dy 3+ /Eu 3+ samples, with identified mechanism as the quadrupole-quadrupole interaction. Tunable luminescence ranging from white to yellow then to orange-red is achieved by adjusting the doping concentration of Eu 3+ in KSrGd(PO 4 ) 2 :5%Dy 3+ , x%Eu 3+ . Under the excitation of 980 nm near-infrared light, the prepared KSrGd(PO 4 ) 2 :Yb 3+ /Ho 3+ , KSrGd(PO 4 ) 2 :Yb 3+ /Er 3+ and KSrGd(PO 4 ) 2 :Yb 3+ /Tm 3+ samples showed up-conversion emissions of red, yellow and blue, respectively, with two-photon processes. In KSrGd(PO 4 ) 2 matrix, multi-color and multi-security marks were successfully prepared by utilizing up/down conversion luminescence characteristics, which obtained good anti-counterfeiting effect and improved anti-counterfeiting security. Therefore, the obtained KSrGd(PO 4 ) 2 : Ln 3+ phosphors exhibit promising application prospects in the fields of WLED and multiple anti-counterfeiting.

Photocatalytic hydrogen production with simultaneous degradation of organic pollutants by using solar energy provide a promising strategy to solve the energy and environmental issue in the future. However, for onefold wide band-gap semiconductor, the poor photocatalytic efficiency limits its application on a large scale due to its easy recombination of photo-generated electron-hole pairs and low solar energy utilization. Herein, a novel Z-scheme BiVO 4 (040)-Bi-(NaYF 4 :Er,Yb,Tm@BiOBr)/Bi photocatalyst is prepared via photo-assisted isoelectric point method. In this composite photocatalyst, up-convention luminescence material, NaYF 4 :Er,Yb,Tm, can offer more ultraviolet light and visible light to intensify the activations of BiOBr and metal Bi nanoparticles, respectively. Furthermore, the formation of Z-scheme BiVO 4 (040)-BiOBr photocatalystic sytem, the spatial separation of the BiVO 4 (040) and BiVO 4 (110) facets and the presence of metal Bi surface plasmon resonance effect are contributed to the high-efficiency separation of the photo-generated electron-hole pairs. The experimental results show that as-prepared Z-scheme BiVO 4 (040)-Bi-(NaYF 4 :Er,Yb,Tm@BiOBr)/Bi photocatalyst exhibits advanced photocatalytic hydrogen production amount (192.0 μm) and excellent degradation ratio of malachite green (88.51 %) under simulated sunlight irradiation for 5.0 h. The apparent quantum yield of the Z-scheme BiVO 4 (040)-Bi-(NaYF 4 :Er,Yb,Tm@BiOBr)/Bi photocatalyst at 420 nm reaches a specific value of 5.8 %. In addition, Z-scheme BiVO 4 (040)-Bi-(NaYF 4 :Er,Yb,Tm@BiOBr)/Bi photocatalyst also remains a high stability in the photocatalytic hydrogen production with simultaneous degradation of malachite green within six reuse. At last, the possible reaction mechanism and degradation pathways of malachite green caused by Z-scheme BiVO 4 (040)-Bi-(NaYF 4 :Er,Yb,Tm@BiOBr)/Bi photocatalyst under simulated sunlight irradiation are put forward.

Rare-earth doped upconversion nanoparticles (UCNPs) have attracted wide-ranging interest due to their potential applications in bioimaging and biolabeling, where a high emission intensity and small particle size are needed. However, common synthesis processes can be somewhat complex or require additional reactants, and the use of small-size NPs is limited by their weak luminescence intensity. In this work, uniform, monodisperse ∼5 nm β-NaGdF4:20% Yb/2% Er NPs were synthesized via a simple one-pot synthesis method in the high boiling-point solvents 1-octadecene (ODE) and oleic acid (OA) using a single dose of NaF at an ultralow NaF concentration of NaF/RE = 4. The effects of NaF/RE, the reaction temperature, and the reaction time on the size and phase of the particles were investigated. High-quality β-NaGdF4:20% Yb/2% Er@β-NaGdF4:20% Yb/10% Nd core–shell NPs with a size of ∼13 nm, enhanced UC luminescence, long UC lifetimes (τ) for green (τ ≈ 150 μs) and red (τ ≈ 105 μs) emission under 980 and 808 nm excitation, revealed via lifetime imaging, and obvious emission in the near-infrared (NIR) II region were synthesized. The synthesized core–shell NPs may result in a decrease in the overheating effect and allow for a deep penetration depth when further used for bioimaging applications. This work could inspire the synthesis of other high-quality RE-doped UCNPs via adding a single dose of a small amount of NaF and could lay the foundation for the use of Gd-based UCNPs in biomedical applications.

Exendin-4 (ex-4) is a long-acting glucagon-like peptide-1 receptor (GLP-1R) agonist which exerts beneficial effects on glycemic control and promotes cell viability. In the present study, we investigated the anti-apoptotic effects of ex-4, as well as the potential mechanisms responsible for these effects in rat bone marrow-derived mesenchymal stem cells (BM-MSCs) under conditions of oxygen, glucose and serum deprivation (OGD). The apoptosis of the MSCs was induced by subjecting the cells to OGD conditions for 4 h and was detected by Annexin V/PI and Hoechst 33258 staining. The MSCs were pre-conditioned with ex-4 for 12 h prior to being subjected to OGD conditions, and the expression levels of an apoptotic marker (cleaved caspase-3), endoplasmic reticulum (ER) stress markers [phosphorylated (p-)protein kinase RNA-like endoplasmic reticulum kinase (PERK), PERK, binding immunoglobulin protein (BIP), activating transcription factor 4 (ATF-4) and C/EBP homologous protein (CHOP)], as well as those of a survival marker (Bcl-2) were measured by western blot analysis. Furthermore, the mRNA levels of ATF-4 and CHOP were determined by RT-qPCR. ELISA was used to examine the activity of intracellular cAMP. Moreover, the GLP-1R antagonist, exendin9-39 (ex9-39), the protein kinase A (PKA) inhibitor, H89, and small interfering RNA (siRNA) targeting rat ATF-4 and CHOP were co-incubated with the MSCs. The apoptotic rate was markedly diminished following pre-conditioning with ex-4 in a dose‑dependent manner (P<0.05). The ER stress markers, p-PERK, BIP, ATF-4 and CHOP, were upregulated in the cells subjected to OGD conditions. Ex-4 pre-conditioning significantly decreased the mRNA and protein levels of ATF-4 and CHOP (P<0.05), and increased the activity of intracellular cAMP (P<0.05). Furthermore, the anti-apoptotic effects of ex-4 were almost reversed by treatment with either H89 or ex9-39 (P<0.05); transfection with siRNA-CHOP significantly reduced the apoptotic rate of the MSCs and did not impair the cytoprotective effects of ex-4. Taken together, these findings suggest that ex-4 protects rat BM-MSCs from OGD-induced apoptosis through the activation of the PKA/cAMP pathway and the attenuation of the ER stress signaling pathway. Ex-4 may thus prove to be a therapeutic agent with the potential to improve the viability of MSCs in the ischemic milieu, and consequently, to optimize the therapeutic effects of MSC therapy in acute myocardial infarction.
