Skin-interfaced self-powered pressure and strain sensors based on fish gelatin-based hydrogel for wireless wound strain and human motion detection

Ren Yan,Qizeng Sun,Xuewen Shi,Zhaoqing Sun,Shenxing Tan,Biao Tang,Wenting Chen,Fei Liang,Hai-Dong Yu,Wei Huang

Biomass-based hydrogels, due to their excellent biocompatibility, can behave as wearable monitoring platforms for healthcare applications and triboelectric sensing devices. However, low tissue adhesiveness, poor stretchability, and bacterial susceptibility cause them to fail to adapt to specific joints with complex movements. Herein, we report a novel biomass-based hydrogel by integrating fish gelatin into polymer networks with in-situ formation of silver nanoparticles (denoted as FG-Ag hydrogel), which achieves great stretchability (2600%), excellent sensitivity (gauge factor = 4), and strong self-adhesion simultaneously. The FG-Ag hydrogel also features a robust antibacterial activity for Escherichia coli and Staphylococcus aureus . Benefitting from these advantages, we have developed a FG-Ag hydrogel-based wearable strain sensor for diverse human-motion detections such as elbow, knee, finger, and wrist bending. Moreover, we have demonstrated an FG-Ag hydrogel-based prototype with an integrated wireless system for real-time strain monitoring on the wound sites. The FG-Ag hydrogel could significantly reduce bacterial infection in vivo and effectively promote wound healing. Additionally, the self-powered pressure sensor and the biomechanical energy harvester also have been demonstrated by the FG-Ag hydrogel based triboelectric nanogenerator (TENG). Accordingly, such FG assistance of hydrogel-based skin-interfaced electronics could provide adequately delicate biomechanical information related to the general health, which furnishes essential technical support for cost-effective, all-green, and highly precise personalized health assessment.

Determination of 26 endocrine disrupting chemicals in fish and water using modified QuEChERS combined with solid-phase extraction and UHPLC-MS/MS

Mei Yang,Youning Ma,Wenjun Gui,Yiping Ren,Guonian Zhu,Yihua Liu

Endocrine-disrupting chemicals (EDCs) in the environment have adverse effects on humans and wildlife. A method based on ultrahigh performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) for the determination of 26 EDCs (including five estrogens, eight androgens, three progestogens, six glucocorticoids, two mineralocorticoids and two thyroid hormones) in fish and water was developed. Various experimental parameters that could affect the extraction efficiencies were investigated in detail. The sample was extracted by a modified QuEChERS (Quick, Easy, Cheap, Effective, Rugged, and Safe) method with 20 mL of acetonitrile (for fish) or 5 mL of ethyl acetate (for water), and then cleaned-up using an Oasis HLB SPE (solid-phase extraction) cartridge. The analytes were quantified using an isotope-labelled internal standard and recoveries between 69.1% and 120.5% were obtained. The relative standard deviation of inter- and intra-day analyses for all the compounds was below 20%. The detection limits ranged from 0.01 to 0.98 ng mL−1 for water and 0.01 to 9.04 ng g−1 for fish. For real samples, progesterone and trenbolone were detected in zebrafish (Danio rerio) samples at 5.73 ± 0.21 and 7.45 ± 0.34 ng g−1, respectively. There was no target analyte detected in tap-water samples. The developed method would be useful for monitoring EDC abuse in fishery, potential EDC screening and risk assessment in aquatic toxicology.

Comparison and enrichment of sn-2 palmitoyl triacylglycerols (OPO/OPL) in fish oil for its potential application as human milk fat substitutes

Dongyin Liu,Jingtao Cui,Ruibing Zhou,Chen He,Jun Cao,Chuan Li

Triacylglycerols (TAG) are differences in fatty acid distributions between infant formula and human milk. In this study, fish oil (Tilapia, Golden pompano, Tiger grouper, and Basa) showed the potential as the source of human milk fat substitutes by comparing TAG profiles with infant formula and human milk. The total lipids and TAG of fish were concentrated in the by-products of fish (head and viscera) and contained high levels of palmitic acid, oleic acid, and linoleic acid. Compared with infant formula, fish oil was closer to human milk in sn -2 fatty acid distribution, and sn -2 palmitic acid level in fish oil exceeded 52 % of total palmitic acid, Golden pompano head was the highest (64.46 %). Further research showed that the content of sn -2 palmitoyl TAG (OPO and OPL dominated) increased from 157.16 mg/g TAG to 305.18 mg/g TAG by isopropanol enrichment (solid–liquid ratio: 1:4, temperature: −12 °C, time: 4 h).

Self-assembly PS@dual-emission ratiometric fluorescence probe coupled with core-shell structured MIP for the detection of malachite green in fish

Hui Ran,Zheng-zhong Lin,Cheng-yi Hong,Jun Zeng,Qiu-Hong Yao,Zhi-Yong Huang

Rapid detection of malachite green (MG) by fluorescence probes of CdTe quantum dots (QDs) coated with molecularly imprinted polymer (MIP) has been reported, but low sensitivity, slow response and narrow linear range were the main disadvantages for these MIP@QDs probes. Here, an ultra-sensitive ratiometric fluorescence probe based on self-assembly functionalized polystyrene (PS) and core-shell structured MIP was developed for the detection of MG in fish. After modified with poly(diallyldimethylammonium chloride) and hexadecyl trimethyl ammonium bromide, CdTe QDs and silica encapsulated carbon quantum dots (CDs@SiO 2 ) with negative charges, used as detection and reference fluorophores, were respectively self-assembled on the surface of PS to form PS-QDs&CDs. And a core-shell structured MIP layer was fabricated on the surface of PS-QDs&CDs to form a ratiometric fluorescence probe of MIP@PS-QDs&CDs with sol-gel method using MG, (3-aminopropyl)triethoxy-silan and tetraethyl orthosilicate as template, functional monomer and cross-linker, respectively. With intensive fluorescence densities of QDs&CDs in nucleus, the fluorescence emission of probe at λ em of 630 nm could be linearly quenched by MG, while the fluorescence emission at λ em of 430 nm remained unchanged. The probe of MIP@PS-QDs&CDs owned good fluorescence stability (RSD ≤2.3%), fast response time (5 min), wide linear range (0.1–32 μmol L −1 ) and low detection limit (5.6 μg kg −1 ), and has been applied to the detection of MG in fish samples with recoveries of 94.3–110.2%. The result validated that the as-prepared probe of MIP@PS-QDs&CDs had an excellent performance for MG detection in fish samples.

Preservative effects of fish gelatin coating enriched with CUR/βCD emulsion on grass carp (Ctenopharyngodon idellus) fillets during storage at 4 °C

Xinyu Sun,Xiaoban Guo,Mingyu Ji,Jiulin Wu,Wenjin Zhu,Jianhua Wang,Cui Cheng,Li Chen,Qiqing Zhang

This study investigated the effects of a novel edible coating combining fish gelatin with curcumin/β-cyclodextrin (CUR/βCD) emulsion on the quality of grass carp fillets (GCFs) during storage at 4 °C. For all samples, the quality parameters, including weight loss, pH, total volatile basic nitrogen (TVB-N), peroxide value (PV), thiobarbituric acid (TBA) value, SDS-PAGE, free amino acids (FAA), microbiological (total viable counts (TVC), Pseudomonas counts, yeasts and molds, and H 2 S-producing bacteria), color and sensorial characteristics, were tested periodically. The coatings containing CUR/βCD emulsions exhibited better preservative effects than gelatin/βCD coating. Therefore, fish gelatin coating enriched with CUR/βCD emulsion can be used as an effective way to maintain the quality of GCF and extend its shelf life during storage at 4 °C.

Ion-pairing reversed-phase chromatography coupled to inductively coupled plasma mass spectrometry as a tool to determine mercurial species in freshwater fish

Heyong Cheng,Xiaopan Chen,Lihuan Shen,Yuanchao Wang,Zigang Xu,Jinhua Liu

Most of analytical community is focused on reversed phase high performance liquid chromatography (RP-HPLC) for mercury speciation by employing mobile phases comprising of high salts and moderate amounts of organic solvents. This study aims at rapid mercury speciation analysis by ion-pairing RP-HPLC with inductively coupled plasma mass spectrometry (ICP-MS) detection only using low salts for the sake of green analytical chemistry. Two ion-pairing HPLC methods were developed on individual usage of positively and negatively charged ion-pairing reagents (tetrabutylammonium hydroxide –TBAH and sodium dodecylbenzene sulfonate –SDBS), where sodium 3-mercapto-1-propysulfonate (MPS) and l -cysteine (Cys) were individually added in mobile phases to transform mercury species into negative and positive Hg-complexes for good resolution. Addition of phenylalanine was also utilized for rapid baseline separation in combination of short C 18 guard columns. Optimum mobile phases of 2.0   mM SDBS   +   2.0   mM Cys   +   1.0   mM Phe (pH 3.0) and 4.0   mM TBAH   +   2.0   mM MPS   +   2.0   mM Phe (pH 6.0) both achieved baseline separation of inorganic mercury (Hg 2+ ), methylmercury (MeHg), ethylmercury (EtHg) and phenylmercury (PhHg) on two consecutive 12.5-mm C 18 columns. The former mobile phase was selected for mercury speciation in freshwater fish because of short separation time (3.0   min). Detection limits of 0.015 for Hg 2+ , 0.014 for MeHg, 0.028 for EtHg and 0.042   μg   L −1 for PhHg were obtained along with satisfactory precisions of peak height and area (1.0-2.8% for 5.0   μg   L −1 Hg-mixture standard). Good accordance of determined values of MeHg and total mercury in certified reference materials of fish tissue (GBW 10029) and tuna fish (BCR-463) with certified values as well as good recoveries (91–106%) proved good accuracy of the proposed method. An example application to freshwater fish indicated its potential in routine analysis, where MeHg was presented at 3.7–20.3   μg   kg −1 as the dominate species.

A highly selective dual-signal response ratiometric fluorescence sensing strategy for malachite green in fish based on carbon dots/copper nanoclusters nanocomposite

Jun-Qiu Zhang,Xiao-Fang Shen,Jun Liu

Malachite green (MG), a widely used antiparasitic agent, poses health risks to human due to its genotoxic and carcinogenic properties. Herein, a stable dual-emission fluoroprobe of carbon dots/copper nanoclusters is prepared for highly selective detection of MG based on the inner filter effect. This probe exhibits characteristic emission bands at 435 and 625 nm when excited at 376 nm. After adding MG, the both emission signals were significantly quenched, and the ratio of fluorescence intensity (F 435 /F 625 ) was linearly related to the concentration of MG in the range of 0.05–40 μmol L −1 with a limit of detection of 18.2 nmol L −1 . Meanwhile, the two signals exhibit linear relationships with the concentration of MG, respectively, and the corresponding detection results were consistent. The fluoroprobe was successfully used for the detection of MG in fish samples with the recoveries ranging from 96.0% to 103.8% and a relative standard deviation of <3.3%.

Construction of magnetic azo-linked porous polymer for highly-efficient enrichment and separation of phenolic endocrine disruptors from environmental water and fish

Guijiao Zhao,Chenhuan Wang,Min Kang,Lin Hao,Weihua Liu,Zhi Wang,Xiaodong Shi,Qiuhua Wu

Developing effective methods for highly sensitive detection of phenolic endocrine disruptors (EDCs) is especially urgent. Herein, a magnetic hydroxyl-functional porous organic polymer (M-FH-POP) was facilely synthesized by green diazo-couple reaction using basic fuchsin and hesperetin as monomer for the first time. M-FH-POP delivered superior adsorption performance for phenolic EDCs. The adsorption mechanism was hydrogen bonds, hydrophobic interaction and π-π interplay. With M-FH-POP as adsorbent, a magnetic solid phase extraction method was established for extracting trace phenolic EDCs (bisphenol A, 4- tert -butylphenol, bisphenol F and bisphenol B) in water and fish before ultra-high performance liquid chromatography tandem mass spectrometry analysis. The method displayed low detection limit (S/N = 3) of 0.05–0.15 ng mL −1 for water and 0.08–0.3 ng g −1 for fish. The spiked recoveries were 88.3 %–109.8 % with the relative standard deviations of 2.4 %–6.4 %. The method offers a new strategy for sensitive determination of phenolic EDCs in water and fish samples.

Partial replacement of fish-meal by soy protein concentrate and soybean meal based protein blend for juvenile Pacific white shrimp, Litopenaeus vannamei

Shi-wei Xie,Yong-jian Liu,Shuailin Zeng,Jin Niu,Li-xia Tian

An 8-week feeding trial was conducted to evaluate the effect of fish-meal replacement on growth performance, whole body composition, apparent digestibility coefficients and anti-oxidative ability of juvenile Pacific white shrimp , Litopenaeus vannamei . Five practical diets were formulated to contain graded levels (5, 10, 15, 20 and 25%) of fish-meal. Soy protein concentrate and soybean meal based protein blend was used to replace fish-meal, amino acids were supplemented in the low fish-meal diet to get similar amino acid profile as the high fish-meal diet. Weight gain, survival, feed conversion rate (FCR), feed intake and protein efficiency ratio (PER) did not show any significant differences between any of the groups. Alanine ratio in whole body were higher in shrimp fed high fish-meal diets while arginine decreased with the increasing fish-meal levels. The Fe content in whole body was significantly lower in shrimp fed the low fish-meal diets, while the other minerals showed no difference among all the groups. Apparent digestibility coefficients (ADC) of dry matter, protein, Ca, P, Cu, Zn and Mg were equal in all the treatments, while ADC of Fe decreased with the decreasing fish-meal levels. When dietary fish-meal decreased, the anti-oxidant related enzymes activity (Total antioxidant capacity (T-AOC) and Catalase (CAT)). Superoxide dismutase (SOD) was unaffected by treatments. Nitric oxide (NO) contents in hemolymph significantly decreased with the decreasing fish-meal levels, while Phenoloxidase (PO) was unaffected. The results indicated that growth performance and apparent digestibility coefficients of nutrients were not affected when fish-meal was replaced from 25% to 5%, but anti-oxidative ability and immune response were damaged after fish-meal level decreased to 15%. Statement of relevance The results of this manuscript contribute to the commercial feed industry of white shrimp, it will be helpful in fish meal replacement, which is very important to the sustainable development of aquaculture.

Fabricating photoelectrochemical aptasensor for selectively monitoring microcystin-LR residues in fish based on visible light-responsive BiOBr nanoflakes/N-doped graphene photoelectrode

Xiaojiao Du,Ding Jiang,Liming Dai,Lei Zhou,Nan Hao,Jing Qian,Baijing Qiu,Kun Wang

The presence of microcystins in fish has been augmenting the risk of toxicity to animal and human health. Herein, a selective and sensitive method for detecting microcystin-LR (MC-LR) in fish samples by integrating the photoelectrochemical (PEC) technique and the specific recognition ability of aptamer was developed. Specifically, as an efficient PEC transducer, the BiOBr nanoflakes/N-doped graphene p–n heterojunction electrode was utilized as the aptamer immobilization platform via the π–π stacking interaction, which would be a biosensor enabling the convenient and exquisite PEC analysis. Subsequently, the PEC response of constructed aptasensor was specific binding to MC-LR. Other isoforms did not interfere with the detection process, and thus, it could be applied for the highly selective determination of MC-LR level. Under the optimized condition, the PEC signal versus the logarithm of the MC-LR concentration was in good linear relationship ranging from 0.1   pM to 100   nM with detection limit about 0.03   pM. The constructed method was employed to analyze fish samples collected from the local supermarket. The overall analytical recovery of MC-LR in the fish matrices ranged from 97.8 to 101.6%, with relative standard deviation (RSD) of 2.52–5.14%, implying it would have great potential in farm product analysis.

Li Luo,Yuan Yang,Qiang Wang,Hai-pu Li,Zhou-fei Luo,Zhi-peng Qu,Zhao-guang Yang

The presence of endocrine disrupting chemicals such as 4- n -octylphenol, 4- n -nonylphenol and bisphenol A in environmental samples and artificial products may cause health problems for human and organisms. This developed method was efficient for monitoring of those tracing emerging contaminates and the Plackett-Burman factorial design introduced in extraction step was timesaving. Matrix effect was also considered to ensure proper quantitation. Good linearity (r 2   >   0.99), low Method Detection Limits (MDL) and Method Quantitation Limits (MQL) range from 0.18 to 0.54   ng/g and 0.60 to 1.80   ng/g were obtained. Three level spiking experiment (4, 10, 15   ng/g) showed recoveries range from 74% to 113% for 4- n -octylphenol and 4- n -nonylphenol with RSD range from 2% to 11%. The method was finally applied to the analysis of real fish samples taken from Hunan Province, China, offering a reliable and effective means for monitoring of trace emerging contaminates in fishes largely consumed by residents.

Fast Derivatization Followed by Gas Chromatography–Mass Spectrometry for Simultaneous Detection of Melamine, Ammeline, Ammelide, and Cyanuric Acid in Fish and Shrimp

Fan Yingming,Ma Xiaoguo,Li Zhongyang,Chen Meiqin

A sensitive and reliable method was proposed for the simultaneous detection of melamine, ammeline, ammelide, and cyanuric acid in fish and shrimp by gas chromatography–mass spectrometry after fast derivatization. The analytes were derivatized using a 99:1 mixture of N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA) and trimethylchlorosilane (TMCS) under microwave irradiation for 2 min with a power of 700 W, followed by determination with gas chromatography–mass spectrometry in selected ion monitoring mode. Conditions for extraction and derivatization of melamine and related compounds were investigated and optimized. Under the optimal experimental conditions, the limits of detection (LODs, S/N = 3) for melamine, ammeline, ammelide, and cyanuric acid were 0.006, 0.012, 0.013, and 0.010 mg kg −1 , respectively. The developed method was successfully applied to determine melamine and related compounds in fish and shrimp samples, and the spike recoveries were 82.5–103.1 % with relative standard deviations (RSDs) ( n  = 3) of 0.6–4.1 %.
