Doping of Cu(I) ions into CdS/ZnS core/shell nanocrystals through a cation exchange strategy

Mingrui Liu,Lijin Wang,Zhongyuan Guan,Zhenyang Liu,Yongsheng Zhu,Aiwei Tang

The optical properties of doped semiconductor nanocrystals (NCs) are strongly affected by the dopant position inside the host lattice. However, controlling the dopant position in Cu(I) doped core/shell semiconductor NCs is still unexplored due to the high ionic mobility of Cu(I) ions at the doping temperature. In this paper, we report the doping of Cu(I) ions into CdS/ZnS core/shell NCs via a cation exchange strategy. During the doping, the exciton emission of CdS NCs is reduced and the dopant emission becomes intensified as the doping level is increased from 0 to 1.1%, and the PL profile of the final product is very similar to that of uncoated CdS:Cu(I) NCs. Such an optical change is very different from that observed in the products synthesized by a conventional method, in which no exciton emission of CdS NCs is observed. During the Cu(I) doping process, the variation of the dopant emission may arise from the doping position change from the outside ZnS shell to the inside CdS core in the CdS/ZnS core/shell NCs. The deduction can be further verified by the optical change by direct heating of a small amount of Cu(I) doped CdS/ZnS NCs in a non-coordinating solvent. This work offers a cation exchange strategy for fabricating Cu(I) doped core/shell NCs with tuned dopant positions, which will open up rich opportunities for tuning the optoelectronic properties of Cu(I) doped semiconductor NCs.

Reporter Molecules Embedded Au@Ag Core-Shell Nanospheres as SERS Nanotags for Cardiac Troponin I Detection

Ding Wang,Yiru Zhao,Shen Zhang,Liping Bao,Huijun Li,Jingcheng Xu,Bin He,Xumin Hou

Rapid and accurate detection of acute myocardial infarction can improve patients’ chances of survival. Cardiac troponin I (cTn I) is an important diagnostic biomarker for acute myocardial infarction. However, current immunoassays are insufficient to accurately measure cTn I, as they have limited detection sensitivity and are time-consuming. Surface-enhanced Raman scattering (SERS) is a brilliant fingerprints diagnostic technique characterised by ultrasensitivity, fast response, and qualitative and quantitative analysis capabilities. In this study, reporter molecules (4-Mercaptobenzoic acid, 4-MBA) embedded[email protected]core-shell nanospheres as SERS nanotags were prepared for the detection of cTn I. As the Raman reporters were embedded between the core and the shell, they could be protected from the external environment and nanoparticle aggregation. Excellent SERS performances were obtained due to the enhanced local electromagnetic field in the gap of core and shell metals. In a standard phosphate buffered saline (PBS) environment, the limit of detection for cTn I was 0.0086 ng mL−1(8.6 ppt) with a good linear relationship. The excellent Raman detection performance was attributed to the localized surface plasmon resonance effect and strong electromagnetic field enhancement effect produced by the gap between the Au core and the Ag shell. The SERS nanotags we prepared were facile to synthesize, and the analysis procedure could be completed quickly (15 min), which made the detection of cTn I faster. Therefore, the proposed SERS nanotags have significant potential to be a faster and more accurate tool for acute myocardial infarction diagnostics. Keywords:[email protected]nanospheres;GERTs;SERS;cTn I detection;AMI

Enhanced Expression of Alcohol Dehydrogenase I in Pichia pastoris Reduces the Content of Acetaldehyde in Wines

Kun Geng,Ying Lin,Xueyun Zheng,Cheng Li,Shuting Chen,He Ling,Jun Yang,Xiangyu Zhu,Shuli Liang

Acetaldehyde is an important carbonyl compound commonly detected in wines. A high concentration of acetaldehyde can affect the flavor of wines and result in adverse effects on human health. Alcohol dehydrogenase I (ADH1) inSaccharomyces cerevisiaecatalyzes the reduction reaction of acetaldehyde into ethanol in the presence of cofactors, showing the potential to reduce the content of acetaldehyde in wines. In this study,ADH1was successfully expressed inPichia pastorisGS115 based on codon optimization. Then, the expression level ofADH1was enhanced by replacing its promoter with optimized promoters and increasing the copy number of the expression cassette, with ADH1 being purified using nickel column affinity chromatography. The enzymatic activity of purified ADH1 reached 605.44 ± 44.30 U/mg. The results of the effect of ADH1 on the content of acetaldehyde in wine revealed that the acetaldehyde content of wine samples was reduced from 168.05 ± 0.55 to 113.17 ± 6.08 mg/L with the addition of 5 mM NADH and the catalysis of ADH1, and from 135.53 ± 4.08 to 52.89 ± 2.20 mg/L through cofactor regeneration. Our study provides a novel approach to reducing the content of acetaldehyde in wines through enzymatic catalysis. Keywords:alcohol dehydrogenase I;Pichia pastoris;cofactor regeneration;multicopy;acetaldehyde;wine

A new pore-strength model for the quantitative strength prediction of ceramics under mode-I loading

Shuai Wang,Anzhe Wang,Pan Gao,Rui Bai,Junjie Liu,Peng Zhou,Dazhao Liu

Strength prediction under the action of pores is crucial for the safety of ceramics. Here, a quantitative prediction model of pore-strength response under mode-I loading was developed based on the new mapping relationship between stress intensity factor and tip radius. The new model was proved to have great application after comparison and analysis of the pore-strength test data of three typical ceramics as well as the pertinent data as-reported in the literature. Results indicated that when the tip radius of pore ( ρ ) exceeds the critical value ( ρ c  =  K Ic 2 /(π σ 0 2 )), the stress intensity factor increases as a power function with increasing ρ , at which point the strength decay is significantly weakened, and when the ρ is lower than the ρ c , the pore can be equated to a crack problem. This work can not only forecast and assess the failure strength of ceramics containing pores, but also provide guidance for the design of extremely dependable ceramics.

Supramolecular self-assembled nanoparticles camouflaged with neutrophil membrane to mitigate myocardial ischemia/reperfusion (I/R) injury

Xinman Hu,Fang-Yuan Chen,Jie Ding,Zihe Zhai,Huang Yang,Liyin Shen,Yang Zhu,Dong-Sheng Guo,Changyou Gao

The clinically restored blood supply post myocardial infarction (MI) will break the physiological and metabolic balance of the ischemic myocardial region, leading to a new injury known as myocardial ischemia/reperfusion (I/R) injury. Abundant oxygen brought by the fresh blood would trigger the damaged myocardium to produce excessive reactive oxygen species (ROS) and thereby inflammation along with the recruitment and infiltration of activated neutrophils and macrophages. Inspired by the active targeting process of inflammatory cells, a novel “neutrophil-like” nanoassembly (At@NQA) was prepared by modifying neutrophil membrane fragments onto At@QA NPs containing 1:1 (molar ratio) calixarene molecule (QA) and anti-inflammatory and anti-fibrotic atorvastatin calcium (At). Tail vein injection of the At@NQA could efficiently target the damaged myocardium, inhibit oxidative damage and inflammation, reduce myocardial fibrosis, increase myocardial troponin T (cTnT) and connexin 43 (Cx43) expression, and thus improve cardiac functions effectively.

Cu(I)/Cu(II) Released by Cu Nanoparticles Revealed Differential Cellular Toxicity Related to Mitochondrial Dysfunction

Xiangrui Wang,Wen-Xiong Wang

Cu-based nanoparticles (NPs) have been increasingly fabricated, and different Cu species (i.e., Cu+ and Cu2+) of these NPs are tuned to achieve differential physicochemical properties. Although ion release is one of the major toxic mechanisms of Cu-based NPs, differences in cytotoxicity between released Cu(I) and Cu(II) ions are largely unknown. In this study, the A549 cells exhibited a lower tolerance to Cu(I) compared with Cu(II) accumulation. Bioimaging of labile Cu(I) indicated that the change of the Cu(I) level upon CuO and Cu2O exposure displayed different trends. We then developed a novel method to selectively release Cu(I) and Cu(II) ions within the cells by designing CuxS shells for Cu2O and CuO NPs, respectively. This method confirmed that Cu(I) and Cu(II) exhibited different cytotoxicity mechanisms. Specifically, excess Cu(I) induced cell death through mitochondrial fragmentation, which further led to apoptosis, whereas Cu(II) resulted in cell cycle arrest at the S phase and induced reactive oxygen species generation. Cu(II) also led to mitochondrial fusion, which was likely due to the influence of the cell cycle. Our study first uncovered the difference between the cytotoxicity mechanisms of Cu(I) and Cu(II), which could be greatly beneficial for the green fabrication of engineered Cu-based NPs.

The Various Packing Structures of Tb@C82 (I, II) Isomers in Their Cocrystals with Ni(OEP)

Wei Dong,Qin Zhou,Wangqiang Shen,Le Yang,Peng Jin,Xing Lu,Yongfu Lian

Soot-containing terbium (Tb)-embedded fullerenes were prepared by evaporation of Tb4O7-doped graphite rods in an electric arc discharge chamber. After 1,2,4-trichlorobenzene extraction of the soot and rotary evaporation of the extract, a solid product was obtained and then dissolved into toluene by ultrasonication. Through a three-stage high-pressure liquid chromatographic (HPLC) process, Tb@C82(I, II) isomers were isolated from the toluene solution of fullerenes and metallofullerenes. With the success of the growth of cocrystals of Tb@C82(I, II) with Ni(OEP), the molecular structures of Tb@C82(I) and Tb@C82(II) were confirmed to be Tb@C2v(9)-C82and Tb@Cs(6)-C82, respectively, based on crystallographic data from X-ray single-crystal diffraction. Moreover, it was found that Tb@C82(I, II) isomers demonstrated different packing behaviors in their cocrystals with Ni(OEP). Tb@C2v(9)-C82forms a 1:1 cocrystal with Ni(OEP), in which Tb@C2v(9)-C82is aligned diagonally between the Ni(OEP) bilayers to form zigzag chains. In sharp contrast, Tb@Cs(6)-C82forms a 2:2 cocrystal with Ni(OEP), in which Tb@Cs(6)-C82forms a centrosymmetric dimer that is aligned linearly with Ni(OEP) pairs to form one-dimensional structures in the a–c lattice plane. In addition, the distance of a Ni atom in Ni(OEP) to the Cs(6)-C82cage is much shorter than that to the C2v(9)-C82one, indicative of a stronger π-π interaction between Ni(OEP) and the C82carbon cage in the cocrystal of Tb@CS(6)-C82and Ni(OEP). Density functional theory calculations reveal that the regionally selective dimerization of Tb@CS(6)-C82is the result of a dominant unpaired spin existing on a particular C atom of the CS(6)-C82cage.Keywords:metallofullerene;terbium;cocrystals;centrosymmetric dimer;unpaired spin

Enzyme activity termination by titanium carbide nanosheet and its application for the detection of deoxyribonuclease I

Guibin Peng,Bixia Lin,Manli Guo,Yujuan Cao,Ying Yu,Yumin Wang

Deoxyribonuclease I (DNase I) is a typical nuclease that plays key roles in many physiological processes and the development of a novel biosensing strategy for DNase I detection is of fundamental significance. In this study, a fluorescence biosensing nanoplatform based on a two-dimensional (2D) titanium carbide (Ti 3 C 2 ) nanosheet for sensitive and specific detection of DNase I was reported. Fluorophore-labeled single-stranded DNA (ssDNA) can be spontaneously and selectively adsorbed on Ti 3 C 2 nanosheet through the hydrogen bond and metal chelate interaction between phosphate groups of ssDNA and titanium of Ti 3 C 2 nanosheet, resulting in effective quenching of the fluorescence emitted by fluorophore. Notably, it was found the enzyme activity of DNase I will be terminated by the Ti 3 C 2 nanosheet. Therefore, the fluorophore-labeled ssDNA was firstly digested by DNase I and the “post-mixing” strategy of Ti 3 C 2 nanosheet was chosen to evaluate the enzyme activity of DNase I, which provided the possibility of improving the accuracy of the biosensing method. Experimental results demonstrated that this method can be utilized for quantitative analysis of DNase I activity and exhibited a low detection limit of 0.16 U/ml. Additionally, the evaluation of DNase I activity in human serum samples and the screening of inhibitors with this developed biosensing strategy were successfully realized, implying that it has high potential as a promising nanoplatform for nuclease analysis in bioanalytical and biomedical fields.

Simultaneous removal of Cs(I) and U(VI) by a novel magnetic AMP@PDA@Fe3O4 composite

Jiayi Li,Weixing Han,Heyao Liu,Minhua Su,Diyun Chen,Gang Song

The development and use of clean energy produced by nuclear power plants is becoming a major international trend, but it will inevitably lead to the problem of radionuclide pollution. Radioactive nuclear wastewater containing Cs and U and produced as a byproduct of coastal nuclear power production can cause substantial environmental pollution . In order to simultaneously remove radioactive cesium and uranium, polydopamine (PDA) inlaid ammonium phosphomolybdate (AMP) has established a hydrophilic functional structural film on the surface of Fe 3 O 4 , forming AMP@PDA@Fe 3 O 4  (APF) which is used to simultaneous remove Cs(I) and U(VI). Using the pH sensitive range of APF adsorption in the process of adsorbing Cs(I) and U(VI), synergistic adsorption can be achieved under the condition of weak acidity to neutral. The effects of temperature, and common cations on Cs(I) and U(VI) adsorption by APF were investigated, the adsorption was found to be sensitive to the presence of K + . The efficiency of Cs(I) and U(VI) adsorption by APF was 99.88% and 98.39%, respectively. The Cs(I) adsorption process was primarily ion exchange with NH 4 + within APF composite, whereas that for U(VI) was primarily C-O complexation. Both the Cs(I) and U(VI) adsorption reactions were consistent with second-order kinetic and Langmuir isothermal adsorption models. After five cycles of experiments the removal efficiency of Cs(I) and U(VI) can reach 64.88% and 79.81%, respectively. APF was also found to have high selectivity for Cs and U in seawater with high salt content at pH 6.0–7.0. In mimic contaminated seawater, the efficiency of Cs(I) and U(VI) removal was 97.65% and 84.63%, respectively. These findings provide references for the remove of Cs(I) and U(VI) from radioactive wastewater , making the process of energy production safer.

Dual-signal aptamer sensor based on polydopamine-gold nanoparticles and exonuclease I for ultrasensitive malathion detection

Guoli Xu,Jingzhou Hou,Yanan Zhao,Jing Bao,Mei Yang,Huanbao Fa,Yixia Yang,Li Li,Danqun Huo,Changjun Hou

A highly sensitive dual-signal aptamer sensor based on polydopamine-gold nanoparticles (PDA-AuNPs) and exonuclease I (Exo I) was developed for detection of malathion. Compared with traditional sensing elements, aptamer has many advantages, such as high affinity, superior specificity, strong stability and easy modification. The electrodeposition synthesis of PDA-AuNPs gave excellent biocompatibility and electrical conductivity on the sensor. With the addition of malathion, the specific interaction between malathion and its aptamer forced the aptamer to detach from the electrode surface and induced the capture probe to form a hairpin structure on the electrode surface. Exo I was added to motivate the autocatalytic target cycling which remarkably increased the current change of electrochemical signal over 2 times. Therefore, the promising strategy gave rise to an optional dual-signal current readout in both the signal-on of Fc and the signal-off of Tn. In this work, the prepared biosensor exhibited high sensitivity to malathion via the combination of dual-signal design and autocatalytic target cycling amplification. Under the optimized conditions, the proposed sensor showed a wide linear range from 0.5 to 600 ng/L malathion. It also exhibited excellent specificity, acceptable repeatability and good stability. The application of real samples obtained satisfactory recovery results, demonstrating a promising potential in food safety analysis.

A type I Bi2S3@ZnS core–shell structured photocatalyst for the selective photoelectrochemical sensing of Cu2+

Jiajia Xi,Hao Wang,Bihong Zhang,Xiaopeng Hu,Faqiong Zhao,Baizhao Zeng

A type I core/shell structured Bi2S3@ZnS photocatalyst was synthesized by an electrostatic interaction mechanism and used for the photoelectrochemical detection of Cu2+. The Bi2S3@ZnS composite showed excellent photoelectrochemical activity and stability due to the high photoelectrochemical activity of Bi2S3 and the anti-photocorrosion function of the ZnS shell. In addition, the ZnS shell also provided a transfer platform for the photogenerated holes through its acceptor states (i.e. VZn and IS) and suppressed the recombination of photogenerated carriers. The photocurrent signal of the obtained Bi2S3@ZnS/ITO electrode was much higher than that of pure Bi2S3 under visible-light irradiation. When it was used for the photoelectrochemical sensing of Cu2+, a wide detection range (0.003–30.0 μM) and a low detection limit (0.001 μM) were observed. The sensor also displayed high stability and acceptable selectivity. It was applied to detect real samples and good results were achieved.

Injectable self-crosslinking HA-SH/Col I blend hydrogels for in vitro construction of engineered cartilage

Yafang Chen,Junhui Sui,Qing Wang,Yijia Yin,Jun Liu,Qiguang Wang,Xianglong Han,Yong Sun,Yujiang Fan,Xingdong Zhang

The injectable self-crosslinking blend hydrogel by combination of collagen I and thiolated hyaluronic acid could alleviate collagen I contraction in vitro and overcome weak cell adhesive sites of hyaluronic acid. Five groups of injectable hydrogels with different ratios were prepared to investigate their gelation time, injection force, mechanical properties, swelling capacity and disintegration performance. These results indicated that Col 7 HA-SH 3 hydrogel achieved the optimal controlled and injectable effect, the gelation time was just ten seconds with injection force at 3.5 N, and the storage modulus of hydrogel could reach 11 kPa with frequency at 10 Hz. Furthermore, the phenotype maintaining, biocompatibility and chondrocytes proliferation were administrated by CLSM, SEM, histological staining, immuohistochemical staining, MTT test and glycoaminoglycans quantification. Similarly, the Col 7 HA-SH 3 blend hydrogel encapsulated chondrocytes presented most excellent proliferation potential, phenotype maintaining, biocompatibility and convenient operational characteristics. These findings might approach the underlying clinical application of blend hydrogel in cartilage repair.
