Thermosensitive nanoparticles can be activated by externally applying heat, either through laser irradiation or magnetic fields, to trigger the release of drug payloads. This controlled release mechanism ensures that drugs are specifically released at the tumor site, maximizing their effectiveness while minimizing systemic toxicity and adverse effects. However, its efficacy is limited by the low concentration of drugs at action sites, which is caused by no specific target to tumor sties. Herein, hyaluronic acid (HA), a gooey, slippery substance with CD44-targeting ability, was conjugated with a thermosensitive polymer poly(acrylamide-co-acrylonitrile) to produce tumor-targeting and thermosensitive polymeric nanocarrier (HA-P) with an upper critical solution temperature (UCST) at 45 °C, which further coloaded chemo-drug doxorubicin (DOX) and photosensitizer Indocyanine green (ICG) to prepare thermosensitive nanoreactors HA-P/DOX&ICG. With photosensitizer ICG acting as the “temperature control element”, HA-P/DOX&ICG nanoparticles can respond to temperature changes when receiving near-infrared irradiation and realize subsequent structure depolymerization for burst drug release when the ambient temperature was above 45 °C, achieving programmable and on-demand drug release for effective antitumor therapy. Tumor inhibition rate increased from 61.8 to 95.9% after laser irradiation. Furthermore, the prepared HA-P/DOX&ICG nanoparticles possess imaging properties, with ICG acting as a probe, enabling real-time monitoring of drug distribution and therapeutic response, facilitating precise treatment evaluation. These results provide enlightenment for the design of active tumor targeting and NIR-triggered programmable and on-demand drug release of thermosensitive nanoreactors for tumor therapy.

Cationic polymers have great potential for cancer therapy due to their unique interactions with cancer cells. However, their clinical application remains limited by their high toxicity. Here we show a cell membrane-targeting cationic polymer with antineoplastic activity (P mt ) and a second near-infrared (NIR-II) fluorescent biodegradable polymer with photosensitizer Bodipy units and reactive oxygen species (ROS) responsive thioketal bonds (P Bodipy ). Subsequently, these two polymers can self-assemble into antineoplastic nanoparticles (denoted mt-NP Bodipy ) which could further accumulate at the tumor and destroy cell membranes through electrostatic interactions, resulting in cell membrane destabilization. Meanwhile, the photosensitizer Bodipy produces ROS to induce damage to cell membranes, proteins, and DNAs to kill cancer cells concertedly, finally resulting in cell membrane lysis and cancer cell death. This work highlights the use of near-infrared light to spatially and temporarily control cationic polymers for photodynamic therapy, photo-immunotherapy, and NIR-II fluorescence for bio-imaging.

Although acceptor-donor-acceptor (A-D-A)-type molecules show advantages in constructing NIR absorbing photothermal agents (PTAs) due to their strong intramolecular charge transfer and molecular planarity, their applications in photothermal therapy (PTT) of tumor are insufficiently explored. In particular, the influence of ESP distribution on optical properties of A-D-A photosensitizers has not been investigated. Herein, we utilized the advantages of A-D-A-type molecules with large ESP difference to construct NIR absorbing PTA with sufficient extinction coefficient (ɛ) and high photothermal conversion efficiency (PCE). The calculation results of density functional theory (DFT) indicate that the large ESP difference of Y6 may increase intermolecular interactions and induce tight packing of the A-D-A photosensitizer. As a result, Y6 NP exhibited an obvious bathochromic shift of absorption peak from 711 nm to 822 nm, with a NIR-II emission extended to 1400 nm. Moreover, a sufficient ɛ of 3.34 L·g-1·cm-1 and PCE of 66.3% were attained, making Y6 NP suitable for PTT. With a concentration of 100 μg/mL, Y6 NPs in aqueous dispersion yielded a death rate of 93.4% for 4T1 cells upon an 808 nm laser irradiation (1 W·cm-2) for 10 min, which is comparable with the best results of recently reported PTT agents.

The second near-infrared window (NIR-II, 1000–1700 nm) emitting phosphor is a key component for the phosphor-converted light-emitting diode (pc-LED), which has garnered significant attention. However, it remains a major challenge to discover efficient NIR-II broadband phosphors that can be excited by blue LEDs. Herein, Cr3+ and Ni2+ ions were co-doped into the Li2ZnSn3O8 (LZSO) host. LZSO:Cr3+,Ni2+ phosphor achieves a broadband NIR-II emission peak at 1465 nm with a large full width at half maximum (FWHM) of 300 nm and internal quantum efficiency (IQE) of 43.31% under excitation at 426 nm because of the efficient energy transfer (ET) from Cr3+ to Ni2+. In addition, a dual-emissive doped phosphor across the NIR-I (700–1000 nm) and NIR-II regions with a higher sensitivity at physiological temperatures was obtained. The relative sensitivity value (Sr) reaches 1.44% K−1 at 293 K, indicating that the LZSO:Cr3+,Ni2+ phosphor is a potential candidate for NIR optical thermometers in biotechnological applications. The manufactured NIR-II pc LED provides a radiative flux of 8.26 mW with a photoelectric conversion efficiency of 2.71% at a drive current of 100 mA and has promising potential in night vision, medical imaging, and spectral analysis applications.

Magnesium phosphate bone cement (MPC) has gained widespread usage in orthopedic implantation due to its fast-setting and high initial strength benefits. However, the simultaneous attainment of drug-controlled release and osteogenic potential in MPC remains a significant challenge. Herein, a strategy to create a smart injectable cement system using nanocontainers and chondroitin sulfate is proposed. It employs nanocontainers containing alendronate-loaded mesoporous silica nanoparticles, which are surface-modified with polypyrrole to control drug release in response to near-infrared (NIR) stimulation. The alendronate-incorporated cement (ACMPC) exhibits improved compressive strength (70.6 ± 5.9 MPa), prolonged setting time (913 s), and exceptional injectability (96.5% of injection rate and 242 s of injection time). It also shows the capability to prevent degradation, thus preserving mechanical properties. Under NIR irradiation, the cement shows good antibacterial properties due to the combined impact of hyperthermia, reactive oxygen species, and alendronate. Furthermore, the ACMPC (NIR) group displays good biocompatibility and osteogenesis capabilities, which also lead to an increase in alkaline phosphatase activity, extracellular matrix mineralization, and the upregulation of osteogenic genes. This research has significant implications for developing multifunctional biomaterials and clinical applications.

Deep tissue bacterial infections, especially methicillin-resistant Staphylococcus aureus (MRSA) infections, pose challenges to clinical therapy due to their low debridement efficiency and relapsing. Molybdenum disulfide (MoS 2 ) is used in the antibacterial field as a classic photothermal agent (NIR-I) with good biocompatibility. However, due to its limited NIR-I tissue penetration ability and single treatment mode, MoS 2 has poor therapeutic effects on deep tissue infection. Herein, we prepared a defect-type hybrid 2H-MoS 2 nanozyme (MoWS 2 ) using hydrothermal method fabricate the MoWS 2 composite, which is a new antibacterial strategy involving photothermal and enzyme catalysis, and further enhances the activity of the nanozyme through overheating. The regulation of 2H-MoS 2 defects through tungsten ion doping endows MoWS 2 with better near-infrared two-region absorption (NIR-II) and enzyme catalytic performance. Antibacterial activity experiments in vitro have shown that MoWS 2 can achieve efficient bactericidal activity and biofilm clearance through hyperthermia and reactive oxygen species (ROS). Deep MRSA infection experiments have shown that MoWS 2 rapidly removes bacteria from subcutaneous infected tissues through photothermal therapy (PTT) and chemodynamic therapy (CDT), accelerates the dissipation of abscesses, and promotes the healing of infected wounds. Additionally, the versatile treatment mode of MoWS 2 was further confirmed through tissue sectioning and immunofluorescence staining analysis. Overall, these results provide a feasible approach for achieving efficient treatment of deep tissue infections through tungsten ion doping to regulate defective 2H-MoS 2 . Statement of significance The photothermal effect of MoS 2 nanosheets in the NIR-I (650-900 nm) window in anti-MRSA therapy is considered to be highly reliable and efficient in PTA. However, most of the developed PPT therapies or antimicrobial systems based on PTT therapies developed with 1T-MoS 2 have in vivo sterilization temperatures of more than 55°C, which have the risk of damaging the normal tissues of the skin. In this study, we prepared W@MoS 2 with a good photothermal effect (36.9%) in the NIR-II window and good peroxidase-like activity. The combined effect of PTT and CDT has a stronger bactericidal effect while avoiding high-temperature damage, which makes the W@MoS 2 material more advantageous in terms of antimicrobial effect.

The severe urban heat island effect emphasizes the heightened importance of reducing building energy consumption. Cool pigments with higher near-infrared reflectivity contribute to the overall coating performance of the building. In this work, a novel intense blue pigment with superior infrared reflectance based on Ni-doped Zn2SiO4 was successfully synthesized via the simple solid-state method. The crystal structure, oxidation states, and optical properties are analyzed. The oxidation state of Ni was confirmed to be +2 using magnetic analysis and XPS. The intense blue color exhibited by the solid solution is due to the 3T1(3F) → 3T1(3P) transition of Ni2+ at the tetrahedral sites. UV–vis–NIR spectra demonstrate that this pigment exhibits superior near-infrared reflectance compared to cobalt blue in the range of 900–1700 nm, indicating its potential application in the realm of building energy conservation.

Cisplatin resistance is a major challenge in laryngeal cancer treatment. Tumor necrosis factor-alpha-induced protein 2 (TNFAIP2) has been implicated in chemoresistance, though its role in laryngeal cancer remains unclear. We identified TNFAIP2 as a key gene associated with cisplatin resistance and developed a second near-infrared (NIR-II) light-responsive CRISPR-Cas9 nanosystem (APC@RBCs) to target it. In cisplatin-resistant Tu177/CDDP cells, light-activated knockout of TNFAIP2 significantly increased cisplatin sensitivity (IC50 reduced from 12.55 to 4.37 µg/mL). Mechanistic studies revealed that TNFAIP2 modulates cisplatin resistance via the NRF2 pathway, affecting oxidative stress response and epithelial-mesenchymal transition. TNFAIP2 deletion also suppressed cancer cell migration and invasion. Our results establish TNFAIP2 as a critical mediator of chemoresistance and demonstrate the potential of APC@RBCs as a precise, non-invasive gene therapy platform. This strategy offers a promising approach to overcoming cisplatin resistance and improving treatment outcomes in laryngeal cancer patients.

Methylglyoxal is considered a key indicator in evaluating wine flavor and quality, as well as an important marker for diabetic pathological syndromes. Rapid and accurate quantitative detection of methylglyoxal is essential in scenarios of wine production standards and human health monitoring. Herein, we report a visual method for detecting methylglyoxal via an NIR-excitable reversible ratiometric fluorescent hydrogel sensor, where NIR-excited upconversion nanoparticles serve as energy donors and eosin B acts as the energy acceptor, together forming an integrated ratiometric nanophotonic probe that ensures the accuracy of detection without being affected by various background fluorescence interference in different scenarios. The integrated optical probe is combined with a 3D network hydrogel to design a sensing patch that can be easily regenerated through simple treatment, exhibiting a distinct optical color response. Upon the addition of methylglyoxal, the G/R value of the sensing patch changes, enabling the real-time quantitative detection of methylglyoxal. Additionally, we combined the hydrogel sensing patch with a smartphone to create a portable sensing platform for the convenient visual detection of methylglyoxal. The probe and hydrogel sensing patch have detection limits for methylglyoxal as low as 59 and 75.4 nM, respectively. The portable sensing patch designed here provides an effective strategy for standardizing the wine production process and monitoring patient health.

As the gold-standard neuroprotective intervention against cerebral ischemia-reperfusion injury, therapeutic hypothermia (TH) exhibits clinical efficacy while the current TH strategies are limited in application due to their non-specific cooling effect causing severe side effects. Developing targeted TH strategies still remains a problem. In this work, a near infrared emissive, ischemic targeted nanocomposite is demonstrated to achieve localized TH at the ischemic-reperfusion site. Rare earth doped nanoparticle as the core of the nanocomposite generates near infrared II luminescence for the guidance of delivery process. 2H,3H-decafluoropentane and ammonium nitrate loaded in the mesoporous silica layer of the nanocomposite serve as physical and chemical cooling agents for temperature reduction in the brain. Surface ligands modified with stroke-homing peptide and reactive oxygen species cleavable ketone moieties of the nanocomposite can target the inflammatory ischemic region and the promote cooling effects. By the intranasal administration of the nanocomposite in ischemic stroke mice model, infract area and pro-inflammatory factors levels of the brain are effectively reduced and the neurological functions are preserved. This work points to a concept of localized hypothermia method via nanomaterials, which may inspire the development of TH therapy for stroke in the future.

Persistent Luminescence (PersL) materials, which use traps to store energy and emit photons over a long period, have found important applications in the fields of optical information storage, security labeling, and biological imaging. The trap depth is a crucial factor determining the performance of these materials; however, achieving the desired trap depth with high precision remains a great challenge. Here, we provide double perovskite phosphors (Cs 2 SnCl 6 -Cs 2 ZrCl 6 -Cs 2 HfCl 6 series) with highly compatible crystal structures, enabling continuous and precise tuning of trap depth over an ultra-wide range of 0.11 to 1.25 eV. By incorporating W 4+ as the luminescent centers, these phosphors exhibit outstanding near-infrared (NIR) PersL performance at approximately 900 nm and a lasting emission duration exceeding 10 h. The underlying mechanism of PersL is elucidated, and the wide-range tunability of trap depth is attributed to the universal applicability of band-gap engineering in the entire material system. Furthermore, we demonstrate the practical application of these materials by designing a flexible detector plate for X-ray imaging. The detector plate exhibits a storage time of more than 1 week, a detection limit of 0.83 μGy air ·s -1 in the near-infrared region, and real-time and delay-time imaging resolutions of 14.2 lp·mm -1 and 2.5 lp·mm -1 , respectively. These attributes demonstrate strong potential for X-ray luminescence extension imaging.

Surgical precision in tumor resection critically relies on real-time intraoperative imaging, yet conventional probes face limitations in specificity and spatiotemporal control. Here, we present a tumor microenvironment (TME)-activated near-infrared (NIR) fluorescent nanoprobe (DNS–DYE/PEG–NI) that integrates dual responsiveness to hypoxia and glutathione (GSH) for submillimeter-level surgical navigation. The system comprises a GSH-activatable NIR fluorophore (λex/em = 679/730 nm) quenched by 2,4-dinitrobenzenesulfonyl (DNS) moieties and hypoxia-sensitive amphiphilic PEG–NI micelles. Upon tumor accumulation via the enhanced permeability and retention (EPR) effect, a hypoxic TME triggers micelle disassembly through nitroimidazole (NI) reduction, releasing DNS–DYE. Subsequent GSH cleavage restores fluorescence via intramolecular charge transfer (ICT) recovery, achieving a 12.3-fold tumor-to-normal tissue signal ratio and >90% reduction in off-target activation compared to non-responsive controls. Systematic validation demonstrates: (1) dose-dependent fluorescence recovery (35-fold intensity increase at 10 mM GSH); (2) hypoxia-driven micelle destabilization (800% hydrodynamic diameter expansion); (3) sustained colloidal stability (12.9% size variation over 15 days); and (4) low cytotoxicity (cell viability >90% at 125 μg mL−1). In vivo studies reveal precise tumor delineation within 12 h post-injection, enabling real-time resection of submillimeter lesions. By coupling TME-specific activation with prolonged tumor retention, this dual-responsive nanoprobe advances fluorescence-guided surgery toward precision oncology, reducing positive margin rates from 70% to <5% in preclinical models.
