Sensitive and accurate methods for early detection of acute ischemic stroke (AIS) are essential for timely treatment and prognostic assessment of patients. In this study, we report a microfluidics-based ultrasensitive surface-enhanced Raman scattering (SERS) immunoassay device for the quantitative determination of multiplex biomarkers in AIS. By preparing 5,5′-dithiobis-2-nitrobenzoic acid (DTNB) antibody-modified gold nanoparticles (AuNPs) on SERS devices as SERS probes, the biomarkers in whole blood of AIS were accurately captured and further visualized for SERS signal intensity quantitative analysis of six biomarkers in the blood samples. It is worth mentioning that the limit of detection (LOD) of the method can reach the level of fg/mL, with excellent sensitivity and selectivity. Meanwhile, the analytical comparison with ELISA method showed that the detection results of both methods were consistent, which verified the feasibility of the assembled device. The SERS immunoassay device detection provides a powerful strategy for the prediction, early diagnosis and dynamic monitoring of prognosis of AIS with a wide range of clinical practice prospects.

A signal amplification strategy is always required to improve sensing performance, especially for mass-prepared, miniature and highly sensitive testing equipment. Herein, an on-chip integrated biosensor combining microcavity lasing and surface enhanced Raman scattering (SERS) for label-free biochemical analysis is developed. The gallium nitride (GaN) microrings array is fabricated as a whispering gallery mode microlaser based on totally internal wall reflection, and gold nanoparticles (AuNPs) are further assembled on the cavity surface to confine the optical field synergistically. The improvement of lasing capability indicates the strong light-matter interaction that is conducive to superior response of biomolecules. As a functional example, a GaN/AuNPs substrate is employed in a urine assay, which avoids the shortcomings of requiring specific reactive reagents on different display modules. Ultimately, the chip not only realizes a wide range of pH (3.6–7.8) identification depending on the ultra-sensitivity feedback of lasing to the refractive index of liquids, but also significantly enhances SERS signals to enable real-time determination of the biomolecules in human urine. Even the lowest levels of creatinine (0.02%) can be quantitatively detected in less than a minute; thus this work with dual-mode spectral analyses realizes rapid diagnosis for urinary systems, as well as provides a more accurate reference for the screening of other diseases.

As a reactive oxygen species (ROS), excessive production of H 2 O 2 contributes to the development of several diseases such as, inflammation, cancer, and respiratory diseases. Supplementation with endogenous or exogenous antioxidants can scavenge ROS and reduce the oxidation of cellular molecules, thus alleviating the generation of diseases. Therefore, the determination of H 2 O 2 content and its antioxidant activity is of great importance in disease diagnosis and treatment. In this paper, a ratiometric SERS sensor with a flexible cellulose membrane was designed for quantitative detection of H 2 O 2 and assessment of antioxidant activity. First, gold seeds were reduced on bacterial cellulose membrane (BCM) and Au NPs were smoothly deposited on the bacterial cellulose membrane (BCM) using halides to reduce the reduction potential in the growth solution to form a flexible BCM@Au NPs SERS substrate. Afterwards, the oxidation of H 2 O 2 was used to convert 3-mercaptophenylboronic acid (3-MPBA) to the corresponding phenol form 3-hydroxyphenylethanol (3-HTP). The change of substance resulted in a good linear relationship between the intensity ratio corresponding to the two Raman shifts of 881 cm −1 and 995 cm −1 and the H 2 O 2 concentration with a detection limit of 0.0186 μM. This opens up a new method for the detection of H 2 O 2 with high sensitivity and accuracy. In addition, this SERS platform was successfully used for the determination of antioxidant activity. It is promising to be applied to disease diagnosis and efficacy evaluation.

As one of the most widely distributed microRNAs, microRNA-21 (miRNA-21) significantly regulates target genes’ expression levels and participates in many cellular and intercellular activities, and its abnormal expression is always related to some diseases, especially cancer. Hence, detecting miRNA-21, as a biomarker, at the single-cell level helps us to reveal cell heterogeneity and expression level variation during the state change of cells. In this study, we constructed a gold nanoparticles nanomembrane (AuNPs-NM)-modified plasmonic glass nanopipette (P-nanopipette) surface-enhanced Raman scattering (SERS) sensing platform to sensitively detect content variation of the intracellular miRNA-21 during the electrostimulus (ES)-induced apoptosis process. The cytoplasm-located miRNA-21 was first extracted by using the extraction DNA (HP1)-modified P-nanopipette through a hybridization chain reaction (HCR). The nanopipette was then incubated with a labeling DNA (HP2) and reporter 4-MBA-modified Raman tag. The Raman signal (collected from the tip area near the orifice within 1 μm) showed a good response to the content variation of intracellular miRNA-21 under ES, and the proposed single-cell SERS detection platform provides a simple way to study intracellular substance change and evaluate cancer treatment outcomes.

This study focuses on the development of a highly sensitive surface-enhanced Raman scattering (SERS) sensor for detecting homocysteine (Hcy) molecules. The Hcy sensor was created by depositing silver nanoparticles (AgNPs) onto the surface of graphene oxide (GO) film to form a dense AgNPs@GO composite film. The AgNPs on the composite film interacted with sulfur atoms (S) of Hcy molecules to form Ag–S bonds, which boosted the chemisorption of Hcy molecules and enabled them to be specifically recognized. The SERS sensor exhibited a maximum enhancement factor of up to 1.1 × 104, with a reliable linear response range from 1 to 60 ng mL−1. The limit of detection (LOD) for Hcy molecules was as low as 1.1 × 10−9 M. Moreover, Hcy molecules were successfully distinguished in a mixed solution of γ-aminobutyric acid and Hcy molecules. In this study, a simple preparation process of SERS substrate and a novel detection method for Hcy molecules provided a new pathway for the rapid and effective detection of Hcy molecules in the food and biomedicine fields.

Volatile organic compounds (VOCs) are common environmental pollutants and important biomarkers for early diagnosis of lung cancer. However, aldehydes are difficult to detect directly due to their small Raman scattering cross-section and gaseous phase. Here, a Ag-coated ternary layered double hydroxide (LDH) was designed for the detection and identification of various aldehydes. The specific surface area of CoNi-LDH was increased by doping Fe3+, which provides abundant active sites to capture gas molecules. Furthermore, the energy band gap (Eg) was decreased due to the local amorphous FeCoNi-LDH with an extended band tail, promoting the excitonic transition of Fe0.07(CoNi)0.93-LDH. In addition, the Fermi level of Ag prevented the recombination of electron–hole pairs of Fe0.07(CoNi)0.93-LDH, providing a new bridge for charge transfer between the substrate and the molecule. Ag/Fe0.07(CoNi)0.93-LDH presented excellent surface-enhanced Raman scattering (SERS) performance for aldehyde VOCs by modification with 4-aminothiophenol (4-ATP) to capture aldehydes and realized the detection of benzaldehyde (BZA) at 10 ppb. The enhancement and Raman shift of the b2 mode indicated the contribution of chemical enhancement to the SERS system, so the substrate presented good uniformity. The recycling of the SERS substrate is realized based on the reversibility of the Schiff base reaction. These results manifested that Ag/FeCoNi-LDH has a wide prospect in the application in the trace detection of aldehydes.

Rapid extraction and analysis of target molecules from irregular surfaces are in high demand in the field of on-site analysis. Herein, a flexible platform used for surface-enhanced Raman scattering (SERS) based on an ordered polymer pyramid structure with half-imbedded silver nanoparticles (AgNPs) was prepared to address this issue. The fabrication includes the following steps: (1) creating inverted pyramid arrays in silicon substrate, (2) preparing a layer of AgNPs on the surface of the inverted pyramids, and (3) obtaining a substrate with an ordered polymer pyramids array with half-imbedded AgNPs by the molding method. This flexible substrate is capable of rapid extraction via a simple and convenient “paste and peel off” method. In addition, the substrate exhibits great repeatability and good sensitivity thanks to the uniformity and larger surface area of the ordered pyramids. The density of “hot spots” (local electromagnetic field with high intensity) is increased on the structured surface. Semi-imbedding silver particles in the polymer pyramids makes “hot spots” robust on the substrate. In addition, the preprepared silicon template with the inverted pyramids can be reused, which greatly reduces the production cost. With this substrate, we successfully analyzed thiram molecules on the epidermis of apples, cucumbers, and oranges, and the detection limits are 2.4, 3, and 3 ng/cm2, respectively. These results demonstrate the great potential of the substrate for in situ analysis, which can provide reference for the design of ideal SERS substrates.

The detection of biomolecules utilizing surface-enhanced Raman spectroscopy (SERS) is highly trending in research, but the unlabeled and sensitive detection of saccharides in body fluids is challenging. Here, a novel SERS substrate (AgNS 600 ) is fabricated by physical scratching of smooth silver sheets, which successfully achieves enhanced SERS fingerprinting by capturing glucose molecules through nanoscale grooves on the surface of the substrate to form multi-dimensional “hot spot” regions. The platform proves to be rapid, reliable, and reproducible, with a lower detection limit of 0.5 amol L −1 in deionized water. Further detection of multiple monosaccharides, polysaccharides, and mix samples, which are distinguished with machine learning and heat map, demonstrates the versatility and specificity of the platform. Importantly, real samples are explored and the results show that the platform is able to detect and recognize SERS signals of glucose in the blood, urine, tears, and perspiration, and non-destructive predict diabetes by analyzing the signal intensity of different characteristic peaks. In this work, it is the first non-chemical, un labeled determination of glucose with high sensitivity via SERS, which provides a potential strategy for advancing the clinical development of SERS technology and the early diagnosis of glucose metabolic diseases.

The novel histone deacetylase drug chidamide (CHI) has been proven to regulate gene expression associated with oncogenesis via epigenetic mechanisms . However, huge side effects such as non-targeting, poor intracellular accumulation and low nuclear entry efficiency severely restrict its therapeutic efficacy. Dual-targeted nanodrug delivery systems have been proposed as the solution. Herein, we developed a CHI-loaded drug delivery nanosystem based on Prussian blue (PB) nanocarrier , which combines surface-enhanced Raman scattering (SERS) tracking function with cancer cell/nuclear-targeted chemotherapy capability. With the property of background-free SERS mapping, PB nanocarriers can serve as tracking agents to localize intracellular CHI. The incorporation of targeted molecules specifically enhances the cancer cell/nuclear internalization and chemotherapeutic effects of CHI-loaded PB nanocarriers. In vitro cytotoxicity assay clearly shows that the constructed CHI-loaded PB nanocarriers have significant inhibitory on Jurkat cell proliferation . Furthermore, SERS spectral analysis of Jurkat cells incubated with the CHI-loaded PB nanocarriers reveals obvious features of cellular apoptosis: DNA skeleton fragmentation, chromatin depolymerization , histone acetylation, and nucleosome conformation change. Importantly, this CHI-loaded PB nanocarrier will provide a new insight for lymphoblastic leukemia targeted chemotherapy.

Shewanella plays an important role in geochemical cycle, biological corrosion, bioremediation and bioenergy. The development of methods for identifying Shewanella can provide technical support for its rapid screening, in-depth research into its extracellular respiratory mechanism and its application in ecological environment remediation. As a tool for microbial classification, identification and detection, Surface-enhanced Raman scattering (SERS) has high feasibility and application potential. In this work, bio-synthesized silver nanoparticles (AgNPs) were used as SERS substrates to effectively distinguish different types of Shewanella bacteria based on the difference in bacterial extracellular electron transfer (EET) ability. AgNPs were combined with the analyzed bacteria to prepare “Bacteria-AgNPs” SERS samples, which can strongly enhance the Raman signal of the target bacteria and reliably obtain spatial information of different molecular functional groups of each bacteria. Our developed approach can effectively distinguish between non-metal reducing and metal-reducing bacteria, and can further distinguish the three subspecies of Shewanella ( Shewanella oneidensis MR-1, Shewanella decolorationis S12, and Shewanella putrefaciens SP200) at the genus and species level. The Raman signal enhancement is presumably caused by the excitation of local surface plasma (LSP) and the enhancement of surrounding electric field. Therefore, our developed method can achieve interspecific and intraspecies discrimination of bacteria. The proposed method can be extended to distinguish other metal-reducing bacteria, and the novel SERS active substrates can be developed for practical applications.

Tobramycin aptamer was adsorbed on the surface of gold nanoparticles (Au NPs) by van der Waals forces , hydrogen bonds , and other factors, limiting nanozyme peroxidase activity. Because tobramycin has a higher affinity for the tobramycin aptamer, including tobramycin in Au@Apts nanozymes was able to dislodge the aptamer from the surface of Au NPs and restore the catalytic activity of nanozymes. Raman spectroscopy of the enzyme-catalyzed product TMB ox can detect this shift in enzyme activity . The restored value of nanozyme activity increased with tobramycin concentration and had a good linear correlation in the concentration range of 10 −10 ∼10 −1 M with a detection limit of 2.04 × 10 −11 M. Aptamer biosensors are built using the SERS platform, which has the advantages of high sensitivity, high precision, and a wide detection range. It is prospectively employed in food safety and the detection of antibiotic residues.

Perfluoro- and polyfluoroalkyl substances (PFAS) are widely used throughout the world and are extensively found in the environment. The detection of PFAS has become essential to ensuring environmental sustainability and human health. Herein, AuNRs were synthesized by the seed growth method and then the samples were coated with silver shells to form Au@Ag core–shell nanorods (Au@AgNRs). Au@AgNRs monolayer was prepared by oil–water self-assembly on a silicon wafer and then was constructed as a plasmonic AgNPs/Au@AgNRs sandwich structure for surface-enhanced Raman scattering (SERS) detection of PFAS. The shell thickness of Au@AgNRs was optimized and displayed a 11.9-fold SERS intensity higher than the pristine AuNRs. Moreover, the hotspots generated by the electromagnetic field coupling between the interlayer gap achieved 3.6-fold SERS signal enhancement compared to that of the Au@AgNRs monolayer. Besides, the excellent structural uniformity (RSD ∼ 8.0%) makes it ideal for quantitative SERS detection applications. More importantly, the use of the AgNPs/Au@AgNRs sandwich structure sensor enables the highly sensitive detection of model molecules (10–11 M) and quantitative detection of a wide range of fluorinated alkyl substances of perfluorooctanoic acid (PFOA), perfluorohexanoic acid (PFHxA), and potassium perfluorobutanesulfonate (PPFBS), even with a portable Raman device. And the detection concentration can be down to 0.1 ppm for PFAS. This sensitive and quantitative SERS technique has promising prospects in the monitoring of fluorinated alkyl substance contaminations.
