5-(Hydroxymethyl)-2′-deoxycytidine (5-hmdC), 5-(formyl)-2′-deoxycytidine (5-fodC), and 5-(carboxyl)-2′-deoxycytidine (5-cadC) are crucial intermediate products of the DNA demethylation pathway, which can also act as potential biomarkers reflecting the diagnosis and prognosis in multiple tumors. Detecting 5-hmdC, 5-fodC, and 5-cadC in human urine has various advantages including readily available samples and being noninvasive to patients. However, few works have reported the detection of 5-fodC and 5-cadC due to their trace amounts. Here we developed a novel magnetic hyper-cross-linked microporous polymer (HMP) material based on polyionic liquid (PIL) for the enrichment of 5-hmdC, 5-fodC, and 5-cadC. These magnetic PIL-HMP materials provided specific enrichment superiority for three modified cytidines. After enrichment, the signal intensity of 5-hmdC, 5-fodC, and 5-cadC increased 10–75-fold with lower limits of quantitation (LLOQ) of 0.049, 0.781, and 0.781 ng/mL, respectively. The recoveries were approximately 86.5–95.2% for 5-hmdC, 95.2–107.8% for 5-fodC, and 99.4–102.4% for 5-cadC under the relative standard deviation (RSD) of 0.2–10.3%. Finally, we successfully applied magnetic PIL-HMP materials coupled with high-performance liquid chromatography–electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS) in enrichment and quantitative determination of 5-hmdC, 5-fodC, and 5-cadC in human urine of 10 breast cancer patients and 10 healthy people. We found that the level of 5-hmdC decreased in breast cancer patients (p < 0.05), while the levels of 5-fodC and 5-cadC increased (p < 0.05, p < 0.01). Our results demonstrated that the levels of metabolic 5-hmdC, 5-fodC, and 5-cadC in human urine are closely related to breast cancer, which could contribute to the clinical diagnosis and investigation of breast cancer and its occurrence and development mechanisms.

A colorimetric assay is presented for the determination of ascorbic acid (AA). Manganese(II) doped carbon dots (Mn-CDs) were prepared by a convenient hydrothermal route and are shown to possess oxidase-like catalytic ability. They catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) by dissolved oxygen to form a blue colored product (oxTMB). AA can reduce blue oxTMB to colorless TMB. The fading of the blue color (measured at 652 nm) can be applied for quantifying AA in the 50 to 2500 nM concentration range and with a 9 nM detection limit. The method was successfully used for the quantitation of AA in real samples.
Schematic representation of a colorimetric assay platform for the sensitive detection of ascorbic acid (AA) in view of inhibitory effect of AA on the 3,3′,5,5′-tetramethylbenzidine (TMB) oxidation, benefitting from excellent oxidase-like catalytic activity of manganese(II) doped carbon dots (Mn-CDs).

The oxidation of 5-hydroxymethylfurfural (HMF) to 2,5-furandicarboxylic acid (FDCA) is one of the most attractive reactions to establish a sustainable chemical process based on biomass resources. In this work, a CNT-supported ruthenium–manganese oxide nano-catalyst (RuO2/MnO2/CNT) was employed for the electro-catalytic oxidation of HMF in base-free aqueous solution. The activity test showed that α-MnO2 can effectively promote the activity of RuO2 in the oxidation of HMF. In comparison with RuO2/CNT, RuO2/MnO2/CNT possessed a lower activation energy and more than twice the FDCA formation rate. Under the optimized reaction conditions, the RuO2/MnO2/CNT catalyst afforded a FDCA yield of 72.1% in a 0.1 M K2SO4 aqueous solution at a 0.9 V (vs. Ag/AgCl) applied potential. To our knowledge, this is the first demonstration of FDCA formation as the primary product with high yield in an initially neutral electrolyte. The product FDCA can be easily separated after cooling the reaction solution to room temperature.

Six series of 4-(benzo[ c ][1,2,5]thiadiazol-5-yl)-3(5)-(6-methylpyridin-2-yl)- pyrazoles 18a – d , 19a – d , 22a – d and 3(5)-(6-methylpyridin-2-yl)-4-(thieno[3,2,- c ]- pyridin-2-yl)pyrazoles 20a – d , 21a – d , 23c , 23d have been synthesized and evaluated for their activin receptor-like kinase 5 (ALK5) and p38α mitogen activated protein (MAP) kinase inhibitory activities in enzymatic assays. Among these compounds, the most active compound, 22c , inhibited ALK5 phosphorylation with an IC 50 value of 0.030 μM in the enzymatic assay. Compound 22c showed four-fold more potent activity against ALK5 kinase than the clinical candidate, compound LY-2157299 . The selectivity index of 22c against p38α MAP kinase is 235, which is much higher than that of LY-2157299 (4) and equally selective to that of EW-7197 (218). Compound 22c effectively suppressed protein and mRNA expression of collagen I and α-SMA in TGF-β-induced LX-2 human hepatic stellate cell (HSC), this result shows that compound 22c has the ability to inhibit the activation of HSC. Compound 22c is expected to be a preclinical candidate for the treatment of hepatic fibrosis .

A series of new two-dimensional copolymers, PBDTT-TABT, PBDTT-TANT and PBDTT-TSNT, with conjugated benzodiathiazole (BT) or naphthobisthiadiazole (NT) side chains were successfully synthesized and characterized for high performance polymer solar cells (PSCs). The NT unit showed a stronger electron-withdrawing ability and a larger conjugation than the BT unit, which induced a stronger ICT process between the benzodithiophene (BDT)-thiophene (T) backbone and the conjugated side chain for NT containing copolymers. As a result, PBDTT-TANT and PBDTT-TSNT showed lower-lying bandgaps and more red-shifted absorption than PBDTT-TABT. The alkylthio modification effect of conjugated side chains was also investigated in this work. This effect showed a positive role in lowering the HOMO energy level, and a negative role in elevating carrier mobility and molecular stacking properties in our two-dimensional polymeric systems. Bulk heterojunction (BHJ) PSCs were fabricated using these copolymers as the donor materials to evaluate their photovoltaic properties. The PBDTT-TABT, PBDTT-TANT and PBDTT-TSNT devices exhibited PCEs of 4.60%, 5.65% and 4.01%, respectively. In spite of the Voc values, the highest Jsc, FF and PCE were achieved for the PBDTT-TANT device, which was attributed to its red-shifted absorption, improved carrier mobility and well-defined phase separation. It is interesting that the Jscs, FFs and PCEs of all these devices were elevated significantly when made using the solvent vapor annealing (SVA) method. The THF–SVA process would provide a driving force to facilitate the formation of a much more well-defined surface morphology, resulting in the enhanced Jsc and FF values. The PBDTT-TANT device showed the highest PCE of 8.04%, which is the top efficiency for this type of two-dimensional copolymer with donor (D)–donor (D) polymer backbones and donor (D)–acceptor (A) conjugated side chains. Our design strategy would give an instructive guide to developing high performance two-dimensional polymer donors to be used in organic photovoltaic applications.

JZ-1201, targeting SERT, NET, and 5-HT 1A receptors, attenuates depressive-like behaviors. • The antidepressant effect of JZ-1201 is partially mediated by activation of the 5-HT 1A receptor. • JZ-1201 enhances PACAP-related neuroplasticity in the hippocampus. Depression has become a significant public health issue, highlighting the need for novel antidepressants. Based on the monoaminergic mechanisms of classical antidepressant drugs and the therapeutic potential of 5-HT 1A receptor activation, we designed a novel compound, JZ-1201, which acts as a selective serotonin (5-HT) and norepinephrine (NE) reuptake inhibitor and 5-HT 1A receptor partial agonist. This study aimed to evaluate its antidepressant-like effects and explore the underlying mechanisms involving 5-HT 1A receptor activation and enhanced neuroplasticity. The results demonstrated that JZ-1201 exhibited high affinity for the serotonin transporter (SERT) and norepinephrine transporter (NET), and 5-HT 1A receptors, inhibited 5-HT and NE reuptake. It enhanced the 5-hydroxytryptophan (5-HTP)-induced head-twitch response (HTR) and inhibited forskolin-stimulated cAMP formation. In male ICR mice, acute administration of JZ-1201 reduced immobility time in the tail suspension test (TST) and forced swim test (FST), an effect that was attenuated by the selective 5-HT 1A receptor antagonist WAY-100635. Chronic administration of JZ-1201 significantly ameliorated depressive-like behaviors in male Wistar rats subjected to chronic unpredictable mild stress (CUMS). In CUMS rats, JZ-1201 increased dendritic spine density and the expression of PACAP/mTOR/BDNF signaling pathway proteins in the hippocampus. These findings indicate that JZ-1201 is a triple-target antidepressant compound and modulates hippocampal PACAP-related neuroplasticity. Thus, JZ-1201 could be developed as a novel therapeutic agent for the treatment of depression.

The highly-efficient conversion of 5-hydroxymethylfurfural (HMF) to 2,5-bis(propoxymethyl)furan was achieved via the one-pot route, which were performed by the combination of supported copper catalyst and hierarchical pore ZSM-5 zeolite. Among the supported copper catalyst, the Cu/Al 2 O 3 catalyst exhibited the excellent activity and selectivity in the hydrogenation of HMF, obtaining a 98.7% yield of BHMF at 100 °C for 6 h under 2.5 MPa of H 2 . The reason might be attributed to that the highly dispersed copper particles and relatively weaker acid sites, which improved the hydrogenation activity of catalyst and suppressed the occurrence of side reaction. In the one-pot synthesis of BPMF, the Cu/Al 2 O 3 catalyst and HP-ZSM-5 zeolite were responsible for HMF hydrogenation and BHMF etherification , respectively, and the competition between them is negligible. This work provides a simple route to produce furanic diether from HMF.

The Cd x In y S ( x +1.5 y ) catalysts with gradient Cd:In ratios were controllably synthesized via an one-step hydrothermal approach. Particularly, the hydrothermal synthesis conferred those Cd x In y S ( x +1.5 y ) catalysts with Cd:In ratios over 1:2 with a one-dimensional–three-dimensional heterojunction structure composed of CdS nanorods and cubic-phase CdIn 2 S 4 . Our characterization evidences reflected that the CdS nanorods effectively promoted the separation of photoexcited electron–hole pairs in Cd x In y S ( x +1.5 y ) and enabled transboundary transfer of photogenerated carriers. Spectroscopic and experimental results were further employed to develop a detailed catalytic mechanism, in which the superoxide anion (•O 2 − ) plays an important role in the catalytic oxidation of HMF to DFF by Cd x In y S ( x +1.5 y ) . Unlike other photocatalysts with •O 2 − as the main active species, Cd 1.5 In 2 S 4.5 gives an excellent performance in HMF conversion. This shows the great potential of cadmium–indium sulfides as photocatalysts for biomass conversion under ambient conditions.

Photoreforming cellulose to simultaneously produce 5-hydroxymethylfurfural (5-HMF) and hydrogen represents a promising strategy for harnessing solar energy. However, the development of highly efficient photocatalysts for this reaction remains a challenge, and the influence of cellulose’s crystalline structure on conversion efficiency is not well understood. In this study, a Pt-modified g-C 3 N 4 –Bi 3 TiNbO 9 heterojunction photocatalyst was synthesized to explore the photocatalytic reforming of four crystalline forms of cellulose. The results demonstrate that the crystalline structure of cellulose significantly affects hydrogen and 5-HMF production, with the order of activity being cellulose III > IV > II > I. Among the cellulose types tested, cellulose III exhibited the highest performance, achieving 51.8 μmol g⁻ 1 h⁻ 1 of hydrogen and 113.4 μmol g⁻ 1 h⁻ 1 of 5-HMF, surpassing the yields from cellulose I, II, and IV when using the Pt 1.0 –g-C 3 N 4 –Bi 3 TiNbO 9 (2:1) heterojunction. Additionally, a lower degree of polymerization of cellulose was found to favor the coproduction of hydrogen and 5-HMF, as it enhances the breakdown of cellulose via reactive species. Factors such as cellulose powder size, solution pH, and catalyst-cellulose interactions were also shown to influence the yields. Density functional theory (DFT) calculations revealed an electron migration of 0.019684e between the O 2 p and Bi 6 p orbitals of Bi 3 TiNbO 9 (220) and the C 2 p and N 2 p orbitals of g-C 3 N 4 , confirming interfacial electron transfer. The calculated adsorption energies followed the trend: cellulose IV (− 1.72 eV) > I (− 1.73 eV) > II (− 1.94 eV) > III (− 2.07 eV), consistent with experimental results, except for cellulose IV. 5-Hydroxymethylfurfural (5-HMF) is an important biomass-based platform molecule with various commercial applications, and its green, efficient production is the key for value-added applications of lignocellulosic biomass. This work studied the preparation of 5-HMF from glucose using autogenic H+ and OH– as the catalysts under hydrothermal conditions without additional catalysts. The effects of reaction temperature, time, pressure, and glucose concentration on the 5-HMF yield were discussed by single-factor experiments. The results show that the highest yield of 5-HMF is 70.2% and the selectivity is 80% at 180 °C for 6 h. The kinetics for glucose and fructose conversions was investigated in detail. It was found that the processes of glucose and fructose conversion to 5-HMF conform to the first-order reaction rate equation, showing the activation energies for 5-HMF formation of 94.5 and 89.6 kJ/mol, respectively. Moreover, 77% of the furfural yield could also be obtained by using xylose as the feedstock. This work offers an alternative, versatile, and eco-friendly strategy for furanic compound production from lignocellulosic biomass and sugar derivatives.

A cyanostilbene-based guest forms a stable hydrogel with Me 10 Q [5] via portal interactions. • Efficient FRET enables emission color tuning from green to red. • Near-white-light emission achieved at optimized S–RhB doping ratio. • Hydrogel shows strong mechanical stability and ordered microstructure. • Strategy simplifies preparation of tunable multicolor luminescent materials. This study aims to develop a multicolor-tunable luminescent supramolecular hydrogel through host-guest assembly between decamethylcucurbit[5]uril (Me 10 Q[5]) and 4,4′-diaminocyanostilbene hydrochloride (CN-DAB). The hydrogel formation was driven by portal-specific interactions, confirmed via Proton Nuclear Magnetic Resonance spectra ( 1 H NMR), Fourier-Transform infrared (FT-IR) spectra, and Ultraviolet-Visible (UV-vis) spectra, with a high binding constant (K a1 = 3.12 × 10 3 L/mol and K a2 = 3.12 × 10 5 L/mol). The resulting hydrogel exhibited aggregation-induced emission at 530 nm. By doping sulforhodamine B (S–RhB) as an acceptor, efficient förster resonance energy transfer (FRET) was achieved, enabling wide-range emission color tuning from green to red. At 0.09 wt% S–RhB, near-white-light emission was obtained with CIE coordinates of (0.35, 0.38) and an energy transfer efficiency of up to 52%. Rheological and Scanning Electron Microscope (SEM) analyses revealed excellent mechanical strength and a porous microstructure. This work provides a simple yet effective strategy for designing multicolor luminescent hydrogels with potential applications in sensing and optoelectronics. 5-Hydroxymethylfurfural (HMF) is a high value-added platform compound, converted into bio-based polyesters and biofuels etc. In this paper, we used two commercially available hexamethylenimine and cetyltrimethylammonium bromide (CTAB) as the structure-directing agents to synthesize HMWW zeolite nanosheets via one step. A series of HMWW zeolite nanosheets with different disorder index ( DI ) were controllably prepared by adjusting the amount of CTAB, operating also as an in situ exfoliating agent, converting fructose into HMF in the tetrahydrofuran/NaCl-H 2 O biphasic solvent with low boiling point. Successful construction of disordered HMWW zeolite nanosheets was verified by PXRD. Further characterizations by BET, SEM and TEM etc. show that HMWW 4.0 with the optimum DI of 0.67 has a disordered network-like arrangement of layers with the external surface area of 224 m 2 /g and the mesoporous size of 3.5 nm. Catalytic evaluations showed that HMF selectivity was closely related to DI of the catalysts. Under the optimized DI , fructose molecules are readily accessible to acidic sites on the external surface while larger external surface area is exposed. The HMWW 4.0 zeolite catalyzed fructose dehydration in the biphasic solvent, producing 90% HMF selectivity with 93% fructose conversion at 140 °C for 6 h. In the reuses of the HMWW 4.0 catalyst, HMF selectivity was around 90% with fructose conversion decreasing by ca. 10% after six runs, indicating that it possesses the promising stability.
