Microplitis bicoloratus parasitism promotes cyclophilin D-p53 interaction to induce apoptosis of hemocytes in Spodoptera litura

Pan Zhang,Zi-Yan Liu,Dan Yu,Cui-Xian Xu,Yan Hu,Ya-Ping Liang,Jie Jin,Ming Li

Microplitis bicoloratus parasitism can induce apoptosis of hemocytes in the M. bicolortus host, Spodoptera litura . However, it is unclear how M. bicolortus parasitism regulates host signaling pathways to induce apoptosis. Expression of cyclophilin D (CypD) and p53 was significantly upregulated in S. litura hemocytes at 6 days postparasitization. In the parasitized hemocytes, there was mitochondrial membrane potential (△Ψ m ) loss, cytochrome c (Cyt C) release from mitochondria, and caspase-3 activation. These occurred while hemocytes were undergoing upregulation of CypD and p53. Parasitism also promoted the interaction between CypD and p53. CypD silencing could rescue the apoptotic phenotypes induced by parasitism, but had no effect on apoptosis in unparasitized S. litura . These findings suggest that the CypD-p53 pathway may be an important component of the parasitism-induced immunosuppressive response and establish a basis for further studies of parasitoid/host interactions. Research highlights Microplitis bicoloratus parasitism upregulates the expression of CypD and p53 and induces host hemocyte apoptosis. M. bicoloratus parasitism promotes the CypD-p53 interaction, resulting in Cyt C release from mitochondria and caspase-3 activation.

Xiaolan Li,Yufei Zhang,Hossain M. Zabed,Junhua Yun,Guoyan Zhang,Mei Zhao,Yuvaraj Ravikumar,Xianghui Qi

d -Arabitol is a top value-added compound with wide applications in the food, pharmaceutical and biochemical industries. Nevertheless, sustainable biosynthesis of d -arabitol is limited by lack of efficient strains and suitable fermentation process. Herein, metabolic engineering and process optimization were performed in Zygosaccharomyces rouxii to overcoming these limitations. Adopting systems metabolic engineering include enhancement of innate biosynthetic pathway, supply of precursor substrate d -ribulose-5P and cofactors regeneration, a novel recombinant strain ZR-5A with good performance was obtained, which boosted d -arabitol production up to 29.01 g/L, 59.31 % higher than the parent strain. Further with the optimum medium composition and fed-batch fermentation, the strain ZR-5A finally produced 149.10 g/L d -arabitol with the productivity of 1.04 g/L/h, which was the highest titer ever reported by Z.rouxii system. This is the first report on the use of metabolic engineering to construct Z. rouxii chassis for the sustainable production of d -arabitol.

D-mannitol-based eutectic composite phase change materials with high thermal conductivity and solar-thermal conversion

Yu Bai,Wenjuan Qiu,Shuangfeng Wang

Summary At present, there are few kinds of phase change materials that suitable for medium temperature heat storage. Among them, sugar alcohols have broad application prospects in the heat storage at medium temperature due to their suitable phase transition temperature. In this work, eutectic phase change material (PCM) was prepared with D-mannitol as PCM and KBr as melting point modifier. Expanded graphite (EG) was used as thermally conductive support network to prepare shape-stable composite phase change material (CPCM). The prepared CPCM—10% EG/PCM has an appropriate phase transition temperature (136.88°C), high phase melting enthalpy (215.7 J/g), and good photothermal conversion (81.9%). The thermal conductivity of 10% EG/PCM reached 6.01 W/(m K), which is approximately 10 times higher than D-mannitol (0.603 W/(m K)). Because of its excellent thermal properties, the applications of CPCM in energy storage and photothermal conversion can be further broadened.

Engineering oxygen vacancies in Co3O4 to boost p-d hybridization for hydrogeneration of nitrogen and oxygen

Xianwei Li,Shuangxiu Ma,Fang Luo,Zehui Yang

Electrochemical hydrogenerations of nitrogen and oxygen molecules are of significance for nitrogen reduction and oxygen reduction reactions. Strengthening the binding between active sites and gaseous molecules is beneficial for enhancing the catalytic performance, especially in NRR catalysis. In this work, we have engineered the oxygen vacancies in Co 3 O 4 nanoparticles to boost the catalytic activity towards NRR and ORR. Ascribing to the p-d hybridization between Co and N atoms, N 2 electrochemical adsorption is promoted and the related energy barrier of N 2 protonation is significantly decreased by 50%; as a consequence, Faradaic efficiency is 11.2% at −0.4 V vs. RHE, 3-fold higher than Co 3 O 4 /CNT; moreover, due to the oxygen vacancies in Co 3 O 4-x /CNT leading to a higher oxophilicity, Co 3 O 4-x /CNT showed a better selectivity in ORR catalysis and kinetic current density at 0.85 V vs. RHE is boosted by 2.4 times. Consequently, Co 3 O 4-x /CNT exhibits a better rechargeable zinc air battery performance than Pt/C-IrO 2 by a factor of 2.2.

Dual-Atomic Catalysts Deduced from d−π Conjugated Metal−Organic Frameworks for Efficient Oxygen Evolution Reaction

Jiameng Liu,Shuangrun Zhao,Changbao Wang,Bin Hu,Linghao He,Minghua Wang,Zhihong Zhang,Miao Du

A series of semiconductive cobalt-based metal–organic framework (MOFs) are prepared in this work using 2,3,6,7,10,11-hexaiminotriphenylene (HATP) as ligand and diverse metal ions as the second coordination centers (M = Mn, Ni, and Zn) (denoted as Co x M 3-x (HITP) 2 MOFs). The series of bimetallic Co x M 3-x (HITP) 2 MOFs exhibit ultrathin graphene-like nanostructure, highly d–π conjugated network, large specific surface areas, and diverse metal coordination modes. Density functional theory calculations reveal the unique synergism between M (M = Mn, Ni, and Zn) and Co nodes, in which adjacent layers of M atoms can efficiently modulate the electron densities of Co atoms in Co 3 (HITP) 2 . As compared, Co x Zn 3-x (HITP) 2 MOF possesses substantially enhanced electron density around the Co atoms, dual-metal atomic sites, and rich metal coordination modes, and exhibits superior electrochemical activity. Among them, Co x Zn 3-x (HITP) 2 displays the outperformed electrocatalytic performances of the oxygen evolution reaction (OER). Particularly, Co x Zn 3-x (HITP) 2 shows the lowest overpotential of 210 mV and good long-term stability. This work develops an efficient strategy for designing and regulating atomically dispersed catalysts in conductive MOFs toward clean energy applications.

Nicotinamide mononucleotide supplementation protects the intestinal function in aging mice and D-galactose induced senescent cells

Meng Ru,Wanwan Wang,Zhenya Zhai,Ruxia Wang,Yumeng Li,Jiang Liang,Damini Kothari,Kaimin Niu,Xin Wu

The nicotinamide adenine dinucleotide (NAD+) level shows a temporal decrease during the aging process, which has been deemed as an aging hallmark. Nicotinamide mononucleotide (NMN), a key NAD+ precursor, shows the potential to retard the age-associated functional decline in organs. In the current study, to explore whether NMN has an impact on the intestine during the aging process, the effects of NMN supplementation on the intestinal morphology, microbiota, and NAD+ content, as well as its anti-inflammatory, anti-oxidative and barrier functions were investigated in aging mice and D-galactose (D-gal) induced senescent IPEC-J2 cells. The results showed that 4 months of NMN administration had little impact on the colonic microbiota and NAD+ content in aging mice, while it significantly increased the jejunal NAD+ content and improved the jejunal structure including increasing the villus length and shortening the crypt. Moreover, NMN supplementation significantly up-regulated the mRNA expression of SIRT3, SIRT6, nuclear factor E2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), the catalytic subunit of glutamate-cysteine ligase (GCLC), superoxide dismutase 2 (SOD2), occludin, and claudin-1, but down-regulated the mRNA expression of tumor necrosis factor alpha (TNF-α). Specifically, in the D-gal induced senescent IPEC-J2 cells, 500 μM NMN restored the increased mRNA expression of interleukin 6 (IL6ST), IL-1A, nuclear factor (NF-κB1), and claudin-1 to normal levels to some extent. Furthermore, NMN treatment significantly affected the mRNA expression of antioxidant enzymes including NQO1, GCLC, SOD 2 and 3, and GSH-PX1, 3 and 4. In addition, 200 μM NMN enhanced the cell viability and total antioxidant capacity and lowered the reactive oxygen species level of senescent IPEC-J2 cells. Notably, NMN restored the down-regulated protein expression of occludin and claudin-1 induced by D-gal. The above data demonstrated the potential of NMN in ameliorating the structural and functional decline in the intestine during aging.

Chitosan and D-fructose 1,6-bisphosphate differ in their effects on soil acidity and aluminum activation

Nkoh Jackson Nkoh,He Xian,Lu Hai-long,Li Ke-wei,Shi Ren-yong,Li Jiu-yu,Xu Ren-kou

Purpose This study aims to evaluate the effects of chitosan and D-fructose 1,6-bisphosphate (F16BP) trisodium salt on the acidity of selected Ultisols (Ultisol-QY1, Ultisol-QY2, Ultisol-LX, and Ultisol-YT) and their resistance to soil acidification. Materials and methods The effect of adsorbed chitosan and F16BP on soil acidity was evaluated by interacting different concentrations of the organic substrates with 4.0 g soil under different agitation times (2 to 264 h, 200 rpm, 25 °C). Also, the soils were amended with the organic substrate and incubated for 14 days to determine their effects on soil physicochemical properties. Afterwards, the soil solid phases from the adsorption studies and incubated soil were acidified with different concentrations of nitric acid to simulate soil acidification and activate soil aluminum (Al). Results and discussion The adsorption interactions of chitosan and F16BP with Ultisols demonstrated different potentials in their ability to displace mineral-bound hydroxyl groups and ameliorate soil acidity. The R-PO 4 3− group of F16BP was more effective in displacing the hydroxyl groups through a ligand exchange mechanism and thus showed greater ameliorating effect on soil acidity than chitosan. Compared to lime and F16BP, chitosan-treated soils exhibited the least decrease in pH during soil acidification due to the significant improvement in soil pH buffering capacity (pHBC). Specifically, when amended with chitosan, the pHBC of Ultisol-QY1, Ultisol-QY2, Ultisol-LX, and Ultisol-YT was increased by 262.2%, 88.9%, 337.7%, and 320.6% as opposed to 22.1%, 18.4%, 27.4%, and 83.0% for these Ultisols by F16BP, respectively. Thus, F16BP is a better material to improve soil pH, while chitosan is a better option to improve soil pHBC, inhibit soil acidification, and retard the activation of phytotoxic Al 3+ . By the nature of their functional groups, we deduced that F16BP ameliorated soil acidity by displacing the hydroxyl groups on soils due to adsorption of R-PO 4 3− group through ligand exchange mechanism, while chitosan directly improved pHBC by providing abundant R-NH 2 groups to accommodate excess exogenous H + . Conclusions Chitosan is a promising soil amendment material for use in the management of acidic soils due to its high N content and potential to inhibit soil acidification. Given the readily available raw materials and ease of producing chitosan, long-term incubation studies should be carried out to assess the effect of chitosan on soil pH variations, carbon, and N mineralization.

Bo Kong,Xiang Yang,Huiqian Dai,Yu Wu,Hong-Bing Lu,Wei Liu,Xiaoying Liu

In this work, an electrochemical sensor was developed based on the delaminated titanium carbide nanosheet (d-Ti 3 C 2 T x ) and fullerene (C 60 ) composite modified glassy carbon electrode (GCE). The composite materials d-Ti 3 C 2 T x /C 60 were characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The prepared modified electrode (d-Ti 3 C 2 T x /C 60 /GCE) exhibited commendable electrocatalytic activity for the oxidation of baicalein. Under the optimum detection conditions, the detection of baicalin showed a wide linear range of 0.015–4.0 μM with a detection limit of 0.005 μM. In addition, the d-Ti 3 C 2 T x /C 60 /GCE showed good reproducibility, stability and anti-interference. It was also successfully used for the determination of baicalin in the commercially available Shuanghuanglian oral liquid with satisfactory results.

Ni–Zn Dual Sites Switch the CO2 Hydrogenation Selectivity via Tuning of the d-Band Center

Shuangxi Lin,Qiang Wang,Maoshuai Li,Ziwen Hao,Yutong Pan,Xiaoyu Han,Xiao Chang,Shouying Huang,Zhenhua Li,Xinbin Ma

Controlling the selectivity of CO2 hydrogenation by catalysis is a fundamental challenge. This study examines the interrelation between active sites and reaction pathways in Ni-catalyzed CO2 hydrogenation. The alloying of Ni with Zn to charged (Niσ––Znσ+) active sites modifies the electronic structure and d-band center, weakens the interaction with CO/H2, and preferentially catalyzes the reverse water gas shift to CO with the thermodynamically favored methanation pathway switched off. The charged dual sites can stabilize the activated CO2 species in a η2(C, O) bridge configuration, directly dissociate the C═O bond to *CO, and promote CO desorption. The mechanistic investigation has elucidated the reaction pathways in the Ni-catalyzed CO2 hydrogenation and identified the crucial intermediates that impacted the product selectivity, which can provide a theoretical guide for the Ni-based catalyst design.

Noncanonical Wnt5a/JNK Signaling Contributes to the Development of D-Gal/LPS-Induced Acute Liver Failure

Ji Xiang-Fen,Fan Yu-Chen,Sun Fei,Wang Jing-Wei,Wang Kai

Acute liver failure (ALF) is a deadly clinical disorder with few effective treatments and unclear pathogenesis. In our previous study, we demonstrated that aberrant Wnt5a expression was involved in acute-on-chronic liver failure. However, the role of Wnt5a in ALF is unknown. We investigated the expression of Wnt5a and its downstream c-Jun N-terminal kinase (JNK) signaling in a mouse model of ALF established by coinjection of D-galactosamine (D-Gal) and lipopolysaccharide (LPS) in C57BL/6 mice. We also investigated the role of Box5, a Wnt5a antagonist, in vivo. Moreover, the effect of Wnt5a/JNK signaling on downstream inflammatory cytokine expression, phagocytosis, and migration in THP-1 macrophages was studied in vitro. Aberrant Wnt5a expression and JNK activation were detected in D-Gal/LPS-induced ALF mice. Box5 pretreatment reversed JNK activation and eventually decreased the mortality rate of D-Gal/LPS-treated mice, with reduced hepatic necrosis and apoptosis, serum ALT and AST levels, and liver inflammatory cytokine expression, although the latter was not significant. We further demonstrated that recombinant Wnt5a (rWnt5a)-induced tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) mRNA expression and increased THP-1 macrophage phagocytosis in a JNK-dependent manner, which could be restored by Box5. In addition, rWnt5a-induced migration of THP-1 macrophages was also reversed by Box5. Our findings suggested that Wnt5a/JNK signaling plays an important role in the development of ALF and that Box5 could have particular hepatoprotective effects in ALF.

Enantioselective recognition of L/D-amino acids in the chiral nanochannels of a metal-organic framework

Xiaohui Niu,Simeng Yan,Jinliang Chen,Hongxia Li,Kunjie Wang

Chiral metal-organic frameworks (MOF) have received extensive attention from researchers due to their chiral properties and potential applications in chiral-related fields. However, the construction of chiral metal-organic framework remains challenging due to the unpredictability of the synthesis process and the difficulty in keeping chirality during the growth of chiral MOF. Herein, a chiral MOF (D-His-ZIF-8) is reported for the construction of electrochemical sensing interface, which was synthesized via in-situ substitution of 2-methylimidazole on ZIF-8 by D-histidine (D-His). The introduction of Ketjen Black (KB) to the D-His-ZIF-8 sensing interface can improve its conductivity. The electrochemical recognition results show that KB/D-His-ZIF-8 has enantiodiscrimination ability for amino acids in voltammetry experiments. The best recognition efficiency is observed with Trp enantiomers . The recognition mechanism shows that the chiral nanochannel of D-His-ZIF-8 is conducive to the binding of L-amino acid, which will lead to the recognition of chiral molecules of different configurations. This research points to the possibility of developing further chiral recognition protocols based on D-His-ZIF-8, particularly with the suggested improvement in the preparation procedure of chiral MOF.

Cannabidiol-Decorated Berberine-Loaded Microemulsions Improve IBS-D Therapy Through Ketogenic Diet-Induced Cannabidiol Receptors Overexpression

Xinyu Fan,Jiachen Shi,Ye Liu,Mengqiu Zhang,Min Lu,Ding Qu

Background Berberine (BR) shows promise as a candidate for treating irritable bowel syndrome with diarrhea (IBS-D). However, the undesired physicochemical properties and poor oral absorption limit its clinical translation. A ketogenic diet (KD) can induce intestinal overexpression of cannabidiol (CB) receptors, which may offer a potential target for IBS-D-specific delivery of BR.Methods The microemulsions loaded with BR and decorated with cannabidiol (CBD/BR-MEs) were developed through a one-step emulsion method. The pharmaceutical behaviors of the CBD/BR-MEs were measured using dynamic light scattering and high-performance liquid chromatography. The efficacy of the anti-IBS-D therapy was evaluated by assessing fecal water content, Bristol score, and AWR score. The intestinal permeability were assessed through immunofluorescent staining of CB1 and ZO-1, respectively. The signaling of CREB/BDNF/c-Fos was also studied along with immunofluorescent and immunohistochemical examination of brain sections. Results The CBD/BR-MEs, which had a particle size of approximately 30 nm and a surface density of 2% (wt%) CBD, achieved greater than 80% (wt%) encapsulation efficiency of BR. The pharmacokinetics performance of CBD/BR-MEs was significantly improved in the KD-fed IBS-D rats than the standard diet-fed ones, which is highly related to intestinal expression of CB1 receptors. The treatment with CBD/BR-MEs and KD exhibited evident comprehensive advantages over the other groups in terms of anti-IBS-D efficacy. CBD/BR-MEs and KD synergistically decreased intestinal permeability. Moreover, the treatment with CBD/BR-MEs and KD not only blocked the CREB/BDNF/c-Fos signaling in the brain but also decreased the levels of neurotrophic factors, neurotransmitters, and inflammatory cytokines in the serum of IBS-D model rats. Conclusion Such a design represents the first attempt at IBS-D-targeted drug delivery for improved oral absorption and efficacy through KD-induced target exposure, which holds promising potential for the treatment of IBS-D.
