In this work, a target-triggered three-way junction (3-WJ) structure was combined with enzyme-powered cascade amplification strategy for ultrasensitive electrochemiluminescence (ECL) detection of microRNA via free-running DNA walker. In the presence of target microRNA-21 (miRNA-21), a three-way junction structure was formed to trigger enzyme-aided multiple DNA amplification, and thus exponentially amplified triggers were generated. Then, the trigger DNA could hybridize with the blocking DNA on the electrode to release walker DNA, which specifically combined with the quantum dots (QDs)-DNA probe, and formed a recognition site for Nt.BbvCI. The movement of DNA walker is powered by the nicking endonuclease that cleaves specific QDs-DNA probes on the track. Thus one walker DNA can automatically cleave multiple QDs probes on the electrode during the movement, resulting in significantly amplified changes of ECL signal, which could be used for the highly sensitive detection of target miRNA-21. Under the optimal conditions, the ECL signal change is linear with the concentration of miRNA-21 in the range from 10 −14 M to 10 -7 M with a detection limit of 1.5 fM. This newly established strategy could achieve rapid, isothermal, and homogeneous signal amplification for specific nucleic acids in complicated biomatrix, which hold great potential for application in early clinical diagnosis.

Bioinspired nanofluidic systems that utilize ions as signal carriers hold great promise for emulating neural processing in biochemical sensing and neuromorphic computing. However, achieving parallel, brain-like processing of multiple biochemical signals remains a significant challenge. Herein, we present a nanofluidic artificial postsynaptic membrane (APM) functionalized with confined DNA aptamers to construct a neuromorphic signal processing platform. Target-induced conformational switching of DNA aptamers dynamically modulates ionic transport through nanochannels, effectively mimicking synaptic information transmission. The integration of cross-responsive aptamer-based APM units into a cascaded logic system enables signal processing without relying on the physical series network of nanochannels. By independently addressing and reading each unit, dendritic multi-input integration and brain-like information fusion are achieved at the signal-algorithm level, and 100% accurate discrimination of multiple targets is reached. This approach marks a conceptual shift from the traditional “one-probe-one-target” model toward a brain-inspired, multitarget recognition architecture. The fusion of DNA probes with nanofluidic logic and their cascade at the signal level enables the development of neuromorphic biochips with integrated processing capabilities for multiplexed signals.

A DNA-polyphenol wet-adhesive hydrogel is developed for rapid hemostasis. • The hydrogel exhibits robust wet adhesion and favorable coagulation-related performance. • Effective hemostasis is achieved in multiple bleeding models, including high-pressure arterial injury. Rapid hemostasis under wet and dynamic conditions remains a major challenge for wound management. Herein, a DNA-polyphenol wet-adhesive hydrogel was developed by integrating DNA with tannic acid (TA), dopamine (DOPA), and mesoporous bioactive glass (MBG). The resulting hydrogel exhibited robust wet adhesion, enabling effective sealing on various substrates and biological tissues. The integration of DNA, polyphenol components, and MBG endowed the hydrogel with enhanced adhesive properties, with an adhesive strength exceeding 35 kPa after prolonged immersion. In vitro studies demonstrated enhanced coagulation-related performance, including a reduced blood clotting index and shortened clotting time, together with high fluid absorption capacity. In vivo studies in rat liver injury, rat tail amputation, and rabbit femoral artery models further demonstrated effective hemostatic performance, with reduced blood loss and shortened hemostasis time, even under high-pressure bleeding conditions. In addition, the hydrogel exhibited good cytocompatibility, distinct in vitro release behavior, and progressive in vivo resorption with favorable tissue compatibility. These results suggest that the DNA-polyphenol hydrogel is a promising biomacromolecule-based material for rapid hemostasis and wound sealing applications.

A molecular beacon (MB) is an oligonucleotide probe with a stem-loop structure designed to fluoresce upon hybridization with a specific target nucleic acid. Herein, we introduce a novel i-motif DNA-based molecular beacon (iMB) designed for nucleic acid detection. DNA chemical sequencing showed that the iMB can be selectively opened when its target is present, thanks to the formation of a duplex between this target and the iMB recognition loop. Phase diagrams were introduced to analyze the binding properties between the iMB and its target, and to optimize the detection conditions with different combinations of pH and temperature. Benefiting from the environment-sensitive stability and special structural feature inherent to the i-motif, this iMB exhibits a controllable sensitivity and specificity through choosing appropriate temperature/pH combination and recognition loop position in the i-motif probe. Leveraging the duplex-specific nuclease (DSN) resistance of the i-motif, we significantly enhanced the sensitivity by more than four orders of magnitude through the implementation of a DSN-assisted amplification strategy. Finally, miRNAs in cell lines and patient plasma were detected by the iMBs, as validated via quantitative reverse transcription polymerase chain reaction (RT-qPCR). This research successfully broadens the structural repertoire of MB stems to include a pH- and temperature-responsive i-motif stem.

Disrupted potassium (K+) homeostasis is an early biochemical hallmark of altered membrane potential, electrolyte imbalance, and malignant transformation, yet in vivo K+ mapping remains unrealized. Optical sensors can report K+ fluctuations but lack the penetration depth required for deep-tissue imaging. Although magnetic resonance imaging (MRI) enables noninvasive, high-resolution visualization of biochemical processes, no contrast agent has achieved K+-dependent modulation of relaxivity, precluding true K+-selective MRI. Here we report a mechanistically defined K+-activated magnetic resonance sensor (KMRS) that translates G-quadruplex folding into magnetic relaxation control. KMRS links a Gd-DOTA reporter to an iron oxide (IO) quencher through a K+-selective DNA linker. In the low-K+ state, the short Gd-IO distance enforces strong dipolar coupling that suppresses spin fluctuations and minimizes longitudinal relaxivity (r1). K+ binding induces G-quadruplex formation, increases interparticle separation, restores Gd-centered rotational dynamics, and yields a pronounced enhancement in r1 value. This mechanism-driven MRI activation enables deep-tissue visualization of pathological K+ elevations, supporting early detection of malignant foci and ion-based discrimination between tumor and benign tissues. The design establishes a generalizable framework for engineering ion-responsive MRI probes.

TLR9 agonists hold significant potential in vaccine development and cancer immunotherapy. While synthetic CpG oligodeoxynucleotides (ODNs) have been employed in immunotherapies, their clinical application remains constrained by dose-limiting toxicity and metabolic instability. DNA nanotechnology provides a promising strategy for the precise modulation of ligand-TLR9 interactions. In this study, DNA nanostructure-based TLR9 agonists were rationally designed by optimizing the spatial orientation of 5′-TCG ligands based on the two DNA-binding sites revealed in the TLR9 receptor crystal structure. Fine-tuning DNA nanostructure configurations enabled controllable modulation of TLR9 activation with structural flexibility identified as a critical determinant. Notably, the oligomerization of DNA nanostructures markedly enhanced TLR9 stimulation efficacy, establishing a paradigm for the rational design of TLR9 agonists. These findings advance innovative approaches for developing next-generation immunotherapeutic agents.

Porous silicon (PSi) layers are prepared on the single-crystal silicon surface by electrochemical etching, and PSi membranes (PSiMs) are fabricated by separating the PSi layers from the single-crystal silicon substrate using high-current pulses. PSiMs have better adsorption and response time than nonseparated PSi layers, making them more suitable for sensing applications. PSi microcavity (PSM) structures are widely used for the detection of biomolecules in biosensors because of their high transmission resonance peaks with narrow full-width at half-maximum (FWHM) in the reflection spectra. However, when compared to PSiMs, the permeability of the PSM on the silicon substrate is low, resulting in a significant reduction in its detection sensitivity. In this study, an efficient self-supported Bragg stacked microcavity (SBSM) structure was prepared for the detection of DNA biomolecules by combining the advantages of PSM and PSiMs. The redshift of the reflectance spectra obtained in the concentration range of 0.1– $0.5 ~\mu \text{M}$ shows a good linear relationship, with a detection limit of 2.2 nM. The detection sensitivity is improved by more than a factor of ten when compared to the PSM structure on the silicon substrate. The novel device allows DNA biomolecules to enter the porous layers more easily, especially the defective layer, thus improving its detection sensitivity.

Thermotropic liquid crystals (TLCs) have created unpredictable applications ranging from sensor to electronic devices. DNA can interact with positively charged cationic and anionic (catanionic) vesicles by electrostatic interaction to form DNA-based TLCs. In the present work, the polarizing optical microscopy (POM) images clearly demonstrate the formation of DNA-involved liquid crystals, we explore the general role of DNA-involved TLC formation and the regulating factors of properties of DNA-based TLCs. We verify the effect of the chain lengths of surfactants and DNA on the formation and properties of TLCs. We found that DNA-involved TLCs constructed by the different surfactants with same chain length present the excellent properties. The position of functional group of ferrocenyl surfactants greatly affects the formation of TLCs. The bulky group closing to the headgroup of surfactant and the strong electrostatic interaction between positively charged catanionic vesicles and DNA affect the formation of TLCs. Furthermore, we found that the shorter DNA strand length, the more consistent fit better with the vesicles to enhance the thermal stability of DNA-based TLCs. The influence factors of the formation of DNA-based TLCs were explored and the importance of interactions to the architecture of DNA-based TLCs also was underscored. Our results may direct the design and synthesis of functional DNA-based TLCs and lay the foundation of applications in the field of electrochemistry and flexible electronic devices.

Plastics play important roles in modern life and currently the development of plastic recycling is highly demanding and challenging. To relieve this dilemma, one option is to develop new sustainable bioplastics that are compatible with the environment over the whole material life cycle. We report a sustainable bioplastic made from natural DNA and biomass-derived ionomers, termed as DNA plastics. The sustainability involves all aspects of the production, use, and end-of-life options of DNA plastics: (1) the raw materials are derived from biorenewable resources; (2) the water-processable strategy is environmentally friendly, not involving high-energy consumption, the use of organic solvents, and the production of byproducts; (3) recyclable and nondestructive use is achieved to significantly prolong the service lifetime of the plastics; and (4) the disposal of waste plastics follows two green routes including the recycling of waste plastics and enzyme-triggered controllable degradation under mild conditions. Besides, DNA plastics can be “aqua-welded” to form arbitrary designed products such as a plastic cup. This work provides a solution to transform biobased hydrogel to bioplastic and demonstrates the closed-loop recycling of DNA plastics, which will advance the development of sustainable materials.

The study aimed to achieve enhanced targeted cytotoxicity and cell-internalization of cisplatin-loaded deoxyribonucleic acid-nanothread (CPT-DNA-NT), mediated by scavenger receptors into HeLa cells. DNA-NT was developed with stiff-topology utilizing circular-scaffold to encapsulate CPT. Atomic force microscopy (AFM) characterization of the DNA-NT showed uniformity in the structure with a diameter of 50–150 nm and length of 300–600 nm. The successful fabrication of the DNA-NT was confirmed through native-polyacrylamide gel electrophoresis analysis, as large the molecular-weight (polymeric) DNA-NT did not split into constituting strands under applied current and voltage. The results of cell viability confirmed that blank DNA-NT had the least cytotoxicity at the highest concentration (512 nM) with a viability of 92% as evidence of its biocompatibility for drug delivery. MTT assay showed superior cytotoxicity of CPT-DNA-NT than that of the free CPT due to the depot release of CPT after DNA-NT internalization. The DNA-NT exhibited targeted cell internalizations with the controlled intracellular release of CPT (from DNA-NT), as illustrated in confocal images. Therefore, in vitro cytotoxicity assessment through flow cytometry showed enhanced apoptosis (72.7%) with CPT-DNA-NT (compared to free CPT; 64.4%). CPT-DNA-NT, being poly-anionic, showed enhanced endocytosis via scavenger receptors.

A variety of three-dimensional DNA assemblies have been proposed as drug carriers owing to their good biocompatibility and easy fabrication. In this study, inspired by the structure of cockleburs, a novel aptamer-tethered DNA assembly was developed for effective targeted drug delivery. The Apt-nanocockleburs were fabricated via a facile process of DNA base pairing: four complementary DNA single strands, including one aptamer-ended strand and three sticky-end strands, were applied to pair with each other. The main body of the nanocockleburs can load doxorubicin (Dox) whilst the covered aptamer spines bind to the target MCF-7 cells. The self-assembled Apt-nanocockleburs exhibit higher cell uptake as well as increased cytotoxicity to MCF-7 cells than DNA nanocockleburs without aptamers. This study provided a DNA constructing platform to produce new drug carriers with high selectivity for cancer targeted drug delivery.
