Silicon is an essential component of various environmental processes and biogeochemical cycles with silicate (H 4 SiO 4 ) representing the bioavailable form of silicon in waters and soils. This study aimed to develop a method for accurately measuring labile silicate levels in waters and soils using diffusive gradients in thin-films (DGT) with a hydrogel diffusive gel and a modified precipitated zirconia (mPZ) binding gel. The diffusion coefficient of silicate in the diffusive gel, measured using an independent diffusion cell, was 6.77 × 10 −6 cm 2 s −1 at 25 °C. The mPZ gels were found to be effective at quickly and efficiently binding silicate, and a mixed solution of 0.5 mol L −1 NaOH and 0.5 mol L −1 H 2 O 2 was determined to effectively elute silicate from mPZ gels, with an elution factor of 1.0. Furthermore, mPZ-DGT was found to be able to measure silicate at various pH levels (4.03–7.93) and ionic strengths (0.1–1000 mmol L −1 NaNO 3 ). Field application of mPZ-DGT devices in an urban river revealed a decrease in silicate concentration downstream compared to upstream, likely due to increased human activity. Finally, mPZ-DGT was used in four sandy or loam soils to measure silicate concentration, and the results showed relatively small variations compared to those measured by CaCl 2 and ammonium oxalate extractions. The findings suggest that mPZ-DGT is a useful tool for monitoring labile silicate concentrations and assessing its bioavailability in waters and soils.

The breakup of water-in-water droplets in a Y-junction microchannel was experimentally investigated, focusing on the unobstructed breakup mode due to its broad applicability. This process was divided into four stages: entering, squeezing, pinch-off, and filament rupture, based on the evolution of the neck width and its thinning rate. In the entering stage, neck thinning accelerates with increasing dispersed-phase flow rate Q d , droplet length l 0 , and two-phase viscosity ratio λ , and slows down with increasing continuous phase flow rate Q c . In the squeezing stage, the minimum neck width follows a power-law relation with time. The thinning rate increases with higher Q d , l 0 , and λ , whereas increasing Q c inhibits thinning. In the pinch-off stage, neck width decreases linearly with time, and the thinning is enhanced by higher Q c , Q d , and l 0 , but remains nearly independent of λ . The filament rupture stage follows a power-law thinning trend, and exhibits similar parameter dependencies, though satellite droplets may form at high Q d and Q c , and show increased instability at low viscosity. These findings offer insights into neck dynamics and interfacial rupture in low interfacial tension systems.

Peptide-drug conjugates (PDCs) are an important class of therapeutic agents that combine one or more active molecules with a short peptide via a biodegradable spacer to generate prodrugs. Here an antioxidant (i.e., 3,5-dihydroxybenzoic acid), a non-steroidal anti-inflammatory drug (i.e., ibuprofen, IPF), and a self-assembled peptide motif (i.e., Gly-Phe-Phe-Tyr-Asp, GFFYD) are integrated to produce a peptide-drug amphiphile, which self-assembles to form hydrogel and further exerts the synergistic effects, and consequently emerges high therapeutic efficacy. In the in vitro study toward lipopolysaccharide-activated RAW264.7 macrophages, the resulting hydrogel significantly reduces the production of cytokines (e.g., IL-1β, IL-6 and tumor necrosis factor-α (i.e., TNF-α)) compared with that of native IPF via the inhibition of nuclear factor-κB (i.e., NF-κB), janus kinase-signal transducer and activator of transcription (i.e., JAK-STAT) signals and NLR family pyrin domain containing 3 (i.e., NLRP3) inflammasome activation. Moreover, a rabbit model of endotoxin induced uveitis evidences the superior therapeutic efficacy of the resulting hydrogel over that of clinically used 0.1 wt% diclofenac sodium eyedrop. Overall, this work offers new insights into the anti-inflammatory mechanism of PDCs hydrogel and presents the therapeutic values in the treatment of ocular disorders, and ultimately fulfils the potential clinical benefits of PDCs.

The increasing power density of chips poses a significant challenge in the form of material aging for aluminum-filled polydimethylsiloxane (Al/PDMS) composites, which are widely used in thermal interface materials. Despite the growing importance of this issue, the specific mechanisms behind the interfacial aging process remain elusive, hindering a comprehensive grasp of the aging dynamics in these composites. In our research, we have developed an in-situ Raman aging monitoring system that leverages the non-contact and high-resolution capabilities of Raman spectroscopy to study the interface aging process. Our findings indicate a notable decrease in the intensity of the Raman peak as further cross-linking of the molecules during aging, with the most pronounced decline occurring at the interface between aluminum and PDMS. This insight could potentially elucidate why the interface in composite materials is frequently the site of failure during aging. Our study offers a versatile methodology for investigating the interfacial aging of polymer composites, contributing to a broader understanding of the interface behavior in composite materials at the molecular level.

Milk powder is a common food in most families. It is of great significance to accurately detect the quality and safety of milk powder to mitigate food safety problems. This paper presents a method for the determination of vanillin and ethyl vanillin in milk powder based on terahertz (THz) spectroscopy. Samples with varying concentration gradients of these two additives were prepared, and a terahertz time-domain spectrometer was used to collect spectral data from the samples in the 0.2 to 1.5 THz range. Seven spectral preprocessing algorithms were evaluated using the partial least squares (PLS) method, and it was found that the combination of multivariate scattering correction (MSC) and Savitzky-Golay (SG) smoothing preprocessing yielded the best results, significantly improving the accuracy of the test sets for both additives. Subsequently, nine quantitative detection methods were constructed by combining three dimensionality reduction algorithms (ant colony algorithm (ACO), competitive adaptive reweighted sampling (CARS), and successive projection algorithm (SPA)) with three regression models (support vector regression (SVR), long short-term memory (LSTM), and particle swarm optimization-back propagation (PSO-BP)). The results showed that the LSTM regression model, with dimensionality reduction performed by the CARS algorithm, performed best for detecting vanillin in milk powder, achieving a recognition rate of 94.49%. Compared to the other eight methods, this increased the recognition rate by 7.69%. Similarly, the LSTM regression model, combined with the SPA algorithm for dimensionality reduction, performed best for detecting ethyl vanillin in milk powder, reaching a recognition rate of 98.37%. This represented a 6.59% increase in recognition rate over the other eight methods, providing a novel technical approach for non-destructive testing and analysis of milk powder quality and safety.

The objective of the present study was to evaluate structure–activity relationships of four sanshools on antioxidant activities. In vitro, hydroxy-α-sanshool (HAS), hydroxy-β-sanshool (HBS), and hydroxy-γ-sanshool (HRS) had stronger antioxidant capacities than hydroxy-γ-sanshool (RS), attributed to the hydroxyl group. Furthermore, HRS had the greatest oxygen radical absorbance capacities (ORAC) and free radical scavenging (DPPH and ABTS) capacities, attributed to more cis-double bonds. Sanshools could effectively decrease the level of oxidative stress in colonic tissue and HCT-116 cells, including increased enzyme activities of catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) and decreased concentrations of malondialdehyde (MDA). After intragastric administration of sanshool, HRS increased the the level of hepatic Total antioxidant capacity (T-AOC) and colon GSH-Px and SOD. Moreover, sanshools significantly increased Nrf2, HO-1 proteins and decreased Keap1 protein. The protective effect of HRS was much higher than HAS, HBS, and RS, apparently due to its amide and hydroxyl groups. The binding energies of Nrf2 with HAS, HBS, HRS, and RS were −5.17, −5.11, −5.75, and −4.16 kcal/mol, respectively. HRS is more easily combine with Nrf2. In summary, sanshools acted as substitutes for natural antioxidants, in which HRS has the greatest antioxidant activity. This study provided a basis for future research on relationships between structure and activity.

Global warming and acidification, induced by a substantial increase in anthropogenic CO2 emissions, are expected to have profound impacts on biogeochemical cycles. However, underlying mechanisms of nitrous oxide (N2O) production in estuarine and coastal sediments remain rarely constrained under warming and acidification. Here, the responses of sediment N2O production pathways to warming and acidification were examined using a series of anoxic incubation experiments. Denitrification and N2O production were largely stimulated by the warming, while N2O production decreased under the acidification as well as the denitrification rate and electron transfer efficiency. Compared to warming alone, the combination of warming and acidification decreased N2O production by 26 ± 4%, which was mainly attributed to the decline of the N2O yield by fungal denitrification. Fungal denitrification was mainly responsible for N2O production under the warming condition, while bacterial denitrification predominated N2O production under the acidification condition. The reduced site preference of N2O under acidification reflects that the dominant pathways of N2O production were likely shifted from fungal to bacterial denitrification. In addition, acidification decreased the diversity and abundance of nirS-type denitrifiers, which were the keystone taxa mediating the low N2O production. Collectively, acidification can decrease sediment N2O yield through shifting the responsible production pathways, partly counteracting the warming-induced increase in N2O emissions, further reducing the positive climate warming feedback loop.

Background and Objective Ciclopirox is a widely used antifungal drug, redisposition of which has drawn increasing attentions due to multiple promising activities. The drug undergoes extensive glucuronidation, which acts as a major obstacle in the ongoing novel application and still remains poorly understood. The current study aims to phenotype ciclopirox glucuronidation pathway and as well to decipher the related species differences. Methods Ciclopirox glucuronidation was investigated in liver microsomes from humans (HLM) and various experimental animals. Assays with recombinant uridine diphosphate glucuronosyltransferases (UGTs), enzyme kinetic analyses and selective inhibitors were used to determine the role of individual UGTs in ciclopirox glucuronidation. Results HLM is highly active in ciclopirox glucuronidation with Michaelis–Menten constant ( K m ), maximum velocity ( V max ), and intrinsic clearance (CL int ) values of 139 μM, 7.89 nmol/min/mg, and 56 μL/min/mg, respectively. UGT1A9 displays by far the highest activity, whereas several other isoforms (UGT1A6, UGT1A7, and UGT1A8) catalyze formation of traced glucuronides. Further kinetic analysis demonstrates that UGT1A9 has a closed K m value (167 μM) to HLM. UGT1A9 selective inhibitor (magnolol) can potently inhibit ciclopirox glucuronidation in HLM with the IC 50 value of 0.12 μM. The reaction displays remarkable differences across liver microsomes from mice, rats, cynomolgus monkey, minipig, and beagle dog, with the CL int values in the range of 26–369 μL/min/mg. In addition, ciclopirox glucuronidation activities of experimental animals’ liver microsomes were less sensitive to magnolol than that of HLM. Conclusions Ciclopirox glucuronidation displays remarkable species differences with UGT1A9 as a dominant contributor in humans. It is suggested that the pharmacological or toxicological effects of ciclopirox may be UGT1A9 and species dependent.

Itaconate serves as an immune-specific metabolite that regulates gene transcription and metabolism in both host and pathogens. S -itaconation is a post-translational modification that regulates immune response; however, its antimicrobial mechanism under the physiological condition remains unclear. Here, we apply a bioorthogonal itaconate probe to perform global profiling of S -itaconation in living pathogens, including S. Typhimurium, S. aureus , and P. aeruginosa . Some functional enzymes are covalently modified by itaconate, including those involved in the de novo purine biosynthesis pathway. Further biochemical studies demonstrate that itaconate suppresses this specific pathway to limit Salmonella growth by inhibiting the initiator purF to lower de novo purine biosynthesis and simultaneously targeting the guaABC cluster to block the salvage route. Our chemoproteomic study provides a global portrait of S -itaconation in multiple pathogens and offers a valuable resource for finding susceptible targets to combat drug-resistant pathogens in the future.

Cycling is not just a skilled accomplishment by individual cyclists, it can also be social. Cycling to work with other commuters or learning to ride a bike with a caregiver, for example, both involve a keen attention to negotiating and maintaining being together in and through cycling and being seen to be together in a “mobile with”. This article reports on an investigation of some of the different vélomobile formations-in-action that involve specific arrangements of bodies on bikes and configurations of a “vélomobile with”. Video recordings of commuter and family bike rides using consumer and micro video cameras from multiangles were made to capture aural and visual features of the local organization of the ride from the participants’ perspective(s). Several phenomena are presented and discussed, including starting, stopping, and maintaining a vélomobile formation; singling up and tucking in; and stretchy mobile formations. The analysis starts by analyzing side-by-side arrangements, which are then temporarily disrupted, and finishes with formations that are more extensively stretched, but which still afford opportunities for social interaction.

Electrospinning is now commercially used for the fabrication of nano/micro fibers. Compared with spider dragline silk, artificial fibers have poor mechanical properties. Unlike natural silk, which has a hierarchical structure with an approximate 3-fold symmetry, the molecular structure of spun fiber has neither folding nor orientation. To date, it is almost impossible to control molecule orientation during the spinning process. Here, we show that macromolecule orientation can be easily controlled using the laminar flow of fluid mechanics. A lasting laminar flow in a long needle can order macromolecules. We find that the orientation of macromolecules can greatly affect the morphology and mechanical properties of fibers. We expect our technology to be helpful for more sophisticated fabrication of fibers with ordered macromolecules and DNA-like twists. Keywords:macromolecule;laminar flow;electrospinning;hierarchical structure

The structure of DNA is susceptible to alterations at high temperature and on changing pH, irradiation and exposure to DNase. Options to protect and preserve DNA during storage are important for applications in genetic diagnosis, identity authentication, drug development and bioresearch. In the present study, the stability of total DNA purified from human dermal fibroblast cells, as well as that of plasmid DNA, was studied in silk protein materials. The DNA/silk mixtures were stabilized on filter paper (silk/DNA + filter) or filter paper pre-coated with silk and treated with methanol (silk/DNA + PT-filter) as a route to practical utility. After air-drying and water extraction, 50–70% of the DNA and silk could be retrieved and showed a single band on electrophoretic gels. 6% silk/DNA + PT-filter samples provided improved stability in comparison with 3% silk/DNA + filter samples and DNA + filter samples for DNA preservation, with ∼40% of the band intensity remaining at 37 °C after 40 days and ∼10% after exposure to UV light for 10 hours. Quantitative analysis using the PicoGreen assay confirmed the results. The use of Tris/borate/EDTA (TBE) buffer enhanced the preservation and/or extraction of the DNA. The DNA extracted after storage maintained integrity and function based on serving as a functional template for PCR amplification of the gene for zinc finger protein 750 (ZNF750) and for transgene expression of red fluorescence protein (dsRed) in HEK293 cells. The high molecular weight and high content of a crystalline beta-sheet structure formed on the coated surfaces likely accounted for the preservation effects observed for the silk/DNA + PT-filter samples. Although similar preservation effects were also obtained for lyophilized silk/DNA samples, the rapid and simple processing available with the silk–DNA–filter membrane system makes it appealing for future applications.
