Se@SiO2@Au-PEG/DOX NCs as a multifunctional theranostic agent efficiently protect normal cells from oxidative damage during photothermal therapy

Wenyan Kong,Qi Wang,Guoying Deng,Hang Zhao,Linjing Zhao,Jie Lu,Xijian Liu

Photothermal therapy (PTT) is a promising treatment for tumors due to its efficiency and non-invasiveness. However, during the PTT treatment, reactive oxygen species (ROS) are produced in response to hyperthermia and thus harm the neighboring normal cells. In this work, a multifunctional theranostic agent (Se@SiO2@Au-PEG/DOX NCs) was exploited to solve this problem by introducing selenium, which can efficiently prevent normal cells from oxidative damage by scavenging reactive oxygen species during photothermal therapy. In addition, the Se@SiO2@Au-PEG/DOX nanocomposites (NCs) not only exhibited excellent properties of combined chemo-thermal synergistic therapy, but also showed no appreciable toxicity towards normal tissues due to the protective effect for continuous release of selenium. Thus, the fabricated Se@SiO2@Au-PEG/DOX NCs provide an integrated solution to overcome the limitations of selenium and PTT, and demonstrate great prospects as a safe and highly reliable theranostic agent.

Cell-penetrating peptide together with PEG-modified mesostructured silica nanoparticles promotes mucous permeation and oral delivery of therapeutic proteins and peptides

Xinyi Tan,Yan Zhang,Qian Wang,Tianyang Ren,Jingxin Gou,Wei Guo,Tian Yin,Haibing He,Yu Zhang,Xing Tang

Poor permeation across intestinal mucous barriers often limits the oral delivery of prospective therapeutic proteins and peptides (TPPs). In order to address this issue, cell penetrating peptide (CPP) together with PEG modified and pore-enlarged mesostructured silica nanoparticle (NP) were constructed to form the mucus-penetrating electrostatic particle-complexes, CPP/TPP/NP. Alone, CPP and TPP often present with poor stability, and their traditional electrostatic complex shows reduced pharmacodynamics. To provide satisfactory protection, silica NPs were loaded with CPP and TPP (CPP@NP and TPP@NP), respectively, and then CPP@NP and TPP@NP could together form CPP/TPP/NP via electrostatic interaction. As a result, CPP involvement with PEG modification showed an 8.45-, 1.62- and 5.09-fold increase in cellular uptake, exocytosis and final transcellular permeation in mucous conditions, respectively. It was found that CPP involvement mainly affected transport and exocytosis, and the PEG polymer significantly influenced mucous penetration and cellular uptake, which could further promote CPP ability for uptake and exocytosis. Additionally, NP-mediated CPP/TPP/NP showed a similar uptake mechanism with supporting carriers (clathrin-mediated endocytosis), and could strengthen transcellular routes (the endoplasmic reticulum–Golgi apparatus pathway and the lysosome route). Utilizing recombinant growth hormone (RGH) as a model TPP, oral administration of the RGH-loaded CPP/TPP/LMSN-PEG10k with hydrophilic and electroneutral properties induced 5.41- and 4.91-fold increases in pharmacodynamics in vitro and in vivo, respectively. Thus, CPP/TPP/NP significantly promoted mucous permeation and shows promising potential for oral delivery of TPPs.

Thermochromic and tunable emissivity properties of BCG/CaCl2/PEG-g-CDA composites for smart adaptive camouflage in visible and infrared wavebands

Honghan Bu,Chen Xu,Guoyue Xu,Chuyang Liu,Shujuan Tan,Tengchao Guo,Shanshan Xiang

BCG/CaCl 2 /PEG-g-CDA composites with different mass ratios of BCG and CaCl 2 under constant PEG-g-CDA were synthesized via liquid-phase reaction method. The composites were analyzed and characterized using FT-IR, XRD , DSC and UV–vis. The results show that the color changes visibly from green to yellow by improving temperature from 50 °C to 62 °C, and turns back to green reversibly with temperature cooling back to room temperature. The infrared emissivity values increase rapidly at the temperature range of 50 °C–62 °C, and the maximum variation reaches 0.23. Furthermore, the mechanisms for thermochromic phenomenon and tunable infrared emissivity were also proposed. The reversible thermochromic phenomenon is attributed to the molecular structure transformation of BCG between ring-opened quinone structure and ring-closed lactone structure. The tunable emissivity at different temperatures is ascribed to the changes of resistivity under temperature fluctuations. Our results suggest that BCG/CaCl 2 /PEG-g-CDA composites are a potential candidate for smart adaptive camouflage in visible and infrared waveband.

Polymeric micelles with α-glutamyl-terminated PEG shells show low non-specific protein adsorption and a prolonged in vivo circulation time

Xiaoju Wang,Cuiping Yang,Chenhong Wang,Leijia Guo,Tianhong Zhang,Zhenqing Zhang,Husheng Yan,Keliang Liu

Although PEG remains the gold standard for stealth functionalization in drug delivery field up to date, complete inhibition of protein corona formation on PEG-coated nanoparticles remains a challenge. To improve the stealth property of PEG, herein an α-glutamyl group was conjugated to the end of PEG and polymeric micelles with α-glutamyl-terminated PEG shells were prepared. After incubation with bovine serum albumin or in fetal calf serum, the size of the micelles changed slightly, while the size of the micelles of similar diblock copolymer but without α-glutamyl group increased markedly. These results indicated that the micelles with α-glutamyl-terminated PEG shells showed low non-specific protein adsorption. In vivo blood clearance kinetics assay showed that the micelles with α-glutamyl-terminated PEG shells exhibited a longer in vivo blood circulation time compared with similar micelles but without α-glutamyl groups. The better stealth property of the micelles with α-glutamyl-terminated PEG shells was presumably attributed to the zwitterionic property of the α-glutamyl groups.

One-pot preparation of boronic acid and PEG bi-functionalized silica particles for separation and purification of catecholamine from rat serum

Yan Wang,Lei Sun,Bingqing Yu,Shaotong Li,Shian Zhong

A facile one-pot preparation of aminophenylboronic acid (APBA) and poly(ethyleneglycol) (PEG) bi-functionalized silica particles named SiO2@APBA@PEG by the aid of epoxy silanes is described in this work. The as-prepared particles could not only selectively extract the catecholamine through boronate binding but could also effectively resist protein adsorption using PEG brushes; FT-IR spectra and contact angle measurements proved that APBA and PEG groups were successfully introduced into the surface of silica. The maximum adsorption capacities towards dopamine (DA) and levodopa (L-DOPA) were found to be 254.45 and 104.71 μmol g−1, respectively. The as-prepared material was used as a solid phase extraction (SPE) sorbent for the extraction of catecholamine from rat serum. The recoveries of DA and L-DOPA were 95 ± 3.21% and 93 ± 3.57%, meanwhile, proteins were eliminated. The obtained results indicated that the SiO2@APBA@PEG particles were qualified for efficient purification of catecholamine from complex biological samples.

A well-defined biodegradable 1,2,3-triazolium-functionalized PEG-b-PCL block copolymer: facile synthesis and its compatibilization for PLA/PCL blends

Wang Ping,Zhang Di,Zhou Yiyang,Li Yu,Fang Huagao,Wei Haibing,Ding Yunsheng

A novel biodegradable 1,2,3-triazolium-functionalized PEG- b -PCL copolymer (TAPEC) was synthesized by the “click” coupling of methoxypolyethylene glycol azide and α-propargyl-ω-hydroxyl-poly(ε-caprolactone), followed by the quaternization of the 1,2,3-triazole moiety with iodomethane. All the intermediates and TAPEC were characterized by 1 H NMR, FT-IR, and gel permeation chromatography (GPC). Taking advantage of the characteristics of ionic liquid and block copolymer, this ion-containing diblock copolymer is expected to be used as a novel compatibilizer in mixed biopolyester for regulating the interface and crystallization behaviors. Hence, the TAPEC was evaluated as a compatibilizer and an interface emulsifier in the blends of polylactic acid (PLA) and poly(ε-caprolactone) (PCL). Non-isothermal crystallization experimental results showed that the TAPEC with the higher amount of ε-caprolactone units induces a plasticization and nucleate effect that increased the crystallization ability of the PLA phase; meanwhile, in the PCL phase, the agminated ionic cluster acting as a nucleating agent significantly increased the crystalline of PCL.

Influence of pre-gelation temperature on mechanical properties of cellulose aerogels based on a green NaOH/PEG solution—a comparative study

Wan Caichao,Jiao Yue,Li Jian

Cellulose aerogels with porous three-dimensional network structure often suffer from weak mechanical strength, which imposes huge restrictions on their applications. In this work, an easily-operated pre-gelation treatment was introduced to the conventional fabrication technique of regenerated cellulose aerogels (i.e., dissolution, regeneration, and drying treatment) to improve their structure uniformity, crystallinity, and mechanical strength. These properties were found to be closely related to the pre-gelation temperature, and a relatively lower temperature (50 °C) makes the greatest contribution. Especially for the mechanical strength, the elasticity modulus and energy absorption of the aerogels treated at 50 °C reach the maximums of 1.03 MPa and 476.2 kJ m −3 , which are 5.4 times and 94.3 % higher than those of the untreated aerogels. As an example of potential application, the surface of the aerogels was decorated with multiwalled carbon nanotubes. The obtained composite displays a good electromagnetic interference shielding ability.

Preparation and characterization of Fe3O4-NH2@4-arm-PEG-NH2, a novel magnetic four-arm polymer-nanoparticle composite for cellulase immobilization

Juan Han,Li Wang,Yun Wang,Jian Dong,Xu Tang,Liang Ni,Lei Wang

A geometrically symmetric four – arm dendritic polymer 4-arm-PEG-NH 2 was employed to modify on the surface of Fe 3 O 4 as support for cellulase immobilization. Glutaraldehyde was used as a coupling agent of this magnetic support to covalently immobilize cellulase. The morphology and properties of the synthesized magnetic material were characterized by Fourier transform infrared (FT-IR) spectroscopy, vibrating-sample magnetometer (VSM), thermogravimetric analysis (TGA), transmission electron microscopy (TEM) and X-ray diffraction (XRD). The properties of immobilized enzyme such as the optimum temperature and pH, Michaelis constant, thermal stability, reusability and storage stability were investigated. The loading capacity of cellulase was 132   mg/g. Compared with free cellulase, immobilized cellulase showed wider pH and temperature ranges, higher operational stability and good storage stability. Besides, the results of hydrolysis filter paper and microcrystalline cellulose showed that the catalytic effect of immobilized cellulase on the free cellulase was increased by about 76%.

Co-delivery of Paclitaxel/Atovaquone/Quercetin to regulate energy metabolism to reverse multidrug resistance in ovarian cancer by PLGA-PEG nanoparticles

Qingyu Lu,Wenhao Gao,Zhenzhen Chen,Zhihong Liu,Jie Wang,Lingjun Zeng,Xiaomu Hu,Enqin Zheng,Qian Zhang,Hongtao Song

Ovarian cancer is a malignant tumor that seriously endangers the lives of women, with chemotherapy being the primary clinical treatment. However, chemotherapy encounters the problem of generating multidrug resistance (MDR), mainly due to drug efflux induced by P-glycoprotein (P-gp), which decreases intracellular accumulation of chemotherapeutic drugs. The drugs efflux mediated by P-gp requires adenosine triphosphate (ATP) hydrolysis to provide energy. Therefore, modulating energy metabolism pathways and inhibiting ATP production may be a potential strategy to reverse MDR. Herein, we developed a PTX-ATO-QUE nanoparticle (PAQNPs) based on a PLGA-PEG nanoplatform capable of loading the mitochondrial oxidative phosphorylation (OXPHOS) inhibitor atovaquone (ATO), the glycolysis inhibitor quercetin (QUE), and the chemotherapeutic drug paclitaxel (PTX) to reverse MDR by inhibiting energy metabolism through multiple pathways. Mechanistically, PAQNPs could effectively inhibit the OXPHOS and glycolytic pathways of A2780/Taxol cells by suppressing the activities of mitochondrial complex III and hexokinase II (HK II), respectively, ultimately decreasing intracellular ATP levels in tumor cells. Energy depletion can effectively inhibit cell proliferation and reduce P-gp activity, increasing the chemotherapeutic drug PTX accumulation in the cells. Moreover, intracellular reactive oxygen species (ROS) is increased with PTX accumulation and leads to chemotherapy-resistant cell apoptosis. Furthermore, PAQNPs significantly inhibited tumor growth in the A2780/Taxol tumor-bearing NCG mice model. Immunohistochemical (IHC) analysis of tumor tissues revealed that P-gp expression was suppressed, demonstrating that PAQNPs are effective in reversing MDR in tumors by inducing energy depletion. In addition, the safety study results, including blood biochemical indices, major organ weights, and H&E staining images, showed that PAQNPs have a favorable in vivo safety profile. In summary, the results suggest that the combined inhibition of the two energy pathways, OXPHOS and glycolysis, can enhance chemotherapy efficacy and reverse MDR in ovarian cancer.

A Novel Degradable Three-Dimensional Binder DSM-g-PAA-c-PEG for a High-Performance Si Anode in Lithium-Ion Batteries

Zijian Zhao,Zhenyang Zheng,Kaixiang Chen,Mengqi Ma,Mingqiu Zhang,Wenhong Ruan

Silicon anodes have been extensively researched due to their high capacity and potential to revolutionize energy storage. However, during lithiation and delithiation cycles, the volume expansion of silicon anodes can lead to mechanical damage and significant capacity decay. To address this issue, three-dimensional polymer binders have been introduced to suppress volume changes by providing mechanical support. Although cross-linking in these binders improves their robustness, current options with high molecular weights and cross-linked structures are not biodegradable. To provide an environmentally friendly solution, we propose grafting degradable starch microspheres (DSM) with poly(acrylic acid) (PAA) and cross-linking with polyethylene glycol (PEG) to form DSM-g-PAA-c-PEG, a binder that demonstrates strong adhesive properties and superior mechanical strength compared to PAA. The UV-Visible spectrum confirms DSM’s continued degradability. After cycling the Si@DSM-g-PAA-c-PEG anode 100 times at 0.25 C, it achieves a capacity retention rate of 58.1 % with a reversible capacity of 2127.8 mAh g −1 . This work provides new insight into the engineering of next-generation silicon electrode using an environmentally-friendly binding strategy, which may advance the large-scale industrial application of lithium-ion batteries.

Low-dose STS–PEG–chitosan coating on PLCL scaffolds: A multifunctional strategy for endothelialization, anti-inflammation, and antithrombosis in vascular grafts

Yutong Liu,Xiaolin Sun,Yunhuan Li,Kuihua Zhang,Zhiyong Yan,Changlin Zhai,Jinglei Wu,Anlin Yin

The long-term success of small-diameter vascular grafts remains limited by thrombosis, chronic inflammation, and poor endothelialization. To address these challenges, we developed a multifunctional coating strategy by integrating polyethylene glycol–chitosan (PEG–CS) with low-dose sodium tanshinone IIA sulfonate (STS, 0.01 %) on poly(L-lactide-co-ε-caprolactone) (PLCL) scaffolds. The PEG–CS/STS coating significantly improved hydrophilicity and structural stability, while providing bioactivity that promoted endothelial cell proliferation and alignment. In vitro studies demonstrated markedly reduced platelet adhesion and activation, along with a favorable shift in macrophage polarization toward the anti-inflammatory M2 phenotype. These synergistic effects create a vascular-friendly microenvironment that promotes endothelialization and suppresses thrombosis and inflammation. PEG reduces platelet adsorption, allowing STS to exert its anti-inflammatory effects, while chitosan’s electrostatic interaction with STS supports its stable loading and controlled release, enhancing its bioactivity. Our findings highlight that the PEG-CS/STS modified scaffold effectively integrates endothelialization, immunomodulation, and antithrombotic protection, representing a promising next generation strategy for small diameter vascular grafts.

A PEGylated alternating copolymer with oxidation-sensitive phenylboronic ester pendants for anticancer drug delivery

Yu Zhang,Pan He,Xinming Liu,Huailin Yang,Hongyu Zhang,Chunsheng Xiao,Xuesi Chen

To target a response to a high oxidative stress environment of inflammatory or tumor sites, various reactive oxygen species (ROS) sensitive polymers have been developed as drug delivery systems. In this study, a novel oxidation sensitive copolymer, phenylboronic acid pinacol ester-functionalized methoxyl poly(ethylene glycol)-block-poly(phthalic anhydride-alter-glycidyl propargyl ether) (mPEG-b-P(PA-alt-GPBAe)), was designed and synthesized by ring-opening alternating copolymerization (ROAP) and click reaction. The copolymers could self-assemble into micelles in aqueous solution with an average size of 20.3 ± 9.3 nm, and are able to load hydrophobic anticancer drug (doxorubicin, DOX) with a high encapsulation efficiency of 75.2%. Interestingly, the encapsulated drug showed accelerated release in the trigger of H2O2, or at low pH values. The copolymers have low cytotoxicity indicated by the 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay towards 4T1 cells, which showed cell viabilities of more than 80% with treatment of our copolymers at concentrations up to 0.5 mg mL−1. The effective uptake of the drug-loaded micelles by 4T1 cells was investigated by confocal laser scanning microscopy (CLSM) and flow cytometry (FCM) analysis. Finally, compared with free DOX, the DOX-loaded nanoparticles exhibited a better antitumor effect and had lower systemic toxicity in 4T1 tumor-bearing mice. Therefore, this new kind of copolymer acting as a stimuli-responsive nanocarrier should represent a promising therapeutic platform for cancer therapy.
