Excess Molar Volumes and Viscosities of Binary Systems of Butylcyclohexane with n-Alkanes (C7 to C14) at T = 293.15 K to 313.15 K

Hong Liu,Lin Zhu

Experimental values of densities and viscosities have been measured in the binary mixtures of butylcyclohexane (BCH) with n-heptane, n-octane, n-nonane, n-decane, n-dodecane, and n-tetradecane at temperatures T = (293.15 to 313.15) K and pressure p = 0.1 MPa over the entire mole fraction range. The excess molar volumes (VmE) and viscosity deviations (Δη) are calculated, and then the changes of VmE and Δη with the composition of BCH are fitted to the Redlich–Kister equation. The values of densities increase continuously with the increase of mole fraction of BCH while the values of viscosities show different trends. All the densities and viscosities decrease with the rise of temperature. The variations of VmE and Δη are discussed with the change of mole fraction of BCH and temperature. This work is useful to understand the molecular interaction in binary systems of liquid.

Near-infrared II fluorescence imaging of PD-L1 and VEGF for assessing and early prediction of “T+A” therapeutic responsiveness in hepatocellular carcinoma

Jukai Feng,Yuting Zhang,Wenhan Liu,Pu Zhang,Jianbin Xiao,Qiang Feng,Zhiyu Wang,Jinyu Zhang,Yang Zhong,Zhongying Shi,Meiqi Pan,Xiaolong Zhang,Jingfeng Liu,Jianmin Wang

The combination of atezolizumab (T, anti-PD-L1) and bevacizumab (A, anti-VEGF) (“T+A”) are recommended as first line treatment in patients with unresectable hepatocellular carcinoma (HCC). However, there is a lack of reliable methods to accurately screen beneficiary populations and predict therapeutic outcomes for “T+A” treatment. In this study, we developed two antibody-labelled quantum dot (QD)-based nanoprobes with non-overlapping near-infrared II (NIR-II) fluorescence for simultaneous in vivo imaging of VEGF and PD-L1 levels within tumors. These nanoprobes utilized NIR-II QDs (emission: ∼950 nm and 1550 nm) as cores, while coating with 4-Arm-PEG-NH 2 as a linker to accomplish highly efficient conjugation of anti-VEGF or anti-PD-L1 antibodies via amide condensation. By using tumor-bearing mouse model, we confirmed positive correlation between the in vivo fluorescence signal and the corresponding protein expression level of tumors as determined by immunohistochemistry. Importantly, our results demonstrated that high fluorescence signal of VEGF and PD-L1 levels within tumors indicated more immunosuppressive microenvironment and was associated with faster tumor progression. At meanwhile, fluorescent imaging for VEGF and PD-L1 levels before treatment could predict the endpoint of therapeutic response to “T+A” regimen. Indeed, high fluorescence signal suggested a better therapeutic response versus that with low fluorescence. Overall, our findings established an in vivo dual-targets fluorescent molecular imaging technique as a promising clinical decision-making tool that can predict response to “T+A” regimen in a minimally invasive manner prior to treatment initiation, therapy enabling personalized therapy selection for patients with advanced HCC.

Superior Antitumor Efficacy of IFN-α2b-Incorporated Photo-Cross-Linked Hydrogels Combined with T Cell Transfer and Low-Dose Irradiation Against Gastric Cancer

Qin Liu,Dinghu Zhang,Hanqing Qian,Yanhong Chu,Yan Yang,Jie Shao,Qiuping Xu,Baorui Liu

Introduction The exhaustion and poor homing of activated lymphocytes are critical obstacles in adoptive cell immunotherapy for solid tumors. In order to effectively deliver immune cells into tumors, we encapsulated interferon-α2b (IFN-α2b) into macroporous hydrogels as an enhancement factor and utilized low-dose irradiation (LDI) as a tumoral attractor of T cells. Methods Hydroxypropyl cellulose hydrogels were prepared by irradiation techniques, and the cross-sectional microstructure was characterized by scanning electron microscopy. The synergistic antitumor mechanism of combination of IFN-α2b and CIK cells was evaluated by detecting the expression of activation marker CD69 on CIK cell surface and IFN-γ production by CIK cells. The in vivo antitumor activity of IFN-α2b-incorporated hydroxypropyl cellulose hydrogels combined with CIK and radiation was evaluated in an MKN-45 xenografted nude mice model. Results The bioactivity of IFN-α2b was well maintained in ultraviolet-reactive, rapidly cross-linkable hydroxypropyl cellulose hydrogels. In vitro studies demonstrated IFN-α2b-activated T cells, as evidenced by upregulating early activation marker CD69 and secretion inflammatory cytokine IFN-γ. In vivo real-time image showed our hydrogels kept a higher amount of drug delivery at the tumor site for a long time compared with free drug injection. Low-dose irradiation promoted T cell accumulation and infiltration in subcutaneous tumors. Combination of IFN-α2b-loaded hydrogels (Gel-IFN) with T cells and LDI exhibited higher efficacy to eradicate human gastric cancer xenograted tumors with less proliferating cells and more necrotic regions compared with IFN-α2b or T cells alone.Discussion HPC hydrogels kept the activity of IFN-α2b and stably release of IFN-α2b to stimulate T cells for a long time. At the same time, low-dose radiation recruits T cells into tumors. This innovative integration mode of IFN-α2b-loaded hydrogels and radiotherapy offers a potent strategy to improve the therapeutic outcome of T cell therapy.

The consensus Nglyco-X-S/T motif and a previously unknown Nglyco-N-linked glycosylation are necessary for growth and pathogenicity of Phytophthora

Can Zhang,Meng Cai,Shanshan Chen,Fan Zhang,Tongshan Cui,Zhaolin Xue,Weizhen Wang,Borui Zhang,Xili Liu

Summary Asparagine (Asn, N )-linked glycosylation within N glyco -X-S/T; X ≠ P motif is a ubiquitously distributed post-translational modification that participates in diverse cellular processes. In this work, N -glycosylation inhibitor was shown to prevent Phytophthora sojae growth, suggesting that N -glycosylation is necessary for oomycete development. We conducted a glycoproteomic analysis of P. sojae to identify and map N -glycosylated proteins and to quantify differentially expressed glycoproteins associated with mycelia, asexual cyst, and sexual oospore developmental stages. A total of 355 N -glycosylated proteins was found, containing 496 glycosites, potentially involved in glycan degradation, carbon metabolism, glycolysis, or other metabolic pathways. Through PNGase F deglycosylation assays and site-directed mutagenesis of a GPI transamidase protein (GPI16) upregulated in cysts and a heat shock protein 70 (HSP70) upregulated in oospores, we demonstrated that both proteins were N -glycosylated and that the N glyco -N motif is a target site for asparagine – oligosaccharide linkage. Glycosite mutations of Asn 94 N glyco -X-S/T in the GPI16 led to impaired cyst germination and pathogenicity, while mutation of the previously unknown Asn 270 N glyco -N motif in HSP70 led to decreased oospore production. In addition to providing a map of the oomycete N -glycoproteome, this work confirms that P. sojae has evolved multiple N -glycosylation motifs essential for growth.

A zero-background fluorescent aptasensor for ultrasensitive detection of pesticides based on magnetic three-dimensional DNA walker and poly(T) -templated copper nanoparticles

Yunwei Zhao,Ying Wang,Ruimei Yang,Hui Zhang,Yanfang Zhao,Xiangmin Miao,Lihua Lu

A zero-background fluorescent aptasensor for the ultrasensitive detection of pesticides has been successfully constructed, using poly(T) stabilized copper nanoparticles (CuNPs) as a fluorescent signal and three-dimensional (3-D) DNA walker as a signal amplifier. To fabricate the aptasensor, one strand of dsDNA and one strand of ssDNA are immobilized onto magnetic Fe 2 O 3 beads. The dsDNA is hybridized by the target aptamer contained DNA (Apt-1) and the partial complementary DNA which named as WS since it acts as a walking strand (WS) in DNA walker. Upon the specific binding between omethoate with Apt-1, WS becomes a free walking ssDNA strand. With the aid of nicking endonuclease, the 3-D DNA walker can produce a great quantity of poly(T) DNA during the WS walking. When the supernatant is added into ascorbic acid (AA) and Cu 2+ , poly(T) templated CuNPs forms and strong fluorescence generates. The developed aptasensor has a zero-background, and can ultrasensitively detect omethoate in the linear range of 0–200 nM with the detection limit of 0.22 nM. This zero-background aptasensor provides a facile strategy for pesticides determination in the fields of food safety and environmental monitoring.

Volumetric properties and viscosity for the ternary system of (1-pentanol + ethylcyclohexane + methyl myristate) and corresponding binary systems at T = 293.15–323.15 K

Dan Li,Yujia Zheng,Jingying Wang,Yan Pang,Meifang Liu

Densities and viscosities of ternary system (1-pentanol + ethylcyclohexane + methyl myristate) and the corresponding binary systems, (1-pentanol + ethylcyclohexane), (1-pentanol + methyl myristate), and (ethylcyclohexane + methyl myristate) were determined at different temperatures from 293.15 to 323.15 K. The experimental data were further employed to calculate excess molar volume ( V m E ) and viscosity deviation (Δ η ). Over the whole composition range, the V m E values show positive deviations from ideal behavior and the Δ η values are negative. The V m E values for the ternary mixtures were also correlated with the corresponding binary data using Cibulka and Redlich–Kister equations, respectively. The experimental data and results provide useful base data for further thermodynamic studies of alcohol–biodiesel–diesel blends.

A Volumetric and Viscosity Study for the Binary Mixtures of Ammonium-Based Asymmetrical Gemini Ionic Liquids with Alcohols at T = 293.15–333.15 K

Xuzhao Yang,Yun Fang

In this work, the experimental densities and dynamic viscosities of binary mixtures of ammonium-based room-temperature asymmetrical Gemini ionic liquids (GILs) 1-trimethylammonium-3-(pyridinium) propane bisdicyanamide ([N111C3Py][DCA]2) and 1-trimethylammonium-3-(1-methylpiperidinium) propane bisdicyanamide ([N111C3MPi][DCA]2) with methanol (MeOH) or ethanol (EtOH) over the entire concentration range were, respectively, determined experimentally at temperatures of (293.15, 303.15, 313.15, 323.15, and 333.15) K in the atmospheric pressure of 0.1 MPa. The excess molar volumes (VmE), excess dynamic viscosities (Δη), and excess Gibbs energy of activation (ΔG*E) of the mixtures were evaluated according to the experimental ρ and η values of pure [N111C3Py][DCA]2, [N111C3MPi][DCA]2, MeOH, EtOH, and their binary solutions at the investigated temperatures and pressure. The change of VmE and Δη were correlated with the Redlich–Kister-type expression and optimal fitting parameters were evaluated. It was observed that values of VmE and Δη are negative deviations from ideal behaviors for the four binary systems. With the mole fraction of alcohol decreasing, these excess properties for these mixtures exist as more negative deviations from ideal solutions, until minimum excess values are reached. Negative values of VmE and Δη for the four binary solutions indicate stronger interactions between GIL and alcohol molecules compared to the pure components and better packing owing to the differences in size and shape of the investigated molecules. Positive values of ΔG*E can be ascribed to the size effect of mixing components. Meanwhile, the dependence of these excess properties on temperature for all binary systems indicates that the temperature has a great influence on intermolecular interactions in the studied mixtures.

Densities and Viscosities of Ternary System n-Dodecane (1) + Bicyclohexyl (2) + n-Butanol (3) and Corresponding Binaries at T = (293.15 to 333.15) K

Yitong Dai,Wenqi Zhao,Haiyun Sun,Yongsheng Guo,Wenjun Fang

Densities (ρ) and viscosities (η) of ternary system n-dodecane + bicyclohexyl + n-butanol and three corresponding binary systems, n-dodecane + bicyclohexyl, n-dodecane + n-butanol, and bicyclohexyl + n-butanol, were measured at temperature range from 293.15 to 333.15 K in detail. Densities were obtained using a vibrating-tube densimeter. Viscosities were determined using an automatic microviscometer based on the rolling ball principle. The excess molar volumes (VmE) and the viscosity deviations (Δη) of each mixture were calculated with the measured data. The VmE and Δη values of three binary systems were fitted to the Redlich–Kister equation, and those of the ternary system were fitted to Nagata–Tamura, Singh, Clibuka, and Redlich–Kister semiempirical equations. The VmE values are negative for the binary system of n-dodecane + bicyclohexyl with the minimum at about x = 0.5, while those of binary systems n-dodecane + n-butanol and bicyclohexyl + n-butanol are positive over the entire concentration range. For the ternary system, the VmE values are subtotal positive and the Δη are negative over the entire concentration range. The nonideal behaviors of the mixtures are explained by physical interactions and structural effects between mixture components.

Measurement and Correlation of Solubility of Resorcinol Bis(cyclic 2,2-dimethyl-1,3-propanediol phosphate) in Selected Solvents from T = 293.15 to 333.15 K

Dong-Fang Zhao,Li-Sheng Wang,Jian Sun,Chuan-Ming Chen

A phosphorus flame retardant named resorcinol bis(cyclic 2,2-dimethyl-1,3-propanediol phosphate) (RCDMPP) was synthesized and characterized by high-performance liquid chromatography (HPLC), elemental analysis (EA), Fourier transform infrared spectroscopy (FTIR), mass spectra (MS), and nuclear magnetic resonance (NMR). Its thermal stability was evaluated via thermogravimetric analysis (TGA), and the fusion enthalpy and melting point of RCDMPP were detected by differential scanning calorimetry (DSC). The solubilities of RCDMPP in selected solvents at T = 293.15 to 333.15 K and atmospheric pressure were measured via a static-analytic method. The solubility data were correlated with modified Apelblat, Scatchard–Hildebrand, Buchowski-Ksiazczak (λh), nonrandom two liquid (NRTL), Wilson, and universal quasichemical (UNIQUAC) equations. The calculated results obtained from these models showed good agreement with the experimental data, and the NRTL, Wilson, and UNIQUAC models give better correlation results compared with modified Apelblat and λh models. The solubility parameter of RCDMPP was calculated by use of the Scatchard–Hildebrand methodology.

Cerium Oxide-Loaded Exosomes Derived From Regulatory T Cells Ameliorate Inflammatory Bowel Disease by Scavenging Reactive Oxygen Species and Modulating the Inflammatory Response

Simei Yue,Lingjiao Gong,Yulin Tan,Xiaodan Zhang,Fei Liao

Background Abnormal immune homeostasis, which leads to the accumulation of reactive oxygen species (ROS) and an inflammatory response, plays a crucial role in accelerating the progression of inflammatory bowel disease (IBD). The lack of targeted therapeutic strategies significantly hampers the efficacy of clinical treatments for IBD. This study presents cerium oxide nanoparticle-loaded regulatory T cell-derived exosomes (exo@nCeO) as innovative anti-inflammatory and antioxidant agents specifically designed to address the effects of immune dysregulation. Methods In this work, the morphology and antioxidant properties of nano-cerium oxide were characterized using transmission electron microscopy, as well as hydroxyl radical and 1,1-diphenyl-2-picrylhydrazyl radical assays. Tumor necrosis factor-α and dextran sulfate sodium were employed to establish cellular and animal models of IBD. The impact of exo@nCeO on ROS scavenging and anti-inflammatory activity in intestinal epithelial cells was assessed using dihydroethidium and 2,7-dichlorodihydrofluorescein staining, Western blotting, and apoptosis flow cytometry analysis. Hematoxylin and eosin staining, along with immunohistochemistry and immunofluorescence staining, were utilized to evaluate intestinal epithelial inflammation and ROS levels in the IBD mouse model. Results The findings demonstrate that exo@nCeO possesses augmented anti-inflammatory properties and ROS scavenging abilities in intestinal epithelial cells. In murine models of IBD, exo@nCeO effectively maintained the integrity of the intestinal epithelial barrier and impeded the progression of IBD.Conclusion This study introduces a novel therapeutic approach for IBD and underscores a potential strategy for addressing diseases associated with inflammation and oxidative stress.

Icaritin-curcumol activates CD8+ T cells through regulation of gut microbiota and the DNMT1/IGFBP2 axis to suppress the development of prostate cancer

Xu Wenjing,Li Yingqiu,Liu Lumei,Xie Jing,Hu Zongren,Kuang Shida,Fu Xinying,Li Bonan,Sun Tiansong,Zhu Congxu,He Qinghu,Sheng Wen

Background Prostate cancer (PCa) incidence and mortality rates are rising. Our previous research has shown that the combination of icariin (ICA) and curcumol (CUR) induced autophagy and ferroptosis in PCa cells, and altered lipid metabolism. We aimed to further explore the effects of the combination of ICA and CUR on gut microbiota, metabolism, and immunity in PCa. Methods A mouse subcutaneous RM-1 cell tumor model was established. 16 S rRNA sequencing was performed to detect changes in fecal gut microbiota. SCFAs in mouse feces, and the effect of ICA-CUR on T-cell immunity, IGFBP2, and DNMT1 were examined. Fecal microbiota transplantation (FMT) was conducted to explore the mechanism of ICA-CUR. Si-IGFBP2 and si/oe-DNMT1 were transfected into RM-1 and DU145 cells, and the cells were treated with ICA-CUR to investigate the mechanism of ICA-CUR on PCa development. Results After treatment with ICA-CUR, there was a decrease in tumor volume and weight, accompanied by changes in gut microbiota. ICA-CUR affected SCFAs and DNMT1/IGFBP2/EGFR/STAT3/PD-L1 pathway. ICA-CUR increased the positive rates of CD3 + CD8 + IFN-γ, CD3 + CD8 + Ki67 cells, and the levels of IFN-γ and IFN-α in the serum. After FMT (with donors from the ICA-CUR group), tumor volume and weight were decreased. SCFAs promote tumor development and the expression of IGFBP2. In vitro, DNMT1/IGFBP2 promotes cell migration and proliferation. ICA-CUR inhibits the expression of DNMT1/IGFBP2. Conclusions ICA-CUR mediates the interaction between gut microbiota and the DNMT1/IGFBP2 axis to inhibit the progression of PCa by regulating immune response and metabolism, suggesting a potential therapeutic strategy for PCa.
