Preparation of magnetic ZIF-8/reduced graphene oxide composite aerogels for efficient removal of ofloxacin

Zhangxu Chen,Fanli Meng,Qihong Cai,Minglian Fu,Danchen Zhu

To solve the problem of ofloxacin pollution in contaminated water, Fe3O4 and ZIF-8 were prepared by a solvothermal method and stirring at normal temperature, respectively. Furthermore, a series of magnetic ZIF-8/reduced graphene oxide (Fe3O4/ZIF-8/rGO) composite aerogels with different mass ratios were synthesized by reduction and freeze-drying methods. The products were characterized using a Fourier transform infrared spectrometer, an X-ray diffractometer, a field emission scanning electron microscope, and a thermogravimetric analyzer. Taking ofloxacin (OFL) as the target pollutant, an orthogonal test was carried out to explore the optimum conditions for removing ofloxacin with the composite aerogel: under the conditions of an illumination time of 8.0 h, a mass ratio of 10 : 15 : 20 for Fe3O4/ZIF-8/rGO, a composite aerogel dosage of 8 mg, and an initial concentration of ofloxacin of 50 mg L−1, the removal rate of ofloxacin reached 98.87%. After being recycled five times, the removal rate remained at 90.23%. The prepared Fe3O4/ZIF-8/rGO composite aerogel and the developed ofloxacin removal procedure could provide a reference for the treatment of quinolone antibiotics and PPCPs in polluted water. Moreover, they can be expected to alleviate the energy crisis and enhance the application of solar energy to deal with environmental remediation in the future.

ZIF-8-Based Dual-Modal Smart Responsive Nanoplatform for Overcoming Radiotherapy Resistance in Advanced Tumors

Yi Feng,Zijing Li,Luqi Song,Shu Zhang,Bin Chen,Guanglin Wang,Kai Yang,Yan Lu,Ran Zhu

Radiation therapy is one of the core means of tumor treatment, especially in local control and radical treatment has an irreplaceable role. However, radiotherapy resistance is one of the major challenges in current clinical practice. Tumor cells have a strong ability to repair DNA damage, which can effectively resist DNA double-strand breaks caused by X-ray, thus weakening the killing effect of radiotherapy. In addition, the complexity of the tumor microenvironment (TME) will further reduce the sensitivity of radiotherapy, leading to poor treatment results. With the rapid development of nanotechnology, multi-modal combined therapy nanoplatform has gradually become a new strategy to overcome radiotherapy resistance. These nanoplatforms achieve synergies by integrating multiple therapeutic approaches, such as radiation sensitization, photothermal therapy and chemotherapy. In this study, we utilized ZIF-8, a member of the metal-organic frameworks, to simultaneously load ICG and Rapamycin for X-ray sensitization and combined photothermal therapy. In this formulation, Rapamycin enhances tumor cells' sensitivity to radiotherapy by inhibiting the mTOR signaling pathway, increasing DNA damage, regulating the cell cycle, and stimulating the STING pathway, which amplifies the tumor immune response. Meanwhile, ICG, as a photosensitizer, effectively converts light energy into heat, achieving tumor photothermal ablation. The modified drug-delivery system becomes a tumor-microenvironment-responsive smart carrier, increasing tumor cell uptake, prolonging retention at the tumor site, and achieving targeted drug delivery. It releases drugs in the specific tumor microenvironment, enhancing photothermal and radiotherapy sensitization effects. The results show that the smart dual-loaded nanoplatform effectively combines photothermal therapy and external-beam sensitization, reshapes the immunosuppressive tumor microenvironment and significantly inhibits tumor proliferation in the HepG2 subcutaneous xenograft model, demonstrating marked antitumor activity and good biosafety.

Magnetically-drivable, pH-responsive Fe3O4/ZIF-8 coating on implant for biofilm prevention and treatment

Kecheng Quan,Yupu Lu,Ting Zhang,Shuren Wang,Hao Tang,Chunhao Yu,Yongsheng Zhou,Yunsong Liu,Dandan Xia,Yufeng Zheng

Antimicrobial coatings with the capabilities to prevent and treat biofilm infections on implant are promising surface treatment strategies, but it is difficult to eradicate an already-formed biofilm by the coating. Magnetically-drivable coatings can physically destruct the surface-attached biofilm to enhance antimicrobial killing. However, current magnetic coatings cannot inhibit the initial bacteria growth and the demand of coating stability restricts their magnetic controllability. Herein, a magnetically-drivable, pH-responsive coating is developed by simply mixing Fe 3 O 4 and ZIF-8 nanoparticles on titanium surface. In physiological pH, Fe 3 O 4 nanoparticles are stable and non-magnetically drivable on surface due to charge attractions and electrostatic interactions. While in acidic pH caused by bacterial growth, ZIF-8 degradation is significantly accelerated, releasing Zn 2 + and recovering the magnetic controllability of Fe 3 O 4 nanoparticles. The released Zn 2+ kills the initially-attached bacteria, inhibiting subsequent biofilm formation. Once a biofilm has formed, Fe 3 O 4 nanoparticles can be magnetically pulled-off to destruct the biofilm structure and then enhance antibiotic killing. The Fe 3 O 4 /ZIF-8 coating shows excellent biocompatibility and osteogenesis to compare with bare Ti disc. Animal experiments further proved the in-vivo antimicrobial and osteogenic performances of the coating. Therefore, this work put forward a new surface strategy to solve biofilm-related infections on implants.

Highly stable and luminescent ZIF-8@MAPbBr3 enhanced “turn on” fluorescent aptasensor for azlocillin monitoring

Yu Li,Anqi Sun,Yufan Yang,Zhouping Wang,Junsong Yang

As emerging luminescent materials, perovskite quantum dots (PQDs) have attracted considerable attention due to their exceptional fluorescence properties. However, their practical application is hindered by inherent instability and high toxicity. In this study, we developed a “turn-on” fluorescent aptasensor for azlocillin detection by integrating aptamer A58-modified ZIF-8-encapsulated MAPbBr 3 (ZIF-8@MAPbBr 3 @A58) with complementary DNA-functionalized magnetic beads (MBs@cDNA). We first rationally designed a 2-aminobenzimidazole and Mn 2+ co-doped ZIF-8@MAPbBr 3 composite, exhibiting enhanced stability, strong luminescence, low biotoxicity and facile modification. The ZIF-8@MAPbBr 3 was further functionalized with A58 to serve as a highly specific signal probe (ZIF-8@MAPbBr 3 @A58), while MBs@cDNA acted as a magnetic separation probe. In the present of azlocillin, the ZIF-8@MAPbBr 3 @A58 probe displayed high affinity toward the targe, leading to the dissociation of the ZIF-8@MAPbBr 3 @A58-MBs@cDNA complex and resulting in enhanced fluorescence intensity. Subsequently, the complementary region between aptamer and cDNA was optimized guided by the molecular docking. Under optimal conditions, the proposed aptasensor achieved a limit of detection of 0.014 ng/mL, with a linear range of 0.1-100 ng/mL. This work demonstrates that ZIF-8 encapsulation combined with ion doping effectively addresses PQD instability. And the strategy paves the way for developing stable PQD-based aptasensors for trace antibiotic monitoring.

Antifouling Mussel-Inspired Hydrogel with Furanone-Loaded ZIF-8 for Quorum Sensing-Mediated Marine Antifouling

Yanbin Xiong,Junnan Cui,Xiaodan Liu,Haobo Shu,Pan Cao

Marine biofouling, the process of marine microorganisms, algae, and invertebrates attaching to and forming biofilms on ship hulls, underwater infrastructure, and marine equipment in ocean environments, severely impacts shipping and underwater operations by increasing fuel consumption, maintenance costs, and corrosion risks, and by threatening marine ecosystem stability via invasive species transport. This study reports the development of a hydrogel-metal-organic framework (MOF)-quorum sensing inhibitor (QSI) antifouling coating on 304 stainless steel (SS) substrates. Inspired by mussel adhesion, a hydrophilic bionic hydrogel was first constructed via metal ion coordination. The traditional metal ion source was replaced with a zeolitic imidazolate framework-8 (ZIF-8) loaded with 2-(5H)-furanone (HF, a QSI) without altering coating formation. Physicochemical characterization using Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), thermogravimetric analysis (TGA), scanning electron microscopy (SEM), high-resolution transmission electron microscopy (HRTEM), the Brunauer–Emmett–Teller (BET) method, and the diffraction of x-rays (XRD) confirmed successful HF loading into ZIF-8 with intact crystal structures. Antifouling tests showed HF@ZIF-8 enhanced antibacterial inhibition againstStaphylococcus aureus(97.28%) andEscherichia coli(>97%) and suppressedChromobacterium violaceumCV026 pigment synthesis at 0.25 mg/mL (sub-growth concentration). The reconstructed PG/PVP/PEI/HF@ZIF-8 coating achieved 72.47% corrosion inhibition via synergistic anodic protection and physical shielding. This work provides a novel green approach for surface antifouling and drag reduction, highlighting MOF-loaded QSIs as promising additives to enhance the antifouling performance of hydrogel coatings, anti-corrosion performance, and QSI performance for sustainable marine engineering applications.

Hierarchically ordered truncated single crystal mesoporous ZIF-8 as a solid platform for lipase immobilization

Yang Zhou,Zhenzhu xia,Frank Peprah Addai,Jinping Chen,Chengxiang Feng,Zongjian Zhen,Juan Han,Feng Lin,Zhirong Wang,Yun Wang

Most pristine metal organic frameworks (MOFs) are inherently microporous. To introduce desired porosity for easy enzyme infiltration and immobilization, polystyrene spheres was used as sacrificial template to design hierarchical ordered micro-mesoporous truncated single-crystalline ZIF-8 (SOM-ZIF-8). The surface area and pore diameter of the SOM-ZIF-8 were 821 m 2 /g and 7.09 nm which changed to 669 m 2 /g and 4.98 nm respectively, after lipase immobilization, suggesting the pores and surface of the SOM-ZIF-8 served as binding sites for the enzyme, and could reach a loading capacity of 134 mg/g after 24 h. The optimal conditions for achieving maximum lipase activity for free LipaseELP, ZIF-8@LipaseELP and SOM-ZIF-8@LipaseELP were pH 7.8 at 45 °C, and maintained 33.67 %, 45.6 % and 57.78 % residual activity after incubation at 80 °C for 2 h. The specific activity towards tributyrin and p-nitrophenyl acetate were observed to be SOM-ZIF-8@LipaseELP (0.25 and 45.85 U/mg) compared to ZIF-8@LipaseELP (0.211 and 43.62 U/mg) and LipaseELP (0.179 and 41.09 U/mg) respectively. The SOM-ZIF-8@LipaseELP could further be recovered and reused for 9 rounds while maintaining 73.8 % of its original activity. This study demonstrates that introducing mesoporous structures into ZIF-8 could improve its binding enzyme property for enhanced hydrolytic function.

Cur@ZIF-8@BA nanomaterials with pH-responsive and photodynamic therapy properties promotes antimicrobial activity

Shang Xiujuan,Wang Hongdong,Yu Yongbo,Gu Jin,Zeng Jian,Hou Sinan

Antimicrobial photodynamic therapy (aPDT) has emerged as a highly promising strategy for non-antibiotic treatment of infections due to its unique advantages in efficient bactericidal action and reduction of drug resistance. The natural photosensitizing properties of curcumin (Cur) are widely acknowledged; however, its limited bioavailability has impeded its practical application. In this study, we developed a nanomaterial called Cur@ZIF-8@BA by encapsulating Cur within ZIF-8 and modifying the surface with boric acid (BA). The Cur@ZIF-8@BA exhibits pH-responsive properties and enhances bacterial binding, thereby effectively promoting photodynamic therapy. Moreover, its antibacterial activity against E. coli, Staphylococcus aureus and A. baumannii is significantly increased in the presence of light compared to a dark environment. The mechanism behind this may be that BA increases the affinity of Cur@ZIF-8@BA towards bacteria, and making released Zn2+ and BA from the nanomaterial increase bacterial cell membrane permeability. This facilitates efficient delivery of Cur into bacterial cells, resulting in generation of abundant reactive oxygen species (ROS) and subsequent bactericidal activity. In conclusion, our prepared Cur@ZIF-8@BA holds great promise as a photodynamically mediated antimicrobial strategy.

Facile Fabrication of a Continuous PVDF/ZIF-8-Lignosulfonate Membrane for Efficient Organic Pollutants Adsorption

Juan Dai,Qixu Chen,Yue Li,Jin Wang,Yanchao Yin,Peiyu Zhao,Peichuang Li,Luying Wang

Graphical A defect-free PVDF/ZIF-8-LS membrane prepared by loading a continuous ZIF-8-LS film layer on the PVDF membrane exhibited high and stable adsorption performance for CR and TC removal, because of a good compatibility between PVDF membrane and ZIF-8-LS layer. Lignosulfonate (LS) enhances ZIF-8 dispersion and active sites but faces challenges in recovery and secondary pollution due to its powdered form. To address this, a polyvinylidene fluoride (PVDF)/ZIF-8-LS composite membrane was fabricated via vapor deposition, where LS promoted dense ZIF-8 growth and interfacial compatibility with PVDF. The resulting PVDF/ZIF-8-LS membrane shows a higher continuous, uniform and dense ZIF-8-LS layer without macroscopic cracks or obvious defects. In addition, the PVDF/ZIF-8-LS exhibits the best adsorption capacity of 110.6 mg g −1 for Congo Red, about 8.8 mg g −1 higher than the PVDF/ZIF-8, and 110.2 mg g −1 for tetracycline, respectively. With 92% capacity retention after three cycles and stability across pH 2–10. The more active adsorption sites facilitates the contact between the ZIF-8-LS layer and the adsorbed substance, in addition to the electrostatic, H-bonding, π-π interactions, and coordination bond are also conductive to the adsorption of PVDF/ZIF-8-LS. The excellent adsorption performance and good recyclability of the PVDF/ZIF-8-LS composite membranes exhibit a potential for adsorbing other contaminants from water.

Bimetallic engineering in Ag/Zn-ZIF-8/Pebax mixed matrix membranes for enhanced CO2 separation

Qian-Qian Li,Yang Li,Heng Mao,Yan-Mei Zhang,Qiu-Ying Zhang,Xin-Ru Chen,Zi-Cong Shan,Xian-Zhe Zhou,Li-Hao Xu,Zhi-Ping Zhao

Mixed matrix membranes (MMMs) have emerged as a critical platform for molecular separation, especially carbon capture from point-emission sources. In this study, a series of novel MMMs were fabricated by incorporating the bimetallic Ag/Zn-ZIF-8 nanoparticles into the poly (ether-block-amide) (Pebax) matrix for efficient CO 2 /N 2 separation. The SEM, EDS, XRD, FTIR, XPS, TGA, N 2 adsorption-desorption measurements were employed to elucidate the microstructures and physicochemical properties of the resultant nanoparticles and membranes. The bimetallic coordination-induced optimization modulated the inherent structure of ZIF-8 framework and enhanced CO 2 affinity, thereby forming the CO 2 -selective transport channels over N 2 . The incorporation of Ag + promoted the uniform dispersion of Ag/Zn-ZIF-8 nanoparticles in Pebax matrix, indicating the excellent polymer-filler interfacial compatibility. Impressively, the resulting membrane achieved the superior separation performance with a CO 2 permeability of 359 Barrer and a CO 2 /N 2 selectivity of 53, which were 2.5 and 1.4 times higher than that of the pristine Pebax membrane. Besides, the introduction of Ag/Zn-ZIF-8 nanoparticles enhanced the thermal and mechanical stability of the membrane, ensuring the potential for long-term operation. These findings herein advance the rational design and preparation of high-performance MMMs for sustainable carbon capture.

Humidity-Resistant Def-ZIF-8/PPY/BC@PDMS Composite for Dual NO2 and Strain Detection

Mingyin Qiu,Ying Men,Zhe Lin,Lipiao Bao,Ziyu Qin,Xing Lu

The growing demand for multifunctional sensors in wearable systems has prompted the need for materials that simultaneously support gas sensing and mechanical strain detection. However, current flexible sensing materials still suffer from issues such as response hysteresis, poor humidity resistance, and lack of antibacterial properties. Herein, we developed a ternary flexible composite by integrating defect-engineered ZIF-8, conductive polypyrrole (PPY), and bacterial cellulose (BC), which was further encapsulated with polydimethylsiloxane (PDMS) to form the Def-ZIF-8/PPY/BC@PDMS composite. The results demonstrated that with 2–3 layers of PDMS, the sensor achieved a fast and stable NO 2 response (9.3% at 10 ppm), with reliable recovery. Under 80% relative humidity, PDMS reduced humidity-induced signals from 34% to 10.3%, confirming markedly improved moisture resistance. The incorporation of a PDMS layer enabled enhanced stress sensing, reducing the response/recovery times to 0.4/0.3 s from 0.9/0.8 s in the uncoated structure. The sensor demonstrated high fidelity in recognizing complex biomechanical signals such as swallowing, walking, and finger articulation. Furthermore, antibacterial testing confirmed 42.18% and 64.69% inhibition against Escherichia coli and Staphylococcus aureus , respectively. This multifunctional composite simultaneously optimizes gas sensing, mechanical flexibility, humidity resistance, and antibacterial efficacy, paving the way for advanced materials in fully integrated wearable sensing technologies.

Mn Oxide Nanowire/ZIF-8 Composites with Multiple Enzyme-like Activities for Enantioselective Glutamate Sensing

Guo-Ying Chen,Mao-Ling Luo,Jing-Jing Dai,Feng-Qing Yang

A composite material of Mn oxide nanowires and ZIF-8 (MnxOyNWs@ZIF-8-RD) with controllable sizes and morphologies (rhombic dodecahedron-shape) was successfully synthesized under mild reaction conditions. The systematic investigation into the effects of synthesis conditions of the material on their microstructure and crystalline morphology was conducted. The material function as “tandem enzymes”, exhibiting multiple enzyme-like activities, such as peroxidase (POD)- and glutamate-oxidase (Glu OXD)-like activities. Kinetic studies reveal that the MnxOyNWs@ZIF-8-RD has excellent enzyme-like catalytic activity, with high substrate affinity and a maximum reaction rate of (H2O2: 840.52 × 10−8M·S−1). MnxOyNWs@ZIF-8-RD shows remarkable enantioselectivity for Glu enantiomers based on its POD- and Glu OXD-like activities. By integrating theoretical and experimental approaches, the recognition mechanism was preliminarily elucidated. In short, this study offered valuable insights for developing sophisticated functional materials and provided methodological references for Glu enantiomer recognition and quantitative detection.

A zeolite imidazolate (ZIF-8)-based system for synergistic photodynamic/starvation therapy of breast cancer

Li Fenglan,Zhang Yuanyuan,Ye Chenliu,Zhuang Junyang

The treatment of breast cancer (BC) faces multiple challenges, including high recurrence rates, drug resistance, and toxic side effects. This study constructed a cell membrane-mimicking biomimetic cascade nanoreactor, mCGH@ZIF, which is loaded with the photosensitizer chlorin e6 (Ce6), glucose oxidase (GOx), and hemin, enabling synergistic starvation-photodynamic therapy. Within the tumor microenvironment, GOx depleted glucose and generated H₂O₂; hemin converted H₂O₂ into O₂ in situ, effectively alleviated hypoxia and amplified the generation of Ce6-mediated singlet oxygen; and the cancer cell membrane coating enabled both immune evasion and homologous targeting. In vitro experiments demonstrated that mCGH@ZIF significantly suppressed 4T1 breast cancer cell proliferation while preserving excellent biocompatibility. This strategy established a closed-loop system integrating glucose depletion, oxygen production, and reactive oxygen species (ROS) generation, overcame the limitations of monotherapies, and offered a novel approach for precision combination therapy in breast cancer.
