NIR-triggered Tandem Photothermal and Photochemical Cancer Therapy Based on Nanoscale MOFs under Hypoxic Conditions

Miao He,Jian Yang,Ke Li,Qinghua Wang,Jinlou Gu

Graphical The nanoscale zeolitic imidazole framework-8 (ZIF-8) is adopted to co-assemble a temperature-sensitive free radical initiator (AIPH) and a photothermal molecule (ICG) to realize the NIR-triggered tandem photothermal and oxygen-independent photodynamic therapy. This as-prepared ICG/AIPH@ZIF-8 nanoplatform displays an excellent killing effect of cancer cells in both normoxia and hypoxia. Due to the limited penetration depth of visible light and insufficient oxygen supply in hypoxic tumor tissues, the development of near-infrared (NIR) triggered and oxygen-independent photodynamic therapy (PDT) to generate active radicals is necessary for cancer treatment. Herein, we developed a NIR-triggered photothermal therapy (PTT) and oxygen-independent PDT system based on metal-organic frameworks (MOFs), which integrated indocyanine green (ICG) and 2,2’-azobis[2-(2-imidazolin-2-yl)propane] dihydrochloride (AIPH) as NIR-triggered photothermal units and temperature-sensitive free radical initiator into ZIF-8, respectively. AIPH was introduced into ZIF-8 via a one-pot strategy with high loading capacity (125 mg g −1 ). Through the irradiation of 808 nm laser, the ICG units could efficiently absorb NIR light to rapidly raise the local temperature to 44 °C within 3 minutes. Meanwhile, the local high temperature further induced the AIPH to continuously generate toxic alkyl radicals (R−C⋅). The combination of PTT and free radical therapy is significantly better than the single treatment mode. More importantly, the NIR-triggered synergistic therapy could induce extensive apoptosis of cancer cells in hypoxic conditions, which sheds new light on the development of MOFs-based photochemical therapy platform in the actual tumor microenvironment.

A novel macrophage-mediated biomimetic delivery system with NIR-triggered release for prostate cancer therapy

Qiang Lei,Cai Zheng,Jiang Wenjun,Liu Jiyong,Tai Zongguang,Li Guorui,Gong Chunai,Gao Shen,Gao Yuan

Background Macrophages with tumor-tropic migratory properties can serve as a cellular carrier to enhance the efficacy of anti neoplastic agents. However, limited drug loading (DL) and insufficient drug release at the tumor site remain the main obstacles in developing macrophage-based delivery systems. In this study, we constructed a biomimetic delivery system (BDS) by loading doxorubicin (DOX)-loaded reduced graphene oxide (rGO) into a mouse macrophage-like cell line (RAW264.7), hoping that the newly constructed BDS could perfectly combine the tumor-tropic ability of macrophages and the photothermal property of rGO. Results At the same DOX concentration, the macrophages could absorb more DOX/PEG-BPEI-rGO than free DOX. The tumor-tropic capacity of RAW264.7 cells towards RM-1 mouse prostate cancer cells did not undergo significant change after drug loading in vitro and in vivo. PEG-BPEI-rGO encapsulated in the macrophages could effectively convert the absorbed near-infrared light into heat energy, causing rapid release of DOX. The BDS showed excellent anti-tumor efficacy in vivo. Conclusions The BDS that we developed in this study had the following characteristic features: active targeting of tumor cells, stimuli-release triggered by near-infrared laser (NIR), and effective combination of chemotherapy and photothermotherapy. Using the photothermal effect produced by PEG-BPEI-rGO and DOX released from the macrophages upon NIR irradiation, MAs-DOX/PEG-BPEI-rGO exhibited a significant inhibitory effect on tumor growth.

Tetraphenylporphine-Modified Polymeric Nanoparticles Containing NIR Photosensitizer for Mitochondria-Targeting and Imaging-Guided Photodynamic Therapy

Pan Yuan,Zheng Ruan,Lifeng Yan

Near-infrared (NIR) photodynamic therapy (PDT) is a promising antitumor strategy under NIR light irradiation to kill cancer cells. Mitochondria has a critical function in sustaining cellular viability and death, which is the ideal organelle for PDT. Here, we reported a tetraphenylporphine (TPP)-conjugated amphiphilic copolymer and an iodinated boron dipyrromethene photosensitizer (BDPI) with high singlet oxygen yield to form nanoparticles (PBDPI-TPP), which could realize mitochondria-targeting and improve the NIR imaging-guided PDT. The as-prepared mitochondria-targeting nanoplatform could show effective subcellular localization and bring about significant irreversible mitochondrial injury for enhanced PDT. Both in vitro and in vivo experiments revealed that the mitochondria-targeting PDT system could achieve a remarkable therapeutic effect, indicating that it is a promising nanoplatform for NIR imaging-guided PDT in cancer therapeutics.

Full-spectrum responsive ZrO2-based phototheranostic agent for NIR-II photoacoustic imaging-guided cancer phototherapy

Chengyuan Zhu,Zhaoyang Ding,Zhengxi Guo,Xiaolu Guo,Aijia Yang,Zhilang Li,Bang-Ping Jiang,Xing-Can Shen

Second near-infrared (NIR-II) window responsive phototheranostic agents have a precise spatiotemporal potential for the diagnosis and treatment of cancer. In this study, a full-spectrum responsive ZrO2-based phototheranostic agent was found to achieve NIR-II photoacoustic (PA) imaging-guided tumour-targeting phototherapy. Initially, the ZrO2-based phototheranostic agent was fabricated through NaBH4 reduction to form boron-doped oxygen-deficient zirconia (ZrO2−x-B), an amino-functionalised SiO2 shell and a further covalent connection with hyaluronic acid (HA), namely, ZrO2−x-B@SiO2-HA. In the ZrO2−x-B@SiO2-HA system, the oxygen vacancy and boron doping resulted in full-spectrum absorption, enabling a high NIR-II photothermal conversion, high-resolution PA imaging ability and a remarkable production of reactive oxygen species (ROS). The surface modification of HA provided ZrO2−x-B@SiO2-HA with water dispersibility and a targeting capability for CD44 overexpressed cancer cells. Furthermore, in vitro and in vivo experiments showed that NIR-II activated ZrO2−x-B@SiO2-HA had a targeted photothermal/photodynamic effect for cancer elimination under the real-time guidance of NIR-II PAs. Hence, ZrO2−x-B@SiO2-HA displays a precise NIR-II radiation-activated phototheranostic potential for PA imaging-guided cancer-targeting photothermal/photodynamic therapy.

Injectable, NIR/pH-Responsive Nanocomposite Hydrogel as Long-Acting Implant for Chemophotothermal Synergistic Cancer Therapy

Xiaoyu Xu,Ziyuan Huang,Zeqian Huang,Xuefei Zhang,Siyu He,Xiaoqi Sun,Yifeng Shen,Mina Yan,Chunshun Zhao

In this study, gold nanorods (GNRs) were incorporated into the hydrogel networks formed by the copolymerization of N-isopropylacrylamide (NIPAm) and methacrylated poly-β-cyclodextrin (MPCD)-based macromer to fabricate an injectable and near-infrared (NIR)/pH-responsive poly(NIPAm-co-MPCD)/GNRs nanocomposite hydrogel, which could serve as a long-acting implant for chemophotothermal synergistic cancer therapy. The nanocomposite hydrogel showed superior mechanical and swelling properties, gelation characteristics, and excellent NIR-responsive property. A hydrophobic acid-labile adamantane-modified doxorubicin (AD-DOX) prodrug was loaded into the hydrogel efficiently by host–guest interaction. The nanocomposite hydrogel exhibited a manner of sustained drug release and could sustain the slow and steady release of DOX for more than 1 month. The pH-responsive release of DOX from the nanocomposite hydrogel was observed owing to the cleavage of acid-labile hydrazone bond between DOX and the adamantyl group in acidic environment. NIR irradiation could accelerate the release of DOX from the networks, which was controlled by the collapse of the hydrogel networks induced by photothermal effect of GNRs. The in vitro cytotoxicity test demonstrated the excellent biocompatibility and photothermal effect of the nanocomposite hydrogel. Moreover, the in situ-forming hydrogel showed promising tissue biocompatibility in the mouse model study. The in vivo antitumor test demonstrated the capacity of the nanocomposite hydrogel for chemophotothermal synergistic therapy with reduced adverse effects owing to the prolonged drug retention in the tumor region and efficient photothermal effect. Therefore, this injectable and NIR/pH-responsive nanocomposite hydrogel exhibited great potential as a long term drug delivery platform for chemophotothermal synergistic cancer therapy.

Ruojing Qian,Zhaoyu Xu,Xingyu Hu,Suru Liu,Yao Mai,Xinzhi Tan,Xiaofan Su,Mingyan Jiang,Wei Tang,Weidong Tian,Li Xie

Purpose A synergistic antibacterial system employing photocatalytic performance and low-temperature photothermal effect (LT-PTT) with the potential for infectious skin wound healing promotion was developed. Methods Ag/Ag2O was synthesized with a two-step method, and its physicochemical properties were characterized. After its photocatalytic performance and photothermal effect were evaluated under 0.5 W/cm2 808 nm NIR laser irradiation, its antibacterial activities in both planktonic and biofilm forms were then studied in vitro targeting Staphylococcus Aureus (S. aureus), and the biocompatibility was tested with L-929 cell lines afterward. Finally, the animal model of dorsal skin wound infection was established on Sprague-Dawley rats and was used to assess infectious wound healing promotion of Ag/Ag2O in vivo. Results Ag/Ag2O showed boosted photocatalytic performance and local temperature accumulation compared with Ag2O when exposed to 0.5 W/cm2 808 nm NIR irradiation, which therefore endowed Ag/Ag2O with the ability to kill pathogens rapidly and cleavage bacterial biofilm in vitro. Furthermore, after treatment with Ag/Ag2O and 0.5 W/cm2 808 nm NIR irradiation, infectious wounds of rats realized skin tissue regeneration from a histochemical level. Conclusion By exhibiting excellent NIR-triggered photocatalytic sterilization ability enhanced by low-temperature photothermal effect, Ag/Ag2O was promising to be a novel, photo-responsive antibacterial agent.

A continuous stimuli-responsive system for NIR-II fluorescence/photoacoustic imaging guided photothermal/gas synergistic therapy

Ziliang Zheng,Qi Chen,Rong Dai,Zhuo Jia,Chenhua Yang,Xiaoyang Peng,Ruiping Zhang

Nanosystems responsive to a tumor microenvironment (TME) have recently attracted great attention due to their potential in precision cancer theranostics. However, theranostic nanosystems with a TME-activated consecutive cascade for the accurate diagnosis and treatment of cancer have rarely been exploited. Herein, an activatable theranostic nanosystem (Bi2S3-Ag2S-DATS@BSA-N3 NYs) is designed and constructed on the basis of a one-pot biomineralization method and surface functional modification to improve second near-infrared (NIR-II) fluorescence/photoacoustic (PA) imaging-guided photothermal therapy (PTT)/gas therapy (GT). Based on enhanced penetration and retention (EPR) effect-mediated tumor accumulation, the tumor-overexpressed glutathione (GSH) can accelerate hydrogen sulfide (H2S) generation from the nanoparticles by reacting with the encapsulated diallyl trisulfide (DATS). Meanwhile, the in situ released H2S can be used not only for gas therapy, but also to start the reduction of –N3(−) to –NH2(+), thereby enhancing the tumor-specific aggregation of NYs. As a result, the activatable nanosystems with excellent tumor accumulation and biodistribution could achieve an accurate NIR-II/PA dual-modality imaging for guiding the synergistic anticancer efficacy (PTT/GT). Thus, this work provides a promising TME-mediated continuously responsive strategy for efficient anticancer therapy.

Composite Nanomaterials of Conjugated Polymers and Upconversion Nanoparticles for NIR-Triggered Photodynamic/Photothermal Synergistic Cancer Therapy

Xueyuan Guo,Ling Li,Wenhua Jia,Chen Zhang,Wei Ren,Chenghui Liu,Yanli Tang

Phototherapies such as photodynamic therapy (PDT) and photothermal therapy (PTT) have attracted great attention in the field of cancer treatment. However, the individual PDT or PTT makes it difficult to achieve optimal antitumor effects compared to the PDT/PTT combined therapy. Also, the effect of PDT is usually limited by the penetration depth of the UV–vis light source. Herein, we designed and synthesized novel composite nanoparticles UCNPs-CPs, which are constructed from two conjugated polymers and upconversion nanoparticles β-NaYF4:Yb,Tm (UCNPs) via a coordination reaction. By virtue of the excellent spectral overlap between absorption of conjugated polymers and emission of UCNPs, the UCNPs can absorb NIR light and effectively excite conjugated polymers by energy transfer to produce massive reactive oxygen species under 980 nm excitation and heat energy under 808 nm laser irradiation, achieving photodynamic/photothermal synergistic therapy. The in vitro cellular investigation proves that the dual modal phototherapy exhibits enhanced antitumor ability compared to single PDT or PTT. Furthermore, UCNPs-CPs inhibit tumor growth 100% in a 4T1 breast tumor mice model with both NIR laser irradiation, indicating that UCNPs-CPs is an excellent platform for synergistic PDT/PTT treatment. Thus, this study provides a promising strategy for NIR-triggered dual modal phototherapy.

Synthesis and characterization of novel yellow-green Al-doped Y3Fe5O12 nano-pigments with high NIR reflectance

Wenwu Zhou,Jianyong Ye,Sheng Zhuo,Danxia Yu,Ping Fang,Rong Peng,Yue Liu,Weifan Chen

The pigments with high near-infrared (NIR) reflectance hold great promise toward both fundamental types of researches and applications for the realization of comfortable indoor-temperature by mitigating urban heat island effect. However, the facile yet simple method for the NIR reflective pigments with bright color and non-toxic feature remains a huge challenge, limiting their practical application. Herein, we proposed a mild and universal method of solution combustion for constructing the non-toxic and NIR reflective yellow-green pigments of Al-doped Y 3 Fe 5 O 12 . The Y 3 Fe 4 AlO 12 pigment presented a high NIR reflectance of 87.66%, and an outstanding acid/alkali resistance. Moreover, the pigment had an impressive color performance (L* = 78.72, a* = − 4.05, b* = 40.04). Besides, the practical near-infrared reflection performance revealed that the transient surface temperature of the Y 3 Fe 4 AlO 12 pigment was 7.6 °C lower than that of the pigment with similar color on the market. This research supplied high NIR reflecting pigments of Y 3 Fe 5−x Al x O 12 as promising coatings for roofs and exterior walls of buildings, acting as a favorable solution for urban heat island effect.

Sb doping facilitates particle refinement and improves transparent NIR insulation of CsxWO3 for energy-saving coating

Qianyi Li,Yuxuan Zheng,Jingyao Zhang,Zhiyong Xiong,Haibo Jin,Yuefeng Su,Caihong Feng,Ning Li,Chengzhi Wang,Jingbo Li

Cesium tungsten bronze (Cs x WO 3 ) is of significant interest due to its unique spectral selectivity, which is utilized in transparent thermal insulation coatings. However, the low-cost batch preparation of Cs x WO 3 nanopowders remains a challenge. In this study, we performed an Sb doping modification of Cs x WO 3 . It was observed that Sb doping makes Cs x WO 3 more prone to breakage, facilitating the preparation of nanoparticle Cs x WO 3 through a simple solid-state reaction followed by mechanical milling. A 7 % Sb doping results in a small average particle size of 108 nm using moderate ball milling. Meanwhile, appropriate Sb doping enhances the transparent thermal insulation properties of Cs x WO 3 . A 3 % Sb doping raises the integrated visible light transmittance ( T lum , 380–780 nm) of Cs x WO 3 from 61.7 % to 65.40 %, and improves the near-infrared shielding efficiency ( Ψ NIR , 780–2500 nm) from 61.20 % to 65.92 %, increasing by approximately 4 % and 5 %, respectively. The reasons for these property improvements are discussed based on experiments concerning bandgap ( E g ) and oxygen vacancy concentrations induced by Sb doping. This study indicates that Sb doping of 1–3 at% can produce Cs x WO 3 nanopowders with better performance in a low-cost method, which is of great significance for advancing the production and practical application of nano-sized tungsten bronze powder in energy-saving coatings.

In situ fabrication of NIR-II responsive TiO2 bio-metasurface for photothermal antibacterial and enhanced osseointegration

Fan Xiao,Chen-xiao Huang,Jun-hao Dai,Jun-hui Ye,Kang-jie Cheng,Xu Xu,Li-quan Deng,Jia You,Wei Wang,Wan-bing Jin

Titanium implants, widely used in orthopedic and dental applications, often face challenges due to inadequate osseointegration and the risk of postoperative infections , which can potentially lead to implant failure. To address this issue, we developed a titanium implant with an in-situ fabricated anatase TiO 2 metasurface , characterized by a subwavelength thickness and a preferential orientation of the (001) facets. This metasurface, which consists of uniformly distributed nano-crystallites of approximately 30 nm in size, aims to enhance both osseointegration and antibacterial properties. The addition of isopropanol (IPA) to the precursor solution used in the fabrication process played a crucial role in controlling the grain size and coating thickness of the metasurface. This process led to a redshift of the light absorption peak into the near-infrared (NIR)-II region, thereby increasing the penetration depth of photothermal therapy . The implants with TiO 2 metasurface exhibited increased hydrophilicity, rapid apatite formation in simulated body fluid, and an effective photothermal response under NIR-II light, showing significant antibacterial activity against E. coli and S. aureus . In vivo experiments in rabbits indicated superior bone regeneration and osseointegration around the implants with TiO 2 metasurface compared to the control titanium implants. At 4 weeks postoperatively, the bone volume to tissue volume (BV/TV) ratio within the implant was 53.94 %, and by 8 weeks, the implants were nearly entirely filled with new bone, with a BV/TV ratio of approximately 70 %. The findings suggest that the TiO 2 metasurface, which combines antibacterial and bioactive features, has the potential to reduce the risk of postoperative infections and improve clinical outcomes.

Acceptor-engineering tailored type-I photosensitizer with aggregation-induced NIR-II emission for cancer multimodal phototheranostics

Li Jiangao,Niu Niu,Wang Deliang,Liu Xue,Qin Yi,Wang Lei,Tang Ben Zhong,Wang Dong

Exploration of single molecular species synchronously featured by long excitation/emission wavelength, accurate diagnosis, and effective therapy, remains supremely appealing to implement high-performance cancer phototheranostics. However, those previously established phototheranostic agents are undiversified and stereotyped in terms of structural skeleton, and generally exhibit insufficient phototheranostic outcomes. Herein, we innovatively utilized indanone-condensed thiadiazolo[3,4-g]quinoxaline (ITQ) as electron acceptor to construct novel photosensitizer with second near-infrared (NIR-II) emission. Experimental study and theoretical calculation demonstrated that comparing with the counterparts constituting by widely employed NIR-II building block benzobisthiadiazole (BBTD) and 6,7-diphenylthiadiazoloquinoxaline (DPTQ), ITQ-based photosensitizer (TITQ) showed superior aggregation-induced emission (AIE) characteristics, much stronger type-I reactive oxygen species (ROS) production, and prominent photothermal conversion capacity. Furthermore, TITQ nanoparticles with excellent biocompatibility were capable of effectively accumulating in the tumor site and visualizing tumor through fluorescence-photoacoustic-photothermal trimodal imaging with highly spatiotemporal resolution, and completely eliminating tumor by type-I photodynamic-photothermal therapy.
