Acidity-responsive polyphenol-coordinated nanovaccine for improving tumor immunotherapy via bidirectional reshape of immunosuppressive microenvironment and controllable release of antigen

Huimin Qiu,Shuman Wang,Rimei Huang,Xingyu Liu,Liqun Li,Zheng Liu,Aihui Wang,Shi-Chen Ji,Hong Liang,Bang-Ping Jiang,Xing-Can Shen

The tumor immunosuppressive microenvironment (TIME) and uncontrollable release of antigen can cause poor efficacy of nanovaccine-based immunotherapy (NBI). It is necessary to develop new strategy for TIME reshape and controllable release of antigen to improve the NBI efficacy. Herein, an acidity-responsive Schiff-base conjugated polyphenol-coordinated nanovaccine was constructed to realize bidirectional TIME reshape and controllable release of antigen for activating T cells for the first time. Specially, the acidity-responsive tannic acid-ovalbumin (TA-OVA) nanoconjugate was prepared via Schiff-base reaction. FeⅢ was coordinated with TA-OVA to produce FeⅢ-TA-OVA nanosystem, and 1-methyltryptophan (1-MT) as indoleamine 2,3-dioxygenase inhibitor was loaded to form polyphenol-coordinated nanovaccine. The coordination between FeⅢ and TA could cause the photothermal ablation of primary tumor, and the acidity-triggered Schiff-base dissociation of TA-OVA could controllably release OVA to realize lysosome escape, initiating the body’s immune response. More importantly, oxidative stress generated by tumor-specific Fenton reaction of Fe ion could promote the polarization of tumor-associated macrophages from M2 to M1 phenotype, resulting in the upregulation of cytotoxic T cells and helper T cells. Meanwhile, 1-MT could downregulate immunosuppressive regulatory T cells. Overall, such skillful combination of bidirectional TIME reshape and controllable antigen release into one coordination nanosystem could effectively enhance NBI of tumors.

A self-assembled FeNPs/metal polyphenol structured superhydrophilic PVDF membrane with capability to activate PMS for efficient oil-water separation

Yinan Chen,Qi Peng,Chengwen Song,Yuanlu Xu,Zaijin You,Xinfei Fan

Despite the membrane technology as an efficient method for treating emulsified oily wastewater, the issue of membrane fouling remains an enormous challenge during extended operation. Hence, a unique self-assembled method was proposed to clean membrane fouling for oil/water (O/W) separation, where the tannic acid/iron (TA/Fe) coating layer improved hydrophilicity and doped with iron nanoparticles to activate peroxymonosulfate (PMS) for removing of the irreversible oil fouling. The TA/Fe coating, which contained phenolic hydroxyl groups, rendered the modified membrane superhydrophilic and underwater oleophobic properties to resist membrane surface oil droplets. The iron nanoparticles/TA/polyvinylidene fluoride (FeNPs/TA/PVDF) achieved notable permeation flux of 1571.13 L m −2 h −1 and 1490 L m −2 h −1 in O/W mixture and oil-in-water emulsion (liquid paraffin as oil phase), respectively. Meanwhile, the membrane exhibited a distinguished oil removal rate, surpassing 98 % for various emulsions (e.g. N-hexane, isooctane, soybean oil). Notably, in the pollution and cyclic experiments, the regeneration water flux of FeNPs/TA/PVDF increased from 71.7 % of pristine PVDF to 91.5 % after final modification, which proved the enhancement of the membrane anti-oil-fouling capability through the deposition of the hydrophilic coating. Moreover, after catalytic cleaning, the water permeation flux of FeNPs/TA/PVDF could reach approximately 90 % of its original value. Obviously, we observed that FeNPs/TA/PVDF exhibited a high regenerative capacity during the PMS-assisted catalytic cleaning process, where the radical and nonradical pathways coexisted. Generally, the research offered a simple approach to creating functional membranes suitable for efficient O/W separation.

Multifunctional Conjugated Polyphenol “Bridge” Capped the Upper Interface of Highly Efficient and Stable Inorganic Perovskite Solar Cells Fabricated in Air

Yifei Shi,Lei Zhang,Shuming Hu,Xu Wang,Jincheng Huang,Xinlong Zhang,Yuanfang Zhang,Jintao He,Kai Liao,Ruoxi Yang,Hengzhi Zuo,Wenqi Wu,Fei Jiang,Jianlin Chen,Zhuoyin Peng

Carbon-based all-inorganic perovskite solar cells (C-PSCs) are promising alternatives compared with conventional organic–inorganic halide PSCs because of their good thermal stability. However, low carrier transport efficiency and energy level mismatch of the upper interface of the device limit the further improvement of power conversion efficiency (PCE), and all-inorganic perovskites are extremely sensitive to oxygen and humidity, which seriously damages the long-term stability of the device. Upper interface modification is an effective solution to improve the PCE and the stability of devices. Unfortunately, many traditional interface modifiers exhibit insulated and hygroscopic characteristics, which may be harmful to the performance of the device. Herein, a multifunctional conjugated polyphenol “bridge” strategy was adopted on the upper interface of the device fabricated in air, that is, stilbene and polyhydroxy stilbene were capped on perovskite. The results demonstrate that they can effectively enhance the energy level alignment, improve charge transport, passivate defects, suppress nonradiative recombination of interface, and resist oxygen and moisture in air, and the positive effects of modification become more and more pronounced with the increment of the number of hydroxyl groups. The PCE of PSCs was improved from 11.10 to 13.10% after modification. In addition, the unencapsulated modified devices exhibited better stability than the control devices under different environmental conditions.

Enhancing Mechanical Properties and Oil Resistance of Carbon Black/Hydrogenated Nitrile Butadiene Rubber Composites via Tea Polyphenol-Assisted Interfacial Modification

Hang Yin,Jingkai Nie,Yifan Wang,Shicheng Zhang,Ming Sun,Yi Tian,Chao Fan,Qiang He

Rubber vibration dampers in transformers require high oil resistance to ensure long-term stability in power facilities. Hydrogenated nitrile butadiene rubber (HNBR), characterized by its strong polarity and superior oil resistance, serves as an ideal matrix material. However, its high polarity often leads to poor carbon black (CB) dispersion and weak interfacial interactions with conventional oil-resistant additives, causing additive migration and reduced oil resistance. To address these challenges, tea polyphenol (TP), a polar small-molecule compound, was introduced as a multifunctional modifier and oil-resistant additive to improve the performance of CB/HNBR composites. The study demonstrated that TP acts as an interfacial bridge, where hydrogen bonding between the phenolic hydroxyl groups in TP and the nitrile groups in HNBR, along with π–π interactions between TP and CB, significantly enhances the CB-HNBR interface and promotes uniform CB dispersion. At an optimal TP content of 2 ppm rubber (phr), the CB/HNBR composites exhibited remarkable improvements in mechanical properties, with the 300% modulus, tensile strength, and tear strength increasing by 8.9, 9.4, and 18.5%, respectively, compared to the CB/HNBR composite without TP. Moreover, the formation of a polar network between TP and HNBR further enhanced the oil resistance of the CB/HNBR composites. Oil resistance tests revealed that the volume and mass change rates of the CB/HNBR composite with 2 phr TP decreased by 48.1 and 57.1%, respectively, indicating that TP effectively mitigates additive migration while reinforcing the oil resistance of the composites.

Probiotics functionalized with a gallium-polyphenol network modulate the intratumor microbiota and promote anti-tumor immune responses in pancreatic cancer

Han Zi-Yi,Fu Zhuang-Jiong,Wang Yu-Zhang,Zhang Cheng,Chen Qi-Wen,An Jia-Xin,Zhang Xian-Zheng

The intratumor microbiome imbalance in pancreatic cancer promotes a tolerogenic immune response and triggers immunotherapy resistance. Here we show that Lactobacillus rhamnosus GG probiotics, outfitted with a gallium-polyphenol network (LGG@Ga-poly), bolster immunotherapy in pancreatic cancer by modulating microbiota-immune interactions. Upon oral administration, LGG@Ga-poly targets pancreatic tumors specifically, and selectively eradicates tumor-promoting Proteobacteria and microbiota-derived lipopolysaccharides through a gallium-facilitated disruption of bacterial iron respiration. This elimination of intratumor microbiota impedes the activation of tumoral Toll-like receptors, thus reducing immunosuppressive PD-L1 and interleukin-1β expression by tumor cells, diminishing immunotolerant myeloid populations, and improving the infiltration of cytotoxic T lymphocytes in tumors. Moreover, LGG@Ga-poly hampers pancreatic tumor growth in both preventive and therapeutic contexts, and amplifies the antitumor efficacy of immune checkpoint blockade in preclinical cancer models in female mice. Overall, we offer evidence that thoughtfully designed biomaterials targeting intratumor microbiota can efficaciously augment immunotherapy for the challenging pancreatic cancer.

Protein–polyphenol nanoassemblies modulated construction of zeolitic imidazole framework-67 based nanofiltration membrane for effective separation of dye/salt mixtures

Faizal Soyekwo,Xin Mao,Ruixin Nie,Changkun Liu

Metal–organic frameworks (MOFs) show great potential in various separation applications. However, fast nucleation rate and crystallization of MOFs, produce non-uniform and uncontrollable sized crystals, which are not suitable for the formation of MOF-based membranes. Herein, we propose the concept of utilizing protein polyphenol nanoassemblies (PPNs) as the building blocks for the confined growth of MOFs to fabricate zeolitic imidazolate frameworks (ZIF-67)-based membranes. We first demonstrated that colloidal PPNs as modulators in solution enables the formation of ZIF-67 MOFs with uniform and controllable crystal sizes. Subsequently, we utilized the metal salt coordinated PPNs (Co 2+ @PPN) as the precursor layer matrix deposited on porous polymeric support to fabricate ZIF-67 membranes via in-situ growth strategy at room temperature. The resultant membrane features a robust heterogeneous and well-integrated ZIF-67-reinforced PPN (ZIF-67@PPN) microporous networks. Benefiting from the rigid and multidimensional micropores of ZIF-67, the optimized ZIF-67@PPN1 composite membrane with improved water stability exhibited good water permeance (45.68 L m −2  h −1  bar −1 ), excellent dye desalination performance (Na 2 SO 4 /Congo red selectivity of 1271.47), and good antibiotic desalination performance (NaCl/Erythromycin selectivity of 21.46). The findings in this work provide important engineering concepts for the development of MOF-based membranes using sustainable nature-derived building blocks for emergent environment-relevant desalination applications in the chemical process industry.

Comparative analysis of natural and recombinant CMd from barley malt in the synthesis of gold nanoclusters: The influence of polyphenol binding

Yang Gao,Mingyang Sun,Hanhan Liu,Chenyi Su,Tuo Zhang,Hao Zhang,Chenyan Lv

Protein-templated gold nanoclusters (Au NCs) are promising fluorescent nanomaterials owing to their biocompatibility, stability, and eco-friendly synthesis. CMd, a cysteine-rich subunit of the barley α-amylase/trypsin inhibitor tetramer, is well-suited as a protein template due to its high thiol content. However, native CMd (nCMd) interacts with polyphenolic compounds, potentially altering its structure and function. To enable direct comparison, recombinant CMd (rCMd) was heterologously expressed in E. coli . Spectroscopic analyses (UV–Vis, fluorescence, CD, and FTIR) revealed pronounced structural differences: nCMd displayed extensive polyphenol binding and markedly lower surface hydrophobicity. Both proteins were further employed as templates for gold nanoclusters (Au NCs) synthesis. rCMd-Au NCs exhibited a higher fluorescence quantum yield (∼8.2 %) and enabled ultrasensitive Hg 2+ detection with a detection limit of 2.95 nM (3σ). The rCMd-Au NCs probe also achieved reliable visual detection of Hg 2+ in Chinese herbal medicines, including Astragalus and Codonopsis . Overall, this study demonstrates the impact of polyphenol binding on CMd structural and functional properties and expands the application potential of CMd-based systems in bioanalytical sensing.

A dual-responsive metal-polyphenol nanomedicine based on targeting LAT1 via levodopa and ROS amplification for precise treatment of breast cancer

Xiao He,Zhongmin Wang,Huayang Bai,Yaqin Tang,Daimeng Feng,Shuyu Zhang,Robert J. Lee,Jing Xie,Yansun Sun

Breast cancer remains a major global health challenge, with triple-negative breast cancer (TNBC) posing a particular therapeutic dilemma due to its lack of targetable receptors and reliance on chemotherapy, which is often thwarted by drug resistance. To overcome these limitations, we developed a novel metal-polyphenol nano-platform (DLF@NPs) via a one-pot coordination assembly of L-dopa (L-DA), doxorubicin (DOX), and ferrous ions (Fe 2+ ). This GSH/pH-dual-responsive nanoparticle exploits the overexpression of L-type amino acid transporter 1 (LAT1) on breast cancer cells for targeted delivery and tumor-specific enrichment, effectively addressing the off-target toxicity and resistance associated with free DOX. Upon internalization into the acidic and high-GSH tumor microenvironment, DLF@NPs rapidly disassemble, releasing their payload. The liberated DOX not only exerts its apoptotic effect but also activates NADPH oxidases (NOXs) to elevate intracellular H 2 O 2 levels. This endogenous H 2 O 2 supply fuels a Fenton reaction catalyzed by the co-released Fe 2+ , generating highly toxic hydroxyl radicals. Furthermore, L-DA reduces the resultant Fe 3+ back to Fe 2+ , establishing a catalytic cycle that robustly amplifies reactive oxygen species (ROS). The resultant ROS burst synergizes with DOX by inducing severe mitochondrial damage, leading to potentiated apoptosis. In summary, this work proposes a novel targeted nanoplatform that utilizes the synergy between chemodynamic therapy and chemotherapy to provide a promising strategy for combating drug-resistant breast cancer.

Efficient Removal of Tetracycline from Aqueous Solutions by a Green Biomass Adsorption Material: Cotton Fabric-Supported Fe@ Tannin-Based Polyphenol Network

Mo Jianing,Huang Qianlan,Peng Yu,Peng Zhiyuan

To address the challenges in antibiotic wastewater treatment, this study developed a green and efficient adsorbent based on biomass material. A polyphenol network was formed by the grafting polymerization of hydroxymethyl tannin on the surface of cotton fabric. The cotton fabric-supported Fe@tannin-based polyphenol network (CF@Fe–MPNs) was prepared by the complexation of the polyphenol network with iron ions, and the adsorption performance of CF@Fe–MPNs for tetracycline (TC) was investigated. The adsorption of CF@Fe–MPNs for TC primarily occurred through hydrogen bonding, π–π interactions, and electrostatic interactions, with a maximum adsorption capacity of 146.52 mg/g. The adsorption process followed the Langmuir isothermal model and pseudo-second-order kinetics. Thermodynamic analysis revealed that the adsorption was spontaneous and endothermic. Moreover, CF@Fe–MPNs exhibited excellent reusability over multiple adsorption–desorption cycles, demonstrating their potential as an efficient and sustainable adsorbent for antibiotic removal.

The preparation and physicochemical properties of biodegradable konjac glucomannan-based films reinforced with bacterial cellulose and tea polyphenol for food packaging

Lan Ma,Pengyu Hu,Yihao Zhang,Yunchuan Dai,Yanzhu Guo

Biodegradable and edible packaging films derived from konjac glucomannan (KGM) are gaining popularity as promising candidates to replace petroleum-based plastics with the aim of alleviating environmental problems. However, their poor mechanical strength and lack of antioxidant activity have restricted their practical application in food packaging. Bacterial cellulose (BC), with a three-dimensional nanostructure and biodegradable and biocompatible properties, was chosen as a reinforcing agent to enhance the mechanical properties of KGM film. Film prepared with 15% BC displayed the highest tensile strength of 62.87 MPa and elongation at break of 38.17%, owing to the strong hydrogen bond interactions between the BC and KGM chains. Furthermore, tea polyphenols (TP), a type of natural antioxidant agent, were incorporated into the BC and KGM composite films and endowed films with excellent antioxidant properties, with scavenging rates toward DPPH and ABTS free radicals of up to 96.74% and 97.50%, respectively. Meanwhile, the mechanical strength, UV shielding performance, and water-vapor and oxygen barrier properties of the films were also improved through the incorporation of TP. The film was successfully applied as a packaging material for fresh-cut apple preservation. The prepared film demonstrated nontoxicity and excellent biodegradability over 28 days, with no significant impact on plant growth. Therefore, the prepared composite film exhibited great prospects as a novel type of biodegradable, sustainable and bioactive film for food preservation.

Elucidating the astringency perception mechanism of protocatechuic acid, a key astringent component in black rice, based on polyphenol–salivary protein interaction

Qian Huang,Zijun Liu,Wangyang Shen,Weiping Jin,Guoyan Yang,Wenjing Huang,Jinling Li,Bin Li,Cheng Guo

Black rice is rich in bran layer polyphenols, which enhance bioactivity but may reduce its consumer acceptability due to astringency. Polyphenol–salivary protein aggregation is the molecular basis of astringency. In this study, a saliva-induced precipitation approach with liquid chromatography-tandem mass spectrometry was used to identify protocatechuic acid (PA) as the key astringent component in black rice, accounting for 68% of total precipitated polyphenols. With α-amylase as a representative, the PA–salivary protein interaction was analyzed using fluorescence spectroscopy, isothermal titration calorimetry (ITC), and molecular docking. Fluorescence spectroscopy showed a quenching constant ( K q ) of 7.8451 × 10 11  L⋅mol −1 ⋅s −1 , and ITC revealed a binding constant ( K ) of 2.65 × 10 3  L/mol with 1.32 binding sites, indicating the interaction was characterized by strong, spontaneous, noncovalent binding, static quenching, and multiple binding sites. Atomic force microscopy, contact angle, and small amplitude oscillatory shear rheology revealed that PA increased salivary film surface roughness (contact angle from 36.2° to 43.3°) and weakened viscoelasticity, thereby facilitating film rupture and loss of lubrication underlying astringency perception through tactile stimulus pathway. Molecular dynamics simulations further revealed stable PA binding to human astringency receptor human taste 2 receptor member 16 (hTAS2R16), indicating an additional astringency perception gustatory pathway.
