Characterization of lipid compositions, minor components and antioxidant capacities in macadamia (Macadamia integrifolia) oil from four major areas in China

Xixiang Shuai,Taotao Dai,Mingshun Chen,Cheng-mei Liu,Roger Ruan,Yuhuan Liu,Jun Chen

Macadamia is rich in lipids and considered as a source of high-quality oil. In this study, fatty acids, triacylglycerols , minor components and antioxidant activities of macadamia oils (MOs) from four cultivated regions (YN: Yunnan Province; GZ: Guizhou Province; GX: Guangxi Province; GD: Guangdong Province) in China were systematically analyzed by modern chromatography (Gas chromatography, high performance liquid chromatography , inductively coupled plasma mass spectrometry and ultraviolet spectroscopy). The analysis indicated macadamia oil enriched in unsaturated fatty acids (UFA), including 62.4%–65.6% oleic acid and 11.4%–17.3% palmitoleic acid . Among the four cultivated regions, the MO from YN exhibited the highest contents of UFA (85.0%) and palmitoleic acid. In addition, the minor components and antioxidant activities of MO from four cultivation areas showed a significant difference, which GX-MO had the highest contents of α-tocotrienols (58.9 mg/kg) and phytosterols (2310.4 mg/kg), while GZ-MO was rich in squalene (626.7 mg/kg) and YN-MO had the highest levels of polyphenols (71.0 mg GAE/kg) and antioxidant capacity. Based on the correlation analysis, the polyphenols and α-tocotrienols had a remarkable contribution on the antioxidant activities of MO. Furthermore, the cultivation areas of MO can be distinguished according to the analysis of hierachical cluster analysis and principal components. Our studies offer a potential guidance for the identification of MO and its application in food industry.

Yaling He,Hui Chen,Ziyang Ye,Xinmiao Zhang,Hongling Ye,Ming Ye

To prepare zinc and selenium modified polysaccharides derivatives (ZnLEP-1a and SeLEP-1a), a novel polysaccharide (LEP-1a) isolated from Lachnum YM38 was chosen. Following that, the structural characterization and in vitro bioactivities of LEP-1, ZnLEP-1a and SeLEP-1a were investigated. Results revealed that LEP-1a had a molecular weight of 5.69 × 10 4 Da and was composed of mannose , galactose and glucose. The linkage types of LEP-1a were proved to be (1→)-α- D -Manp, (1→3)-α- D -Manp, (1→2)-α- D -Galp, (1→2)-α- D -Glcp, (1→2,6)-α- D -Manp. Zinc content of ZnLEP-1a was 0.69 mg/g, while selenium content of SeLEP-1a was 206.99 µg/g. Compared with LEP-1a, ZnLEP-1a and SeLEP-1a had a lower molecular weight, same monosaccharides in different molar ratios. The results of FT-IR, DSC , NMR, SEM and AFM suggested that there were obvious changes in structural characteristics after LEP-1a was modified with zinc and selenium. ZnLEP-1a and SeLEP-1a had stronger antioxidant, hypoglycemic, antitumor activities in vitro in comparison with LEP-1a. As a result, ZnLEP-1a and SeLEP-1a showed great potential to develop into a natural antioxidant, hypoglycemic and low-toxic antitumor drug.

Gelatin-based nanofiber membranes loaded with curcumin and borneol as a sustainable wound dressing

Yarong Lv,Zhongxun Yu,Chenxi Li,Jianwei Zhou,Xujin Lv,Jia Chen,Ming Wei,Jun Liu,Xiaomin Yu,Ce Wang,Ping Hu,Yong Liu

Infection is a huge obstacle to wound healing. Thus, to enhance the healing of infected wounds, wound dressings that permit the dual delivery of antimicrobials and antioxidants are highly desirable. In this study, a series of gelatin-based nanofiber membranes with different curcumin contents were fabricated via solution electrospinning. The obtained membranes were characterized in terms of their morphologies, in addition to their physical, mechanical, and in vitro properties. The results showed that the membranes maintained an integrated morphology, excellent water absorption capability, satisfactory mechanical properties, and a high dissolution rate of curcumin. The addition of curcumin and borneol conferred the membranes the ability to inhibit Staphylococcus aureus and eliminate free radicals. Furthermore, cytocompatibility testing using the L929 cell line confirmed the excellent biocompatibility of the membranes. These gelatin-based nanofiber membranes loaded with curcumin and borneol can therefore be considered as promising materials for dressing wounds. Moreover, the use of biodegradable polymers and environmentally sustainable production techniques in this system render it suitable for the commercial manufacture of composite membranes.

Li Dan,Xu Xin-yu,Yang Yang,Wu Na,Ma Zhan-qian,Zuo Feng,Zhang Na

The protein hydrolysate of purple speckled kidney bean (PSKB) was used as the raw material in this study, and the antioxidant peptide of the PSKB protein hydrolysate was purified using macroporous resin. The XAD-7HP macroporous resin was selected as the best purification material, and the static adsorption-desorption of the purified PSKB antioxidant peptide was optimized. The optimum static adsorption and desorption conditions were as follows: the adsorption capacity reached 11.93 ± 0.11 mg/ml at pH 7 for 24 h, and the desorption capacity was 5.24 ± 0.04 mg/ml with 60% ethanol for 30 min. Under this condition, the amount of antioxidant peptide obtained by adsorption-desorption was the highest. The optimum process conditions were as follows: the appropriate flow rate was 1 ml/min, and the optimal injection volume was 40 ml. The adsorption amount at this time can reach 12.19 ± 0.15 mg/ml. The components with an elution time of 10–30 min were separated using the reversed-phase high-performance liquid chromatography (RP-HPLC) technique to obtain three main components, namely, RP1, RP2, and RP3. The DPPH free radical scavenging ability reached 56.26 ± 0.56, 66.42 ± 0.56, and 78.57 ± 0.56%, respectively, which were 36.65, 46.34 ± 0.56, and 54.39 ± 0.56% higher than those before purification. The amino acid sequences of the three components were identified as Phe-Leu-Val-Asp-Arg-Ile, Phe-Leu-Val-Ala-Pro-Asp-Asp, and Lys-Asp-Arg-Val-Ile-Ser-Glu-Leu.

Phenolic features and anthocyanin profiles in winemaking pomace and fresh berries of grapes with different pedigrees

Zhu Lei,Wu Xin,Hu Xixi,Li Xinyue,Lv Shanshan,Zhan Chuan,Chen Yunhua,Wang Changyuan,Xu Jingyu

The total contents and antioxidant activities of phenolic compounds as well as anthocyanin profiles were analyzed and compared in fresh berries and fermented pomace of three grape cultivars with different pedigrees. The phenolic contents and antioxidant activities decreased significantly in skins ( p  < 0.05), while relatively large amounts of them were retained in seeds after fermentative maceration. Fermentative maceration also had a significant impact on the anthocyanin compositions. The proportions of anthocyanins with more stable structures, such as malvidin derivatives, methylated, diglucosides and nonacylated anthocyanins, increased significantly in the pomace skins ( p  < 0.05). There were obvious differences in phenolic features and anthocyanin profiles among the three cultivars. ‘NW196’, a wine hybrid of Vitis vinifera and V. quinquangularis , was characterized by the highest total anthocyanin contents and degree of diglucosylation. The results obtained in this study could contribute to the primary data for the development and utilization of winemaking pomace, especially from local non- Vitis vinifera grapes.

Free, Conjugated, and Bound Phenolics in Peel and Pulp from Four Wampee Varieties: Relationship between Phenolic Composition and Bio-Activities by Multivariate Analysis

Xue Lin,Yousheng Shi,Pan Wen,Xiaoping Hu,Lu Wang

Free, conjugated, and bound phenolic fractions of peel and pulp in four wampee varieties from South China were analyzed for their contents, composition, antioxidant capacities, and inhibitory activities againstα-glucosidase. We found that there were significant differences in phenolic/flavonoid contents among diverse varieties and different parts (peel and pulp), and the contents were highest in the peel’s bound form. The results of UHPL-Q-Exactive HF-X and HPLC showed that chlorogenic acid, gentisic acid, and rutin were abundantly distributed over the three phenolic fractions in peel and pulp of all wampee samples, while isoquercitrin was the most abundant in the conjugated form of peel/pulp and myricetin had the richest content in the free form of peel/pulp. Wampee peel had stronger antioxidant capacities of ABTS+, DPPH, ·OH, and FRAP than the pulp, and the bound phenolic fraction of the peel/pulp had much higher antioxidant activities than FP and CP fractions. It is interesting that the same phenolic fraction of the wampee peel displayed roughly close IC50values ofα-glucosidase inhibition to those from the pulp samples. The relationship between individual phenolic and TPC/TFC/the bio-activities and the similarity among the free, conjugated, and bound phenolic fractions in peel and pulp samples were explored by using Pearson correlation analysis, principal component analysis, and hierarchical cluster analysis. This work provides a systematic and comprehensive comparison of the three phenolic fractions of diverse wampee varieties and different parts, and a rationale for applying phenolics from wampee fruits. Keywords:wampee varieties;free phenolic;conjugated phenolic;bound phenolic;antioxidant capacities;inhibitory activity againstα-glucosidase;multivariate analysis

Ultrastretchable, Self-Healable, and Tissue-Adhesive Hydrogel Dressings Involving Nanoscale Tannic Acid/Ferric Ion Complexes for Combating Bacterial Infection and Promoting Wound Healing

Ke Yang,Xueyao Zhou,Zhaoli Li,Zefeng Wang,Yuze Luo,Le Deng,Dinggeng He

Preventing bacterial infections and accelerating wound closure are essential in the process of wound healing. Current wound dressings lack enough mechanical properties, self-healing ability, and tissue adhesiveness, and the bacterial killing also relies on the use of antibiotic drugs. Herein, a well-designed hybrid hydrogel dressing is constructed by simple copolymerization of acrylamide (AM), 3-acrylamido phenylboronic acid (AAPBA), chitosan (CS), and the nanoscale tannic acid (TA)/ferric ion (Fe3+) complex (TFe). The resulting hydrogel possesses lots of free catechol, phenylboronic acid, amine, and hydroxyl groups and contains many reversible and dynamic bonds such as multiple hydrogen bonds and boronate ester bonds, thereby showing satisfactory mechanical properties, fast self-healing ability, and desirable tissue-adhesive performance. Benefiting from the high photothermal conversion efficiency of the TFe, the hydrogel exhibits satisfactory antibacterial activity against both Gram-positive and Gram-negative bacteria. Moreover, the embedded TFe also endows the hydrogel with good antioxidant activity, anti-inflammatory property, and cell proliferation to promote tissue regeneration. Remarkably, in vivo animal assays reveal that the hybrid hydrogel effectively eliminates biofilm bacteria in the wound sites and accelerates the healing process of infected wounds. Taken together, the developed versatile hydrogels overcome the shortcomings of traditional wound dressings and are expected to become potential antibacterial dressings for future biomedical applications.

Optimization of ultrasonic-assisted extraction model of Pinus koraiensis seed scales oil induced by tea saponin

Zhanjun Li,Yibin Xu,Yunwei Liu,Ke Ma,Kunlun Wang,Fengjian Yang

Tea saponin was used for UAE of oil from Pinus koraiensis seed scales for the first time, and the extraction kinetic model was fitted and compared with that of traditional SE. Considering the relationship among the oil yield, proanthocyanidins content and solvent properties, ethanol was selected as the optimum extraction solvent. PBD and BBD were used to determine the optimal conditions: 63% aqueous ethanol solution, liquid-to-material ratio was 11:1 mL/g, ultrasonic power was 100 W, ultrasonic duration was 15 min, temperature was 45 °C, and the dose of tea saponin was 0.3%. Under the optimal conditions, the oil yield and proanthocyanidins content from P. koraiensis seed scales were 22.73 ± 0.42% and 3.04 mg/g ± 0.08%, respectively. The actual operating value was consistent with the theoretically predicted value. Comparison of the extraction kinetics model, oil components, physical and chemical properties, antioxidants, microstructure of oil residue, energy consumption and ecological emission characteristics of UAE and SE shows that UAE is a more advantageous technique.

Enhancement of functional activity and biosynthesis of exopolysaccharides in Monascus purpureus by genistein treatments

Liuming Xie,Gang Wang,Jianhua Xie,XianXiang Chen,Jiayan Xie,Xiaoyi Shi,Zhibing Huang

The exopolysaccharides (EPS) produced by the edible medicinal fungus Monascus purpureus (EMP) become the center of growing interest due to their techno-functional properties and their numerous applications in the food industries; however, the low EPS yields limit its application. In this study, the effect of genistein supplementation on the production, rheological and antioxidant properties of EPS by M . purpureus and its biosynthesis mechanism were explored. The results indicated that the addition of genistein (3 g/L) generated a 110% and 59% increase in the maximum mycelial biomass and EPS yield, respectively. The genistein supplementation group (G-EMP) had higher molar percentages of Xyl and Man, and significantly decreased molecule weight and particle size of EPS, which resulted in stronger antioxidant effect and cell growth promotion. Rheological analysis showed that both EMP and G-EMP demonstrated pseudoplastic fluid behavior and G-EMP exhibited strong gel-like elastic behavior (G' > G"). Furthermore, genistein not only facilitated the production of EPS by regulating cell membrane permeability, enhancing cellular respiratory metabolism and monosaccharide precursor synthesis pathways, and enhancing antioxidant enzyme activity to reduce oxidative stress damage, but also affected the composition of the monosaccharides by increasing enzyme activity in the underlying synthesis pathways. These findings expand the application of M . purpureus resources and provide a paradigm for future study of the structural and functional characteristics of EPS.

Natural Small Biological Molecule Based Supramolecular Bioadhesives with Innate Photothermal Antibacterial Capability for Nonpressing Hemostasis and Effective Wound Healing

Xiang Ke,Shuxian Tang,Hao Wang,Yusong Cai,Zhiyun Dong,Mingjing Li,Jiaojiao Yang,Xinyuan Xu,Jun Luo,Jianshu Li

Bioadhesives with immediate wound closure, efficient hemostasis, and antibacterial properties that can well integrate with tissue are urgently needed in wound management. Natural small biological molecule based bioadhesives hold great promise for manipulating wound healing by taking advantage of integrated functionalities, synthetic simplification, and accuracy, cost efficiency and biosafety. Herein, a natural small biological molecule based bioadhesive, composed of natural small biological molecules (α-lipoic acid and tannic acid) and a small amount of ferric chloride, was prepared via an extremely simple and green route for wound management. In this system, covalent and noncovalent interactions between each component resulted in the self-healing supramolecular bioadhesive. It possessed appropriate wet-tissue adhesion, efficient nonpressing hemostasis and free radical scavenging abilities. More importantly, the interaction between tannic acid and Fe3+ endowed the bioadhesive with innate and steady photothermal activity, which showed excellent photothermal bactericidal activity to both E. coli and S. aureus. The bioadhesive promoted wound healing for linear and circular wounds in vivo, especially for infectious wounds under near-infrared (NIR) irradiation. This bioadhesive will have promising value as a safe and effective antimicrobial adhesive for infectious wound management.

Salicylaldehyde and D-(+)-galactose functionalized chitosan oligosaccharide nanoparticles as carriers for sustained release of pesticide with enhanced UV stability

Shaochun Su,Li Hao,Huayao Chen,Xinhua Zhou,Hongjun Zhou

Sustainable food consumption is intertwined with sustainable agriculture development, which focuses on the production and protection of agricultural crops in a safe and effective fashion. Herein, amphiphilic functionalized chitosan oligosaccharide nanoparticles (CSD) were formed by a Schiff base reaction grafted with salicylaldehyde and D-(+)-galactose ( D -Gal). Avermectin (AVM) was used as a model pesticide and self-assembled onto CSD to obtain AVM@CSD nanoparticles with an encapsulation efficiency of 74.94%. AVM@CSD nanocarrier exhibited excellent anti-UV property to retain more AVM, due to their Schiff base structure and salicylaldehyde for UV shielding. Compared with bare AVM and commercial AVM emulsifiable concentrates, AVM@CSD preserved high insecticidal activity after UV irradiation . Additionally, CSD carrier promoted cucumber seed germination and seedling growth. These data suggested the potential utilization of the amphiphilic chitosan oligosaccharide as carriers for pesticides to improve their delivery and release properties and opens a novel avenue towards the application of water-dispersible and biological CSD in the field of pest management and plant growth.
