Low-salt cured fish is prone to deterioration due to lipid oxidation and microbial proliferation during refrigeration. Chlorogenic acid (CGA), with excellent antioxidant and antimicrobial activity, is a promising candidate for the preservation of cured fish. However, its instability in the presence of environmental factors significantly confines its direct application. In this research, CGA was encapsulated in liposomes and utilized as a dip-coating for cured fish. The effects of varying concentrations of CGA-loaded liposomes (L-CGA) coating on the physicochemical quality and microbial diversity of cured fish were rigorously compared to those treated with CGA solutions, blank liposomes, and distilled water throughout 32 days’ storage at 4 °C. The results showed that L-CGA exhibited a higher lipid oxidation-inhibiting capacity (generation of hydroperoxides and their secondary oxidation products) than the corresponding free CGA at fixed concentrations. Furthermore, the liposomal formulation showed significantly enhanced inhibitory activity against dominant spoilage-associated bacterial genera (e.g.,Staphylococcus,Macrococcus, andRothia), with the L-CGA loaded at 800 mg/L of CGA showing optimal effectiveness. This enhanced preservation effect can be attributed to the protective and controlled release properties of the liposomes, which facilitate improved preservation outcomes for CGA. These findings demonstrate that L-CGA could be used as a promising preservative for low-salt cured fish or some similar products.

gcSGs and smSGs exhibited more susceptibility to CaHMB than scSGs or sbSGs. • The water retention ability of surimi gels decreased due to CaHMB hydration effect. • The substation of NaCl by CaHMB promoted the protein aggregation with dense network. • CaHMB promoted the unfolding of MP heads and tails in different type of surimi gels. • CaHMB could inhibit the MHC degradation of scSGs, smSGs, and sbSGs. The improvement of gel properties in low-salt surimi is vital for constructing health-oriented products. In this work, calcium β-hydroxy-β-methylbutyrate (CaHMB) was used to substitute sodium chloride to fabricate various low-salt surimi gels derived from freshwater and marine fish. Results indicated that grass carp surimi gels (gcSGs) and Spanish mackerel surimi gels (smSGs) presented superior gel performance with homogeneous microstructures in the presence of CaHMB. Remarkably, they exhibited the greatest textual properties in Na-L/Ca-H groups with increments of 32%–56%, compared with those in Na groups. The water retention capacity and thermal stability of samples were affected by CaHMB, as corroborated by 12.1%–19.4% reduction in water holding capacity and 5.4%–31.4% decrease in critical temperature than those of Na groups. CaHMB promoted unfolding of protein heads and tails in freshwater fish and marine fish surimi gels, respectively. Overall, replacing sodium salts with CaHMB was effective for producing surimi with high quality and nutrition. Download: Download high-res image (287KB) Download: Download full-size image

Color indicator films for fish freshness were fabricated by incorporating κ-carrageenan (CAR) polymer with red grape skin extract (GSE) as a pH-sensing agent and silver nanoparticles (AgNPs) as an antimicrobial agent. Anthocyanins in GSE exhibited distinguished pH responsive color changes. GSE and AgNPs were well compatible with CAR with intramolecular interactions, approved by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) analysis, thermo-gravimetric analysis (TGA) and differential scanning calorimetry (DSC). GSE-containing films displayed distinguished color changes in response to pH variations and volatile ammonia. Enhanced UV blocking ability and strong antioxidant activity were revealed for GSE included films without sacrificing the physico-chemical properties of the CAR film. Films containing AgNPs showed improved mechanical strength and strong antimicrobial ability against both Escherichia coli and Staphylococcus aureus . The CAR/AgNPs/GSE film displayed a distinctive color change corresponding to changes in the total volatile basic nitrogen (TVB-N) of fish during storage. In addition, the CAR/AgNPs/GSE film showed excellent color stability to consecutive UV exposure and its storage time at 25 °C is expected to be at least 240 days, which indicates that it has high potential as an intelligent food freshness indicator film.

Since excess histamine (His) in seafood poses a significant health threat, the development of intelligent, on-site, and visual multimodal methods for detecting fish freshness has attracted widespread attention. In this study, we developed a Lewis base-induced paper-based chip coupled with smartphone RGB (Red Green Blue) recognition for the real-time colorimetric/fluorescent dual-mode detection of His in fish meat. The sensing mechanism was based on the catalytic role of His in rapidly forming a green fluorescent compound (GFC) from dopamine and 3-ethylamino-4-methylphenol. Interestingly, the introduction of 1% Tween (TW) 80 was found to significantly enhance the fluorescence signal and real-time stability of the system, achieving an exceptional detection limit of 16.67 nM for His. By immobilizing the GFC probe on Whatman 1 filter paper, a paper-based test strip for His was further developed. His quantification is achieved through RGB analysis with a smartphone, allowing visual and real-time assessment of fish meat freshness directly in the field. The developed sensor exhibits outstanding selectivity, stability, and sensitivity, and has been successfully applied to monitor His levels in fish meat samples (crucian, saury and golden pomfret) over time, providing a reliable means to prevent His poisoning. Therefore, this work presents a robust, low-cost, and user-friendly point-of-care testing platform that holds significant potential for applications in intelligent food monitoring and environmental sensing.

Examined flavor and lipid profiles during thermal processing of fish oil and algal oil. • The potential of AO as an alternative to FO in food processing was investigated. • Higher temperatures induced similar trends in flavor and lipid profiles of both oils. • Key volatile markers in FO were predominantly derived from phospholipid oxidation. • Key volatile compounds in AO were primarily produced via triacylglycerol oxidation. As a major source of essential polyunsaturated fatty acids, fish oil faces increasing sustainability constraints, underscoring the need for viable lipid alternatives. Volatile organic compounds and lipidomic transformations of fish oil (FO) and algal oil (AO) were characterized. In both oils, aldehydes, ketones, and alcohols constituted the predominant classes, and their relative proportions were broadly comparable. As oxidation temperature increased, FO and AO displayed similar trends in flavor development and lipid compositional changes. FO preferentially developed fatty–green and roasted notes dominated by aldehydes, benzenoids, and furans. AO exhibited a broad and complex flavor profile enriched in alcohols, ketones, sesquiterpenoids, and indole derivatives, imparting enhanced herbal, fruity, and floral characteristics during thermal processing. Volatile formation in FO was dominated by phospholipid oxidation, yielding key markers such as (E)-2-nonenal and (Z)-3-hexenal. Triacylglycerol oxidation predominated in AO, producing characteristic compounds including (E, E)-2,4-nonadienal and 2-methylbutanal. Despite distinct oxidation mechanisms, both oils exhibited comparable thermal flavor evolution, supporting algal oil as a sustainable and vegetarian-friendly alternative to fish oil. Zebrafish ( Danio rerio ), Marine medaka ( Oryzias melastigma ), and freshwater medaka ( Oryzias latipes ) are three major animal species in evaluation of environmental pollutant toxicity and building of human disease models with conserved physiological and molecular pathways. Bacterial strains from gut microbiota should firstly be isolated for their functions’ exploration. In this study, we optimized a culture-based workflow under aerobic and anaerobic conditions to recover intestinal bacteria from adult zebrafish (ZF), marine medaka (MM), and freshwater medaka (OL) using both dissection-based and in vivo sampling. The isolates were then identified by Gram staining, 16S rRNA gene sequencing, phylogenetic analysis, and API 20E-based biochemical characterization. The culturable bacteria isolated from gut microbiota were identified belonged to 14 genera in ZF, 12 genera in MM, and 18 genera in OL. Comparative analysis showed clear differences in the composition of cultured gut bacteria among the three fish species. Overall, the library and the differences of gut bacteria in three key fish models provided the potential applications of bacterial strains as the probiotics, to against the fish pathogens and to increase the pollutant toxicity-resistant. Moreover, the bacterial library will support the deep researches combined with germ-free (GF) animals to clarify the relationships of intestinal microbiota to host health.

This study evaluated the effects of co-inoculation with Latilactobacillus sakei A20 and Lactiplantibacillus plantarum LJ1–1 at different ratios on the quality and safety of fermented mandarin fish ( Siniperca chuatsi ). Compared with the C group without starter culture, the inoculated groups effectively delayed lipid oxidation and decreased TVB-N content by 14.00%. Notably, the accumulation of biogenic amines was markedly inhibited, with tyramine and cadaverine levels decreasing by over 30.00%. It also reshaped the microbial community by suppressing spoilage bacteria such as Shewanella baltica and Aeromonas sobria , while promoting the accumulation of esters and aldehydes. Sensory evaluation confirmed a 15.44% increase in overall acceptability. Furthermore, a fermentation period of 7 days is recommended to ensure product safety and quality. These results indicate that Lat. sakei A20 and Lpb. plantarum LJ1–1 have great potential as mixed starter cultures for improving the safety, flavor, and sensory acceptability of fermented mandarin fish.

Histamine food poisoning (HFP) or scombrotoxic fish poisoning is a major public health concern associated with seafood and seafood products containing copious amounts of free histidine , which may be decarboxylated into scombrotoxic histamine, the main culprit of HFP. Moreover, the deamination product of the histidine, trans -urocanic acid, is assumed to be scombrotoxic by degranulating mast cells to release endogenous histamine after isomerizing to the cis -form, as well as known to have immunosuppressive effects. In recognition of the potential health risks associated with the histidine degradatives, this work aimed to provide a method for the simultaneous determination of histidine, histamine, and urocanic acid isomers in food, based on precolumn dansylation and subsequent reversed-phase HPLC analysis. The dansylation conditions were optimized for yield with the design of experiments approach consisted of a center point-enhanced 2 4 factorial design followed by sequential steepest ascent searches. Satisfactory chromatographic separation of fish sauce was achieved within 9 min by retention modeling of both the analytes and potentially interfering peaks. The method was fully validated with limits of detection between 0.004 and 0.017 mg 100 g −1 fish sauce, and successfully applied to several fish sauce samples, in which the histamine and urocanic acid levels were found low and unlikely of concern, with correspondingly low levels of the precursor histidine.

Concerns about food safety have been arisen due to the improper use of chemicals in aquaculture. Malachite green (MG) has attracted attention because of its illegal usage and its potential negative impacts on the environment and public health. Surface-enhanced Raman scattering (SERS) platforms coupled with different SERS substrates have been employed for rapid analysis of MG residues in food. However, the most commonly used SERS substrates were non-reusable and showed limited detection sensitivity. In this study, a novel SERS substrate with a good recyclability and a high sensitivity was prepared by electrostatically assembling together a metal–organic framework material called materials of institute lavoisie-100(Fe) (MIL-100(Fe)) and Au NPs. The lowest detectable concentration of MG was 10 −13 M based on the optimal substrate. The SERS sensor was applied for the detection of the trace MG in fish pond water, which was accomplished with the correlation coefficients R 2 = 0.991–0.996 in a concentration range of 10 −6 –10 −13 M. Moreover, MIL-100(Fe)/Au was recycled at least five times, realizing a “detection to degradation”, showing great potential for food contamination monitoring due to its distinguished performance.

A novel type of PEG-modified halloysite was prepared and used as a hydrophilic interaction and cation exchange mixed-mode sorbent for solid-phase extraction of biogenic amines in fish samples. The eluates were analyzed by high-performance liquid chromatography-ultraviolet detection after the derivatization with benzoyl chloride. The developed sorbent was characterized by scanning electron microscopy, infrared spectroscopy, X-ray diffraction, zeta potential analyzer, and thermo-gravimetric analysis. After the optimization of various parameters influencing the extraction efficiency, the PEG-modified halloysite-based SPE method was evaluated. The adsorption capacities of putrescine, spermine, phenethylamine, and histamine were as high as 9.3, 8.5, 5.7, and 5.6 mg g −1 , respectively. Satisfactory reproducibility of sorbent preparation was obtained with within-batch and batch-to-batch relative standard deviations (RSDs) lower than 3.9% and 8.6%, respectively. The biogenic amine spiking recoveries in fish samples ranged from 84.3 to 105.5% with good RSDs lower than 7.8%. Intra-day and inter-day precision, expressed as RSDs, were better than 8.8%. The limits of detection of histamine, putrescine, phenethylamine, and spermine were 9.4, 1.9, 0.5, and 0.9 μg L −1 , respectively. This work provides a new hydrophilic interaction and cation exchange mixed-mode sorbent and is successfully applied to the extraction of trace biogenic amines from fish samples. Graphical Abstract

It is important to detect the presence of biogenic amines (BAs) as indicators of food freshness. The purpose of this study was to develop a novel ultrasonic–microwave synergistic supramolecular solvent liquid–liquid microextraction based on solidification of floating organic droplet (UMS-SUPRAS-SFO-LLME) combined with high-performance liquid chromatography for the determination of BAs. The physical properties and microstructure of SUPRAS based on 1-dodecanol and tetrahydrofuran were studied, and the extraction conditions such as the SUPRAS volume, the UMS process, and the centrifugal conditions were optimized. The results for the extraction kinetics and thermodynamics showed that UMS-SUPRAS-SFO-LLME is a spontaneous, endothermic diffusion process. The linear ranges of this method are 0.1–2.0 × 10 5 ng·mL −1 ( R 2 > 0.994), the limits of detection are 4.0 × 10 -3 -6.0 × 10 -2 ng·mL −1 , and the recoveries were 96.28–103.15%. Compared with existing analysis methods, UMS-SUPRAS-SFO-LLME is a sensitive, green and economical sample pretreatment method for analyzing the enrichment of BAs in beer and fish.

Background Wound closure in the complex body environment places higher requirements on suture’s mechanical and biological performance. In the scenario of frequent mechanical gastric motility and extremely low pH, single functional sutures have limitations in dealing with stomach bleeding trauma where the normal healing will get deteriorated in acid. It necessitates to advance suture, which can regulate wounds, resist acid and intelligently sense stomach pH. Methods Based on fish swim bladder, a double-stranded drug-loaded suture was fabricated. Its cytotoxicity, histocompatibility, mechanical properties, acid resistance and multiple functions were verified. Also, suture’s performance suturing gastric wounds and Achilles tendon was verified in an in vivo model. Results By investigating the swim bladder’s multi-scale structure, the aligned tough collagen fibrous membrane can resist high hydrostatic pressure. We report that the multi-functional sutures on the twisted and aligned collagen fibers have acid resistance and low tissue reaction. Working with an implantable “capsule robot”, the smart suture can inhibit gastric acid secretion, curb the prolonged stomach bleeding and monitor real-time pH changes in rabbits and pigs. The suture can promote stomach healing and is strong enough to stitch the fractured Achilles tendon. Conclusions As a drug-loaded absorbable suture, the suture shows excellent performance and good application prospect in clinical work.
