Amplified circularly polarized luminescence in assembly of terpyridine-ZnII and GTP induced by silver(I) ion

Fan Huahua,Wang Runjia,Zhang Li,Liu Minghua

The G-quartet, a hydrogen-bonded macrocycle formed by cation-templated assembly of guanosine derivative, plays a vital role in areas ranging from structural biology to supramolecular chemistry and nanotechnology. Herein, we show a zinc-terpyridine derivative (Ftpy) that could form complex with various guanosine phosphates, exhibiting relatively weak circularly polarized luminescence CPL (dissymmetry factor, g lum ≈ 1 × 10 −3 ). Interestingly, when Ag + ions are involved, significantly boosted circularly polarized luminescence (CPL) is obtained in the assemblies of Ftpy with GTP, which the g lum can reach to 0.15. In contrast, no significant difference is observed in the presence of other cations (Na + , K + , Rb + , NH 4 + , Cu 2+ , and Sr 2+ ). It reveals that CPL signal is amplified due to the formation of G-quartet in the presence of Ag + ions, which further hierarchically self-associate into helically stacked supramolecular aggregates with the cooperation and π - π of Ftpy, contributing to the expression of chirality information. Furthermore, the co-assemblies of Ftpy/GTP/Ag + exhibit an enantioselective recognition of enantiomeric penicillamine by CPL spectra. This work provides a straightforward avenue to fabricate supramolecular assemblies with strong CPL based on G-quartet formation.

New Rhenium(I) Complexes: Synthesis, Photophysics, Cytotoxicity, and Functionalization of Gold Nanoparticles for Sensing of Esterase

Mei-Jin Li,Xing Liu,Mei-Juan Nie,Zhao-Zhen Wu,Chang-Qing Yi,Guo-Nan Chen,Vivian Wing-Wah Yam

A series of the luminescent Re(I) complexes, [Re(bpy-(CH2OH)2)(CO)3(L1)], [Re(bpy-(CH2OH)2)(CO)3(L2)]+, [Re(bpy-(CH2OH)2)(CO)3(L3)]+, and [Re(bpy-(CH2OH)2)(CO)3(L4)] ((bpy-(CH2OH)2 = 4,4′-bis(hydroxymethyl)-2,2′-bipyridine; L1 = 4-pyridylmethyl lipoate; L2 = [1,2]dithiolane-3-pentyl 4-pyrindylacetate; L3 = 4-pyridinyl-methanol; L4 = 4-pyridinylmethyl pentanoate), were synthesized and characterized. The photophysical properties and cytotoxicity of the complexes and the sensing of esterase based on the gold nanoparticles (Au-NPs) modified with the complexes were also studied. The intense luminescence of the Re(I) complexes was quenched efficiently by Au-NPs due to the energy and electron transfer between the Re(I) fluorophores and Au-NPs. Upon addition of esterase to the mixture, the emission of the Re(I) complexes 1 and 2 system was enhanced significantly by the release of the Re(I) complexes from the surface of the Au-NPs.

Astragaloside I Promotes Lipophagy and Mitochondrial Biogenesis to Improve Hyperlipidemia by Regulating Akt/mTOR/TFEB Pathway

Jie Zhao,Gai Gao,Jing Ding,Wei Liu,Tao Wang,Liang Zhao,Jiangyan Xu,Zhenqiang Zhang,Xiaowei Zhang,Zhishen Xie

The simultaneous enhancement of lipophagy and mitochondrial biogenesis has emerged as a promising strategy for lipid lowering. The transcription factor EB (TFEB) exhibits a dual role, whereby it facilitates the degradation of lipid droplets (LDs) through the process of lipophagy while simultaneously stimulating mitochondrial biogenesis to support the utilization of lipophagy products. The purpose of this study was to explore the effect of astragaloside I (AS I) on hyperlipidemia and elucidate its underlying mechanism. AS I improved serum total cholesterol and triglyceride levels and reduced hepatic steatosis and lipid accumulation in db/db mice. AS I enhanced the fluorescence colocalization of LDs and autophagosomes and promoted the proteins and genes related to the autolysosome. Moreover, AS I increased the expression of mitochondrial biogenesis-related proteins and genes, indicating that AS I promoted lipophagy and mitochondrial biogenesis. Mechanistically, AS I inhibits the protein level of p-TFEB (ser211) expression and promotes TFEB nuclear translocation. The activation of TFEB by AS I was impeded upon the introduction of the mammalian target of rapamycin (mTOR) agonist MHY1485. The inhibition of p-mTOR by AS I and the activation of TFEB were no longer observed after administration of the Akt agonist SC-79, which indicated that AS I activated TFEB to promote lipophagy-dependent on the Akt/mTOR pathway and may be a potentially effective pharmaceutical and food additive for the treatment of hyperlipidemia.

Polyethylene Terephthalate Fiber Modified with Type I Collagen as a 3D Scaffold Material for Bioartificial Liver

Yang Li,Yang Zhang,Jianping Gao,Shuguang Liao,Guifeng Zhang

Acute and chronic liver failure are clinically significant conditions, and the artificial liver support system (ALSS) is emerging as a novel and effective approach for the clinical management of liver failure. Within this framework, scaffold materials occupy a pivotal position as integral components of the bioreactor. Elevating the performance capabilities of these scaffolds not only augments the therapeutic efficacy of the artificial liver but also lays the groundwork for refining and selecting large-scale hepatocyte culture models. In this study, we introduced a novel hepatocyte scaffold material designated as PET-COL, crafted by coating polyethylene terephthalate (PET) with collagen. This involved a sequence of modifications, including alkaline hydrolysis, EDC/NHS activation and crosslinking, as well as collagen conjugation. The physicochemical attributes of the scaffold were thoroughly characterized by Fourier-transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), second harmonic generation (SHG), water contact angle measurements, and high-performance liquid chromatography–mass spectrometry (HPLC-MS). Furthermore, an investigation into the material’s biological properties was conducted that encompassed SEM (HepaRG growth), fluorescence staining (assessment of cell viability), staining by trypan blue (HepaRG counting), CCK-8 (proliferation of cells), biochemical testing, and immunosorbent assay. Our findings revealed that collagen was covalently bonded to the PET surface, leading to a substantial enhancement in the material’s hydrophilicity (p< 0.001). The quantity of collagen coating was determined to be precisely 33.30 μg per scaffold. Human liver progenitor HepaRG cells thrived on the PET-COL material. Compared with the untreated group, cell viability, albumin secretion, urea synthesis, and the expression levels of CYP3A4 and CPS1 increased significantly (p< 0.001), demonstrating remarkable biological vitality. The PET-COL scaffold, as developed in this study, holds immense potential for application in bioartificial livers.

QSAR prediction, synthesis, anticancer evaluation, and mechanistic investigations of novel sophoridine derivatives as topoisomerase I inhibitors

Lin Zhu,Yongle Yu,Youfu Ma,Yenong Shi,Jamal Alzobair Hammad Kowah,Lisheng Wang,Mingqing Yuan,Xu Liu

Sophoridine, which is derived from the Leguminous plant Sophora alopecuroides L., has certain pharmacological activity as a new anticancer drug. Herein, a series of novel N-substituted sophoridine derivatives was designed, synthesized and evaluated with anticancer activity. Through QSAR prediction models, it was discovered that the introduction of a benzene ring as a main pharmacophore and reintroduced into a benzene in para position on the phenyl ring in the novel sophoridine derivatives improved the anticancer activity effectively. In vitro, 28 novel compounds were evaluated for anticancer activity against four human tumor cell lines (A549, CNE-2, HepG-2, and HEC-1-B). In particular, Compound 26 exhibited remarkable inhibitory effects, with an IC 50 value of 15.6 μM against HepG-2 cells, surpassing cis-Dichlorodiamineplatinum (II). Molecular docking studies verified that the derivatives exhibit stronger binding affinity with DNA topoisomerase I compared to sophoridine. In addition, 26 demonstrated significant inhibition of DNA Topoisomerase I and could arrest cells in G 0 /G 1 phase. This study provides valuable insights into the design and synthesis of N-substituted sophoridine derivatives with anticancer activity.

Using N–I–N Photodiodes Made of Perovskite Single Crystals for Low Noise Gamma-Ray Spectroscopy

Xin Wang,Yubing Xu,Yuzhu Pan,Shunjie Chai,Jie Wu,Jingda Zhao,Yuwei Li,Zhiwei Zhao,Qing Li,Jun Wu,Jing Chen,Byung Seong Bae,Jianming Zhou,Ying Zhu,Wei Lei,Xiaobao Xu

Solution-processed lead halide perovskite single crystals (LHPSCs) are believed to have great potential in gamma-ray spectroscopy. However, obtaining low-defect LHPSCs from a solution at low temperatures is difficult compared to obtaining Bridgman single crystals such as CdTe and Si. Herein, noise from the intrinsic defects of LHPSCs is considered as the main problem hindering their gamma-ray detection performance. By isolating the defect-induced holes in LHPSCs via energy barriers, we show that NIN photodiodes based on three types of LHPSCs, i.e., MAPbBr3 (MA = CH3NH3), MAPbBr2.5Cl0.5, and cascade LHPSCs, have demonstrated good energy resolution in the range of 6.7–10.3% for 662 keV 137Cs gamma-ray photons. The noise for >10 mm3 devices is low, in the order of 340–860 electrons, and the electron collection efficiency reaches 23–43%. These results pave the way for obtaining low-cost, large, high energy-resolution gamma-ray detectors at room temperature (300 K).

Visual aptasensor based on PtNPs/g-C3N4 and DNase I for the field detection of acetamiprid

Wang Linhai,Hu Xiao,Yao Guoqing,Chen Qinhua,Wang Sicen

A visual aptasensor based on the combination of graphitic carbon nitride loaded with platinum nanoparticles (PtNPs/g-C 3 N 4 ) and deoxyribonuclease I (DNase I) was developed for detecting acetamiprid. The prepared PtNPs/g-C 3 N 4 exhibited excellent peroxidase (POD)-like activity, demonstrating the capacity to oxidize the colourless o-phenylenediamine (OPD) to produce the coloured product diaminophenazine (DAP) in the presence of hydrogen peroxide (H 2 O 2 ). The DNA aptamer of acetamiprid can adsorb onto the surface of PtNPs/g-C 3 N 4 , thereby inhibiting its POD activity. The binding of acetamiprid to its aptamer results in the aptamer desorbing from the surface of PtNPs/g-C 3 N 4 and subsequent digestion by DNase I. Ascribed to the synergistic effect of these factors, the aptasensor can achieve the rapid on-site detection of acetamiprid based on the acetamiprid-induced the colour change of the solution just with the smartphone. Furthermore, due to the remarkable fluorescence signal of DAP at 570 nm, the aptasensor under fluorescence mode enables highly sensitive and quantitative detection of acetamiprid with a linear range of 1 ng/mL to 4 µg/mL and a detection limit as low as 0.31 ng/mL. The aptasensor has successfully realized the detection of acetamiprid in river water and cucumber samples, offering a novel perspective for the rapid assessment of environmental pollution and food safety risks associated with acetamiprid residues. Theoretical and Experimental Study of Functional Additives in Au(I)-DMH Cyanide-Free Gold Electroplating System

Linghui Meng,Jie Jiang,Xuesong Peng,Lin Zhu,Ruopeng Li,Yaqiang Li,Maozhong An

Developing a cyanide-free gold electrodeposition system is significant for replacing the cyanide electroplating gold plating system, which is highly harmful to the environment and toxic to human health. In our study, a cyanide-free system for electrodepositing gold (Au) coatings was proposed with AuCl as the main salt and 5,5-dimethyl hydantoin (DMH) as the Au (I) complexing agent. Through theoretical calculations and electrochemical test screening, 4,6-dimethyl-2-mercaptopyrimidine (DMP) is suitable for DMH-based gold electrodeposition and was selected as an electroplating additive. The role of DMP in gold electrodeposition was further studied. In particular, the adsorption–coordination mechanism was proposed to elucidate the role of DMP, based on calculations of the Bader charge density difference for DMP adsorbed on the gold surface and the interaction between DMP and Au+. Electrochemical tests such as cyclic voltammetry and chronoamperometry were utilized to study the impact of varying DMP concentrations on gold electrodeposition behavior. Chronoamperometric analysis indicates that Au electrodeposition obeys a 3D diffusion-controlled progressive nucleation process, independent of DMP concentration and applied potential, while the presence of DMP markedly suppresses grain growth. The addition of DMP significantly improved the surface morphology and brightness of the gold coating, resulting in a smooth and dense layer with a golden-yellow appearance. XRD results showed that the addition of DMP reduced the grain size of the Au coating from 22.1 to 15.7 nm, with the gold coating primarily growing along the Au (111) plane. The XPS test results show that the purity of the gold plating layer is high, and there is no inclusion of additives in the coating.

Cationization-Enhanced Type I and Type II ROS Generation for Photodynamic Treatment of Drug-Resistant Bacteria

Shanshan Liu,Bingnan Wang,Yuewen Yu,Yubo Liu,Zeyan Zhuang,Zujin Zhao,Guangxue Feng,Anjun Qin,Ben Zhong Tang

Photodynamic therapy as an emerging phototheranostic approach holds great potential for antibacterial treatment, but is limited by compromised reactive oxygen species (ROS) generation in an aggregate and hypoxic microenvironment. Herein, we report a molecular cationization approach to boost the ROS, especially type I ROS generation of aggregation-induced emission (AIE) photosensitizers for photodynamic treatment of drug-resistant bacteria. Such cationization reinforces the electron-accepting ability of the cationic moiety, promotes intersystem crossing (ISC), and increases electron separation and transfer processes. The resultant CTBZPyI exhibits largely enhanced ROS generation ability with predominant hydroxyl radical generation over its neutral counterpart in aggregate. Moreover, cationization also confers CTBZPyI with the bacterial binding ability and a moderate bacterial inactivation ability in the dark. Further light irradiation leads to superb antibacterial performance, which largely promotes the healing process of a MRSA-infected wound. Such a cationization strategy is expected to be a general strategy for the design of highly effective type I photosensitizers for bacterial infection treatment.

Stress triggers irritable bowel syndrome with diarrhea through a spermidine-mediated decline in type I interferon

Li Zhang,Hao-li Wang,Ya-fang Zhang,Xin-tao Mao,Ting-ting Wu,Zhi-hui Huang,Wan-jun Jiang,Ke-qi Fan,Dan-dan Liu,Bing Yang,Mei-hui Zhuang,Guang-ming Huang,Yinming Liang,Shu Jeffrey Zhu,Jiang-yan Zhong,Guang-yin Xu,Xiao-ming Li,Qian Cao,Yi-yuan Li,Jin Jin

Irritable bowel syndrome with diarrhea (IBS-D) is a common and chronic gastrointestinal disorder that is characterized by abdominal discomfort and occasional diarrhea. The pathogenesis of IBS-D is thought to be related to a combination of factors, including psychological stress, abnormal muscle contractions, and inflammation and disorder of the gut microbiome. However, there is still a lack of comprehensive analysis of the logical regulatory correlation among these factors. In this study, we found that stress induced hyperproduction of xanthine and altered the abundance and metabolic characteristics of Lactobacillus murinus in the gut. Lactobacillus murinus -derived spermidine suppressed the basal expression of type I interferon (IFN)-α in plasmacytoid dendritic cells by inhibiting the K63-linked polyubiquitination of TRAF3. The reduction in IFN-α unrestricted the contractile function of colonic smooth muscle cells, resulting in an increase in bowel movement. Our findings provided a theoretical basis for the pathological mechanism of, and new drug targets for, stress-exposed IBS-D.

Structural diversity of copper(I) alkynyl cluster-based coordination polymers utilizing bifunctional pyridine carboxylic acid ligands

Zheng Liu,Jun-Jie Fang,Zhi-Yi Wang,Yun-Peng Xie,Xing Lu

The utilization of bifunctional ligands, specifically pyridine carboxylic acids, endowed with dual coordination sites, has been instrumental in the assembly of polymer materials. The ambidentate characteristics of these ligands play a crucial role in shaping the structure and framework of cluster-based polymers. In this study, we have synthesized a diverse array of multidimensional copper(I) alkynyl cluster-based polymers (CACPs) by employing four distinct pyridine carboxylic acids – namely, isonicotinic acid (INA), 6-isoquinolinecarboxylic acid (IQL), 4-pyridin-4-yl-benzoic acid (4-PyBA), and 3-pyridin-4-yl-benzoic acid (3-PyBA) – as linking ligands. These pyridine carboxylic acids not only serve as protective ligands but also act as pivotal linkers in constructing the cluster-based framework materials, exerting significant influence on the overall framework structures. Furthermore, the incorporation of auxiliary ligands has been shown to markedly impact the structural integrity and framework architecture of the CACPs. This study elucidates the indispensable role of pyridine carboxylic acids in the construction and stabilization of cluster-based framework materials, thereby advancing the frontier of research in metal cluster-based framework material synthesis.

Advancement of doxorubicin monitoring with a DNA fragmentation strategy for SYBR Green I-based aptamer biosensors

Luke Wei,Yuxin Hu,Jiamin Wu,Jing Mao,Tianqin Yin,Jieqiong Qiu

Doxorubicin (DOX), a widely utilized chemotherapeutic antibiotic, increasingly threatens environmental and public health due to its persistence in marine ecosystems, bioaccumulation in organisms, and contamination of agricultural systems. In response to the urgent demand for effective DOX detection, we have developed a label-free fluorescent aptamer biosensor (FAB) that employs SYBR Green I (SG I)-mediated signal amplification. A key innovation in this design is the strategic fragmentation of the 41-mer complementary strand (CP) of the DOX aptamer into three 11-mer fragments. It significantly mitigates nonspecific SG I binding interference caused by the longer ssDNA CP, enhancing the fluorescence signal ratio (F dsDNA /F ssDNA ) from 4.6-fold to 12.6-fold. Upon DOX introduction, competitive binding between DOX, SG I, and DNA further suppresses background noise, yielding an exceptional fluorescence quenching ratio (F 0 /F) of 52.8. The optimized FAB demonstrates ultra-sensitive DOX detection, with a detection limit of 1.41 nM and a linear response between 1.41 nM and 300 nM. The recoveries of DOX spiked into pond water and drinking water samples ranged from 95.1% to 101.8%, with relative standard deviations (RSDs) below 3.46%, demonstrating negligible matrix effects and confirming the FAB method’s robustness for environmental monitoring applications. This strategy synergistically enhances signal transduction efficiency, and strengthens detection reliability, all while maintaining operational simplicity. As a result, this FAB for DOX detection holds transformative potential, poised to revolutionize biosensor technology in the fields of food safety, environmental monitoring, and biosecurity.
