A Hydrophilic/Hydrophobic Janus Membrane Used as Wound Dressings with Enhanced Antibacterial Properties

Song Mengqi,Zhao Qian,Wang Xiuting,Shi Chuan,Hu Xiao,Li Jiwei

In this study, an asymmetric wettable composite wound dressing consists of hydrophobic polyvinylidene fluoride (PVDF) outer layer and a hydrophilic polydopamine (PDA) coated Ag nanoparticles (Ag NPs)/PVDF layer (PVDF-Ag-PDA) was successfully prepared through electrospinning and PDA coating. The results show that the contact angle of the PVDF surface is 132.8°, while the PDA-coated surface is nearly 0°, indicating an asymmetric wettability. The highly hydrophilic PVDF-Ag-PDA layer can significantly reduce bacterial growth, while the PVDF layer with a small pore size and high hydrophobicity can prevent bacterial adherence. In addition, cytotoxic assays showed that PVDF/PVDF-Ag-PDA present a high cell viability percentage (above 80 %), which is attributed to that the thin layer of PDA can alleviate the cytotoxicity of Ag NPs. Therefore, the Janus wound dressing noted as PVDF/PVDF-Ag-PDA has the potential for practical applications with the advantages mentioned above.

ATP and ssDNA aptamer-mediated peroxidase-like activity of rGO@PDA@CeO2 nanozyme: Exosomal proteins profiling and detection at physiological pH for colorimetric sensor

Jingjing Kuang,Shengli Ruan,Yangkun Sun,Zeyu Wu,Jiayao Xu,Ting Zhang,Xiaofan You,Shiman Yang,Min Zhang,Hongyang Zhang,Ping Hu

Exosomes are extracellular vesicles that inherit molecular markers from their parent cells for the transfer and delivery of membrane and cytosolic molecules between cells. Exosome detection and protein profiling provide noninvasive access to disease diagnosis and treatment. The nanozyme designed in this paper contains polydopamine-functionalization of graphene oxide (rGO@PDA) units that can adsorb ssDNA aptamers and CeO 2 metal oxide nanosheet units with peroxidase activity. As auxiliary regulator, adenosine triphosphate (ATP) enhances peroxidase activity of rGO@PDA@CeO 2 under a wide range of pH (4–7.4), which breaks through the limitation that traditional nanozymes can only exhibit peroxidase activity under acidic conditions. The ssDNA aptamers, which are recognition probes of exosome, can be adsorbed on the surface of rGO@PDA by aromatic stacking, competing with ATP molecules. Thus, the ssDNA aptamer acts as the switch for colorimetric sensor to flexibly regulate the peroxidase activity of the rGO@PDA@CeO 2 under physiological conditions , and realizes sensitive detection of exosomes. This assay achieves a detection linear range of 0.46 × 10 7 –10 8 particle/mL and a limit down to 3.81 × 10 5 particles/mL. In addition, this aptasensor sensitively profiles six exosomal protein across five cell types and clinical samples, suggesting their potential in clinic diagnosis.

Photothermal-Controlled Release of IL-4 in IL-4/PDA-Immobilized Black Titanium Dioxide (TiO2) Nanotubes Surface to Enhance Osseointegration: An In Vivo Study

Bo Chen,Yu Liang,Yunjia Song,Yunkai Liang,Jian Jiao,Hong Bai,Ying Li

Host immune response has gradually been accepted as a critical factor in achieving successful implant osseointegration. The aim of this study is to create a favorable immune microenvironment by the dominant release of IL-4 during the initial few days after implant insertion to mitigate early inflammatory reactions and facilitate osseointegration. Herein, the B-TNT/PDA/IL-4 substrate was established by immobilizing an interleukin-4 (IL-4)/polydopamine (PDA) coating on a black TiO2nanotube (B-TNT) surface, achieving on-demand IL-4 release under near infrared (NIR) irradiation. Gene Ontology (GO) enrichment analyses based on high-throughput DNA microarray data revealed that IL-4 addition inhibited osteoclast differentiation and function. Animal experiment results suggested that the B-TNT/PDA/IL-4+Laser substrate induced the least inflammatory, tartrate-resistant acid phosphatase, inducible nitric oxide synthase and the most CD163 positive cells, compared to the Ti group at 7 days post-implantation. In addition, 28 days post-implantation, micro-computed tomography results showed the highest bone volume/total volume, trabecular thickness, trabecular number and the lowest trabecular separation, while Hematoxylin-eosin and Masson-trichrome staining revealed the largest amount of new bone formation for the B-TNT/PDA/IL-4+Laser group. This study revealed the osteoimmunoregulatory function of the novel B-TNT/PDA/IL-4 surface by photothermal release of IL-4 at an early period post-implantation, thus paving a new way for dental implant surface modification. Keywords:photothermal;controlled release;IL-4;PDA;TiO2nanotubes;osseointegration;near-infrared laser;titanium

Multifunctional stable PDA/RGO/MOFs&SiO2-COOH membrane with excellent flux and anti-fouling performance for the separation of organic dye and oil/water

Yucheng Liu,Zhi Huang,Wei He,Mingyan Chen,Wenwen Tu,Meng Zhu,Dong Gan,Shumeng Liu

ABSTRACT In this work, bimetallic MIL53(Fe-Co) nanorod was modified by SiO 2 -COOH nanoparticles to obtain a novel MOF with low aggregation, and then intercalated into the reduced graphene oxide (RGO) layer crosslinked by polydopamine (PDA) to form a super-hydrophilic membrane with selective adsorption. The composite material was characterized by various analytical techniques. Owing to the low agglomeration of bimetallic MIL53(Fe-Co) and improved layer spacing of RGO , the stable composite membrane exhibited a water contact angle of approximate 0°, with the dye solution flux of 1721.46 L·m −2 ·h −1 ·bar −1 for methylene blue (MB) and 1834.88 L·m −2 ·h −1 ·bar −1 for malachite green (MG), the dye solution flux was increased by 31 and 33 times compared with M0, respectively. Meanwhile, the rejection rates of MB and MG were 99.46% and 99.4% with an excellent tolerance to a wide range of pH. Furthermore, the composite membrane shows a huge adsorption capacity in treating dye solution with high concentration and large volume for reusability due to the improvement of hydrophilicity. It also performed excellently in the field of separating oil/water emulsion with the flux of 539.79 L·m −2 ·h −1 ·bar −1 and rejection of 98.12%. In conclusion, the composite membrane with extraordinary hydrophilicity, strong adsorption capacity, and superior anti-fouling performance with low costs endows possibility of massive manufacture in commercial production for wastewater separation and purifications.

Polydopamine intimate contacted two-dimensional/two-dimensional ultrathin nylon basement membrane supported RGO/PDA/MXene composite material for oil-water separation and dye removal

Xiaofang Feng,Zongxue Yu,Runxuan Long,Yuxi Sun,Ming Wang,Xiuhui Li,Guangyong Zeng

Titanium carbide (MXene) sheet membranes have become attractive candidates in terms of solvent nanofiltration performance. Whereas, the low contaminant removal ability limited the practical application of MXene membranes in efficient separation. Here, the reduced graphene oxide (RGO)/polydopamine (PDA)/titanium carbide (MXene) composites were prepared by dopamine modification method. Then, the RGO/PDA/MXene composites were suction filtrated on a nylon membrane to construct the two-dimensional-two-dimensional (2D-2D) laminated composite membrane. The effects of MXene content on the surface hydrophilicity, dye retention and oil-water separation properties of the modified membrane were systematically studied. Among them, the RPM-8 membrane exhibited ultra-high separation effect (>96%) and excellent throughput (>200 L/m 2 ·h) for Methylene Blue (MB), Methyl Orange (MO), Methyl Red (MR), Congo Red (CO) and Evans Blue (EB). At the same time, the modified membrane shows very high oil-water separation effect on the emulsion (>97%). In addition, long-term cycle experiments have also revealed the stability of RGO/PDA/MXene composite membranes. The above results further illustrated the importance of the regular straight rigid channels of the composite membrane in separation performance. Therefore, RGO/PDA/MXene composite membrane has tremendous potential in practical applications.

A novel analytical method based on HPLC-PDA coupled post-column derivatization to evaluate the ability to inhibit tyrosine nitration in lotus leaf extracts

Yin Qian,Xi Chen,Jin Qi,Xuming Liu

Protein tyrosine nitration plays a key role in many inflammatory and cardiovascular diseases and diabetes. Many natural products are used to treat these diseases through their ability to potentially interfere this reaction. Here, we describe a novel method to provide active fingerprinting of inhibition of tyrosine nitration by natural products based on post-column tyrosine nitration reaction analysis using high-performance liquid chromatography coupled to a photometric diode array. Results indicated that lotus leaf extracts exhibited obvious inhibitory activity against tyrosine nitration by peroxynitrite, and that chemical and active fingerprints were simultaneously established, with the active fingerprints indicating the active compounds of the lotus leaves. Additionally, flavonoids were screened as the principal active compounds involved in inhibiting tyrosine nitration in the lotus leaf extracts, with quercetin-3-O-glucuronide and quercetin-3-O-glucoside exhibiting the greatest contributions. Moreover, our results suggested that lotus leaves from three regions (Nanjing, Suzhou, and Hangzhou) exhibited the best inhibitory activity. These findings indicated the usefulness of this method for screening active compounds involved in inhibiting protein tyrosine nitration, and that similar strategies can likely be applied to evaluate the inhibitory activity against tyrosine nitration of other natural products.

In-Situ construction of Lithium-Ion channel within PVDF/VNs@PDA composite separators for remarkable enhanced cycle stability and safety for lithium batteries (LIBs)

Ziqiang Yang,Yingdong Yang,Bin Yang,Mengning Luo,Yue Zhang,Jibin Miao,Shijun Kang,Zhengzhi Zheng,Jiasheng Qian,Ru Xia,Yuchao Ke,Yang Pan

In this work, a non-solvent induced phase separation (NIPS) technique was used to prepare polyvinylidene difluoride (PVDF) lithium battery (LIB) separators. Firstly, vermiculite nanosheets (VNs) coated by polydopamine (PDA), i.e., VNs@PDA, was ultrasonically dispersed in DMAc and PVDF was solution-blended with VNs@PDA with final porous composite films obtained after the extraction of DMAc. Effects of the presence of VNs@PDA on both structure and properties of PVDF films were intensively studied, and the SEM observations further suggested that the addition of VNs@PDA heavily affected the thermodynamic parameters between the polymer and solvent, causing a remarkable enhancement on the thermodynamic instability of the PVDF solution system. The polymer–solvent phase separation by the addition of less nonsolvent tended to induce the formation of long finger-like pores. An incorporation of 7.0 wt% VNs@PDA resulted in an increase in ionic conductivity of the composite membrane from 0.320 mS·cm −1 to 1.515 mS·cm −1 via AC impedance tests. The incorporation of VNs@PDA not only enhanced the hydrophilicity of the composite films (as judged by water contact angle dropping from 112.7° to 74.2°), but also significantly improved both battery performance and mechanical properties. The present study provides a facile way of fabricating LIB separators with sufficiently high safety and superior performance.

Sequential grafting of PEI on PDA-anchored graphene oxide for enhanced xylose dehydrogenase Catalysis: Positively charged surface interfaces and mechanistic insights via molecular simulation

Yifan Gui,Gen Lu,Chengyu Sun,Ya Wu,Xu Wang,Yuanmin Zhu,Wenwen Jin,Longjiang Yu

In this work, graphene oxide (GO) composites, sequentially enhanced with polydopamine (PDA) and polyethyleneimine (PEI), were developed for the innovative immobilization of xylose dehydrogenase (XylB). These composites were designed to achieve positively charged surfaces, aiming to explore the potential interactions between the enzyme and the modified composites. The focus was particularly on determining whether these modifications could improve the enzyme's catalytic performance and stability. Characterization was performed using Fourier Transform Infrared Spectroscopy (FT-IR), X-ray Photoelectron Spectroscopy (XPS), and Scanning Electron Microscopy (SEM). Molecular dynamics simulations elucidated the structure-function correlations of immobilized XylB, revealing significant enhancements in thermal stability and catalytic performance compared to its free counterpart. Notably, the immobilized enzyme retained over 85 % of its activity after four consecutive usage cycles and exhibited double the residual activity compared to free XylB after 30 days of storage at 4 °C. Additionally, the immobilized system achieved a 1.4-fold increase in xylonic acid yield relative to the free system. This approach not only highlights the potential of advanced material composites in industrial enzyme applications but also sets a foundation for further research into sustainable biocatalyst technologies.

Combination of 3-methyladenine therapy and Asn-Gly-Arg (NGR)-modified mesoporous silica nanoparticles loaded with temozolomide for glioma therapy in vitro

Peng Zhang,Menghuan Tang,Qin Huang,Guanjian Zhao,Ning Huang,Xiang Zhang,Ying Tan,Yuan Cheng

Mesoporous silica nanoparticles (MSNPs) of a small diameter were loaded with the anticancer drug temozolomide (TMZ), coated with polydopamine (PDA), and conjugated with Asn-Gly-Arg (NGR) for use in the treatment of glioma. The accumulation of NGR-MSNPs in C6 cells was shown to be higher than that of unmodified MSNPs. Anticancer drugs can cause autophagy in tumor cells, whereas autophagy inhibitors can block this reaction and enhance the therapeutic effect of the drugs. In this study, we demonstrated that MSNP-TMZ-PDA-NGR had stronger autophagy- and apoptosis-inducing effects in C6 cells than TMZ alone, and its anticancer effect was further enhanced when combined with autophagy inhibition. These results demonstrate that the combination of targeting vehicles and autophagy inhibitors may have research value in the treatment of gliomas.

Novel phase-change PDA-Ni@GNS/CNF-C/SA/PEG composites with ultrahigh shape stability and latent heat as thermal management material for microelectronic devices

Xiaohong Wang,Bin Yang,Yue Zhang,Yuqing Yang,Huijie Lu,Hao Wang,Ziyi Wang,Jiasheng Qian,Ru Xia,Ning Jia,Yuchao Ke

Phase-change materials (PCMs) have become a hot spot in the development of modern thermal management materials because of their absorbing and releasing heat while keeping the temperature constant during phase change process. However, their relatively low thermal conductivity (TC) considerably restricts their further application. In this work, graphene nanosheets (GNS) were initially nickel-plated to increase the contact area between adjacent GNS (in the form of Ni@GNS) by constructing a “point-surface” structure. Then, the prepared Ni@GNS was coated with polydopamine (PDA) to prepare PDA-Ni@GNS, which helped to effectively reduce the interface thermal resistance (ITR) between the filler and the matrix, and also made the prepared filler more easily dispersed in the polymer matrix. By constructing well-defined three-dimensional (3D) thermally conductive pathways in the matrix, the PCMs exhibited excellent TC and effectively prevented the leakage of polyethylene glycol (PEG) even at low filler loading. The results of thermogravimetry (TG), differential scanning calorimeter (DSC), and cyclic DSC tests jointly showed that the PDA-Ni@GNS/CNF-C/SA/PEG PCMs displayed excellent thermal storage properties, high latent heat as well as good cyclic thermal stability. The present study provides a facile way of preparing PEG-based PCMs, which are applicable in thermal management area.

Identification and determination of major constituents in a traditional Chinese medicine compound recipe Xiongdankaiming tablet using HPLC-PDA/ESI-MS n and HPLC-UV/ELSD

Wang Shu-fang,Leng Jing,Xu Yi-min,Feng Mei-ling

Xiongdankaiming tablet (XDKMT), a well-known compound in traditional Chinese medicine, is widely used for the treatment of acute iridocyclitis and primary open-angle glaucoma. In this paper, accurate and reliable methods were developed for the identification of 20 constituents using high-performance liquid chromatography with photodiode array and electron spray ionization-mass spectrometry (HPLC-PDA/ESI-MS n ), and determination of nine of the constituents (chlorogenic acid, gentiopicroside, isochlorogenic acid B, diosmetin-7- O -β- d -glucopyranoside, apigenin, diosmetin, tauroursodeoxycholic acid, acacetin, and taurochenodeoxycholic acid) was developed using HPLC with ultraviolet absorption detector and evaporative light scattering detector (HPLC-UV/ELSD) for the first time. The best results were obtained on a Zorbax SB-C18 column with gradient elution using water (0.1% formic acid) (A) and methanol (0.1% formic acid) (B) at a flow rate of 0.7 ml/min. Tauroursodeoxycholic acid and taurochenodeoxycholic acid, owing to their low UV absorption, were detected by ELSD. The other seven compounds were analyzed by HPLC-UV with variable wavelengths. The calibration curves of all nine constituents showed good linear regression ( R 2 >0.9996) within the linearity ranges. The limits of detection and quantification were in the ranges of 0.0460–9.90 μg/ml and 0.115–24.8 μg/ml, respectively. The accuracy, in terms of recovery, varied from 95.3% to 104.9% with relative standard deviations (RSDs) less than 4.4%. Precision (with the intra- and inter-day variations less than 4.4%) was also suitable for its intended use. The developed method was successfully applied for the analysis of major components in XDKMT, which provides an appropriate method for the quality control of XDKMT.

DAHA/CMCS/Ag@PDA dual-dynamic crosslinked injectable hydrogels with NIR-II photothermal synergistic antibacterial and antioxidant properties for accelerated healing of bacterial-infected wounds

Junqi Zhao,Zicheng Chen,Jiahao Huang,Jingxian Zhang,Langhuan Huang,Shaozao Tan,Yongqiang Xiong

Bacterial infections and excessive oxidative stress severely impede the wound healing process, and the development of multifunctional wound dressings with antibacterial, antioxidant, and biosafety properties is of great clinical significance. In this study, DAHA/CMCS/Ag@PDA (DCA) injectable hydrogels were successfully prepared by dual-dynamic crosslinking of Schiff base and hydrogen bonds. Dopamine graft-modified oxidized hyaluronic acid (DAHA) and carboxymethyl chitosan (CMCS) constructed a dynamic and reversible cross-linking network of the hydrogel, which endowed the hydrogel with excellent injectable, self-healing, shape-adaptive, and high adhesion properties. Meanwhile, DAHA and Ag@PDA, which are rich in catechol groups, provided the hydrogels with highly efficient synergistic antioxidant properties, which could effectively scavenge various free radicals. In addition, the loading of Ag@PDA endowed the hydrogel with excellent Ag + release and NIR-II photothermal conversion properties, and DCA2 could realize efficient killing (99.6%) of S. aureus and E. coli through the synergistic action of Ag  +  slow release and NIR-II photothermal, which could effectively promote the healing of bacterial-infected wounds. In conclusion, this study provides a new design strategy for hydrogel dressings for the treatment of bacterial-infected wound healing through the multifunctional integration of NIR-II photothermal synergistic antibacterial and antioxidant properties.
