Protection and embedding of hydrophobic bioactive compounds using protein hydrogels are emerging focus during the latest decade. In present study, we fabricated the porcine plasma protein (PPP) cold-set gel by microbial transglutaminase (MTGase) and glucono-δ-lactone (GDL) as coupling precursors. As a result, the embedding, protection and controlled-release effect of the gel on vulnerable hydrophobic bioactive components (quercetin (Que) as representative) with proposed molecular mechanisms were investigated in detail. The results showed that high concentration of Que (5 mmol/L) could be loaded with PPP cold-set gel and embedding efficiency (EE) was over 98%. Compared with free Que, the embedded one exhibited significantly higher thermostability , photochemical stability and storage stability ( P < 0.05). In addition, the gel loaded with 5 mmol L −1 of Que had higher swelling potency under gastric (low pH) media and controlled release performance following simulated intestinal digestive pathway. Water holding capacity (WHC) implied that free water molecules played less role in the retention ability of Que in fabricated gel network. Spectral-assisted structural characterization proved that Que was efficiently embedded mainly by generating PPP-Que complex through hydrogen bond and van der Waals force, and the binding site was mainly near Trp residue. This work gave novel insight into the potential use of PPP cold-set gel as an excellent carrier towards protection and selective delivery of vulnerable small hydrophobic nutraceutical compounds.

Herbicides pollution is currently a big threat to the environment due to their widespread use in the agricultural field. Herein, we proposed the creation of bifunctional porous polyethyleneimine-grafted lignin microspheres (PLMs) by grafting polyethyleneimine onto lignin via an inverse suspension polymerization method by using polyethylene glycol as a porogen. The porous structure and abundant functional groups of PLMs allowed it to efficiently capture the representative herbicide, 2,4-dichlorophenoxyacetic acid (2,4-D), steadily over a wide pH range of 4–10. The maximum adsorption capacity was 909.09 mg/g at 45 °C that was much higher than the reported activated carbon (515.46 mg/g) and metal-organic framework (MIL-53, 556 mg/g). The adsorption process followed the Langmuir isotherm model and pseudo-second-order kinetic model, respectively. More importantly, the 2,4-D-loaded PLMs showed a distinct sustained release behavior as the environmental pH values varied. Under acidic (pH = 2) or alkaline (pH = 11) conditions, 57% or 88% of 2,4-D was released after 72 h, respectively, while only 10% of 2,4-D was released even after 96 h under pH = 7. FTIR and XPS analysis indicated that the adsorption mechanism was mainly involved with the electrostatic attraction, hydrogen bonding, π-π stacking and van del Waals forces between the PLMs and 2,4-D molecules. This work proposed an environmentally-friendly biomass-based bifunctional microspheres that could be potentially applied in the management of herbicide pollution through adsorption and controlled release formulations.

With the increasing demand for treating cancer and cardiovascular diseases in recent years, drug delivery therapy using intelligent nanocarriers has been investigated intensely. Usually, hydrophobic dyes were used as drug models for the development of drug-release carriers. In this study, light and temperature dual-responsive micelles were designed and fabricated, and the controlled release property was investigated using the hydrophobic fluorescent dye coumarin 102 as the model. A carbamate-containing spiropyran methacrylate monomer (SPMA) was adopted as the photosensitive unit because it is more conducive to stabilize the micelle structure compared to traditional spiropyran. Firstly, a light and temperature dual-responsive copolymer P[DMA 50 - b -(SPMA 20 - co -NIPAAm 17 )] was synthesized through reversible addition-fragmentation chain transfer (RAFT) polymerization. Then, the copolymer was self-assembled into micelles with coumarin 102 encapsulated in the hydrophobic core above the lower critical solution temperature (LCST) and under visible light (Vis) irradiation. When the visible light was switched to ultraviolet (UV) radiation, the micelles' permeability increased as a result of the change from closed, three-dimensional spiropyrans (SP) to open, planar merocyanines (MC), resulting in the release rate of 51% for coumarin 102 in 15 min with “low” release capability. Lowering the temperature to room temperature (RT), the hydrophobic nuclei were partially destructed as a result of the changing of PNIPAAm segment from hydrophobicity to hydrophilicity. Meanwhile, the release rate was observed to be 77% in 15 min showing “medium” release capability. The micelles were almost wholly disintegrated under the dual stimulation of both UV irradiation and RT, achieving the release rate of 96% within the same time exhibiting “high ” release capability. This study offered an approach for the construction of controllable drug release carriers.

Active packaging with antimicrobial functions to improve the quality and extend the shelf life of food products has gained great interest. Because commercial plastic packaging materials are not biodegradable and cause great environmental problems, plant-derived natural materials have been widely studied for the application of biodegradable packaging materials. Herein, we reported a study of essential oils (EOs)-loaded zein film. Cinnamaldehyde (CIN), carvacrol, and eugenol were added to equip the films with antimicrobial effects, while polyethylene glycol (PEG) and oleic acid (OA) were selected for the improvements of mechanical properties. The results showed that PEG efficiently improves the tensile strength and elongation (%E) of zein films compared to OA, although PEG induced weaker water barrier properties of the films than OA. FTIR spectra confirmed the formation of the hydrogen bonds between zein and PEG/OA. The EO-embedded zein film showed better antimicrobial effects than EO themselves. CIN-embedded films showed the highest antimicrobial effect among the three EOs. The sizes of the inhibition zones against Staphylococcus aureus of PEG-added zein films with 1%, 3%, and 5% CIN were 5.67, 12.67, and 16.67 mm, which were larger than that of pure CIN, with the sizes of 0.00, 3.00, and 4.67 mm, respectively. The developed films demonstrate a gradual release of EOs and show antimicrobial effects up to 96 h, indicating their high potential for the applications as active food packaging.

Phenolipids such as hydroxytyrosol-SCFA acyl esters (HTy-SEs) and tyrosol-SCFA acyl esters (TYr-SEs) with various alkyl chains lengths (C1–C4) and different isomers (branched-chain and straight-chain) were successfully synthesized. All esters were hydrolyzed by pancreatic lipase to produce polyphenols (HTy and TYr) and SCFAs (iso-butyric acid, acetic acid, propionic acid, and n-butyric acid). Moreover, HTy-SEs (and TYr-SEs) could also be hydrolyzed to free HTy (and TYr) and SCFAs by gut microbiota and Lactobacillus from mice feces. Especially, the hydrolysis rates showed positive correlation with the carbon skeleton length, and the hydrolysis degree (DH) of ester with a branched-chain fatty acid was weaker than that of ester with a straight-chain fatty acid. Besides, the DH values of TYr -SEs were significantly higher than those of HTy-SEs. Therefore, through regulating the structures of polyphenols, carbon skeleton lengths, and isomers, controlled-release of polyphenols and SCFAs from phenolipids will be easily achieved.

Targeted and controlled nitric oxide (NO) release is critical due to an extremely short half life and low bioavailability for treating cardiovascular diseases. To address this challenge, various sustained-release precursors and NO donors activated by light, enzyme, or pH are developed. However, their efficacy is limited by the deep location and rapid blood flow of the heart. Herein, a platelet membrane-coated nanoparticle (B-P@PLT) is designed with a polymeric core loaded with BNN6, an ultrasound-responsive NO donor, for the targeted treatment of myocardial ischemia reperfusion injury (MIRI). B-P@PLT can specifically target the ischemic myocardium and release NO during ultrasound (US) irradiation, thereby increasing the local concentration of NO. B-P@PLT + US shows promising results in promoting angiogenesis, reducing reactive oxygen species production, protecting cardiomyocytes both in vitro and in vivo, and ultimately decreasing cardiac injury and improving heart function in MIRI mice. These findings demonstrate a simple and noninvasive strategy for targeted delivery and controlled release of NO, highlighting its potential therapeutic application in MIRI.

Dihydromyricetin (DHM) is an important natural flavonoid . However, most of DHM preparations have shown shortcomings such as low drug loading, poor drug stability, and/or large fluctuations in blood concentration. This study aimed to develop a gastric floating tablet with a double-layered structure for zero-order controlled release of DHM ( [email protected] ). The final product [email protected] showed a high average cumulative drug release at 24 h that best fit the zero-order model, and had a good floating ability in the stomach of the rabbit with a gastric retention time of over 24 h. The FTIR , DSC , and XRPD analyses indicated the good compatibility among the drug and the excipients in [email protected] The pharmacokinetic study revealed that [email protected] could prolong the retention time of DHM, reduce the fluctuation of blood drug concentration, and enhance the bioavailability of DHM. The pharmacodynamic studies demonstrated that [email protected] had a potent and long-term therapeutic effect on systemic inflammation in rabbits. Therefore, [email protected] had the potential to serve as a promising anti-inflammatory agent and may develop into a once-a-day preparation, which was favorable to maintain a steady blood drug concentration and a long-term drug efficacy. Our research provided a promising development strategy for DHM and other natural products with a similar structure to DHM for improving their bioavailability and therapeutic effect.

A novel series of quadruple responsive copolymers, poly(ethylene glycol)- ss -[poly(dimethylaminoethyl methacrylate)- co -poly(2-nitrobenzyl methacrylate)] [PEG- ss -(PDMAEMA- co -PNBM)], were synthesized via atom transfer radical polymerization mediated by home-made PEG-based macro-initiator labeled with disulfides. The obtained copolymers could self-assemble in aqueous solution forming micelles with the disulfide bridge linking the hydrophilic coronas (PEG) and the hydrophobic cores (PDMAEMA- co -PNBM). Investigation on the resulted micelles indicated that the micelles could respond to various stimuli, that is, temperature, pH, the presence of dithiothreitol (DTT), and UV irradiation. Moreover, the responsive behavior of the micelles depends on the type of stimuli, that is, temperature change causes size change of the micelles, while UV irradiation leads to dissolution of the self-assembled structures. Such stimulus-dependent responsive behavior could be applied in smart materials that deal with multi-tasks or in the construction of complex logic gate. The potential application of the multi-responsive micelles in cargo release system was also evaluated using Nile Red (NR) as model molecule. © 2018 Wiley Periodicals, Inc. J. Appl. Polym. Mesocellular silica foam (MCF) grafted with thermo-responsive and pH-responsive copolymer poly( N -isopropylacrylamide- co -acrylic acid), was prepared by precipitation polymerization using supercritical carbon dioxide (scCO 2 ) as entrainer and solvent. Vinyl-functionalized MCF was first synthesized through post-synthesis via using vinyltriethoxysilane as a coupling agent for surface modification. Entrapped monomers of N-isopropylacrylamide (NIPA) and acrylic acid (AA) were then introduced and assembled onto the vinyl-modified MCF with the aid of scCO 2 . The grafting copolymer performed here as an on/off switch in the release of 5-fluorouracil, as confirmed by using UV-visible absorption spectroscopy. The grafted organic content and drug loading efficiency were 65.2 wt% and 11.5 wt%, respectively. The resultant grafting composite showed potential for application in drug delivery and exhibited thermo- and pH-responsiveness to external environmental changes.

Fucoxanthin (FUCO) is a marine xanthophyll carotenoid with widespread potential physiological functions. However, poor water solubility and chemical instability limit FUCO's processing adaptability and bioavailability. In this study, chitosan surface-modified bovine serum albumin (BSA)-oleic acid (OA) self-assembled complexes were fabricated to improve the thermal stability as well as the controlled release and bioaccessibility of FUCO during gastrointestinal digestion. The results showed that glucose-grafted chitosan (GGC) could surface modify BSA-OA complexes by non-covalent affinity adhesion, resulting in a stable hydrophilic dispersion system with a polydispersity index <0.25 and a Zeta potential > −20 mV. Fourier transform infrared spectroscopy and transmission electron microscope observations revealed that GGC covered the surface of BSA-OA particles through hydrogen bonding, hydrophobic and electrostatic interactions. The GGC-BSA-OA delivery system captured FUCO with >88.00% encapsulation efficiency. Additionally, embedding FUCO in the GGC-modified BSA-OA complex improved its thermal stability. In vitro, gastrointestinal digestion simulations showed that the GGC-modified protein shell had a controlled-release effect on FUCO, which prevented acidic pH from destroying FUCO due to premature release during gastric digestion and released it during intestinal digestion. As expected, GGC-BSA-OA, an encapsulated carrier, dramatically improved the FUCO's digestive stability (retention rate >74.00%), bioaccessibility (>54.42%), and micellization rate (>70.00%) in simulated gastrointestinal digestion. These findings suggested that GGC-BSA-OA as a delivery vehicle could effectively improve FUCO's thermal processing stability and absorbability.

Smart drug carriers with intrinsic photoluminescence (PL) tracking of controllably released drugs are of great significance to indicate in real-time where, when and how the drug was released. Herein, we present a novel multifunctional poly(N-isopropylacrylamide)–Mn–ZnS (PMZS) nanocomposite for thermo- and pH-controlled release of doxorubicin (Dox) and real-time tracking of the released Dox in a PL-enhanced manner. The simple coupling of Mn–ZnS quantum dots and poly(N-isopropylacrylamide) resulted in the unique thermo- and pH-responsive PL of PMZS. There was a much higher PL intensity at 37 °C than 25 °C and a much higher PL intensity at weakly acidic pH than at neutral pH, which is promising for PL imaging in tumor microenvironments. The PL of the PMZS composite was greatly quenched upon loading of Dox (the loading content and encapsulation efficiency of PMZS composites toward Dox were 15.2% and 84.1%, respectively), but was gradually restored with the controllable release of Dox, and the restored PL intensity was proportional to the cumulative amount of Dox released. The versatility of the PMZS composite as an effective Dox carrier and real-time PL-enhanced tracking of the controllably released Dox were further proved by cell viability assays and in vitro cell imaging experiments. The simple PMZS nanocomposite was easily-synthesized, biocompatible, and promising for PL tracking of when, where and how Dox was released.

A novel thermal-responsive and magnetic molecularly imprinted polymers (TMMIPs) were prepared based on mag-yeast via a mild and effective method; the obtained TMMIPs were used for selective adsorption and release of tetracycline from aqueous solution. Imprinted polymerization directly occured on the surface of microspheres, and core–shell imprinted polymers were developed via in situ precipitation polymerization. The properties of obtained TMMIPs were characterized by scanning electron and transmission electron microscopy, X-ray diffraction, Fourier transform infrared spectra, thermo-gravimetric analysis and so on. The adsorption equilibrium data were well described by Langmuir isotherm model. Kinetic experiments showed the adsorption process reached the equilibrium within 60 min, and the pseudo-second-order kinetic model was used to fit the adsorption data well. TMMIPs exhibited magnetic sensitivity, magnetic stability and thermal stability. Reversible recognition and release of template molecule were realized by changing environmental temperatures. Several other antibiotics were selected as model analytes to evaluate the selective recognition performance of TMMIPs. The TMMIPs have good temperature response, selectivity and reusability, making them possible in applying for antibiotics adsorption and controlled release.
