Pseudo-interpenetrating network viscoelastic surfactant fracturing fluid formed by surface-modified cellulose nanofibril and wormlike micelles

Yang Yang,Hao Zhang,Hao Wang,Jiang Zhang,Yunfei Guo,Bing Wei,Yangbing Wen

Nano-material enhanced viscoelastic surfactant (VES) systems have attracted considerable attention as clean fracturing fluids due to their improved rheological properties compared to conventional VES systems. In this study, a pseudo-interpenetrating network VES (PINVES) fracturing fluid, which was composed of sodium oleate (NaOA), potassium chloride (KCl) and surface-modified cellulose nanofibril (SMCNF), was proposed. The rheological behavior of the PINVES fracturing fluid and the interaction mechanism between SMCNF and wormlike micelles (WLMs) were comprehensively studied. The zero-shear viscosity of PINVES increased with appropriate SMCNF addition but decreased with excess SMCNF addition. Meanwhile, the typical viscosity-flat of WLMs gradually disappeared when the concentration of SMCNF exceeded the optimum value. The optimum value was closely related to the functional groups on the surface of SMCNF. The SMCNF physically crosslinked the WLMs through hydrogen bonding but the sulfonic groups and hydrophobic groups on the surface of SMCNF destroyed the network of WLMs at relatively high SMCNF concentration. The network formed by SMCNF dominated the rheological behavior of the PINVES fracturing fluid simultaneously. In addition, due to the competition between the network of WLMs and the network of SMCNF, the PINVES fracturing fluid exhibited higher concentration dependence of surfactant in rheological behavior than conventional VES fracturing fluid. The SMCNF network also endowed the PINVES with better temperature-resistance. Furthermore, the relaxation time and elasticity of PINVES fracturing fluid increased with the increase in SMCNF concentration. The static proppant settling tests demonstrated that the addition of SMCNF improved the sand-carrying capability of PINVES fracturing fluid. The results of the study could provide some enlightenments on the application of SMCNFs in reservoir stimulation.

The effect of topology of PEG chain on the stability of micelles in brine and serum

Bin Wu,Wei-Qiang Huang,Xuan Nie,Ze Zhang,Guang Chen,Hai-Li Wang,Fei Wang,Shen-Gang Ding,Zong-Yao Hao,Ye-Zi You

Polymeric micelles are promising vehicles for drug delivery these years. The stability of micelles in blood system is very important for the encapsulated therapeutic agents reaching the targeting area. However, generally, micelles are not stable in the presence of proteins and ions after being injected intravenously. The topology of polymer chain on the surface of micelles plays a key role on the stability of micelles. Here we have prepared flower micelles and the traditional micelles via self-assembly of triblock and diblock copolymer of polyethylene glycol (PEG) and poly(ε-caprolactone) (PCL) in water. The special PEG shell of flower micelles prevents micelles from ions and proteins, which endows the micelles with the ability to resist aggregation. The experiment results show that flower micelles are much more stable in the presence of serum and salt than traditional micelles, which indicates these micelles would be promising in drug delivery in the future.

Wen Li,Danxiu Dong,Jinxuan Zou,Huiqiang Zhao,Weilu Zhang

To broaden the applications of alkyl gadolinium (Gd) compounds, a new enhanced hydrophilic soft congeries were fabricated with nonionic surfactant alcohol ethoxylate (AEO) when the micelles constructed in aqueous media. Polyamidoamine (PAMAM) dendrimer was also added to the micellar solution to improve their stability. It was found that the aggregates showed high nuclear magnetic resonance (NMR) contrast effect and hydrophilicity. Meanwhile, the release of gadolinium ions from the aggregates can be controlled by adjusting the solution pH, the temperature or the concentration PAMAM, respectively, suggested the great potential of the congeries as magnetic resonance imaging (MRI) contrast agent.

Preparation of camptothecin micelles self-assembled from disodium glycyrrhizin and tannic acid with enhanced antitumor activity

Qihong Zhang,Zongmiao Feng,Hui Wang,Chen Su,Zhaohui Lu,Jingbo Yu,Alexandr V. Dushkin,Weike Su

Natural compounds as carriers for hydrophobic drugs have been increasingly used in drug delivery systems . In this study, disodium glycyrrhizin (Na 2 GA), tannic acid (TA) and camptothecin (CPT) were firstly used to prepare the camptothecin solid dispersion (CPT SD). When dissolved in a solution medium, Na 2 GA self-assembled to form micelles and CPT was encapsulated in micelles, meanwhile, TA connected with Na 2 GA through hydrogen bonds to form a contract shell. The average diameter of the CPT-loaded micelles is 80  nm with the critical micellar concentration of 0.303 mg/mL, the zeta potential of −33 mV, the PDI of 0.25 and drug loading 6.22%. In vitro experiments confirmed that the drug-loaded micelles exhibited excellent stability and permeability in the intestinal environment. Furthermore, the formulation showed excellent anti-tumor activity in vitro and in vivo . These findings imply that this nano-micelles provide a more potential and efficacious oral drug formulation for chemotherapy.

Polymeric micelles amplify tumor oxidative stresses through combining PDT and glutathione depletion for synergistic cancer chemotherapy

Ting Su,Furong Cheng,Yuji Pu,Jun Cao,Shuibin Lin,Guizhi Zhu,Bin He

Cancer has been one of the major healthcare burdens, which demands innovative therapeutic strategies to improve the treatment outcomes. Combination therapy hold great potential to leverage multiple synergistic pathways to improve cancer treatment. Cancer cells often exhibit an increased generation of reactive oxygen species (ROS) and antioxidant species compared with normal cells, and the levels of these species can be further elevated by common therapeutic modalities such as photodynamic therapy (PDT) or chemotherapy. Taking advantage that cancer cells are vulnerable to further oxidative stress, we aim to design a drug delivery system by simultaneously increasing the cellular ROS level, reducing antioxidative capacity, and inducing anticancer chemotherapy in cancer cells. Here, we designed a star-shape polymer, PEG(- b -PCL-Ce6)- b -PBEMA, based on the Passerini three-component reaction, which can both enhance ROS generation during PDT and decrease the GSH level in cancer cells. The polycaprolactone conjugated with photosensitizer Ce6 served as hydrophobic segments to promote micelle formation, and Ce6 was used for PDT. The H 2 O 2 -labile group of arylboronic esters pendent on the third segment was designed for H 2 O 2 -induced quinone methide (QM) release for GSH depletion. We thoroughly investigated the spectral properties of blank micelle during its assembling process, ROS generation, and H 2 O 2 -induced QM release in vitro . Moreover, this polymeric micelle could successfully load hydrophobic anti-cancer drug Doxorubicin (DOX) and efficiently deliver DOX into cancer cells. The triple combination of ROS generation, GSH elimination, and chemotherapy dramatically improved antitumor efficiency relative to each of them alone in vitro and in vivo .

Co-delivery of doxorubicin and SIS3 by folate-targeted polymeric micelles for overcoming tumor multidrug resistance

Wang Shuanghu,Tan Xueying,Zhou Quan,Geng Peiwu,Wang Jinhui,Zou Ping,Deng Aiping,Hu Jingbo

Multidrug resistance (MDR) is considered as a critical limiting factor for the successful chemotherapy, which is mainly characterized by the overexpression of ATP-binding cassette (ABC) transporter ABCB1 or ABCG2. In this study, folate-targeted polymeric micellar carrier was successfully constructed to co-delivery of doxorubicin (DOX) and SIS3 (FA/DOX/SIS3 micelles), a specific Smad3 inhibitor which sensitizes ABCB1- and ABCG2-overexpressing cancer cells to chemotherapeutic agents. The ratio of DOX to SIS3 in polymeric micelles was determined based on the anti-tumor activity against resistant breast cells. In addition, FA/DOX/SIS3 micelles exhibited a much longer circulation time in blood and were preferentially accumulated in resistant tumor tissue. Pharmacodynamic studies showed that FA/DOX/SIS3 micelles possessed superior anti-tumor activity than other DOX-based treatments. Overall, FA/DOX/SIS3 micelles are a promising formulation for the synergistic treatment of drug-resistant tumor. Graphical abstract

Thermosensitive Micelles Gel to Deliver Quercetin Locally for Enhanced Antibreast Cancer Efficacy: An In Vitro Evaluation

Yanxue Sun,Yun Bai,Silu Liu,Shuxia Cui,Pengcheng Xu

Although quercetin is low cytotoxicity to normal human cells, quercetin is effective against the growth of some tumors. Given the poor blood stability in vivo, insolubility, low delivery efficiency, and poor medicinal properties of quercetin, we developed a local drug delivery system comprising quercetin core’s polymer micelles and F127 hydrogel stroma. In vitro evaluation revealed that quercetin core’s polymer micelles have excellent antitumor activity and could inhibit the multiplication of 4T1 breast cancer cells through the apoptosis pathway. Meanwhile, a rheological study revealed that the quercetin core’s micelles gel possessed excellent properties of hydrogel formation and injectability of liquid preparation as a local drug delivery system after the quercetin core’s polymer micelles were loaded into the F127 hydrogel stroma. Our study findings indicated that the drug stability and stable release capacity of quercetin were vastly improved with the composite formulation of the micelles gel. This not only realized drug injectability but also drug storage in the semisolid form, which is a more comfortable and slower drug-releasing form that will eventually exert a proper therapeutic effect. In conclusion, quercetin micellar hydrogel system has better antitumor activity and excellent hydrogel properties.

Polymeric Micelles with Chiral Diamine-Ru(II) Catalysts for Asymmetric Transfer Hydrogenation of Ketones in Water

Zhou Lianpei,Ji Peipei,Wang Xiuwu,Qi Dongming,Chen Tao

Chiral aromatic alcohols with unique structures have attracted wide attention in asymmetric catalysis for decades. In this study, polymeric micelles with chiral diamine-Ru(II) catalysts in the hydrophobic core for asymmetric transfer hydrogenation of ketones affording chiral aromatic alcohols were presented. Firstly, block copolymers with (1R,2R)-(+)-1,2-diphenyl ethylenediamine (DPEN) in the pendant chain were successfully synthesized by Ring-Opening Metathesis Polymerization (ROMP) using 1,1-dimethyl ethyl N-(2-amino-1,2-diphenylethyl) carbamate (NB-DPEN-Boc) and polyethylene glycol (NB-PEG 2000 ) with norbornene groups as raw materials. The structures of block copolymers P(DPEN) -co- P(PEG 2000 ) were characterized by FT-IR and 1 H NMR. Next, Block copolymers with chiral DPEN ligand in the pendant chain coordinate with [RuCl 2 (p-cymene)] 2 to provide amphiphilic polymers with chiral diamine-Ru(II) catalyst. Then, the self-assembly of the amphiphilic polymers in water produced the polymeric micelles with chiral diamine-Ru(II) catalysts in the hydrophobic core. The particle sizes were found to be 65 nm by DLS analysis. The polymeric micelles as catalytic nanoreactors were successfully applied in the asymmetric transfer hydrogenation (ATH) of various ketones in water. The heterogenization of homogeneous catalysts and construction of catalysts for aqueous catalysis demonstrate potential application prospects in the preparation of chiral aromatic alcohols. Procedurally Targeted Delivery of Antitumor Drugs Using FAPα-Responsive TPGS Dimer-Based Flower-like Polymeric Micelles

Xinping Hu,Chuhang Zhou,Leqi Wang,Qi Liu,Yining Ma,Yingwei Tang,Xiaoxiao Wang,Kanghao Chen,Xinyu Wang,Yan Liu

To overcome the intestinal epithelium barrier and achieve a better antitumor effect, the procedurally targeting flower-like nanomicelles for oral delivery of antitumor drugs were designed based on FAPα-responsive TPGS1000 dimer (TPGS-Gly-Pro-TPGS) and L-carnitine linked poly(2-ethyl-2-oxazoline)-b-poly(D, l-lactide) (Car-PEOz-b-PLA). As expected, compared with unmodified polymeric micelles (TT-PMs) composed of TPGS-Gly-Pro-TPGS, L-carnitine conjugated polymeric micelles (CTT-PMs) formed from both TPGS-Gly-Pro-TPGS and Car-PEOz-b-PLA with favorable stability in simulated gastrointestinal fluid and FAPα-dependent release capability exhibited remarkably enhanced cellular uptake and transmembrane transport through OCTN2 mediation confirmed by fluorescence immunoassay, which was intuitively evidenced by stronger fluorescence within epithelial cells, and the basal side of small intestinal epithelium of mice being given intragastric administration of DiI-labeled micelles. The transport of CTT-PMs across the intestinal epithelium in an intact form was mediated by clathrin along the intracellular transport pathway of endosome-lysosome-ER-Golgi apparatus. Furthermore, both the increased uptake by FAPα-positive U87MG cells and unchangeable uptake by FAPα-negative C6 cells for coumarin-6 (C-6)/CTT-PMs compared with C-6/TT-PMs evidenced the targeting ability of CTT-PMs to FAPα-positive tumor cells. Both OCTN2-mediation and FAPα-responsiveness were beneficial for polymeric micelles to improve the delivery and therapeutic efficiency of antitumor agents, which was further supported by the remarkable enhancement in in vivo antitumor efficacy via promoting apoptosis of tumor cells for paclitaxel (PTX)-loaded CTT-PMs (PTX/CTT-PMs) with low toxicity compared with PTX/TT-PMs. Our findings offered an alternative design strategy for procedurally targeted delivery of chemotherapeutics by an oral route.

TPGS/soluplus® blended micelles: an effective strategy for improving loading capacity of ferroptosis inducer erastin

Junwei Liu,Meihong Zhang,Chuanbin Wu,Xin Pan,Zhengwei Huang

Erastin is the most classical small molecular ferroptosis inducer but its clinical application is hindered due to its poor solubility. Nanotechnology has been used to improve ferroptosis inducer erastin’s solubility. The recently reported erastin-incorporated nanosystems (e.g., nanoparticulate ferritin, poly(ethylene glycol) methyl ether-blockpoly(lactide-co-glycolide) (mPEG-PLGA) nanoparticle, exosome, and liposome) could to some degree elevate its solubility, but were still poor in loading capacity. To be specific, the maximum entrapment efficiency and drug loading in previous studies were approximately 36% and 3.7%, respectively. In order to improve drug loading capacity, we constructed a novel carrier based on Vitamin E d-α-Tocopherol polyethylene glycol 1000 succinate (TPGS) and polyvinyl caprolactam-polyvinyl acetate-polyethylene glycol graft copolymer (Soluplus®) blended micelles. The erastin@TPGS/Soluplus® micelles were prepared by thin film hydration method, and formulation details including dosage of Soluplus®, the weight ratio of TPGS to Soluplus®, the weight ratio of erastin to TPGS/Soluplus® were optimized. The physicochemical properties of the erastin micelles were measured by DLS, TEM, FTIR, and TG-DSC analysis. The products exhibited a nanoscale and uniform particle size (78.11 ± 0.54 nm) and neutral surface charge (-0.43 ± 0.32 mV). Significantly improved entrapment efficiency (89.63 ± 1.81%) and drug loading rate (10.03 ± 0.14%) were obtained, which was over 2-fold of current reports. The in vitro release study demonstrated that erastin could release in a sustained yet complete manner, and the drug release might be governed by multiple mechanisms including anomalous diffusion of non-Fickian diffusion, matrix erosion and sustained release. In summary, this proposed erastin nanosystem provides a new strategy for improving loading capacity, and may probably augment ferroptosis-inducing therapy for cancer treatment.

pH-sensitive charge-conversion cinnamaldehyde polymeric prodrug micelles for effective targeted chemotherapy of osteosarcoma in vitro

Jiapeng Deng,Su Liu,Guoqing Li,Yien Zheng,Weifei Zhang,Jianjing Lin,Fei Yu,Jian Weng,Peng Liu,Hui Zeng

Introduction: Chemotherapy is a common strategy for the treatment of osteosarcoma. However, its therapeutic efficacy is not ideal due to the low targeting, lowbioavailability, and high toxicity of chemotherapy drugs. Nanoparticles can improve the residence time of drugs at tumor sites through targeted delivery. This new technology can reduce the risk to patients and improve survival rates. To achieve this goal, we developed a pHsensitive charge-conversion polymeric micelle [mPEG- b -P(C7- co -CA) micelles] for osteosarcoma-targeted delivery of cinnamaldehyde (CA). Methods: First, an amphiphilic cinnamaldehyde polymeric prodrug [mPEG- b -P(C7- co -CA)] was synthesized through Reversible Addition-Fragmentation Chain Transfer Polymerization (RAFT) polymerization and post-modification, and self-assembled into mPEG- b -P(C7- co -CA) micelles in an aqueous solution. The physical properties of mPEG- b -P(C7- co -CA) micelles, such as critical micelle concentration (CMC), size, appearance, and Zeta potential were characterized. The CA release curve of mPEG- b -P(C7- co -CA) micelles at pH 7.4, 6.5 and 4.0 was studied by dialysis method, then the targeting ability of mPEG- b -P(C7- co -CA) micelles to osteosarcoma 143B cells in acidic environment (pH 6.5) was explored by cellular uptakeassay. The antitumor effect of mPEG- b -P(C7- co -CA) micelles on 143B cells in vitro was studied by MTT method, and the level of reactive oxygen species (ROS) in 143B cells after mPEG- b -P(C7- co -CA) micelles treatment was detected. Finally, the effects of mPEG- b -P(C7- co -CA) micelles on the apoptosis of 143B cells were detected by flow cytometry and TUNEL assay. Results: An amphiphilic cinnamaldehyde polymeric prodrug [mPEG- b -P(C7- co -CA)] was successfully synthesized and self-assembled into spheric micelles with a diameter of 227 nm. The CMC value of mPEG- b -P(C7- co -CA) micelles was 25.2 mg/L, and it showed a pH dependent release behavior of CA. mPEG- b -P(C7- co -CA) micelles can achieve chargeconversion from a neutral to a positive charge with decreasing pHs. This charge-conversion property allows mPEG- b -P(C7- co -CA) micelles to achieve 143B cell targeting at pH 6.5. In addition, mPEG- b -P(C7- co -CA) micelles present high antitumor efficacy and intracellular ROS generation at pH 6.5 which can induce 143B cell apoptosis. Discussion: mPEG- b -P(C7- co -CA) micelles can achieve osteosarcoma targeting effectively and enhance the anti-osteosarcoma effect of cinnamaldehyde in vitro . This research provides a promising drug delivery system for clinical application and tumor treatment.

Acid/GSH Dual-Responsive Endosome Escape Dual Pro-drug Micelles Targeted Synergistic Treatment for A549/ADR

Hailiang Chen,Chenyu Liu,Simiao Yu,Hengjun Zhou,Farishta Shafiq,Weihong Qiao

The resistance of cancer cells to anticancer drugs has been recognized as one of the main reasons for chemotherapy failure. Multidrug combination therapy is one of the most effective ways to solve this problem. Therefore, in this article, we designed and synthesized a pH/GSH dual-responsive camptothecin/doxorubicin (CPT/DOX) dual pro-drug synergistic treatment system with the aim of overcoming the resistance of non-small cell lung cancer A549/ADR cells to DOX. The pro-drug cRGD-PEOz-S-S-CPT (cPzT) was obtained by linking CPT to poly(2-ethyl-2-oxazoline) (PEOz) with endosomal escape properties through a GSH-responsive disulfide bond and modifying it with the targeted peptide cRGD. The pro-drug mPEG–NH–N=C-DOX (mPX) was synthesized by attaching DOX to polyethylene glycol (PEG) through acid-sensitive hydrazone bonds. The dual pro-drug micelles cPzT/mPX configured according to the CPT/DOX mass ratio of 3:1 showed a strong synergistic therapeutic effect at IC50 with a combined therapy index CI = 0.49, far less than 1. Moreover, with the further improvement of the inhibition rate, the 3:1 ratio showed a stronger synergistic therapeutic effect than other ratios. The cPzT/mPX micelles not only had better targeted uptake ability but also showed a better therapeutic effect in both 2D and 3D tumor suppression assays relative to free CPT/DOX and significantly enhanced the penetration ability into solid tumors. In addition, the results of confocal laser scanning microscopy (CLSM) showed that cPzT/mPX could effectively overcome the resistance of A549/ADR cells to DOX by delivering DOX into the nucleus to exert its effect. Thus, this dual pro-drug synergistic therapy system combining targeting and endosomal escape ability provides a possible strategy to overcome tumor drug resistance.
