Wenwen Chen,Rong Chen,Yutong Sun,Han Wang,Zhenyu Chen,Liqun Chen,Guoming Huang,Guifang Lin,Dengke Hong

The gut bacteria not only play a crucial role in maintaining human health but also exhibit close associations with the occurrence of numerous diseases. Understanding the physiological and pathological functions of gut bacteria and enabling early diagnosis of gut diseases heavily relies on accurate knowledge about their in vivo distribution. Consequently, there is a significant demand for noninvasive imaging techniques capable of providing real-time localization information regarding gut bacteria. In this work, we developed a second near-infrared (NIR-II) fluorescent nanoprobe labeling-based visualization strategy for real-time tracking of the biodistribution of Gram-negative bacteria in the gastrointestinal tract of mice. By utilizing positively charged silver sulfide quantum dots (Ag2S QDs) as NIR-II nanoprobes and exploiting electrostatic interactions to efficiently label the Gram-negative probiotic Escherichia coli Nissle 1917 with negative surface charges, we have achieved rapid and effective labeling. Leveraging the exceptional NIR-II fluorescent performance of Ag2S QDs, our approach enables high spatiotemporal resolution visualization via NIR-II imaging in mouse gastrointestinal areas where Ag2S QD-labeled probiotics are present, facilitating real-time in vivo tracking capabilities for these labeled probiotics. This work not only establishes a powerful intestinal bacterial imaging strategy but also introduces novel concepts for constructing nanomaterial-bacteria hybrid systems.

Fluoride nanocrystals and quantum dots nanocomposite for efficient luminescence nanothermometer in NIR-II biological window

Zhengying Du,Denghao Li,Lei Lei,Gongxun Bai,Huanping Wang,Xianghua Zhang,Shiqing Xu,Jianrong Qiu

Biosensors working in the second near-infrared (NIR-II) spectral region have recently gained great attention owning to their deep tissue bioimaging and high spatiotemporal resolution . However, there is still a challenge to develop efficient nanothermometers based on NIR-II luminescence. Herein, we present a pioneering advancement in the field of nanothermometry by synthesizing a novel nanothermometer, a composite of fluoride nanocrystals and quantum dots . This NaYF 4 :1Nd-Ag 2 S QDs@SiO 2 nanocomposite demonstrates exceptional functionality, being excited by a single light irradiation at 808 nm for NIR-II luminescence imaging. Notably, the composite emits at two distinct wavelengths-1064 nm and 1125 nm, attributable to Nd 3+ and Ag 2 S QDs, respectively. This unique emission profile enables deeper tissue penetration, mitigating overheating risks compared to conventional excitation sources at 980 nm. Furthermore, the nanomaterials exhibit remarkable temperature-sensing capabilities, with maximum sensitivities of 0.81%K −1 (S R ) at 333 K. These superior sensitivities outperform many existing temperature sensors relying on thermally coupled Stark sublevels. The SiO 2 matrix not only encapsulates and protects the fluoride nanocrystals and quantum dots but also shields them from aqueous environments, ensuring outstanding luminescence stability. Our findings highlight the potential of this nanothermometer as a highly efficient temperature sensor with promising applications in cutting-edge bioapplications . This development introduces a new frontier in temperature sensing and bioimaging, showcasing its potential for transformative contributions to the field.

Iron-Single-Atom Nanozyme with NIR Enhanced Catalytic Activities for Facilitating MRSA-Infected Wound Therapy

Qian Liu,Xueliang Liu,Xiaojun He,Danyan Wang,Chen Zheng,Lin Jin,Jianliang Shen

Patients with methicillin-resistant Staphylococcus aureus (MRSA) infections may have higher death rates than those with non-drug-resistant infections. Nanozymes offer a promising approach to eliminating bacteria by producing reactive oxygen species. However, most of the conventional nanozyme technologies encounter significant challenges with respect to size, composition, and a naturally low number of active sites. The present study synthesizes a iron-single-atom structure (Fe-SAC) via nitrogen doped-carbon, a Fe-N 5 catalyst (Fe-SAC) with a high metal loading (4.3 wt.%). This catalyst permits the development of nanozymes consisting of single-atom structures with active sites resembling enzymes, embedded within nanomaterials. Fe-SAC displays peroxidase-like activities upon exposure to H 2 O 2 . This structure facilitates the production of hydroxyl radicals, well-known for their strong bactericidal effects. Furthermore, the photothermal properties augment the bactericidal efficacy of Fe-SAC. The findings reveal that Fe-SAC disrupts the bacterial cell membranes and the biofilms, contributing to their antibacterial effects. The bactericidal properties of Fe-SAC are harnessed, which eradicates the MRSA infections in wounds and improves wound healing. Taken together, these findings suggest that single Fe atom nanozymes offer a novel perspective on the catalytic mechanism and design, holding immense potential as next-generation nanozymes.

Inhaled NIR-II Nanocatalysts for Real-Time Monitoring and Immunomodulatory Therapy of Acute Lung Injury

Yi Li,Dongfang Liu,Ting Chen,Jianwen Song,Xuya Yu,Qian Liu,Ji Qi,Wen Li

Acute lung injury (ALI) poses a significant and escalating medical challenge, where precise diagnosis and timely intervention hold utmost importance in halting its deterioration. Nevertheless, persistent obstacles arise from the lack of agents proficient in both real-time diagnosis and efficient mitigation of ALI. Here, a biocatalytic and second near-infrared (NIR-II) fluorescence-illuminating nanoplatform is designed to simultaneously facilitate real-time monitoring and robust inflammation alleviation in ALI. The study first develops a new aggregation-induced emission luminogen with trifluoromethyl substitutions, which simultaneously increase NIR-II emission wavelength and fluorescence brightness. The molecular probe is further integrated into biocatalytic hollow ceria nanostructures, and cloaked with pre-activated macrophage membranes for targeted inflammation intervention. Upon inhalation administration in ALI mice, the theranostic nanoagents leverage the bright NIR-II emission and active inflammation-tropic properties for in vivo sensitive NIR-II imaging of ALI, which also facilitates real-time tracking of the nanoagents’ distribution and dynamic fate within pneumonia milieu. Concurrently, the catalytic prowess of the nanoplatforms efficaciously scavenges excess reactive oxygen species, dampens proinflammatory cytokines, and promotes macrophage repolarization, substantially alleviating acute lung damage. The multifaceted nanoplatform integrates NIR-II bioimaging with nanocatalysis-mediated immunoregulation, offering a versatile and promising approach for addressing the intricate challenges posed by acute inflammation diseases.

NIR II Laser-Triggered Photothermal Nanoplatform for Multimodal Imaging-Guided Synergistic Therapy toward Colon Cancer

Guodong Ren,Xuewei Wang,Jianbo Cao,Haolin Pu,Jinyao Li,Lili Yan,Sufang Ma,Lihong Li,Lixia Guo,Boye Zhang,Haojiang Wang,Bin Wang,Haipeng Diao,Wen Liu,Chengwu Zhang

Colon cancer is one kind of malignant digestive tract tumor with high morbidity and mortality worldwide, treatments for which still face great challenges. Recently emerged intervention strategies such as phototherapy and gas therapy have displayed promising effects in the treatment of colon cancer, but their application are still hindered due to insufficient tumor targeting and deeper tissue penetrating capacity. Herein, in the present study, we developed one theranostic nanoplatform Cet-CDs-SNO (CCS) to realize multimodal imaging-guided synergistic colon cancer therapy. Among the CCS, Cetuximab (Cet), one first-line clinical drugs for colorectal cancer, endowed CCS with tumor-targeting capacity and enhanced drug accumulation in tumor cells; CDs doped by Ni2+ and Mn2+ served as NIR-II photothermal therapy (PTT), chemodynamic therapy (CDT), and photothermal/magnetic resonance/fluorescence imaging (PTI/MRI/FLI) agents; SNO, a nitric oxide (NO) donor, exerted gas therapeutic (GT) effects under thermal stimulation derived from PTT. In vitro and in vivo experiments proved that CCS had excellent colon cancer-targeting ability. Proliferation of colon cancer cells and tumor growth were significantly inhibited by the administration of CCS without detectable cytotoxicity. This study presented one strategy for developing a multifunctional nanoplatform to be applied in imaging-guided precise tumor therapy.

NIR-Activated Hydrogel with Dual-Enhanced Antibiotic Effectiveness for Thorough Elimination of Antibiotic-Resistant Bacteria

Fengjiao Yang,Yuqi Ge,Yue Zhang,Zhongqi Cui,Shiyang Lin,Wenxuan Ni,Zijiu Sun,Dandan Shen,Jichao Zhu,Li Liu,Shasha Zhao,Nan Huang,Fenyong Sun,Yingying Lu,Shuo Shi,Jinghua Li

Antibiotic resistance has become a critical health crisis globally. Traditional strategies using antibiotics can lead to drug-resistance, while inorganic antimicrobial agents can cause severe systemic toxicity. Here, we have developed a dual-antibiotic hydrogel delivery system (PDA-Ag@Levo/CMCS), which can achieve controlled release of clinical antibiotics levofloxacin (Levo) and classic nanoscale antibiotic silver nanoparticles (AgNPs), effectively eliminating drug-resistant P. aeruginosa. Benefiting from the photothermal (PTT) effect of polydopamine (PDA), the local high temperature generated by PDA-Ag@Levo/CMCS can quickly kill bacteria through continuous and responsive release of dual-antibiotics to restore sensitivity to ineffective antibiotics. Moreover, AgNPs could significantly improve the efficiency of traditional antibiotics by disrupting bacterial membranes and reducing their toxicity to healthy tissues. A clever combination of PTT and drug-combination therapy can effectively eliminate biofilms and drug-resistant bacteria. Mechanism studies have shown that PDA-Ag@Levo might eliminate drug-resistant P. aeruginosa by disrupting biofilm formation and protein synthesis, and inhibit the resistance mutation of P. aeruginosa by promoting the expression of related genes, such as rpoS, dinB, and mutS. Collectively, the synergistic effect of this dual-antibiotic hydrogel combined with PTT provides a creative strategy for eliminating drug-resistant bacteria in chronic infection wounds.

NIR-enhanced multi-mode catalytic activity over Pd nanocrystals sandwiched within magnetic polydopamine hollow spheres

Jie Jin,Haoran Li,Shanshan Wu,Jing Wang,Qunling Fang,Chunyan Xu,Yunqi Xu,Weili Kong,Di Wang,Shouhu Xuan

With the rapid development of social industrialization, energy crisis and environmental problems are becoming increasingly serious. Therefore, there is an urgent need to synthesize multi-mode nanocatalysts for general application. In this paper, a h-Fe3O4@Pd/PDA nanocatalyst with Pd nanocrystals sandwiched between a h-Fe3O4 hollow sphere and a polydopamine (PDA) shell was prepared by a simple redox-oxidative polymerization method. The catalytic activity of h-Fe3O4@Pd/PDA hollow spheres was evaluated using selective reduction hydrogenation of 4-nitrostyrene, reduction of 4-nitrophenol and oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) as catalytic models. h-Fe3O4@Pd/PDA has reduction kinetics of 4-nitrophenol up to 0.77 min−1 mg−1. Importantly, the rate of 4-nitrophenol reduction catalyzed by h-Fe3O4@Pd/PDA was enhanced by 1.43 times under near infrared light (NIR, 808 nm) irradiation. In addition, NIR laser can induce the selective reduction of 4-nitrostyrene to 4-aminostyrene by h-Fe3O4@Pd/PDA with a selectivity of 84.8%. Under different reaction scenarios, h-Fe3O4@Pd/PDA hollow spheres could be recycled by magnetic recovery, and the catalytic reaction activity was above 70% after six cycles. As a result, owing to the simple fabrication, good magnetic separation and high NIR-enhanced catalytic properties, h-Fe3O4@Pd/PDA exhibited a wide range of potential in nanocatalysis.

NIR-IIb-triggered photodynamic therapy combined with chemotherapy platform based on rare-earth-doped nanoparticles

Tu Zhuo,Wan Yong,Ge Juan,Li Chen-Chen,Liang Tao,Li Zhen

Abstract Photodynamic therapy (PDT), with high spatiotemporal selectivity, minimal side effects and less drug resistance, has emerged as a promising approach for the treatment of cancer. However, inadequate penetration depth and poor efficacy seriously restrict its practical application. To address these challenges, we developed a combined therapy platform (RENP@C/D-FA) based on rare-earth-doped nanoparticles (RENPs) with a structure of NaErF 4 :Tm(0.5%)@NaYF 4 , on which photosensitizer Ce6 (chlorin e6) and chemo-therapeutic drug (DOX) (doxorubicin) were loaded by encapsulating with an amphiphilic polymer distearoyl phosphatidylethanolamine-polyethylene glycol-folic acid (DSPE-PEG-FA). RENPs with NaErF 4 as the matrix possess multiple excitation/emission peaks, enabling activation of PDT under near-infrared-IIb (NIR-IIb, 1500–1700 nm) excitation and luminescence imaging in NIR-IIb region by absorbing 808 nm near-infrared photons. Furthermore, DOX can be selectively released in the acidic microenvironment of tumors. Benefiting from the tumor targeting capability of folic acid, this platform can successfully ablate tumor cells and inhibit tumor growth by combining PDT and chemotherapy. This innovative approach holds great promise as a powerful tool for cancer treatment, showing the potential to revolutionize PDT. Graphical abstract

NIR-Triggered Thermosensitive Nanoreactors for Dual-Guard Mechanism-Mediated Precise and Controllable Cancer Chemo-Phototherapy

Guoxi Qiu,Wentao Zhou,Yupeng Liu,Tingting Meng,Fangying Yu,Xiangyu Jin,Keke Lian,Xueqing Zhou,Hong Yuan,Fuqiang Hu

Thermosensitive nanoparticles can be activated by externally applying heat, either through laser irradiation or magnetic fields, to trigger the release of drug payloads. This controlled release mechanism ensures that drugs are specifically released at the tumor site, maximizing their effectiveness while minimizing systemic toxicity and adverse effects. However, its efficacy is limited by the low concentration of drugs at action sites, which is caused by no specific target to tumor sties. Herein, hyaluronic acid (HA), a gooey, slippery substance with CD44-targeting ability, was conjugated with a thermosensitive polymer poly(acrylamide-co-acrylonitrile) to produce tumor-targeting and thermosensitive polymeric nanocarrier (HA-P) with an upper critical solution temperature (UCST) at 45 °C, which further coloaded chemo-drug doxorubicin (DOX) and photosensitizer Indocyanine green (ICG) to prepare thermosensitive nanoreactors HA-P/DOX&ICG. With photosensitizer ICG acting as the “temperature control element”, HA-P/DOX&ICG nanoparticles can respond to temperature changes when receiving near-infrared irradiation and realize subsequent structure depolymerization for burst drug release when the ambient temperature was above 45 °C, achieving programmable and on-demand drug release for effective antitumor therapy. Tumor inhibition rate increased from 61.8 to 95.9% after laser irradiation. Furthermore, the prepared HA-P/DOX&ICG nanoparticles possess imaging properties, with ICG acting as a probe, enabling real-time monitoring of drug distribution and therapeutic response, facilitating precise treatment evaluation. These results provide enlightenment for the design of active tumor targeting and NIR-triggered programmable and on-demand drug release of thermosensitive nanoreactors for tumor therapy.

Nanoparticles destabilizing the cell membranes triggered by NIR light for cancer imaging and photo-immunotherapy

Tang Dongsheng,Cui Minhui,Wang Bin,Liang Ganghao,Zhang Hanchen,Xiao Haihua

Cationic polymers have great potential for cancer therapy due to their unique interactions with cancer cells. However, their clinical application remains limited by their high toxicity. Here we show a cell membrane-targeting cationic polymer with antineoplastic activity (P mt ) and a second near-infrared (NIR-II) fluorescent biodegradable polymer with photosensitizer Bodipy units and reactive oxygen species (ROS) responsive thioketal bonds (P Bodipy ). Subsequently, these two polymers can self-assemble into antineoplastic nanoparticles (denoted mt-NP Bodipy ) which could further accumulate at the tumor and destroy cell membranes through electrostatic interactions, resulting in cell membrane destabilization. Meanwhile, the photosensitizer Bodipy produces ROS to induce damage to cell membranes, proteins, and DNAs to kill cancer cells concertedly, finally resulting in cell membrane lysis and cancer cell death. This work highlights the use of near-infrared light to spatially and temporarily control cationic polymers for photodynamic therapy, photo-immunotherapy, and NIR-II fluorescence for bio-imaging.

Acceptor-donor-acceptor-type Molecules with large electrostatic potential difference for Effective NIR Photothermal Theranostics

Kexin Fan,Ludan Zhang,Qinqiu Zhong,Yanhe Xiang,Bowei Xu,Yuguang Wang

Although acceptor-donor-acceptor (A-D-A)-type molecules show advantages in constructing NIR absorbing photothermal agents (PTAs) due to their strong intramolecular charge transfer and molecular planarity, their applications in photothermal therapy (PTT) of tumor are insufficiently explored. In particular, the influence of ESP distribution on optical properties of A-D-A photosensitizers has not been investigated. Herein, we utilized the advantages of A-D-A-type molecules with large ESP difference to construct NIR absorbing PTA with sufficient extinction coefficient (ɛ) and high photothermal conversion efficiency (PCE). The calculation results of density functional theory (DFT) indicate that the large ESP difference of Y6 may increase intermolecular interactions and induce tight packing of the A-D-A photosensitizer. As a result, Y6 NP exhibited an obvious bathochromic shift of absorption peak from 711 nm to 822 nm, with a NIR-II emission extended to 1400 nm. Moreover, a sufficient ɛ of 3.34 L·g-1·cm-1 and PCE of 66.3% were attained, making Y6 NP suitable for PTT. With a concentration of 100 μg/mL, Y6 NPs in aqueous dispersion yielded a death rate of 93.4% for 4T1 cells upon an 808 nm laser irradiation (1 W·cm-2) for 10 min, which is comparable with the best results of recently reported PTT agents.

Blue light-excited broadband NIR-II-emitting Li2ZnSn3O8:Cr3+,Ni2+ phosphor for multifunctional optical applications

Zhexuan Gao,Yi Zhang,Yinyan Li,Shilong Zhao,Peng Zhang,Xiaolong Dong,Degang Deng,Shiqing Xu

The second near-infrared window (NIR-II, 1000–1700 nm) emitting phosphor is a key component for the phosphor-converted light-emitting diode (pc-LED), which has garnered significant attention. However, it remains a major challenge to discover efficient NIR-II broadband phosphors that can be excited by blue LEDs. Herein, Cr3+ and Ni2+ ions were co-doped into the Li2ZnSn3O8 (LZSO) host. LZSO:Cr3+,Ni2+ phosphor achieves a broadband NIR-II emission peak at 1465 nm with a large full width at half maximum (FWHM) of 300 nm and internal quantum efficiency (IQE) of 43.31% under excitation at 426 nm because of the efficient energy transfer (ET) from Cr3+ to Ni2+. In addition, a dual-emissive doped phosphor across the NIR-I (700–1000 nm) and NIR-II regions with a higher sensitivity at physiological temperatures was obtained. The relative sensitivity value (Sr) reaches 1.44% K−1 at 293 K, indicating that the LZSO:Cr3+,Ni2+ phosphor is a potential candidate for NIR optical thermometers in biotechnological applications. The manufactured NIR-II pc LED provides a radiative flux of 8.26 mW with a photoelectric conversion efficiency of 2.71% at a drive current of 100 mA and has promising potential in night vision, medical imaging, and spectral analysis applications.
