Unintended changes in ocular biometric parameters during a 6-month follow-up period after FS-LASIK and SMILE

Wang Junjie,Lopes Bernardo T.,Li Hechen,Vinciguerra Riccardo,Cao Si,Wu Songan,Zhu Rong,Wang Qinmei,Zheng Xiaobo,Bao Fangjun,Elsheikh Ahmed

Background Corneal refractive surgery has become reliable for correcting refractive errors, but it can induce unintended ocular changes that alter refractive outcomes. This study is to evaluate the unintended changes in ocular biometric parameters over a 6-month follow-up period after femtosecond laser-assisted laser in situ keratomileusis (FS-LASIK) and small incision lenticule extraction (SMILE). Methods 156 consecutive myopic patients scheduled for FS-LASIK and SMILE were included in this study. Central corneal thickness (CCT), mean curvature of the corneal posterior surface (K pm ), internal anterior chamber depth (IACD) and the length from corneal endothelium to retina (ER) were evaluated before and after surgery over a 6-month period. Results Both the FS-LASIK and SMILE groups (closely matched at the pre-surgery stage) experienced flatter K pm , shallower IACD and decreased ER 1 week post-surgery ( P  < 0.01), and these changes were larger in FS-LASIK than in SMILE group. During the 1 week to 6 months follow up period, K pm , IACD and ER remained stable unlike CCT which increased significantly ( P  < 0.05), more in the FS-LASIK group. Conclusions During the follow up, the posterior corneal surface became flatter and shifted posteriorly, the anterior chamber depth and the length from the corneal endothelium to retina decreased significantly compared with the pre-surgery stage. These unintended changes in ocular biometric parameters were greater in patients undergoing FS-LASIK than SMILE. The changes present clear challenges for IOL power calculations and should be considered to avoid affecting the outcome of cataract surgery.

Hydrated perovskite-based MgSn(OH)6/4CuS@HC nanostructures: Z-scheme photocatalysis for water pollutants degradation in wastewater

Omar S.,Omar M.,El-Subruiti G. M.,Attia N. F.,Eltaweil A.

This study presents a novel MgSn(OH) 6 /4CuS@HC photocatalyst developed via hydrothermal treatment followed by chemical precipitation. MgSn(OH) 6 /4CuS@HC was applied for wastewater treatment, targeting eight pollutants: Tetracycline, Doxycycline, Ofloxacin, 3-Nitroaniline, 2-Nitrophenol, Methyl red, Congo red, and Methylene blue. The novel photocatalyst was studied using FTIR, EDX, DRS, SEM, and PL techniques. To gain further insight into the reaction mechanism, scavenger experiments were conducted in addition to operational parameters such as pH, dosage, CuS loading on MgSn(OH) 6 @HC, pollutant initial concentration, selectivity, reusability, water source, and kinetic studies. The performance test highlighted the outstanding photocatalytic activity of templated MgSn(OH) 6 /4CuS@HC, exhibiting remarkable stability over five cycles. This performance is attributed to the direct Z-scheme mechanism and the hydrochar template, which improve charge separation, extend photocharge lifetime, and reduce the agglomeration of nanosemiconductors, thereby enhancing overall stability. This study offers a new perspective on green photocatalysts for diverse environmental applications.

Preparation of a composite dust suppressant and investigation of its toxicity to lung tissue in C57BL/6 mice

Rongxia Yu,Zhian Huang,Hao Ding,Longsheng Huang,Junchuan Zhang,Xiangrui Meng,Shunye He,Chao Huan,Yinghua Zhang,Asim Farooq,Jinhui Li,Yingjie Gao

Excessively high dust concentrations can induce pneumoconiosis, a condition presenting considerable therapeutic challenges. Dust suppressants constitute one effective means of dust control; however, research on dust suppressants has predominantly focused on characterizing their performance properties, with no systematic evaluations to date of their toxic effects on lung tissue. Therefore, through modification and response surface analysis, this study determined the optimal formulation for a composite dust suppressant to be as follows: 0.47 g modified guar gum, 0.24 mL rhamnolipid, and 5.36 g polyethylene glycol. Three toxic-exposure models were established to investigate the effects of dust and dust suppressants—both individually and in combination—on the lung tissue of C57BL/6 mice. The results indicated that low concentrations of dust suppressant (4 mg/m3) reduced total protein and immunoglobulin levels, thereby lowering mucosal infection risk; moderate concentrations (4–20 mg/m3) inhibited M1-type alveolar macrophage polarization and inflammatory factor expression (IL-6, TNF-α, etc.); and high doses (100 mg/m3) reduced the pulmonary fibrosis positivity rate from 11.31% to 1.30%, showing a tendency to reduce the probability of fibrosis. This study preliminarily complements the toxicological research system of dust suppressants and offers preliminary reference for the safe application of composite dust suppressants.

Polyamide 6 Composites With Balanced Flame Retardancy and Mechanical Properties via Epoxy Silicone Oil/Aluminum Diethylphosphinate Synergistic System

Shuo Zhang,Linlin Jiang,Ruiping Wang,Chuang He,Miaojun Xu,Zhichao Tian,Yue Xu,Zhiyong Zhang,Bin Li

Polyamide 6 (PA6) composites offer great promise for industrial applications, yet balancing flame retardancy with mechanical properties is an ongoing challenge. Epoxy silicone oil (ESO) acted as a reactive compatibilizer to prepare flame-retardant PA6/ADP/ESO composites. The fire safety of composites was evaluated using standard flame-retardant tests and combustion behavior tests. Meanwhile, the effects of ESO incorporation on the rheological behavior, mechanical properties, and thermal stability of PA6/ADP composites were systematically studied by rheological tests, impact tests, and thermogravimetric analysis (TGA). The results showed that even with a total addition of only 11% of the ADP/ESO (96:4) system, the PA6 composites still passed the UL-94 V-0 rating with a limiting oxygen index (LOI) of 30.7%. Compared to the pure PA6/ADP system, the PA6/ADP/ESO composites had a 43.11% lower peak of smoke production rate (PSPR) and excellent fire resistance. Furthermore, the impact strength of the PA6/ADP 96 ESO 4 composites was significantly improved, which was attributed to the chemical reaction between the epoxy functionalities in ESO and the terminal groups of PA6 chains, thereby promoting chain extension and the establishment of cross-linked networks. Meanwhile, the melt viscosity of the system also increased. Interestingly, the increased shear force exerted by the matrix on the ADP aggregates not only significantly improved the dispersibility of ADP within the matrix but also helped to alleviate stress concentration. A simple, effective method produces high-performance PA6 composites that balance flame retardancy with mechanical integrity.

Ultrasensitive LC-MS analysis of saccharides using 2-hydrazinyl-6-methoxy-1,3-benzothiazole as a novel derivatization reagent

Xin Xin,Qi Zhang,Sujun Yan,Zilong Chen,Xinhai Zhu

Background Monosaccharides and disaccharides are central components of carbohydrate metabolism, and fluctuations in their abundances are closely associated with disease onset, progression, and diagnostic outcomes. However, accurate analysis of trace-level saccharides in complex biological matrices remains challenging due to their limited chromatographic retention, modest ionization efficiency, and limited MS/MS fragmentation. Derivatization has been widely explored to improve analytical performance in LC-MS-based saccharide analysis; nevertheless, further development of simple, sensitive, and broadly applicable strategies is desirable, particularly for small-volume and low-abundance biological samples. This work presents an efficient derivatization-LC-MS approach enabling ultrasensitive quantification of saccharides under microscale analytical conditions. Results A derivatization strategy based on 2-hydrazinyl-6-methoxy-1,3-benzothiazole (HMBT) was developed to enhance LC-MS analysis of monosaccharides and disaccharides. The mild reaction generated stable derivatives directly compatible with LC-MS. HMBT tagging significantly improved chromatographic retention and separation on conventional C18 columns and markedly enhanced ionization efficiency. The derivatives produced high-intensity precursor ions and a dominant, highly responsive product ion, enabling ultrasensitive MRM detection on a triple-quadrupole system. Using nine representative saccharides, the method achieved excellent quantitative linearity down to 10 pg·mL −1 . Application to mouse serum enabled the quantification of glucose at single-cell-equivalent levels, and spiked galactose showed a linear range of 0.05–100 ng·mL −1 . Importantly, the workflow enabled quantification of five endogenous saccharides in individual HeLa cells. Significance The HMBT derivatization workflow provides an ultrahigh-sensitivity LC-MS platform for saccharide analysis, enabling robust MRM-based quantification at pg·mL −1 levels. Its operational simplicity, minimal sample handling, and full compatibility with conventional LC-MS instrumentation facilitate practical implementation. The demonstrated applicability to true single-cell measurements highlights its potential for glycometabolic studies in microscale biological systems.

Extracellular Vesicle-Mediated Delivery of VEGF and NGF Protects Dopaminergic Neurons in 6-OHDA-Induced Parkinson’s Disease Models

Wenyu Yan,Xianyue Meng,Yan Wang,Chuanfei Wei,Fabin Han,Xueli Li

Background: Parkinson’s disease (PD) is a neurodegenerative disorder marked by motor dysfunction. No definitive methods exist to repair damaged neurons. Vascular endothelial growth factor (VEGF) and nerve growth factor (NGF) are two neuroprotective agents that work synergistically. However, these large molecular proteins have difficulty crossing the blood–brain barrier (BBB). Extracellular vesicles (EVs) offer superior targeting and low immunogenicity, making them excellent carriers. In this study we examined the protective effects of VEGF and NGF in a cell model and evaluated the therapeutic potential of VEGF–NGF contained within EVs in PD rats. Methods: EVs were isolated using sequential differential centrifugation and characterized using transmission electron microscopy, nanoparticle tracking analysis, and western blotting (WB). VEGF and NGF were loaded into the EVs using a saponin-assisted method to create VEGF@EVs, NGF@EVs, and VEGF/NGF@EVs. The viability of 6-hydroxydopamine hydrochloride (6-OHDA)-induced SH-SY5Y cells was measured using the cell counting kit-8 assay before and after treatment with VEGF and NGF. Autophagy levels were assessed using WB, and the role of autophagy was further explored using the autophagy inhibitor chloroquine. Unilateral PD rat models were established via stereotactic injection of 6-OHDA into male Sprague–Dawley rats. Behavioral changes were monitored before and after treatment. Neuronal recovery, neurotransmitter levels, and autophagy levels in the rat brains were evaluated using immunohistochemistry, enzyme-linked immunosorbent assay, and WB. Results: VEGF/NGF@EVs significantly enhanced the viability of 6-OHDA-induced SH-SY5Y cells. A complete autophagic process was identified as essential for this protective effect. The intranasal administration of VEGF/NGF@EVs improved motor behavior in PD rats, with performance better than that of single growth factor treatments. The number of tyrosine hydroxylase (TH)–positive neurons, TH protein expression, and dopamine content were significantly increased. In addition, the level of autophagy in the rat substantia nigra was elevated. Conclusions: VEGF/NGF@EVs exert protective effects in both in vitro and in vivo 6-OHDA-induced PD models by promoting autophagy, demonstrating greater efficacy than either growth factor alone. By transplanting VEGF/NGF@EVs into PD rats, we showed that these vesicles can effectively cross the BBB and deliver targeted therapy to the central nervous system. This study highlights the significant potential of EV-mediated protein transplantation strategies for treating neurological disorders.

Ac EL-6 Reduces Methylglyoxal-Associated Glycation and NF-κB-Mediated Inflammatory Responses While Supporting Acute Skin Repair

Jiang Xinyi,Li Junxiang,Lu Yi,Ni Fangyuan,Li Sen

Purpose Advanced glycation end products (AGEs) and their downstream inflammatory consequences contribute to skin dysfunction and aging-related deterioration. EQ-9 has been identified as a novel functional peptide with anti-aging and skin repair potential. In this study, we investigated the effects of EQ-9 and its two derivatives (EL-6 and ac EL-6) on Methylglyoxal (MGO)-induced glycation and inflammatory damage. Methods We exposed HaCaT cells to MGO to simulate hyperglycemic conditions. The protective effects of these peptides were assessed through measurements of cell viability, migration, CML levels, inflammatory cytokine secretion, RAGE expression, and NF-κB pathway activation. Additionally, TNF-α/IFN-γ were used to induce inflammatory injury and evaluate the anti-inflammatory effectiveness of the peptides. A murine excision wound model was also established to examine the effects of ac EL-6 on skin inflammation, collagen deposition, angiogenesis, and wound healing. Results All three peptides significantly reduced MGO-induced CML generation, downregulated RAGE, inhibited NF-κB activation, attenuated proinflammatory cytokine secretion, alleviated cell damage, and promoted migration. They also mitigated TNF-α/IFN-γ-evoked inflammatory responses via NF-κB suppression. Topical application of ac EL-6 decreased cytokine levels, increased VEGF expression and collagen deposition, and consequently accelerated wound healing in mice. Conclusion Together, these findings identify ac EL-6 as a stabilized lead candidate derived from EQ-9, which exhibits anti-glycation activity and modulates NF-κB-mediated inflammatory signaling in keratinocytes, and demonstrates initial skin-repair potential in vivo. The data support further development of ac EL-6 for skin-directed applications. Graphical

Lipase-catalyzed synthesis of 4-amino-3,5-dicyano-6-arylphthalates and in silico assessment of anti-Leishmanial activity

Dantong Wang,Hanqing Xie,Yongqi Zeng,Fengxi Li,Lei Wang,Peng Chen,Zhi Wang

This study presented, for the first time, a method for synthesizing 4-amino-3,5-dicyano-6-aryl phthalates using porcine pancreatic lipase (PPL) as a catalyst. We synthesized 18 derivatives of 4-amino-3,5-dicyano-6-aryl phthalates employing aryl aldehyde, acetylenic esters, and malononitrile under optimal conditions for 4 h, with water serving as the primary reaction medium. The yields of the target products ranged from moderate to high, achieving 43 % to 95 %. This method demonstrates an excellent tolerance for various functional groups. Furthermore, this paper elucidates the reaction mechanism through molecular docking studies and confirms the interaction between the substrate and lipase. The data indicate that the measured distance from the substrate to the catalytic triad is 3.8 Å, with a binding energy of -4.6 kJ/mol, suggesting that PPL demonstrates the strongest catalytic potential in this specific reaction. Additionally, the aforementioned products underwent molecular docking analysis with three selected enzyme targets from Leishmania, highlighting their potential as lead compounds for anti-Leishmania activity.

UiO-66-SO3H@nylon-6 Membrane for Rapid Adsorption and Removal of Rhodamine B (RhB) from Aqueous Solutions

Heng Li,Chengzhong Zhang,Yuhang Jiang,Wenwu Ni,Mingshuo Li,Jing Wang

Widespread dye contamination poses a serious threat to humans and aquatic environments. Metal–organic frameworks (MOFs) with abundant reactive groups and high specific surfaces are candidates for the adsorption of dyes. In this study, good growth of UiO-66-SO3H crystals on the surface of nylon-6 fibers was achieved based on the idea of in situ growth. UiO-66-SO3H with a sulfonic acid group was successfully introduced into nylon-6 electrostatic spinning fiber, and the UiO-66-SO3H@nylon-6 membrane with high specific surface area and a three-dimensional mesh structure was obtained, which effectively solved the problems of clogging of the equipment, difficulty in being separated and recovered from the water, the possible secondary pollution of the water environment caused by the powder form of the MOF material, etc. The UiO-66-SO3H@nylon-6 membrane has a good adsorption capacity for rhodamine B (RhB), and the experimental data are consistent with the pseudo-second-order kinetic model and Langmuir model, which shows that the adsorption is a spontaneous heat absorption process. Pore-filling, hydrogen bonding, π–π interactions, and electrostatic attraction induced by –SO3H are the adsorption mechanisms of RhB on this adsorbent. In conclusion, the UiO-66-SO3H@nylon-6 membrane is an adsorbent capable of rapid adsorption of RhB and can be easily separated from water for recovery, regeneration, and utilization. This study may provide ideas for the application of MOFs in the field of pollutant removal.

Miniemulsion polymerization-formulated poly(fluorene-alt-6-(2-ethylhexyl)-[1,2,5]thiadiazole[3,4-f]benzotriazole) for cancer cell imaging

Zhiguo He,Xuejiao Han,Hui Xie,Zifeng Yan,Bing Guo,Youwei Yao

Near-infrared (NIR) conjugated polymer nanoparticles (CPNPs) possesses advantages for bioimaging because of their merits in low autofluorescence interference, good biocompatibility, large light extinction coefficient, improved photostability, and facile functionalization capability. Different from conventional post-polymerization methods for fabricating NIR CPNPs, like nanoprecipitation, emulsion and solvent evaporation/miniemulsion, and self-assembly, herein, an efficient one-pot strategy by in-situ miniemulsion polymerization is demonstrated to synthesize CPNPs for NIR fluorescence imaging of cancer cells. Via adjusting the donor-acceptor strength , the new CP, poly(fluorene- alt -6-(2-ethylhexyl)-[1,2,5]thiadiazole[3,4- f ]benzotriazole) ( PFTA ), show absorption and emission maximum at 648 nm and 730 nm, respectively, which is much more red-shifted than its counterpart, poly(fluorene- alt -benzo-2,1,3-thiadiazole) ( PFBT ). DFT calculation demonstrates that the repeating unit of PFTA has a smaller dihedral angel to 33.2° and much reduced energy gap to 2.46 eV, as compared to that of PFBT NPs. Importantly, through controlling surfactant concentrations from 40 g·L −1 to 2.5 g·L −1 during the miniemulsion polymerization, the particles size can be accordingly changed from even 88 to 210 nm. Furthermore, the particles show extinction coefficient of 2.82 × 10 4  M −1 ·cm −1 at 648 nm, photoluminescence quantum yields c.a. 2.4% and fluorescence lifetime c.a. 1.26 ns, respectively. Moreover, the PFTA NPs exhibit biocompatibility, good colloidal stability and bright fluorescence signals to clearly pinpoint breast cancer cells, with a trend that smaller NP size, higher cellular uptake. This work demonstrates the promise of miniemulsion polymerization as a facile method to fabricate NIR CPNPs for bioimaging.

Magnetic nanoparticle carrying verbascoside suppresses oral squamous cell carcinoma progression by targeting ICAM1 via IL-6/STAT3 axis

Mifang Yang,Jingjing Wu,Yuan Jing,Siyu Chen,Rongrong Cao,Heming Wu,Hongming Du,Zhirui Guo,Fan Zhang,Wei Shu,Yi Yuan

Oral squamous cell carcinoma (OSCC), the most common oral malignancy, requires innovative therapeutic strategies. We developed a magnetic nanoparticle carrying verbascoside (VB-Fe 3 O 4 ) to enhance the bioavailability and antitumor efficacy of VB, a bioactive compound from Rehmannia glutinosa. VB-Fe 3 O 4 demonstrated superior tumor-targeting capabilities, achieving higher drug accumulation compared with free VB. In the human OSCC cell line HSC-3, VB-Fe 3 O 4 markedly induced apoptosis, inhibited proliferation and invasion through IL-6/STAT3 pathway. Mechanistic studies screened out intercellular cell adhesion molecule 1 (ICAM1) as the primary target of VB, with VB-Fe 3 O 4 suppressing IL-6-induced STAT3 phosphorylation. VB-Fe 3 O 4 downregulated metastasis-related genes, such as vascular cell adhesion molecule 1, mucin 1, ezrin, and invasion factor matrix metalloproteinase-9, were markedly decreased by VB-Fe 3 O 4 . VB-Fe 3 O 4 showed greater tumor suppression in OSCC transplanted tumors and experimental metastasis tumors compared with free VB. This nanoplatform synergistically combines precision delivery and molecular targeting, offering a promising treatment for OSCC management.

6-Shogaol ameliorates experimental colitis and colitis-associated colorectal cancer in mice through the Nrf2-Keap1 signaling pathway

Na Li,Hehe Shi,Wei Du,Siqian Luo,Xianyue Yang,Xinyue Yang,Bingyue Jiang,Peng Xia,Guangji Zhang,Wanghui Jing

The progression from ulcerative colitis to colitis-associated colorectal cancer (CAC) represents a significant clinical challenge. 6-Shogaol, a bioactive phenolic compound of ginger, exhibits anti-inflammatory, antioxidant, and anti-tumor properties. This study aims to investigate the therapeutic efficacy of 6-shogaol against colitis and CAC, and explore the underlying mechanisms through network pharmacology and in vivo validation. Putative hub targets and pathways associated with 6-shogaol were identified through network pharmacology. Murine models of dextran sulfate sodium (DSS)-induced colitis and azoxymethane/DSS-induced CAC were established, and the mice were treated with 6-shogaol to assess the latter’s therapeutic effects. The targets of 6-shogaol predicted through network pharmacology were mainly associated with oxidative stress and reactive oxygen species (ROS) signaling pathways. I n vivo , 6-shogaol relieved colitis symptoms and reduced CAC tumor burden. Furthermore, 6-shogaol alleviated intestinal inflammation by decreasing the colonic levels of TNF-α and MPO, and reduced oxidative stress in the colon tissues, as indicated by lower ROS production and malondialdehyde content. Moreover, 6-shogaol restored intestinal barrier function by upregulating claudin-4, occludin, and ZO-1, and decreasing intestinal permeability. Mechanistically, 6-shogaol promoted Nrf2 nuclear translocation by upregulating nuclear Nrf2 expression while downregulating cytoplasmic Keap1, thereby activating antioxidant response element driver genes, including Hmox1 , Nqo1 , and Gclc . Molecular docking confirmed strong binding affinity between 6-shogaol and Keap1 receptor, with a low binding energy of -6.89 kcal/mol. These findings collectively suggested that 6-shogaol alleviated colitis and CAC in murine models by inhibiting colonic inflammation, relieving oxidative stress, and enhancing intestinal barrier function through the activation of the Nrf2-Keap1 pathway.
