Asymmetric mesoporous silica nanoparticles with anisotropic geometry and dual-compartments are highly desired for loading and release of dual-drugs in separated storage spaces. In this study, an asymmetric lollipop-like mesoporous silica nanoparticle Fe 3 O 4 @SiO 2 &EPMO (EPMO = ethane bridged periodic mesoporous organosilica) was successfully developed via an anisotropic epitaxial growth strategy. The asymmetric nanoparticles show a uniform lollipop shape with a head of spherical Fe 3 O 4 @SiO 2 core-shell that is 200 nm in diameter and a tail of EPMO nanorods with a length of ∼90 nm, and a specific surface area of ∼650.3 m 2 g −1 . Most importantly, the asymmetric nanoparticles possess the unique dual independent (hydrophilic/hydrophobic) spaces with good loading capacities and are significantly more efficient for cancer cell killing than pure drug based on in vitro studies. Additionally, the dual-drug-loaded nanoparticles exhibited excellent antibacterial activity.

Alcohol content is one of the most important quality control metrics in Chinese liquor production, and its accurate determination is highly desired in safeguarding product quality, consumer health, and tax assessment. This study presents an innovative fluorescence sensing platform for alcohol content determination. Curcumin was used as a fluorescent indicator, which demonstrated distinct fluorescence responses to hydrogen bonding interaction with ethanol and water. Experimental results demonstrated an excellent linear correlation between fluorescence intensity changes and alcohol content (20–80 %, vol%) with a strong correlation coefficient (R 2 = 0.991), excellent precision (relative standard deviation <1.67 %) and accuracy (recovery rate 96.78–102.79 %, relative error ± 3.22 %). Further integrated with smartphone technology, a portable fluorescence sensing platform was developed, which enables accurate alcohol content determination through RGB analysis of fluorescent images. This novel method demonstrates promising potential in practical applications, offering a portable and user-friendly solution for alcohol content determination in food and beverage industries.

Targeted and stimuli-responsive drug delivery systems (DDS) hold significant potential for improving therapeutic efficacy and mitigating adverse effects in cancer treatment. Here, we developed a pH-responsive nanocarrier through a surface modification strategy, which enables active targeting of hepatocellular carcinoma (HCC) cells for the delivery of drug curcumin (Cur), a poorly water-soluble therapeutic agent. Briefly, a polydopamine (PDA) layer was first deposited onto the surface of cellulose nanocrystals (CNCs) via the self-polymerization of dopamine hydrochloride. Subsequently, 3-carboxyphenylboronic acid (CA) was grafted onto the PDA-coated surface to obtain CNCs@PDA-CA. After that, Cur was loaded onto CNCs@PDA-CA by forming a boronate ester linkage with the boronic acid groups of CA, resulting in the final nanocomposite CNCs@PDA-CA-Cur with a drug loading capacity of 10.58 ± 1.32 %. This nanocarrier not only exhibited active targeting ability toward HCC cells, but also achieved controlled Cur release in response to the acidic tumor microenvironment. Specifically, CNCs@PDA-CA-Cur showed high pH-responsive ability that released 3.18-folds of Cur in acidic environments (pH = 5.4) more than in neutral environments (pH = 7.2). Furthermore, the nanocarrier demonstrated efficient active targeting to HCC cells, leading to elevated intracellular Cur accumulation and enhanced cytotoxicity in both 2D and 3D HCC models in vitro. Collectively, these results highlight the potential of the targeted and pH-responsive nanocarrier to improve the therapeutic delivery of poorly soluble drugs for HCC treatment.

Ischemic stroke represents one of the leading causes of disability and death worldwide. Neuroprotection aimed at mitigating oxidative stress and inflammation is crucial for improving the prognosis of patients. However, the inadequate accumulation of drugs at the ischemic site significantly restricts their clinical efficacy. We found that platelet membrane (PLTM)-biomimetic nanosystems loaded with curcumin (Cur@PLTM) and tideglusib (Tid@PLTM) actively targeted the ischemic brain and facilitated transcytosis into the ischemic parenchyma via caveolin-dependent transcytosis, mimicking the recruitment of platelets in damaged cerebral vessels. This represented the first application of tideglusib nanoformulations in treating ischemic stroke, further demonstrating that the therapeutic effects were associated with M2 microglia regulation. Additionally, Cur@PLTM and Tid@PLTM synergistically scavenged reactive oxygen species (ROS) and promoted the secretion of neuroprotective cytokines via redox and cellular regulatory mechanisms to mitigate ischemia/reperfusion (I/R) injury. Overall, this platelet membrane-biomimetic nanosystem offers a prospective strategy for targeted brain delivery and combined treatment through antioxidative and anti-inflammatory approaches against ischemic stroke.

Objectives: Alzheimer’s disease (AD) is a progressive neurodegenerative disorder marked by cognitive impairment, amyloid-beta (Aβ) accumulation, tau hyperphosphorylation, and chronic neuroinflammation. Despite symptomatic treatments, no current therapies halt disease progression. Traditional Chinese Medicine (TCM) offers potential multi-targeted interventions in complex diseases like AD. In this study, we evaluated the individual and combined neuroprotective effects of curcumin and Glycyrrhiza glabra in a D-galactose sodium nitrite-induced mouse model of AD. Behavioral analysis, biochemical assays, and molecular profiling were conducted to assess cognitive function, inflammatory cytokine expression, oxidative stress, and AD-related protein levels. Methods: Mice were randomly assigned to five groups: control, AD model, curcumin-treated, Glycyrrhiza glabra-treated, and combination-treated groups. Cognitive function was assessed using the Morris water maze. Levels of neuroinflammatory cytokines interleukin-6(IL-6) and tumor necrosis factor-alpha (TNF-α), AD-related proteins amyloid precursor protein (APP) and microtubule-associated protein tau (MAPT),oxidative stress (MDA), and antioxidant capacity (SOD) were evaluated via quantitative PCR and Western blotting. Results: Curcumin alone modestly improved spatial learning and reduced IL-6 (p < 0.05), whereas Glycyrrhiza glabra had limited effects. The combination therapy yielded the strongest outcomes, significantly reducing escape latency (p < 0.01), IL-6 and TNF-α levels (p < 0.01 and p < 0.05), and downregulating APP and MAPT expression. Additionally, oxidative damage was attenuated, as indicated by decreased MDA and elevated SOD activity (p < 0.001). Although direct measurement of TLR4/NF-κB phosphorylation was not performed, the observed anti-inflammatory and antioxidant effects suggest possible modulation of this pathway. Conclusion: Co-administration of curcumin and Glycyrrhiza glabra exerts synergistic neuroprotective effects by attenuating neuroinflammation, oxidative stress, and AD-related protein expression, surpassing monotherapy outcomes. These findings suggest a synergistic neuroprotective mechanism via modulation of TLR4/MyD88/NF-κB and redox pathways. This study provides preclinical evidence supporting the development of TCM-based combination strategies for AD intervention.

N,S-doped carbon dots (N,S-CDs) were fabricated by utilizing m-phenylenediamine and β-mercaptoethylamine as precursors and designed as colorimetric/fluorometric dual-mode nanosensors. The curcumin (CUR) could quench the green fluorescence around 509 nm. The mechanism of fluorescence quenching was mainly attributed to the static quenching effect. Meanwhile, the color alterations of sensing solution were acquired with CUR contents changing from 0 to 50 μM. Thereby, the quantitative measurement of CUR could be materialized based on colorimetric and fluorometric dual-mode sensing methods. The wide linearity and low detection limit (LOD) were achieved through this dual-mode nanosensor. The feasibility of this system was continuously confirmed via the assay of CUR in fruit samples. Moreover, a portable smartphone-assisted detection system based on N, S-CDs suggested a facile, inexpensive, and efficient approach for detecting CUR in actual fruit samples.

Inflammatory bowel disease (IBD) is a continuously increasing chronic inflammation, and it has been a worldwide health threat. Nevertheless, long-term treatments for IBD remain limited in clinical application. Curcumin is an ideal anti-oxidation and anti-inflammatory drug candidate for chronic inflammation and yet is still pharmacologically safe. However, its clinical application is limited by low solubility and poor bioavailability. To this end, we developed a colon-targeted curcumin carrier using β-glucan, sodium alginate (Alg) and chitosan (CS) named bionic yeast (BY) to improve the anti-inflammatory activity in IBD therapy. Saccharomyces cerevisiae -derived β-glucan greatly enhanced the bio-accessibility and colon-targeting release ability of curcumin. Moreover, the cell viability of bionic yeast demonstrated almost no toxicity on Caco-2 and RAW 264.7cells. The NO, iNOS, and proinflammatory factor including TNF-α, IL-1β, IL-6 were measured in LPS-induced RAW 264.7 cell line. These results indicated that bionic yeast containing β-glucan has a more pronounced anti-inflammatory effect than curcumin does. Therapeutic efficacy of bionic yeast was evaluated in DSS-induced IBD mouse model. It maintained the body weight, liver and spleen index triggered by DSS. The colon proinflammatory factor and inflammatory cell infiltration were significantly alleviated after receiving bionic yeast treatment. Furthermore, the bioavailability of bionic yeast was determined by pharmacokinetics. Compared with free curcumin, the concentration of bionic yeast increased with 11.8-fold in bloodstream and 14.1-fold in colon, demonstrating that bionic yeast is readily absorbed after oral administration. This bionic yeast delivery system combines the immune-enhancing properties of β-glucan with the colon-targeting capabilities of Alg and CS, bridging the gap in ‘natural carrier synergistic therapy’ and addressing the challenge of long-term management in acute bowel inflammation.

Diabetic wound healing remains a challenge due to high levels of oxidative stress, excessive inflammation, and bacterial infection. Smart dressings loaded with natural active monomers are proving to be effective strategies for enhancing diabetic wound healing. Herein, the bio-composites (PTIGA-Cur and PTIGA-Cur-Ag) with curcumin (Cur) responsive release were reported for promoting angiogenesis and diabetic wound repair, showing excellent anti-inflammatory, antioxidant, and antibacterial properties. The integration of three-dimensional networks and chemical bonds endowed the bio-composites with superior thermodynamic, mechanical, and self-healing properties. Notably, pH-responsive Schiff base as well as ester groups in the matrix enable the Cur to be released in a controlled manner. A biosensor assembled from PTIGA-Cur-Ag demonstrated electronic conductivity based on in-situ synthesis of AgNPs, which enabled sensitive monitoring of blood glucose levels. In addition, the release of AgNPs enhanced the antibacterial and anti-inflammatory properties. Expectedly, the bio-composites exhibited remarkable biocompatibility, effectively promoting the polarization of macrophages to M2 phenotype, and reducing the expression of proinflammatory cytokines. The full-thickness diabetic wound model revealed that PTIGA-Cur and PTIGA-Cur-Ag were able to effectively promote collagen deposition, neovascularization, and granulation tissue regeneration through their anti-inflammatory, antioxidant, and antibacterial properties. This study provides evidence supporting the potential utility of bio-composites with both pro-healing properties and monitoring functions in the management of diabetic wounds.

Developing environmentally friendly delivery systems capable of encapsulating, protecting, and promoting the absorption of bioactive components in the human body is crucial for those with low oral bioavailability and poor stability. This study developed W/O/W emulsion-filled sodium alginate (SA) hydrogel for the co-delivery of hydrophilic riboflavin and hydrophobic curcumin. Firstly, W/O/W emulsion was prepared using peanut protein isolate (PPI)-bacterial cellulose nanofibers (BCNFs) as the hydrophilic emulsifier and lecithin as the lipophilic emulsifier, and the effect of stabilizer concentrations on emulsion stability was investigated. Results showed that PPI and BCNFs self-assembled into stable complexes through electrostatic interaction and hydrogen bonding, thereby improving the emulsification properties of PPI. Subsequently, SA was added to the outer aqueous phase of emulsions, and its influence on emulsion properties and stabilization mechanism was investigated. SA significantly improved the stability and rheological properties of PPI-BCNFs-stabilized W/O/W emulsions. Finally, the loading capacity of this system for riboflavin and curcumin and its gastrointestinal fate were evaluated. We found this delivery system efficiently co-delivered both hydrophilic and hydrophobic bioactive components and improved their photostability and thermal stability. Moreover, the bioaccessibilities of riboflavin and curcumin were significantly increased to 61.97% and 45.66%, respectively. This was attributed to SA forming a composite interfacial layer on droplets and a dense gel network in the continuous phase, thereby acting as a physical barrier to better protect the encapsulated components. This delivery system holds promise for the development of functional foods incorporating multiple nutraceuticals.

Illegal addition of curcumin (Cur) in food poses a significant public health threat, creating an urgent need for sensitive, reliable detection methods. In this work, dual-emission carbon dots (DCDs) were synthesized via a one-step hydrothermal method with Rose Bengal and melamine as raw materials. Based on the distinct fluorescence responses of DCDs to Cur at two characteristic wavelengths, a novel ratiometric fluorescence sensor for Cur detection was constructed. The emission at 432 nm is primarily governed by inner filter effect, while quenching at 532 nm is associated with the fluorescence resonance energy transfer. The sensor shows a robust linear range of 0.1–20 μM with a limit of detection of 0.089 μM. The sensor was successfully employed for the detection of Cur in various food samples, including chili, ginger, and mustard powder, achieving recovery rates between 92.75% and 110.00%. This work enriches CDs synthesis strategies and provides a sensitive, cost-effective, rapid method for Cur detection, supporting food industry monitoring needs.

Purpose Current antidepressants are limited by insufficient efficacy of conventional monoaminergic drugs and poor brain penetration across the blood-brain barrier. This study designed pure curcumin loaded lipid nanoparticle (CNP) with optimized brain-targeting delivery for depression therapy. Methods Cur molecules first self-assembled into carrier-free drug nanoparticles. Subsequently, CNP were then prepared via thin-film dispersion and fully characterized in terms of particle size, PDI, DSC, XRD, TEM. The antidepressant effect of CNP was systematically investigated via in vitro and in vivo assays, including cellular uptake, LPS-induced stress model in BV2 cells, and in vivo CUMS depression model. Results CNP displayed uniform spherical morphology with an average size of 115.8 ± 18.3 nm, PDI of 0.216 ± 0.015 and zeta potential of −27.1 mV, along with high encapsulation efficiency (86.11 ± 4.28%), drug loading (6.62 ± 0.45%) and sustained release behavior. The cellular uptake efficiency of the CNP group reached 41.47 ± 1.45%, which was more than double that of the Cur group (17.21 ± 0.54%). In vitro studies showed that CNP not only rescued the viability of cells damaged by corticosterone and hydrogen peroxide but also exerted significantly enhanced anti-inflammatory and antioxidant effects in lipopolysaccharide induced cellular stress models. In vivo studies indicated that CNP alleviated depressive-like behaviors more effectively. Conclusion CNP exhibits significantly enhanced antidepressant efficacy, thus providing a promising approach for developing brain-targeted therapeutics for MDD.

Corneal neovascularization (CoNV) is a major cause of visual impairment and presents significant challenges in corneal transplantation. Current treatments are limited by side effects, poor bioavailability, and the need for frequent administration. Curcumin, a natural polyphenol with potent anti-inflammatory, antioxidant, and anti-angiogenic properties, shows promise for CoNV therapy but suffers from low water solubility and rapid clearance. To address these limitations, we developed an injectable self-healing hydrogel incorporating curcumin-copper nanoparticles (CurNPs) within a thiolated alginate (SH-Alg) and oxidized dextran (ODex) matrix. This hydrogel provides localized and sustained drug release, enhancing curcumin bioavailability while minimizing systemic exposure. Its self-healing property ensures mechanical stability and consistent therapeutic delivery. In an alkali burn-induced rat model of CoNV, the OS/CurNPs (ODex/SH-Alg/CurNPs) hydrogel significantly reduced oxidative stress and inhibited corneal neovascularization, demonstrating superior antioxidant and anti-angiogenic efficacy. These results highlight the potential of this hydrogel system as a minimally invasive strategy for treating CoNV and other oxidative stress–related corneal disorders.
