Introduction The study aimed to explore the association of serum 25(OH)D 3 and hepatic steatosis in non-alcoholic fatty liver disease (NAFLD) patients and to determine whether the effect of vitamin D (VD) is mediated by activation of the peroxisome proliferator-activated receptor α (PPARα) pathway. Methods The study contained a case-control study, in vivo and in vitro experiments. A case-control study was conducted to compare serum parameters between NAFLD patients and controls and to evaluate the association of 25(OH)D 3 and NAFLD. In vivo study, male Wistar rats were randomly divided into control and model groups, fed a standard chow diet and a high-fat diet (HFD), respectively, for 7 weeks to generate an NAFLD model. Then, the rats were treated with VD and a PPARα antagonist (MK886) for 7 weeks. Tissue and serum were collected and assessed by biochemical assays, morphological analysis, histological analysis, and western blot analysis. In vitro , HepG2 cells were incubated with oleic acid (OA) to induce steatosis, which was evaluated by staining. HepG2 cells were pretreated with MK886 followed by calcitriol treatment, and differences in lipid metabolism-related proteins were detected by western blot. Results NAFLD patients were characterized by impaired liver function, dyslipidemia, and insulin resistance. Serum 25(OH)D 3 was negatively associated with alanine aminotransferase (ALT) in NAFLD. VD deficiency was a risk factor for patients with no advanced fibrosis. Adequate VD status (25(OH)D 3 >20 ng/mL) had a protective effect in patients after adjustment for confounding variables. NAFLD rats showed hyperlipidemia with severe hepatic steatosis, systematic inflammation, and lower serum 25(OH)D 3 . VD treatment ameliorated hepatic steatosis both in NAFLD rats and OA-induced HepG2 cells. Further, MK886 inhibited the anti-steatosis effect of VD. Conclusion The study revealed that an adequate VD level may act as a protective factor in NAFLD and that VD may alleviate hepatic steatosis via the PPARα signaling pathway.

Herein, the synergistic inhibition of two typical corrosion inhibitors benzotriazole (BTA) and sodium D -gluconate (SD) on the corrosion of Q235 steel in simulated concrete pore solution (SCP) were systematically studied. The results demonstrated that the mixed BTA/SD showed an excellent anti-corrosion performance and the inhibition efficiency of (BTA/SD, 1ː1) was as large as 98.33 % which is higher than that of BTA (88.65 %) and SD (92.16 %). Furthermore, the synergistic inhibition effect of BTA/SD was also researched by surface analysis technologies, the X-ray photoelectron spectroscopy (XPS) result verified that the BTA/SD adsorbed on steel, while both scanning electron microscopy (SEM) and atomic force microscopy (AFM) images proved that the mixed BTA/SD can effectively protect the steel from corrosion in SCP solution which containing corrosive ions. In addition, the anti-corrosion mechanism of the mixed BTA and SD was revealed by adsorption model and molecular dynamics (MD) simulation, the mixed BTA/SD corrosion inhibitors showed the largest K ads , the lowest ∆ G ads and the largest binding energy values , demonstrating the strong adsorption on steel and the best anti-corrosion performance. The results of this work can provide experimental and theoretical references for the corrosion protection of reinforced concrete structures while using synergistic inhibitors.

D-amino acid (DAA) an environmentally friendly biofilm inhibitor, has low efficiency for membrane biofouling control due to its instability. To address this challenge, a novel nanocomposite was prepared with DAA adhering to polydopamine (PDA)-coated halloysite nanotube (HNTs) through the interactions of H-bonding and π-π stacking between PDA and DAA. Membrane modified with such nanocomposite was fabricated via blending phase inversion, and the nanocomposite was uniformly distributed in the modified membrane matrix. In comparison with the pristine membrane, the addition of nanocomposites resulted in robust mechanical property for modified membrane with the ultimate stress and strain increased by 23.97% and 35.62%, respectively. Moreover, an excellent tradeoff between water flux (2.5 folds of pristine membrane) and selectivity was achieved, probably due to the improved membrane hydrophilicity. Meanwhile, bovine serum protein (BSA) static adsorption as well as dynamic filtration experiments exhibited excellent antifouling ability of the modified membrane. Most importantly, a superior anti-biofouling stability over a 10-day period was obtained for the membrane modified with nanocomposite, indicating that the activity of DAA to mitigate biofouling was effectively maintained. This study developed a novel and promising strategy for membrane biofouling mitigation.

Acute liver failure (ALF) is a life-threatening clinical condition resulting from massive apoptosis of hepatocytes. This study aims to examine the protective effects of Lactobacillus plantarum C88 on LPS/D-GalN-induced ALF and elucidate its underlying molecular mechanisms in mice. The results showed that L. plantarum C88 pretreatment significantly reduced the levels of plasma ALT and AST in mice with LPS/D-GalN–induced liver injury. L. plantarum C88 pretreatment decreased the secretion of inflammatory cytokines. In addition, L. plantarum C88 remarkably increased the expression levels and nuclear translocations of Nrf2 mRNA in liver tissues, but inhibited NF-κB nuclear translocations and P65, P38 phosphorylation levels. Moreover, L. plantarum C88 enhanced the intestinal mucosal barrier by upregulating the expressions of tight junction proteins as well as HIF-1α. These results suggest that L. plantarum C88 protects against ALF associated with Nrf2 and the MAPK/NF-κB signaling pathways.

A series of novel C15 urushiol derivatives were designed by introducing a pechmann structure and F-, Cl-, and Br-nitro substituents with different electronic properties into its alkyl side chain, as well as a triazolyl functional group in its aromatic oxide. Their chemical structures were determined based on the analysis of the NMR (nuclear magnetic resonance) spectroscopic and mass spectrometric data. The results showed that compound4exhibited a strong inhibition of the HepG2 cell proliferation (half maximal inhibitory concentration (IC50): 2.833 μM to human hepatocellular carcinoma (HepG2), and 80.905 μM to human normal hepatocytes (LO2)). Furthermore, it had an excellent synergistic effect with levopimaric acid. The nitrogen atom of the triazole ring formed a hydrogen-bonding interaction with Gly103, Gly154, and Tyr308, which made compound4bind to histone deacetylase (HDAC)2 more tightly. One triazole ring and His33 formed a π–π stacking effect; the other, whose branches were deep into the pocket, further enhanced the interaction with HDAC2. Meanwhile, compound4involved a hydrophobic interaction with the residues Phe210 and Leu276. The hydrophobic interaction and π–π stacking provided powerful van der Waals forces for the compounds.Keywords:urushiol;triazole;diels-alder;pechmann;anti-tumor;HDAC2

Papain-stabilized gold nanoclusters ( [email protected] ) with a red fluorescence emission at 639 nm have been generated successfully in aqueous solution. The fluorescence of [email protected] could be quenched in the addition of Cu 2+ . Subsequently, a unique fluorescent probe based on [email protected] 2+ complex has been constructed for sensitive, selective and “turn-on” detection of D-penicillamine (D-pen). The sensing probe has exhibited a remarkable fluorescence enhancement in the presence of D-pen, which can be measured ranging from 30.0 μM to 2.0 mM (r 2 = 0.991) with the detection limit of 5.0 μM. Furthermore, the developed assay has been utilized to explore the metabolic process of D-pen in rats after intraperitoneal injection. As far as we concerned, the present study has reported the first analytical method for sensing serum D-pen in real time by using the fluorescent [email protected] 2+ complex. Our strategy paves a way for digging up the biological and clinical application of the fluorescent AuNCs.

Electrochromic (EC) devices with reversible transmittance switching have promising prospects in the applications of photoelectric displays. Herein, a novel air-stable and high contrast solution-phase electrochromic device based on new viologen derivative (DPB) with A-D-A structure (A: acceptor; D: donor) was demonstrated. Devices possessed simple configuration of ITO/EC solution/ITO. Ferrocene and polycarbonate (PC) were selected as the complementary material and solvent, respectively. All of the fabrications and performance testing work were conducted in normal air condition. The DPB-based device showed deep magenta color in colored state and exhibited high optical contrast of ΔT of 72.2% (517 nm) and good coloration efficiency of 102.2 cm 2 /C at 1.8 V. The device appeared only slight degradation of ΔT (ΔT = 69.6% or retained 96.7% of its initial ΔT after 5000 s constant switching) under the normal air atmosphere, which was even better than some reported well-packaged viologen-based solution ECDs. Additionally, the UV–vis reflective spectra were also conducted to assess the reflectance property of the devices. The reflectance peak (ΔR = 2.6%) and absorption peak matched accurately, which indicated that the new viologen derivative possesses the potential application in reflective electrochromic devices.

Biodegradable organic-inorganic composites composed of polylactide (PLA) and hydroxyapatite (HA) are important bone repairing materials, while the dispersibility of nanoscaled HA in PLA and the interfacial adhesion between HA and PLA remained unsatisfactory. In this study, poly(D-lactide) (PDLA) oligomers with different molecular weights were grafted onto HA nanorods (HA-PDLA), and the HA-PDLA hybrids were mixed with poly(L-lactide) (PLLA). Dispersibility of HA-PDLA hybrids in PLLA matrix was investigated, and the formation of PDLA/PLLA stereocomplex at the interface of HA-PDLA and PLLA was studied by characterizations including crystallization, mechanical and thermal properties, taking PLLA grafted HA (HA-PLLA) and unmodified HA as comparisons. Surface grafting of PLLA or PDLA oligomers both significantly improved the dispersibility of HA nanorods in PLLA matrix. Benefiting from PDLA/PLLA stereocomplexation, HA-PDLA could increase the tensile strength and the elongation of resulting PLLA/HA nanocomposites more significantly than HA-PLLA. The interfacial interaction between HA-PDLA and PLLA matrix could be further strengthened by increasing the molecular weights of those grafted PDLA chains on HA, which was verified by changes in crystallization and glass transition behaviors. To sum up, the concept of using PDLA/PLLA stereocomplexation was feasible and effective in preparing PLA-based organic-inorganic nanocomposites targeting bone repairing.

Two pairs of amino-acid functionalized poly(3,4-ethylenedioxythiophene) (PEDOT) derivatives, namely, poly( N -(tert-butoxycarbonyl)- L -methionyl (3,4-ethylenedioxythiophene-2’-yl)methylamide) ( L -PEDOT-Boc-Met) and poly( N -(tert-butoxycarbonyl)- D -methionyl (3,4-ethylenedioxythiophene-2’-yl)methylamide) ( D -PEDOT-Boc-Met); poly( L -methionyl (3,4-ethylenedioxythiophene-2’-yl)methylamide) ( L -PEDOT-Met) and poly( D -methionyl (3,4-ethylenedioxythiophene-2’-yl)methylamide) ( D -PEDOT-Met) were synthesized via chemical oxidative polymerization of corresponding monomers. The structural characterization, spectroscopic properties and thermal stability of these monomers and polymers were systematically explored by FTIR spectra, Raman spectra, XRD spectra, UV-Vis spectra and thermogravimetric analysis. As chiral electrode materials, these polymers were employed to successfully recognize 3,4-dihydroxyphenylalanine (DOPA) enantiomers by cyclic voltammetry (CV) in sulphuric acid solution. The measurement results reveal that the tendency was hetero-chiral interaction between L -PEDOT-Met/PVA/GCE and D -DOPA, D -PEDOT-Met/PVA/GCE and L -DOPA, respectively. Also, the mechanism of chiral discrimination was discussed. All the results implied that the combination of electrochemical molecular recognition technology and chiral PEDOT materials can be a promising approach for chiral recognition and may open new opportunities for facile, biocompatible, sensitive and robust chiral assays in biochemical applications.

Electrochemical sensors now play an important role in analysis and detection of nucleic acids. In this work, we present a novel double-signal technique for electrochemically measuring the sequence and length of the d(CAG)n repeat. The double-signal technique used an electrochemical molecular beacon (a hairpin DNA labeled with ferrocene), which was directly modified on the surface of a gold electrode, while a reporter probe (a DNA sequence labeled with horseradish peroxidase) was hybridized to the target DNA. First a simple single-signal sensor was characterized in which d(CAG)n repeats were detected using a short reporter DNA strand labeled with horseradish peroxidase. To obtain a reliable signal that was dependent on repeat number, a double-signal biosensor was created in which the single strand capture DNA in single-signal sensor was replaced by an electrochemical molecular beacon labeled with ferrocene. When the hairpin DNA hybridized to the target–reporter DNA complex, it opened, resulting in a decreased ferrocene current. Both electrochemical biosensors exhibited high selectivity and sensitivity with low detection limits of 0.21 and 0.15 pM, respectively, for the detection of d(CAG)n repeats. The double-signal sensor was more accurate for the determination of repeat length, which was measured from the ratio of signals for HRP and ferrocene (H/F). A linear relationship was found between H/F and the number of repeats (n), H/F = 0.1398n + 9.89788, with a correlation coefficient of 0.974. Only 10 nM of target DNA was required for measurements based on the value of H/F in the double-signal technique. These results indicated that this new double-signal electrochemical sensor provided a reliable method for the analysis of CAG trinucleotide repeats.

Photoirradiation surface molecularly imprinted polymers for the separation of 6- O -α- d -maltosyl-β-cyclodextrin were synthesized using functionalized silica as a matrix, 4-(phenyldiazenyl)phenol as a light-sensitive monomer, and 6- O -α- d -maltosyl-β-cyclodextrin as a template. Fourier transform infrared spectroscopy results indicated that 4-(phenyldiazenyl)phenol was grafted onto the surface of functionalized silica. The obtained imprinted polymers exhibited specific recognition toward 6- O -α- d -maltosyl-β-cyclodextrin. Equilibrium binding experiments showed that the photoirradiation surface molecularly imprinted polymers obtained the maximum adsorption amount of 6- O -α- d -maltosyl-β-cyclodextrin at 20.5 mg/g. In binding kinetic experiments, the adsorption reached saturation within 2 h with binding capacity of 72.8%. The experimental results showed that the adsorption capacity and selectivity of imprinted polymers were effective for the separation of 6- O -α- d -maltosyl-β-cyclodextrin, indicating that imprinted polymers could be used to isolate 6- O -α- d -maltosyl-β-cyclodextrin from a conversion mixture containing β-cyclodextrin and maltose. The results showed that the imprinted polymers prepared by this method were very promising for the selective separation of 6- O -α- d -maltosyl-β-cyclodextrin.

In this work, a unique D-amino acid oxidase reactor for enhanced enzymolysis efficiency is presented. A kind of magnetic polymer matrices, composed of iron oxide nanoparticles and porous polymer membrane (poly styrene-co-maleic anhydride), was prepared. With covalent bonding D-Amino acid oxidase on the surface of the matrices and characterization of scanning electron microscope and vibrating sample magnetometer, it demonstrated that the membrane enzyme reactor was successfully constructed. The enzymolysis efficiency of the enzyme reactor was evaluated and the apparent Michaelis-Menten constants of D-Amino acid oxidase were determined ( K m was 1.10 mM, V max was 23.8 mM min −1 ) by a chiral ligand exchange capillary electrophoresis protocol with methionine as the substrate. The results indicated that the enzyme reactor could exhibit good stability and excellent reusability. Importantly, because the enzyme and the substrate could be confined into the pores of the matrices, the enzyme reactor displayed the improved enzymolysis efficiency due to the confinement effect. Further, the prepared enzyme reactor was applied for D-Amino acid oxidase inhibitors screening. It has displayed that the proposed protocol could pave a new way for fabrication of novel porous polymer membrane based enzyme reactors to screen enzyme inhibitors .
