Antioxidant activity, storage stability and in vitro release of epigallocatechin-3-gallate (EGCG) encapsulated in hordein nanoparticles

Hongdong Song,Qingyu Wang,Aijing He,Sen Li,Xiao Guan,Yawen Hu,Siyi Feng

Hordein nanoparticles have been successfully fabricated to encapsulate EGCG. The present study aimed to investigate the effect of hordein nanoparticles encapsulation on antioxidant activity, storage stability and in vitro release of EGCG. The antioxidant activities of EGCG before and after encapsulation by hordein nanoparticles had no significant difference. The 28-day storage experiments indicated that EGCG-hordein nanoparticles had a good storage stability in terms of average diameter and zeta potential. Meanwhile, the stability of EGCG was significantly improved by hordein nanoparticles encapsulation. The remaining amount of encapsulated EGCG was 1.31-fold and 1.52-fold higher than that of free EGCG at 4 °C and 25 °C, respectively. The release study in the simulated gastric or intestinal fluids suggested that hordein nanoparticles could slow down the release of EGCG in the simulated gastric or intestinal fluids. These results showed that encapsulation of EGCG using hordein nanoparticles could be a promising approach to improve its stability.

A consecutive extraction of pectin and hesperidin from Citrus aurantium L.:Process optimization, extract mechanism, characterization and bio-activity analysis

Peng Zhou,Min Zheng,Xiangzhou Li,Jun Zhou,Yaxuan Shang,ZhaoShuang Li,Limin Qu

With the proposal of the Paris Climate Agreement and double carbon policy, production enterprise is now facing green and low-carbon transformation. To promote green and low-carbon development, optimizing the production process and improving utilization rate of raw materials for the reduction of production cost is an significant technological path for the plant extract industry. Thus, the aim of this work was to develop a consecutive extraction process of pectin and hesperidin with lower cost and higher purity, as well as better properties of extract. The process parameters will be put forward through single-factor experiments, Plackett-Burman design and Box-Behnken design. The experimental results showed that the consecutive extraction process was effective and feasible. Pectin with excellent properties was obtained, and the solid-liquid ratio of extraction process parameters of hesperidin was reduced to 1:8 (g: mL). The problems of environmental pollution and high energy consumption in traditional extraction could be solved. The consecutive extraction process, an innovative, economic and environmental-friendly process, may be realized easily in industrial application. At the same time, the consecutive extraction process mechanism was revealed with the help of microscopy analysis of the plant tissue structure. The results demonstrated that preferential extraction of pectin process was helpful for the exposure and dissolution of hesperidin crystallization in cells and thus can improve extraction efficiency of hesperidin. The purity of the obtained hesperidin was up to 91.7%, which can be further purified depending on application requirements. A large amount of pectin was obtained (23.3% ± 0.5% yield), and the pectin has superior gel-property, strong antioxidant activity and certain antibacterial activity. The pectin and hesperidin obtained will have better application prospects in medicine and food because of their unique characteristics.

Antioxidant activity of delphinidin and pelargonidin: Theory and practice

Hongan Li,Chengyue Zhang,Zeyuan Deng,Bing Zhang,Hongyan Li

The quantum chemical density functional theory and in vitro chemical-based antioxidant assays were used to research the reaction mechanism of delphinidin/pelargonidin with free radicals including superoxide anion radicals (O 2 − ∙) and hydroperoxide radicals (OOH∙). The geometric configuration, bond dissociation energy, PCM (polarizable continuum model) solvent model reaction enthalpy changes were studied to explain the transition states, and the reaction enthalpy change value was calculated to determine the active site. From the results of spatial configuration, delphinidin showed a stronger conjugation effect than that of pelargonidin. The dihedral angle between the three rings of delphinidin was almost 180°, and the angle between the B and C rings was only −2.81868°. Both coplanar and antioxidant activity of delphinidin was better than pelargonidin. The consequences of reaction enthalpy change in PCM were consistent with the bond dissociation energy. The phenolic hydroxyl bond dissociation energy of delphinidin was slightly smaller than that of pelargonidin. Moreover, the C4' site of delphinidin and the C3 site of pelargonidin were the active sites for scavenging free radicals. The free radical scavenging ability of delphinidin was marginally higher than that of pelargonidin. On the other hand, in vitro antioxidant results proved the scavenging ability of delphinidin and pelargonidin on superoxide anions, DPPH, and ABTS ∙+ free radicals. It was shown that the chemical-based antioxidant activity was consistent with the theoretical calculation results, with delphinidin showing greater antioxidant activity. These results could explain the antioxidant mechanism of delphinidin/pelargonidin in scavenging free radicals from chemical reactions. Practical applications This manuscript explained the antioxidant mechanism of delphinidin/pelargonidin in scavenging free radicals through the analysis of the geometric configuration of delphinidin/pelargonidin and the theoretical calculation of the reaction transition state. It could also speculate on the possible reaction sites, and provide a basis for judging how to efficiently select antioxidants with great antioxidant activity.

Preservation of cardiac functions post myocardial infarction in vivo by a phenylboric acid-grafted hyaluronic hydrogel with anti-oxidation and accelerated degradation under oxidative microenvironment

Shuqin Wang,Yuejun Yao,Tong Zhou,Jieqi Xie,Jie Ding,Wangbei Cao,Liyin Shen,Yang Zhu,Changyou Gao

Excessive oxidative stress and inflammatory responses can exacerbate the pathological conditions of left ventricular (LV) myocardium and even lead to heart failure post myocardial infarction (MI). Intramyocardial injectable antioxidative hydrogels without loading of any cells, genes or cell growth factors are attractive for the translation in clinical therapy. In this study, a self-adaptive, degradable and antioxidative hydrogel (R gel) was prepared by a Schiff base reaction between adipic acid dihydrazide (ADH)-modified and phenylboric acid (PBA)-grafted hyaluronic acid (HA-ADH-PBA) and oxidized dextran (Ox-Dex) with aldehyde groups. Compared to the non-reactive control hydrogel (N-R gel), the R gel exhibited high efficiency in scavenging excessive ROS in vitro and in vivo , and protected cells from oxidative damage in infarcted rat myocardium in vivo , as demonstrated by significantly reduced cell membrane peroxidation and apoptosis, suppressed inflammation, preserved cardiac functions, and promoted LV vascularization. The expressions of key genes associated with the inflammation in short term (3 d) and cardiomyocyte functions in long term (8 w) were investigated to explore the mechanisms in a molecular level.

Effects of simulated saliva-gastrointestinal digestion on the physicochemical properties and bioactivities of Siraitia grosvenorii polysaccharides

Yuxi Guo,Xuefeng Chen,Pin Gong,Mengrao Wang,Wenbo Yao,Wenjuan Yang,Fuxin Chen

Summary The aim of this study was to investigate the effect of simulated saliva-gastric digestion (SGID) on the physicochemical properties and bioactivity of Siraitia grosvenorii polysaccharides (SGPs). The results revealed that the digestibility (bioavailability) of SGPs was approximately 16.81%, 43.86%, and 52.38% after saliva digestion (SD), saliva-gastric digestion (SGD), and SGID, respectively. SGID significantly altered the physicochemical properties of SGPs, such as total polysaccharide content (from 88.25% to 69.26%) and molecular weight (from 23.00 to 7.82). The structure of SGPs was significantly altered after digestion. In addition, SGID affected the biological activity of SGPs. after SGID, SGPs still had strong antioxidant activity, as well as α -glucosidase, α -amylase inhibition, and cellular hypoglycemic activity. However, the ability to bind bile acids and cholesterol in vitro was not affected. In conclusion, these results could provide a theoretical basis for investigating the mechanism of action of SGPs during digestion in vivo .

Enhanced flow electrochemistry for cyclohexane Conversion: From simulation to application

Yujun Zhang,Hao Tian,Zhaowei Cui,Zhen Yin,Hongsen Hui,Hong Wang,Lei Zhang,Hongchang Pei,Zhenhuan Li,Bhekie B. Mamba,Jianxin Li

To further improve the capability of flow electrochemistry, the multistage plug-flow electrochemical membrane reactor (PF-ECMR) with membrane electrode was developed for enhancing the oxidation of cyclohexane (CHA) to produce KA oil (mixture of cyclohexanol and cyclohexanone). The catalytic results demonstrated that the CHA conversion and the selectivity to KA oil were up to 95.6% and > 99%, which was the highest in comparison with most of the state-of-the-art catalytic processes reported in the previous works. The mechanism was explored by density functional theory (DFT) calculations, indicating the auxo-action effect of cyclohexanone was beneficial for the formation of cyclohexyl hydroperoxide so as to enhance the CHA conversion. The excellent performance of PF-ECMR was also attributed to the staggered electric field and the accumulative effect of the multi-electrodes. This study provided a successful case of a combination of theoretical prediction and experimental verification together for organic synthesis.

Effects of Crosslinking Sericin on the Color Fastness and Antioxidant Activity of Naturally Colored Silk

Lv Zongyu,Tan Ting,Hussain Munir,Zhou Wenlong,Ma Mingbo

Naturally colored silk (NCS) fades easily during wet processing and home washing due to the loss of pigment, which is accompanied by dissolution of the sericin layer. This study proposed a green and effective strategy for fixing the sericin of NCS to improve the color fastness of NCS textiles under wet conditions. The sericin of NCS was crosslinked with a bifunctional epoxy resin, poly(ethylene glycol) diglycidyl ether (PEGDE), in the presence of calcium chloride as a catalyst. The influences of the concentrations of PEGDE and the catalyst, pH, temperature, and reaction time on the crosslinking effect were investigated. The results showed that crosslinking NCS under optimum conditions could result in a 5.11 % weight gain. After crosslinking, the pigment in the sericin layer exhibited a considerable fixation fastness under wet conditions due to the low solubility of the crosslinked sericin. The K/S value of crosslinked NCS decreased by only 15.9 % after washing 15 times, which is much lower than that of uncrosslinked NCS (43.8 %). Crosslinking of NCS also improved its antioxidant capacity retention against repeated home washing. This study proposed an effective sericin-crosslinking strategy that could be used not only for fixing the color of NCS but also for sericin-grafted textiles and sericin-based biomaterials.

An Organic Solvent-Free Method for the Extraction of Ellagic Acid Compounds from Raspberry Wine Pomace with Assistance of Sodium Bicarbonate

Ning Jin,Shouyu Zhang,Shibo Sun,Minghuo Wu,Xiaojing Yang,Jianqiang Xu,Kun Ma,Shui Guan,Weiping Xu

Industrial processing of raspberry juice and wine generates considerable byproducts of raspberry pomace. Ellagic acids/ellagitannins, being characterized by their antioxidant and antiproliferation properties, constitute the majority of polyphenolics in the pomace and are valuable for recovery. In the present study, we developed a novel procedure with sodium bicarbonate assisted extraction (SBAE) to recover ellagic acid from raspberry wine pomace. Key parameters in the procedure, i.e., sodium bicarbonate concentration, temperature, time and solid/liquid (S/L) ratio, were investigated by single factor analysis and optimized subsequently by Response Surface Methodology (RSM). Optimal parameters for the SBAE method here were found to be 1.2% (w/v) NaHCO3, 1:93 (w/v) S/L ratio, 22 min and 100 °C. Under these conditions, the ellagic acid yield was 6.30 ± 0.92 mg/g pomace with an antioxidant activity of 79.0 ± 0.96 μmol Trolox eq/g pomace (DPPH assay), which are 2.37 and 1.32 times the values obtained by extraction with methanol–acetone–water solvent, respectively. The considerable improvement in ellagic acid extraction efficiency could be highly attributed to the reactions of lipid saponification and ellagitannin hydrolysis resulted from sodium bicarbonates. The present study has established an organic solvent-free method for the extraction of ellagic acid from raspberry wine pomace, which is feasible and practical in nutraceutical applications. Keywords:ellagic acid;extraction;raspberry;wine pomace;sodium bicarbonate;response surface methodology

A change-prone zwitterionic hyperbranched terpolymer-based diabetic wound dressing

Xianhua Xie,Xin Jin,Binbin He,Yang Zou,Jumin Yang,Changjun Liu,Xiaoling Kong,Wenguang Liu,Wei Wang

The long inflammation state and susceptibility to infection of a diabetic ulcer impairs wound healing. In this report, an in situ formed hydrogel dressing with antibacterial and anti-inflammatory activities is designed to nurse diabetic wounds. Firstly, a hyperbranched terpolymer (termed as HBPVD) is synthetized by a unique dynamically controllable reversible addition-fragmentation chain transfer (RAFT) polymerization of poly(ethylene glycol) diacrylate (PEGDA), 1-vinylimidazole (VI), and 2-(N-3-sulfopropyl-N,N-dimethyl ammonium) ethyl methacrylate (DMAPS). The resulted HBPVD is equipped with multi-acrylate end group that can be crosslinked with gelatin methacrylate (GelMA) to form a hydrogel in situ under UV light. The imidazole group in VI can chelate metal ions to enhance the mechanical strength of the hydrogels. The introduction of zwitterion DMAPS imparts the hydrogels thermosensitive and anti-fouling capability to get rid of the bacterial debris from wound beds. Meanwhile, the self-assembled ascorbyl palmitate (AP) nanosheet is encapsulated into the hydrogels, affording a strong anti-inflammatory capability and serving as a reservoir of reducing agent. Based on an Ag + -AP in situ -reduction method, Ag + complexed by VI in the hydrogel can be homogeneously reduced to Ag nanoparticles. The rheological results confirm that a solid HBPVD/GelMA hydrogel is obtained by UV light irradiation within 1 min. The strength of hydrogels is sharply increased by complexation of metal ions. A strong anti-bacterial ability against both gram-positive and gram-negative bacteria is achieved in HBPVD/GelMA/Ag/AP hydrogel. The HBPVD/GelMA/Ag/AP hydrogel dressing is administrated on humanized wound beds in diabetic rats, which can be conveniently changed by spraying 4 °C Cu 2+ solution that fleetly triggers both ion-strengthening and thermosensitive responses. The designed dressing presents a faster wound closure ratio with promoting epithelialization and the density of collagen, decreasing the expression level of pro-inflammatory factors (IL-1β, TNF-α), and improving the expression level of anti-inflammatory factor (IL-10). This change-prone hydrogel dressing with antibacterial and anti-inflammatory capability can provide a promising alternative method to repair diabetic wounds.

Grafting resveratrol onto mesoporous silica nanoparticles towards efficient sustainable immunoregulation and insulin resistance alleviation for diabetic periodontitis therapy

Yujie Tan,Jing Feng,Yu Xiao,Chongyun Bao

Host-modulation therapy is generally accepted as a novel promising method for diabetic periodontitis (DP) treatment and screening an appropriate drug model is the key to success. Resveratrol (RSV), because of its viable antioxidative and anti-inflammatory properties and its ability to control glucose metabolism, is considered a potential candidate. However, poor water solubility, rapid decomposition and short serum half-life period significantly limit its application. Therefore, in this study, we designed a RSV-grafted mesoporous silica nanoparticle (MSN-RSV) drug carrier system to enhance RSV's stability effectively and prolong its duration. Further analyses have verified the indispensable role of MSNs in improving the bioavailability of RSV, which could result in a more favorable therapeutic efficacy in DP related to regulating the polarization of the macrophage. The reason for this could be explained by activating the SIRT1/AMPK signaling pathway and inhibiting the NF-κB signaling pathway. This study also focused on the auxiliary effect of MSN-RSV on alleviating insulin resistance (IR) and controlling glucose metabolism. In brief, the study has provided a potential alternative strategy for DP therapy. It is also helpful for future intensive research topics like the immunoregulatory mechanisms in the bidirectional relationship between diabetes and periodontitis.

Phytochemical Compositions and Antioxidant Activities of Essential Oils Extracted from the Flowers of Paeonia delavayi Using Supercritical Carbon Dioxide Fluid

Xiao Yu,Huaibi Zhang,Juan Wang,Junming Wang,Zhenxing Wang,Jinbo Li

Essential oils were extracted from dark-purple, red and yellow petals ofPaeonia delavayiusing Supercritical Carbon Dioxide method. The compositions of essential oils were analyzed using gas chromatography-mass spectrometry (GC-MS). Antioxidant activity assays were carried out using DPPH, ABTS- and FRAP methods. Total polyphenols and total flavonoids were measured to evaluate the in vitro antioxidant activity in addition to the volatile compounds contained in the essential oils extracted from the flower petals ofP. delavayiwith the three flower colors. A total of 194 compounds were detected from essential oils ofP. delavayiflowers, including 83 in dark-purple petals, 90 in red petals and 80 in yellow petals. These compounds mainly include alcohols, aldehydes, ketones, alkenes, alkanes, esters and polyphenols. The results showed that the volatile compounds accumulated differentially among the essential oils from the different colors of flower petals. Principal component analysis (PCA) indicated that essential oils derived from dark-purple and red petals were more closely clustered while the yellow petal essential oil was very different with both the purple-red and red. Antioxidant assays suggested that the radical scavenging activity and the iron reduction antioxidant activity in the essential oils were highly correlated with the flower petal colors. These results suggestP. delavayiflower petals are potentially good resources for high quality essential oils and natural antioxidants. Keywords:Paeonia delavayi;flower colors;essential oil;antioxidant activity
