Efficient extraction of melon (Cucumis melo L.) seed husk cellulose and saccharification enhancement by E-beam pretreatment combined with alkaline deep eutectic solvents

Qian Lin,Yupeng Fan,Yanliang Guo,Li Niu,Hanlin Guan,Wenhao Li,Huijun Zhang

Melon seed husk is an agricultural waste rich in lignocellulose, and its high-value utilization is of great significance for sustainable development. However, the recalcitrant structure of lignocellulose poses a major bottleneck to achieving efficient bioconversion. This study aims to elucidate the mechanism of efficient deconstruction of melon seed husk cellulose via synergistic pretreatment with electron beam irradiation (EBI) and a deep eutectic solvent (DES), and to develop a green biorefinery process. We applied synergistic pretreatment of melon seed husk using varying doses of EBI and choline chloride/urea DES. The structural evolution of the pretreated material was analyzed at multiple scales, and its saccharification efficiency was evaluated via enzymatic hydrolysis. The results revealed that EBI dose-dependently disrupted the hydrogen-bonding network and the crystalline structure of cellulose, reducing its crystallinity by 0.95 %–11.54 % and forming a porous network. The synergistic pretreatment exhibited a significant dose-threshold effect. At the optimized dose of 150 kGy, the cellulose-binding rate was significantly enhanced, resulting in a saccharification yield of 48.29 %. This study provides a theoretical foundation and a feasible strategy for the industrialization of green biomass refining.

Phytoremediation of antibiotic-contaminated wastewater: Insight into the comparison of ciprofloxacin absorption, migration, and transformation process at different growth stages of E. crassipes

Yan Yan,Yang Deng,Wenjing Li,Wei Du,Yangyang Gu,Jiayi Li,Xiaoguang Xu

The selection of aquatic plants at different growth stages and their absorption, migration, and transformation mechanisms has yet to be clarified. In this study, Eichhornia crassipes at the seedling and mature stages were selected to uptake antibiotics under hydroponic conditions. The results showed that the enrichment of ciprofloxacin (CIP) in roots at the seedling and mature stages were 7.72~2114.39 μg g −1 and 0.07~3711.33 μg g −1 , respectively. The enrichment of CIP in aerial parts at the seedling and mature stages were 16.38~24.24 μg g −1 and 9.55~20.13 μg g −1 , respectively. The translocation from roots to aerial parts at the seedling stage was high, as evidenced by the relatively higher transfer factor (TF). In addition, eight and ten major metabolic products were observed in the tissues of seeding and mature stage of E. crassipes , respectively. The metabolic pathway of CIP was short at the maturity stage, and CIP had a strong upward migration ability at the seedling stage, facilitating long-time photodegradation . However, E. crassipes exhibited a poor CIP tolerance at the mature stage and decayed relatively early. Therefore, the seedling stage of E. crassipes was proposed to be applied for phytoremediation , and these findings might improve the ability to phytoremediation of antibiotic-contaminated water.

Efficient Conversion of All-E-Lutein Esters to Z-Lutein Esters and Physiological Activities and Physicochemical Properties of Free and Esterified Lutein Isomers

Liangman Shi,Lunhui Zhu,Jialing Cao,Xiaoyan Xu,Xiaoshu Tang,Shuang Zhang,Lianfu Zhang,Cheng Yang

A rapid method for producing cis-(Z)-lutein esters from all-E-lutein esters was developed using I2, AlCl3, and sulfides as catalysts. Z-Isomer-rich lutein (38.6%) and Z-isomer-rich lutein esters (34.8%) by I2 were obtained. The antioxidant activities of Z-isomer-rich lutein and Z-isomer-rich lutein esters were higher than those of their all-E-isomers by Ferric Reducing Antioxidant Power, ORAC-L, and ABTS assays. The UV-A and UV-B blocking activities of Z-isomer-rich lutein esters were higher than those of all-E-lutein esters. After short-term ingestion by rats, the in vivo absorption concentration of Z-isomer-rich lutein (222.3 pmol/mL) in a high dose was higher than those of other forms of lutein (<125.6 pmol/mL). Enrichment of Z-lutein (Z-ratio: 59.5%) and Z-lutein esters (Z-ratio: 44.4%) was achieved in hexane and ethanol, respectively. We conclude, for the first time, that Z-isomer-rich lutein esters play a more important role than all-E-lutein esters in preventing oxidation and UV-blocking activity and are estimated to exhibit higher bioavailability than all-E-lutein esters and all-E-lutein.

E-/VE-Cadherin-Fc Interface Programs Gli-1+ Stromal Cells for Assembly With Functionalized Microparticles Into 3D Renal Fibrotic Microtissues for Drug Screening

Zhong Cheng,Zhanyuan Qu,Jiaxin Sun,Zhihua Duan,Yong An,Yan Zhang,Jun Yang

Renal fibrosis lacks experimentally tractable human-relevant models that integrate defined stromal programming with spatially controlled profibrotic cues. Here, we developed a dual E-/VE-cadherin-Fc (EVE) interface to condition human mesenchymal stem cells (MSCs) under CTGF/TGF-β stimulation. Cells showed increased expression of Gli-1, NG2, PDGFRβ, and FAP and activated transcription of extracellular matrix remodeling. These changes were consistent with the acquisition of a Gli-1 + perivascular-like profibrotic stromal phenotype, and were accompanied by reorganization of cadherin-catenin complexes, actin cytoskeletal rearrangement, and increased YAP nuclear localization. Cell-sized PLGA/chitosan-heparin microparticles were subsequently functionalized with E-/VE-cadherin-Fc and loaded with CTGF/TGF-β. Co-assembly of these microparticles with the conditioned stromal cells (gMSCs) and renal epithelial cells (HK-2) generated 3D renal fibrotic microtissues that combined cadherin-mediated adhesive presentation with localized cytokine delivery. The resulting microtissues exhibited a broader distribution of α-SMA, Collagen I, and Fibronectin, together with reciprocal epithelial-stromal signaling and spatially organized fibrotic activation. As a proof of concept for pharmacological evaluation, the microtissues responded to pirfenidone (PFD) in a dose-dependent manner, with 100 µM PFD reducing α-SMA expression by ∼73% while preserving microtissue viability. Together, these findings establish a material-enabled, cadherin-guided strategy for constructing disease-relevant stromal organization and spatially controlled profibrotic signaling in human renal fibrosis models.

Tunable solid-state fluorescence of charge-transfer cocrystal self-assembled by (E)-N’-(9-phenanthrylmethylene) benzohydrazide with TCNB: Stacking mode and theoretical analysis

Jinxing Xu,Zhouyu Jiang,Mingliang Wang

A CT cocrystal FPTC of donor FPHD and acceptor TCNB was formed via self-assembly. • Donor-acceptor stacking mode and π⋯π interactions were quantified by SCXRD. • CT effect was analyzed through detailed DFT calculation. • Band-gap and optical modulation were induced by CT via the incorporation of TCNB. Investigation of the structure and optical property of charge transfer (CT) cocrystals comprising electron donors and acceptors is indispensable to clarify intrinsic relationships between donors and acceptors and construct new organic fluorescent materials. Herein, the electron donor ( E )-N’-(9-phenanthrylmethylene) benzohydrazide (FPHD), and acceptor 1,2,4,5-benzenetetracarbonitrile (TCNB) formed cocrystal FPTC (FPHD-TCNB) through supramolecular self-assembly. The crystal structure and π⋯π stacking mode of cocrystal FPTC were identified through single-crystal X-ray diffraction. The variation of C ≡ N stretch wavenumbers of TCNB and FPTC in Fourier-transform infrared spectra and the 75 nm red-shift in solid-state ultraviolet visible diffuse reflectance spectra confirmed the formation of cocrystal FPTC. Additionally, the 119 nm red-shift in solid-state fluorescence emission spectra indicated enhanced fluorescence, prolonged average fluorescence lifetime and elevated quantum yield of cocrystal FPTC. Through density functional theory (DFT), the weak interactions in cocrystal FPTC were analyzed, confirming the CT effect in FPTC. Furthermore, the narrowed band gap due to CT effect resulted in the change of transition dipole moment under excited state in cocrystal FPTC, directly leading to improved optical property of FPTC. This study illustrated CT effect on the structure and optical property of cocrystal FPTC via experimental exploration and theoretical analysis, providing a new strategy for novel organic fluorescent materials and endowing the cocrystal with promising research value for optoelectronic applications. Xiaojian Wen,Kailang Yang,Jaime C. Piñero,Junbao Wen

Simple SummaryEucryptorrhynchus scrobiculatus(Motschulsky) andE. brandti(Harold) are significant pests of tree of heaven,Ailanthus altissima, often leading to tree death. Monitoring systems that involve host–plant based attractants need to be developed for both insect pest species. Here, we compared the behavioral and electrophysiological responses ofE. scrobiculatusandE. brandtito volatiles emitted by various parts of the host plant. Host plant-derived volatiles showed to play a greater role in the foraging behavior ofE. brandtithan inE. scrobiculatus. Volatile components of phloem were found to be particularly attractive toE. brandti.AbstractEucryptorrhynchus scrobiculatusandE. brandti(Coleoptera: Curculionidae) are host-specific pests ofAilanthus altissima(Mill.) Swingle (Sapindales: Simaroubaceae), causing extensive damage to the host. There are no effective attractants available for pest management. The main aim of this study was to explore the role of host plant-derived volatiles in the behavioral response of both weevil species. In a field experiment, both weevil species showed positive response to phloem, and there was no preference for phloem associated with healthy or injured trees. Significantly moreE. brandtiadults responded to the olfactory treatments compared toE. scrobiculatus. In a large-arena experiment, both males and females ofE. scrobiculatussignificantly preferred phloem from the tree trunk while adults ofE. brandtiresponded in significantly greater numbers to tree limbs than to any other parts of host. Females and males ofE. scrobiculatusresponded positively to all parts of host tested in the Y-tube bioassay, whileE. brandtiadults were only attracted by the phloem from healthy and injured trees. There were dissimilar electroantennographic responses to compounds such as 1-hexanol and (1S)-(−)-β-pinene between the two weevil species. This study represents the first report documenting behavioral and electrophysiological responses ofE. scrobiculatusandE. brandtito volatiles from various parts ofA. altissimaand findings may aid efforts to develop attractants. Keywords:host plant volatiles;semiochemical;feeding selection;behavior;physiology

Rapid detection of Aspergillus spp. infection levels on milled rice by headspace-gas chromatography ion-mobility spectrometry (HS-GC-IMS) and E-nose

Shuang Gu,Wei Chen,Zhenhe Wang,Jun Wang,Yujia Huo

This study described the rapid detection of milled rice infected with Aspergillus spp. species based on headspace-gas chromatography ion-mobility spectrometry (HS-GC-IMS) and electronic nose (E-nose) combined with chemometrics, namely principal component analysis (PCA), k-nearest neighbor (kNN) and partial least squares regression (PLSR). 3D HS-GC-IMS imaging and their response differences enabled the discrimination among fungal species. kNN was used to differentiate rice samples with cdifferent levels of fungal infection and achieved correct classified rate of 94.44% and 91.67% by HS-GC-IMS and E-nose, respectively. PLSR method was used for quantitative regression of fungal colony counts in rice samples and good prediction performances were achieved by HS-GC-IMS (R p 2  = 0.909, RMSEP = 0.202) and E-nose (R p 2  = 0.864, RMSEP = 0.235). The results indicated that both HS-GC-IMS and E-nose approaches can potentially be implemented for the detection of fungal contamination levels in milled rice, and HS-GC-IMS fingerprinting coupled with chemometrics might be used as an alternative tool for a highly sensitive method. This research might provide scientific information on the rapid, non-destructive, and effective fungal detection system for rice grains .

Biometric-Tuned E-Skin Sensor with Real Fingerprints Provides Insights on Tactile Perception: Rosa Parks Had Better Surface Vibrational Sensation than Richard Nixon

Senlin Hou,Qingyun Huang,Hongyu Zhang,Qingjiu Chen,Cong Wu,Mengge Wu,Chen Meng,Kuanming Yao,Xinge Yu,Vellaisamy A. L. Roy,Walid Daoud,Jianping Wang,Wen Jung Li

The dense mechanoreceptors in human fingertips enable texture discrimination. Recent advances in flexible electronics have created tactile sensors that effectively replicate slowly adapting (SA) and rapidly adapting (RA) mechanoreceptors. However, the influence of dermatoglyphic structures on tactile signal transmission, such as the effect of fingerprint ridge filtering on friction-induced vibration frequencies, remains unexplored. A novel multi-layer flexible sensor with an artificially synthesized skin surface capable of replicating arbitrary fingerprints is developed. This sensor simultaneously detects pressure (SA response) and vibration (RA response), enabling texture recognition. Fingerprint ridge patterns from notable historical figures – Rosa Parks, Richard Nixon, Martin Luther King Jr., and Ronald Reagan – are fabricated on the sensor surface. Vibration frequency responses to assorted fabric textures are measured and compared between fingerprint replicas. Results demonstrate that fingerprint topography substantially impacts skin-surface vibrational transmission. Specifically, Parks' fingerprint structure conveyed higher frequencies more clearly than those of Nixon, King, or Reagan. This work suggests individual fingerprint ridge morphological variation influences tactile perception and can confer adaptive advantages for fine texture discrimination. The flexible bioinspired sensor provides new insights into human vibrotactile processing by modeling fingerprint-filtered mechanical signals at the finger-object interface.

Investigating the role and mechanism of water in E-beam modified sweet potato starch: Multi-scale structure, physicochemical properties, and in vitro digestibility

Wei Liang,Wenqing Zhao,Xinyue Liu,Jiayu Zheng,Zhuangzhuang Sun,Xiangzhen Ge,Huishan Shen,Gulnazym Ospankulova,Marat Muratkhan,Wenhao Li

Aiming to investigate the role and mechanism of the effect of water on the structural, physicochemical, and in vitro digestible properties of starch during irradiation processing. This work used water pretreatment of sweet potato starch (12%, 25%, 50%, 75% and 100% water content) and then modification by E-beam (6, 12 and 24 kGy). The results showed that E-beam treatment preserved the polarization cross, growth rings, crystal types and FT-IR patterns of sweet potato starch. Also, E-beam induced the water molecule activation and free radical production, thus promoting starch degradation. Moreover, the short-range ordered structure, relative crystallinity, molecular weight, long-chain proportion, ΔH, viscosity and swelling power of starch decreased with increasing E-beam irradiation dose. In contrast, the short-chain proportion, solubility, and resistant starch content were improved. In addition, a synergistic effect was found between the water pretreatment and E-beam, i.e., the higher the water content of the starch, the deeper the modification by E-beam. Results obtained in this study can partially elucidate the role of water content in E-beam-modified starch and provide a basis for E-beam treatment of starch-based foods under different water levels.

Natural food flavour (E)-2-hexenal, a potential antifungal agent, induces mitochondria-mediated apoptosis in Aspergillus flavus conidia via a ROS-dependent pathway

Weibin Ma,Luling Zhao,Eric T. Johnson,Yanli Xie,Mingming Zhang

Natural food flavour ( E )-2-hexenal, a green leaf volatile, exhibits potent antifungal activity on Aspergillus flavus , but its antifungal mechanism has not been fully elucidated. In this study, we evaluated ( E )-2-hexenal-induced apoptosis in A. flavus conidia and explored the underlying mechanisms of action. Evidence of apoptosis in A. flavus conidia were investigated by methods including fluorescent staining, flow cytometry, confocal laser scanning microscope, and spectral analysis. Results indicated that 4.0 μL/mL (minimum fungicidal concentration, MFC) of ( E )-2-hexenal application induced early markers of apoptotic cell death in A. flavus conidia with a rate of 38.4% after 6 h exposure. Meanwhile, typical hallmarks of apoptosis, such as decreased mitochondrial membrane potential (MMP), activated metacaspase activity, fragmented DNA, mitochondrial permeability transition pore (MPTP) opening and cytochrome c (Cyt C) release from mitochondria to the cytosol were also confirmed. Furthermore, intracellular ATP levels were reduced by 63.3 ± 3.6% and reactive oxygen species (ROS) positive cells increased by 31.1 ± 3.1% during A. flavus apoptosis induced by ( E )-2-hexenal. l -Cysteine (Cys), an antioxidant, could strongly block the excess ROS generation caused by ( E )-2-hexenal, which correspondingly resulted in a significant inhibition of MPTP opening and decrease of apoptosis in A. flavus , indicating that ROS palys a pivotal role in ( E )-2-hexenal-induced apoptosis. These results suggest that ( E )-2-hexenal exerts its antifungal effect on A. flavus conidia via a ROS-dependent mitochondrial apoptotic pathway.

Boosting the Electrochemical Performance of PI-5-CA/C-SWCNT Nanohybrid for Sensitive Detection of E. coli O157:H7 From the Real Sample

Wang Huan,Fan Yanmiao,Yang Qiaoli,Sun Xiaoyu,Liu Hao,Chen Wei,Aziz Ayesha,Wang Shenqi

Redox activity is an important indicator for evaluating electrochemical biosensors. In this work, we have successfully polymerized indole-5-carboxylic acid into poly-5-carboxyindole nanomaterials (PI-5-CA), using its superior redox activity, and introduced carboxylated single-walled carbon nanotubes (C-SWCNTs) to synthesize a composite material. Finally, a synthesized composite material was used for the modification of the glass carbon electrode to fabricate the PI-5-CA/C-SWCNTs/GCE-based immunosensor and was successfully applied for the sensitive detection of E. coli O157:H7. The fabricated immunosensor exhibited an outstanding electrocatalytic activity toward the detection of E. coli O157:H7 with a remarkably lowest limit of detection (2.5 CFU/ml, LOD = 3 SD/k, n = 3) and has a wide linear range from 2.98×101 to 2.98×107 CFU/ml. Inspired from the excellent results, the fabricated electrode was applied for the detection of bacteria from real samples (water samples) with a good recovery rate (98.13–107.69%) as well as an excellent stability and specificity. Owing to its simple preparation, excellent performance, and detection time within 30 min, our proposed immunosensor will open a new horizon in different fields for the sensitive detection of bacteria from real samples.

Folic acid-decorated vitamin E Poly(ethylene glycol) monoplatinum ester with disulfide bond as theranostic nanoparticle for drug resistant tumor diagnosis and treatment

Zhuoran Li,Huikang Yang,Fan Xu,Xuwen Zeng,Haowei Huang,Xinqing Jiang

Vitamin E Poly(ethylene glycol) monoplatinum ester (TPGS) nanoparticles have attracted much attention in recent years for overcome multidrug resistance. Herein, a well-defined folic acid (FA)-conjugated and disulfide bond-linked polymer (FA-SS-TPGS) was synthesized. These polymer nanoparticles were utilized as theranostic agents for tumor-targeted magnetic resonance imaging (MRI) and chemotherapy. By loading doxorubicin (DOX) and superparamagnetic iron oxide (SPIO) particles into TPGS nanoparticles, FA-SS-TPGS@DOX/SPIO nanoparticles are obtained. In vitro drug release studies revealed that under a reducing environment in the presence of glutathione (GSH), approximately 100% of the doxorubicin (DOX) was released from the disulfide bond-linked theranostic nanoparticles within 24 h. DOX and SPIO were efficiently delivered into HepG2-ADM cells due to the folate receptor-mediated endocytosis process of the nanoparticles. Additionally, the presence of glutathione (GSH) triggered the cleaving of the disulfide bonds, further facilitating the delivery of DOX and SPIO into the cells. Furthermore, the FA-SS-TPGS @DOX-SPIO nanoparticles exhibited strong MRI contrast enhancement properties. In conclusion, FA-SS-TPGS@DOX/SPIO are potential nanoparticles for tumor-targeted MRI and chemotherapy, which can also overcome multidrug resistance.
