Comparison of different extraction methods of polysaccharides from cup plant (Silphium perfoliatum L.)

Hongyan Wu,Hongmei Shang,Yang Guo,Hexiang Zhang,Hongxin Wu

The purpose of the present research was to compare the effects of four extraction methods on the extraction yields, physicochemical characteristics, and antioxidant activities of polysaccharides from the cup plant ( Silphium perfoliatum L.) (CPPs). The extraction methods included the hot water extraction method (HEM), ultrasonic-assisted extraction method (UEM), enzyme-assisted extraction method (EEM), and the ultrasonic-enzyme-assisted extraction method (UEEM). Four corresponding CPP extracts, HEM-CPPs, UEM-CPPs, EEM-CPPs, and UEEM-CPPs, were obtained. The results indicated that there were notable differences between the extraction methods regarding the yield, average molecular weight, uronic acid content, monosaccharide proportions, and the antioxidant activities of the CPPs. HEM-CPPs were extracted with the lowest yield (6.44%). EEM-CPPs had the largest extraction yield (9.87%) and the most notable antioxidant abilities of ferric reducing power and free radical scavenging activity. According to correlation analysis, the higher antioxidant abilities of EEM-CPPs might be due to their smaller average molecular weight and a higher content of uronic acid than those of the other extracted CPPs. Therefore, EEM could serve as a promising technique for extracting CPPs.

Ultrasound-induced destruction of heparin-loaded microbubbles attenuates L-arginine-induced acute pancreatitis

Lili Lin,Saili Xie,Yingzheng Zhao,Zenghui Liang,Qilong Wu,Mingling Fang,Xiaolu Teng,Binbin Shi,Yunjun Yang,Bin Chen

Purpose Acute pancreatitis (AP) involves sudden inflammation caused by abnormal activation of pancreatic enzymes. The mechanisms underlying AP include oxidative stress, high levels of inflammatory mediators and inflammatory cell infiltration. Heparin, a key therapeutic drug, exerts anti-inflammatory, antioxidative, and anticoagulative effects. However, safe and effective drug delivery remains an obstacle. This study is the first to investigate the therapeutic effects of heparin-loaded microbubbles (HPMB) combined with ultrasound (UHPMB) and the role of heparin in acoustic cavitation. Methods The characteristics of the microbubbles, including particle size, concentration, release, stability, and development, were studied. Heparin concentration in the HPMB was measured, and heparin-induced anticoagulation was evaluated. Drug safety was explored using hemolysis and cell viability assessments. The ability of HPMB to alleviate oxidative stress and inflammation were investigated in vitro . L -arginine induces AP in vivo . UHPMB was used for AP treatment. Serum amylase levels were measured and pancreatic architecture and pathological features were evaluated to determine AP severity. In vivo efficacy was evaluated, and the underlying mechanism of heparin action during acoustic cavitation was explored. Results HPMB was spherical and presented as an emulsion-like solution without aggregation. HPMB was visible and stable and effectively released the drug under ultrasound (US). HPMB and UHPMB led to lower AP severity than in the untreated group. US-targeted microbubble destruction (UTMD) enhanced the therapeutic effect by decreasing oxidative stress and inflammation in AP models without injuring vital organs. UHPMB regulated VEGF/Flt-1 and SOD-1 expression. HPMB can also mitigate oxidative stress and inflammation in H 2 O 2 -pretreated cells. Conclusion UHPMB exhibits a strong ability not only to selectively target pancreatic lesions and release heparin but also to provide efficient protection by inhibiting oxidative stress and inflammation.

Unveiling a trans-4-hydroxy-L-proline substrate binding protein in Vibrio spp.

Wang Yan,Zhu Yanping,Liu Ning-Hua,Zhao Fang,Cao Hai-Yan,Chen Yin,Zhang Yu-Zhong,Chen Xiu-Lan,Zhang Yu-Qiang,Gao Chao

Free trans -4-hydroxy-L-proline (T4LHyp), primarily released during the natural degradation of collagen, is absorbed and catabolised by heterotrophic bacteria, contributing to the global recycling of carbon and nitrogen. While bacterial T4LHyp catabolism is well documented, our understanding on how bacteria recognize and import T4LHyp remains limited. Here we show that HatA, the substrate binding protein (SBP) of an ATP-binding cassette transporter within the T4LHyp gene cluster of Vibrio alginolyticus 1A11092, recognizes T4LHyp during its uptake by the strain. HatA possessed significant binding affinity towards T4LHyp and L-proline and a binding signature (W 33 F 37 D 86 V 135 F 240 R 243 Y 265 N 267 Y 303 ), acting as the structural determinant for the specific ligand recognition, was defined. This signature is strictly conserved among HatA and its homologs, representing a subgroup of cluster D-II of SBPs. Genes encoding HatA-like SBPs are often located adjacent to collagenase and T4LHyp catabolic enzyme genes, and are nearly exclusively found in the genomes of Vibrio spp. in hosts and marine environments, which helps them exert pathogenicity when encountering host or environmental collagen. This study reveals a T4LHyp SBP in cosmopolitan Vibrio spp., offering better insights into the global T4LHyp catabolism and recycling driven by bacteria.

The Metabolomics Response of Solanum melongena L. Leaves to Various Forms of Pb

Siyu Zhang,Bing Zhao,Xuejiao Zhang,Fengchang Wu,Qing Zhao

Due to activities like mining and smelting, lead (Pb) enters the atmosphere in various forms in coarse and fine particles. It enters plants mainly through leaves, and goes up the food chain. In this study, PbXn(nano-PbS, mic-PbO and PbCl2) was applied to eggplant (Solanum melongenaL.) leaves, and 379 differential metabolites were identified and analyzed in eggplant leaves using liquid chromatography–mass spectrometry. Multivariate statistical analysis revealed that all three Pb treatments significantly altered the metabolite profile. Compared with nano-PbS, mic-PbO and PbCl2induced more identical metabolite changes. However, the alterations in metabolites related to the TCA cycle and pyrimidine metabolism, such as succinic acid, citric acid and cytidine, were specific to PbCl2. The number of differential metabolites induced by mic-PbO and PbCl2was three times that of nano-PbS, even though the amount of nano-PbS absorbed by leaves was ten times that of PbO and seven times that of PbCl2. This suggests that the metabolic response of eggplant leaves to Pb is influenced by both concentration and form. This study enhances the current understanding of plants’ metabolic response to Pb, and demonstrates that the metabolomics map provides a more comprehensive view of a plant’s response to specific metals. Keywords:metabolite;absorption;metabolic pathway;amino acid;antioxidation

Construction of metal–organic framework-based multienzyme system for l-tert-leucine production

Wang Ru,Jia Jianyao,Liu Xue,Chen Yaru,Xu Qing,Xue Feng

Chiral compounds are important drug intermediates that play a critical role in human life. Herein, we report a facile method to prepare multi-enzyme nano-devices with high catalytic activity and stability. The self-assemble molecular binders SpyCatcher and SpyTag were fused with leucine dehydrogenase and glucose dehydrogenase to produce sc-LeuDH (SpyCatcher-fused leucine dehydrogenase) and GDH-st (SpyTag-fused glucose dehydrogenase), respectively. After assembling, the cross-linked enzymes LeuDH-GDH were formed. The crosslinking enzyme has good pH stability and temperature stability. The coenzyme cycle constant of LeuDH-GDH was always higher than that of free double enzymes. The yield of l - tert -leucine synthesis by LeuDH-GDH was 0.47 times higher than that by free LeuDH and GDH. To further improve the enzyme performance, the cross-linked LeuDH-GDH was immobilized on zeolite imidazolate framework-8 (ZIF-8) via bionic mineralization, forming LeuDH-GDH @ZIF-8. The created co-immobilized enzymes showed even better pH stability and temperature stability than the cross-linked enzymes, and LeuDH-GDH@ZIF-8 retains 70% relative conversion rate in the first four reuses. In addition, the yield of LeuDH-GDH@ZIF-8 was 0.62 times higher than that of LeuDH-GDH, and 1.38 times higher than that of free double enzyme system. This work provides a novel method for developing multi-enzyme nano-device, and the ease of operation of this method is appealing for the construction of other multi-enzymes @MOF systems for the applications in the kinds of complex environment. Synergistic Fertilizers Improved Chemical Properties of Soil in Wheat (Triticum aestivum L.) Field

Shuang Chen,Yihui Wang,Yan Shi

In this study, the efficiency of fertilizers was improved by coating the fertilizers with a hydrogel formed by the polymerization of itaconic acid (IA), maleic acid (MA), acrylic acid (AA), and potassium persulfate (KPS) to form a synergistic fertilizer and improve its slow-release performance. The effect of synergistic fertilizers on the changes in soil nutrients in wheat fields was studied through field and soil column experiments using ordinary and commercial synergistic fertilizers as controls and different application rates of `synergistic fertilizers as treatments. The results revealed that the alkali-hydrolyzed nitrogen (N) and total N contents were significantly higher in T1 than that in CK, and the treatments were ranked as T3 > T2 > T5 > T1 > CK > T4. The difference in the alkali-hydrolyzed N and total N contents between CK and T3 was significant. In terms of available phosphorus (P) and potassium (K) contents, the treatments were ranked as T1 > T4 > T2 > CK > T3 in all soil layers and T3 > T2 > T5 > T1 > CK > T4 in the soil column. The treatments were ranked as T4>CK>T3>T1>T2 in terms of soil organic matter (SOM) content. Compared with ordinary fertilizers, synergistic fertilizers could significantly increase the soil total N, alkali-hydrolyzed N, available P, and available K contents. Compared with ordinary slow-release fertilizers in the market, the effect of synergistic fertilizers is better in improving the use efficiency of fertilizers. By slowly releasing nutrients, synergistic fertilizers can continuously provide nutrients for the growth and development of wheat and improve the use efficiency of fertilizer.

l-Arginine-Modified CoWO4/FeWO4 S-Scheme Heterojunction Enhances Ferroptosis against Solid Tumor

Zhuoran Yang,Chunyu Yang,Dan Yang,Ye Zhang,Qingzhu Yang,Fengyu Qu,Wei Guo

Ferroptosis has recently attracted much attention as an anti-tumor therapy. Evidence suggests that ferroptosis can induce oxidative stress and accumulation of lethal lipid peroxides in cancer cells, leading to cell damage. However, unsuitable pH, H 2 O 2 levels, and high glutathione (GSH) expression in the tumor microenvironment hinder the development of ferroptosis-mediated therapy. In this study, an l -arginine ( l -arg)-modified CoWO 4 /FeWO 4 (CFW) S-scheme heterojunction is strategically designed and constructed for ultrasound (US)-triggered sonodynamic- and gas therapy-induced ferroptosis. CFW not only has excellent Fenton-catalytic activity, outstanding GSH consumption capacity, and excellent ability to overcome tumor hypoxia, but its S-scheme heterostructure can also avoid the rapid combination of electron (e) and hole (h + ) pairs, thereby enhancing the sonodynamic effects. As a precursor of nitric oxide (NO), l -arg is modified on the surface of CFW (CFW@ l -arg) to achieve controlled NO release under US irradiation, thereby enhancing ferroptosis. In addition, poly(allylamine hydrochloride) is further modified on the surface of CFW@ l -arg to stabilize l -arg and achieve controllable NO release. Both in vitro and in vivo results demonstrate that such a multifunctional therapeutic nanoplatform can achieve high therapeutic efficacy through sonodynamic and gas therapy-enhanced ferroptosis. This designed oncotherapy nanoplatform provides new inspiration for ferroptosis-mediated therapy.

Synthesis and carbon dioxide capture properties of flower-shaped zeolitic imidazolate framework-L

Shaozhen Wang,Biao Zang,Yueyue Chang,Hongqi Chen

Flower-shaped zeolitic imidazolate framework-L (ZIF-L) nanostructures were synthesized by a coordination control method. The various factors influencing the growth of the flower-shaped ZIF-L include the quantity of H2O2, reaction time and temperature, and the formation process and the mechanism were investigated. The results showed that H2O2 plays an induction role in the growth of crystal nuclei. Using the cleaning regulator, H2O2, flower-like ZIF-L with a high yield was produced in a short time at room temperature, and the fluorescence enhancement products were prepared at high temperature. Meanwhile, the CO2 adsorption capacity of flower-shaped ZIF-L was 1.15 mmol g−1 at room temperature and 1 bar, which was higher than that of the two-dimensional ZIF-L. This outstanding CO2 capture performance is attributed primarily to the hierarchical flower-shaped micro/nanostructure which is resistant to leaf aggregation and stacking, thus increasing the CO2 adsorption capability. These results may open a new avenue for the development of ZIF-L nanocrystals with a controlled morphology, which will be widely applied in luminescence analysis, gas sensing, separation and storage.

Antibacterial Activity and Mechanisms of Essential Oil from Citrus medica L. var. sarcodactylis

Ze-Hua Li,Ming Cai,Yuan-Shuai Liu,Pei-Long Sun,Shao-Lei Luo

In this work, antibacterial activity of finger citron essential oil (FCEO,Citrus medica L. var. sarcodactylis) and its mechanism against food-borne bacteria were evaluated. A total of 28 components in the oil were identified by gas chromatography-mass spectrometry, in which limonene (45.36%), γ-terpinene (21.23%), and dodecanoic acid (7.52%) were three main components. For in vitro antibacterial tests, FCEO exhibited moderately antibacterial activity against common food-borne bacteria:Escherichia coli,Staphylococcus aureus,Bacillus subtilisandMicrococcus luteus. It showed a better bactericidal effect on Gram-positive bacteria than Gram-negative. Mechanisms of the antibacterial action were investigated by observing changes of bacteria morphology according to scanning electron microscopy, time-kill analysis, and permeability of cell and membrane integrity. Morphology of tested bacteria was changed and damaged more seriously with increased concentration and exposure time of FCEO. FCEO showed a significant reduction effect on the growth rate of surviving bacteria and lead to lysis of the cell wall, intracellular ingredient leakage, and consequently, cell death. Keywords:finger citron;essential oil;antibacterial;mechanism

ε-Poly(L-lysine)-based Hydrogels with Fast-acting and Prolonged Antibacterial Activities

Zou Yi-Jie,He Shi-Sheng,Du Jian-Zhong

Bacterial infections and the associated morbidity and mortality due to bacterial pathogens in wounds and medical implants have been increasing as most of current coatings cannot fulfill all the requirements including excellent intrinsically antibacterial activity, low cytotoxicity, and favorable physical properties. Herein, we present a kind of antibacterial hydrogel based on ε -poly(L-lysine) (EPL) grafted carboxymethyl chitosan (CMC- g -EPL) as the inherently antibacterial matrix and the surplus EPL as highly efficient antimicrobial agent. Such hydrogels possess tunable swelling abilities with water absorption percentages of 800%-2000% and modulus varying from 10 kPa to 100 kPa, and exhibit two-stage excellent antibacterial behavior. First, the free EPL can be released from the hydrogel network for quick and highly efficient bacteria killing with 99.99% of efficacy; second, the grafted EPL endows hydrogel matrix with prolonged intrinsically antibacterial activity, especially when most of free EPL is released from the hydrogel. Overall, we provide a new insight for preparing highly effective antibacterial hydrogels.

Synthesis of l-Histidine-Attached Graphene Nanomaterials and Their Application for Steel Protection

Chengbao Liu,Peng Du,Haichao Zhao,Liping Wang

Graphene-based carbonaceous materials have aroused great attention among the material protection fields due to their excellent impermeability. However, graphene nanosheets tend to aggregate in a polymer matrix and trigger microgalvanic corrosion, which potentially aggravates metal deterioration. Herein, we present a facile strategy to improve the protective property of graphene by tailoring graphene oxide with l-histidine molecules. The resultant functionalized graphene nanomaterials, without direct connections, can be well-dispersed in a polymer matrix. Electrochemical impedance measurements demonstrated that embedding a small percentage of well-dispersed functionalized graphene in epoxy coatings significantly enhanced the impermeable properties of the as-prepared composite coatings. The probability of galvanic corrosion of graphene with metal was largely inhibited and verified through the scanning vibrating electrode technique. The protection mechanism of composite coatings is interpreted as the barrier property of graphene nanosheets by suppressing the diffusion of corrosive species and the isolate function of l-histidine at the edge of graphene lamellae via increasing the electrical resistance.

Interspecies Developmental Differences in Metabonomic Phenotypes of Lycium ruthenicum and L. barbarum Fruits

Qi Wang,Shaohua Zeng,Xiangyu Wu,Hehua Lei,Ying Wang,Huiru Tang

Fruits of Lycium ruthenicum (LR) and L. barbarum (LB) in Solanaceae family contain abundant bioactive metabolites used widely as functional food and natural medicine. To characterize the fruit developmental molecular phenotypes, we comprehensively analyzed metabolite composition of both Lycium fruits at three developmental stages using the combined NMR, liquid chromatography–tandem mass spectrometry, and gas chromatography-flame ionization detector/mass spectrometry methods. The metabonomes of these fruits were dominated by over 90 metabolites including sugars, amino acids, tricarboxylic acid (TCA) cycle intermediates, fatty acids, choline metabolites, and shikimate-mediated plant secondary metabolites. Metabolic phenotypes of two species differed significantly at all three developmental stages; LB fruits contained significantly more sugars and amino acids but less TCA cycle intermediates, fatty acids, and secondary metabolites than LR. Interspecies differences for fatty acid levels were much greater after color-breaking than precolor-breaking. Furthermore, LR fruits contained more osmolytes than LB fruits indicating different osmoregulation requirements for these fruits during development. Significant differences were also present in biosynthesis of shikimate-mediated plant secondary metabolites in LR and LB. These findings provided essential metabolic information for plant physiology of these Lycium species and their utilizations demonstrating the usefulness of this metabonomic phenotyping approach for studying fundamental biochemistry of the plant development.
