Nature-derived bioactive components and photothermal synergistic therapy bring potential strategies for fighting bacterial infection and accelerating would healing by virtue of their excellent therapeutic efficiencies and ignorable side effects, where photothermal property not only acts as sterilization energy but also as a doorkeeper to control the natural component release. Herein, by integrating the excellent antibacterial property of cinnamaldehyde (CA) and the outstanding photothermal performance of copper sulfide nanoparticles (CuS NPs), a multifunctional nanoplatform of SiO 2 @CA@CuS nanospheres (NSs) is constructed with silica nanosphere (SiO 2 NSs) as carrier. SiO 2 @CA@CuS NSs exhibit photothermal property, bacterial absorption capacity, extraordinary antibacterial activity and antioxidant property. Mechanism characteriazation and antibacterial experiment indicate that positive charged SiO 2 @CA@CuS can adhere to the negative charged surface of bacteria, and quickly kill bacteria through the synergistic action of the released CA and heat produced under near infrared light (NIR) irradiation at 980 nm. The sterilization efficiencies for Escherichia coli ( E. coli ) and S. aureus reach 99.86% and 99.84%, respectively. Furthermore, NIR-regulated SiO 2 @CA@CuS perform great biocompatibility, as well as effective effects for accelerating S. aureus -infected wound healing at a low photothermal temperature (45 °C) relying on synergistic sterilization and anti-oxidation.

In PDT, high doses of photosensitizers can lead to prolonged patient photosensitivity and undesired side effects. As a classic fluorescent dye, BODIPY has attracted much attention in optical diagnosis and therapy due to its excellent stability, effective singlet oxygen production, and flexible modification. Herein, we synthesized successfully two organic near-infrared BODIPY derivatives with active groups. NO 2 -BDPI could be transformed into NH 2 -BDPI after responding to hydrogen sulfide in solution and brought an apparent color change. And both dyes possessed excellent single oxygen production capacity ( Φ Δ = 0.629 for NO 2 -BDPI and Φ Δ = 0.688 for NH 2 -BDPI). After being encapsulated with an amphiphilic polypeptide , NH 2 -BDPI could be assembled into nano–NH 2 –BDPI particles that own good water solubility and passively tumor-targeting via the enhanced permeability and retention (EPR) effect. Benefiting from the admirable cellular uptake and excellent intracellular ROS generation in vitro , the nano–NH 2 –BDPI shows splendid PDT effect towards 4T1 and HeLa cells under very low doses (0.24 μM) and NIR power density (660 nm 10 mW/cm 2 ). In vivo experiment, the nano-photosensitizer exhibited highly effective tumor suppression and ablation under a 40 mW/cm 2 NIR irradiation. The exploration demonstrated the commendable PDT effect of NH 2 -BDPI and made it be a potential candidate for further NIR imaging-guided PDT research in clinical application.

Chemodynamic therapy (CDT) has attracted considerable attention recently, but the poor reaction kinetics restrict its practical utility in clinic. Herein, glucose oxidase (GOx) functionalized ancient pigment nanosheets (SrCuSi 4 O 10 , SC) for programmable near-infrared II (NIR-II) photothermal-enhanced starvation primed CDT is developed. The SC nanosheets (SC NSs) are readily exfoliated from SC bulk suspension in water and subsequently functionalized with GOx to form the nanocatalyst (denoted as SC@G NSs). Upon laser irradiation, the photothermal effect of SC NSs can enhance the catalytic activity of GOx for NIR-II photothermal-enhanced starvation therapy, which effectively eliminates intratumoral glucose and produces abundant hydrogen peroxide (H 2 O 2 ). Importantly, the high photothermal-conversion efficiency (46.3%) of SC@G NSs in second biological window permits photothermal therapy of deep-seated tumors under the guidance of NIR-II photoacoustic imaging. Moreover, the acidity amplification due to gluconic acid generation will in turn accelerate the degradation of SC NSs, facilitating the release of strontium (Sr) and copper (Cu) ions. Both the elevated H 2 O 2 and the released ions will prime the Cu 2+ /Sr 2+ -H 2 O 2 reaction for enhanced CDT. Thus, a programmable NIR-II photothermal-enhanced starvation primed CDT is established to combat cancer with minimal side effects.

Titanium implants, widely used in orthopedic and dental applications, often face challenges due to inadequate osseointegration and the risk of postoperative infections , which can potentially lead to implant failure. To address this issue, we developed a titanium implant with an in-situ fabricated anatase TiO 2 metasurface , characterized by a subwavelength thickness and a preferential orientation of the (001) facets. This metasurface, which consists of uniformly distributed nano-crystallites of approximately 30 nm in size, aims to enhance both osseointegration and antibacterial properties. The addition of isopropanol (IPA) to the precursor solution used in the fabrication process played a crucial role in controlling the grain size and coating thickness of the metasurface. This process led to a redshift of the light absorption peak into the near-infrared (NIR)-II region, thereby increasing the penetration depth of photothermal therapy . The implants with TiO 2 metasurface exhibited increased hydrophilicity, rapid apatite formation in simulated body fluid, and an effective photothermal response under NIR-II light, showing significant antibacterial activity against E. coli and S. aureus . In vivo experiments in rabbits indicated superior bone regeneration and osseointegration around the implants with TiO 2 metasurface compared to the control titanium implants. At 4 weeks postoperatively, the bone volume to tissue volume (BV/TV) ratio within the implant was 53.94 %, and by 8 weeks, the implants were nearly entirely filled with new bone, with a BV/TV ratio of approximately 70 %. The findings suggest that the TiO 2 metasurface, which combines antibacterial and bioactive features, has the potential to reduce the risk of postoperative infections and improve clinical outcomes.

Exploration of single molecular species synchronously featured by long excitation/emission wavelength, accurate diagnosis, and effective therapy, remains supremely appealing to implement high-performance cancer phototheranostics. However, those previously established phototheranostic agents are undiversified and stereotyped in terms of structural skeleton, and generally exhibit insufficient phototheranostic outcomes. Herein, we innovatively utilized indanone-condensed thiadiazolo[3,4-g]quinoxaline (ITQ) as electron acceptor to construct novel photosensitizer with second near-infrared (NIR-II) emission. Experimental study and theoretical calculation demonstrated that comparing with the counterparts constituting by widely employed NIR-II building block benzobisthiadiazole (BBTD) and 6,7-diphenylthiadiazoloquinoxaline (DPTQ), ITQ-based photosensitizer (TITQ) showed superior aggregation-induced emission (AIE) characteristics, much stronger type-I reactive oxygen species (ROS) production, and prominent photothermal conversion capacity. Furthermore, TITQ nanoparticles with excellent biocompatibility were capable of effectively accumulating in the tumor site and visualizing tumor through fluorescence-photoacoustic-photothermal trimodal imaging with highly spatiotemporal resolution, and completely eliminating tumor by type-I photodynamic-photothermal therapy.

As a prominent class of 2D transition metal dichalcogenides (TMDCs), niobium diselenide nanosheets (NbSe 2 NSs) have garnered tremendous interest on account of promising applications pertaining to optoelectronics and energy storage. Although NbSe 2 NSs have many unique advantages such as inherent biocompatibility and broad absorption in the NIR region, their biomedical applications have rarely been reported, especially as therapeutic agents for the second near-infrared (NIR-II) range. Herein, a biodegradable nanotherapeutic platform consisting of NbSe 2 NSs is designed and demonstrated for NIR-II light-triggered photothermal therapy. NbSe 2 NSs synthesized by grinding and liquid exfoliation exhibit superior photothermal conversion efficiency (48.3%) and remarkable photothermal stability in the NIR-II region. In vitro assessment demonstrates that NbSe 2 NSs have favorable photothermal cell ablation efficiency and biocompatibility. After intravenous injection in vivo, the NbSe 2 NSs accumulate passively in tumor sites to facilitate fluorescence imaging and tumor ablation by NIR-II illumination. Furthermore, as a result of gradual degradation in the physiological environment, NbSe 2 NSs can be excreted from the body to avoid potential toxicity caused by long-term retention in vivo. The results reveal a promising NIR-II light-triggered PTT strategy with the aid of NbSe 2 NSs and the platform is expected to have large potential in cancer theranostics.

Hydrophobic CuS nanodisk assemblies and the anticancer drug paclitaxel (PTX) have been co-encapsulated into hybrid silica nanocapsules using a triblock copolymer®F127 (PEO106PPO70PEO106) micelle based soft-template method under mild conditions. The as-prepared CuS–PTX/SiO2 composite nanocapsules have excellent biocompatibility, solubility and high colloidal stability in physiological environments and also exhibit an enhanced photothermal conversion efficiency (PCE) of up to 31.2% as a result of the localized surface plasmon resonance (LSPR) effects. Owing to the hydrophobic–hydrophobic interaction and the silica shell protection, the PTX encapsulation ratio could reach 41.25% and the loaded PTX can undergo sustained release and controlled by near infrared (NIR) laser irradiation. In addition, in vitro and in vivo studies demonstrated the remarkable antitumor effects of the CuS–PTX/SiO2 nanocapsules under NIR laser irradiation (980 nm, 1 W cm−2) for 5 minutes. Hence, the designed novel CuS nanodisk assembly based CuS–PTX/SiO2 nanoplatform shows great potential for synergetic chemo-photothermal cancer therapy.

Combination of different therapy modalities with multiple tumoricidal mechanisms has emerged as a promising anticancer strategy. Herein, we reported an aldehyde/catechol-functionalized hyaluronic acid (DAHA) and hydroxyethyl chitosan (HECS) decorated gold nanorod (GNR) platform for combined chemo-photothermal therapy of breast cancer. The DAHA was synthesized by conjugating dopamine onto oxidized hyaluronic acid. The nanoplatform was prepared by successively modifying GNR with DAHA via Au-catechol bonds, conjugating DOX onto DAHA moiety through Schiff base linkage, and coating HECS interlayer for charge-reversal and hyaluronic acid corona for tumor cell targeting. The resulting nanoplatform GNR-HA DOX CH exhibited acid-triggered surface charge-reversal and pH/NIR dual-responsive drug release behaviors. The nanoplatform could be efficiently internalized into MCF-7 breast cancer cells and displayed greater cancer cell killing than individual modalities. Therefore, polysaccharide decoration could ensure the co-delivery of GNR and DOX into cancer cells, and the developed GNR-HA DOX CH holds great potential for breast cancer treatment.

Hypoxia is a typical feature of solid tumors, which highly limits the application of the oxygen-dependent therapy. Also, the dense and hyperbaric tumor tissues impede the penetration of nanoparticles into the deep tumor. Thereby, we designed a novel localized injectable hydrogel combining the photothermal therapy (PTT) and the thermodynamic therapy (TDT), which is based on the generation of free radicals even in the absence of oxygen for hypoxic tumor therapy. In our study, gold nanorods (AuNRs) and 2,2′-Azobis[2-(2-imidazalin-2-yl)propane] dihydrochlaride (AIPH) were incorporated into the hydrogel networks, which were formed by the copolymerization of hydrophobic N-isopropyl acrylamide (NIPAM) and hydrophilic glycidyl methacrylate modified hyaluronic acid (HA-GMA) to fabricate an injectable and near-infrared (NIR) responsive hydrogel. The crosslinked in situ forming hydrogel could not only realize PTT upon the NIR laser irradiation, but also generate free radicals even in hypoxic condition. Meanwhile the shrink of hydrogels upon thermal could accelerate the generation of free radicals to further damage the tumors, achieving the controlled drug release on demand. The designed hydrogel with a sufficient loading capacity, excellent biocompatibility and negligible systemic toxicity could serve as a long-acting implant for NIR-triggered thermo-responsive free radical generation. The in vitro cytotoxicity result and the in vivo antitumor activity illustrated the excellent therapeutic effect of hydrogels even in the absence of oxygen. Therefore, this innovative oxygen-independent platform combining the antitumor effects of PTT and TDT would bring a new insight into hypoxic tumor therapy by the application of alkyl free radical.

Most of chemotherapies are less effective when using alone. Exosomes (Exos) are cell derived nanovesicles that can serve as delivery nanoplatforms. Thus, Exos-based synergistic treatment is emerging as a promising strategy. Nevertheless, it is unknown about the underlying mechanism of Exos-based synergistic chemo-phototherapy at the gene level, especially in a glioma model. Here, we demonstrate that the combined therapy leads a superior chemo-phototherapy effect compared with chemotherapy or phototherapy alone. With the aid of Exos, the encapsulated agents can circulate for a longer time in vivo and effectively accumulated at tumor site. The local hyperthermia induced under near-infrared irradiation (NIR), impacts the permeability of Exos membrane and triggers drug release for maintaining a sustained chemotherapeutic efficacy. More importantly, the possible molecular mechanisms of Exos-based synergistic chemo-phototherapy are found to be involved in specific pathways including apoptosis, self-renewal repression and EMT program reversion. These data indicate that the engineered Exos can effectively combine chemotherapy and phototherapy to advance treatment of glioma and applied to other cancers as well, provide a new therapeutic strategy and potential molecular targets for clinical treatment.

A synergistic nanoprobe enables precise, safe, non-radiative skeletal imaging. • Multi-targeting enables precise enrichment throughout the skeleton. • AA@TFN provides a versatile platform for bone disease diagnosis and monitoring. Early diagnosis of bone-related diseases remains a critical clinical challenge, attributable to persistent limitations in imaging accuracy and safety. While near-infrared Region II fluorescence (NIR-II FL) imaging confers substantial advantages for deep-tissue visualization, its clinical translation is hampered by the inadequate targeting specificity of conventional contrast agents. Here, we developed Au-Ag@Apo-Tf/-N 3 (AA@TFN), a synergistic targeting gold–silver nanocluster probe engineered with glutathione and azide functionalities, which achieved bone-specific enrichment via a hierarchical multi-targeting mechanism. AA@TFN first anchored to hydroxyapatite in the bone matrix through glutathione carboxyl-mediated chelation, then targeted transferrin receptors highly expressed in bone marrow, and finally endured macrophage-mediated uptake facilitated by azide-enhanced bone affinity. A synergistic targeting strategy enabled real-time, high-resolution NIR-II FL imaging across the complete skeleton, with enhanced probe enrichment at lesion sites driven by microenvironmental changes. AA@TFN enabled visualization of synovial inflammation in rheumatoid arthritis models and millimeter‑scale microlesions in multiple myeloma models. Furthermore, the probe exhibited excellent biocompatibility and favorable safety profiles, underscoring its strong translational potential. A versatile platform was established for the diagnosis of bone diseases and non-invasive monitoring of therapeutic efficacy. The precise activation of the cGAS-STING pathway for effective tumor immunotherapy remains a significant challenge due to the complexity of immune responses and tumor microenvironment (TME) limitations. Here, a multifunctional nanoagonist, cRGD-PDA/ZIF8@ICG/TPT (cDZ@IP), was developed to realize nano-metabolite–driven multimodal synergistic activation of the STING pathway and enhanced immune recognition. The agonist combines cRGD peptide-targeted polydopamine-coated zeolitic imidazolate framework-8 with the photosensitizer indocyanine green and the chemotherapeutic drug topotecan. Upon near-infrared laser irradiation, cDZ@IP degrades within the TME, generating high levels of reactive oxygen species, inducing mitochondrial stress, and releasing endogenous mitochondrial DNA. Additionally, topotecan enhances DNA damage accumulation by inhibiting nuclear DNA repair. Zn 2 + released from the agonist further amplifies cGAS-STING pathway activation, thereby ensuring a robust immune response. The mild photothermal therapy-induced immunogenic cell death promotes the initiation of antitumor immunity, while also enhancing the effectiveness of immune checkpoint blockade. In vivo studies show that cDZ@IP significantly inhibits primary and distant tumor growth and prevents lung metastasis, providing a promising strategy for STING pathway-targeted cancer immunotherapy.
