Fabrication of nano-ZnS coated PEDOT-reduced graphene oxide hybrids modified glassy carbon-rotating disk electrode and its application for simultaneous determination of adenine, guanine, and thymine

Xiaoliang Ye,Yongling Du,Kaiyue Duan,Daban Lu,Chunming Wang,Xuezhao Shi

We proposed a facile approach for the synthesis of zinc sulfur-coated poly (3,4-ethylenedioxythiophene)-reduced graphene oxide hybrids film (ZnS-PEDOT-rGO) by using thioacetamide (TAA) as the reducing agent. The ZnS-PEDOT-rGO hybrids film modified glassy carbon-rotating disk electrode (GC-RDE) was then developed for the sensitive simultaneous determination of three deoxyribonucleic acid (DNA) bases: guanine (G), adenine (A), and thymine (T). By cyclic voltammetry (CV), this electrochemical sensor showed a good photoelectronic effect, and the peak currents of G, A, and T increased obviously. Some kinetic parameters were estimated by linear sweep voltammetry (LSV). Under the optimal conditions, the magnified anodic peak currents represented the excellent analytical performance of simultaneous detection of G, A, and T in a wide linear range and low detection limit. This proposed method also has been successfully applied to the assessment of G, A, and T contents in real-life samples, such as herring sperm DNA samples, milk powder and urine sample from human beings, with satisfactory results. The obtained results were also compared to HPLC of analysis for those three DNA bases and no significant differences were found. By the treatment of the experimental data, the electrochemical reaction mechanisms of G, A, and T all involved a two-electron-two-proton-transfer process.

Prussian blue analogue in situ derived hierarchical flower-like ZnS/FeS2 Z-scheme heterojunction for boosted visible-light responsive photocatalytic degradation of Rhodamine B with synergistic peroxydisulfate activation

Hancheng Tan,Xiaolan Zhou,Haiyan Zhu,Wenxiu Gu

A micro/nano hierarchical flower-like ZnS/FeS2 direct Z-scheme heterojunction visible-light catalyst was in situ derived from a Prussian blue analogue cube (Zn–Fe PBA) using a facile one-pot hydrothermal sulfidation strategy. The as-prepared ZnS/FeS2 catalyst shows extraordinarily boosted visible-light photocatalytic degradability for Rhodamine B (RhB) with the synergism of peroxydisulfate (PDS). Under visible-light illumination, 98.5% degradation efficiency can be achieved within 15 min and 100% within 20 min with excellent structural stability of ZnS/FeS2. The degradation kinetic constant of ZnS/FeS2 + PDS is 9.33 times that of ZnS/FeS2, 23.02 times that of p-ZnS/FeS2, 24.05 times that of FeS2, and 28.25 times that of ZnS, respectively. The ZnS/FeS2 heterojunction shows significant synergistic benefits in the photocatalytic degradation reaction. More advantageously, ZnS/FeS2 + PDS shows excellent photocatalytic degradation performance in a wide pH range (1–10). A reasonable direct Z-scheme heterojunction mechanism for visible-light responsive photocatalytic degradation of RhB with ˙OH, h+, ˙O2−, and ˙SO4− as the crucial active species is recommended for the ZnS/FeS2 + PDS composite system. The micro/nano hierarchical flower-like structure of ZnS/FeS2 simultaneously combines the high activity of nano-materials with the advantages of easy recycling of micro-sized materials. The in situ transformation characteristics of ZnS/FeS2 from the PBA template contribute to the advantageous high performance and structural stability.

InP/ZnSe/ZnS quantum dots with strong dual emissions: visible excitonic emission and near-infrared surface defect emission and their application in in vitro and in vivo bioimaging

Jie Zhang,Jie Wang,Tong Yan,Yanan Peng,Dajun Xu,Dawei Deng

Quantum dots (QDs) exhibit many unique optical properties, and show great promise as fluorescent markers in molecular, cellular and in vivo imaging. In this thematic issue, a major concern is their cytotoxicity. Among various Cd-free alternatives, InP-based QDs without highly toxic heavy metal elements have received the most attention. This article first focuses on the synthetic control of oil-soluble InP/ZnSe/ZnS QDs, exhibiting strong dual emissions, namely, visible excitonic emission and near-infrared (NIR) surface defect emission. Next, the organic-to-aqueous phase transfer of the dual emissive QDs was explored systematically. It was found that the dual emissions are relatively stable against the water transfer strategies used here; among them, aqueous dual emissive QDs obtained by wrapping oil-soluble QDs with a poly(acrylic acid)-octylamine (PAA-based) amphiphilic polymer (or modified with the cRGD peptide) exhibit enhanced NIR emission. Finally, using in vitro cell and in vivo small animal optical imaging techniques, the bioactivities of the cRGD-modified amphiphilic polymer-wrapped QDs were also investigated. The results confirm that single-wavelength excitation with strong dual emissions ranging from 550 to >1000 nm will endow the InP-based QDs with the capability for biomedical optical imaging across different spatial scales, as a promising alternative for Cd- and Pb-based QDs.

Fluorescent ligand fishing combination with in-situ imaging and characterizing to screen Hsp 90 inhibitors from Curcuma longa L. based on InP/ZnS quantum dots embedded mesoporous nanoparticles

Yue Hu,Anchen Fu,Zhaoyi Miao,Xiaojing Zhang,Tianlin Wang,An Kang,Jinjun Shan,Dong Zhu,Wei Li

Although ligand fishing has been shown to be an efficient technique for the identification of bioactive components from complex mixtures such as natural products , it cannot be applied to biomedical image processing. Herein, a specific fluorescent ligand fishing combined with in situ imaging approach is presented for the identification of heat shock protein 90 (Hsp 90) inhibitors from complex matrixes, Curcuma longa L. , using N-terminus immobilized Hsp 90α functionalized InP/ZnS quantum dots embedded mesoporous nanoparticles ( i.e. Hsp 90α (NT)-FQDNs) as extraction sorbents and fluorescent tracer. The fished ligands were identified by liquid chromatography time-of-flight/mass spectrometry (LC-TOF/MS) and gas chromatography-mass spectrometry (GC-MS). Moreover, in situ imaging by confocal laser scanning microscopy (CLSM) was applied for evaluating the effect of fished-ligands on bioactivity-induced apoptosis morphologically in HeLa cells. MTT assay verified the bioactivity of the ligands and molecular docking results further provided convincing information to verify the feasible binding mode between ligands and protein. Twelve ligands as potential Hsp 90 inhibitors were ultimately fished and identified from Curcuma longa L. crude extracts. The proposed approach based on Hsp 90α functionalized nanocomposites is superior in the combination of highly specific screening efficiency and concurrent visual in situ imaging, which could have great promise for the development of other plant-derived Hsp 90 inhibitors, and providing a rapid and reliable platform for discovering biologically active molecules in natural products.

Turn-on Room Temperature Phosphorescence Assay of Heparin with Tunable Sensitivity and Detection Window Based on Target-Induced Self-Assembly of Polyethyleneimine Capped Mn-Doped ZnS Quantum Dots

Huan Yan,He-Fang Wang

Room-temperature phosphorescence (RTP) turn-on assay of heparin with tunable sensitivity and detection window was demonstrated on the basis of the target induced self-assembly of polyethyleneimine capped Mn-doped ZnS (PEI-Mn-ZnS) QDs. The proposed method can cover the whole therapeutic dosing concentration range in postoperation and long-term therapy (0.2–1.2 U/mL, 1.7–10 μM) and cardiovascular surgery (2–8 U/mL, 17–67 μM) in 10 mM Tris–HCl (pH 7.4) buffer and can be applied for heparin determination in 100-fold diluted human serum samples. The tunable sensitivity and detection window was ascribed to the tunable particle size and ligand loading amount of PEI-Mn-ZnS QD arising from the chain length (or molecule weight) and feed amount of PEI. The chain length of PEI exhibited significant effect on the particle size and ligand loading amount and ultimately had important influence on both the sensitivity and detection window. The feeding amount of PEI, however, greatly affected the ligand loading amount alone and, in turn, affected only the detection window.

A visible light-driven photoelectrochemical sensor for mercury (II) with “turn-on” signal output through in-situ formation of double type-II heterostructure using CdS nanowires and ZnS quantum dots

Luping Feng,Lixiang Zhang,Xi Chen,Chunxian Zhang,Guojiang Mao,Hua Wang

An ultrasensitive and selective photoelectrochemical (PEC) platform has been developed for determining Hg 2+ with “turn-on” signal output by synergistically using CdS nanowires (NWs) and ZnS quantum dots (QDs). CdS NWs was first prepared and then deposited with ZnS QDs to yield ZnS/CdS nanocomposite with type-II heterostructure. It was found that the as-constructed ZnS/CdS heterostructure can exhibit excellent photoelectrochemical characteristics by separately employing CdS NWs and ZnS QDs as the photosensitizer and Hg 2+ -recognition probe. What's more, the constructed analysis platform could display specifically enhanced photocurrent responses to Hg 2+ under the visible light via the selective ion-exchange reaction. The in-situ formation of ternary HgS/ZnS/CdS could be thus expected with double type-II heterostructures so as to further enhance the transferring and separation of charges. A linear response can be achieved for Hg 2+ in the concentration range from 0.0010 to 5.0 nM, with a detection limit of 0.40 pM. Moreover, the application feasibility of the developed sensor with ZnS/CdS heterostructure could be testified by detecting Hg 2+ ions in environmental waters. Such a route of in-situ formation of double type-II heterostructure might promise the wide applications for designing different visible light-driven PEC sensors for probing heavy metals like Hg 2+ with the “turn-on” signal output.

A QDs Nanocomposites-Based Photoluminescence Ratiometric Method for Selective and Visual Cadmium Detection Combining with Smartphone-Based PL E-Eye

Xinyi Wang,Liubing Kong,Shuqi Zhou,Tao Liang,Hao Wan,Ping Wang

Photoluminescence (PL) sensors based on quantum dots (QDs) are difficult to achieve exact detection in sample waters. In this paper, polyvinyl alcohol (PVA) based CdTe/ZnS/CdS QDs and fluorescein isothiocyanate (FITC) fluorophores were used to build a ratiometric PL sensor. Combining with the homemade smartphone-based PL E-eye, the Cd2+ exact detection can be achieved. PVA was used to connect QDs and FITC fluorophores without unnecessary ligand exchange and purification. For QDs fluorophore, ethylenediaminetetraaceticacid (EDTA) was used on the surface to induce the specifically Cd2+ recognition site. For FITC fluorophore, the PL remains unchanged in this experiment. Thus, the ratio of two fluorophores can be used to provide a built-in correction. The PL changes with the increase of Cd2+ concentrations could be displayed as a visual color change on the smartphone. Further quantitative analysis could be carried out by the RGB value of the picture through the App in less than 1 min The ratio of R/G is linear to Cd2+ concentration in the range of 1–2000 μg l−1 with a low LOD of 0.057 μg l−1 (S/N). Compared with traditional analysis methods, the PL ratiometric method with PL E-eye is portable, rapid, visible and highly selective especially in discriminate Cd2+ from Zn2+.

CdSe/ZnS quantum dots coated with carboxy-PEG and modified with the terbium(III) complex of guanosine 5′-monophosphate as a fluorescent nanoprobe for ratiometric determination of arsenate via its inhibition of acid phosphatase activity

Wen Shao-Hua,Liang Ru-Ping,Zeng Hui-Hui,Zhang Li,Qiu Jian-Ding

A ratiometric fluorometric method is described for the determination of arsenate via its inhibitory effect on the activity of the enzyme acid phosphatase. A nanoprobe was designed that consists of CdSe/ZnS quantum dots (QDs) coated with the terbium(III) complex of guanosine monophosphate (Tb-GMP). The nanoprobe was synthesized from carboxylated QDs, Tb(III) and GMP via binding of Tb(III) by both the carboxy and the phosphate groups. The nanoprobe, under single-wavelength excitation (at 280 nm), displays dual (red and green) emission, with peaks at around 652 nm from the QDs, and at 547 nm from the Tb-GMP coordination polymers. It is shown to be a viable nanoprobe for fluorometric determination of As(V) detection through it inhibitory action on the activity of acid phosphatase (ACP). The enzyme destroys the Tb-GMP structure via hydrolysis of GMP, and hence the fluorescence of the Tb-GMP complex is quenched. In contrast, the fluorescence of the CdSe/ZnS QDs remains inert to ACP. It therefore can serve as an internal reference signal. In the presence of arsenate (an analog of phosphate), the activity of ACP is inhibited due to competitive binding. Thus, hydrolysis of GMP is prevented. These findings were used to design a ratiometric fluorometric method for the quantification of As(V). The ratio of fluorescences at 547 and 652 nm increases linearly in the 0.5 to 200 ppb As(V) concentation range, and the limit of detection is 0.39 ppb. Under a UV lamp, the probe shows distinguishable color from green to red on increasing the concentration of As(V). Schematic illustration of CdSe/ZnS quantum dot coated with carboxy-PEG and modified with the terbium(III)-GMP complex as a fluorescent nanoprobe for ratiometric determination of arsenate via its inhibition of ACP activity.
