Core–shell thermal-responsive and magnetic molecularly imprinted polymers based on mag-yeast for selective adsorption and controlled release of tetracycline

Ma Wei,Li Songtian,Chen Long,Sun Jun,Yan Yongsheng

A novel thermal-responsive and magnetic molecularly imprinted polymers (TMMIPs) were prepared based on mag-yeast via a mild and effective method; the obtained TMMIPs were used for selective adsorption and release of tetracycline from aqueous solution. Imprinted polymerization directly occured on the surface of microspheres, and core–shell imprinted polymers were developed via in situ precipitation polymerization. The properties of obtained TMMIPs were characterized by scanning electron and transmission electron microscopy, X-ray diffraction, Fourier transform infrared spectra, thermo-gravimetric analysis and so on. The adsorption equilibrium data were well described by Langmuir isotherm model. Kinetic experiments showed the adsorption process reached the equilibrium within 60 min, and the pseudo-second-order kinetic model was used to fit the adsorption data well. TMMIPs exhibited magnetic sensitivity, magnetic stability and thermal stability. Reversible recognition and release of template molecule were realized by changing environmental temperatures. Several other antibiotics were selected as model analytes to evaluate the selective recognition performance of TMMIPs. The TMMIPs have good temperature response, selectivity and reusability, making them possible in applying for antibiotics adsorption and controlled release.

Chitosan surface-modified bovine serum albumin-oleic acid self-assembled complexes: Improving the thermal stability, controlled release and bioaccessibility of fucoxanthin

Huiying Kuang,Liyuan Ma,Yuanjie Guo,Yixiang Liu

Fucoxanthin (FUCO) is a marine xanthophyll carotenoid with widespread potential physiological functions. However, poor water solubility and chemical instability limit FUCO's processing adaptability and bioavailability. In this study, chitosan surface-modified bovine serum albumin (BSA)-oleic acid (OA) self-assembled complexes were fabricated to improve the thermal stability as well as the controlled release and bioaccessibility of FUCO during gastrointestinal digestion. The results showed that glucose-grafted chitosan (GGC) could surface modify BSA-OA complexes by non-covalent affinity adhesion, resulting in a stable hydrophilic dispersion system with a polydispersity index <0.25 and a Zeta potential  > −20 mV. Fourier transform infrared spectroscopy and transmission electron microscope observations revealed that GGC covered the surface of BSA-OA particles through hydrogen bonding, hydrophobic and electrostatic interactions. The GGC-BSA-OA delivery system captured FUCO with >88.00% encapsulation efficiency. Additionally, embedding FUCO in the GGC-modified BSA-OA complex improved its thermal stability. In vitro, gastrointestinal digestion simulations showed that the GGC-modified protein shell had a controlled-release effect on FUCO, which prevented acidic pH from destroying FUCO due to premature release during gastric digestion and released it during intestinal digestion. As expected, GGC-BSA-OA, an encapsulated carrier, dramatically improved the FUCO's digestive stability (retention rate >74.00%), bioaccessibility (>54.42%), and micellization rate (>70.00%) in simulated gastrointestinal digestion. These findings suggested that GGC-BSA-OA as a delivery vehicle could effectively improve FUCO's thermal processing stability and absorbability.

Optimization of Zn biodegradable cardiac stent performance through near-infrared light-controlled release dual responsive cerium oxide loaded PLGA coating growth

Yuemei Li,Yongmei Li,Yunfei Qu

Zn alloy stents, as implantable cardiac devices, have broad application prospects in the field of heart. However, excessive Zn 2+ release during degradation remains a challenge for biocompatibility. Here, the Yb/Er/CeO 2 coating is grown on the Zn substrate, which inhibits the release of Zn 2+ and enhances the corrosion resistance of Zn stent. Under 980 nm excitation, the Yb/Er/CeO 2 coating enables electron transitions from the valence band to the conduction band. This process catalyzes H 2 O and O 2 to generate reactive oxygen species (ROS), such as ∙OH. These ROS act as chemical linkers to accelerate the deposition of the drug-eluting PLGA coating. The Zn@Yb/Er/CeO 2 /PLGA composite demonstrates excellent biodegradability. It shows great potential for treating cardiovascular diseases.

Grafting of thermo- and pH-responsive polymer inside mesoporous silica foam in supercritical carbon dioxide for controlled release of 5-fluorouracil

Hao Youwei,Zhao Jie,Wang Gang,Cao Liqin,Wang Jide,Yue Fan

Mesocellular silica foam (MCF) grafted with thermo-responsive and pH-responsive copolymer poly( N -isopropylacrylamide- co -acrylic acid), was prepared by precipitation polymerization using supercritical carbon dioxide (scCO 2 ) as entrainer and solvent. Vinyl-functionalized MCF was first synthesized through post-synthesis via using vinyltriethoxysilane as a coupling agent for surface modification. Entrapped monomers of N-isopropylacrylamide (NIPA) and acrylic acid (AA) were then introduced and assembled onto the vinyl-modified MCF with the aid of scCO 2 . The grafting copolymer performed here as an on/off switch in the release of 5-fluorouracil, as confirmed by using UV-visible absorption spectroscopy. The grafted organic content and drug loading efficiency were 65.2 wt% and 11.5 wt%, respectively. The resultant grafting composite showed potential for application in drug delivery and exhibited thermo- and pH-responsiveness to external environmental changes.

A novel strategy for encapsulation and controlled release of Schisantherin a employing biofriendly polydopamine microparticles built on biomineralized calcium carbonate crystals

Zhaoyu Wang,Qingxia Ji,Tingting Wang,Weiliang Hua,Yanhong Bi,Yongzhou Chi,Rongling Yang

Schisantherin A (SCA) has recently attracted increasing attention because of its promising biological profile. However, poor aqueous solubility and bioavailability have limited the efficacy of SCA in pharmaceutical applications. In this study, novel complex CaCO 3 -based biomineralized microparticles that demonstrate significant potential as carriers for loading and delivering SCA were constructed for the first time by employing gum arabic as the stable template and polydopamine as the functional coating agent. The results show that these composite microparticles have a high SCA loading rate of 15.6 ± 0.4 % and a pH-responsive drug release characteristic under simulated in vitro conditions. Further investigation revealed that the encapsulated forms of SCA exhibit significantly enhanced synergistic antioxidant activity and antibacterial efficacy against Staphylococcus aureus compared to the carrier-free SCA. Furthermore, a 4′,6-diamidino-2-phenylindole (DAPI) staining experiment and a rhodamine 123 (Rh123) fluorescence staining test of lipopolysaccharide (PSL) induced BV-2 microglial cells demonstrated that the encapsulated form of SCA exhibits considerable efficacy in inducing changes in the morphology and structure of cells. With these findings, this study provides a new type of CaCO 3 -mediated particle carrier that is useful for targeted and controlled release applications.

Application of hollow mesoporous organosilica nanoparticles as pH and redox double stimuli-responsive nanocontainer in the controlled release of corrosion inhibitor molecules

Chunling Li,Xiyu Zhao,Cheng Meng,Tengfang Zhang,Shuangqing Sun,Songqing Hu

The stimuli-responsive release could reduce the waste of the corrosion inhibitor and realize the accurate repair of corroded region. In this work, hollow mesoporous organosilica nanoparticles (HMON) were first synthesized by silica-etching chemistry tailoring method. Then, HMON were encapsulated with 2-mercaptobenzothiazole (MBT) corrosion inhibitor molecules to build an intelligent corrosion inhibitor system, i.e., MBT@HMON. The microstructure and frame composition of the synthesized HMON were characterized. After the successful encapsulation of MBT into HMON, the release kinetics of MBT from HMON under different dithiothreitol (DTT) concentration and pH conditions were investigated. Finally, the corrosion inhibition performance of MBT@HMON for copper alloy was evaluated in 3.5 wt% NaCl solution by weight loss test and electrochemical impedance spectroscopy (EIS). The results show that the controlled release of corrosion inhibitor MBT from HMON could be realized by tuning the redox and pH conditions of corrosion medium. When the appropriate concentration of MBT@HMON was added into the corrosion medium (1.6 g·L −1 in this work), the corrosion of cooper alloy was greatly inhibited. This simple and general strategy of introducing corrosion inhibitors loaded HMON directly into the corrosion medium offers a new prospect for the application of nanocontainers in the intelligent corrosion protection of metallic materials.

Regulation of the encapsulation, protection, and controlled release properties of naringin by sodium alginate oxidized hydrogel as a steady-state delivery system

Shuxian Bai,Lei Chen,Hui Teng,Hesham R El-Seedi,Junyan Yu,Chao Ai

Introduction Addressing the application challenges of low oral bioavailability and poor chemical stability associated with the citrus flavonoid naringin, hydrogels featuring unique three-dimensional network structures have garnered significant attention in the field of bioactive compound delivery. Methods In this study, a natural polysaccharide hydrogel delivery system based on dynamic imine bonds was designed and constructed. By regulating the oxidation degree of oxidized sodium alginate (OSA), hydrogels with distinct pore sizes were engineered to achieve stable encapsulation and controlled release of naringin. Dynamic covalent crosslinking between the aldehyde groups on OSA chains and the amino groups of carboxymethyl chitosan (CMC) endowed the hydrogels with exceptional adaptability. Results Research indicates that increasing the oxidation state of OSA can form a denser cross-linked network, thereby significantly regulating the swelling behavior, biodegradation rate, and drug release kinetics of hydrogels. Among these, 6OSA-Hy exhibits the most ideal sustained-release properties, enabling continuous and controlled release of naringin in simulated environments. In vitro cell experiments confirmed that this delivery system exhibits excellent biocompatibility, and its released active components effectively promote cell migration, with the 6OSA-Hy group achieving a scratch closure rate of 43.5%. Discussion The above studies demonstrate that the hydrogel system developed in this research provides a sustained-release stabilization strategy for enhancing the delivery of bioactive compounds such as naringin. This approach holds potential for improving bioavailability and is expected to find applications in the development of functional foods and nutritional supplements.

Encapsulation of Zanthoxylum schinifolium essential oil by complex coacervation with whey protein isolate and gum Arabic for improved stability and controlled release

Jie Zhao,Jiating Chen,Tanghui Liu,Dong Zhang,Chang Su,Hai Chi,Hongrui Chen,Jie Tang,Xingzhong Zhang

Zanthoxylum schinifolium essential oil (ZSEO) exhibits remarkable antioxidant and antibacterial biological activities and is widely used in the food industry. However, its low water solubility, high volatility, and susceptibility to decomposition by light, heat, and oxygen complicate its transport and storage. Microencapsulation of ZSEO is an effective method that can overcome these limitations and expand its application scope. To enhance stability and preserve functionality of ZSEO, ZSEO-loaded microcapsules were produced by complex coacervation of whey protein isolate (WPI) and gum Arabic (GA), followed with freeze-drying. So far, no comprehensive research has been conducted on the method of microencapsulating ZSEO using WPI/GA. Through research, the optimal pH for the formation of WPI/GA (1:1, w/w) complex coacervates was found to be 3.9. The ZSEO-in-water (O/W) emulsions stabilized by whey protein isolate (WPI) were prepared, which exhibited a uniform size of around 200 nm and showed enhanced stability with increasing WPI contents (3.0–6.0 wt%). After complex coacervation with GA, the resulting microcapsules showed improved physical properties and higher encapsulation efficiency as the ratio of WPI/GA complex wall material increased. The ZSEO-loaded microcapsules still maintained their ability to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) free radicals compared to free ZSEO, especially performing the optimal antioxidant activity at a core-to-wall ratio of 5:8 (w/w). Release kinetics studies confirmed controlled and delayed release of ZSEO from the microcapsules in various food simulants (aqueous, ethanolic and aerial environments). This work demonstrated that microencapsulation via WPI/GA complex coacervation was an effective strategy to encapsulate essential oils with high antioxidant activity and controlled release.

Controlled release of hypoxic exosomes from PLGA microspheres attenuates IVDD progression through activating PPAR-γ and suppressing oxidative stress-mediated NPCs senescence

Weiqi Zhang,Han Yin,Chenchen Wang,Xiaowei Ma,Dazhuang Miao,Guimiao Li,Wei Chen,Zhiyong Hou,Xianda Gao,Yingze Zhang,Di Zhang

Oxidative stress and apoptosis mediated senescence of nucleus pulposus cells (NPCs) and the degradation of extracellular matrix (ECM), which represent a key mechanism underlying intervertebral disc degeneration (IVDD). Mesenchymal stem cell-derived exosomes hold therapeutic potential for tissue repair; however, their clinical application is limited by rapid clearance, short half-life, and poor target specificity. To overcome these challenges, this study developed poly(lactic-co-glycolic acid) (PLGA) microspheres as a sustained-release carrier for exosomes derived under either normoxic (Exos, 21% O₂) or hypoxic (H-Exos, 1% O₂) conditions. The PLGA microspheres demonstrated high exosome-loading efficiency and provided controlled release for over four weeks. Released hypoxic exosomes activate PPAR-γ pathway, exerting better effects by reducing NPCs SA-β-gal positive rate by 59.8% and elevates Col II and ACAN levels by 30.7% and 25.5%. Animal experiments confirmed that this system effectively attenuated IVDD progression indicating its potential for clinical translation.

A pH/ROS dual-responsive hydrogel promotes diabetic wound healing by controlled release of O2 and EGCG to induce angiogenesis and immunomodulation

Yongcen Chen,Xuemin Ma,Yajuan Hu,Huiyue Wang,Peirong Zhou,Xiaoping Tian,Haofeng Wang,Yun He,Gang Tao,Rui Cai,Junliang Chen

Diabetic wounds are marked by persistent infection, hypoxia, excessive reactive oxygen species (ROS), and impaired angiogenesis, which present significant challenges for clinical management. In this study, a pH/ROS dual-responsive multifunctional hydrogel (QOF-EGCG/Hb@PDA) was constructed by grafting 3-carboxyl-4-ffuorophenylboronic acid (FPBA) onto Quaternary ammonium chitosan (QCS) to form Schiff base bonds with Oxidized gellan gum (OGG) (pH response), and by reacting FPBA with Epigallocatechin gallate (EGCG) to form borate bonds (ROS response). Hb@PDA nanoparticles were then loaded into the hydrogel. Under acidic conditions, the cleavage of Schiff base bonds triggers the release of Hb@PDA, providing sustained oxygen delivery to alleviate hypoxia. Additionally, in ROS-rich environments, the cleavage of borate ester bonds promotes EGCG release, exerting antioxidant and anti-inflammatory effects. QOF-EGCG/Hb@PDA displays excellent self-healing capacity and tissue adhesion, as well as remarkable photothermal activity. Furthermore, it demonstrates excellent antibacterial, antioxidant, anti-inflammatory, pro-angiogenic, and hemostatic properties. In vivo studies confirmed that this hydrogel markedly accelerated diabetic wound healing by enhancing epithelial regeneration, promoting collagen deposition, regulating inflammatory responses, and facilitating angiogenesis. Transcriptomic analysis revealed that the hydrogel promotes angiogenesis by activating the HIF-1α/VEGF signaling pathway, which plays a pivotal role in vascular formation and tissue regeneration, thereby accelerating the healing of diabetic wounds. Collectively, QOF-EGCG/Hb@PDA provides a promising and comprehensive strategy with great potential for application in diabetic wound treatment.

Synthesis of Reduction-Sensitive Brush-Arm Star Polymers via Combined Ring-Opening Metathesis Polymerization–Atom Transfer Radical Polymerization and Controlled-Release Properties

Xuhui Liang,Yuping Liu,Zhen Dong,Nanting Qiu,Fuwei Kong,Kun Cao,Zhong-Ren Chen

Mild reaction conditions and excellent functional group tolerance make ring-opening metathesis polymerization (ROMP) a widely used technique to synthesize functional materials. In this work, we used ROMP and atom transfer radical polymerization (ATRP) to prepare a reduction-sensitive brush-arm star polymer (BASP) drug carrier by a one-pot method and further conducted in vitro studies. First, a bifunctional norbornene-based ATRP initiator NB-Br and a reduction-sensitive dinorbornene cross-linker (DNC) were synthesized. Subsequently, NB-Br was subjected to ATRP using the hydrophilic monomer oligoethylene glycol monomethyl ether methacrylate (OEGMA) at different polymerization times to obtain the norbornenyl macromonomer NB-P(OEGMA)n with different degrees of polymerization. In the same reaction flask, NB-P(OEGMA)n underwent an ROMP reaction to generate a brush-shaped polymer. Next, DNC was added to the reaction flask, and the reaction was continued to obtain BASP. This one-pot method could successfully be used to achieve the transformation of the macromonomer to the brush-shaped polymer and subsequently to the star polymer. In vitro biological experiments demonstrated that BASP micelles exhibited reduction-triggered cleavage of disulfide bonds and degradation of polymers after coculture with 10 mM glutathione, promoting the release of intracellular doxorubicin and achieving the controlled release of drugs. We expect the findings of our study to serve as an efficient synthetic strategy to prepare carriers for biomedical applications.

A new pH/redox/light stimuli-responsive magnetic molecularly imprinted polymer based on β-cyclodextrin for recognition and controlled release of andrographolide

Lijie Liu,Yaxin Tan,Yuqi Bao,Sen Liao,Duoduo Wang,Qiaorong Ye,Chensi Zeng,Ni Tan

To improve the recognition ability and the controlled release of andrographolide (ADR), it is necessary to prepare a better intelligent drug delivery carrier. In this paper, the magnetic andrographolide imprinted polymer (ADR-MMIP) with pH/redox/light triple stimulus response was successfully prepared by reverse atom transfer radical polymerization (RATRP), where ADR was as the template, β-Cyclodextrin (β-CD) and 4-amino-4′-methacrylamide azobenzene (AMAB) as the bi-functional monomers , and both of N, N′-bis (acryloyl) cystamine (BAC) and ethylene glycol dimethacrylate (EGDMA) as cross-linkers. A series of adsorption experiments indicated that ADR-MMIP had excellent adsorption and selectivity capacities with the maximum adsorption capacity 41.43 mg g −1 and the factors α 4.256 and β 1.940 for the selectivity and relative selectivity, respectively. The adsorption mechanism of ADR-MMIP was investigated the multiphase adsorption with the chemical action on heterogeneous material surface by the linear fitting with the isothermal and kinetic adsorption models, and the in-vitro ADR release showed ADR-MMIP was sensitive to the triple-stimuli of pH/redox/light with the maximum cumulative drug release rate 91.53% when the above three conditions were applied concurrently. The above information demonstrated the ADR-MMIP should be of great application potential in the ADR extraction and isolation from mixture and the intelligent drug delivery system.
