Apple peel polyphenol alleviates antibiotic-induced intestinal dysbiosis by modulating tight junction proteins, the TLR4/NF-κB pathway and intestinal flora

Ziqian He,Na Deng,Bisheng Zheng,Yingjie Gu,Jian Chen,Tong Li,Rui Hai Liu,Ling Yuan,Wenzhi Li

The intestine and its flora have established a strong link with each other and co-evolved to become a micro-ecological system that plays an important role in human health. Plant polyphenols have attracted a great deal of attention as potential interventions to regulate the intestinal microecology. In this study, we investigated the effects of apple peel polyphenol (APP) on the intestinal ecology by establishing an intestinal ecological dysregulation model using lincomycin hydrochloride-induced Balb/c mice. The results showed that APP enhanced the mechanical barrier function of mice by upregulating the expression of the tight junction proteins at the transcriptional and translational levels. In terms of the immune barrier, APP downregulated the protein and mRNA expression of TLR4 and NF-κB. As for the biological barrier, APP promoted the growth of beneficial bacteria as well as increasing the diversity of intestinal flora. In addition, APP treatment significantly increased the contents of short-chain fatty acids in mice. In conclusion, APP can alleviate intestinal inflammation and epithelial damage as well as inducing potentially beneficial changes in the intestinal microbiota, which helps to reveal the potential mechanisms of host–microbial interactions and polyphenol regulation of intestinal ecology.

Polyphenols from pigmented quinoa as potential modulators of maize starch digestion: Role of the starch-polyphenol inclusion and non-inclusion complexes

Qiaozhi Zhang,Shiyu Fan,Hujun Xie,Yan Zhang,Linglin Fu

This study aimed to investigate the interaction modes between maize starch (MS) and quinoa polyphenols (QPs) under hydrothermal treatments and its influence on the multi-scale structure and digestive behaviors of MS through mechanistic analyses. Thermodynamic and microscopic results revealed that during starch gelatinization , the starch chains interacted with QP polyphenols to form new aggregates with retarded gelatinization. The X-ray diffraction and 13 C nuclear magnetic resonance spectroscopy further confirmed the presence of both V-type inclusion complexes and non-inclusion complexes in QP-complexed MS. Through multiple spectroscopic approaches, it was demonstrated that complexation with QPs promoted the formation of local helical and short-range ordered structures and crystallization in MS and led to resistance of the starch chains to digestive enzymes. As a consequence, the MS–QP complexes showed substantially reduced digestion rates with increases in the contents of resistant starch and slowly digestible starch, as well as lower values of the predicted glycemic index . Compared to white quinoa polyphenols, polyphenols from red and black quinoa displayed a greater efficiency in modulating the starch structure and digestive properties. This study provides a valuable basis for the application of pigmented quinoa polyphenols as functional ingredients in starchy foods with desired glycemic response.

Effect of polyphenolic structure and mass ratio on the emulsifying performance and stability of emulsions stabilized by polyphenol-corn amylose complexes

Huan Xie,Xianling Wei,Xiaoyan Liu,Weidong Bai,Xiaofang Zeng

O/W emulsions stabilized by polyphenol/amylose (AM) complexes with several polyphenol/AM mass ratios and different polyphenols (gallic acid (GA), epigallocatechin gallate (EGCG) and tannic acid (TA)) were prepared by a high-intensity ultrasound emulsification technique. The effect of the pyrogallol group number of polyphenols and the mass ratio of polyphenols/AM on polyphenol/AM complexes and emulsions was studied. The soluble and/or insoluble complexes gradually formed upon adding polyphenols into the AM system. However, insoluble complexes were not formed in the GA/AM systems because GA has only one pyrogallol group. In addition, the hydrophobicity of AM could also be improved by forming polyphenol/AM complexes. The emulsion size decreased with increasing pyrogallol group number on the polyphenol molecules at a fixed ratio, and the size could also be controlled by the polyphenol/AM ratio. Moreover, all emulsions presented various degrees of creaming, which was restrained by decreasing emulsion size or the formation of a thick complex network. The complex network was enhanced by increasing the ratio or pyrogallol group number on the polyphenol molecules, which was because the increasing number of complexes was adsorbed onto the interface. Altogether, compared to GA/AM and EGCG/AM, the TA/AM complex emulsifier had the best hydrophobicity and emulsifying properties, and the TA/AM emulsion had the best emulsion stability.

Sea bass protein-polyphenol complex stabilized high internal phase of algal oil Pickering emulsions to stabilize astaxanthin for 3D food printing

Lijuan Zhang,Chengfu Zhou,Shanghua Xing,Yannan Chen,Wentao Su,Haitao Wang,Mingqian Tan

The protective effect of sea bass protein (SBP)-(−)-epigallocatechin-3-gallate (EGCG) covalent complex-stabilized high internal phase (algal oil) Pickering emulsions (HIPPEs) on astaxanthin and algal oils was demonstrated in this study. The SBP-EGCG complex with better wettability and antioxidant activity was formed by the free radical-induced reaction to stabilize HIPPEs. Our results show that the SBP-EGCG complex formed dense particle shells surrounding the oil droplets, and the shells were crosslinked with the complex in the continuous phase to produce a network structure. The rheological analysis demonstrated that the SBP-EGCG complex endowed HIPPEs with high viscoelasticity, high thixotropic recovery, and good thermal stability, which were beneficial for three-dimensional (3D) printing applications. HIPPEs stabilized by SBP-EGCG complex were applied to improve the stability and bioaccessibility of astaxanthin and to delay algal oil lipid oxidation. The HIPPEs might become a food-grade 3D printing material served as a delivery system for functional foods.

Construction of a Tannin-Based Polyphenol Network on the Surface of a Filter Paper and Its Adsorption Performance for Methyl Violet

Qianlan Huang,Jialing Li,Zhiyuan Peng

An active hydroxymethyl group was introduced to the phenolic ring structure of tannin, and then the polyphenol network from hydroxymethyl tannin was immobilized on the surface of the filter paper (FP) by the reaction between hydroxymethyl tannin and cellulose of FP. Due to the introduction of the polyphenol structure on the surface of the FP, the ultraviolet absorption of the FP was broadened, the thermal stability of the FP was increased, and the FP was endowed with the antibacterial properties. The inhibition efficiency for Escherichia coli and Staphylococcus aureus was 82.3 and 80.1%, respectively. The FP-immobilized polyphenol network of tannin had good adsorption performance for methyl violet (MV), and the maximum adsorption capacity for MV was 131.5 mg·g–1. The adsorption efficiency could reach more than 98% when the initial mass concentration of MV was lower than 50 mg·L–1. The adsorption process conformed to the Langmuir isothermal adsorption model and quasi-secondary adsorption kinetic model. The FP-immobilized polyphenol network of tannin has good adsorption repeatability, and the preparation method is facile, green, and ecofriendly, which can be considered as a new potential adsorbent for dye wastewater.

Fully Biodegradable Films Based on Functionalized Natural Polyphenol/Poly(3-Hydroxybutyrate-co-3-Hydroxyvalerate) as a Potential Material for Food Packaging

Heng Xue Xiang,Zi Ye Chen,Wei Chen,Zhe Zhou,Bin Sun,Mei Fang Zhu

As a potential material for food packaging, fully biodegradable composite films based on novel functionalized natural polyphenol/Poly (3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) were successfully fabricated by solution casting, and also the functionalized natural polyphenol (C18TAx) was synthesized by grafting fatty acid onto natural polyphenol Tannic Acid (TA). The resultant C18TAx with better hydrophobicity was uniformly dispersed into a PHBV matrix. The influence of C18TAx dosage on the mechanical behavior, crystallization behavior and thermal properties of the C18TAx/PHBV composite films was investigated by uniaxial tensile mechanical testing, Differential Scanning Calorimeter (DSC), Polarizing Optical Microscope (POM) and Wide Angle X-ray Diffractometry (WAXD). The results revealed that the x value was about 0.09, showing that about 11/20 phenol hydroxy groups in TA molecules were replaced with fatty acid chains. Compared with those of neat PHBV, the tensile strength and elongation at break of the C18TA0.09/PHBV composite films with 10 wt.% C18TA0.09 content were enhanced by 121%, and 458%, respectively. This improvement was primarily ascribed to intermolecular hydrogen bonding interactions in composite films. This new type of fully biodegradable bio-based composite films shows great potential for food packaging.

Improving the hydrophilicity and fouling resistance of RO membranes by surface immobilization of PVP based on a metal-polyphenol precursor layer

Junhui Wu,Zhi Wang,Wentao Yan,Yao Wang,Jixiao Wang,Shichang Wang

A commercial polyamide reverse osmosis (RO) membrane was surface modified in a sequential two-step dip coating process for improvement of the hydrophilicity and fouling resistance. In the first step, an ultrathin metal-polyphenol precursor layer was deposited onto the membrane surface via the self-assembly of tannic acid (TA) and Fe(III) ions, and polyethylenimine (PEI) was added as a covalent cross-linker. In the second step, poly(N-vinylpyrrolidone) (PVP), a common hydrophilic polymer , was immobilized onto the metal-polyphenol precursor layer through the strong hydrogen bonding interactions between the lactam groups in PVP and the phenolic hydroxyl groups in the precursor layer. X-ray photoelectron spectroscopy (XPS), scanning electron microscope (SEM), transmission electron microscopy (TEM) and zeta potential measurements confirmed the successful immobilization of the precursor layer and PVP. Atomic force microscopy (AFM) test indicated a smoother surface of the modified membrane. Furthermore, the water flux of the modified membrane was only slightly decreased and the salt rejection was almost unchanged. More importantly, static contact angle measurement showed that the membrane surface hydrophilicity was significantly improved after the PVP immobilization (contact angle from 59.0±1.2° to 14.5±2.4°). Membrane fouling experiments were performed over three “filtration–rinsing” cycles using two proteins (lysozyme and bovine serum albumin) and one polysaccharide (sodium alginate) as model foulants. The results of the fouling experiments revealed that the fouling resistance of the reverse osmosis membrane was obviously improved by the surface immobilization of PVP. Besides, cross-flow filtration test under high hydrodynamic shear condition for 15 days indicated the excellent stability of the immobilized PVP. This work provided a new potential strategy for improving the fouling resistance of RO membranes.

Green and efficient extraction of polyphenols from the leaves of Quercus dentata Thunb and in vitro antioxidant activities of polyphenol extract

xiaofeng Yu,Jiahui Wang,min Zhang,Xianfeng Ma,xiuquan Xu

The ultrasonic-coupled deep eutectic solvents method was firstly applied to extract phenolic compounds efficiently from the leaves of Quercus dentata Thunb. Five factors including type of deep eutectic solvent, water content, solid-liquid ratio, extraction time and extraction temperature were investigated based on total polyphenol content. The optimal extraction parameters were obtained through response surface methodology , that is, the deep eutectic solvent was composed of choline chloride and 1, 4-butanediol (molar ratio 1:2) with water content of 38%, the solid-liquid ratio was 1:30 g/mL, the extraction time was 31 min, the extraction temperature was 55 °C. At this condition, total polyphenol content reached the highest value of 78.56 ± 2.44 mg gallic acid/g. In addition, eleven phenolic compounds were identified by high performance liquid chromatography quadrupole/time-of flight mass spectrometry as flavonol glycosides . And then, the extraction showed good antioxidant activities in reducing transition metals (Fe 3+ , Cu 2+ , Mo 6+ ) and scavenging free radicals (DPPH·, ABTS + ·, O 2 − · and ·OH). Among them, the IC 50 values of scavenging DPPH·, ABTS + ·, O 2 − · and ·OH free radicals were 10.87 ± 0.25 μg mL −1 , 8.04 ± 0.24 μg/mL, 149.47 ± 5.27 μg/mL and 131.73 ± 5.31 μg/mL, respectively. On the other hand, the extraction at the cellular level exerted its protective effect on PC12 cells by increasing the cell survival rate of H 2 O 2 -induced damaged cells, weakening the level of reactive oxygen species in damaged cells. Overall, this study provided a green and high-efficient strategy for extracting phenolic compounds with great antioxidant activities from Q. dentata leaves.

Local delivery of stem cell spheroids with protein/polyphenol self-assembling armor to improve myocardial infarction treatment via immunoprotection and immunoregulation

Chuanfeng An,Fei Shao,Canling Long,Yujie Zhang,Wen Nie,Rui Zeng,Zhenzhen Dou,Yuan Zhao,Yuanyuan Lin,Shiying Zhang,Lijun Zhang,Changle Ren,Yang Zhang,Guangqian Zhou,Huanan Wang,Jia Liu

Stem cell therapies have shown great potential for treating myocardial infarction (MI) but are limited by low cell survival and compromised functionality due to the harsh microenvironment at the disease site. Here, we presented a Mesenchymal stem cell (MSC) spheroid-based strategy for MI treatment by introducing a protein/polyphenol self-assembling armor coating on the surface of cell spheroids, which showed significantly enhanced therapeutic efficacy by actively manipulating the hostile pathological MI microenvironment and enabling versatile functionality, including protecting the donor cells from host immune clearance, remodeling the ROS microenvironment and stimulating MSC's pro-healing paracrine secretion. The underlying mechanism was elucidated, wherein the armor protected to prolong MSCs residence at MI site, and triggered paracrine stimulation of MSCs towards immunoregulation and angiogenesis through inducing hypoxia to provoke glycolysis in stem cells. Furthermore, local delivery of coated MSC spheroids in MI rat significantly alleviated local inflammation and subsequent fibrosis via mediation macrophage polarization towards pro-healing M2 phenotype and improved cardiac function. In general, this study provided critical insight into the enhanced therapeutic efficacy of stem cell spheroids coated with a multifunctional armor. It potentially opens up a new avenue for designing immunomodulatory treatment for MI via stem cell therapy empowered by functional biomaterials.

Schiff base crosslinking based dialdehyde-chitosan/gelatin/tea polyphenol electrospun nanofibrous membranes with excellent antibacterial and antioxidant activity for food packaging

Yu Ma,Ru Tian,Yudong Song,Mengxiao Liu,Jie Li,Zhichun Bi,Hui Huang,Yongxin Li

Chitosan and gelatin (GEL) are the two most common substrates for green active food packaging. However, chitosan is difficult to solute in water, and gelatin-based films are limited in application by gelatin’s poor active and basic properties. In this work, chitosan was modified to dialdehyde-chitosan (DCS), then it was blended with gelatin and doped with tea polyphenol (TP). Finally, the above materials were electrospined and Schiff-base crosslinked to dialdehyde-chitosan/gelatin/tea polyphenol nanofibrous membranes (DCS/GEL/TP-M) with antibacterial and antioxidant properties and good basic properties. The crosslinking process greatly enhanced the basic properties of the membranes, the addition of TP improved the activity of the membranes, with the inhibition of E. coli and S. aureus up to 91.08 % and 89.31 %, respectively, the scavenging of 1,1-diphenyl-2-picryl-hydrazyl radical (DPPH) free radicals of 85.43 %, and practical preservation experiments showed that the prepared film successfully extended the shelf life of strawberries to 7 days at 25 ℃. The results show that the DCS/GEL/TP-M have good application prospects in active food packaging.

A Natural Polyphenol-Regulated Microporous Soybean Protein Injectable Hydrogel Loaded with Placental Stem Cell-Derived Exosomes for Promoting Chronic Wound Healing

Hui Liu,Wen Jiang,Lu Han,Xiong Lu

Diabetic chronic wounds exhibit difficulties in healing due to prolonged high glucose and inflammatory conditions. Suppression of inflammation and restoration of the epithelial layer are crucial for preventing infections, reducing moisture loss, and restoring structural and functional integrity to the wound. Currently, there are numerous studies on dressings for diabetic chronic wounds, most of which can inhibit inflammation, but the effect on epithelial regeneration is limited. In this study, we developed a novel therapeutic system utilizing a microporous hydrogel regulated by the natural tea polyphenol EGCG for the treatment of chronic wounds. This hydrogel, made from soy protein isolate and dextran, exhibits anti-inflammatory properties and excellent biocompatibility, and is designed to be both injectable and biodegradable. This hydrogel facilitated the efficient loading and release of human placental stem cell-derived exosomes. Then the exosome-loaded hydrogel was used for full-thickness diabetic skin wound healing. Histological characterization confirmed that the exosome-loaded hydrogel promoted wound healing and epidermal regeneration. Immunofluorescence staining and Western blot experiments demonstrated the anti-inflammatory effect of the hydrogel. In short, this study proposed an Exo-hydrogel therapy as a promising potential for effective treatment of chronic wounds.

A time-saving pH-ultrasonic-shifting method for insoluble pea protein isolate to encapsulate polyphenol: Binding mechanism, structure, and functional characteristics

Linrong Yu,Qingchen Cui,Xiaolu Zhang,Liping Zhou,Mengshuang Li,Lei Tian,Xianggen Wu,Ying Jie

The application of the commercial pea protein isolate (CPPI) and polyphenols are often limited by their poor solubility. To address it, a time-saving pH-ultrasonic-shifting method was skillfully developed by combining the pH-shifting and ultrasonic methods to efficiently encapsulate polyphenols, and this final obtained nanoparticles was named CPPI-NAR-4. CPPI-NAR-4 has shown higher encapsulation efficiency (EE, 93.81 ± 0.56%) and loading capacity (LC, 8.58 ± 0.05%) for NAR , smaller particle size (126.40 ± 3.84 nm) and lower zeta potential (−18.50 ± 0.98 mV) as compared to nanoparticles prepared by other three known methods. This high EE and LC of CPPI-NAR-4 were attributed by both CPPI-NAR-4 and CPPI-4 having the higher surface hydrophobicity , the higher fluorescence intensity, and more protein expansion in their comparison. Multi-spectra evaluations indicated that the pH-ultrasonic-shifting process might produce more hydrogen bonding in the combination of CPPI and NAR . As to the interaction mechanisms between CPPI and NAR in CPPI-NAR nanoparticles, molecular docking results showed that hydrogen bonds , van der Waals forces , and hydrophobic interactions were dominant in the binding of NAR and CPPI; further molecular dynamics simulations revealed that NAR and CPPI had more interactions at pH 12 than at pH 7. Based on these experimental and simulative results, it could be suggested that the effects of ultrasonic on the binding between CPPI and NAR might be better at pH 12 than at pH 7. In conclusion, this pH-ultrasonic-shifting method was expected to be further developed for industrial fabricating protein-polyphenol nanoparticles such as CPPI-NAR.
