Increasing research has recognized that the ubiquitous presence of microplastics in terrestrial environments is undeniable, which potentially alters the soil ecosystem properties and processes. The fact that microplastics with diverse characteristics enter into the soil may induce distinct effects on soil ecosystems. Our knowledge of the impacts of microplastics with different polymers, shapes, and concentrations on soil bacterial communities is still limited. To address this, we examined the effects of spherical microplastics (150 μm) with different polymers (i.e., polyethylene (PE), polystyrene (PS), and polypropylene (PP)) and four shapes of PP microplastics (i.e., fiber, film, foam, and particle) at a constant concentration (1%, w /w) on the soil bacterial community in an agricultural soil over 60 days. Treatments with different concentrations (0.01–20%, w/w) of PP microplastic particles (150 μm) were also included. The bacterial communities in PE and PP treatments showed a similar pattern but separated from those in PS-treated soils, indicating the polymer backbone structure is an important factor modulating the soil bacterial responses. Fiber, foam, and film microplastics significantly affected the soil bacterial composition as compared to the particle. The community dissimilarity of soil bacteria was significantly ( R 2 = 0.592, p < 0.001) correlated with the changes of microplastic concentration. The random forest model identified that certain bacteria belonging to Patescibacteria were closely linked to microplastic contamination. Additionally, analysis of the predicted function further showed that microplastics with different characteristics caused distinct effects on microbial community function. Our findings suggested that the idiosyncrasies of microplastics should not be neglected when studying their effects on terrestrial ecosystems.

Soils and waters are heavily contaminated by antimony in Xikuangshan (XKS) mine area. It is widely accepted that oxidative dissolution of sulfide minerals and aqueous dissolution are the most prevalent geochemical mechanisms for the release of Sb to the environment. Bosea sp. AS-1 is an antimonite-oxidizer isolated from the mine slag in Xikuangshan Sb mine. Whole genome sequencing revealed the presence of multiple sulfur-oxidizing genes, antimony (Sb) metabolism genes and carbon fixation genes in AS-1′s genome. We therefore hypothesized that under oxic conditions, AS-1 could mediate the oxidation of sulfide and Sb(III) in stibnite (Sb 2 S 3 ) and lead to the release of Sb. Indeed, strain AS-1 was discovered as an autotrophic Sb(III)-oxidizer. Antimony mobilization studies conducted with strain AS-1 showed significantly enhanced mobilization of Sb, and complete oxidation of released Sb and sulfur to Sb(V) and sulfate. In addition, AS-1 induced a faster release of Sb under heterotrophic condition, and new acicular minerals might form. These findings support the hypothesis that microorganisms play an important role in the mobilization and transformation of Sb in XKS mine area and may contribute to our further understanding of the Sb biogeochemical redox cycle in natural environment.

Mercury and ferrous iron commonly co-exist in anaerobic environments, the hotspots of microbial mercury methylation that leads to formation of neurotoxic methylmercury. However, the influence of ferrous iron on mercury bioavailability and methylation remains unclear. Here, we discover that microbial methylmercury production is inhibited by iron addition in a non-monotonical manner, reaching the minimal level at the initial molar ratio of dissolved Fe/Hg as 1/3, when highly aggregated Hg–S–Fe nanoparticles occur and prevent the nanoparticle–bacteria interactions. The dominant mercury species formed upon Hg–S–Fe co-precipitation is iron-incorporated nano-metacinnabar with short-range structural disorder. The methylation potential of this mercury species is inversely correlated with the aggregation state of nanoparticles. The non-monotonical trends of the aggregation state and subsequent methylation of nano-metacinnabar are due to the opposite effects of ferrous iron on the nucleation and particle attachment that determine the formation of nanoparticle aggregates. Increased Fe/Hg ratio hinders particle nucleation and thus decreases the density of particle monomers for aggregation. However, Fe substitution of sub-surface Hg weakens the inner-sphere coordination of surface Hg with natural thiol ligands and consequently enhances nano-metacinnabar aggregation. This new mechanistic insight is important for assessing and minimizing the risks of mercury methylation, and informs how intertwined iron and sulfur cycling may influence metal bioavailability in the changing environment.

Objective Bladder cancer(BCa) was a disease that seriously affects patients’ quality of life and prognosis. To address this issue, many researches suggested that the gut microbiota modulated tumor response to treatment; however, this had not been well-characterized in bladder cancer. In this study, our objective was to determine whether the diversity and composition of the gut microbiota or the density of specific bacterial genera influence the prognosis of patients with bladder cancer. Methods We collected fecal samples from a total of 50 bladder cancer patients and 22 matched non-cancer individuals for 16S rDNA sequencing to investigate the distribution of Parabacteroides in these two groups. Further we conducted follow-up with cancer patients to access the impact of different genera of microorganisms on patients survival. We conducted a Fecal Microbiota Transplantation (FMT) and mono-colonization experiment with Parabacteroides distasonis to explore its potential enhancement of the efficacy of anti-PD-1 immunotherapy in MB49 tumor-bearing mice. Immunohistochemistry, transcriptomics and molecular experiment analyses were employed to uncover the underlying mechanisms. Results The 16S rDNA showed that abundance of the genus Parabacteroides was elevated in the non-cancer control group compared to bladder cancer group. The results of tumor growth curves showed that a combination therapy of P. distasonis and ICIs treatment significantly delayed tumor growth and increased the intratumoral densities of both CD4 + T and CD8 + T cells. The results of transcriptome analysis demonstrated that the pathways associated with antitumoral immune response were remarkably upregulated in the P. distasonis gavage group. Conclusion P. distasonis delivery combined with α-PD-1 mAb could be a new strategy to enhance the effect of anti-PD-1 immunotherapy. This effect might be achieved by activating immune and antitumor related pathways.

Sulfamethoxazole (SMX) is an extensively consumed sulfonamide antimicrobial agent and is frequently detected in surface water. This work studied the degradation process of SMX in anodic chamber of microbial fuel cell (MFC) reactors. We found that the biodegradation of SMX could be achieved after acclimation and even high concentrations of SMX (e.g. 200 ppm) could be rapidly degraded. Excitation and emission matrix fluorescence spectroscopy analysis revealed that the chemical structure of SMX was altered during the process. Q-Exactive hybrid quadrupole-Orbitrap mass spectrometry was used to identify the degradation byproducts of SMX. The activity of electrode biofilm was examined afterwards and it was found that the microbe was in an active state. High-throughput sequencing analysis suggested that the microbial community structure was greatly changed during the process; some reported SMX scavengers, such as Achromobacter and Pseudomonas, were abundant in the reactors. Some metazoans were also recognized in the biofilm samples, which indicates that the operation of the MFC reactors was in a steady state. This study discusses the degradation mechanism of SMX and explores the microbial community response during the process, which provides useful information for the application of MFC in antibiotic elimination.

An integrated constructed wetland-microbial electrolysis cell (ICW-MEC) system was investigated for nitrogen removal under different pollution loads, hydraulic loads (HLRs), and aeration conditions. The treatment performance of each unit and the microbial community characteristics for nitrogen removal were elucidated. The results showed that, on average, 80% of NH4+-N, around 70% of nitrate nitrogen (NO3−-N), and 70% of total nitrogen (TN) were removed by the system under three pollution loads, with less influence by pollution loads. The high removal efficiencies of NH4+-N (81.8%), NO3−-N (71.4%), and TN (72.8%) indicated tolerable to high HLRs. The intermittent aeration negatively affected NH4+-N removal, while increasing NO3−-N and TN removals by 3.2–13.0% and 3.7–16.7%. The contribution efficiencies of the secondary unit to the removal of NH4+-N, NO3−-N, TN, and total organic carbon (TOC) reached 47.4%, 55.0%, 45.9%, and 38.8%, respectively. The distinct microbial communities existed in various units of the ICW-MEC system, which were strongly affected by environmental factors and shaped by diverse fillers and structures of the system. The dominant bacteria contributed to the efficient nitrogen removal performance of the ICW-MEC system. The three units exerted their advantages to ensure efficient and stable system operation.

We selected an array of whole grains (WGs), including highland barley, buckwheat, coix seed, foxtail millet, barley, oats, proso millet and sorghum to determine their carbohydrates profile before and after extrusion-expansion, estimated glycemic index (GI) and fermentation characteristics. Our findings reveal that the extrusion processing can significantly influence the structural properties and digestibility of starch. A notable rise in the total dietary fiber (TDF) content was found in foxtail millet, barley and sorghum ( p < 0.05) after extrusion. Digestion results of extruded whole grains (EWGs) suggested that coix seeds, oats, and sorghum after extrusion can better manage glycemic index (GI) values due to low starch hydrolysis degree and higher resistant starch (RS) and slowly digestible starch (SDS) content. EWGs with higher TDF content (HE, FE, SE, OE and BE) demonstrated greater SCFA yield, with extruded highland barley exhibiting a superior capacity for generating propionic acid and butyric acid. Extruded barley, buckwheat, oats, and proso millet significantly boosted microbial diversity. All eight EWGs notably curtailed Bilophila growth and fostered beneficial bacteria such as Prevotella , Megamonas , Megasphaera , Bifidobacterium , Parabacteroides and Phascolarctobacterium . This study provides a robust scientific foundation for the application of extrusion technology in functional WGs products.

High myopia (HM) is a leading cause of blindness worldwide with currently no effective interventions available. A major hurdle lies in its often isolated perception as a purely ocular morbidity, disregarding potential systemic implications. Recent evidence suggests the existence of a gut-eye axis; however, the role of gut microbiota in the pathogenesis of HM remains largely unexplored. Herein, we provide a potential crosstalk among HM’s gut dysbiosis, microbial metabolites, and scleral remodeling. Utilizing 16S rRNA gene sequencing, we observed an altered gut microbiota profile in HM patients with a significant reduction in probiotic abundance compared with healthy controls. Subsequent targeted metabolic profiling revealed a notable decrease in plasma levels of the gut microbiota-derived metabolite indole-3-acetic acid (3-IAA) among HM patients, which is closely associated with the reduced probiotics, both negatively correlated with HM severity. Genetic analyses determined that gut microbiota are causally associated with myopia risk. Importantly, when mice subjected to HM modeling receive fecal microbiota transplantation from healthy donors, there is an increase in 3-IAA plasma levels and simultaneous retardation of HM progression along with better maintenance of collagen type I alpha 1 (COL1A1) expression in the sclera. Furthermore, 3-IAA gavage achieves similar effects. Mechanistic investigations confirm the transcriptional activation of COL1A1 by 3-IAA via promoting the enrichment of SP1 to its promoter. Together, our findings provide novel insights into the gut microbiota-eye axis in the pathogenesis of HM and propose new strategies for HM intervention by remodeling the gut microbiota and indole supplementation.

Microplastics, as emerging pollutants, can adsorb polycyclic aromatic hydrocarbons (PAHs) in aqueous environments. However, the coeffects of PAHs and microplastics on aquatic organisms are vague. In this study, nanoscale and micron-scale microplastics (30 mg/kg) and phenanthrene (10 mg/kg) were added to aquatic sediment to explore the effects of different particle sizes of microplastics and typical PAHs on invertebrates ( Tubifex tubifex ) and microbes after 28 days operation. The fluorescence signal of fluorescently labelled micron-scale and nanoscale microplastics were detected in T. tubife x, especially nanoscale microplastics. The reduced activities of CAT, GST, SOD and MDA in T. tubifex suggested that the micron-sized microplastics and PAHs exacerbated the oxidative damage to T. tubif ex. Microbial analysis confirmed that microplastics and PAHs decreased the abundance of Proteobacteria , Fusobacteriota and Bacteroidetes in the gut of T. tubifex and thereby affected the degradation of phenanthrene. Microplastics also reduced the abundance of Bacteroidetes , Acidobacteria and Proteobacteria in sediment. The abundance of salicylate hydroxylase enzyme that related to PAHs metabolism was increased with the addition of phenanthrene. The results in this study would be useful for further risk assessment of PAHs and microplastics in aquatic environments.

The use of antibacterial and disinfection products is increasing in recent years. Para-chloro-meta-xylenol (PCMX), a widely used antimicrobial agent, has been detected in various environments. Herein, the impacts of PCMX with long-term exposure on anaerobic sequencing batch reactors were investigated. The high concentration (50 mg/L, GH group) PCMX severely inhibited the nutrient removal process, and the low concentration group (0.5 mg/L, GL group) slightly affected the removal efficiency which was recovered after 120 days of adaptation compared to the control group (0 mg/L, GC group). Cell viability tests indicated that PCMX inactivated the microbes. A significant reduction in bacterial α-diversity was observed in the GH but not the GL group. The microbial communities were shifted upon PCMX exposure, among which Olsenella , Novosphingobium, and Saccharibacteria genera incertae Sedis became the predominant genera in the GH groups. Network analyses showed that PCMX significantly reduced the complexity and interactions of the microbial communities, consistent with the negative impacts on bioreactor performance. Real-time PCR analysis indicated that PCMX affected the behavior of antibiotic resistance genes (ARGs), and the relationship between ARGs and bacterial genera gradually became complicated after long-term exposure. Most detected ARGs decreased on Day 60 but increased on Day 120 especially in the GL group, implying the potential risk of environment-relevant concentration of PCMX in the ecosystems. This study provides new insights into the understanding of the impacts and risks of PCMX on wastewater treatment processes.

Considering the inevitable release of antibiotics and nanoparticles (NPs) into the nitrogen containing wastewater, the combined impact of CuO NPs and sulfamethoxazole (SMX) antibiotic on partial nitrification (PN) process was investigated in four identical reactors. Results showed that the bioactivity of the aerobic ammonia-oxidizing bacteria (AOB) decreased by half after they were exposed to the combination of CuO NPs and SMX for short-term; however, there was no obvious variation in the bioactivity of AOB when they were exposed to either CuO NPs or SMX. During long-term exposure, the ammonia removal efficiency (ARE) of CuO NPs improved whereas that of SMX decreased, while the combination of CuO NPs and SMX significantly decreased ARE from 62.9% (in control) to 38.2% and had an unsatisfactory self-recovery performance. The combination of CuO NPs and SMX significantly changed the composition of microbial community, decreased the abundance of AOB, and significantly suppressed PN process. Reegarding the resistance genes, the CuO NPs-SMX combination did not improve the expression of copA , cusA , sul1 and sul2 ; however, it significantly induced the expression of sul3 and sulA .

Background The gut microbiome plays a potential role in clinical events in preterm infants and may affect their lateral development. Understanding the initial colonization of microbes in the gut, their early dynamic changes, and the major factors correlated with these changes would provide crucial information about the developmental process in early life. Methods The present study enrolled 151 preterm infants and examined the longitudinal dynamics of their fecal microbiome profiles during the period of hospitalization using 16S ribosomal RNA gene sequencing. Random forest modeling was used to predict postnatal age (Age), postmenstrual age (PMA), and gestational age (GA), using gut microbiome features. Results Principal coordinate analysis revealed that the gut microbiome of the preterm infants displayed an obvious time-dependent change pattern, which showed the strongest association with Age, followed by PMA, and a much weaker association with (GA). Random forest modeling further evidenced the time-dependent change pattern, with the Pearson’s correlation coefficients between the actual values and the gut microbiome-predicted values being 0.68, 0.53, and 0.38 for postnatal, postmenstrual, and gestational age, respectively. The microbiome dynamism could be further divided into four Age stages, each with its own characteristic microbial taxa. The first 1–4 days (T1 stage) represented the meconium microbiome, with colonization of a high diversity of microbes before or during delivery. During 5–15 days (T2 stage), the gut microbiome of the preterm infants underwent a rapid turnover, in which microbial diversity declined, and stabilized afterward. Enterobacteriaceae , Enterococcaceae , Streptococcaceae , Staphylococcaceae , and Clostridiaceae were the major classes in the gut microbiome in the lateral stages of development (T3–T4 stage). Conclusions Postnatal age, rather than the gestational age, is significantly correlated with the gut microbiome of preterm infants, suggesting that clinical interventions contribute more to the early dynamics of gut microbiome in preterm infants than the natural development of the gut.
