Precise isolation of circulating tumor cells (CTCs) is proved to be significant for early cancer diagnosis and downstream analysis. Most of the existing strategies yield low purity or cause unexpected damage to cells because of foreign material introduction. To avoid foreign material caused damage and achieve high efficiency simultaneously, this work presents an innovative strategy using tumor cell targeting molecules to bind homologous red blood cells (RBCs) with tumor cells, which results in obvious optical constant differences (both size and mean refractive index) between CC-RBCs (RBC conjugated CTCs) and other blood cells. Then the modified CTCs can be precisely separated under laser illumination in an optofluidic system. Experiments show that CTCs are efficiently modified with erythrocytes and finally isolated from blood at high purity (more than 92%) and a high recovery rate (over 90%). In the whole process, CTCs are proved to keep membrane and function integrity. The combination of homologous RBC binding and an optofluidic system will provide a convenient tool for cancer early diagnosis and treatment monitoring, which exhibits good performance in CTC non-invasive and precise isolation, thus showing great potential.

Nanoparticle shape has been recognized as a crucial parameter to affect the transport across various biological barriers, but its impact on drug release and the resulting therapeutic efficacy is less understood. Inspired by erythrocytes with shape-facilitated oxygen-carrying and penetrating abilities, we constructed artificial erythrocyte-like nanoparticles (RNDs) by wrapping discoidal mesoporous silica nanoparticles with red blood cell membrane . We observed that, compared with their spherical and rod-shaped counterparts with monotonic drug release profiles, RNDs displayed an on-demand drug release pattern mimicking natural erythrocytes, that is, they could rapidly release loaded oxygen and doxorubicin (DOX) in hypoxic condition but were relatively stable in high oxygen areas. Besides, the discoidal shape also endowed RNDs with facilitated transport capability in tumor extracellular matrix, contributing to increased tumor permeability. In tumor models, systemically administrated RNDs efficiently infiltrate throughout tumor tissue, successfully relieve tumor hypoxia , and further altered the cancer cell cycle status from G1 to G2 phase, enhancing cancer cell sensitivity to DOX correlated with improved chemotherapy efficacy. In contrast, nanospheres show hampered permeability, and nanorods suffer from insufficient intratumoral drug accumulation. These findings can offer guidelines for the use of particle shape as a design criterion to control drug release, transportation, and therapeutics delivery.

Two-photon polymerization (TPP) microfabrication is an advanced technology to fabricate precise three-dimensional (3D) hydrogel micro/nanostructure. 3D hydrogel microstructures fabricated by TPP with sophisticated details and appropriate stiffness are able to effectively simulate the microenvironment used in tissue engineering and drug delivery. The mechanical property of the microstructures, for instance, the Young’s modulus is crucial to achieve the microstructures with high fidelity. In this study, the mechanical property of the poly(ethylene glycol) (PEG) 3D microstructures fabricated with various laser powers, writing speeds and layer distances in the air was investigated by characterizing the Young’s modulus. Meanwhile, the Young’s modulus of the microstructure with different layer distances in water was determined as 3.50–6.52 MPa. Furthermore, 3D PEG microstructures simulating red blood cell morphology of different postures and sizes were successfully fabricated.

Nanoformulations for combining chemotherapy, chemodynamic therapy, and photothermal therapy have enormous potential in tumor treatment. Coating nanoformulations with cell membranes endows them with homologous cellular mimicry, enabling nanoformulations to acquire new functions and properties, including homologous targeting and long circulation in vivo , and can enhance internalization by homologous cancer cells. Herein, we fused multifunctional biomimetic nanoformulations based on Cu-doped zeolitic imidazolate framework-8 (ZIF-8). Hydroxycamptothecin (HCPT), a clinical anti-tumor drug, was encapsulated into ZIF-8, which was subsequently coated with polydopamine (PDA) and red blood cell membrane. The as-fabricated biomimetic nanoformulations showed an enhanced cell uptake in vitro and the potential to prolong blood circulation in vivo , producing effective synergistic chemotherapy, chemodynamic therapy, and photothermal therapy under the 808 nm laser irradiation. Together, the biomimetic nanoformulations showed a prolonged blood circulation and evasion of immune recognition in vivo to provide a bio-inspired strategy which may have the potential for the multi-synergistic therapy of breast cancer.

Fluorescent carbon dots (CDs) have a variety of biomedical applications such as bio-imaging. Successful application of CDs in clinic greatly depends on their biocompatibility and favorable bio-functions. In the present study, we report a facile and efficient method to synthesize a CDs-based nanohybrid bearing a multi-hydroxy hyperbranched polyglycerol (HPG) shell by combing a “grafting from” method with an anionic ring-opening polymerization technique. The resultant CDs-g-HPG had high water-dispersibility, low cytotoxicity, improved hemocompatibility and strong green fluorescence. The results demonstrate that the CDs-g-HPG possesses promising potential for bio-imaging.

A novel microcarrier based on red blood cells was prepared and characterized. Proof of principle was demonstrated by using sheep red blood cells that were treated with glutaraldehyde to render the cells generally resistant to hemolysis. Magnetic beads of approximately 10 nm in diameter were prepared and attached to the surface of glutaraldehyde-treated sheep red blood cells by electrostatic interaction at various concentrations. The composites were characterized by scanning electron microscopy. Dilute coverage of magnetic beads allowed the red blood cells to be separated from solution in the presence of a magnetic field, eliminating the need for centrifugation. The magnetized glutaraldehyde-treated red blood cells were then functionalized with human immunoglobulin and used in flow cytometry for the determination of antihuman immunoglobulin. The results indicated that such magnetized glutaraldehyde-treated red blood cell may be used as carriers in flow cytometry. The approach may allow the preparation of carriers using other red blood cells.

Acute hemorrhage that occurs after trauma is a life-threatening condition. Hence, to halt massive bleeding, there is a critical need to develop a suitable therapy. In this study, we developed self-propelling chestnut-like particles (Pro-MAS) comprising a macro-acanthosphere (MAS) coated with calcium carbonate and protonated tranexamic acid to puncture red blood cells (RBCs) and thus activate hemostasis. In vitro assessments revealed that Pro-MAS was biocompatible, biodegradable, and nontoxic; furthermore, it was capable of puncturing RBCs to release procoagulants and activate platelet aggregation for hemostasis. Animal tests showed that self-propelling Pro-MAS effectively traveled through blood flow to the deep ends of wounds; hemorrhage was controlled within 90 s and 4 min in the injured liver and bleeding femoral artery, respectively. Compared with a commercial hemostat, superior hemostasis was achieved with Pro-MAS, which could be ascribed to its functional and structural features. Overall, traveling Pro-MAS possessed sufficient impact force to puncture RBCs and sufficient momentum to reach the targeted bleeding sites. The present study demonstrated the ability of a novel platform, self-propelling MAS particles, to trigger hemostasis by puncturing RBCs. To the best of our knowledge, this is the first trial in which the release of endogenous procoagulants is promoted without the addition of exogenous procoagulants for severe hemorrhage control.

Liver fibrosis is characterized by the aberrant accumulation of extracellular matrix (ECM) components, primarily due to the persistent activation of hepatic stellate cells (HSCs). It is well-known that chronic liver inflammation from various etiologies leads to hepatocellular damage, which activates HSCs, initiating and exacerbating fibrosis. This presents a significant challenge for therapeutic interventions aimed at restoring ECM balance. Here, we present CPT-pH@RLs, a novel drug delivery system consisting of pH-sensitive liposomes modified with erythrocyte membranes (RBCm), designed to deliver cryptotanshinone to the liver. The results of cellular uptake and real-time in vivo imaging indicate that the modification of RBCm reduces the uptake of CPT-pH@RLs by macrophages and prolongs the in vivo cycling time of these particles. CPT-pH@RLs effectively reduce liver inflammation by inhibiting secretion of interleukin-1β (IL-1β). Moreover, they improve liver function, decrease ECM deposition, and alleviate liver fibrosis. Our findings demonstrate that CPT-pH@RLs represent a safe and effective passive targeting drug delivery system for anti-inflammatory and ECM normalization therapies, offering a promising approach for the reversal of liver fibrosis.

Challenges for glioma treatment with nanomedicines include physio-anatomical barriers (the blood-brain barrier and blood-brain tumor barrier), low drug loading capacity, and limited circulation time. Here, a red blood cell membrane-coated docetaxel drug nanocrystal (pV-RBCm-NC(DTX)), modified with pHA-VAP (pV) for all-stage targeting of glioma, was designed. The NC(DTX) core exhibited a high drug loading capacity but low in vivo stability, and the RBCm coating significantly enhanced the stability and prolonged in vivo circulation. Moreover, the Y-shaped targeting ligand pV was modified by a mild avidin-biotin interaction, which endowed RBCm-NC(DTX) with superior barrier-crossing ability and therapeutic efficacy. The integration of nanocrystal technology, cell membrane coating, and the avidin-biotin insertion method into this active targeting biomimetic formulation represents a promising drug delivery strategy for glioma.

Maternal anemia has been identified as a contributing factor to adverse reproductive outcomes associated with cadmium (Cd) exposure, a common heavy metal. Our recent findings suggest that inhibited erythroid differentiation and enucleation also play significant roles in the direct embryonic toxicity resulting from maternal Cd exposure. However, the effects of Cd exposure on lipid metabolism remodeling, which is essential for physiological erythropoiesis, remain poorly understood. In the present study, pregnant mice were administered low doses of CdCl 2 via oral exposure from early to late gestation to mitigate Cd-induced maternal anemia. Compared to vehicle-treated controls, embryos from Cd-treated mice exhibited a slight decrease in weight, though without signs of atrophy. Consistent with our previous observations, fetal livers from Cd-exposed embryos demonstrated a dose-dependent inhibition of erythroid differentiation, as confirmed by ex vivo analysis. Notably, an intrinsic decrease in lipid peroxidation during erythroid differentiation was observed in the bone marrow and fetal livers of vehicle-treated mice, attributed to diminished lipid content. In contrast, this decrease in lipid peroxidation was absent in fetal liver erythroblasts from Cd-treated mice, where an increase in lipid peroxidation was instead noted. These findings elucidate a potential mechanism, lipid peroxidation, underlying Cd-induced embryonic toxicity.

Macrophages play pivotal roles in the regulation of inflammatory responses and tissue repair, making them a prime target for inflammation alleviation. However, the accurate and efficient macrophages targeting is still a challenging task. Motivated by the efficient and specific removal of apoptotic cells by macrophages efferocytosis, a novel biomimetic liposomal system called Effero-RLP (Efferocytosis-mediated Red blood cell hybrid Liposomes) is developed which incorporates the membrane of apoptotic red blood cells (RBCs) with liposomes for the purpose of highly efficient macrophages targeting. Rosiglitazone (ROSI), a PPARγ agonist known to attenuate macrophage inflammatory responses, is encapsulated into Effero-RLP as model drug to regulate macrophage functions in DSS-induced colitis mouse model. Intriguingly, the Effero-RLP exhibits selective and efficient uptake by macrophages, which is significantly inhibited by the efferocytosis blocker Annexin V. In animal models, the Effero-RLP demonstrates rapid recognition by macrophages, leading to enhanced accumulation at inflammatory sites. Furthermore, ROSI-loaded Effero-RLP effectively alleviates inflammation and protects colon tissue from injury in the colitis mouse model, which is abolished by deletion of macrophages from mice model. In conclusion, the study highlights the potential of macrophage targeting using efferocytosis biomimetic liposomes. The development of Effero-RLP presents novel and promising strategies for alleviating inflammation.

Breast cancer is the most prevalent fatal cancer among women worldwide and the leading cause of death for women. Ferroptosis is a form of programmed cell death that relies on iron and is non-apoptotic, triggered by the inhibition of the cellular antioxidant system. Photodynamic therapy (PDT) employs photosensitizers to produce reactive oxygen species (ROS), increasing oxidative stress in tumor cells. When combined with ferroptosis, PDT can work synergistically to regulate intracellular redox balance. In this study, we designed engineered nano-erythrocyte membranes for targeted delivery of Chlorin e6 (Ce6) and cisplatin (DDP) to enhance breast cancer treatment. By using mild ultrasound, Ce6 and DDP were co-loaded onto the nano-erythrocyte membranes, combining ferroptosis inducers and photosensitizers to combat breast cancer. To improve targeting capability towards breast cancer, RGD cyclic peptides were modified onto the nano-erythrocyte membranes through a thiol–maleimide coupling reaction. The RGD-modified nano-erythrocyte membranes co-loaded with Ce6 and DDP not only inherited the good stability and significant biocompatibility of red blood cell membranes but also promoted the uptake by breast cancer cells, effectively inducing ferroptosis in these cells. In conclusion, this multifunctional ‘natural’ nanodrug delivery system provides an effective and safe method for PDT combined with ferroptosis for breast cancer treatment.
