Internal standard method for the measurement of doxorubicin and daunorubicin by capillary electrophoresis with in-column double optical-fiber LED-induced fluorescence detection

Xiupei Yang,Huanhuan Gao,Fan Qian,Chuan Zhao,Xiangjun Liao

An internal standard method has been developed for the simultaneous determination of anthracycline antibiotics, doxorubicin (DOX) and daunorubicin (DAN), in rabbit plasma using capillary electrophoresis (CE) with in-column double optical-fiber LED-induced fluorescence detection (CE-ICDOF-LED-FLD). Rhodamine B (RhB) was selected as an internal standard because its emission wavelength is similar to that of the anthracycline antibiotics. Parameters including buffer pH, buffer concentration, organic solvents and separation voltage have been investigated to explore the sensitivity and separation efficiency of DOX and DAN. The optimal electrophoretic separation conditions were a borate buffer (15   mM, pH 9.0) containing 50% acetonitrile (v/v), 10   s hydrodynamic injection at a height of 20   cm and a separation voltage of 15   kV. The developed CE-ICDOF-LED-FLD method provides limits of detection of 18 and 13   ng/mL for DOX and DAN in rabbit plasma samples, respectively. The recoveries ranging from 93.7 to 104.8% and the relative standard deviations at 1.1–1.7% were achieved for DOX and DAN in spiked rabbit plasma samples.

Effects of Erythromycin on Nereis succinea and the Intestinal Microbiome across Different Salinity Levels

Dali Wang,Qi Ding,Yuheng Zhong,Xiaofeng Han,Long Fu,Jing Yu,Kunting Li,Jing You

The exposure of aquatic organisms to pollutants often occurs concomitantly with salinity fluctuations. Here, we reported the effects of erythromycin (0.250, 7.21, and 1030 μg/L) on marine invertebrate N. succinea and its intestinal microbiome under varying salinity levels (5‰, 15‰, and 30‰). The salinity elicited significant effects on the growth and intestinal microbiome of N. succinea. The susceptibility of the intestinal microbiome to erythromycin increased by 8.7- and 6.2-fold at salinities of 15‰ and 30‰, respectively, compared with that at 5‰ salinity. Erythromycin caused oxidative stress and histological changes in N. succinea intestines, and inhibited N. succinea growth in a concentration-dependent manner under 30‰ salinity with a maximum inhibition of 25%. At the intestinal microbial level, erythromycin enhanced the total cell counts at 5‰ salinity but reduced them at 15‰ salinity. Under all tested salinities, erythromycin diminished the antibiotic susceptibility of the intestinal microbiome. Two-way ANOVA revealed significant interactive effects (p < 0.05) between salinity and erythromycin on various parameters, including antibiotic susceptibility and intestinal microbial diversity. The present findings demonstrated the significant role of salinity in modulating the impacts of erythromycin, emphasizing the necessity to incorporate salinity fluctuations into environmental risk assessments.

Interactive effects of roxithromycin and freshwater microalgae, Chlorella pyrenoidosa: Toxicity and removal mechanism

Jiping Li,Zhongfang Min,Wei Li,Lijie Xu,Jiangang Han,Pingping Li

Roxithromycin (ROX) has received increasing concern due to its large usage, ubiquitous detection in environment and high ecotoxicology risk. This study investigated the acute and chronic effects of ROX on the growth, chlorophyll, antioxidant enzymes, and malonaldehyde (MDA) content of Chlorella pyrenoidosa , as well as the removal mechanism of ROX during microalgae cultivation. The calculated 96 h median effective concentration of ROX on yield (E y C 50 ) and specific growth rate (E r C 50 ) of C. pyrenoidosa was 0.81 and 2.87 mg/L, respectively. After 96 h exposure, 1.0 ~ 2.0 mg/L of ROX significantly inhibited the synthesis of chlorophyll and promoted the activities of SOD and CAT ( p  < 0.05). The MDA content increased with the ROX concentration increasing from 0.5 ~ 1.0 mg/L, and then decreased to 105.76% of the control exposure to 2.0 mg/L ROX, demonstrating the oxidative damage could be moderated by the upregulation of SOD and CAT activities. During the 21 d chronic exposure, low concentration of ROX (0.1 and 0.25 mg/L) showed no significant effect on the growth and chlorophyll content of algae during the first 14 d, but significantly inhibited the growth of algae and the synthesis of chlorophyll at 21 d ( p  < 0.05 or p  < 0.01). 1.0 mg/L ROX significantly inhibited the growth of microalgae during 3 ~ 21 d and the synthesis of chlorophyll at 7 ~ 21 d. High concentration and long-term exposure of low concentration of ROX caused the SOD and CAT activities and MDA content to increase, demonstrating a higher level of oxidative damage of microalgae. During the first 14 d, abiotic removal of ROX played a more important role, contributing about 12.21% ~ 21.37% of ROX removal. After 14 d, the biodegradation of ROX by C. pyrenoidosa gradually became a more important removal mechanism, contributing about 45.99% ~ 53.30% of ROX removal at 21 d. Bio-adsorption and bioaccumulation both played minor roles in the removal of ROX during algae cultivation.

Effect of erythromycin exposure on the growth, antioxidant system and photosynthesis of Microcystis flos-aquae

Jinjin Wan,Peiyong Guo,Xiaofang Peng,Keqi Wen

Erythromycin, a macrolide antibiotic, is commonly used in human life. This compound and its derivatives have been detected in various aquatic compartments and may pose a serious threat to aquatic organisms. This study investigated the effects of erythromycin on the growth, antioxidant system and photosynthesis of Microcystis flos-aquae . The results showed that at 0.001–0.1   μg   L −1 , erythromycin could stimulate the growth of M. flos-aquae and increase its photosynthetic activity; however, it did not significantly increase the activities of superoxide dismutase (SOD) and catalase (CAT) or the levels of malondialdehyde (MDA) and reactive oxygen species (ROS). In contrast, the growth of M. flos-aquae was significantly inhibited ( p   <   0.01) at high levels of erythromycin, reaching an inhibition rate of 81.6% at 40   μg   L −1 erythromycin. At the same time, the activities of SOD and CAT along with MDA content also increased significantly ( p   <   0.01), indicating that the high concentrations of erythromycin caused a severe oxidative stress on algae. However, the balance between oxidants and antioxidant enzymes were disrupted because ROS content simultaneously increased. In addition, the fluorescence parameters of M. flos-aquae decreased significantly with both exposure time and increasing concentration of erythromycin, indicating that photosynthesis was inhibited.

An electrochemical daunorubicin sensor based on the use of platinum nanoparticles loaded onto a nanocomposite prepared from nitrogen decorated reduced graphene oxide and single-walled carbon nanotubes

Kong Fen-Ying,Li Rong-Fang,Yao Lei,Wang Zhong-Xia,Lv Wei-Xin,Wang Wei

A glassy carbon electrode (GCE) was modified with a nanocomposite prepared from nitrogen-doped reduced graphene oxide (N-rGO) and single walled carbon nanotubes (SWCNTs), and then loaded with platinum nanoparticles (Pt NPs) to obtain a voltammetric sensor for daunorubicin (DNR). Reductive doping of GO and the crystallization of the Pt NPs were carried out in a one-step hydrothermal process. The modified electrode was characterized by cyclic voltammetry and differential pulse voltammetry. It exhibited high sensitivity compared with unmodified electrode. Some experimental parameters which affected sensor response were optimized. Under optimum conditions and at a working voltage of typically −0.56 V (vs. Ag/AgCl), the sensor has a low detection limit (3 ng mL −1 ), a wide linear range (0.01–6 μg mL −1 ) and good long-term stability. The method was successfully applied to the sensitive and rapid determination of DNR in spiked human serum samples. Platinum nanoparticles were loaded onto a nanocomposite prepared from nitrogen decorated reduced graphene oxide and single-walled carbon nanotubes (N-rGO-SWCNTs-Pt) and then used for electrochemical determination of daunorubicin (DNR).

Toxic effects of chlorantraniliprole on zebrafish (Danio rerio) at different developmental stages under antibiotic pressure

Xinlei Shi,Yimu Wei,Jingna Cui,Xueke Liu,Fanrong Zhao,Li Zheng,Peng Wang,Donghui Liu

Pesticides and antibiotics have been frequently reported in the environment, but it remains unclear whether antibiotics affect the toxicity of pesticides to aquatic organisms. In this study, the acute, developmental and reproductive toxicity effects of the pesticide chlorantraniliprole on zebrafish at different developmental stages under pressure of ciprofloxacin and erythromycin at environmental concentration were explored. Chlorantraniliprole, ciprofloxacin, and erythromycin are all low toxic to zebrafish (LC 50  > 100 mg/L), and environmental concentrations of antibiotics have no effect on the acute toxicity of chlorantraniliprole to zebrafish. However, ciprofloxacin and erythromycin at environmental concentrations exacerbate the early zebrafish embryo development toxicity of chlorantraniliprole, including abnormal heart and yolk sac development and oxidative stress damage. Additionally, co-exposure of chlorantraniliprole and ciprofloxacin or erythromycin can cause reproductive toxicity in adult zebrafish, resulting in a decrease in offspring quantity and hatching rate, as well as an increase in malformation rate. Greater disruption in the levels of reproductive hormones was observed, suggesting that co-exposure may lead to reproductive risks via endocrine disruptions. Therefore, the increased risks of pesticides to aquatic ecosystems due to the co-existed compounds should be taken into accounts.

Simultaneous detection of three amphenicol antibiotics in shrimp and surface water samples by LC–MS/MS using two-antibodies-immobilized immunoaffinity clean-up technique

Enhui Guo,Linchuan Zhao,Kang Wu,Wen Huang,Kang Zhao,Jianguo Li,Anping Deng

Amphenicol antibiotics including chloramphenicol (CAP), florfenicol (FF) and thiamphenicol (TAP) have been widely used in aquaculture and veterinary to treat various infections. Because of their high toxicity, it is important to detect these amphenicol antibiotics simultaneously. Using the monoclonal antibody against CAP (mAbCAP) binding CAP only and the mAbFF recognizing both FF and TAP, the two antibodies were covalently coupled to CNBr-activated Sepharose-4B for the simultaneous extraction of CAP, FF and TAP from samples. The bound CAP, FF and TAP were eluted from the Sepharose-4B and measured by LC–MS/MS. The extraction conditions of the two-antibodies-immobilized immunoaffinity clean-up (IAC) for CAP, FF and TAP were optimized and the maximum capacity was investigated. The extraction efficiency of IAC for CAP, FF and TAP was 1.1–2.1 times higher than that of SPE. The LC–MS/MS coupled with IAC was a powerful analytical method for the detection of CAP, FF and TAP simultaneously.

Hydrothermal treatment of erythromycin fermentation residue: Harmless performance and bioresource properties

Chen Cai,Yu Hua,Huiping Li,Lei Li,Lingling Dai,Huiling Liu,Xiaohu Dai

Erythromycin fermentation residue (EFR) is a byproduct of bio-fermentation process for producing erythromycin, representing both an economic and environmental burden for the pharmaceutical industry. While hydrothermal treatment (HT) can solve the pollution and toxic problems, the impacts of HT on harmless performance and the bioresource properties of the complex EFR matrix remain unclear. In this study, the abatement of residual erythromycin, its associated antibiotic resistance genes and antimicrobial activity were investigated to evaluate the harmless performance of HT for EFR, and the feasibility of recycling hydrolysates as bioenergy was also assessed. The results showed that raising HT temperature significantly improved the harmless performance. The removal rate of residual erythromycin in EFR reached over 88% and the total absolute abundance of erythromycin resistance genes decreased by more than 3 logs with HT at 160°C for 60 min, but the hydrolysates still existed specific antimicrobial activity against S. aureus. . Additionally, the HT temperature remarkably influenced the bioresource properties of EFR-hydrolyzed products. The release of the protein-like polymers into the liquid phase was enhanced, while high HT temperature worsened the nutrient substances. The elevated drying property of solid products from EFR was observed along with the higher heating value of around 20 MJ/kg, indicating a potential of recycling solid products as biofuel. Total energy production from biogas conversion and biofuel incineration treating 1.0 kg raw EFR was 1875 kJ. Hydrothermal technology was proved to realize the harmless treatment of EFR and meanwhile to be promising for recycling EFR as bioenergy.

Roxithromycin and rhEGF Co-loaded Reactive Oxygen Species Responsive Nanoparticles for Accelerating Wound Healing

Ding Jun,Chen Dan,Hu Jun,Zhang Dinglin,Gou Yajun,Wu Yaguang

Background: Bacterial infection can delay wound healing and is therefore a major threat to public health. Although various strategies have been developed to treat bacterial infections, antibiotics remain the best option to combat infections. The inclusion of growth factors in the treatment approach can also accelerate wound healing. The co-delivery of antibiotics and growth factors for the combined treatment of wounds needs further investigation. Objective: Here we aimed to develop antibiotic and growth factor co-loaded nanoparticles (NPs) to treat Staphylococcus aureus-infected wounds. Methods: By using our previously prepared reactive oxygen species-responsive material (Oxi-αCD), roxithromycin (ROX)-loaded NPs (ROX/Oxi-αCD NPs) and recombinant human epidermal growth factor (rhEGF)/ROX co-loaded NPs (rhEGF/ROX/Oxi-αCD NPs) were successfully fabricated. The in vivo efficacy of this prepared nanomedicine was evaluated in mice with S. aureus-infected wounds. Results: ROX/Oxi-αCD NPs and rhEGF/ROX/Oxi-αCD NPs had a spherical structure and their particle sizes were 164 ± 5 nm and 190 ± 8 nm, respectively. The in vitro antibacterial experiments showed that ROX/Oxi-αCD NPs had a lower minimum inhibitory concentration than ROX. The in vivo animal experiments demonstrated that rhEGF/ROX/Oxi-αCD NPs could significantly accelerate the healing of S. aureus-infected wounds as compared to the free ROX drug and ROX/Oxi-αCD NPs (P < 0.05).Conclusion: ROX and rhEGF co-loaded NPs can effectively eliminate bacteria in wounds and accelerate wound healing. Our present work could provide a new strategy to combat bacteria-infected wounds.

Impact of application of heat-activated persulfate oxidation treated erythromycin fermentation residue as a soil amendment: Soil chemical properties and antibiotic resistance

Yanxiang Zhang,Huiling Liu,Xiaohu Dai,Chen Cai,Jing Wang,Mengmeng Wang,Yunpeng Shen,Peng Wang

Erythromycin fermentation residue (EFR) is the precipitation of fermentative biowaste used for extracting erythromycin (ERY) and may be disposed via land application after heat-activated persulfate (PS) oxidation treatment. However, the effects of the treated EFR as a soil amendment on soil chemical properties and the potential resistance risks caused by introduced ERY remain unclear. Here, a laboratory soil incubation experiment was performed to investigate the soil pH, salinity, introduced antibiotics, antibiotic resistance genes (ARGs) and mobile genetic elements (MGEs), as well as bacterial community structure in the treated EFR-amended soil. The results indicated that pH in treated EFR-amended soil decreased firstly and then increased. The salinity of soil increased but soil was still non-saline soil. In addition, the introduced ERY in the treated EFR-amended soil decreased with the half-life of 12.3 d. Moreover, the relative abundances of ERY resistance genes and MGEs in the treated EFR-amended soil were much lower than those in the control at the end of incubation. Bacterial community structure in the treated EFR-amended soil converged to similar structure in control soil after 49 d incubation. Our results showed that heat-activated PS oxidation treatment of EFR prior to application to soil might be in favor of limiting the spread of ERY resistance genes and MGEs.

Structure-activity relationship and mechanism of flavonoids on the inhibitory activity of P-glycoprotein (P-gp)-mediated transport of rhodamine123 and daunorubicin in P-gp overexpressed human mouth epidermal carcinoma (KB/MDR) cells

Yajing Fang,Fuqiang Liang,Mengmeng Xia,Weiwei Cao,Siyi Pan,Ting Wu,Xiaoyun Xu

This study was aimed to investigate the inhibitory activity of flavonoids on P-glycoprotein (P-gp). Effects of 39 flavonoids on the cellular uptake (CU) of rhodamine123 (Rho) and daunomycin (DNR) were investigated in both parental KB and P-gp overexpressed KB/MDR cells. The inhibition mechanism of selected flavonoids was further investigated by measuring the ATPase activity and expression level of P-gp. Twelve flavonoids improved the uptake of Rho (↑Rho F ) and nineteen flavonoids increased the uptake of DNR (↑DNR F ) in KB/MDR cells with nine flavonoids overlapped. Structure-activity relationship (SAR) indicated that 8-OCH 3 , and 2′-OH have a negative effect on Rho and DNR transport. Whereas 5-OH, 5-OCH 3 , 6-OH, 7-OCH 3 , 3′-OH, and 4′-OH, are essential for inhibition of flavonoids on P-gp and reversing the resistance of Rho and DNR. Eleven selected flavonoids significantly induced the basal P-gp-ATPase activity but much lower than that induced by verapamil . Tangeretin , galangin , kaempferol , quercetin , and morin significantly reversed the ATPase activity stimulated by verapamil. Six of eleven flavonoids significantly decreased P-gp expression, whereas three flavonoids slightly increased P-gp expression. These results provide valuable information that flavonoids can effectively reverse multidrug resistance of P-gp-mediated transport of nutraceutical and drugs by co-administration.

Engineering the Erythromycin-Producing Strain Saccharopolyspora erythraea HOE107 for the Heterologous Production of Polyketide Antibiotics

Lü Jin,Long Qingshan,Zhao Zhilong,Chen Lu,He Weijun,Hong Jiali,Liu Kai,Wang Yemin,Pang Xiuhua,Deng Zixin,Tao Meifeng

Bacteria of the genus Saccharopolyspora produce important polyketide antibiotics, including erythromycin A (Sac. erythraea) and spinosad (Sac. spinosa). We herein report the development of an industrial erythromycin-producing strain, Sac. erythraea HOE107, into a host for the heterologous expression of polyketide biosynthetic gene clusters (BGCs) from other Saccharopolyspora species and related actinomycetes. To facilitate the integration of natural product BGCs and auxiliary genes beneficial for the production of natural products, the erythromycin polyketide synthase (ery) genes were replaced with two bacterial attB genomic integration sites associated with bacteriophages ϕC31 and ϕBT1. We also established a highly efficient conjugation protocol for the introduction of large bacterial artificial chromosome (BAC) clones into Sac. erythraea strains. Based on this optimized protocol, an arrayed BAC library was effectively transferred into Sac. erythraea. The large spinosad gene cluster from Sac. spinosa and the actinorhodin gene cluster from Streptomyces coelicolor were successfully expressed in the ery deletion mutant. Deletion of the endogenous giant polyketide synthase genes pkeA1-pkeA4, the product of which is not known, and the flaviolin gene cluster (rpp) from the bacterium increased the heterologous production of spinosad and actinorhodin. Furthermore, integration of pJTU6728 carrying additional beneficial genes dramatically improved the yield of actinorhodin in the engineered Sac. erythraea strains. Our study demonstrated that the engineered Sac. erythraea strains SLQ185, LJ161, and LJ162 are good hosts for the expression of heterologous antibiotics and should aid in expression-based genome-mining approaches for the discovery of new and cryptic antibiotics from Streptomyces and rare actinomycetes.
