Highly sensitive fluorescent sensing for intracellular glutathione based on Ti3C2 quantum dots

Xu Xiumei,Zhang Haijiao,Diao Quan,Zhu Yongsheng,Yang Gang,Ma Benyuan

Intracellular glutathione (GSH) sensing was important for monitoring many diseases and health of cell. In this study, we prepared surface-functionalized Ti 3 C 2 quantum dots (QDs) by ultrasonic cutting and hydrothermal method. Bright excitation wavelength-dependent blue emission was observed in Ti 3 C 2 QDs which was attributed to surface defects and quantum confinement effects. Furthermore, we developed a novel fluorescence sensor that can detect GSH with high sensitivity, based on the fluorescence resonance energy transfer process from Ti 3 C 2 quantum dots to GSH, with a detection limit of 0.02 μM approximately. This novel Ti 3 C 2 QD can be used as a promising fluorescent sensor for practical applications.

Glutathione stabilized green-emission gold nanoclusters for selective detection of cobalt ion

Rui-Xian Zhao,An-Yong Liu,Qiu-Lin Wen,Bi-Chao Wu,Jun Wang,Yi-Lin Hu,Zheng-Fen Pu,Jian Ling,Qiue Cao

A glutathione stabilized Au nanoclusters (GSH-Au NCs) was synthesized here and used to selective detection of cobalt ion. The as-prepared GSH-Au NCs had strong green light emission around 500 nm, and the features of the NCs have been systematically characterized by UV–vis absorption, X-ray photoelectronic spectroscopic, Fourier transform infrared spectroscopy and transmission electron microscope characterization. The interactions between the GSH-Au NCs and metal ions was studied, and the results indicated that the fluorescence of the GSH-Au NCs could be quenched in the presence of Co 2+ ion at pH of 6.0. The quenching ratio was linear with the concentration of Co 2+ ions, and the calibration curve was I 0 /I  = 0.1187c co  + 0.6085 in the Co 2+ concentration ranges from 2.0 to 50.0 μM with correlation coefficient ( R 2 ) of 0.9950 and the limit of detection (LOD, 3 σ ) of 0.124 μM. In addition, we collected environmental water samples to test the reliability of the method and demonstrated this method is simple, rapid, and selective.

A sensitive fluorescent probe for imaging biothiol in zebrafish

Jing Cao,Xiaoxue Jiang,Nanyan Fu

Due to their highly significant biological activities, in vivo monitoring biothiols, including glutathione (GSH), cysteine (Cys), and homocysteine (Hcy), draws great attention. Among them, GSH is the representative nonprotein thiol inside the cell with the high abundance, protecting cells from oxidative stress and toxins. In this work, we developed a fluorescent probe NI for sensitive mapping endogenous biothiol distribution in zebrafish. We used 1,8-naphthalimide scaffold as easy modifiable fluorophore and 2-thiophenecarbonxyl as recognition moiety. After the cleavage of ester bond by GSH addition, NI exhibited strong emission with large Stokes shift (110 nm) and its LOD is 0.120 μM. We also studied the difference of kinetic response of GSH over Cys and Hcy. Finally, the described probe was successfully applied in sensing endogenous biothiol in zebrafish, showing its potential utility in vivo imaging.

Modulation of (Homo)Glutathione Metabolism and H2O2 Accumulation during Soybean Cyst Nematode Infections in Susceptible and Resistant Soybean Cultivars

Xi Chen,Shuang Li,Xuebing Zhao,Xiaofeng Zhu,Yuanyuan Wang,Yuanhu Xuan,Xiaoyu Liu,Haiyan Fan,Lijie Chen,Yuxi Duan

In plant immune responses, reactive oxygen species (ROS) act as signaling molecules that activate defense pathways against pathogens, especially following resistance (R) gene-mediated pathogen recognition. Glutathione (GSH), an antioxidant and redox regulator, participates in the removal of hydrogen peroxide (H2O2). However, the mechanism of GSH-mediated H2O2generation in soybeans (Glycine max(L.) Merr.) that are resistant to the soybean cyst nematode (SCN;Heterodera glycinesIchinohe) remains unclear. To elucidate this underlying relationship, the feeding of race 3 ofH. glycineswith resistant cultivars, Peking and PI88788, was compared with that on a susceptible soybean cultivar, Williams 82. After 5, 10, and 15 days of SCN infection, we quantified γ-glutamylcysteine (γ-EC) and (homo)glutathione ((h)GSH), and a gene expression analysis showed that GSH metabolism in resistant cultivars differed from that in susceptible soybean roots. ROS accumulation was examined both in resistant and susceptible roots upon SCN infection. The time of intense ROS generation was related to the differences of resistance mechanisms in Peking and PI88788. ROS accumulation that was caused by the (h)GSH depletion-arrested nematode development in susceptible Williams 82. These results suggest that (h)GSH metabolism in resistant soybeans plays a key role in the regulation of ROS-generated signals, leading to resistance against nematodes. Keywords:soybean;glutathione;reactive oxygen species;soybean cyst nematode;resistance

Glutathione-stabilized Cu nanocluster-based fluorescent probe for sensitive and selective detection of Hg2+ in water

Tingting Luo,Shiting Zhang,Yujue Wang,Meina Wang,Mei Liao,Xingming Kou

In this paper, an innovative and facile one-pot method for synthesizing water-soluble and stable fluorescent Cu nanoclusters (CuNCs), in which glutathione (GSH) served as protecting ligand and ascorbic acid (AA) as reducing agent was reported. The resultant CuNCs emitted blue-green fluorescence at 440 nm, with a quantum yield (QD) of about 3.08%. In addition, the prepared CuNCs exhibited excellent properties such as good water solubility, photostability and high stability toward high ionic strength. On the basis of the selective quenching of Hg 2+ on CuNCs fluorescence, which may be the result of Hg 2+ ion-induced aggregation of the CuNCs, the CuNCs was used for the selective and sensitive determination of Hg 2+ in aqueous solution. The proposed analytical strategy permitted detection of Hg 2+ in a linear range of 4 × 10 −8 to 6 × 10 −5 M, with a detection limit of 2.2 × 10 −8 M. Eventually, the practicability of this sensing approach was confirmed by its successful application to assay Hg 2+ in tap water, Lotus lake water and river water samples with the quantitative spike recoveries ranging from 96.9% to 105.4%.

Copper nanocluster-based fluorescent probe for sensitive and selective detection of Hg2+ in water and food stuff

Xue Hu,Wei Wang,Yuming Huang

In this study, Hg 2+ ions were found to quench the fluorescence of glutathione (GSH)-capped copper clusters (Cu NCs). The Cu NCs were prepared by a simple reduction of CuSO 4 in the presence of GSH serving both as a reducing and protecting agents, and characterized by ultraviolet–visible absorption spectroscopy (UV–vis), high resolution scanning electron microscopy (HRTEM), Fourier transform infrared spectroscopy (FT-IR), and X-ray photoelectron spectrometer (XPS). The GSH-Cu NCs displayed a small size, excellent water-dispersibility, good storage stability, good photostability and were stable in the presence of high concentrations of salt. The GSH-Cu NCs possessed strong blue fluorescence with a quantum yield of 10.6% and exhibited an excitation-independent fluorescence behavior. The zeta potential , TEM, resonance light scattering and dynamic light scattering measurements demonstrated that the Hg 2+ ion-induced aggregation of the Cu NCs contributed to the fluorescence quenching of the dispersed Cu NCs. On these findings, a sensitive and selective fluorescent probe was developed for detecting Hg 2+ in the linear range from 10   nM to 10   μM with a detection limit of 3.3   nM (S/N=3). The proposed method has been successfully applied to determine Hg 2+ content in water sample and food stuff. The results of the proposed method were in good agreement with those obtained by a hydride generation atomic fluorescence spectrometry (HG-AFS).

A “turn-on” fluorescent probe used for the specific recognition of intracellular GSH and its application in bioimaging

Lanfang Pang,Yanmei Zhou,Enze Wang,Fang Yu,Hua Zhou,Wenli Gao

We have designed and synthesized a carbazole-based fluorescent probe (CZ-Nm) for the specific recognition of GSH over Cys and Hcy. The probe is almost non-fluorescent owing to the photoinduced electron transfer (PET) process from the carbazole fluorophore to the nitroolefin moiety. Upon treatment with GSH, owing to the Michael addition of GSH to the double bond of the nitroolefin moiety, the probe shows a fluorescence enhancement and absorption change. The CZ-Nm probe displays desirable properties such as acting as a “naked eye” probe, a wide linear range of 0–0.01 M, high sensitivity, and strong anti-jamming capability. More importantly, the probe can also be successfully applied to the detection of intracellular GSH with a bright fluorescence signal, good cell permeability and bio-compatibility.

An ESIPT-based fluorescent probe for highly selective detection of glutathione in aqueous solution and living cells

Xintong Ren,Fang Wang,Jing Lv,Tingwen Wei,Wei Zhang,Yong Wang,Xiaoqiang Chen

In this paper, highly sensitive and selective fluorescent probe 1 for GSH is designed and synthesized based on modulation of the excited-state intramolecular proton transfer (ESIPT) process of 2-(2′-hydroxy-3′-ethoxyphenyl)benzothiazole. Upon introducing GSH in neutral solution containing CTAB micelles , dinitrophenyl, the protecting group of the probe, is removed via the nucleophilic substitution , thereby retrieving the ESIPT process of 2-(2′-hydroxy-3′-ethoxyphenyl)benzothiazole, which results in a fluorescence enhancement at 485 nm. Additionally, the fluorescence intensity of the probe is linearly proportional to GSH concentration ranging from 0 to 100 μM and the obtained detection limit is as low as 0.81 μM. The enhanced selectivity toward GSH over Cys and Hcy is also attributed to one more carboxyl group in GSH. Importantly, the probe is successfully utilized for monitoring GSH in living HeLa cells .

Glutaminase-1 Mediated Glutaminolysis to Glutathione Synthesis Maintains Redox Homeostasis and Modulates Ferroptosis Sensitivity in Cancer Cells

Changsen Bai,Jialei Hua,Donghua Meng,Yue Xu,Benfu Zhong,Miao Liu,Zhaosong Wang,Wei Zhou,Liming Liu,Hailong Wang,Yang Liu,Lifang Li,Xiuju Chen,Yueguo Li

Glutaminase-1 (GLS1) has garnered considerable interest as a metabolic target in cancer due to its heightened involvement and activity. However, the precise fate of glutaminolysis catalysed by GLS1 in cancer cells remains elusive. We found that GLS1 knockout led to significant suppression of cancer cell proliferation, which can be reversed or partially restored by supplementation of glutamate or non-essential amino acids that can be converted into glutamate. The addition of spliceosomal KGA or GAC ameliorates cancer cell growth in vitro and in vivo, providing both simultaneously completely reverse the effect. The primary metabolic fate of glutamate produced through glutaminolysis in cancer cells is mainly used to produce glutathione (GSH) for redox homeostasis, not entering the tricarboxylic acid cycle or synthesising nucleotides. GSH monoethyl ester (GSH-MEE) effectively rescues the inhibition of cancer cell proliferation caused by GLS1 knockout. Deletion of GLS1 results in an elevation of reactive oxygen species (ROS) and malondialdehyde (MDA), a reduction of NADPH/NADP + ratio, and an augmented susceptibility of cells to ferroptosis. Glutathione Peroxidase 4 (GPX4) and GPX1 exhibit complementary roles in redox regulation, with GLS1 knockout promoting GPX4 degradation. Pharmacological inhibition of GLS1 synergises with GPX4 inhibitor to suppress tumour growth. Dual targeting of GPX4 and GPX1 presents a potent anti-cancer strategy. This metabolic mechanism facilitates a deeper comprehension of the abnormal glutamine metabolism in cancer cells, establishing a theoretical basis for the potential clinical utilisation of GLS1 inhibitors and presenting novel perspectives for advancing combinatorial therapeutic approaches.

Cobalt oxyhydroxide nanoflakes integrating with 7-amino-4-methylcoumarin for sensitive detection of glutathione

Peigang Zhang,Yunhui Fu,Tianyi Zheng,Huihui Ge,Martin M.F. Choi,Li Zhang,Wei Bian

Glutathione (GSH), an essential biothiol, plays a critical role in reversible redox reactions, cellular signaling, detoxification, and metabolism. Fluctuations in GSH levels are closely linked to serious health conditions, underscoring the significance of monitoring its concentration. Fluorescence-based sensing with nanomaterials offers a promising strategy for rapid GSH detection. In this work, we developed a fluorescent probe integrating 7-amino-4-methylcoumarin (AMC) and cobalt oxyhydroxide nanoflakes (CoOOH NFs) for GSH detection through a combined internal-filter effect and static quenching mechanism. The introduction of GSH triggered a redox reaction that degraded CoOOH NFs, suppressing their quenching effect and restoring AMC fluorescence, which enabled GSH quantification through fluorescence recovery. The probe detected GSH in the range of 20–200 μM with a detection limit of 0.13 μM. It was successfully applied to determine GSH in human serum, and the results showed excellent agreement with those measured by a commercial GSH detection kit based on the use of Ellman's reagent.

Carbon quantum dots-based fluorescent probe for sensitive and selective detection of glutathione in bacteria and cells

Lina Wu,Huangpei Xiao,Jiaxin Xue,Xiaoyun Lin,Zhenmao Zhu,Ailin Liu,Liqing Lin

Background The sensitivity of 5,5′-dithiobis-(2-nitrobenzoic acid) (DTNB) method may not be enough to detect low concentration glutathione (GSH), especially in complex samples, which limits its application in some biologic scenarios. Fluorescent carbon quantum dots (CQDs) have garnered significant interest in bioanalytical applications due to their unique properties, particularly the sensitivity in the detection of complex samples. Results In this work, carbon quantum dots (TOCDs) were prepared via pyrolysis method of d -threonine and o -phenylenediamine. Considering advantages of selectivity of DTNB and sensitivity of TOCDs, a simple, fast and sensitive fluorescent sensor based on TOCDs and DTNB was fabricated for selective detection of GSH. In the presence of GSH in the mixture of TOCDs and DTNB, –SH of GSH can interact with DTNB and subsequently affording 5-thio-2-nitrobenzoate (TNB), which can strongly absorb excitation light of TOCDs and lead to fluorescence quenching of TOCDs. As a consequence, quantitative screening of GSH can be achieved through the fluorescence intensity of TOCDs was decreased after the addition of GSH in the system of TOCDs and DTNB. Under optimized conditions, good linear relationships in the range of 2.5–40 nM, 40–150 nM and 150–1000 nM for GSH. TOCDs can effectively detect GSH with a limit of detection (LOD) 1.5408 nM. The feasibility of the TOCDs probe was verified through non-invasive detection of reduced glutathione (GSH) in bacteria and cells. The fluorescence of TOCDs in different living bacteria/cells were yellow fluorescence, while the fluorescence could be ignored after joining GSH and DTNB. The mechanism about quench phenomenon of the fluorescence intensity between TOCDs and DTNB/GSH was the inner-filter effect. These results demonstrated that the presented fluorescent sensor could be a promising sensing platform for GSH analysis. Significance This sensing strategy could be expanded to more access to the GSH-induced acute toxicity and disease. The yellow fluorescence carbon quantum dots demonstrated its potential application on GSH detection in bacteria and cells.

The effect of glutathione metabolism on the release of Wound-Induced Spores from Pyropia yezoensis

Haihong Chen,Ziyan Shi,Zhihai Zhong,Nanjing Ji,Xiaoqian Zhou,Zhijie Dan,Xin Shen

Oxidative stress from mechanical injury can trigger the development of Wound-Induced Spores (WIS) in Pyropia yezoensis , yet the regulatory mechanisms underlying this process require further elucidation. In this study, comparative transcriptomic analysis between injured and uninjured P. yezoensis thalli revealed significant differential gene expression within the glutathione (GSH) metabolism pathway during WIS development. There was a down-regulation in the expression of genes encoding for γ-glutamyl-cystein synthetase (GCL) and glutathione synthetase (GSS), which were pivotal for GSH biosynthesis, during the development of WIS. Moreover, a diminished expression level was also observed in genes associated with oxidative metabolism of GSH during this process, including those encoding glutathione S-transferase (GST), glutathione peroxidase (GPX), glutathione reductase (GR), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), and dihydroascorbate reductase (DHAR). However, this transcriptional regulation does not correspond to a significant decrease in intracellular GSH levels, which may be associated with a reduction in the oxidative metabolism of GSH, leading to decreased cellular utilization of GSH. Treatment injured P. yezoensis with Buthionine sulfoximine (BSO) promoted the release probability of WIS, the release efficiency of WIS and germination rate of asexual spores, while supplementation of GSH produced opposite results, suggesting that modulating GSH levels can regulate the release of WIS. Correlation analysis suggested that there was a significant positive correlation between the release of WIS and the germination rate of asexual spores, suggesting that promoting the release of WIS could lead to the acquisition of a greater number of asexual spore seedlings. Our findings underscore the key role of GSH metabolism in WIS regulation and provide a theoretical foundation for the subsequent development of strategies to obtain asexual spore seedlings.
