A bimodal type of AgPd Plasmonic Blackbody Nanozyme with boosted catalytic efficacy and synergized photothermal therapy for efficacious tumor treatment in the second biological window

Jia Tao,Li Dan,Du Jiarui,Fang Xikui,Gerasimov Valeriy,Ågren Hans,Chen Guanying

Nanozymes are promising for precise cancer treatment, but are typically limited in terms of the low catalytic efficiency and the complexity in tumor microenvironment (TME). Herein, we describe a bimodal type of AgPd plasmonic blackbody (AgPd PB) nanozyme of compact sizes (< 30 nm), which presents not only boosted enzyme efficacy but also efficient photothermal therapy (PTT) for synergized therapy through tissue-penetrating light in the second biological window (1000–1700 nm). The synthesized hyperbranched AgPd PB nanozymes possess intense and broadband localized surface plasmonic resonance absorption of 400–1300 nm, entailing prominent photothermal efficiency (η = 45.1% at 1064 nm) for PTT. Importantly, PTT was found to significantly boost the nanozyme efficacy of both catalase (CAT) and peroxidase (POD) processes, which correspondingly decompose H2O2 to into O2 to relieve tumor hypoxia, and activate H2O2 to generate oxidative •OH radical. While the generated •OH was found to be able to minimize heat shock proteins (HSPs), which plays a vital role to counterbalance PTT effect both in vitro and in vivo. As compared to control ground without treatment, the synergized nanozyme and PTT activities resulted in about 7-fold reduction of tumor volume, thus elevating the survival rate from 0 to 80% at 30 days posttreatment. Besides the synergistic therapy, the AgPd PB nanozyme were shown to own fluorescence, computed tomography (CT), and photoacoustic (PA) imaging abilities, thus having implications for uses in imaging-guided precise cancer therapy. This study provides a paradigm of TME responsive theranostics under NIR-II light irradiation.Graphical Abstract

Biodegradable copper telluride nanosheets for redox-homeostasis breaking-assisted chemodynamic cancer therapy boosted by mild-photothermal effect

Guangzhi Zhou,Ming Li

Chemodynamic therapy (CDT) holds great promise for cancer treatment through employing the Fenton or Fenton-like effects for highly toxic ·OH production exclusively in the tumor microenvironment. Herein, glutathione (GSH)-degradable Cu 2-x Te nanosheets modified with polyethylene glycol (bioCu 2-x Te NSs) as a multifunctional CDT agent are reported for effective cancer treatment via mild-photothermally enhanced CDT. The resulting bioCu 2-x Te NSs exhibit excellent near-infrared II (NIR-II) plasmonic absorption and photothermal properties. Upon endocytosis by cancer cells, bioCu 2-x Te NSs are degraded into Cu + and elemental Te by GSH overexpressed in cancer cells. The generated Cu + and Te together exhibit efficient catalytic decomposition of endogenous H 2 O 2 into ·OH with effective GSH depletion and redox homeostasis breaking of cancer cells. The mild-photothermal effect from the NIR-II irradiation is confirmed to promote intracellular ·OH production and tumor accumulation of bioCu 2-x Te NSs. Owing to all these prominent features, our results on both in vitro and in vivo studies verify the remarkable therapeutic efficacy via mild-photothermally enhanced CDT on a MCF-7 tumor mouse model with intravenous administration of bioCu 2-x Te NSs under NIR-II irradiation. This study demonstrates the potential of bioCu 2-x Te NSs as a potent CDT agent, which features GSH-triggered degradability and excellent therapeutic functions, for efficient cancer treatment.

Single Atom-Engineered NIR-II Gold Clusters with Ultrahigh Brightness and Stability for Acute Kidney Injury

You Huang,Ke Chen,Ling Liu,Huizhen Ma,Xiaoning Zhang,Kexin Tan,Yuan Li,Ying Liu,Changlong Liu,Hao Wang,Xiao-Dong Zhang

Near-infrared-II (NIR-II) imaging has shown great potential for monitoring the pathological progression and deep tissue imaging but is limited to present unmet NIR-II agent. Present fluorophores show a promising prospect for NIR-II imaging, but brightness and photostability are still highly challenging during real-time monitoring. In this work, atom-engineered NIR-II Au 24 Cd 1 clusters with ultrahigh brightness, stability, and photostability are developed via single atomic Cd doping. Single atom Cd substitutions contribute to Cd 4d state in HOMO and redistribution of energy level near the gap, exhibiting 56-fold fluorescence enhancement of Au 24 Cd 1 clusters. Meanwhile, single atomic Cd reinforces Cd  Au bond energy, formation energy, and stabilized cluster structure, leading to persistent stability for up to 1 month without decay, as well as excellent photostability of 1 h without photobleaching, much longer than clinically approved indocyanine green (<5 min). In vivo imaging shows gold clusters can monitor acute kidney injury (AKI) even after 72 h of injury, enabling evaluating progression at a very long window. Meanwhile, the bioactive gold clusters can alleviate AKI-induced oxidative stress damage and acute neuroinflammation. Single atom-engineered gold clusters exhibit molecular tracking and diagnostic prospect in kidney-related diseases.

NIR-II Fluorescent Thermophoretic Nanomotors for Superficial Tumor Photothermal Therapy

Jiwei Jiang,Jing Hu,Mingtong Li,Mingzhi Luo,Bin Dong,Metin Sitti,Xiaohui Yan

Peritumoral subcutaneous injection has been highly envisioned as an efficient yet low-risk administration of photothermal agents for superficial tumor photothermal therapy. However, obstructed by complex subcutaneous tissue, the delivery of injected photothermal agents to the specific tumor remains a critical issue. Herein, the study reports a polydopamine (PDA)-encapsulated spherical core/shell nanomotor with fluorescent indocyanine green (ICG) immobilized on its PDA shell. Upon the first near-infrared (NIR-I) irradiation, this motor can generate favorable photothermal heat, and meantime, emit a robust ICG fluorescence in the second near-infrared window (NIR-II). The heat turns the motor into an active photothermal agent able to perform thermophoretic propulsion along the irradiation direction in subcutaneous tissue, while the ICG fluorescence can direct the subcutaneous propulsion of motors toward specific tumor through real-time NIR-II imaging. These functions endow the motor with the ability of moving to tumor after being injected at peritumoral site, enabling an enhanced photothermal therapy (PTT). The results demonstrated herein suggest an integrated nanorobotic tool for the superficial PTT using peritumoral administration, highlighting an NIR-II imaging-directed subcutaneous propulsion.

Extracellular matrix-degrading STING nanoagonists for mild NIR-II photothermal-augmented chemodynamic-immunotherapy

Zhan Meixiao,Yu Xiangrong,Zhao Wei,Peng Yongjun,Peng Shaojun,Li Jingchao,Lu Ligong

Regulation of stimulator of interferon genes (STING) pathway using agonists can boost antitumor immunity for cancer treatment, while the rapid plasma clearance, limited membrane permeability, and inefficient cytosolic transport of STING agonists greatly compromise their therapeutic efficacy. In this study, we describe an extracellular matrix (ECM)-degrading nanoagonist (dNAc) with second near-infrared (NIR-II) light controlled activation of intracellular STING pathway for mild photothermal-augmented chemodynamic-immunotherapy of breast cancer. The dNAc consists of a thermal-responsive liposome inside loading with ferrous sulfide (FeS2) nanoparticles as both NIR-II photothermal converters and Fenton catalysts, 2′3′-cyclic guanosine monophosphate-adenosine monophosphate (cGAMP) as the STING agonist, and an ECM-degrading enzyme (bromelain) on the liposome surface. Mild heat generated by dNAc upon NIR-II photoirradiation improves Fenton reaction efficacy to kill tumor cells and cause immunogenic cell death (ICD). Meanwhile, the generated heat triggers a controlled release of cGAMP from thermal-responsive liposomes to active STING pathway. The mild photothermal activation of STING pathway combined with ICD promotes anti-tumor immune responses, which leads to improved infiltration of effector T cells into tumor tissues after bromelain-mediated ECM degradation. As a result, after treatment with dNAc upon NIR-II photoactivation, both primary and distant tumors in a murine mouse model are inhibited and the liver and lung metastasis are effectively suppressed. This work presents a photoactivatable system for STING pathway and combinational immunotherapy with improved therapeutic outcome.Graphical Abstract

In Vivo Imaging of Mammalian Embryos by NIR-I Photoacoustic Tomography and NIR-II Optical Coherence Tomography Using Gold Nanostars as Multifunctional Contrast Agents

Sheng Zhang,Zhenyang Liu,Linlin Mao,Jian Wu,Di Zhang,Ruiyuan Liu,Li Qi

High quality visualization of live mammalian embryo is an important requirement for studying fetal development. Photoacoustic tomography (PAT) and optical coherence tomography (OCT) are two advanced imaging modalities that have been utilized for embryonic imaging separately. However, high contrast, multiscale, and deep tissue imaging of embryos in vivo remains challenging. Here, we demonstrate multimodal contrast-enhanced imaging of live embryos by gold nanostar-meditated near-infrared I (NIR-I) PAT and near-infrared II (NIR-II) OCT. We investigate intravenous and intravaginal administration of gold nanostars, and significant enhancement of signal, image contrast, and imaging depth are achieved for both PAT and OCT. Our findings may facilitate detailed studies of the morphological and functional changes during embryonic development.

Biotin-decorated hollow gold nanoshell for dual-modal imaging-guided NIR-II photothermal and radiosensitizing therapy toward breast cancer

Yong-Jian Chen,Wei Men,Ming Chen,Lian-Ying Zhang,Ming-Wa Chen,Xiao-Tong Chen,Jian Peng,Nai-Han Huang,Wen-Hua Zhang,Jin-Xiang Chen

Radiotherapy (RT) is dominantly used in breast cancer therapy but is facing fierce side effects because of the limited difference between tumor and normal tissues in response to ionizing radiation. Herein, we construct a core-shell nanoparticle of UiO-66-NH2@AuNS. Then the solid gold shell was etched into hollow AuNS (HAuNS) and further modified with biotin-PEG-SH (PEG-bio) to obtain HAuNS@PEG-bio. HAuNS@PEG-bio demonstrates effective near infrared II (NIR-II) region photothermal therapy (PTT) performance, and the increase of temperature at the tumor site promotes the blood circulation to alleviate the hypoxia in the tumor microenvironment (TME). Meanwhile, HAuNS exhibits strong X-ray absorption and deposition ability due to the high atomic coefficient of elemental Au (Z = 79) and hollowed-out structure. Through the dual radiosensitization of the high atomic coefficient of Au and the hypoxia alleviation from PTT of HAuNS, the breast cancer cells could undergo immunogenic cell death (ICD) to activate the immune response. The outstanding anticancer effects of HAuNS@PEG-bio by cell targeting to exert IR thermal and CT imaging-guided NIR-II photothermal, radiosensitizing therapy, and ICD in vivo. This work highlights that the constructed biotin-decorated hollow gold nanoshell has a promising potential as a diagnostic and treatment integration reagents for the breast cancer.

Hypoxia-Responsive Covalent Organic Framework Nanoplatform for Breast-Cancer-Targeted Cocktail Immunotherapy via Triple Therapeutic Switch Mechanisms

Guoqin Huang,Lianying Zhang,Jiahao Feng,Dan Wu,Libo Wu,Weilun Pan,Yu Jiang,Ming Chen,Jinxiang Chen,Pixian Shui

Covalent organic frameworks (COFs), known for their exceptional in situ encapsulation and precise release capabilities, are emerging as pioneering drug delivery systems. This study introduces a hypoxia-responsive COF designed to encapsulate the chemotherapy drug gambogic acid (GA) in situ. Bimetallic gold-palladium islands were grown on UiO-66-NH 2 (UiO) to form UiO@Au-Pd islands (UAPi), which were encapsulated with GA through COF membrane formation, resulting in a core-shell structure (UAPiGC). Further modification with hyaluronic acid (HA) created UiO@Au-Pd islands @GA-COF@HA (UAPiGCH) for enhanced tumor targeting. In the hypoxic tumor microenvironment, the COF collapses, releasing GA and UAPi, initiating a triple therapeutic response: nanozyme-catalyzed therapy, near-infrared II (NIR-II) mild photothermal therapy (mild-PTT), and chemotherapy. UAPi exhibits catalase (CAT)-like and peroxidase (POD)-like activities, generating oxygen to alleviate hypoxia and reactive oxygen species (ROS) for tumor destruction. GA acts as a chemotherapeutic agent and inhibits heat shock protein 90 (HSP90), enhancing photothermal sensitivity. In vitro and in vivo studies confirm UAPiGCH’s ability to induce pyroptosis, stimulate dendritic cell maturation, and boost T cell infiltration, demonstrating its potential as a precise therapeutic nanoplatform. This strategy integrates multiple therapies into a hypoxia-responsive system, offering promising applications in cancer treatment.

An NIR-II light-responsive nanoplatform for photothermal and enhanced photodynamic synergistic therapy toward drug-resistant bacterial infection and hypoxia relief in subcutaneous wound healing

Wei Meng,Lian-Ying Zhang,Xiao-Tong Chen,Feng-Jun Ge,Peng Huang,Yong-Jian Chen,Ting Huang,Nai-Han Huang,Yi-Ming Zou,Wen-Hua Zhang,Qun Zhang,Jin-Xiang Chen

The drug-resistant bacterial infection has seriously threatened human health and thus it is urgent to develop new and effective drugs to overcome the antibiotic resistance crisis. Herein, a near-infrared-II (NIR-II) region light-responsive Pd rough nanoshells on metal-organic framework (MOF) of UiO-66-NH 2 (UiO-66-NH 2 @Pd shell ) has been developed for the first time, which was further loaded with the photosensitizer IR780 to ultimately yield UiO-66-NH 2 @Pd shell -IR780 (UPI) as the core-shell theranostic nanoplatform. The Pd rough nanoshells demonstrate promising photothermal conversion efficiency, superoxide dismutase (SOD)-like activity, and light-enhanced catalase (CAT)-like activity. In the non-healing wound inflammatory microenvironment with overexpression of reactive oxygen species (ROS) caused by subcutaneous bacterial infection, Pd rough nanoshells in UPI transforms ROS, such as O 2 •− , into H 2 O 2 and efficiently catalyzes the decomposition of the formed and endogenous H 2 O 2 to produce O 2 . The O 2 production process could be enhanced by NIR-II light to facilitate photodynamic efficacy of IR780 to prime rapid clearance of methicillin-resistant Staphylococcus aureus (MRSA) through biofilm inhibition, along with down-regulation of hypoxia inducible factor-1α (HIF-1α) and relief of inflammation, thus promote wound healing. Moreover, UPI exhibits negligible biotoxicity in vitro and in vivo . This work provides a method for preparing promising Pd-based nanomaterials toward subcutaneous infection treatment and wound healing.

A novel near-infrared-II fluorescence probe for serum albumin biosensing and site-binding mechanism study

Jixiang Song,Pengfei Chen,Longsheng Zhang,Junbo Hu,Ruining Yang,Pengfei Sun,Yanqin Huang,Xingfen Liu,Xiaomei Lu,Quli Fan

Abnormal albumin levels are predictors of nephropathy, liver cirrhosis, coronary heart disease, and diabetes. Sensitive and rapid detection of endogenous albumin is of great significance to disease diagnosis and new drug discovery. Here, the first near-infrared-II fluorescence probe for albumin was developed. Based on the remarkable fluorescence enhancement induced by albumin, both human serum albumin (HSA) and bovine serum albumin (BSA) were quantified in several minutes. The limit of detection for albumin in urine samples is as low as 50 nM, which meets the requirement well for clinical analysis. The fluorescence imaging of drug-induced albumin in living cells verified strong capability for the probe to monitor the endogenous albumin. The site-binding mechanism between the probe and albumin was investigated via Job's plots analysis, drug substitution assays, and molecular docking. The NIR-II probe exhibits a specific and high-affinity binding (K b =5.26×10 11 M −1 ) with albumin, which enabled a further determination of trypsin and a protease inhibitor. This novel versatile biosensor provides a promising strategy for on-site evaluating of endogenous albumin for clinical diagnostics and drug screening.

Hyaluronic Acid-Modified Spherical MgO2/Pd Nanocomposites Exhibit Superior Antitumor Effect through Tumor Microenvironment-Responsive Ferroptosis Induction and Photothermal Therapy

Wenhui Xie,Yilin Lu,Yilin Yuan,Lulu Xiao,Jiaqi Liu,Haofeng Song,Rongcheng Niu,Yanli Liu,Juntang Lin

Metal peroxide nanomaterials as efficient hydrogen peroxide (H2O2) self-supplying agents have attracted the attention of researchers for antitumor treatment. However, relying solely on metal peroxides to provide H2O2 is undoubtedly insufficient to achieve optimal antitumor effects. Herein, we construct novel hyaluronic acid (HA)-modified nanocomposites (MgO2/Pd@HA NCs) formed by decorating palladium nanoparticles (Pd NPs) onto the surfaces of a magnesium peroxide (MgO2) nanoflower as a highly effective nanoplatform for the tumor microenvironment (TME)-responsive induction of ferroptosis in tumor cells and tumor photothermal therapy (PTT). MgO2/Pd@HA NC could be well endocytosed into tumor cells with CD44 expression depending on the specific recognition of HA with CD44, and then, the nanocomposites can be rapidly decomposed in mild acid and hyaluronidase overexpressed TME, and plenty of H2O2 was released. Simultaneously, Pd NPs catalyze self-supplied H2O2 to generate abundant hydroxyl radicals (•OH) and catalyze glutathione (GSH) into glutathione disulfide owing to its peroxidase and glutathione oxidase mimic enzyme activities, while the abundant •OH could also consume GSH in tumor cells and disturb the defense pathways of ferroptosis leading to the accumulation of lipid peroxidation and resulting in the occurrence of ferroptosis. Additionally, the superior photothermal conversion performance of Pd NPs in near-infrared II could also be used for PTT, synergistically cooperating with nanocomposite-induced ferroptosis for tumor inhibition. Consequently, the successfully prepared TME-responsive MgO2/Pd@HA NCs exhibited marked antitumor effect without obvious biotoxicity, contributing to thoroughly explore the nanocomposites as a novel and promising treatment for tumor therapy.

A multifunctional antibacterial coating on bone implants for osteosarcoma therapy and enhanced osteointegration

Guannan Zhang,Zhuangzhuang Wu,Yongqiang Yang,Jing Shi,Jia Lv,Yi Fang,Zhen Shen,Zhi Lv,Pengcui Li,Xiaohong Yao,Weiyi Chen,Xiaochun Wei,Paul K. Chu,Xiangyu Zhang

After prosthesis   replacement for the   treatment   of   bone   tumors, incomplete removal of tumor cells is the main challenge. Moreover, combating implant-associated infection and simultaneously promoting osseointegration are highly demanded for orthopedic implants and therefore, a multifunctional coating that produces tumor therapeutic effects, antibacterial activity, and osteogenesis at the same time is of clinical significance. Herein, a phototherapy platform composed of rare-earth elements - doped titanium dioxide nano-shovel/quercetin/L-arginine (TiO 2 @UCN/Qr/LA) on titanium bone implant is fabricated. The stable nano-shovel structure prepared by a hydrothermal method improves the photothermal conversion ability of titanium. Incorporation of Yb and Er enhances the upconversion capability, thereby facilitating generation of reactive oxygen species (ROS) during exposure to near-infrared II (NIR-II) light. Furthermore, under the action of ROS, L-arginine is catalyzed to release nitric oxide (NO)   free   radicals. In vitro and in vivo experiments indicate that the combined actions of ROS, quercetin, and NO not only inhibit bone tumor growth at a temperature of 48 ℃, but also eliminate Staphylococcus aureus biofilms on the Ti implants at a mild temperature of 45 ℃ upon illumination with a 1,060 nm laser. Moreover, TiO 2 @UCN/Qr/LA promotes angiogenesis and osteogenic differentiation, reduces inflammation, and accelerates formation of new bone tissues.
