Glypican-3 (GPC3), a heparin sulfate proteoglycan, is a potential diagnostic and therapeutic target for hepatocellular carcinoma. In this paper, a novel fluorescent aptasensor for GPC3 detection is constructed via [email protected] quantum dots–labeled GPC3 aptamer ( [email protected] Apt ) as a fluorescence probe. First, [email protected] is screened out with higher fluorescence intensity, which emits bright blue fluorescence under ultraviolet light. Then, the fluorescence-labeled [email protected] Apt probe is formed by the combination of amination GPC3 Apt and [email protected] using EDC/NHS coupled reaction. Under hydrogen bond and π-π interaction/stacking, the fluorescence of [email protected] Apt could be quenched by reductive graphene oxide (RGO) with the help of the photoinduced electron transfer and the fluorescence resonance energy transfer mechanism. In the presence of GPC3, the [email protected] Apt specifically recognizes and binds to GPC3, giving rise to the change of secondary structure of GPC3 Apt to form the GPC3/GPC3 Apt [email protected] complex, which would lead to the disintegration of the [email protected] Apt -RGO compound. Therefore, the energy transfer process is blocked and the fluorescence intensity is restored, enabling a highly sensitive response to GPC3. When the concentration of GPC3 is from 5.0 ng/mL to 150.0 ng/mL, the fluorescence recovery rate is well linearly related to GPC3 concentration with the limit of detection of 2.395 ng/mL (S/N = 3). This strategy shows recoveries from 98.31% to 101.89% in human serum samples and provides simple, fast and cheap analysis of GPC3, which suggests that it has great potential applications in clinical diagnosis for hepatocellular carcinoma.

A novel photonic crystal aptamer biosensor SiO 2 -Au-ssDNA two-dimensional photonic crystal (2D PC), allowing label-free and highly sensitive to kanamycin (KANA), is successfully manufactured. This 2D PC biosensor was prepared via a needle tip flow method, using electrostatic adsorption to introduce negatively charged gold nanoparticles (Au NPs) into the 2D PC, combined with sulfhydryl-modified ssDNA for the rapid measurement. Benefiting from the localized surface plasmon resonance effect of Au NPs and optical response capability of PC, the biosensor has an excellent performance on quantitative analysis of KANA ranging from 5 pg∙mL −1 to 5 μg∙mL −1 , with a limit of detection of 1.10 pg∙mL −1 . The recovery of KANA is between 97 % and 110 % in the milk samples with relative standard deviation less than 4.8 %, which revealing that the 2D PC biosensor has the excellent performance on the KANA detection in complex conditions.

Patulin (PAT) is an unsaturated lactone mycotoxin primarily produced by Penicillium expansum and Aspergillus clavatus . Given the potential health risks and economic losses associated with PAT, the rapid detection of PAT using fluorescent aptasensors is of significant importance in evaluating food safety. However, it easily increases the cost and complexity caused by signal labeling. We combined TCPP/BDC-NH 2 mixed ligands functionalized Zr metal–organic frameworks (Zr-MOF mix ) and terminated three-stranded DNA gates (ttsDNA gates) to fabricate a label-free fluorescent aptasensor for PAT detection. The Zr-MOF mix system was synthesized via a one-pot strategy and could be used to address the problem of pore size limitation and increase the loading amounts of dyes. TtsDNA gate was integrated into the Zr-MOF mix system to control the release of dyes, exhibiting a high signal-to-background ratio. The single-stranded aptamer region in ttsDNA gate situated away from the surface of the Zr-MOF mix , resulting in a natural release of dyes in the absence of PAT. While binding to PAT resulted in target-induced conformational changes that helped form the hairpin structure of the aptamer. This structure hindered the release of dyes from the pores of Zr-MOF mix , thus reducing the fluorescence signals intensity. The stimuli-responsive DNA-gated material provides a platform for PAT analysis under conditions of a low limit of detection (0.871 pg/mL). Furthermore, the excellent specificity and anti-interference of the fluorescent aptasensor make the system suitable for the analysis of apple juice samples. This label-free strategy is cheaper and simper compared with labeled detection, especially for the development of multi-target-detection.

Exosomes are one of the most promising biomarkers for tumor diagnosis and prognosis. Therefore, the development of convenient and sensitive exosome sensing strategies is of great significance. Herein, we integrated aptamer-based spherical nucleic acids (SNAs) and hybridization chain reaction (HCR) into a colorimetric aptasensor platform and applied it to the detection of exosomes. In this design, the CD63-specific aptamer pre-immobilized on the microplate was used to capture target exosomes, while the SNAs conjugated with nucleolin-specific aptamer and trigger probe H1 were designed for amplifying signal. In the presence of target exosomes, the SNAs can be attached to the microplate by the bridge effect of exosomes, resulting in the trigger of HCR. This process is accompanied by the formation of abundant G-quadruplex/hemin DNAzyme, enabling the visual quantitative analysis of exosomes. Featured with the dual amplification of SNAs and HCR, the proposed aptasensor achieved a considerable detection limit of 50 particles/μL. The practicability of this method was further verified by testing the different clinical samples. Given the ability of the aptasensor to visually detect exosomes in scenarios lacking instruments and resources, we believe that the aptasensor can be serve as a potential on-site test for liquid biopsy.

Kanamycin residues accumulate in the human body and endanger human health, so the sensitive detection of kanamycin has important research significance. Herein, we constructed an enzyme-free and label-free fluorescent aptasensor for sensitive detection of kanamycin in milk samples based on hybridization chain reaction. We first designed a kanamycin recognition probe with a blocked aptamer and a blocked trigger sequence. Kanamycin specifically recognized the aptamer and then initiated the conformational transition process of the recognition probe to generate an aptamer-kanamycin complex with an activated trigger sequence. The trigger sequence initiated alternate hybridization of two partially complementary assembled probes to generate a long double-stranded DNA with many intact G-quadruplex structures for amplified, enzyme-free detection of kanamycin. The fluorescent dye N-methylmesoporphyrin IX was inserted into the intact G-quadruplex structures, and then it emitted a strong fluorescent signal for amplified, label-free detection of kanamycin. The linear range was from 2.0 to 200 nM for kanamycin, and the limit of detection (LOD), limit of quantification (LOQ) were 0.562 nM and 1.855 nM respectively. Moreover, the fluorescent aptasensor presented high selectivity for kanamycin compared with other antibiotics. Furthermore, the recoveries of spiked kanamycin in milk samples ranged from 98.66% to 100.7%, indicating that the fluorescent aptasensor had a great application prospect in food safety detection.

In view of the urgent need for fluoroquinolones contamination detection in the fields of food safety, a novel aptasensor based on the fluorescence quenching property of graphene oxide (GO) and the fluorescence characteristic of cadmium selenide quantum dots (CdSe QDs) was developed for fluoroquinolones highly sensitive detection in this work. The CdSe QDs with carboxyl-rich surface were synthesized successfully and fluoresced at 525 nm under the optimal excitation light of 366 nm. Based on the hydrophobic and π-π stacking between GO and aptamer, aptamer labeled by CdSe QDs fluorescence (CdSe QDs-apt) were adsorbed by GO and the fluorescence of CdSe QDs was quenched. After the aptamer combined specifically with fluoroquinolones, greater specific force lead to the desorption of CdSe QDs-apt from GO and fluorescence recovery. Represented by Ciprofloxacin (CIP), a member of fluoroquinolones, the fluorescence emission increased with the increasing of CIP concentrations from 8 nM to 500 nM, and the detection limit was 0.42 nM. The spiked recoveries in real samples of honey and milk were 91.5–96.9 % and 90.3–95.2 %, respectively, indicating that the aptasensor was reliable. Moreover, the fluorescence responses of multiple members of fluoroquinolones were found to be consistent, denoting that the fluorescence aptasensor can be used to detect the total amount of multiple members of fluoroquinolones. These results showed that the aptasensor can be used as a promising platform for fluoroquinolones detection.

Aflatoxin B1 (AFB1) with tremendous toxic effects has caused a serious threat to food security. Accurate quantification of AFB1 in food can effectively prevent the risk of human intake of AFB1. Herein, a colorimetric and fluorescent dual-mode aptasensor for accurate and sensitive detection of AFB1 has been developed based on MnO 2 nanoflowers (MnO 2 NFs) for the first time. MnO 2 NFs could catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) into blue oxidation product (TMBox) by H 2 O 2 , which would be used for visible detection of AFB1. Simultaneously, MnO 2 NFs can be served as a signal amplifier and reduced by ascorbic acid to generate lots of Mn 2+ which would quench the fluorescence of calcein for fluorescent detection of AFB1. Both colorimetric and fluorescent methods have been successfully applied for determination of AFB1 in milk samples with satisfactory results. The proposed dual-mode detection method with high detection sensitivity and accuracy showed great promise for monitoring AFB1 in food.

The detection of glioma cells (GMC) could be used to evaluate the effectiveness of treatment. Based on the advantages of easy synthesis and modification of aptamers and the advantages of high sensitivity, excellent selectivity, and wide application of electrochemical methods, this work constructs an aptamer electrochemical sensor that can be used for rapid and efficient detection of GMC. We used activators for the activation of carboxyl groups after modification of 4-carboxyphenyl diazonium salt. The aptamer, which can specifically recognize GMC, is immobilized on the electrode surface by forming an amide bond between the amino and carboxyl groups on the aptamer. The conditions affecting the sensing of the aptamer sensor were investigated in detail. The current of the aptamer sensor is proportionally with the GMC concentration from 1×10 7 to 1×10 2 cells/mL. Moreover, the sensor can identify GMC with reasonable specificity.

In this paper a photoelectrochemical (PEC) aptasensor based on specific recognition between aptamer and target cadmium ion (Cd(II)) with change of the sensing interface structure was constructed. A ZnO-TiO 2 nanocomposites and Au nanochains (Au NCs) modified electrode as photosensitive substrate was prepared and further covalently bound with graphite-like carbon nitride (g-C 3 N 4 ) on the free end of the aptamers to amplify the detection signal, which was applied to Cd(II) detection. Graphite-like carbon nitride with narrow band gap exhibited unique photoelectric property under UV and visible light irradiation. When the target Cd(II) was detected by the modified interface of the aptasensor, the structure of sensing interface was changed with the signal sensitizer g-C 3 N 4 closed to the interface, which caused the changes of photocurrent signal more sensitive and exhibited a good linear relationship with the Cd(II) concentration, so as to realize the quantitative analysis of the target. This PEC aptasensor showed a high sensitivity with a detection limit as 1.1 × 10 −11 mol/L and was further applied to the water sample detection successfully.

The construction of simple and highly sensitive aptasensors will significantly advance the monitoring of proteins. In this work, we have developed a convenient and label-free protein-induced catalytic hairpin assembly signal amplification approach for aptamer-based sensitive electrochemical detection of thrombin in human serums. The target thrombin binds the aptamer-containing hairpin probes immobilized on the sensor electrodes and triggers catalytic assembly of other two hairpins to form many G-quadruplex Y-junction DNA structures in situ . These G-quadruplex structures further associate with hemin to form G-quadruplex/hemin complexes, which are then subject to electrochemical measurement and generate substantially amplified current output for label-free and sensitive detection of thrombin. The developed aptasensor shows a dynamic range of 0.01–1.0 nM and a detection limit of 6.0 p.m for thrombin detection. The sensor can also discriminate the target thrombin against other non-target proteins and exhibits promising results for detecting thrombin in human serums. With the successful construction of the thrombin aptasensor, our signal amplified method can be potentially applied to detect other target molecules by choosing appropriate aptamer/ligand pairs.

This paper proposes a fluorescent aptasensor for the detection of carbendazim (CBZ) in aqueous solution using CBZ-specific aptamer as sensing probe, gold nanoparticles (AuNPs) and Rhodamine B (RhoB) as indicator, respectively. In the absence of CBZ, CBZ aptamer could wrap AuNPs and maintained it dispersed in NaCl solution basically. Contrarily, the aptamer could specifically combine with CBZ and form a stable aptamer-CBZ complex, leaving AuNPs exposed to be aggregated by NaCl solution. The dispersed AuNPs could efficiently quench the fluorescence of RhoB, but those aggregated AuNPs have poor capability to impair the fluorescent indicator . Thus, the concentration of CBZ could be detected quantitatively through the distinction of the fluorescence intensity . This convenient fluorescent assay for CBZ had a wide linear range from 2.33 to 800 nM and a 2.33 nM limit of detection (LOD). Furthermore, it had high selectivity over pesticides, antibiotics, metal ions and other disrupting chemicals. As for application, the method could determine CBZ in water samples with recoveries in the range of 96.3–111.2%. This fluorescent aptasensor possessed great potential application for CBZ detection in actual aquatic environment.

A novel and label-free electrochemical aptasensor based on exonuclease III (Exo III) and G-quadruplex DNAzyme has been first designed to detect metronidazole (MNZ) sensitively and selectively in this paper. Capture probe (CP), helper probe (HP) and aptamer (Apt) hybridize to form double-stranded DNA (dsDNA) structure with a blunt 3′-terminus, resulting in the guanine (G)-rich CP being enclosed in the dsDNA, followed by specific digestion of the 3′-terminus by Exo III to expose the G-rich sequence. At this time, CP can bind with the added hemin to form G-quadruplex DNAzyme which catalyzes the redox reaction of 3,3′,5,5′-tetramethylbenzidine (TMB) and H 2 O 2 , generating a strong current signal. However, in the presence of MNZ, the binding of MNZ and aptamer breaks the original blunt 3′-terminus of the dsDNA structure, blocking the subsequent series of reactions, and only a weak current signal can be observed. Under the optimal conditions, the aptasensor showed excellent sensitivity and specificity for the detection of MNZ with a wide linear range from 10 pM to 500 nM and a detection limit as low as 3.0 pM (S/N = 3). The aptasensor constructed based on MNZ aptamer can effectively avoid the interference of other nitro compounds and antibiotics to a certain extent. Compared with other specific recognition elements, the aptasensor possesses the advantages of strong specificity, simple preparation and low cost. Moreover, the proposed aptasensor has been successfully applied to the detection of MNZ in tap water and honey samples.
