A novel three-dimensional composite scaffold was fabricated via a carefully designed strategy including preparation of a composite with components of cellulose as backbone and alginate as pore size expander, (bis)phosphonate-modification via oxidation with sodium periodate followed by Schiff-base reaction with sodium alendronate, and final biomimetic mineralization. The scaffolds possess appropriate porous structure and porosity. The chemical structure and morphology of the obtained composite scaffolds suggested that the deposited hydroxyapatite on the surface of the phosphorylated scaffolds was close to that of natural bone, including the content, crystallite size, crystallinity and Ca/P ratio. Moreover, it was also revealed that the scaffolds exhibited no cyto-toxicity and the cultured cells were well attached on the surface. Our investigation demonstrates that the presented (bis)phosphonate-modified cellulose/alginate/hydroxyapatite composite material is an appropriate candidate for the fabrication of scaffolds and has great potential to be applied to bone tissue engineering.

The stiffness of most biomaterials used in bone tissue engineering is static at present, and does not provide an ideal biomimetic dynamical mechanical microenvironment for bone regeneration. To simulate the dynamic stiffness better during bone repair, the preparation of dynamic materials, especially hydrogels, has aroused researchers' interest. However, there are still many problems limiting the development of hydrogels such as small-scale stiffness changes and unstable mechanical properties. Here, magnetic liquid metal (MLM) was introduced into bone tissue engineering for the first time. A MLM scaffold was obtained by adding magnetic silicon dioxide particles ( [email protected] 2 ) into galinstan. Furthermore, a porous MLM (PMLM) scaffold was obtained by adding polyethylene glycol as a template to the MLM scaffold. Both scaffolds can respond to external magnetic fields, so changing the magnetic field intensity can achieve a large-scale of dynamic stiffness change. The results showed that the MLM scaffold has good biocompatibility and can promote the osteogenic differentiation of mesenchymal stem cells (MSCs). The PMLM scaffold with dynamic stiffness can promote new bone regeneration and osseointegration in vivo . Our research will open up a new field for the application of liquid metal and bring new ideas for the development of bone tissue engineering materials.

This study developed a biodegradable composite porous polyurethane scaffold based on polycaprolactone and polyethylene glycol by sequential in-situ foaming salt leaching and freeze-drying process with responsive shape changing performance. Biomineral hydroxyapatite (HA) was introduced into the polyurethane matrix as inorganic fillers. Infrared spectroscopy results proved a successful synthesis, scanning electron microscopy showed that the scaffold's porosity decreased with the addition of HA while the average pore size increased. X-ray diffraction and differential scanning calorimetry showed that the addition of HA lowered the melting point of the scaffold, resulting in a transition temperature close to the human body temperature. From the bending experiments, it could be demonstrated that PUHA20 has excellent shape memory performance with shape fixity ratio >98.9% and shape recovery ratio >96.2%. Interestingly, the shape-changing capacity could be influenced by the porous structures with variation of HA content. The shape recovery speed was further accelerated when the material was immersed in phosphate buffered saline at 37 °C. Additionally, in vitro mineralization experiments showed that the scaffold incorporating HA had good osteoconductivity, and implantation assessment proved that scaffolds had good in vivo biocompatibility. This scaffold is a promising candidate for implantation of bone defects.

Due to the limited ability to self-repair, the regeneration of bone critical-sized defects (CSD) is a significant challenge. Bone tissue engineering scaffolds are considered promising candidates for CSD repair, but low cell infiltration efficiency and a lack of nutrients greatly restrict bone regeneration abilities. Herein, we developed a dynamic culturing of large biomimetic bone scaffolds, PCL/GelMA@cells that combining 3D printed polycaprolactone (PCL) multi-channel cylinder with gelatin methacryloyl (GelMA) encapsulated with bone marrow mesenchymal stem cells (BMSCs) and rat aortic endothelial cells (RAECs). A cell dynamic culture system was fabricated to simulate the dynamic microenvironment. Compared to static culturing, dynamic culturing proved to enhance the nutrient exchange within the large scaffold to promote the cells infiltration, growth, proliferation and induce osteogenic and angiogenic differentiation. Furthermore, a rat cranial CSD (D = 10 mm) repair model verified the accelerated vascular ingrowth and new bone formation with the implantation of dynamic culturing of PCL/GelMA@cells scaffold (∼10 times higher than Blank group), indicating the great potential of dynamical culturing of scaffolds for bone repair. In summary, the results highlight the significant advantages of the dynamical culturing of cell-loaded scaffolds for bone regeneration, offering a promising strategy for addressing critical size bone defects.

Mesoporous bioactive glasses (MBGs) are gaining recognition in bone tissue engineering for their osteoblast differentiation-inducing properties and customizable structures. However, the challenge of peri-implantitis has hindered their broader application. To address this, double transition metal TiNbCT x MXene acted as near-infrared (NIR) photothermal agent, has been integrated with MBG nanospheres to develop novel multifunctional TiNbCT x /MBG (TNC/MBG) nanocomposites in this work. The results demonstrate that the TNC/MBG composites feature uniform mesoporous spherical MBG nanoparticles on TiNbCT x nanosheets, boasting a significantly enhanced specific surface area of up to 444.74 m 2 g −1 and a more negative zeta potential than pristine MBG. Importantly, the TNC/MBG nanocomposites can effectively promote apatite formation, as well as the proliferation and viability of MC3T3-E1 cells. Moreover, it is highlighted that the TNC/MBG nanocomposites display remarkable photothermal conversion efficiency and stability, leading to over 95% antibacterial inhibition rates against both S. aureus and E. coli under NIR irradiation. These findings offer an appealing strategy to develop multifunctional TNC/MBG nanocomposites with enhanced biological activity and robust antibacterial properties, which shows great potential for various biomedical applications.

Addressing the prevalent clinical issue of bone defects, this study successfully developed a novel ternary copolymer material (PLCD) based on polylactic acid (PLA), polycaprolactone (PCL), and polydioxanone (PPDO). This ternary material combines the strength of PLA, the toughness of PCL, and the controllable rapid degradation properties of PPDO. making it a promising candidate for biomedical applications. By using an optimized emulsion method combined with finite element analysis, microspheres with a particle size range of 500–700 μm were prepared and subsequently fabricated into microsphere scaffolds using the solvent sintering method. To enhance osteoinductive and osseointegration capabilities, various proportions of β-tricalcium phosphate (β-TCP) were uniformly dispersed within the microspheres, creating composite scaffolds. Additionally, nano zinc oxide were coated on the microsphere scaffold surface using the in situ polymerization properties of dopamine, imparting antibacterial properties and enhancing functionality. Physical and biological evaluations of the microsphere scaffolds revealed excellent overall performance, including an interconnected porous structure, porosity and mechanical properties matching those of cancellous bone, suitable degradation behavior, and in vitro mineralization ability. Furthermore, scaffolds coated with polydopamine and loaded with nano zinc oxide exhibited strong initial antibacterial effects, and promoted osteogenic differentiation. These results indicate that the developed composite microsphere scaffolds have significant potential in bone tissue engineering applications, offering a promising solution for effective bone regeneration.

In this work, the near-infrared (NIR) light-responsive shape memory scaffolds with hierarchical porous structures are designed and facilely formed by freeze drying of 3D printed viscous gel-like pickering emulsions, which are stabilized by hydrophobically modified graphene oxide (g-GO) and silica nanoparticles, and contain thermo-responsive poly( d , l -lactic acid-co-trimethylene carbonate) (PLMC) in the oil phase. The prepared scaffolds display an interconnected filament structure with hierarchical pores and high porosity. The incorporation of g-GO nanoparticles into PLMC matrix prompts that the scaffold shape memory can be triggered by NIR light with fast shape recovery. Moreover, the in vitro mineralization experiment shows that the scaffolds have biological activity, and the drug release study demonstrates that the scaffolds can be used as drug carriers with efficient drug release capacity. Furthermore, cell culture assays based on mouse bone mesenchymal stem cells exhibit that the scaffolds own good cytocompatibility. Therefore, the facile preparation and remote activation of the shape memory nanocomposite scaffolds with hierarchical porous structure and multifunctionality represents a highly attractive candidate as minimally invasive implantation scaffolds for bone tissue engineering applications.

Bone tissue engineering is sought to apply strategies for bone defects healing biodegradable porous scaffolds without limitations and shortcomings. In this work, we have developed a novel maleic anhydride (MAH) and 1,4-butanediamine modified poly(lactide-co-glycolide) polymer (BMPLGA). The synthesized polymer was characterized by Fourier transform infrared spectrometry (FTIR), Nuclear magnetic resonance spectra (1H NMR), gel permeation chromatography (GPC) and contact angle measurements. In addition, cell morphologies in the extracts and cell cytotoxity were also studied. The results showed that the BMPLGA was successfully obtained by introducing MAH and 1,4-butanediamine into PLGA in bulk. The introduction of anhydride and amino groups improved the hydrophilicity of PLGA. Fibroblastic cells showed normal morphologies in BMPLGA extracts, and the BMPLGA materials showed no cell cytotoxicity. The synthetic BMPLGA material may have potentials for biomedical applications due to improving hydrophilicity.

Three-dimensional (3D) homogenous scaffolds composed of natural biopolymers have been reported as superior candidates for bone tissue engineering. There are still remaining challenges in fabricating the functional scaffolds with gradient structures to similar with natural bone tissues, as well as high mechanical properties and excellent affinity to surround tissues. Herein, inspired by the natural bone structure, a gradient-structural scaffold composed of functional biopolymers was designed to provide an optimized 3D environment for promoting cell growth. To increase the interactions among the scaffolds, dopamine (DA) was employed to modify alginate (Alg) and needle-like nano-hydroxyapatite (HA) was prepared with quaternized chitosan as template. The obtained dopamine-modified alginate (Alg-DA) and quaternized chitosan-templated hydroxyapatite (QCHA) were then used to fabricate the porous gradient scaffold by “iterative layering” freeze-drying technique with further crosslinking by calcium ions (Ca 2+ ). The as-prepared Alg-DA/QCHA gradient scaffolds were possessed seamlessly integrated layer structures and high levels of porosity at around 77.5%. Moreover, the scaffolds showed higher compression modules (1.7 MPa) than many other biopolyermic scaffolds. The gradient scaffolds showed appropriate degradation rate to satisfy with the time of the bone regeneration. Both human chondrocytes and fibroblasts could adhesive and growth well on the scaffolds in vitro . Furthermore, an excellent osteogenetic activity of the gradient scaffold can effectively promote the regeneration of the bone tissue and accelerate the repair of the bone defects in vivo , compared with that of the scaffold with the homogenous structure. The novel multilayered scaffold with gradient structure provided an interesting option for bone tissue engineering. © 2019 Wiley Periodicals, Inc. J Biomed Mater Res Part A: 107A: 1615–1627, 2019.

Natural-synthetic hybrid scaffold has attracted great interest due to its synergistic effect on load-bearing bone reconstruction. Herein, sodium alginate (SA)/gelatin (Gel) composite were combined with poly(ε-caprolactone)/58S bioactive glass (PCL/BG) composite to construct natural-synthetic hybrid scaffold by a modified melt molding method. Our results showed that the SA/Gel microspheres used as porogen remained inside the scaffold, which had potential benefits for cell adhesion. The PCL/BG composite acted as the struts. The incorporation of BG resulted in a significant increase in the mechanical properties of the scaffold. Specially, HS-3 scaffold showed a compressive strength of 1.44 ± 0.02 MPa falling within the range of values reported for the cancellous bone. In addition, the presence of BG improved wettability and biodegradability of the scaffold, thus potentially enhancing osteointegration. In conclusion, the PCL/BG–SA/Gel hybrid scaffold with improved mechanical properties and potential cytocompatibility are promising biomaterials for bone repair.

Three-dimension (3D) porous reduced graphene oxide (RGO) scaffold has attracted increasing attention in bone tissue engineering due to its favorable osteoinductivity. In this paper, a 3D porous RGO composite was prepared from graphene oxide (GO) and nano-hydroxyapatite (nHA) via self-assembly, thus constructing biomimetic scaffold for bone defect reparation. Detailed studies were performed to evaluate its structure, cellular responses, biocompatibility and in vivo bone repair efficiency, emphasizing the influences of the composite on in vivo bone cell growth and mineralization. The as-prepared scaffold was found to significantly enhance the proliferation, alkaline phosphatase activity (ALP) and osteogenic gene expression of rat bone mesenchymal stem cells (rBMSCs). Further in vivo experiment demonstrated that the circular calvarial defects with 4 mm diameter in rabbit were successfully healed by 20% nHA incorporated RGO ( [email protected] ) porous scaffold after 6 weeks implanting, which was visibly quicker than the RGO one. The computed tomography (CT) and histological analysis showed the improved collagen deposition, cell proliferation and new bone formation occurred in the 20% [email protected] treated group. These results indicated that the as-prepared porous scaffold has a promising capacity to stimulate mineralization and promote the in vivo defect healing.

Large bone defects due to accidental injuries and fractures as well as the mismatch between the implant material and the defect site are major problems in biomedicine. Bone tissue engineering offers a solution to this problem. In this paper, a series of three-dimensional porous composite scaffolds with shape memory were prepared using polycaprolactone and polytetrahydrofuran as soft segment materials, hexamethylene diisocyanate as hard segment materials and amorphous calcium phosphate/calcium citrate (CAP) as inorganic fillers. The scaffolds have tunable mechanical properties, melt temperature, surface wettability, a pore size range of 100–800 μm and a high degree of connectivity. In vitro mineralization showed good mineral deposition and energy dispersion spectrometer displayed uniform distribution of Ca and P elements on the surface. The composite scaffold exhibited good shape memory properties at 37°C. The shape fixation rate ( R f ) and shape recovery rate ( R r ) were maintained at 94.1% and 94.4% after five compression cycles. The resulting scaffold promoted MG63 cell proliferation to some extent. The scaffold is expected to be used clinically for the treatment of large bone defects.
