Efficient and sensitive electrochemical biosensors are of great significance for on-site measurement and evaluation of pesticides in the environment, and various functional nanomaterials have been designed and developed to achieve this goal. Herein, a kind of three-dimensional holey hierarchical cobalt-based bimetallic oxides randomly assembled by micro-mesoporous ultrathin nanosheets has been exploited. This unique porous hierarchical structure and bimetal composition not only possesses excellent electron pathways and consequent an enhanced electro-conductivity and better catalytic activity , but also exhibits high electroactive surface area and more vacant sites for enzyme immobilization . Along with the highly conductive and biocompatible layered graphdiyne , the 3D porous Co-based bimetallic oxides have been applied to construct an electrochemical AChE biosensor for monitoring dipterex and omethoate (as model OPs pesticides). As a result, the reported biosensor demonstrates superior sensing performance, for example, the wide detection range of 3.88 × 10 −12 –3.88 × 10 −6 M for diptetex and 4.69 × 10 −13 –4.69 × 10 −7 M for omethoate , together with the relative low detection limit of 0.36 pM and 0.033 pM (S/N = 3), respectively. Additionally, the biosensor is feasible in real samples, and the recoveries (94.8–105.1%) are satisfactory. And a possible mechanism has also been proposed. This study offers a new strategy to modulate the property of metal oxides , which make them more attractive in electrochemical sensors .

Alzheimer’s disease (AD) is a common chronic neurodegenerative disease that manifests as cognitive impairment and behavioral deficits and severely threatens the health of the elderly. Acetylcholinesterase (AChE) plays a vital role in biological signaling and is an essential target for the early diagnosis and treatment of AD. Herein, 2D Zn-TCPP(Fe) nanosheets (NSs) employing Zn2+ and Fe-bound tetrakis(4-carboxyphenyl)porphyrin ligands were prepared through a surfactant-assisted synthetic method. The ultrathin two-dimensional (2D) metal–organic framework structures exhibited high peroxidase-like activity, which allowed the catalysis of the H2O2-initiated oxidation of colorless 3,3′,5,5′-tetramethylbenzidine (TMB) to blue oxidized TMB (ox-TMB). Such catalytic performance inspired us to develop a convenient, rapid, and sensitive acetylcholinesterase activity assay, during which AChE can catalyze the substrate acetylthiocholine (ATCh) to produce thiocholine (TCh), and TCh could especially enable the degradation of 2D Zn-TCPP(Fe) NSs accompanied by the reduction of ox-TMB production. Our proposed sensing system exhibited favorable selectivity and sensitivity (LOD of 0.029 mU/mL) and has excellent potential to evaluate AChE activity in human serum samples and to screen AChE inhibitors. This colorimetric assay could provide an alternative pathway for early diagnosis and drug screening of AD, facilitating the development of AD therapy.

An exceptionally stable biosensor for pesticide monitoring was engineered through the assembly of a multilayered core-shell nanostructure incorporating polymeric ionic liquid (PIL)-encased acetylcholinesterase (AChE). In this design, AChE is encapsulated within PILs to form the inner core, PILs serve as the intermediate layer, and silver nanoparticles (Ag NPs) are uniformly assembled on the outer layer to provide surface enhancement effects. The PILs microspheres provide a biocompatible microenvironment to stabilize AChE activity while enabling tunable charge density and ordered structural frameworks to guide the uniform assembly of Ag NPs into nanoscale arrays. This precise arrangement of Ag NPs amplifies localized electromagnetic coupling, significantly enhancing surface-enhanced Raman scattering (SERS) signals. The composite was further modified with 2-(pyridyldithio)ethylamine (PDEA), which functions as a SERS reporter through its pyridine ring vibrations. The detection mechanism involves two critical steps: (1) AChE hydrolyzes acetylthiocholine (ATCl) to generate thiocholine (TCl), which cleaves PDEA's disulfide bonds, releasing pyridine rings and attenuating SERS signals; (2) Organophosphate pesticides inhibit AChE activity, thereby reducing TCl production and preserving PDEA's signal intensity for indirect quantification. Owing to the synergistic integration of hierarchical stability, electromagnetic enhancement, and molecular recognition capabilities, the multilayer core-shell structure (denoted as AChE@PILs@Ag NPs-PDEA) achieves ultrasensitive detection across a wide linear range from 3.8 × 10 −7 to 3.8 × 10 −12 M, reaching an exceptional detection limit of 1.38 × 10 −13 M. The biosensor demonstrates robust thermal stability, long-term storage reliability, and reliable matrix interference resistance in food/environmental matrices (tomato homogenates, tap water), validating its practicality for on-site pesticide monitoring in agricultural products under variable environmental conditions.

Background Studies have found that α-mangostin (MG) can relieve experimental arthritis by activating cholinergic anti-inflammatory pathway (CAP). It affects the polarization of macrophages and the balance of related immune cell subpopulations, but the specific mechanism is still unclear. It has been found that silent information regulator 1 (SIRT1) is closely related to macrophage activity. The purpose of this study is to explore the mechanism of MG intervening in macrophage polarization during treatment of early adjuvant-induced (AIA) rats through the CAP-SIRT1 pathway. Methods We investigated the polarization of M1 macrophages and the differentiation of Th1 in AIA rats by flow cytometry. Activity of acetylcholinesterase (AChE) and the level of nicotinic adenine dinucleotide (NAD+) in serum were also detected, and immunohistochemical was used to detect the levels of α7 nicotinic cholinergic receptor (α7nAChR) and SIRT1. Then in macrophages, the molecular mechanism of MG regulating the abnormal activation of macrophages in rats with early AIA through the CAP-SIRT1 pathway was studied. Results MG can significantly inhibit the polarization of M1 macrophages and the differentiation of Th1 in AIA rats in the acute phase of inflammation. MG can significantly inhibit the activity of AChE and increase the level of NAD+, thereby further up-regulated the expression levels of α7nAChR and SIRT1. Meanwhile, MG inhibited nuclear factor-κB (NF-κB)-mediated inflammation by activating the CAP-SIRT1 pathway in macrophages. Conclusion In summary, the stimulation of MG induced CAP activation, which up-regulated SIRT1 signal, and thereby inhibited M1 polarization through the NF-κB pathway, and improved the pathological immune environment of early-stage AIA rats.

Lead compound is an important concept for modern drug discovery. In this study, a new concept of lead chemome and an efficient strategy to discover lead chemome were proposed. Compared with the concept of lead compound, lead chemome can provide not only the starting point for drug development, but also the direction for structure optimization. Two traditional Chinese medicines of Mahonia bealei and Mahonia fortunei were used as examples to illustrate the strategy. Based on natural chromatogram-effect correlation (NCEC), berberine, palmatine and jatrorrhizine were discovered as acetylcholinesterase (AchE) inhibitors. Taking the three compounds as template molecules, a lead chemome consisting of 10 structurally related natural compounds were generated through natural structure-effect correlation (NSEC). In the lead chemome, the IC 50 values of jatrorrhizine, berberine, coptisine, palmatine and epiberberine are at nanomolar level, which are comparable to a widely used drug of galantamine. Pharmacophore modeling shows that the positive ionizable group and aromatic rings are important substructures for AchE inhibition. Molecular docking further shows that pi-cation interaction and pi-pi stacking are critical for compounds to maintain nanomolar IC 50 values. The structure–activity information is helpful for drug design and structure optimization. This work also expanded the traditional understanding of “stem is the medicinal part of Mahonia bealei and Mahonia fortunei ”. Actually, all parts except the leaf of Mahonia bealei exhibited potent AchE-inhibitory activity. This study provides not only a strategy to discover lead chemome for modern drug development, but also a reference for the application of different parts of medicinal plants.

Background Non-alcoholic fatty liver disease (NAFLD) poses a significant global public health concern. Liupao tea (LPT) is a Chinese national geographical indication product renowned for its lipid-lowering properties. However, the precise mechanisms and active constituents contributing to the efficacy of LPT against NAFLD remain unclear. Purpose This study aims to comprehensively explore the therapeutic potential of Liupao tea extract (LPTE) in alleviating NAFLD through an integrated strategy. Methods Initially, network pharmacology analysis was conducted based on LPTE chemical ingredient analysis, identifying core targets and key components. Potential active ingredients were validated through chemical standards based on LC-MS/MS. To confirm the pharmacological efficacy of LPTE in NAFLD, NAFLD mice models were employed. Alterations in hepatic lipid metabolism were comprehensively elucidated through integration of metabolomics, lipidomics, network pharmacology analysis, and real-time PCR analysis. To further explore the binding interactions between key components and core targets, molecular docking and microscale thermophoresis (MST) analysis were employed. Furthermore, to investigate LPTE administration effectiveness on gut microbiota in NAFLD mice, a comprehensive approach was employed. This included Metorigin analysis, 16S rRNA sequencing, molecular docking, and fecal microbiome transplantation (FMT). Results Study identified naringenin, quercetin, luteolin, and kaempferol as the potential active ingredients of LPTE. These compounds exhibited therapeutic potential for NAFLD by targeting key proteins such as PTGS2, CYP3A4, and ACHE, which are involved in the metabolic pathways of hepatic linoleic acid (LA) and glycerophospholipid (GP) metabolism. The therapeutic effectiveness of LPTE was observed to be comparable to that of simvastatin. Furthermore, LPTE exhibited notable efficacy in alleviating NAFLD by influencing alterations in gut microbiota composition ( Proteobacteria phylum, Lactobacillus and Dubosiella genus) that perhaps impact LA and GP metabolic pathways. Conclusion LPTE could be effective in preventing high-fat diet (HFD)-induced NAFLD by modulating hepatic lipid metabolism and gut microbiota. This study firstly integrated bioinformatics and multi-omics technologies to identify the potential active components and key microbiota associated with LPTE's effects, while also primally elucidating the action mechanisms of LPTE in alleviating NAFLD. The findings offer a conceptual basis for LPTE's potential transformation into an innovative pharmaceutical agent for NAFLD prevention.
