One-Step Labeling Based on Eu-MOFs to Develop Fluorescence Side-Flow Immunoassay for AFB1 Detection in Corn

Yinjun Li,Hua Ding,Ziyu Wang,Zewei Luo,Xitian Peng

Lateral flow immunoassay (LFIA) is a promising tool for rapid detection in the field of agricultural product analysis due to its advantages of cost-effectiveness and operational simplicity. In this work, Eu metal–organic frameworks (MOFs) were introduced to LFIA as a rapid detection method characterized by high stability and low interference. Key research objectives included strong fluorescence, ease of labeling, and the utilization of fluorescent probes. Eu-MOFs were synthesized in one step via the hydrothermal method, exhibiting a fluorescence lifetime of 163 μs and spherical particles with diameters ranging from 250 to 400 nm. These conditions fulfill the characteristics and requirements of LFIA. Eu-MOFs exploit the porous nature of MOFs to mitigate the drawbacks associated with complex crosslinking agents. This enables antibody proteins to be cross-linked merely upon contact, thereby simplifying the detection process. A time-resolved LFIA method was developed utilizing Eu-MOFs for the detection of aflatoxin B1 (AFB1) in corn, achieving a limit of detection (LOD, IC10) of 0.149 ng/mL. The accuracy and reliability of the Eu-MOFs-LFIA method were validated through comparisons with spiked concentrations during spiking and blind sample analyses, with verification conducted using ultra-high-performance liquid chromatography mass spectrometry (UPLC-MS). Furthermore, testing of real samples demonstrated that the Eu-MOFs-LFIA method can effectively facilitate rapid detection of AFB1 in corn.

Portable electrochemical aptasensor based on gold-nanoparticles/FeCo-MOFs and PtPd nanoflowers for in site detection of AFB1

Tao Lu,Tengfei Yu,Yaqi Yan,Huan Li,Zhiguang Suo,Qian Li,Min Wei,Huali Jin

Aflatoxin B1 is one of the most toxic carcinogens and has been classified as a group I carcinogen by the International Agency for Research on Cancer. AFB1 is widely distributed in the natural environment and may be present in various grains and products. Therefore, it is significant to establish a strategy for in site detection of AFB1 accurately. Herein, a portable electrochemical aptasensor was developed for accurate detection of AFB1. A screen-printed electrode was selected as the working electrode, which improves the portability and practical application of the aptasensor. The use of gold-nanoparticles/FeCo-MOFs as electrode modification materials improves both the electron transfer efficiency and the large number of connected signal probes. Daisy-like bimetallic PtPd nanoflowers have rich specific surface area, providing numerous sites for loading of the signal molecule thionine. Additionally, the catalytic hairpin assembly cycle amplification strategy was implemented to effectively enhance the sensitivity of the aptasensor. Under optimal conditions, the sensor exhibited a detection range of 0.001–500 ng/mL and a limit of detection of 0.818 pg/mL. The developed aptasensor exhibited positive analytical performance in real samples, quality control samples and toxin samples produced by aspergillus flavus. This work advances the development of portable, sensitive and in site analytical methods, with promising applications in food safety field testing.

On-site detection of OTA and AFB1 based on branched hybridization chain reaction coupled with lateral flow assay

Wenjun Jiang,Yidan Zhu,Qu Tang,Xiaoxia Kang,Haiwei Ji,Conglin Guo,Xijuan Gu,Jing Zhang,Zhenzhen Mao,Li Wu,Yuling Qin

Mycotoxins are widely prevalent in various agricultural commodities, whose excessive consumption can pose significant risks to human health. In this study, we developed a facile mycotoxin detection platform based on branched hybridization chain reaction coupled with lateral flow assay. Ochratoxin A/Aflatoxin B1 bind to aptamers triggering the release of initiators, which leads to bHCR amplification and forms three-dimensional dendritic DNA nanostructures. Using the functionalized quantum dots as a fluorescent label, by leveraging smartphones and handheld ultraviolet lamps, the qualitative and quantitative detection of OTA and AFB1 can be achieved with a significantly enhanced sensitivity level, surpassing that of commercial test strips by 2–3 orders of magnitude. The visual detection limits for OTA and AFB1 were 30 pg/mL and 4 pg/mL, respectively. This approach eliminates the necessity for enzyme catalysis or the preparation and purification of antibodies and/or hapten, thereby reducing testing expenses and streamlining operational procedures. Moreover, substituting aptamer and nucleic acid sequences can effectively expand the scope of detection targets. Consequently, the as-proposed strategy exhibits great potential as a versatile technique, suitable for various analytical scenarios due to its sensitivity, accuracy, simplicity, and portability.

On-site AFB1 detection in food via dual-gated system and Zn2+-doped UCNPs embedded in injectable PGB hydrogel

Yu Wu,Arul Murugesan,Yaru Han,Juan Han,Yiping Chen,Huanhuan Li

An injectable BZUCNPs-PACMSNs/PGB sensor was proposed to detect AFB 1 . • BPEI was coated on ZUCNPs to improve the water-soluble ability of ZUCNPs • A dual-gated strategy was presented to improve the stability of UCNPs sensor. • BPEI@Cu 2+ can effectively quench the fluorescence of ZUCNPs due to IFE. • Injectable PGB hydrogel was utilized for stable and on-site AFB 1 detection. An expanding presence of aflatoxin B 1 (AFB 1 ) contamination across the global food supply represents a serious threat to public health. This work developed a portable sensor using Zn 2+ -doped upconversion nanoparticles (ZUCNPs) that Cu 2+ -induced fluorescence quenching with an injectable polyvinyl alcohol/gelatin/borax (PGB) hydrogel. First, ZUCNPs exhibiting enhanced fluorescence were modified with branched polyethylenimine (BPEI) to form signal probes known as BZUCNPs. Additionally, mesoporous silica nanoparticles (MSNs) were fabricated via sol-gel methods, loaded with Cu 2+ and coated with aptamer (Apt) and polydopamine (PDA) to create dual-gated capture probes (PACMSNs). Afterwards, BZUCNPs and PACMSNs were mixed to form sensing probes and then integrated into the PGB hydrogel, forming the final sensor (BZUCNPs-PACMSNs/PGB). In the presence of AFB 1 at pH 5.5, the AFB 1 diffused into the PGB hydrogel, disrupting the PDA and Apt layer due to acid environment and changing the structure of Apt as AFB 1 specifically bound to it. Then, Cu 2+ released and facilitated the formation of BPEI@Cu 2+ complex on the surface of ZUCNPs, which produced inner filter effect (IFE) that quenched fluorescence. When experimental conditions were optimized, a pronounced linear calibration curve ( R 2  = 0.994) was observed, linking fluorescence to the logarithm of AFB 1 levels across a 0.1–100 μg/kg AFB 1 range. And its sensitivity was characterized with a detection limit of 0.03 μg/kg. Furthermore, the sensor's applicability was confirmed through analysis of food matrices, including wheat, corn, peanuts, and water, yielding high analytical accuracy with relative standard deviations below 6.08% and recovery rates ranging from 93.77% to 107.67%.

A SERS and Fluorescence Dual-Signal Aptasensor for AFB1 Detection Based on DTNB Labeled AuNPs and CdTe Quantum Dots

Ma Xiaoyuan,Wei Jinxiang,Jiang Caiyun,Wang Zhouping

Herein, an aptasensor with stable surface-enhanced Raman scattering (SERS) and fluorescence dual-signal output had been developed for the detection of aflatoxin B 1 (AFB 1 ). The quantum dots were encapsulated within silica to ensure the stability of fluorescence signal. The gold nanoparticles (AuNPs) were modified with SERS signal molecule DTNB and the complementary sequence (cDNA) of AFB 1 aptamer and then connected on the surface of silica to obtain the signal probes. In the meantime, Fe 3 O 4 was coated with silica which was modified with aptamer to gain capture probes. The two probes could bind specifically based on the base complementary pairing principle. Then, the mixture was separated and gathered by an external magnet. At this time, the maximum SERS signal could be detected in the precipitate, and there was no fluorescence signal in the supernatant. Upon introduction of AFB 1 into the system, the signal probes would be released, resulting in fluorescence recovery in the supernatant while SERS signal weakened in the precipitate. The developed method showed a linear relationship of AFB 1 for the SERS signal in the range of 10 −4 –10 3  ng/mL, with a detection limit of 0.087 pg/mL. The linear range for the fluorescence signal was from 10 −4 to 10 2  ng/mL, with a detection limit of 0.100 pg/mL. The proposed technique was highly sensitive and accurate, which showed promising application prospects in food safety detection.

Target-Induced Electrochemical Sensor Based on Foldable Aptamer and MoS2@MWCNTs–PEI for Enhanced Detection of AFB1 in Peanuts

Xiaoya Meng,Maosheng Sang,Qi Guo,Zhongyu Li,Quanlong Zhou,Xia Sun,Wenping Zhao

Herein, a sensitive and selective electrochemical sensor based on aptamer folding was constructed to detect aflatoxin B1 (AFB1) in peanuts. Specifically, polyethylenimine-functionalized multiwalled carbon nanotubes modified with molybdenum disulfide (MoS2@MWCNTs–PEI) were used as the electrode matrix to enable a large specific surface area, which were characterized by the Randles–Sevcik equation. Additionally, AuNPs were used to immobilize the aptamer via the Au–S covalent bond and provide a favorable microenvironment for signal enhancement. Methylene blue (MB) was modified at the proximal 3′ termini of the aptamer as the capture probe, while the signal transduction of the sensor was obtained through changes in conformation and position of MB induced by the binding between AFB1 and the probe. Changes in spatial conformation could be recorded by electrochemical methods more readily. This electrochemical aptasensor demonstrated remarkable sensitivity to AFB1 with an extensive detection range (1 pg/mL to 100 ng/mL) and a lower limit detection (1.0 × 10–3 ng/mL). Moreover, using the constructed aptasensor, AFB1 was identified successfully in peanut samples, with recoveries ranging from 95.83 to 107.53%, illustrating its potential use in determining AFB1 in food.

A novel dual-channel immunochromatographic strip using up-conversion nanoparticles for simultaneous detection of AFB1 and ZEN in maize

Chen Yiyi,Shen Yanghong,Wang Haifeng,Zhang Jinzhi,Zhu Junli

Due to universal contamination and synergistic toxicity of multiple mycotoxins in foodstuff, reliable and high-throughput detection methods for multiple mycotoxins are urgently needed in corn products. In this study, a novel dual-channel immunochromatographic assay (ICA) based on improved up-conversion nanoparticles (IUCNPs) was developed for rapidly detecting aflatoxin B1 (AFB1) and zearalenone (ZEN). The synthesized IUCNPs doped by 30% Lu 3+ showed a larger size, more regular structure, and brighter fluorescence intensity than conventional UCNPs. The limits of detection (LODs) of single-channel ICA test strips for AFB1 and ZEN detection were 0.01 and 0.1 ng/mL, respectively. After the optimization, the dual-channel ICA of AFB1 and ZEN in 10 min was conducted, resulting in low detection limits of 0.025 and 0.1 ng/mL, respectively. Moreover, the built assay was revealed to be highly specific for six other food-contaminated mycotoxins, and exhibited excellent accuracy, with corresponding R 2 of 0.9931 and 0.9982 in calibration curves, respectively. Long-term storage experiments indicated that the dual-channel test strips had superior stability and precision. The LODs of AFB1 and ZEN in spiked maize were 0.025 and 0.25 μg/kg, demonstrating great sensitivity and matrix tolerance. Furthermore, the IUNCP-ICA was validated by high-performance liquid chromatography (HPLC) analyses, and a satisfactory consistency was obtained in 15 natural maize samples. Thus, the IUCNPs-ICA proposed in this work realized rapid and sensitive detection of AFB1 and ZEN, providing broad application potential in on-site screening for multiple mycotoxins in agricultural products. A Time-Resolved Fluorescent Lateral Flow Immunoassay for the Rapid and Ultra-Sensitive Detection of AFB1 in Peanuts and Maize

Yunrui Xing,Suzhen Yang,Lu Fan,Xiaofei Hu,Shengnan Liu,Yao Wang,Yaning Sun

Aflatoxin B1(AFB1), a highly toxic and carcinogenic mycotoxin, poses significant public health risks due to its widespread contamination of staple food crops such as peanuts and maize. Although conventional lateral flow immunoassays (LFIAs) are widely employed for rapid on-site screening, their limited sensitivity frequently compromises accurate quantification at trace levels. To improve the analytical performance of LFIAs, we developed a novel time-resolved fluorescence-based lateral flow immunoassay (TRFN-LFIA) by integrating reverse artificial antigen labeling with time-resolved fluorescence signal amplification. This method enhances detection sensitivity and enables rapid, ultra-sensitive, visible, and quantitative determination of AFB1in peanut and maize samples. Under optimized conditions, the TRFN-LFIA achieved a visible limit of detection (vLOD) of 0.30 ng/mL (2.22 µg/kg), a quantitative limit of detection (qLOD) of 0.04 ng/mL (0.30 μg/kg), and a half-maximal inhibitory concentration (IC50) of 0.09 ng/mL. Recoveries from spiked peanut and maize samples ranged from 81.33% to 117.86%, with coefficients of variation (CVs) below 13.04%. Analysis of 21 real samples (13 maize and 8 peanut samples) yielded results highly consistent with those obtained by liquid chromatography–tandem mass spectrometry (LC-MS/MS). Moreover, the method demonstrates significant advantages in terms of detection speed, cost-effectiveness, and operational convenience. Therefore, the results established the TRFN-LFIA method as a reliable and practical tool for on-site rapid detection of AFB1in contaminated food matrices, providing both a rapid and accurate approach for trace-level quantification and a novel strategy for enhancing the sensitivity of lateral flow immunoassays.

Quantitative prediction of AFB1 in various types of edible oil based on absorption, scattering and fluorescence signals at dual wavelengths

Xueming He,Jie You,Xiaoyun Yang,Longwen Li,Fei Shen,Liu Wang,Peng Li,Yong Fang

This study aims to address the challenge of matrix interference of various types of edible oils on intrinsic fluorescence of aflatoxin B 1 (AFB 1 ) by developing a novel solution. Considering the fluorescence internal filtering effect, the absorption ( μ a ) and reduced scattering ( μ’ s ) coefficients at dual wavelengths (excitation: 375 nm, emission: 450 nm) were obtained by using integrating sphere technique, and were used to improve the quantitative prediction results for AFB 1 contents in six different kinds of edible oils. A research process of “Monte Carlo (MC) simulation - phantom verification - actual sample validation” was conducted. The MC simulation was used to determine interference rule and correction parameters for fluorescence, the results indicated that the escaped fluorescence flux nonlinearly decreased with the μ a , μ’ s at emission wavelength ( μ a,em , μ’ s,em ) and μ a at excitation wavelength ( μ a,ex ), however increased with the μ’ s at excitation wavelength ( μ’ s,ex ). And the required optical parameters to eliminate the interference of matrix on fluorescence intensity are: effective attenuation coefficients at excitation and emission wavelengths ( μ eff,ex , μ eff,em ) and μ’ s,ex . Phantom verification was conducted to explore the feasibility of fluorescence correction based on the identified parameters by MC simulation, and determine the optimal machine learning method. The modelling results showed that least squares support vector regression (LSSVR) model could reach the best performance. Three kinds of edible oil (peanut, rapeseed, corn), each with two brands were used to prepare oil samples with different AFB 1 contamination. The LSSVR model for AFB 1 based on μ eff,ex , μ eff,em , μ’ s,ex and fluorescence intensity at 450 nm was calibrated, both correlation coefficients for calibration ( R c ) and the validation ( R v ) sets could reach 1.000, root mean square errors for calibration ( RMSEC ) and the validation ( RMSEV ) sets were as low as 0.038 and 0.099 respectively. This study proposed a novel method which is based solely on the absorption, scattering, and fluorescence characteristics at excitation and emission wavelengths to achieve accurate prediction of AFB 1 content in different types of vegetable oils.

Covalent Organic Framework @ Magnetic Carbon Nanotube Composite Rapidly Prepared by Mechanochemical Method for Sensitive Determination of AFB1 in Real Sample

Liu Tao,Shu Jiaqi,Xiao Jiajun,Zhou Jing,Chen Zhenhua,Yu Qi,Lu Limin,Chen Hui

Aflatoxin B 1 (AFB 1 ), a potent hepatocarcinogen, continually poses a serious threat to food safety and public health owing to its stability and propensity for bioaccumulation within the food chain. Monitoring its trace levels across various food matrices remains challenging, necessitating highly efficient and selective sample pretreatment materials. In this study, a novel covalent organic framework-based magnetic composite (3D-N/OCNS@TpPa-1) was rapidly synthesized via a solvent-free mechanochemical method within 30 min. The mechanochemical grinding method enabled not only the rapid and green synthesis of the COF but also its immediate integration with a carbon matrix. This composite strategy was specifically designed to mitigate the common challenges of COF aggregation and poor dispersibility. When employed as a solid-phase extraction sorbent, it demonstrated rapid adsorption/desorption kinetics (10 min and 8 min, respectively) and high affinity for AFB 1 . Coupled with HPLC-FLD, the method enabled the sensitive detection of AFB 1 across a complete food chain transmission pathway—corn, corn oil, chicken, and human plasma—with a wide linear range (0.25–250 μg·kg⁻ 1 ), a low detection limit (10 ng·kg⁻ 1 ), and high precision. The study not only provides a robust and green analytical approach for tracing AFB 1 propagation in agri-food systems but also highlights critical control points for intervention in food safety management.

A Novel Competitive Aptamer Biosensor Based on Functional Nanomaterials for the Detection of Aflatoxin B1 (AFB1) in Rice and Corn

Xinran Yang,Chuanjin Cui,Hongshuo Chen

Developing a reliable detection method for aflatoxin B 1 (AFB 1 ) is urgent due to its potent hepatocarcinogenic effects. An electrochemical aptamer sensor based on competitive binding was prepared for the rapid detection of AFB 1 in this study. Poly(amideamine) dendrimers were electrodeposited onto a screen-printed gold electrode to enhance aptamer loading via amine group conjugation. The chemically synthesized gold nanostars can further improve electron transfer kinetics and provide additional binding sites for single-stranded DNA probes. Bioprobe and modified electrode were comprehensively characterized by transmission electron microscope, scanning electron microscopy, Zeta potential, energy dispersive spectroscopy, selected area electron diffraction, X-ray photoelectron spectroscopy, and atomic force microscope. Differential pulse voltammetry (DPV) and square wave voltammetry (SWV) of pulse voltametric techniques were executed separately in AFB1 solutions for testing the performance of this prepared biosensor. Comparative analysis revealed that both methods exhibited a linear detection range of 10 −4 to 10 3 ng/mL for AFB 1 . Nevertheless, the DPV method achieved a lower limit of 2.25 × 10 −5 ng/mL than the SWV method, with a detection limit of 3.18 × 10 −5 ng/mL. Meanwhile, the developed biosensor showed excellent stability and reproducibility, establishing its reliability for AFB 1 quantification in complex matrices. Graphical The addition of aflatoxin B1 (AFB1) causes competitive binding, leading to the detachment of the probe AuNs@ssDNA from the AuNs@ssDNA/Aptamer/PAMAM/SPGE sensing platform, thereby altering the electron transfer rate on the electrode surface. The attenuation current of the [Fe(CN)6] 3 -/ 4 - redox signal is monitored via differential pulse voltammetry and square wave voltammetry, enabling the quantitative determination of AFB1.

Lipid droplet-organelle communication: A new perspective on AFB1-induced lipotoxicity in chicken hepatocytes and the protective effect of curcumin

Sihong Li,Jun Jiang,Qian Wu,Muhammad Ishfaq,Jiaqing Cheng,Lu Liu,Dong Niu

Aflatoxin contamination in food and feed is prevalent worldwide and exposure to Aflatoxin B1 (AFB1) can lead to significant lipid accumulation in hepatocytes. However, the underlying mechanism by which curcumin mitigates AFB1-induced hepatotoxicity in chickens remains unclear. In this study, primary chicken embryonic hepatocytes (CEHs) were selected as the experimental model and divided into 1) control, 2) AFB1, 3) AFB1+Cur, and 4) curcumin alone group. AFB1 dose-dependently reduced cell viability, whereas curcumin enhanced it. AFB1 induced severe lipid droplets (LDs) accumulation (confirmed by staining and electron microscopy) and disrupted cellular structures. Transcriptome and proteome analyses further demonstrated that AFB1-induced lipotoxicity both in vivo and in vitro . In contrast, curcumin reversed these effects. Under AFB1 exposure, a significant up-regulation of fatty acid synthesis genes (PTGS2, FAS) and down-regulation of the fatty acid metabolism related gene CPT1A in CEHs were observed. AFB1 also impaired autophagy (decreased p62, increased LC3B/ATG9a), damaged lysosomes (downregulated NPC1/LAMP1, upregulated mTOR/LGALS1a), altered mitochondrial dynamics (upregulated DRP1/FIS1/MFN1, downregulated MFN2), and activated necroptosis (upregulated RIPK1/RIPK3/MLKL). Critically, curcumin co-treatment (AFB1+Cur) reversed these detrimental effects. Furthermore, AFB1+Cur significantly decreased LDs-mitochondria colocalization and increased LDs-lysosome colocalization compared to AFB1 alone group. This study suggests that the changes in lipid metabolism, oxidative stress and necroptosis caused by lipid drop-organelle communication may be a new perspective on AFB1-induced lipotoxicity in chicken hepatocytes and curcumin offers a potential treatment for protecting hepatocytes from lipotoxicity. Future directions should include chicken feeding trials assessing curcumin's ability to AFB1-induced hepatic steatosis and necroptosis under real-world exposure levels.
