Effects of Staged Fermentation With Multistrains on the Physicochemical Characteristics and In Vitro Inhibitory Activity on Advanced Glycation End-Products (AGEs) Formation of Buckwheat

Duqin Zhang,Zhujiang Shen

To utilize various physiological and biochemical properties of multistrains, staged fermentation was carried out on buckwheat dehulled seed (BDS) and whole seed (BWS) with Aspergillus oryzae , Rhizopus oryzae , and Bacillus subtilis , Saccharomyces cerevisiae , and Lactobacillus plantarum . Effects of staged fermentation on physicochemical characteristics and in vitro inhibitory activity on advanced glycation end-products (AGEs) formation of BDS and BWS, as well as their correlations were evaluated. BWS was dehulled to prepare BDS, however, they showed quite different results. The crystallinity of BDS increased from 3.10% to 5.59%, which decreased from 6.87% to 6.15% in BWS after fermentation. Breakdown viscosity was only detected in fermented BWS according to rapid visco-analyzer (RVA) results, suggesting a higher fermentation level extended into endosperm compared to BDS. Staged fermented BWS showed higher and lower in vitro inhibitory activities on AGEs formation in bovine serum albumin/glucose (BSA/glucose) and BSA/MGO systems, respectively, compared to one-time fermentation with mixed strains. This indicated that the ability of BWS in inhibiting AGEs formation at middle stage decreased, but it recovered to maximum at final stage. Correlation analysis showed protein and flavonoid contents played major roles in inhibition of AGE formation in BDS, whereas soluble dietary fiber (SDF), flavonoid, and phenolic contents were more effective for BWS.

The binding mechanism of oat phenolic acid to whey protein and its inhibition mechanism against AGEs as revealed using spectroscopy, chromatography and molecular docking

Lezhen Dong,Yunzhen Zhang,Ying Li,Yahui Liu,Qin Chen,Lingyi Liu,Mohamed Farag,Lianliang Liu

Heat sterilization of dairy products can promote the formation of advanced glycation end products (AGEs), protein oxidation products (POPs) and α-dicarbonyl compounds, which have a significant influence on health due to the close association of these products with diabetes complications. In this study, eight oat phenolic acids were first analyzed for their inhibitory effect against AGEs formation. Due to their strong inhibitory effects and structural differences, caffeic acid (CA) and gallic acid (GA) were further selected to assess their anti-glycosylation mechanisms using spectroscopy, chromatography and molecular docking. CA/GA reduced the production of total AGEs and POPs in various bovine milk simulation models and protected whey proteins from structural modifications, oxidation, and cross-linking. Comparative analyses showed a structure–effect relationship between CA/GA and AGEs inhibition. Oat phenolic acids against AGEs and POPs might be related to the unique bonding of key amino acid residues in whey proteins, the inhibitory role of early fructosamine and the trapping of reactive α-dicarbonyl groups to form adducts. In conclusion, oat phenolic acids might present a promising dietary strategy to alleviate AGEs production and glycation of proteins in dairy products upon storage.

Catalytic elevation effect of methylglyoxal on invertase and characterization of MGO modification products

Yixin Li,Zitong Wang,Jiaqi Wu,Jie Zheng,Fu Liu,Juanying Ou,Caihuan Huang,Shiyi Ou

Reactive carbonyl species can modify digestive enzymes upon intake due to their electrophilic nature. This study evaluated the effects of methylglyoxal (MGO), glyoxal, acrolein, and formaldehyde on invertase, an enzyme presents in digestive tract. Unexpectedly, MGO enhanced, rather than inhibited, invertase activity. Moreover, MGO counteracted the inhibitory effects of the other three carbonyls on invertase activity. Kinetic analyses revealed that 150 mmolLexp.-1 MGO resulted in a 2-fold increase in the Km and a 3.3-fold increase in Vmax, indicating that MGO increased the turnover rate of sucrose while reducing the substrate binding affinity of invertase. Additionally, MGO induced dynamic quenching of fluorescence, reduced free amino groups, increased hydrophobicity, the content of Amadori products, fluorescent and nonfluorescent AGEs, and amyloid fibrils of invertase. The specific modifications responsible for the elevated activity of MGO on invertase require further investigation.

Screening of active components of melastoma dodecandrum lour. against diabetic osteoporosis using cell membrane chromatography-mass spectrometry

Xiaoqin Zhang,Jiale Mao,Lu Shao,Shuang Liu,Jiwang Zhou,Mingrong Mei,Zunjing Zhang

Background: Melastoma dodecandrum Lour. (MD), a traditional botanical drug known for its hypoglycemic, antioxidant, and anti-inflammatory properties, is commonly used to treat diabetes, osteoarthritis, and osteoporosis. However, its specific active components against diabetic osteoporosis remain unclear.Purpose: This study aimed to identify the key active components in MD using cell membrane chromatography coupled with mass spectrometry and validate their effects in vitro. Methods: An AGEs-induced osteoblast injury model was established. MTT assays measured cell viability, and ALP activity was assessed using a biochemical kit. Western blotting was employed to detect the expression levels of osteoblast-related proteins OCN and RUNX2 and the AGE receptor protein RAGE. ELISA was used to determine the levels of SOD, MDA, CAT, and GPx. PCR quantified TNF-α expression to evaluate the protective effects and potential mechanisms of MD. The AGEs-induced osteoblast cell membrane chromatography-mass spectrometry method facilitated the rapid identification of potentially active compounds based on their affinity for the osteoblast cell membrane. Cell experiments further validated the activity of the characteristic component isovitexin. Results: MD significantly improved cell viability in AGEs-damaged osteoblasts, enhanced ALP, SOD, CAT, and GPx activities, reduced MDA levels, increased OCN and RUNX2 protein expression, and decreased TNF-α mRNA and RAGE protein expression. Cell membrane chromatography identified 20 chemical constituents, including 13 flavonoids, 4 organic acids, 1 phenylpropanoids, 1 terpenoids, and 1 alkaloid. Cell experiments have confirmed that isovitexin has significant protective activity against osteoblasts and can inhibit the expression of specific receptor RAGE on the osteoblast membrane, consistent with the effect of MD.Conclusion: MD and its active component, isovitexin, provide protective effects against AGEs-induced osteoblast injury, offering a basis for subsequent drug development.

Metabolomic Profiling Reveals the Quality Variations in Citri Reticulatae Pericarpium (Citrus reticulata Blanco cv. Chachiensis) with Different Storage Ages in Response to “Candidatus Liberibacter Asiaticus” Infection

Jiayin Liang,Yuqing Xi,Jiaming Li,Shugui Xu,Yongqin Zheng,Meirong Xu,Zheng Zheng,Xiaoling Deng

Citri ReticulataePericarpium, especially the pericarp ofCitrus reticulataBlanco cv. Chachiensis (PCRC), is an important edible and medicinal ingredient for health and pharmacological properties. Citrus Huanglongbing, a devastating disease that currently threatens the citrus industry worldwide, is caused by a phloem-limited alpha-proteobacterium, “CandidatusLiberibacter asiaticus” (CLas). The industry of cultivar Chachiensis has been suffering from HLB. Although HLB affected the quality of citrus fruit, whether the quality of PCRC was affected by HLB remains unclear. In this study, we compared the metabolite profiles between HLB-affected and healthy PCRC from three sources: fresh, 6-month-old, and 9-year-old PCRC, through the untargeted LC–MS method. Compared to healthy controls, various types of bioactive compounds, mainly flavonoids, terpenoids, alkaloids, coumarins, polysaccharides, and phenolic acids, accumulated in HLB-affected PCRC, especially in the HLB-affected 9-year PCRC. In particular, isorhamnetin, isoliquiritigenin, luteolin 7-O-beta-D-glucoside, limonin, geniposide, pyrimidodiazepine, scoparone, chitobiose, m-coumaric acid, malonate, and pantothenic acid, which contributed to the pharmacological activity and health care effects of PCRC, were highly accumulated in HLB-affected 9-year-old PCRC compared to the healthy control. Multibioassay analyses revealed that HLB-affected 9-year-old PCRC had a higher content of total flavonoids and total polyphenols and exhibited similar antioxidant capacity as compared to healthy controls. The results of this study provided detailed information on the quality of HLB-affected PCRC.

Resina Draconis Promotes Diabetic Wound Healing by Regulating the AGE-RAGE Pathway to Modulate Macrophage Polarization

Xin Jin,Ang Li,Zhaoyuan Dai,Yi Li,Xinchi Feng,Feng Qiu

Resina Draconis (RD), a traditional Chinese medicine, has been widely used in treating diabetic foot ulcers. However, its specific mechanisms of action remain incompletely understood. First, network pharmacology combined with GEO clinical sample data mining was employed to systematically analyze the therapeutic targets of RD in promoting diabetic wound healing. Second, an AGEs-induced RAW264.7 cell model was utilized to investigate the regulatory effects of RD and its primary active components on the AGE-RAGE signaling pathway, along with their anti-inflammatory and antioxidant activities. Finally, a diabetic wound mouse model was established to validate the efficacy of RD and further explore its underlying molecular mechanisms. Integrated analysis of network pharmacology and GEO database mining identified 492 potential therapeutic targets of RD in diabetic wound healing, primarily involving the AGE-RAGE pathway. In vitro, RD (6.25 μg/mL) significantly suppressed AGE-induced inflammatory factors and ROS production while downregulating AGE-triggered RAGE protein overexpression. In vivo, RD hydrogel accelerated diabetic wound healing by modulating the AGE-RAGE axis and regulating macrophage polarization. RD effectively promotes diabetic wound healing through synergistic regulation of the AGE-RAGE pathway, oxidative stress suppression, and macrophage polarization modulation, providing a novel therapeutic strategy for diabetic wound management.

Selective transportation and energy homeostasis regulation of dietary advanced glycation end-products in human intestinal Caco-2 cells

Yi Wu,Minhua Zong,Zhenhui Zhang,Yongtai Wu,Lin Li,Xia Zhang,Hong Wu,Bing Li

Advanced glycation end-products (AGEs) are a chemically heterogeneous set of modifications widely found in processed foods. Due to uncertain bioavailability, dietary AGEs regulate energy homeostasis through mechanisms that largely remain unclear. In this study, selective transmembrane transport of AGEs with different modification types from glycated β-casein digest were identified and compared. The results showed that only a few types of free and peptide-bound AGEs can easily cross the Caco-2 monolayers and thus exert their effects. A combination of biochemical assays, mitochondrial analyses, and comparative experiments identified that the effect of AGEs on cellular energy homeostasis comes mainly from their free fractions. Mechanistically, free AGEs arrest the mitochondrial differentiation and mtDNA repair by intervening in the function of thymidine phosphorylase, and interfering with mitochondrial energy production by inhibiting the activation of AMPK-SIRT6 signaling pathway. These results demonstrate mechanisms by which processed foods cause mitochondrial dysfunction and lead to dysfunctional energy homeostasis.

Fabrication of conducting polymer/noble metal nanocomposite modified electrodes for glucose, ascorbic acid and tyrosine detection and its application to identify the marked ages of rice wines

Zhenbo Wei,Yana Yang,Xize Xiao,Weilin Zhang,Jun Wang

In previous studies, conductive polymers (CPs)/noble metal nanoparticles (NMNPs) composite materials modified electrodes were always applied to distinguish the trace amounts of specific analytes in complex liquid mixtures. In this paper, polymer sulfanilic acid (PABSA)/AuNPs/glassy carbon electrode (GCE), polymer acid chrome blue K (PACBK)/AuNPs/GCE and polymer aspartic acid (PASP)/PtNPs/GCE were fabricated for the identification of the wine age of rice wines with pattern recognitions. The sensitivity of those modified electrodes was exhibited by cyclic voltammetry, and the parameters of electrochemical behaviors was optimized and confirmed gradually. The original responses were recorded by chronoamperometry with multi-frequency rectangle pulse voltammetry and multi-frequency staircase pulse voltammetry, and the feature data correlated with the wine age were extracted from original responses by ‘area method’. Based on the feature data, principal component analysis (PCA, unsupervised method), locality preserving projections (LPP, semi-supervised method) and differential financial analysis (DFA, supervised method) were applied for the classification of rice wine samples with different marked age, and DFA exhibited the most clear result; least squares support vector machines (LSSVM) and library for support vector machines (LIBSVM) were applied for the prediction of the wine ages, and LIBSVM worked better than LSSVM, the correlations based on the training and testing dataset were R 2   =   0.9999 and R 2   =   0.9998, respectively. In conclusion, the CPs/NMNPs/GCEs with pattern recognitions were powerful tools to identify the marked ages of rice wines.

Inhibitory effect of caffeic acid and dihydrocaffeic acid on the formation of advanced glycation end products: Mechanism analysis based on intermolecular interaction

Yanqing Zhang,Lixia Yuan,Qiulu Wang,Xuejie Zhang,Yiyun Yang,Min Liu

The accumulation of advanced glycation end products (AGEs) generated from glycation reaction accelerates the development and deterioration of some diseases. Herein, the AGEs model of fructose-induced glycation of human serum albumin (HSA) was constructed and characterized. Both caffeic acid (CA) and dihydrocaffeic acid (DHCA) can inhibit the formation of AGEs, and the inhibitory effect of CA was more pronounced, as confirmed by fluorescence spectroscopy, gel electrophoresis, scanning electron microscopy, and carbonyl and thiol contents analysis. The binding of CA and DHCA to HSA was investigated by fluorescence spectroscopy and molecular docking. The primary inhibitory mechanism was the occupation of the glycation site LYS199 of HSA, and the more significant inhibitory effect of CA was related to its stronger binding affinity. Furthermore, the conformational changes caused by glycation and CA/DHCA binding were monitored using fluorescence, circular dichroism, and dynamic light scattering analysis. The results would help to elucidate the inhibitory mechanism of CA and DHCA on AGEs formation based on their different structures and inhibitory effects, and provide guidance for choosing natural inhibitors for diabetes.

Ethanol as an accelerator for the formation of advanced glycation end products in glucose-lysine solution

Qian Wu,Kuoquan Zhao,Yuanyuan Chen,Jinsong Xiao,Mengzhou Zhou,Dongshen Li,Nianjie Feng,Chao Wang

In this study, a glucose-lysine (G-L) model was selected to simulate the dietary system (cooking of alcohol-cured steak) to study the effect of alcohol concentration on the formation of advanced glycosylation products (AGEs) in Maillard reaction . It was first reported that higher alcohol concentrations accelerated the formation of AGEs as well as their intermediates glyoxal (GO) and methylglyoxal (MGO). The contents of GO and MGO increased from 11.74 ± 2.55 to 50.18 ± 0.16 μg/mL and 19.04 ± 1.25 to 58.51 ± 2.56 μg/mL with increasing ethanol concentration, respectively. High-alcohol samples produced higher levels of Nε-(carboxymethyl) lysine (CML) and Nɛ-carboxyethyl lysine (CEL) detected by U-HPLC-MS/MS. The capacity of catechin to inhibit the formation of AGEs and intermediates were significantly related to the concentration of ethanol, which may be due to ethanol accelerated catechin degradation and weakened the ability of catechin to trap GO and MGO. Our study strongly suggests that although we cannot completely avoid the AGEs produced by the Maillard reaction , in daily life, we try to avoid adding alcohol, especially high-concentration alcohol, when cooking and processing ingredients.

Ruichun Wang,Songyi Lin,Junbo Chu,Yi Yuan,Simin Zhang

Eleutheronema Tetradactylum is commercially important marine fish and a fixture at tables. However, nonnegligible hazardous substances, such as heterocyclic aromatic amines (HAAs) and advanced glycation end products (AGEs), generates along during thermal processing. Herein, we determined HAAs and two representative AGEs, Nε-carboxyethyl-lysine (CEL) and Nε-carboxymethyl-lysine (CML) formation in roasted Eleutheronema Tetradactylum , and proposed inhibitory method of tea polyphenol to effectively reduce HAAs, CML and CEL, which caused 51.23, 46.72, 24.06 and 35.48 percentage reduction of Norharman, Harman, CML and CEL, respectively, led IQ, 8-MeIQx below detected limitation. Precursors (amino acids, creatin(in)e, glucose and glyoxal), lipid oxidation, fatty acid analysis and further nontargeted lipidomic approaches are attempted to reveal the inhibition mechanism. This work develops a new method to minimize HAAs and AGEs in Eleutheronema Tetradactylum , provide a comprehensive interpretation of inhibition mechanism by tea polyphenols, which is helpful to food manufacturers for healthier fish production.

Effect of superheated steam on Maillard reaction products, digestibility, and antioxidant activity in β-Lactoglobulin-glucose system

Wen-mei Chen,Yang Wang,Xu-mei Wang,Yan-hong Shao,Zong-cai Tu,Jun Liu

This study analyzes the interactions among Maillard reaction products (MRPs), digestibility and antioxidant activity in a β-lactoglobulin-glucose (βlg-Glu) model system during superheated steam treatment at 120 °C for 1 to 5 min. With an increase in treatment duration, there were significant increases in glucose loss, grafting degree, browning, and the formation of intermediate products in βlg-Glu. Characteristic MRPs, including α-dicarbonyl compounds, 5-hydroxymethylfurfural, and advanced glycation end products (AGEs), were formed through the degradation of sugars and condensation reactions between carbonyls and amines, accompanied by an increase in oxidative products. These changes impacted the molecular weight distribution and conformational structure of βlg-Glu, resulting in decreased digestibility. βLg-Glu with the highest level of glycation exhibited superior antioxidant activity after in vitro digestion, which was closely associated with the increase in AGEs and oxidation products. Therefore, the protein-sugar system treated by superheated steam with different heating time can significantly affect the formation and activity of the products, providing theoretical basis for superheated steam processing nutritious and healthy food.
