FAM134B-mediated ER-phagy degrades APP and suppresses Alzheimer’s disease pathology

Zhang Yuting,Sun Jun,Cai Yang,Xu Ziyan,Li Xiang,Wei Wei,Liu Pai,Sun Qiming,Wang Zhi-Hao,Cui Yixian

Endoplasmic reticulum autophagy (ER-phagy) is a selective autophagy pathway in which receptor proteins target ER membranes and proteins for degradation, yet its role in Alzheimer’s disease (AD) remains unclear. Here, we identify FAM134B/RETREG1 as a specific ER-phagy receptor mediating amyloid precursor protein (APP) degradation. FAM134B directly interacts with ER-localized wild-type and familial mutant APP via their C-terminal domains and recruits LC3 through its LC3-interacting region (LIR) to promote APP delivery to phagophores for lysosomal degradation. In AD, epigenetic silencing at the FAM134B promoter suppresses its transcription by limiting TFEB/TFE3 binding despite their nuclear enrichment. This transcriptional suppression impairs ER-phagy, leading to APP accumulation and exacerbated AD pathology. AAV-mediated hippocampal expression of wild-type, but not LIR-mutant, FAM134B in 5XFAD mice restores ER-phagy, enhances APP clearance, reduces Aβ deposition, preserves synaptic and myelin integrity, and improves cognitive performance. These findings establish FAM134B downregulation as an upstream pathogenic event in AD, suggesting ER-phagy enhancement as a promising strategy to suppress Aβ generation at its source.

Air-processed MAPbI3 perovskite solar cells achieve 20.87% efficiency and excellent bending resistance enabled via a polymer dual-passivation strategy

Pengyun Zhang,Jie Xiong,Wei-Hsiang Chen,Pingfan Du,Lixin Song

In recent years, air-processed MAPbI3 perovskite solar cells (PSCs) have attracted widespread interest from researchers worldwide because of their simple and low-cost fabrication process. Nonetheless, the ambient conditions usually bring about many adverse effects, such as imperfect crystallization and numerous defects in perovskite films, which seriously impact both the photoelectric performance and stability of the device. Therefore, in this work, a polymer dual-passivation strategy was employed by introducing ammonium polyphosphate (APP) as an additive to the green anti-solvent to accurately modify the perovskite layer. APP, which has abundant phosphate and ammonium groups, could simultaneously fill the I/Pb vacancies by Lewis acid–base reactions to restrain defect formation and improve the power conversion efficiency (PCE) of the ultimate device. On the other hand, the long molecular chains of the polymer with a certain flexural ability were easily congregated at the grain boundaries of the perovskite grains, thus enhancing the bending resistance. Consequently, high-quality perovskite films with a dense morphology and large grain size were obtained. Because of the reduced defect density and suppressed non-radiative recombination, the optimal PSC attained a champion PCE of 20.87% with negligible hysteresis. Furthermore, the non-encapsulated APP-modified flexible device also exhibited excellent bending resistance. Only 20% of its normalized PCE was lost after 150 bending cycles at room temperature. This simple, green, low-cost, and reliable strategy for preparing high-efficiency PSCs with good stability can facilitate its commercialization.

Sennoside A alleviating cognitive impairment in APP/PS1 mice via balancing microbiome metabolism

Yulian Shi,Huilin Ma,Huajinzi Li,Yingyue Wang,Chaofan Wang,Na Zhang,Gan Luo,Ying Wang,Xiaoyan Gao

BackgroundThe progression of Alzheimer's disease (AD) is associated with constipation, potentially mediated by gut microbiota. Laxatives have shown potential in improving the cognitive function of AD, but the specific mechanism remains underexplored. Sennoside A (SA), a well-established laxative, is commonly used for treating constipation.ObjectiveThis work used SA as a probe to explore the therapeutic effects and potential mechanisms of laxatives on AD via the gut-brain axis.MethodsFollowing a two-month treatment, behavioral experiments were used to assess the cognitive function. The central pathologies and neuroinflammation were evaluated by histopathology and ELISA. 16S rRNA sequencing, fecal microbiota transplantation and antibiotic treatment were conducted to verify whether SA exerts anti-AD effects via gut microbiota. Further, non-targeted metabolomics coupled with Spearman correlation analysis was employed to elucidate the underlying mechanisms.ResultsSA significantly countered cognitive dysfunction and central pathological damage in APP/PS1 mice. Besides, SA ameliorated gut dysbiosis and affected the metabolic functions of the flora. Furthermore, the therapeutic effects of SA decrease with the depletion of gut microbes and could be transferred with the microbiota. Intriguingly, amino acid metabolism and aminoacyl-tRNA biosynthesis were the main metabolic pathways regulated by SA, consistent with the predicted functions of gut bacteria. Finally, correlation analysis revealed a strong correlation between gut microbes, fecal metabolites, and cognitive ability affected by SA.ConclusionsThe study investigated the efficacy and mechanisms of laxatives represented by SA for AD from the perspective of the gut-brain axis.

Music intervention as a therapeutic approach to cognitive impairments in APP/PS1 mice

Xueshan Bu,Ya Yuan,Wenqin Song,Wei Wang

Music-based intervention ameliorates cognitive deficits in APP/PS1 mice • Music intervention attenuates oxidative stress and neuroinflammation in APP/PS1 mice • Music intervention mitigates synaptic impairment and neuronal death in APP/PS1 mice Alzheimer's disease (AD) is typified by progressive deterioration in memory and executive capacities. Although music-based interventions represent a promising non-pharmacological strategy for AD, the underlying mechanisms remain insufficiently understood. APP/PS1 transgenic mice were subjected to 30 consecutive days of musical stimulation, with wild-type C57BL/6 J and untreated APP/PS1 mice serving as controls. Neurobehavioral assessments revealed that music intervention substantially improved memory performance in transgenic AD mice. Mechanistically, music attenuated oxidative stress in both serum and hippocampus of APP/PS1 mice, as evidenced by decreased nitric oxide (NO) and malondialdehyde (MDA) levels along with enhanced antioxidant activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT), and total antioxidant capacity (T-AOC). Peripheral and hippocampal inflammatory responses were also effectively inhibited following musical treatment. Music restored synaptic plasticity–related protein expression, increased postsynaptic density thickness, narrowed synaptic cleft, and elevated dendritic spine density. Additionally, music exposure alleviated Aβ deposition and neuronal loss in APP/PS1 mice. These findings indicate that music-based auditory enrichment exerts neuroprotective effects against AD through synergistically modulating oxidative stress, neuroinflammation, synaptic plasticity, and neuronal survival, providing mechanistic insights into its intervention potential for AD.

Inhibition effect and mechanism of APP/ZB composite suppressant on coal dust explosion

Hao Liu,Chengjun Xiao,Yansong Zhang,Runzhi Li,Xiangbao Meng

APP/ZB composite suppressant prepared via mechanochemical treatment (1:1). • Pmax reduced by 90.7% and (dp/dt)max by 88.3% at 30 wt% addition. • Flame propagation distance and velocity reduced by 47.5% and 62.5%. • APP/ZB decomposes earlier (256.1 °C) with higher heat absorption (65.6 J/g). • P–B–Zn inorganic barrier layer formed on coal particle surface. • Synergistic inhibition via endothermic cooling, dilution, char promotion, and barrier. To enhance coal dust explosion suppression, an APP/ZB composite suppressant (1:1 mass ratio) was prepared via mechanochemical treatment. Its performance was evaluated using a 20 L explosion sphere and a transparent pipe system. Mechanochemical treatment promoted adhesion of ZB fines onto APP surfaces, improving interfacial contact. APP/ZB exhibited a lower initial decomposition temperature (256.1 °C) than the individual components, while its heat absorption (65.6 J/g) was approximately 9.61% higher than the theoretical value (59.85 J/g) calculated for a simple 1:1 physical mixture of APP and ZB. At 30 wt% addition, the maximum explosion pressure decreased by 90.7% (from 0.54 to 0.05 MPa), the maximum pressure rise rate by 88.3% (from 46.2 to 5.4 MPa/s), and K St from 1.25 to 0.14 MPa·m/s; induction and combustion times were significantly prolonged. At 20 wt%, flame propagation distance and velocity dropped by 47.5% and 62.5%, respectively, with the flame front becoming discontinuous. Residue characterization revealed that polyphosphates from APP crosslinked with boron- and zinc-species from ZB, forming a dense inorganic protective layer (P O P, Zn O P, Zn O B) on coal particle surfaces. APP/ZB acts synergistically through early endothermic decomposition, inert gas dilution, char promotion, and a P B Zn barrier that restricts heat, oxygen, and volatile transfer, interrupting the pyrolysis–combustion feedback loop. These findings offer a useful reference for designing phosphorus‑nitrogen/borate composite suppressants for coal dust explosions. A novel optical strategy for ALP detection and development of matched smartphone app

Li Jiawei,Cheng Yi,Huang Yingchun,Xu Lixin,Zhang Kun,Yan Fei,Chen Jian,Lang Chunhui

Alkaline phosphatase (ALP) is a reliable biomarker for various diseases, making the development of new detection methods highly significant. Herein, we report a simple and accurate dual-signal optical detection strategy for measuring ALP activity across the entire clinical range in adults. Based on this, we designed a smartphone app to eliminate the limitations of large instruments, enabling real-time field detection. In this work, ALP catalyzes the conversion of the phosphate monoester of 2-phospho-L-ascorbic acid (PAA) into ascorbic acid (AA), a compound with strong reducing properties. AA facilitates the in-situ reduction of Ag + to form Au@Ag core-shell nanoparticles, accompanied by significant changes in the ultraviolet-visible absorption spectra and color. These changes allow for the qualitative and quantitative detection of ALP activity. In UV absorption spectroscopy and colorimetric detection, the signal shows good linearity with ALP activity concentrations, with detection ranges of 0 U/L to 200 U/L (R 2 =0.9961) and 25 U/L to 250 U/L (R 2 =0.9771), respectively. Subsequent selective analysis and serum sample analysis demonstrated the high selectivity and accuracy of the proposed method, highlighting its potential for field application.

Preventive effects of xanthohumol in APP/PS1 mice based on multi-omics atlas

Wei Liu,Xiao Chen,Chen Yang,Zequn Lin,Xinfeng Huang,Zhen Zhang,Jianjun Liu

Alzheimer's disease (AD) is a complex disease with unknown etiology and pathogenesis. We described a combined analysis of murine proteomics and microbiomics to find potential therapeutic targets of different doses of xanthohumol (Xn), with the goal of providing a biological basis for the treatment of early AD. Xn improved the spatial learning and memory ability of APP/PS1 mice; this was associated with an increased number of newborn neurons in the subgranular zone (SGZ) and dentate gyrus (DG) and a decreased inflammatory response. 108 proteins were significantly changed after 0.5 mg/kg Xn treatment while only 72 proteins changed by 5 mg/kg Xn. Eight significant microbiota were modulated by different doses of Xn at line discriminant analysis (LDA) score 3.0, but only three of which were regulated by 0.5 mg/kg Xn at LDA score 4.0. In addition, Xn treatment could significantly regulate the pathways of neurodegeneration- multiple diseases in the hippocampus and the penicillin and cephalosporin biosynthesis and atrazine degradation pathways in the gut. Interestingly, Nefl protein validated by correlation analysis was found in the common signaling pathway. 0.5 mg/kg Xn was able to reverse the correlation between hippocampal proteins and gut microbiota. Xn treatment significantly improved cognitive function in AD transgenic mice. Different doses of Xn caused significant differences in protein expression and flora composition and abundance, suggesting that the doses of Xn should be selected with caution, and low dose may be better in the prevention of AD.

Preparation and properties of APP flame-retardant ramie fabric reinforced epoxy resin composites

Xu-Chen Jiang,Ping Li,Yun Liu,Yuan-Wei Yan,Ping Zhu

Ramie fabrics (RFs) as natural fibers have shown promising potential in the field of fiber reinforced composites, but flammability of RFs limits their application in composites. In this study, ammonium polyphosphate (APP) was first coated on the surface of RFs, and then the flame-retardant RF reinforced epoxy resin composites were obtained. When the weight gain rate of APP in RFs was about 20 %, the limiting oxygen index of the flame-retardant composites reached 33.8 %, and achieved UL-94 V-0 rating. At the same time, the peak heat release rate and total heat release values of the flame-retardant composites were reduced by 26 % and 41 % respectively, and the total smoke production was reduced by 43.9 %. In terms of mechanical properties, the presence of APP had a certain negative effect on the internal compatibility of composites. In this work, the flame-retardant composites show excellent flame retardancy and provide a feasible solution to improve the flammability of natural fiber reinforced composites.

Effect of a bio-based copolymer containing lysine, dopamine and triazine on flame retardancy and mechanical properties of thermoplastic polyurethane/ammonium polyphosphate

Yao Xu,Bingtao Wang,Zhenghong Guo,Zhengping Fang,Peng Chen,Juan Li

A bio-based copolymer (TDDL) containing lysine, dopamine, triazine was synthesized using cyanuric chloride, dopamine and lysine, respectively. The TDDL was used as a charring agent and ammonium polyphosphate (APP) was used as an acid source to improve the flame retardancy of thermoplastic polyurethane (TPU). The flame retardancy and thermal degradation behaviors of TPU blends were investigated using UL-94 vertical burning, limiting oxygen index (LOI), cone calorimetry tests and thermal gravimetric analysis (TGA). TPU blend containing 8 wt% APP displays a UL-94 V0 rating and a LOI value 30.0 vol%. While TPU blend containing 6 wt% APP/TDDL displays a UL-94 V0 and a LOI 29.6 vol% suggesting good synergistic effect between TDDL and APP. In addition, TDDL reduces the heat release rate of TPU/APP blend, and promotes the formation of a perfect char layer during the combustion process, and suppresses the release of combustible gas and smoke. Moreover, TPU blends have better modulus and elongation at break than those of TPU.

Self-Assembled Serpentine Ni3Si2O5(OH)4 Hybrid Sheets with Ammonium Polyphosphate for Fire Safety Enhancement of Polylactide Composites

Xiaohong Yi,Jingshu Huang,Yizhang Tong,Hui Zhao,Xianwu Cao,Wei Wu

Biodegradable polylactide (PLA) has been widely utilized in people’s daily lives. In order to improve the fire safety of PLA, ammonium polyphosphate (APP) was self-assembled onto the surface of serpentine Ni3Si2O5(OH)4through the electrostatic method, followed by mixing with PLA by melt compounding. The APP-modified serpentine (serpentine@APP) dispersed uniformly in the PLA matrix. Compared with pure PLA, the PLA composite with 2 wt% serpentine@APP reduced the peak heat release rate (pHRR) and total heat release (THR) by 43.9% and 16.3%, respectively. The combination of APP and serpentine exhibited suitable synergistic flame-retardant effects on the fire safety enhancement of PLA. In addition, the dynamical rheological tests revealed that the presence of APP and serpentine could reduce the viscosity of PLA composites. The plasticizing effects of APP and serpentine benefited the processing of PLA. The mechanical properties of PLA/serpentine@APP maintained suitable performance as pure PLA. This study provided a feasible way to enhance the fire safety of PLA without sacrificing its mechanical properties.

Synthesis of aluminum bis(hydroxy-phenyl-methyl)phosphinate and its synergistic flame retardant mechanism in PLA

Baoming Xu,Jingxue Yang,Xinhui Wang,Na Wang,Guorong Chen,Heng Zhang

In this paper, we report on the synthesis of a new aluminum bis(hydroxy-phenyl-methyl)phosphinate, AlBPi, under mild reaction conditions. The product structure was confirmed via characterization, and its thermal stability and flame retardant properties were assessed. To improve the flame retardant properties, we employed ammonium polyphosphate (APP) as a synergist with AlBPi. The PLA composite material containing 10% AlBPi and 5% APP achieved an LOI of 28.1% and a UL-94 rating of V-1. Cone calorimetry tests showed a significant reduction in PHRR, THR, PSPR, and TSP for the PLA/AlBPi/APP composite compared to the pure and PLA/AlBPi materials. SEM analysis revealed a continuous and dense char layer on the residual surface of the synergistic flame retardant sample. AlBPi and APP exhibited excellent synergistic flame retardant effects, with both releasing a large amount of PO·free radicals. APP was also able to release inert gas, exhibiting excellent flame retardant effects in the gas phase. Additionally, APP improved the charring performance of AlBPi and formed a compact char layer on the material surface, achieving good flame retardant effects in the condensed phase. Therefore, the AlBPi and APP synergistic flame retardant system can be considered a viable flame retardant additive for PLA. Highlights A novel flame retardant AlBPi was synthesized. AlBPi and APP demonstrate good synergistic flame retardant performance in PLA. AlBPi and APP play a flame retardant role in both gas and condensed phases.

Dihydroartemisinin Ameliorates Decreased Neuroplasticity-Associated Proteins and Excessive Neuronal Apoptosis in APP/PS1 Mice

Zhao Yueyang,Long Zhimin,Liu Yuanjie,Luo Min,Qiu Yu,Idris Nur F.B.,Song Aijia,Wang Kejian,He Guiqiong

Background: Alzheimer's disease (AD) is one of the worst neurodegenerative disorders worldwide, with extracellular senile plaques (SP), subsequent intracellular neurofibrillary tangles (NFTs) and final neuron loss and synaptic dysfunction as the main pathological characteristics. Excessive apoptosis is the main cause of irreversible neuron loss. Thus, therapeutic intervention for these pathological features has been considered a promising strategy to treat or prevent AD. Dihydroartemisin (DHA) is a widely used first-line drug for malaria. Our previous study showed that DHA treatment significantly accelerated Aβ clearance, improved memory and cognitive deficits in vivo and restored autophagic flux both in vivo and in vitro. Methods: The present study intended to explore the neuroprotective effect of DHA on neuron loss in APP/PS1 double-transgenic mice and the underlying mechanisms involved. Transmission electron microscope (TEM) analysis showed that DHA significantly reduced the swollen endoplasmic reticulum (ER) in APP/PS1 mice. Western blot analysis indicated that DHA upregulated the level of NeuN, NeuroD, MAP2, and synaptophysin and promoted neurite outgrowth. Meanwhile, DHA greatly corrected the abnormal levels of Brain-derived neurotrophic factor (BDNF) and rescued the neuronal loss in the hippocampal CA1 area. Western blot analysis revealed that DHA notably down-regulated the protein expression of full length caspase-3, cleaved caspase-3 and Bax. In parallel, the expression of the anti-apoptotic protein Bcl-2 increased after oral DHA treatment. Results:Altogether, these results indicate that DHA protected AD mice from neuron loss via promoting the expression of BDNF and other neuroplasticity-associated proteins and suppressing the inhibition of neuronal apoptosis.
