Enrichment of tetracycline-degrading bacterial consortia: Microbial community succession and degradation characteristics and mechanism

Xiuli Chen,Yanchu Ke,Ying Zhu,Mingbang Xu,Chao Chen,Shuguang Xie

Tetracycline (TC) is an antibiotic that is recently found as an emerging pollutant with low biodegradability . Biodegradation shows great potential for TC dissipation. In this study, two TC-degrading microbial consortia (named SL and SI) were respectively enriched from activated sludge and soil. Bacterial diversity decreased in these finally enriched consortia compared with the original microbiota. Moreover, most ARGs quantified during the acclimation process became less abundant in the finally enriched microbial consortia. Microbial compositions of the two consortia as revealed by 16 S rRNA sequencing were similar to some extent, and the dominant genera Pseudomonas , Sphingobacterium, and Achromobacter were identified as the potential TC degraders. In addition, consortia SL and SI were capable of biodegrading TC (initial 50 mg/L) by 82.92% and 86.83% within 7 days, respectively. They could retain high degradation capabilities under a wide pH range (4−10) and at moderate/high temperatures (25–40 °C). Peptone with concentrations of 4–10 g/L could serve as a desirable primary growth substrate for consortia to remove TC through co-metabolism. A total of 16 possible intermediates including a novel biodegradation product TP245 were detected during TC degradation . Peroxidase genes, tet X-like genes and the enriched genes related to aromatic compound degradation as revealed by metagenomic sequencing were likely responsible for TC biodegradation.

Biodegradable microplastics induced the dissemination of antibiotic resistance genes and virulence factors in soil: A metagenomic perspective

Ruiping Song,Yuanze Sun,Xinfei Li,Changfeng Ding,Yi Huang,Xinyu Du,Jie Wang

Concerns about the ecological safety of both conventional and biodegradable microplastics have grown due to the inadequate end-of-life treatments of plastics. In this study, the effects of conventional and biodegradable microplastics on the spread of antibiotic resistance genes (ARGs) and virulence factors (VFs) were estimated in a soil microcosm experiment. The gene profiles and their respective bacterial hosts in soil were evaluated by metagenomic sequencing methods. The abundances of ARGs and VFs in polybutylene succinate (PBS) treated soils were statistically higher than the values in the control and conventional microplastic treatments. In comparison with the control, application of conventional microplastics showed negligible effects on ARG and VF profiles in the soil, while biodegradable microplastic amendments significantly changed the compositions of ARGs and VFs. The host-tracking analysis suggested application of microplastics broadened the bacterial hosts of ARGs and VFs in the soil. The percentage of Proteobacteria as ARG hosts increased from 38.5% in the control soils to 58.2% in microplastic exposed soil. The genus Bradyrhizobium was the dominant host of ARGs and VFs in biodegradable microplastic treatments, while conventional microplastics increased the percentages of Pseudomonas as the bacterial hosts. This study enhances the understanding of the effects of conventional and biodegradable microplastics on the propagation and hosts of ARGs and VFs in the terrestrial environment, providing essential insights into the risk assessment and management of plastics.

Residual chlorine persistently changes antibiotic resistance gene composition and increases the risk of antibiotic resistance in sewer systems

Jingyi Zhang,Zuxin Xu,Wenhai Chu,Feng Ju,Wei Jin,Peng Li,Rong Xiao

During the COVID-19 pandemic, excessive amounts of disinfectants and their transformation products entered sewer systems worldwide, which was an extremely rare occurrence before. The stress of residual chlorine and disinfection by-products is not only likely to promote the spread of antibiotic resistance genes (ARGs), but also leads to the enrichment of chlorine-resistant bacteria that may also be resistant to antibiotics. Therefore, the potential impact of such discharge on ARG composition should be studied and the health risks should be assessed. Thus, this study combined high-throughput 16S rRNA gene amplicon sequencing and metagenomic analysis with long-term batch tests that involved two stages of stress and recovery to comprehensively evaluate the impact of residual chlorine on the microbial community and ARG compositions in sewer systems. The tests demonstrated that the disturbance of the microbial community structure by residual chlorine was reversible, but the change in ARG composition was persistent. This study found that vertical propagation and horizontal gene transfer jointly drove ARG composition succession in the biofilm, while the driving force was mainly horizontal gene transfer in the sediment. In this process, the biocide resistance gene (BRG) subtype chtR played an important role in promoting co-selection with ARGs through plasmids and integrative and conjugative elements. Moreover, it was further shown that the addition of sodium hypochlorite increased the risk of ARGs to human health, even after discontinuation of dosing, signifying that the impact was persistent. In general, this study strengthens the co-selection theory of ARGs and BRGs, and calls for improved disinfection strategies and more environmentally friendly disinfectants.

nZnO-based graphene/graphene oxide compounds inhibit methane metabolic pathways and lower the development of antibiotic resistance genes and virulence factors

Haibo Zhang,Lizhi Li,Qingfeng Xing,Jia Chen,Yihao Fan,Luo Dan,Yuhong Zhong,Yang Wang,Dongsheng Guo,Weikun Guan,Cheng Zhang

Ruminant gastrointestinal fermentation produces a large amount of methane (CH 4 ), antibiotic resistance genes (ARGs), and virulence factors (VFs) pose significant risks to human health. This study utilized metagenomic sequencing to investigate the effect of nZnO-based graphene (nZnOG) and nZnO-based graphene oxide (nZnOGO) on the archaeal community, CH 4 metabolism, ARGs, VFs, and chemotaxis. Results indicated that the levels of zinc in manure were significantly lower in the nZnOG and nZnOGO groups compared to the nano zinc oxide (nZnO) group. The principal component analysis (PCA) of ARGs antibiotic class showed that there was an 80.13% variation between the three groups, suggesting that different zinc sources could influence ARGs related to various classes of antibiotics. The nZnO group exhibited considerably higher resistance levels to antimicrobial peptides and triclosan, in contrast to the nZnOG and nZnOGO groups. The copy number of ARGs followed the order of nZnO > nZnOG > nZnOGO, with a significant difference observed between the nZnO and nZnOGO groups. Moreover, the nZnOGO group displayed a significantly lower copy number of VFs compared to the nZnO group. Furthermore, the nZnOGO group demonstrated inhibition of CH 4 metabolic and chemotaxis pathways, which were associated with reduced transfer of ARGs and VFs. The functional coexistence of ARGs and VFs was driven by the archaeal community, while chemotaxis regulated changes in the archaeal community. These findings suggest that nZnOGO has the potential to inhibit the expression of ARGs and VFs through the inhibition of CH 4 metabolism and chemotaxis pathways. Consequently, feed supplementation with nZnOGO may offer a possible solution for mitigating CH 4 emission and reducing the transmission of ARGs and VFs.

Effects of environmentally relevant concentrations of oxytetracycline and sulfadiazine on the bacterial communities, antibiotic resistance genes, and functional genes are different between maize rhizosphere and bulk soil

Zhang Zekun,Zhao Le,Yang Jie,Pang Jiayin,Lambers Hans,He Honghua

Antibiotic contamination in soil has become a major concern worldwide. At present, it is not clear how two co-existed antibiotics with environmentally relevant concentrations would affect soil bacterial community structure, the abundances of antibiotic resistance genes (ARGs) and functional genes, and whether the effects of antibiotics would differ between rhizosphere and bulk soil. We conducted a greenhouse pot experiment to grow maize in a loess soil treated with oxytetracycline (OTC) or sulfadiazine (SDZ) or both at an environmentally relevant concentration (1 mg kg −1 ) to investigate the effects of OTC and SDZ on the rhizosphere and bulk soil bacterial communities, abundances of ARGs and carbon (C)-, nitrogen (N)-, and phosphorus (P)-cycling functional genes, and on plant growth and plant N and P nutrition. The results show that the effects of environmentally relevant concentrations of OTC and SDZ on bacterial communities and abundances of ARGs and functional genes differ between maize rhizosphere and bulk soil. The effects of two antibiotics resulted in a higher absolute abundances of accA , tet(34) , tnpA-04 , and sul2 in the rhizosphere soil than in the bulk soil and different bacterial community compositions and biomarkers in the rhizosphere soil and the bulk soil. However, OTC had a stronger inhibitory effect on the abundances of a few functional genes in the bulk soil than SDZ did, and their combination had no synergistic effect on plant growth, ARGs, and functional genes. The role of co-existed OTC and SDZ decreased shoot height and increased root N concentration. The results demonstrate that environmentally relevant concentrations of antibiotics shift soil microbial community structure, increase the abundances of ARGs, and reduce the abundances of functional genes. Furthermore, soil contamination with antibiotics can diminish agricultural production via phytotoxic effects on crops, and combined effects of antibiotics on plant growth and nutrient uptake should be considered.

Short-Term Benzalkonium Chloride (C12) Exposure Induced the Occurrence of Wide-Spectrum Antibiotic Resistance in Agricultural Soils

Jieyi Zeng,Yaying Li,Guoping Jin,Jian-Qiang Su,Huaiying Yao

Antibiotic resistance genes (ARGs) are global pollutants that pose a potential risk to human health. Benzalkonium chloride (C12) (BC) disinfectants are thought to exert selection pressure on antibiotic resistance. However, evidence of BC-induced changes in antibiotic resistance in the soil environment is lacking. Here, we established short-term soil microcosms to investigate ARG profile dynamics in agricultural soils amended with sulfamethazine (SMZ, 10 mg kg–1) and gradient concentrations of BC (0–100 mg kg–1), using high-throughput quantitative PCR and Illumina sequencing. With the increase in BC concentration, the number of ARGs detected in the soil increased, but the normalized ARG abundance decreased. The added SMZ had a limited impact on ARG profiles. Compared to broad-spectrum fungicidal BC, the specificity of SMZ significantly affected the microbial community. Network analysis found that low–medium BC exposure concentrations resulted in the formation of small but strong ARG co-occurrence clusters in the soil, while high BC exposure concentration led to a higher incidence of ARGs. Variation partitioning analysis suggested that BC stress was the major driver shaping the ARG profile. Overall, this study highlighted the emergence and spread of BC-induced ARGs, potentially leading to the antimicrobial resistance problem in agricultural soils.

Sulfonamide metabolites enhance resistance transmission via conjugative transfer pathways

Yuan Zhang,Félix Manuel Rosado-García,Yamila Puig Peña,Panagiotis Karanis,Xin Yu,Mingbao Feng,Chengsong Ye

Human beings release thousands of antibiotics into the environment, which could generate the related transformation products (TPs), most of which have yet to be identified and lack rigorous microbial risk information. This study aimed to investigate the impact and mechanisms of 4-nitro sulfamethoxazole, N 4 -acetylated sulfamethoxazole, and N 4 -acetylated sulfadiazine, three typical sulfonamide (SAs) metabolites, on the risk of antibiotic resistance genes (ARGs) transmission. The results revealed that TPs significantly enhance the risk of conjugative transfer of RP4 plasmid at clinically and environmentally relevant concentrations (10 ng/L to 100 μg/L), with a maximum increase of up to 73-fold. These three metabolites’ capabilities to enhance the conjugative transfer of ARGs are more pronounced than the parent sulfonamides. The induction mechanisms of TPs on ARGs transmission are also more complex, which primarily arise from the enhancement of reactive oxygen species, further increased cell membrane permeability and upregulated bacterial secretion systems. Transcriptomic analysis validated the aforementioned biological processes and showed that TPs also increased the activity of toxin-antitoxin system and bacterial intracellular transposon, thereby promoting the spread of ARGs. This research contributes to a better understanding of the antibiotic-like effects of TPs, which is crucial for improving our understanding of non-antibiotic drug-induced bacterial resistance risks.

Efficient persulfate activation by carbon defects g-C3N4 containing electron traps for the removal of antibiotics, resistant bacteria and genes

Boru Gao,Mengmeng Dou,Jin Wang,Shuming Li,Dongying Wang,Lin Ci,Yao Fu

Contaminants such as antibiotics and the resulting antibiotic-resistant bacteria (ARB) and antibiotic-resistant genes (ARGs) have become a hot topic in the field of wastewater purification. In this study, a method was proposed to simultaneously remove antibiotics and reduce ARB and ARGs with C–(N) 3 defect carbon nitride-coupled persulfates (CN-C V2 /PS). The location of the C–(N) 3 defects was determined by a combination of spectroscopic techniques and electron spin resonance tests. Based on photoluminescence and electrochemical tests, and deformation charge density calculations, electron traps were generated at defect sites resulting in enhanced electron–hole separation. The mineralization rates of CN-C V2 /PS for six typical antibiotics were 11–17% and 21–28% higher than those of CN-C V2 and conventional CN , respectively. Over the same reaction time, the inactivation rates of CN-C V2 /PS for E. coli and E. faecalis screened from wastewater treatment plants were 3.6 and 2.7 log higher than that of CN-C V2 . In addition, CN-C V2 /PS showed a reduction rate of more than 2.5 log for tet A and intI 1. DFT calculation showed that due to the formation of defects, the adsorption energy between the PS and catalysts increased from –8.924 eV (CN) to –18.340 eV (CN-C V2 ). At the same time, the effective stretch of peroxide bond of PS changed from 1.505 Å to 1.519 Å. Both results indicated that the activation of PS was significantly enhanced in the presence of the modified CN-C V2 . The synergetic effect of CN-C V2 /PS is expected to remove the antibiotics, resistant bacteria and genes efficiently in future wastewater advanced treatment.

Dissecting the roles of conductive materials in attenuating antibiotic resistance genes: Evolution of physiological features and bacterial community

Qi Zhao,Wanqian Guo,Haichao Luo,Huazhe Wang,Taiping Yu,Banghai Liu,Qishi Si,Nanqi Ren

Supplying conductive materials (CMs) into anaerobic bioreactors is considered as a promising technology for antibiotic wastewater treatment. However, whether and how CMs influence antibiotic resistance genes (ARGs) spread remains poorly known. Here, we investigated the effects of three CMs, i.e., magnetite, activated carbon (AC), and zero valent iron (ZVI), on ARGs dissemination during treating sulfamethoxazole wastewater, by dissecting the shifts of physiological features and microbial community . With the addition of magnetite, AC, and ZVI, the SMX removal was improved from 49.05 to 71.56—92.27 %, while the absolute abundance of ARGs reducing by 26.48 %, 61.95 %, 48.45 %, respectively. The reduced mobile genetic elements and antibiotic resistant bacteria suggested the inhibition of horizontal and vertical transfer of ARGs. The physiological features, including oxidative stress response, quorum sensing, and secretion system may regulate horizontal transfer of ARGs. The addition of all CMs relieved oxidative stress compared with no CMs, but ZVI may cause additional free radicals that needs to be concerned. Further, ZVI and AC also interfered with cell communication and secretion system. This research deepens the insights about the underlying mechanisms of CMs in regulating ARGs, and is expected to propose practical ways for mitigating ARGs proliferation.

Occurrence and removal of antibiotics and the corresponding resistance genes in wastewater treatment plants: effluents’ influence to downstream water environment

Li Jianan,Cheng Weixiao,Xu Like,Jiao Yanan,Baig Shams Ali,Chen Hong

In this study, the occurrence of 8 antibiotics [3 tetracyclines (TCs), 4 sulfonamides, and 1 trimethoprim (TMP)], 12 antibiotic resistance genes (ARGs) (10 tet , 2 sul ), 4 types of bacteria [no antibiotics, anti-TC, anti-sulfamethoxazole (SMX), and anti-double], and intI 1 in two wastewater treatment plants (WWTPs) were assessed and their influences in downstream lake were investigated. Both WWTPs’ effluent demonstrated some similarities, but the abundance and removal rate varied significantly. Results revealed that biological treatment mainly removed antibiotics and ARGs, whereas physical techniques were found to eliminate antibiotic resistance bacteria (ARBs) abundance (about 1 log for each one). UV disinfection did not significantly enhance the removal efficiency, and the release of the abundantly available target contaminants from the excess sludge may pose threats to human and the environment. Different antibiotics showed diverse influences on the downstream lake, and the concentrations of sulfamethazine (SM2) and SMX were observed to increase enormously. The total ARG abundance ascended about 0.1 log and some ARGs (e.g., tet C, intI 1, tet A) increased due to the high input of the effluent. In addition, the abundance of ARB variation in the lake also changed, but the abundance of four types of bacteria remained stable in the downstream sampling sites.

Self-rescue of nitrogen-cycling bacteria under β-lactam antibiotics stress during managed aquifer recharge (MAR): Microbial collaboration and anti-resistance

Yuanping Fang,Congli Chen,Bin Cui,Dandan Zhou

Storing reclaimed water via managed aquifer recharge (MAR) is an effective strategy for alleviating groundwater overdraft and achieving water resource recycling simultaneously. However, β-lactam antibiotics in the reclaimed water can induce stress on aquifer system, reshape microbial community, and affect the emergence and prevalence of antibiotic resistance genes (ARGs). In this study, three sandy soil columns (H 1.5 m, ID 14 cm) were employed to simulate MAR, and synthetic reclaimed water containing either amoxicillin (AMO), ampicillin (AMP) or oxacillin (OXA) was continuously recharged for 120 d The temporal and spatial attenuation of β-lactams and nitrogen was studied, and microbial collaboration and the resistance mechanism were elaborated. Biodegradation is the main pathway for β-lactams elimination, AMO and AMP were eliminated when migrating 30 cm, while the attenuation of OXA experienced in the whole column with final removal efficiency of 82%. Moreover, refractory OXA induced more ARGs production, and approximately 10% and 13% higher than that of AMO and AMP columns. Efflux pump and antibiotics inactivation were the two major resistance mechanisms. NO 3 − -N gradually decreased (by 26%, 38%, and 49% for AMO, AMP, and OXA, respectively) along the recharge direction. Microbial co-occurrence network revealed that nitrogen-cycling bacteria were the keystone species in aquifer community, and ammonation provided NH 4 + -N for the nitrification process of ammonia-oxidizing archaea (AOA), promoting the further denitrification for nitrogen removal in MAR process. Nitrogen-cycling bacteria were the key and active ARG hosts, which could keep nitrogen transformation activity under antibiotics stress. In sum, nitrogen-cycling bacteria exhibited intimate collaboration and elastic resistance in response to the malnutrition environment and β-lactams exposure during MAR.

Sulfonamides repress cell division in the root apical meristem by inhibiting folates synthesis

Guanping Feng,Wenying Zou,Yihui Zhong

Sulfonamides, as broad-spectrum antibiotics, are widely used to protect against bacterial infections in livestock production. A large number of sulfa antibiotics are discharged into soil and water system, which poses a great threat to the safety of organisms. The antibacterial mechanism of sulfonamides is to prevent folate biosynthesis by competitively inhibiting bacterial dihydropteroate synthetase (DHPS). The inhibitory effect of sulfanilamide on root growth has been reported, but the mechanism has not been determined. Here, we used high performance liquid chromatography with ultraviolet detector (HPLC-UV) and transgenic technology to study the effect of sulfonamides on folate synthesis in plants. The results showed that the application of sulfonamide sharply reduced the content of folic acid in plants, resulting in strong inhibition of cell division in the root apical meristem. The inhibition of sulfonamide on cell division can be relieved by supplementing synthetic folic acid and folinic calcium salt hydrate. The transgenic plants expressing the bacterial antibiotic resistance genes (ARGs) Sul1 or Sul2 showed obvious antibiotic resistance. Taking these results together, we suggest that sulfonamides repress cell proliferation in the root apical meristem by competitively inhibiting the biosynthesis of folic acid in plants.
