Novel Cs-Based Upconversion Nanoparticles as Dual-Modal CT and UCL Imaging Agents for Chemo-Photothermal Synergistic Therapy

Yuxin Liu,Luoyuan Li,Quanwei Guo,Lu Wang,Dongdong Liu,Ziwei Wei,Jing Zhou

Lanthanide-based contrast agents have attracted increasing attention for their unique properties and potential applications in cancer theranostics. To date, many of these agents have been studied extensively in cells and small animal models. However, performance of these theranostic nanoparticles requires further improvement. In this study, a novel CsLu 2 F 7 :Yb,Er,Tm-based visual therapeutic platform was developed for imaging-guided synergistic cancer therapy. Due to the presence of the heavy alkali metal Cesium (Cs) in host lattice, the nanoplatform can provide a higher resolution X-ray CT imaging than many other reported lanthanide-based CT contrast agents. Furthermore, by using the targeted RGD motif, chemotherapy drug alpha-tocopheryl succinate ( α -TOS), and photothermal coupling agent ICG, this nanoplatform simultaneously provides multifunctional imaging and targeted synergistic therapy. To demonstrate the theranostic performance of this novel nanoplatform in vivo , visual diagnosis in the small animal model was realized by UCL/CT imaging which was further integrated with targeted chemo-photothermal synergistic therapy. These results provided evidence for the successful construction of a novel lanthanide-based nanoplatform coupled with multimodal imaging diagnosis and potential application in synergistic cancer theranostics.

Efficient oral delivery of water-soluble CT contrast agent using an W1/O/W2 alginate hydrogel matrix

Meng Ren,Na Li,Xuliang Jiang,Xianhe Liu,Aihua Zou

Hypothesis Iohexol (IOH) is a commonly used second-generation nonionic iodinated contrast agent. However, low gastrointestinal mucosal adherence and high downstream speed limit its application in intestinal Computed tomography (CT) imaging. We hypothesize that oral IOH delivery carriers composed of environmentally responsive materials enable the intestinal targeted delivery and prolong the intestinal residence time of IOH, enhancing the intestinal disease detection efficiency. Experiments An emulsion-filled alginate hydrogel system was developed as the intestinal targeting vehicle for IOH. The formulation optimization was determined by response surface analysis . After a thorough study of the physicochemical properties of this hydrogel matrix, the pH sensitivity and the ability to control release were investigated, followed by a vitro cell experiment evaluating its bioactivity and CT imaging capability. Findings This alginate hydrogel matrix was sparsely structured and rapidly released IOH at pH 7.4. Meanwhile the swelling degree was 4.4 times higher than that at pH 1.2, indicating a good selective responsiveness to the gastrointestinal simulated environment. It improved the CT visual contrast of A549 cells without affecting cell morphology, suggesting that it would be an effective oral administration for water-soluble nonionic contrast agents and a potential candidate for intestinal disease detection tools.

Microwave–assisted synthesis of water–disperse and biocompatible NaGdF4: Yb,Ln@NaGdF4 nanocrystals for UCL/CT/MR multimodal imaging

Songxia Yu,Zhiqiang Wang,Ruijun Cao,Lingjie Meng

In this paper, we have developed a series of rare earth fluoride nanocrystals (NCs) that can be used for multimodal imaging applications. Three highly monodisperse and water dispersible NaGdF 4 :Yb,Ln@NaGdF 4 (Ln   =   Er, Tm, Ho) upconversion luminescence (UCL) NCs with an average size of 75   nm were prepared using a facile and efficient microwave–assisted solvothermal reaction in ethylene glycol (EG). The morphology, crystal structures, size distributions and zeta potentials of the resulting NCs were fully characterized by transmission electron microscopy (TEM), X–ray diffraction (XRD) and Nano–ZS90 Zetasizer. The obtained NaGdF 4 :Yb,Ln@NaGdF 4 (Ln   =   Er, Tm, Ho) NCs could emit bright blue, green and red luminescence in water upon laser excitation at 980   nm. Furthermore, these core–shell NCs exhibited low cytotoxicity for HeLa and C9H2 cells, separately. Moreover, these NCs also displayed excellent Hounsfield units (HU) values in X–ray computed tomography (CT) imaging and high relaxivity parameters (r 1 ) in magnetic resonance (MR) imaging in vitro . Thus, these small, water–dispersible and biocompatible NCs show excellent potential as contrast agents for UCL/CT/MR multimodal imaging and other biomedical applications.

A Bi2S3@mSiO2@Ag nanocomposite for enhanced CT visualization and antibacterial response in the gastrointestinal tract

Yan Zu,Liang Yan,Tao Wang,Dongqing Ma,Xinghua Dong,Zhen Du,Wenyan Yin

The non-invasive imaging of the gastrointestinal (GI) tract is highly desired for clinical research due to the various GI tract bacterial infection-induced diseases. To treat GI tract infections, various antibiotics have been used in the clinic. The growing problem of multidrug-resistant bacteria calls for effective antibiotic alternatives. Here, we construct a dual-functional Bi2S3@mSiO2@Ag nanocomposite for simultaneous enhanced X-ray computed tomography (CT) imaging and efficient antibacterial activity in the GI tract. The nanocomposite also has good stability, low cytotoxicity, and negligible hemolysis. Moreover, the investigation of the long-term toxicity and biodistribution of the Bi2S3@mSiO2@Ag nanocomposite after oral administration confirms its safety at the tested dosage. In particular, Ag nanoparticles (NPs) well dispersed on a silica substrate can reduce the antibacterial dosage and enhance the antibacterial activity of the Bi2S3@mSiO2@Ag nanocomposite. Furthermore, we have established bacterially infected enteritis animal models to confirm the antibacterial ability of the nanocomposite. This work opens up a new avenue for the design of a nanotheranostic agent that acts as both a contrast agent for the enhanced visualization of the GI tract and an antibacterial agent as an alternative to antibiotics.

Ultrasmall AgBiSe2 nanodots for CT/thermal imaging-guided photothermal tumor therapy in the NIR-II biowindow

Jingge Zhang,Enna Ha,Danyang Li,Luyang Wang,Junqing Hu

Stimulus-responsive ternary chalcogenide nanomaterials are regarded as promising ‘all-in-one’ nanotheranostics agents on account of their tunable band structures and multi-metal intrinsic properties. Herein, ultrasmall AgBiSe2 nanodots are prepared by a simple thermal injection method. It shows a narrow band gap of 0.91 eV and high absorption coefficient in the NIR-II biowindow, resulting in excellent photothermal performance. Under the irradiation of a 1064 nm laser, AgBiSe2 can induce the overexpression of intracellular heat shock protein (Hsp70) and cell apoptosis to inhibit the growth of tumor cells. The strong signal from CT/thermal imaging also provides guidance for tumor diagnosis. Importantly, AgBiSe2 can be rapidly excreted from the body, thus avoiding long term toxicity. This study presents the first biomedical application of AgBiSe2 nanodots in cancer treatment and extends the development of ternary chalcogenide-based semiconductor nanomedicine.

Simultaneous realization of persistent luminescence and CT dual-mode imaging by x-ray recharged Bi2Ga4O9:Cr nanoprobes in depth-independent tumors

Li Jinlei,Guo Jiaqing,Li Hao,Qu Junle,Song Jun

Synthesis of excellent imaging nanoparticles plays a critical role in disease detection. Here, novel Bi 2 Ga 4 O 9 :Cr (BGOC) nanoparticles were synthesized for persistent luminescence and computed tomography dual-mode imaging. The Cr 3+ ions in the BGOC luminescent nanoparticles were coped as the luminescent center, and the afterglow emission was located in the first biological imaging window. Bi 3+ ions served as the computed tomography (CT) imaging contrast agent. The NH 2 groups were modified on the surface of the nanoparticles to make them highly biocompatible, and folic acid molecules were modified to facilitate the specificity. This modification facilitated simultaneous persistent luminescence and CT imaging in tumor-bearing mice due to the overexpression of the folic acid molecules’ receptors. Furthermore, instead of visible light, x-rays were used as the excitation light source to activate the BGOC-folic acid nanoprobes and realize long time and high sensitivity imaging of tumors in vivo because of their capacity for deep tissue penetration. Therefore, such nanoprobes can provide a promising depth-independent detection mode for high sensitivity diagnosis and long-term monitoring of diseases.

Rhenium Sulfide Nanoparticles as a Biosafe Spectral CT Contrast Agent for Gastrointestinal Tract Imaging and Tumor Theranostics in Vivo

Xiaoyi Wang,Jiaojiao Wang,Jinbin Pan,Fangshi Zhao,Di Kan,Ran Cheng,Xuening Zhang,Shao-Kai Sun

Spectral computed tomography (CT) imaging as a novel imaging technique shows promising prospects in the accurate diagnosis of various diseases. However, clinically iodinated contrast agents suffer from poor signal-to-noise ratio, and emerging heavy-metal-based CT contrast agents arouse great biosafety concern. Herein, we show the fabrication of rhenium sulfide (ReS2) nanoparticles, a clinic radiotherapy sensitizer, as a biosafe spectral CT contrast agent for the gastrointestinal tract imaging and tumor theranostics in vivo by teaching old drugs new tricks. The ReS2 nanoparticles were fabricated in a one-pot facile method at room temperature, and exhibited sub-10 nm size, favorable monodispersity, admirable aqueous solubility, and strong X-ray attenuation capability. More importantly, the proposed nanoparticles possess an outstanding spectral CT imaging ability and undoubted biosafety as a clinic therapeutic agent. Besides, the ReS2 nanoparticles possess appealing photothermal performance due to their intense near-infrared absorption. The proposed nano-agent not only guarantees obvious contrast enhancement in gastrointestinal tract spectral CT imaging in vivo, but also allows effective CT imaging-guided tumor photothermal therapy. The proposed “teaching old drugs new tricks” strategy shortens the time and cuts the cost required for clinical application of nano-agents based on existing clinical toxicology testing and trial results, and lays down a low-cost, time-saving, and energy-saving method for the development of multifunctional nano-agents toward clinical applications.

Bimetallic nanodots for tri-modal CT/MRI/PA imaging and hypoxia-resistant thermoradiotherapy in the NIR-II biological windows

Meng Lyu,Daoming Zhu,Yanhong Duo,Yang Li,Hong Quan

Hypoxic tumor microenvironment leads to resistance or failure of radiotherapy (RT). As a non-invasive therapy, photothermal therapy (PTT) can improve the tumor hypoxic microenvironment in addition to directly killing tumor cells. PTT combined with RT (thermoradiotherapy) becomes an emerging treatment. Multi-functional nanoparticles used for hypoxia-resistant thermoradiotherapy in the second near-infrared (NIR-II) biological windows (1000–1700 nm) are urgently needed to be developed. Here, a facil method synthesis of ultra-small cysteamine (Cys)-coated FePd bimetallic nanodots (NDs) is reported. These NDs can not only produce effective hyperthermia (35.4%) when irradiated in the NIR-II region (1064 nm) but also have an enhanced radiation effect due to i) Hypoxic improvement in tumor tissues by photothermal treatment in the NIR-II Biological Windows can greatly enhance the sensitivity of tumor cells to radiotherapy ii) The ability of NDs to deposit radiation energy in tumors has further enhanced the sensitivity of tumor cells to radiotherapy. Meanwhile, NDs was a contrast agent for tri-modal imaging including computed tomography (CT)/magnetic resonance imaging (MRI)/photoacoustic imaging (PAI) in vitro and in vivo . Both in vitro and in vivo tests demonstrated good biocompatibility and excellent stability of NDs, indicating great potential for clinical applications.

Oxygen-supplementing mesoporous polydopamine nanosponges with WS2 QDs-embedded for CT/MSOT/MR imaging and thermoradiotherapy of hypoxic cancer

Yidan Wang,Shaoze Song,Tong Lu,Yu Cheng,Yilin Song,Siyu Wang,Fengping Tan,Jiao Li,Nan Li

Multifunctional nanoplatforms with flexible architectures and tumor microenvironment response are highly anticipated within the field of thermoradiotherapy. Herein, the multifunctional nanoplatforms for thermoradiotherapy have been successfully constructed by the embedding of tungsten disulfide quantum dots (WS 2 QDs) into mesoporous polydopamine nanosponges (MPDA NSs), followed by integration with manganese dioxide (MnO 2 ). MPDA-WS 2 @MnO 2 , the resultant nanoplatforms, exhibit radiosensitization enhanced behavior and a capacity for responsive oxygen self-supplementation. The ingenious mesoporous structure of MPDA NSs serves as reservoir for the assembly of WS 2 QDs to form MPDA-WS 2 nanoparticles (NPs), in which WS 2 QDs provide the radiation enhancement effect, whereas the MPDA NSs framework endows the MPDA-WS 2 @MnO 2 with an excellent photothermal capability. Additionally, the integration of the MnO 2 component works to decompose the tumor-overexpressed H 2 O 2 and alleviate tumor hypoxia subsequently, which has been demonstrated to enhance radiotherapy performance considerably. Meanwhile, the prepared MPDA-WS 2 @MnO 2 nanoplatforms have been evaluated as trimodality contrast agents for computed tomography (CT), multispectral optoacoustic tomography (MSOT), and tumor microenvironment-responsive T 1 -weighted magnetic resonance (MR) imaging that have the potential for real-time guidance and monitoring during cancer therapy. More importantly, when subjected to near infrared (NIR) laser irradiation and X-ray exposure, the tumor is found to be inhibited significantly through the process of combined thermoradiotherapy. The design concepts of embedding WS 2 QDs into MPDA NSs and oxygen self-supplementing hold great potential for multimodal imaging-guided thermoradiotherapy of hypoxic cancer.

Multiscale X-ray phase-contrast CT uncovers adaptive changes and compensatory mechanisms of circulatory pathways during acute liver injury

Zhao Yuanyuan,Lv Wenjuan,He Yi,Qi Beining,Du Xianqin,Zhao Yuqing,Shan Shan,Zhao Xinyan,Hu Chunhong,Jian Jianbo

Intrahepatic circulation is essential for the repair of acute liver injury (ALI); however, very limited information is available concerning changes in the circulatory pathways during ALI. Therefore, multi-scale X-ray phase-contrast CT combined with three-dimensional (3D) visualization is used to quantitatively analyze the intrahepatic circulation pathway (including the hepatic vein, portal vein and hepatic sinusoid) in the mouse model via the intraperitoneal injection of carbon tetrachloride (CCl 4 ) from acute injury to recovery. The results demonstrate that the liver still preserves some vessel-like channels accessed to the central vein when the injury causes the severe collapse of the hepatic sinusoids that cannot be observed in two-dimensional pathologic slices. Moreover, angiogenesis is observed in the terminal branches of the hepatic vein and portal vein. Additionally, we extend the two-dimensional primary lobule to a 3D model and find that the sinusoids in zone III have the most severe injury. The sinusoids in different zones also show changes in parameters such as density and mean diameter during the ALI. In conclusion, phase-contrast CT can reveal the intact vascular system within the liver lobes, thus providing critical information for studying the mechanisms involved in the evolution of circulatory structures from damage to repair.

A novel bi-modal probe based on BaHoF5 and Cu-doped QDs with enhanced CT contrast efficiency and fluorescent brightness for tumor-targeting imaging

Zheng Xiaoke,Li Chenghui,He Yaqin,Wu Lan,Hou Xiandeng

In this work, a novel bi-modal imaging probe with enhanced CT contrast efficiency and FL brightness was constructed, in which the combination of a binary CT contrast agent BaHoF 5 and Cu-doped QDs served as a vehicle; hyaluronic acid (HA) was employed as a tumor-targeting ligand. With its CT contrast efficiency about 2.1- and 3.9-fold higher than PEG-BaHoF 5 and Iohexol, the CT contrast efficiency and the fluorescent brightness of the bi-modal probe were both enhanced. Likewise, its fluorescent brightness is almost 6-fold brighter after Cu-doped QDs loading. The most important contribution of this work lies on the proposed strategy. The inherent contradiction of the imaging sensitivity of CT and FL imaging is well balanced and a great CT/FL bi-modal imaging performance is simultaneously obtained even at low concentration (400 μg/mL) of the probe, which was superior to the previous CT/FL bi-modal probes. Moreover, since BaHoF 5 as a binary CT contrast agent was introduced instead of conventional Au and Bi 2 S 3 , the CT/FL bi-modal probe would be more suitable for different patients under different operation voltages. In addition, the in vitro tumor cell imaging also demonstrated a good photo-stability, FL brightness, and tumor-targeting capability of the probe, indicating its great potential in practical bi-modal imaging for further tumor diagnosis and therapy. A novel bi-modal imaging probe with enhanced CT contrast efficiency and FL brightness was fabricated, in which its CT contrast efficiency was about 2.1- and 3.9-fold higher than PEG-BaHoF 5 and Iohexol, respectively, and its fluorescent brightness almost 6-fold brighter after Cu-doped QDs loading.

TPGS-stabilized NaYbF4:Er upconversion nanoparticles for dual-modal fluorescent/CT imaging and anticancer drug delivery to overcome multi-drug resistance

Gan Tian,Xiaopeng Zheng,Xiao Zhang,Wenyan Yin,Jie Yu,Dongliang Wang,Zhiping Zhang,Xiangliang Yang,Zhanjun Gu,Yuliang Zhao

Multi-drug resistance (MDR) is a major cause of failure in cancer chemotherapy. Tocopheryl polyethylene glycol 1000 succinate (TPGS) has been extensively investigated for overcoming MDR in cancer therapy because of its ability to inhibit P-glycoprotein (P-gp). In this work, TPGS was for the first time used as a new surface modifier to functionalize NaYbF 4 :Er upconversion nanoparticles (UNCPs) and endowed the as-prepared products (TPGS-UCNPs) with excellent water-solubility, P-gp inhibition capability and imaging-guided drug delivery property. After the chemotherapeutic drug (doxorubicin, DOX) loading, the as-formed composites (TPGS-UCNPs-DOX) exhibited potent killing ability for DOX-resistant MCF-7 cells. Flow-cytometric assessment and Western blot assay showed that the TPGS-UCNPs could potently decrease the P-gp expression and facilitate the intracellular drug accumulation, thus achieving MDR reversal. Moreover, considering that UCNPs process efficient upconversion emission and Yb element contained in UCNPs has strong X-ray attenuation ability, the as-obtained composite could also serve as a dual-modal probe for upconversion luminescence (UCL) imaging and X-ray computed tomography (CT) imaging, making them promising for imaging-guided cancer therapy.
