Insight into volatile metabolites and key odorants in black teas processed from Jianghua Kucha tea germplasm (Camellia sinensis var. assamica cv. Jianghua)

Ying-Lin Zhu,Wei-Xuan Li,Yu-Hui Zhang,Han Yan,Long-Yu Guo,Yue Zhang,Hai-Peng Lv,Ling-Hong Zhou,Zhi Lin,Wen-Liang Wu,Yin Zhu

Jianghua Kucha (JHKC) is an ancient tea germplasm, which presents a high value for resource utilization but was less developed in aroma research. In this study, volatile metabolites in fresh leaves and corresponding black teas of JHKC, along with odorants of JHKC black teas were systematically investigated using stir-bar sorptive extraction in combination with gas chromatography–mass spectrometry (GC–MS) and GC–olfactometry/MS (GC–O/MS) technologies. A total of 128 and 135 volatile compounds were identified in fresh leaves and black teas, respectively, along with 13 key differential volatile components derived from fatty acids, pigments, glycoside precursors, and the terpene pathway. An odorant wheel for JHKC black teas was developed based on 29 important odorants determined through a combination of GC–O/MS perception, aroma extract dilution analysis, odor activity value measurement. Trans - β -Damascenone, cis -jasmone, and cis -jasmin lactone have been identified as significant contributors to the “sweet” flavor of JHKC black teas.

Chemical Composition of Essential Oil from Citrus reticulata Blanco cv. Chachiensis (Chachi) and Its Anti-Mosquito Activity against Pyrethroid-Resistant Aedes albopictus

Jifan Cao,Wende Zheng,Baizhong Chen,Zhenping Yan,Xiaowen Tang,Jiahao Li,Zhen Zhang,Song Ang,Chen Li,Rihui Wu,Panpan Wu,Wen-Hua Chen

Simple SummaryThe essential oil (EO) ofCitrus reticulataBlanco cv. Chachiensis (Chachi) exhibited significant larvicidal activity. The EO from semi-mature fruits and its main components were able to control mosquitoes by effectively inhibiting acetylcholinesterase and three detoxification enzymes and significantly altering the diversity of the internal microbiota in mosquitoes.AbstractThe overuse of synthetic insecticides has led to various negative consequences, including insecticide resistance, environmental pollution, and harm to public health. This may be ameliorated by using insecticides derived from botanical sources. The primary objective of this study was to evaluate the anti-mosquito activity of the essential oil (EO) ofCitrus reticulataBlanco cv. Chachiensis (Chachi) (referred to as CRB) at immature, semi-mature, and mature stages. The chemical compositions of the CRB EO were analyzed using GC-MS. The main components were identified to be D-limonene andγ-terpinene. The contents of D-limonene at the immature, semi-mature, and mature stages were 62.35%, 76.72%, and 73.15%, respectively; the corresponding contents ofγ-terpinene were 14.26%, 11.04%, and 11.27%, respectively. In addition, the corresponding contents of a characteristic component, methyl 2-aminobenzoate, were 4.95%, 1.93%, and 2.15%, respectively. CRB EO exhibited significant larvicidal activity againstAedes albopictus(Ae. albopictus, Diptera: Culicidae), with the 50% lethal doses being 65.32, 61.47, and 65.91 mg/L for immature, semi-mature, and mature CRB EO, respectively. CRB EO was able to inhibit acetylcholinesterase and three detoxification enzymes, significantly reduce the diversity of internal microbiota in mosquitoes, and decrease the relative abundance of core species within the microbiota. The present results may provide novel insights into the utilization of plant-derived essential oils in anti-mosquitoes.

Influence of the stage of ripeness on the phytochemical profiles, antioxidant and antiproliferative activities in different parts of Citrus reticulata Blanco cv. Chachiensis

Hong Wang,Gu Chen,Xinbo Guo,Arshad Mehmood Abbasi,Rui Hai Liu

Citrus is a rich source of nutritive and bioactive compounds . Impact of fruit tissues and ripening stage on phytochemical profile, antioxidant capacity and antiproliferative activity was investigated in Citrus reticulata Blanco cv. Chachiensis (Chachi). Regardless of different ripening stage, peel had the highest contents of phenolics and flavonoids , strongest antioxidant activities resolved by PSC, ORAC and CAA assay and antiproliferative activities against MCF-7, HepG2 and SiHa cancer cell. Increased level of total phenolics, hesperidin and naringin, and elevated antioxidant capacities were detected in peel during maturity, whereas highest amount of nobiletin and strongest antiproliferative activities were explored in immature peel. In flesh the vitamin C content increased during ripening, while lower phenolic content, smallest ORAC value and weakest antiproliferative activity were resolved in mature flesh. The flavonoid content and antioxidant capacity in seed were comparable with flesh and relatively constant during ripening, suggesting seeds as a valuable source of flavonoid extraction for further utilization. The characteristic flavonoids, nobiletin, tangeretin, hesperetin and naringenin inhibited proliferation of SiHa, HepG2 and MCF-7 cell dose-dependently, thus suggesting their contribution and potential application. Results presented here provide the foundation for comprehensive utilization of phytochemicals in different parts of this medicinal and edible citrus fruit.

Geographical Origin Drives Metabolic Divergence in Styphnolobium japonicum cv. Jinhuai: A Widely Targeted Metabolomic Study of Flower Buds from Sichuan and Guangxi, China

Leilei Zuo,Yan Chen,Yuxuan Luo,Huan Yang,Dayi Chen,Ying Zhang,Xiao Meng,Waralee Watcharin

HighlightsWhat are the main findings? We identified 1550 metabolites (flavonoid-dominant), 152 key active ingredients of traditional Chinese medicine (TCM-KAIs), and 204 pharmaceutical disease-resistant ingredients (PDRIs) in the flower bud ofStyphnolobium japonicumcv. Jinhuai (FBSJvJ).Geographic origin drove metabolic divergence. Guangxi samples were enriched in lipids and nucleotides, while Sichuan samples were enriched in flavonoids and phenolic acids. In addition, three core biomarkers were identified. What are the implications of the main findings?Biomarkers enable origin authentication and precision quality control compared to a single marker (rutin), supporting traceability and regulatory compliance for food and medicinal products.Regional metabolic signatures can help identify functional ingredients and guide product formulation, processing optimization, and cultivar selection for food and medicinal development.Background/Objectives:Styphnolobium japonicumcv. Jinhuai (SJvJ) represents a medicinal and edible plant whose metabolite composition is strongly shaped by its growing location. Current quality control methods mainly rely on rutin quantification, lacking comprehensive metabolic markers for origin discrimination. Therefore, this study aimed to profile interregional metabolic differences between Guangxi and Sichuan SJvJ flower buds, identify characteristic differential markers, and clarify relevant metabolic pathways, thereby guiding quality control, germplasm evaluation, and functional food development. Methods: Ultra-high performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS) was employed to identify metabolites. The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) and Cancer HSP were applied to screen the key active ingredients of traditional Chinese medicine (TCM-KAIs) and disease-related pharmaceutical ingredients (PDRIs). Specifically, we targeted six highly prevalent human diseases and another five disorders based on therapeutic indications documented in the Chinese Pharmacopoeia. Multivariate analyses, such as principal component analysis, hierarchical clustering analysis, and other statistical methods, were applied to investigate differential metabolites. The Kyoto Encyclopedia of Genes and Genomes (KEGG) database was utilized for pathway enrichment analysis of marker metabolites. Results: In total, 1550 metabolites were identified across 12 categories, predominantly flavonoids. Additionally, 152 TCM-KAIs and 204 PDRIs against 11 diseases were screened. Multivariate analyses indicated that geographical origin was closely associated with observed metabolic variation among the tested samples: Guangxi samples accumulated higher lipids and nucleotides, whereas Sichuan samples showed higher levels of flavonoids and phenolic acids. Vanilloloside, protocatechuic acid-4-O-glucoside, and gallic acid-4-O-glucoside were identified as key inter-group biomarkers. KEGG enrichment analysis revealed enhanced metabolism of nucleotide/pyrimidine in Guangxi, whereas zeatin biosynthesis was upregulated in Sichuan, consistent with discrepancies in regional climatic patterns. Conclusions: This study established a more comprehensive metabolomic dataset for FBSJvJ. It also clarified the correlations between origin and quality and unraveled the underlying mechanisms. These findings facilitate origin authentication, standardized quality control, and rational exploitation of FBSJvJ as raw materials of functional foods.

Formulation of a Stable Oil-in-Water Microemulsion of Torreya grandis cv. Merrillii Aril Essential Oil and Its Application in Loquat Fruit Preservation

He Wang,Yue Zheng,Xinyue Tang,Ting Zhang

Loquat is a nutrient-rich fruit with juicy and sweet pulp, but it is vulnerable to rot and deterioration without proper postharvest preservation measures. This study aimed to improve the postharvest quality of loquat by developing a microemulsion system based on an essential oil extracted from theTorreya grandiscv. Merrillii aril (TaEO), which has antimicrobial and antioxidant properties. An optimal TaEO microemulsion (TaEO-ME) was formulated, using a mixture of Tween-40 and Tween-80 as the surfactant, 1-butanol as the co-surfactant, and TaEO as the oil phase, with mass ratios of 9:1, 3:1, and 6:1, respectively. Two TaEO-ME formulations with 60% and 70% water contents were stable for 180 days at room temperature, with a mean droplet size below 12 nm and polydispersity index less than 0.24. They also exhibited higher stability and enhanced biological activities compared to free TaEO. Loquat fruit treated with TaEO-ME displayed a reduced decay index and lower membrane lipid peroxidation after 15 days of storage at 15 °C, as indicated by the lower malondialdehyde content and higher peroxidase activity. Moreover, the TaEO-ME treatment preserved the nutrient quality by maintaining the total phenolic compounds and ascorbic acid content. Our findings suggested that TaEO-ME can be used as a substitute for chemical preservatives to keep fruits fresh. Keywords:microemulsion;Torreya grandiscv. Width and length dependent SERS performance of core-shell Au@Ag nanorod self-assembled monolayers

Shuang Lin,Xiang Lin,Siqingaowa Han,Liang He,Haiyan Zhao,Jing Zhang,Wuliji Hasi,Li Wang

An efficient and facile strategy for producing core-shell Au@Ag nanorods (NRs) with controllable width and length as excellent surface enhanced Raman scattering (SERS) substrate was proposed. Au nanodipyramids (NDPs) of three various widths were prepared by overgrowth along the radial orientation, and together with Au NDPs as seed, silver was deposited in the axial orientation to successfully obtain core-shell Au@Ag NRs with diverse length (73 nm–157 nm) and width (25 nm–43 nm). The morphology, optical properties and SERS performance of the as-synthesized Au@Ag NRs were investigated systematically. The broadly tunable UV-Vis absorption range for Au@Ag NRs as far as the near-infrared (NR) region was able to be obtained. Furthermore, the SERS property of Au@Ag NRs two-dimensional monolayer was explored by using crystal violet (CV) as Raman probe. The optimal SERS enhancement effect with enhancement factor (EF) as high as 6.13✕10 5 could be achieved for Au@Ag NRs with wider width and longer length, which was confirmed by the simulation results of electromagnetic (EM) field distribution by FDTD method. Our novel approach provides some facinating insights into adjusting the width of Au@Ag NRs, which also significantly influences the SERS behaviors of the Au@Ag NRs. It allows to fabricating SERS substrate with superior SERS performance compared to previously reported Au@Ag NRs only with tunable length via direct overgrowth. Besides, this as-compounded Au@Ag NRs are expected to have promising application and significant value in other fields.

A novel aptasensor based on MUC-1 conjugated CNSs for ultrasensitive detection of tumor cells

Hongmei Cao,Daixin Ye,Qianqian Zhao,Juan Luo,Song Zhang,Jilie Kong

A novel strategy for the quantitative determination of human colon cancer DLD-1 cells utilizing an electrochemical aptasensor was developed by effective surface recognition between Mucin 1 glycoprotein over-expressed on the cell membrane and MUC-1 aptamer (MUC-1) bound on carbon nanospheres (CNSs). An MTT assay revealed that the as-prepared CNSs by green route exhibited satisfactory biocompatibility for cell viability, providing a suitable platform for the cell adhesion study. Furthermore, using CNSs as a sensing layer accelerated electron transfer and provided a highly stable matrix for the convenient conjugation of target MUC-1 aptamer, considerably amplifying the electrochemical signals. Electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) were applied to assess the optimal conditions and detection performance of the as-fabricated aptasensor. The attachment of colon cancer DLD-1 cells onto the MUC-1 aptamer immobilized CNSs led to increased EIS responses, which changed linearly in cell concentration ranging from 1.25 × 102 to 1.25 × 106 cells per mL with a lower detection limit of 40 cells per mL. With this method, colon cancer DLD-1 cells can be easily distinguished from normal cells, Human astrocytes 1800. The novel aptasensor revealed high specificity to DLD-1 cells. Furthermore, the aptasensor described here showed good reproducibility and high stability because of the CNSs of high stability and biocompatibility. The proposed protocols are a promising technique for the early monitoring of human colon cancer, and might have potential clinical applications such as cancer diagnosis, drug screening.

Dual carminic acid/hemin-marked DNA probes for simultaneously detecting CV-A16 and EV-A71 based on the mechanism of dimer to monomer transition

Lili Chen,Shuang You,Xiaotong Wang,Dong Li,Shuna Ren,Lihua Chen

This study aimed to prepare two hairpin-structure DNA probes by conjugating carminic acid (CA) or hemin into two ends of specific genes of coxsackievirus A16 (CV-A16) and enterovirus A71 (EV-A71) (probe CV-A16-CA and probe EV-A71-hemin ). Then, probe CV-A16-CA and probe EV-A71-hemin as the signal molecules were adsorbed onto NH 2 -MIL-53 (Al) (MOF). Based on these biocomposites, an electrochemical biosensor with dual-signal outputs for simultaneous assay of CV-A16 and EV-A71 was constructed. The stem-loops of probes switched both CA and hemin monomer to dimer, reducing the electrical activity of both CA and hemin. Subsequently, the target-induced opening of the stem-loop switched both CA and hemin dimers to monomers, resulting in two nonoverlapping increasing electrical signals. This sensitively reflected the concentration of target CV-A16 and target EV-A17 ranging from 10 −10 to 10 −15  M with a detection limit of 0.19 and 0.24 fM. This strategy was mainly applied to the simultaneous determination of target CV-A16 and target EV-A17 in 100% serum with satisfactory results. The MOF combined with the high loading capacity broke through the intrinsic limitation on sensitivity using the traditional methods. An increase of three orders of magnitude was observed. This study involved simple one-step detection, and only a simple replacement of a gene could trigger its potential in clinical and diagnostic applications.

Rosa chinensis cv. ‘JinBian’ flowers alleviates brain damage and cognitive deficit by inhibiting ferroptosis via the Keap1/Nrf2/GPX4 pathway and regulating gut microbiota

Li Zhen,Jianhong Peng,Huimin Cui,Junhong Tao,Yaping Liu,Jianxin Cao,Hesham R El-Seedi,Guiguang Cheng,Zhengxuan Wang

Background The flowers of Rosa chinensis cv. ‘JinBian’ (RCF) has been utilized as a herbal tea for millennia to prevent aging-related diseases. RCF is enriched in flavonoids and exhibits significant antioxidant effects for anti-aging potential. However, the specific bioactive compounds and their underlying anti-aging mechanisms remain unclear. Purpose This study aimed to investigate the bioactive ingredients and neuroprotective mechanisms of RCF extracts. Methods The study obtained dichloromethane (RD), ethyl acetate (RE), n-butanol (RB), and residual water (RW) fractions from RCF after liquid-liquid extraction. It investigated their neuroprotective effect in in vitro and in vivo experiments. UHPLC-ESI-HRMS/MS analysis was used to identify phytochemicals in RE. In vitro assays included measuring phenolic and flavonoid contents, ABTS⁺ and DPPH + scavenging capacities, and effects on H₂O₂-induced SHSY-5Y cells. In vivo , D-galactose-induced mice were used and assessed through the Morris water maze test and the crucifixion anxiety test. Molecular pathways and gut microbiota were analyzed. Results Among the fractions, RE had the highest phenolic and flavonoid contents, the strongest ABTS⁺ and DPPH + scavenging capacities, and significantly inhibited ROS accumulation by increasing antioxidants (glutathione, catalase, superoxide dismutase) in H₂O₂-induced SHSY-5Y cells. UHPLC-ESI-HRMS/MS analysis identified 28 phytochemicals in RE, primarily gallic acid derivatives and flavonoid derivatives. In D-galactose-induced mice, RE alleviated memory impairment and anxiety-like behavior. RE ameliorated brain damage by activating the Keap1/Nrf2-regulated antioxidant pathway (increasing T-AOC, GSH, GPX4, NQO1, SOD1, HO-1), suppressing ferroptosis, inhibiting acetylcholinesterase activity, attenuating GSK-3β/Tau/Bcl-2 axis-regulated apoptosis, and modulating AMPK-regulated autophagy. Additionally, RE improved gut microbiota diversity, particularly increasing beneficial bacteria such as Lactobacillus and Bifidobacterium . Conclusions Therefore, RE could alleviate the cognitive deficit associated with age-related neurodegenerative diseases by targeting ferroptosis-related oxidative stress, apoptosis, and autophagy, as well as modulating the gut microbiota.

Influence of leaf tenderness on the aroma and taste characteristics of green tea from Camellia sinensis cv. Chuancha No. 2: integrated sensory and chemical profiling

Chen Liu,Chenbo Wang,Mengyang Wei,Meiyi Ning,Zhiying Xu,Shengxiang Chen,Jilai Cui,Chuankui Song,Qian Tang

Leaf tenderness influences green tea quality, yet its role in flavour formation remains unclear. This study analysed green teas from Camellia sinensis cv. ‘Chuancha No. 2’ with three tenderness levels—single bud (C1B), one-bud with one-leaf (C1B1L), and one-bud with two-leaves (C1B2L)—via sensory evaluation, chemical profiling, and GC–MS analysis. C1B showed the best appearance and a chestnut-like, refreshing aroma. C1B1L featured floral notes and a mellow, umami taste, while C1B2L was stronger and more full-bodied. C1B had higher free amino acids and lower polyphenols and caffeine; C1B1L had the highest polyphenol content. Among 97 volatiles, 11 had ROAV > 1. Five common aroma compounds, namely 1-octen-3-ol, geraniol, hexanal, benzeneacetaldehyde, and 2-pentylfuran, were identified as key contributors to the aroma profile. Geraniol, most abundant in C1B1L, shaped its floral aroma. These origins are supported by established pathways, including lipid-derived oxidation products, glycoside-released geraniol, and amino-acid-derived benzeneacetaldehyde. This study clarifies how tenderness drives flavour divergence and identifies compounds supporting sensory variation in green tea.

Metal ions-enhanced classification of the origin and prediction of antioxidant activity of Chrysanthemum morifolium Ramat. cv. “Hangbaiju” by 3D fluorescence spectroscopy combined with chemometrics

Xueqing Zeng,Guanghua Lei,Juntao Hao,Yanmin Rao,Yuting Guan,Zeyuan Gong,Wanjun Long,Wei Lan,Hengye Chen,Haiyan Fu

This study selected Hangbaiju ( Chrysanthemum morifolium Ramat. cv. “Hangbaiju”) from 5 different origins as the research objects and investigated its 3D (three-dimensional) fluorescence characteristics of organic acids and flavonoids et al. closely related to the origins. Based on the chelating quenching effect between organic acids with Fe 3+ and the ESIPT (excited-state intramolecular proton transfer) effect between 3-hydroxyflavonoids with Al 3+ , a metal ion-enhanced method for antioxidant activity prediction and origin classification of Hangbaiju was established by using 3D fluorescence scanning technology combined with PARAFAC (parallel factor analysis) and RF (random forest) algorithm. The characteristic fluorescent components and their relative concentrations derived from PARAFAC serve as the data sources for RF origin classification and antioxidant activity prediction. The results show that, compared with the original 3D fluorescence spectra, the classification accuracy of Hangbaiju from 5 different origins based on Fe 3+ , Al 3+ -fluorescence spectra has increased from 91.0 % to 97.5 % and 100.0 % respectively. Meanwhile, after interacting with Fe 3+ and Al 3+ , the correlation coefficient between the antioxidant activity of Hangbaiju and its characteristic fluorescent components increased to above 0.85. Furthermore, the correlation coefficients of the antioxidant activity prediction results based on characteristic fluorescent components increased from 0.69 to 0.89 to 0.86–0.93 and 0.85–0.95 respectively, with the RMSE (root mean square error) less than 0.1045. This method ingeniously combines the dimensional advantages of 3D fluorescence with the precise digital separation performance of the second-order correction algorithm, independently isolating the characteristic fluorescence component information in Hangbaiju, achieving its satisfactory origin classification and antioxidant activity prediction results.

Peng Wang,Haifan Wang,Yang Xiao,Jialiang Zou,Hongping Chen,Lin Chen,Fu Wang,Yuan Hu,Youping Liu

In this study, comprehensive and systematic nontargeted metabolomics analysis was performed with the metabolites of Zangju peel ( Citrus reticulata cv. Manau Gan, CRZP, which has been cultivated for over 400 years in Derong County, China.) at four different mature stages. A total of 1878 metabolites were identified, among which flavonoids were the most abundant (62.04 %), and identified 62 key differential metabolites significantly affected by maturity. Based on biological activity measurements, CRZP showed better antioxidant activity, lipase inhibition ability, inhibition of adipogenic differentiation in 3TT-L1 cells and promotion of lipid metabolism, with the biological activity of CRZP at different maturity stages being associated with key differential metabolite. Thus, CRZP is natural antioxidants and possess anti-obesity potential, and industrial production needs to consider the Maturity stage of its collection.
