A sensitive electrochemical biosensor was constructed for Hg 2+ detection based on peonylike Cu-MOFs in situ growth of PtPd NPs bimetallic label S 1 and Mg 2+ -dependent DNAzyme-driven DNA Walker dual amplification signal strategy. The hairpin DNA (HP) and signal label [email protected] NPs/S 1 were first used to self-assemble on the gold nanorods/poly (diallyldimethylammonium chloride) functionalized graphene (AuNRs/PDDA-Gr) modified electrode, at this time, Cu ions can undergo a valence change to produce a larger initial signal. Here, we designed S 2 that can perform dual functions and contains two sequences, on was a substrate sequence of the Mg 2+ -dependent DNAzyme and the other was a sequence complementary to HP. In the presence of target Hg 2+ , HP was opened and hybridized with enzymatically sequenced DNA Walker (S 2 ) to preferentially form thymine-Hg 2+ -thymine (T-Hg 2+ -T). Then, DNA Walker exhibited catalytic activity of DNAzyme in the presence of Mg 2+ to continuously cleave [email protected] NPs/S 1 , resulting in a decrease of electrochemical signal. Under optimal conditions, the change of current was linearly related to the negative logarithm of the Hg 2+ concentration in the detection range of 0.001–100 nM with a low detection limit of 0.52 pM. Moreover, the proposed aptasensor was successfully applied in detection of milk powder samples.

DNA walkers have been widely used to build biosensing systems. However, their moving efficiency was limited, which has hindered practical applications. Here, we for the first time integrated localized CHA with nanoscale AuNPs scaffolds to achieve space double-confined catalytic hairpin assembly (SDCHA) for developing a fast three-dimensional DNA walker. Driven by spatially proximal displacement, the walker possessed a high walking speed, which was about 8 times, 9 times and 23 times faster than LCHA, CHA-AuNPs and conventional CHA, respectively. Moreover, the walking time was about 1.4–8.3 times shorter than most recently reported DNA walkers and space-confined strand displacement reactions. By further integrating it with magnetic probes and g-C 3 N 4 nanosheets , an ultrasensitive and enzyme-free homogeneous electrochemiluminescence (Ho-ECL) aptasensor was firstly constructed for sonic hedgehog (SHh) detection. Profiting from this tailored integration, the Ho-ECL aptasensor avoided multiple interferences from complex milieu as well as tedious procedures of electrode modifying and washing, showing good signal stability, repeatability and reliability. It was also proved to achieve ultrasensitive SHh detection with the detection limit as low as 13.5 fM, superior to previous research on SHh detection. This work might broaden ideas for the construction of fast DNA walkers and facile Ho-ECL bioanalytical methods for clinical diagnosis.

MicroRNAs (miRNAs) play an important role in the process of heart failure (HF) and are emerging biomarkers that can be used for the auxiliary diagnosis of HF. However, it is very challenging to accurately analyze the expression levels of trace miRNAs in complex clinical samples. Here, we developed an enzyme-free colorimetric sensor for the ultrasensitive detection of miRNA-423-5p (HF-associated miRNA) based on three-dimensional DNA walkers constructed from functional nucleic acids and gold nanoparticles (AuNPs). DNAzyme with cleavage activity was specifically activated by miRNA-423-5p to sustainably cleave the substrate, thereby releasing the trigger sequence to initiate the subsequent mismatched catalytic hairpin assembly (MCHA) cycle. Then, as the MCHA cycle proceeded to continuously expose the G-quadruplex (GQ) sequence, the sequence bound with hemin to form a large amount of GQ/hemin DNAzyme on the surface of the AuNPs, which rapidly catalyzed the chromogenic oxidation of 3,3′,5,5′-tetramethylbenzidine to yield an amplified colorimetric signal readout. The colorimetric sensor exhibited an ultralow detection limit (32 fM), showed excellent specificity and performed well in serum samples. The sensor was applied to detect miRNA-423-5p in clinical plasma samples from healthy individuals and HF patients, and the results revealed its good clinical application in HF diagnosis. Thus, the developed colorimetric sensor provides a convenient detection tool for early screening and diagnosis of HF, as well as for pathophysiological studies.

DNA walkers have been recognized as a type of powerful signal amplification tool for biosensors, but how to adopt a proper strategy to increase their amplification efficiency is still highly desirable. Herein we design a dual-catalytic hairpin assembly (CHA)-mediated strategy for the high-efficient formation of a tripedal Mg 2+ -dependent DNAzyme (MNAzyme)-DNA walker, and thus develop a novel proteinase-free dual-mode biosensing method for the kanamycin (Kana) antibiotic assay. The first CHA is initiated by a target-biorecognition reaction, which can produce the DNA walker and also induce the target recycling. The second CHA is initiated by a special base sequence designed as a one-half substrate of the MNAzyme. Upon the first CHA-triggered DNA walking at a magnetic bead (MB) track, this “pseudo-target” sequence can be released to induce another CHA-cycle for the formation of the same DNA walker. Meanwhile, the other one-half substrate strand exposed on the MB surface will trigger the quantitative hybridization chain reaction (HCR)-assembly of a G-quadruplex DNAzyme (G-DNAzyme)-enriched double-stranded DNA polymer. So the enzymatic reaction of G-DNAzymes enabled the convenient colorimetric and photoelectrochemical dual-mode signal transduction of the method. Due to the dual-CHA facilitation to the tripedal and three-dimensional DNA walking and synergetic signal amplification of HCR, this method exhibits very low detection limits of 9.4 and 0.55 fg mL −1 , respectively. In combination with its wide linear range, automated manipulation, and excellent selectivity , repeatability and reliability, the proposed method is expected to be used for the convenient semiquantitative screening and accurate determination of possible antibiotic residues in complicated matrices.

A sensitive dual-target electrochemical biosensor was developed integrating circular bipedal DNA walkers with a three-dimensional hollow carbon nanospheres-tetrametallic nanoparticle (3D HCNs-TT NPs) modified platform, enabling concurrent detection of two critical epidermal growth factor receptor (EGFR) exon 19 mutations: E746_A750 deletion (Mut DNA A) and L747_S752delinsS (Mut DNA B). The circular bipedal DNA walkers address the limitations of low amplification efficiency in traditional single-legged DNA walkers and mitigate the sensitivity reduction caused by excessively long walking strands. Additionally, we used the 3D HCNs-TT NPs nanocomposite materials to modify the electrodes, which significantly increased the electron transfer rate and provided more active sites for molecular probe loading, thereby further enhancing the performance of the detection platform. This biosensor demonstrated good performance, with detection limits as low as 3.4 fM (Mut DNA A) and 2 fM (Mut DNA B) (S/N = 3). The designed sensor was applied for detection using clinical samples, including cells with gene mutations and diluted human serum, and these results confirm the sensor’s potential for clinical translation, offering a potential tool for early non-small cell lung cancer (NSCLC) diagnosis and personalized tyrosine kinase inhibitors (TKI) therapy guidance. In this paper, a photoelectrochemical (PEC)-surface-enhanced Raman scattering (SERS) dual-mode biosensor is constructed coupled with a dual-recognition binding-induced DNA walker with a carbon nitride nanosheet (C3N4)/MXene-gold nanoparticles (C/M-Au NPs) accelerator, which is reliable and capable for sensitive and accurate detection of Staphylococcus aureus (S. aureus). Initially, a photoactive heterostructure is formed by combining C3N4 and MXene via a simple electrostatic self-assembly as they possess well-matched band-edge energy levels. Subsequently, in situ growth of gold nanoparticles on the formed surface results in better PEC performance and SERS activity, because of the synergistic effects of surface plasmon resonance and Schottky barrier. Furthermore, a three-dimensional, bipedal, and dual-recognition binding-induced DNA walker is introduced with the formation of Pb2+-dependent DNAzyme. In the presence of S. aureus, a significant quantity of intermediate DNA (I-DNA) is generated, which can open the hairpin structure of Methylene Blue-tagged hairpin DNA (H-MB) on the electrode surface, thereby enabling the switch of signals for the quantitative determination of S. aureus. The constructed PEC-SERS dual-mode biosensor that can be mutually verified under one reaction effectively addresses the problem of the low detection accuracy of traditional sensors. Experimental results revealed that the effective combination of PEC and SERS is achieved for amplification detection of S. aureus with a detection range of 5–108 CFU/mL (PEC) and 10–108 CFU/mL (SERS), and a detection of limit of 0.70 CFU/mL (PEC) and 1.35 CFU/mL (SERS), respectively. Therefore, this study offers a novel and effective dual-mode sensing strategy, which has important implications for bioanalysis and health monitoring.
