Preparation and characterization of PEGylated magnetic ferrite with a biocompatible delivery for DOX

Ding Feng,Qiu Guobin,Tan Fengliang,Wu Tengyan,Ding Longzhen,Liu Xin,Zhong Hongbin

Characterization results of Fourier transformed infrared spectrometer(FT-IR), thermogravimetry–differential thermal analysis TG–DTA, dynamic stability control (DSC), transmission electron microscopy (TEM), and vibrating sample magnetometer (VSM) indicated that DOX loaded onto PEGylated superparamagnetic Mn 0.6 Zn 0.4 Gd 0.04 Fe 1.96 O 4 (MNPs) was prepared successfully. DOX loading ration and release mechanism was analyzed; the result indicated that the DOX loading ration increased with initial charge DOX increasing, and DOX release was a Fickian diffusion mechanism, with 34.5% of DOX released in PBS solution at 43 °C within 168 h. Consequently, the MNPs@PEG may be a good targeted drug delivery system for tumor targeted and hyperthermia MNPs as therapeutic applications.

Novel catalytic micromotor of porous zeolitic imidazolate framework-67 for precise drug delivery

Linlin Wang,Hongli Zhu,Ying Shi,You Ge,Xiaomiao Feng,Ruiqing Liu,Yi Li,Yanwen Ma,Lianhui Wang

Micromotors hold promise as drug carriers for targeted drug delivery owing to the characteristics of self-propulsion and directional navigation. However, several defects still exist, including high cost, short movement life, low drug loading and slow release rate. Herein, a novel catalytic micromotor based on porous zeolitic imidazolate framework-67 (ZIF-67) synthesized by a greatly simplified wet chemical method assisted with ultrasonication is described as an efficient anticancer drug carrier. These porous micromotors display effective autonomous motion in hydrogen peroxide and long durable movement life of up to 90 min. Moreover, the multifunctional micromotor ZIF-67/Fe3O4/DOX exhibits excellent performance in precise drug delivery under external magnetic field with high drug loading capacity of fluorescent anticancer drug DOX up to 682 μg mg−1 owing to its porous nature, high surface area and rapid drug release based on dual stimulus of catalytic reaction and solvent effects. Therefore, these porous ZIF-67-based catalytic micromotors combine the domains of metal–organic frameworks (MOFs) and micomotors, thus developing potential resources for micromotors and holding great potential as label-free and precisely controlled high-quality candidates of drug delivery systems for biomedical applications.

Construction of Fe3O4-Loaded Mesoporous Carbon Systems for Controlled Drug Delivery

Guowen Qian,Lemin Zhang,Xia Li,Cijun Shuai,Xiupeng Wang

Magnetite (Fe3O4) nanoparticles as drug carriers can achieve precise drug target due to their magnetic property. However, they are easy to aggregate in the physiological environment, which obviously limits their application in drug delivery. The development of the Fe-MIL-88B-derived method to construct the Fe3O4-loaded mesoporous carbon (Fe3O4/carbon) system is a feasible strategy to solve the issue. First, iron atoms evenly distribute in the organic links through coordination bonds in Fe-MIL-88B. After the carbonization of Fe-MIL-88B, mesoporous carbon acts as a barrier to prevent the aggregation of Fe3O4 nanoparticles. Herein, Fe-MIL-88B particles were fabricated by the hydrothermal method and then pyrolyzed to construct Fe3O4/carbon systems. Results showed that Fe3O4 nanoparticles uniformly in situ grew on mesoporous carbon generated by the carbonization of organic components. More encouragingly, the Fe3O4/carbon system loaded with DOX demonstrated pH-responsive DOX release, efficient delivery of DOX into cancer cells, and significant cancer cell killing ability. Therefore, the Fe3O4/carbon systems prepared by the Fe-MIL-88B-derived method might open up a way for targeted and controlled drug delivery.

Polydopamine-drug conjugate nanocomposites based on ZIF-8 for targeted cancer photothermal-chemotherapy

Yuxuan Ma,Chenwei Wang,Lvming Zhu,Chunmei Yu,Bing Lu,Yang Wang,Yue Ding,Chang-Ming Dong,Yong Yao

Stimuli-responsive prodrug-based nanoplatform with synergistic antitumor activity is of central importance to the development of promising nanomedicines for cancer therapy. Here, we describe a polydopamine-drug conjugate nanocomposite (ZP-PDA-DOX) with targeted cancer photothermal-chemotherapy (PTT-CT), which constructed by a gradual copolymerization of dopamine (DA) and pH-sensitive dopamine-derived prodrug (DA-DOX) into the porous channels of zeolite imidazolate frameworks-8 (ZIF-8), followed by PEGylation with amino-terminated folic acid-polyethylene glycol (NH 2 -PEG-FA) to acquire the high biocompatibility, specificity, and excellent tumor-targeting property. The incorporation of polydopamine strengthened the stability and dispersion of ZIF-8, and also conferred photothermal conversion effect. In the tumor acidic microenvironment, the acid-labile hydrazone linker of DA-DOX and ZIF-8 promptly degraded to release activated DOX. Moreover, the generated hyperthermia due to the high photothermal conversion efficiency of PDA component could accelerate drug release, and simultaneously thermally ablate tumor tissue to maximize the DOX-induced CT, which could also assist PTT to eradicate tumor cells. This study provides a promising strategy for targeted cancer PTT-CT with synergistic anti-tumor effect.

Synthesis of drug-loaded H-ZIF-8@CaCO3-PEG nanocarrier for synergistic therapy

Han Song,Jiasheng Fan,Shengkai Liu,Boyu Chen,Jin Huang,Yujie Fu,Zhiguo Liu

In this study, we synthesized DOX-loaded H-ZIF-8@CaCO 3 -PEG nanocarrier, which combined with calcium overload and photothermal therapy (PTT) for synergistic therapy. The nanocarrier comprised a PEG modified calcium carbonate (CaCO 3 ) shell and a hollow imidazolate zeolite skeleton (H-ZIF-8) core, in which the chemotherapy drug doxorubicin hydrochloride (DOX) and photosensitizer indocyanine green (ICG) were simultaneously co-loaded. In vitro cellular experiments revealed that the lethality of DOX&ICG@H-ZIF-8@CaCO 3 -PEG against tumor cells was significantly higher than that of the single chemotherapy owing to the combination with chemotherapy and photothermal therapy as well as calcium overloading. This drug-loaded nanocarrier exhibits notable characteristics such as exceptional stability, elevated drug loading capacity and efficient release of the loaded drug, which provides a promising perspective for efficient synergistic therapy.

Preparation of environmentally responsive PDA&DOX@LAC live drug carrier for synergistic tumor therapy

Liu Lu,Zhao Xuefen

The development of intelligent, environmentally responsive and biocompatible photothermal system holds significant importance for the photothermal combined therapy of tumors. In this study, inspired by Lactobacillus (LAC), we prepared a biomimetic nanoplatform PDA&DOX@LAC for tumor photothermal-chemotherapy by integrating the chemotherapeutic drug doxorubicin (DOX) with dopamine through oxidative polymerization to form polydopamine (PDA) on the surface of LAC. The PDA&DOX@LAC nanoplatform not only achieves precise and controlled release of DOX based on the slightly acidic microenvironment of tumor tissues, but also exhibits enzyme-like properties to alleviate tumor hypoxia. Under near-infrared light irradiation, it effectively induces photothermal ablation of tumor cells, enhances cellular uptake of DOX with increasing temperature, and thus efficiently inhibits tumor cell growth. Moreover, it is further confirmed in vivo experiments that photothermal therapy combined with PDA&DOX@LAC induces tumor cells apoptosis, releases tumor-associated antigens, which is engulfed by dendritic cells to activate cytotoxic T lymphocytes, thereby effectively suppressing tumor growth and prolonging the survival period of 4T1 tumor-bearing mice. Therefore, the PDA&DOX@LAC nanoplatform holds immense potential in precise tumor targeting as well as photothermal combined therapy and provides valuable insights and theoretical foundations for the development of novel tumor treatment strategies based on endogenous substances within the body.

Photothermal-Chemotherapeutic Microneedle Patch Coloaded with HMnO2 and DOX for Postoperative Management of Melanoma

Zhang Xuewei,Zhang Yixun,Mao Lujia,Zhou Xin,Huang Jie,Kang Deni,Liu Sirong,Chen Mingjuan,Yang Ronghua

Surgical resection remains the primary mode of treatment for melanoma. However, postoperative tumor recurrence and impaired wound healing significantly compromise the patient prognosis. In this study, we developed a microneedle (MN) patch coloaded with HMnO 2 and hydroxydaunorubicin (DOX) for the simultaneous management of residual disease and postsurgical wound repair. The MN patch enables efficient transdermal delivery of both agents by penetrating the stratum corneum and targeting the surgical site. Upon near-infrared (NIR) irradiation, HMnO 2 mediates photothermal ablation (PTT), effectively ablating residual melanoma cells while DOX suppresses the proliferation of potential micrometastases. Furthermore, the HMnO 2 /DOX@MN system modulates the local inflammatory microenvironment, promotes angiogenesis, and enhances collagen deposition, thereby accelerating the wound healing. In vivo studies using a murine melanoma model demonstrated that the MN patch significantly reduced postoperative tumor recurrence and improved wound closure rates. This multifunctional microneedle platform presents a promising strategy for integrated postoperative management, following melanoma resection.

Organic Functionalization of Mesoporous Silica Spheres as a Nanovehicle for DOX pH-Triggered Delivery

Shuai Wang,Fang Xiang Song,Li Zhang,Xue Zhang,Yan Li

Mesoporous silica (MS) spheres of different sizes with pH-responsive characteristics were synthesized based on Stöber’s theory. Organic functionalization with aminopropyl and carboxyl groups resulted in different materials, namely, MS@NH 2 @COOH. MS@NH 2 @COOH were observed to have a large number of carboxyl groups and multiamine chains, and were grafted into pore channels and pore outlets through systematic characterization analyses. All modified samples demonstrated the controlling of the delivery rate of DOX from the siliceous matrix. We also compared the drug release behavior of the DOX-loaded materials at high pH (7.4) and low pH (5.5) and studied the cytotoxicity on A549 cells. The experimental results indicated that the drug delivery system can better control drug release and have potential applications in the drug delivery field.

Efficient reduction and pH co-triggered DOX-loaded magnetic nanogel carrier using disulfide crosslinking

Juan Huang,Yanan Xue,Ning Cai,Han Zhang,Kaikai Wen,Xiaogang Luo,Sihui Long,Faquan Yu

To reduce leakage on the drug-delivery pathway to minimize side effect of reduction or pH sensitive drug delivery systems, we designed a glutathione (GSH)/pH co-triggered magnetic nanogel drug delivery system for doxorubicin (DOX) based on the GSH concentration and pH difference between intracellular and extracellular environments. The introduction of superparamagnetic iron oxide nanoparticles (SPION) was intended for magnetic targeting. The magnetic DOX-loaded nanogel was then prepared by the oxidation of thiolated alginate with thiolated SPION in the presence of DOX. The nanogel size can be readily regulated in a range of 120–320   nm upon preparation conditions, with a negative surface charge of around −   40   mV. Saturation magnetization was estimated at 27.4   emu/g Fe by VSM. In vitro release was conducted in simulated cancerous environment conditions such as a high GSH concentration and mild acidity. As a result, the nanogel expressed, upon dual stimuli of pH   5/10   mM GSH, significantly higher accumulative release than upon single stimulus of pH   5 without GSH or pH   7.4/10   mM GSH. In vitro cytotoxicity against HeLa cells clearly illustrated that the nanogel could effectively inhibit cell growth, and the IC 50 was figured out to be 2.3   μg/mL of the nanogel, while the nanogel exclusive of DOX was nontoxic. Confocal laser scanning microscopy observation, combined with the result of Prussian blue staining, indicated that DOX was efficiently internalized into HeLa cells through endocytosis, released into the cytoplasm, and then principally entered the nuclei. The quantitative examination of the iron content revealed an exponential increase in the cellular uptake and an exponential decrease in the uptake efficiency with the fed nanogel. This drug-loaded nanogel could be a promising drug carrier for effective tumor-targeted chemotherapy.

Synergistic chemotherapeutic effect of sorafenib-loaded pullulan-Dox conjugate nanoparticles against murine breast carcinoma

Junhui Sui,Yani Cui,Hanxu Cai,Shaoquan Bian,Zhiyi Xu,Ling Zhou,Yong Sun,Jie Liang,Yujiang Fan,Xingdong Zhang

pH-Sensitive pullulan-doxorubicin conjugates encapsulating sorafenib (P-Dox/S) nanoparticles were developed as a synergistic combinatorial delivery system against murine breast carcinoma. The nanoparticles can encapsulate Dox and sorafenib with ultra-high loading capacity (65.34 wt%) through chemical conjugation and physical loading, whereas can remain stable under physiological conditions and gradually release Dox and sorafenib with the decreasing pH. These conjugates can be effectively internalized and clearly suppress 4T1 cell growth in vitro. Furthermore, research data of in vivo animal models revealed that the synergistic combinatorial P-Dox/S nanoparticles heavily accumulated in solid tumor tissue sites to maximize therapeutic efficacy; they also significantly inhibited solid tumor growth, even remarkably reduced solid tumor volume in comparison to the initial volume, and obviously diminished adverse effects. The anti-tumor therapeutic effect obviously outperformed the delivery of combinational chemotherapy of free drugs or single drug-loaded P-Dox nanoparticles at the same concentration. These promising results indicate the high-efficiency synergistic chemotherapeutic effects of these nanoparticles. Combinational chemotherapy using P-Dox/S nanoparticles has important potential in the clinical treatment of malignancy for overcoming drug resistance and heterogeneity.

Near-infrared mediated chemo/photodynamic synergistic therapy with DOX-UCNPs@mSiO2/TiO2-TC nanocomposite

Ruihan Tong,Huiming Lin,Yuhua Chen,Na An,Gang Wang,Xiaofan Pan,Fengyu Qu

To integrate photodynamic therapy (PDT) with chemotherapy for enhanced anticancer efficiency, near infrared (NIR) light mediated upconversion nanoparticles@mesoporous silica (UCNPs@mSiO 2 ) nanovehicle was constructed as the nanocarrier. Based on the novel ultraviolet (UV) emission, TiO 2 (3.0–3.2   eV) was adopted as the photosensitiver (PS) on account of the high activity and excellent stability. Here, a simple host-assembly method was exploited to graft it onto the inner and outer surface of the mSiO 2 shell. On the other hand, a UV cleavable o -nitrobenzyl derivative linker (TC linker) was prepared as “gate” to encapsulate anticancer agent doxorubicin (DOX) inside mSiO 2 , insuring the few prematurity. Under NIR irradiation, the UV emission can excite TiO 2 to produce reactive oxygen species (ROS) and also can induce the photodegradation of TC linker and drug release as well. The detailed release kinetics was studied to reveal that it is TC linker to manipulate the NIR-sensitive drug release and the amount of TC linker also can be used to adjust the release performance. The synergistic action of PDT and chemotherapy displays the improved cytotoxicity of DOX-UCNPs@mSiO 2 /TiO 2 -TC to HeLa cells under NIR irradiation that makes the potential application in anticancer field due to the superior to any single means.

SPIONs/DOX loaded polymer nanoparticles for MRI detection and efficient cell targeting drug delivery

Le Shang,Qiu-yue Wang,Kang-long Chen,Jing Qu,Qing-han Zhou,Jian-bin Luo,Juan Lin

In this research, reducible polydopamine coated magnetic nanoparticles (SPIONs@PDA) were prepared and doxorubicin (DOX) was encapsulated for both magnetic resonance imaging (MRI) detection and cell targeting drug delivery. The SPIONs@PDA exhibited excellent water-solubility and colloidal stability, because the coating of the magnetic nanoparticle was a crosslinked reducible polydopamine synthesized with N,N-bis(acrylate) cystamine (BACy) as a crosslinker, dopamine methacrylamide (DMA) as an anchor to immobilize SPIONs, and polyethylene glycol (PEG) to provide colloidal stability. The SPIONs@PDA showed reduction-sensitivity due to the reducible disulfide bond in the polymer network. Subsequently, a sustained and high cumulative drug release in glutathione (GSH) was observed, rather than the low drug release in PBS (pH 7.4). Fluorescent imaging studies by confocal laser scanning microscopy indicated that the prepared SPIONs@PDA could specifically target tumor cells. The results of the cytotoxicity assays further demonstrated that the DOX-loaded SPIONs@PDA presented efficient antitumor activity to HeLa cells. These favorable properties suggested SPIONs@PDA as promising targeted magnetic nanoparticles for MRI detection and stimuli-triggered drug delivery in cancer theranostics.
