Fish selectively reserves docosahexenoic acid (DHA) in ovary during gonadal development . However, no direct proof supports this. The present study tried to elucidate the DHA migration to the developing ovary of female zebrafish. An injection study of 13 C-labeled DHA for DHA tracing was conducted, and another injection study of unlabeled-DHA (DHA-injected group) and BSA-saline (control group) was conducted for lipid and DHA content detection, related gene expression analyses, and histological observation. The results showed that the rapid absorption of lipid occurred at stage III with a constant accumulation of DHA in the ovary. The proportion of oocytes at stage III on day 7 and 21, and at stage IV on day 3 and 21 in DHA-injected group was significantly higher than that in control group, respectively ( P < .05). The injected 13 C-labeled DHA was accumulated twice in the ovary respectively on day 1 and 7, and remained at a relatively high level. In DHA-injected group, the fatp4 expression was significantly higher in ovary on day 3, 5 and 7 ( P < .05), and significantly lower ( P < .05) in liver on day 5, 14 and in muscle on day 1, 5 and 7 than that in control group. In conclusion, the present study suggested a migration of DHA from the liver and muscle to the gonads when necessary.

Although it has been widely known that arsenic poses great threat to organisms and arsenic species account for the toxic effect of the metalloid, relatively little attention has been given to the arsenic toxicity to different organisms at different pH values. Three kinds of aquatic organisms including Photobacterium phosphoreum , Daphnia magna , and Danio rerio were used to study the effects of pH on the toxicity of arsenic. Toxicity test results were analyzed by calculating EC 50 values and for arsenic species at five different pH values (5.0, 6.0, 7.0, 8.0, and 9.0). As a result, As(III) is more toxic to the tested organisms than As(V), and P. phosphoreum was more sensitive to the toxicity of arsenic than D. magna and D. rerio . The toxicity of As(III) species is different for different organisms: dihydrogen arsenite, H 2 AsO 3 – , is more toxic to P. phosphoreum and D. rerio than arsenous acid, H 3 AsO 3 , while H 3 AsO 3 is more toxic to D. magna . As for As(V), arsenate, HAsO 4 2− , is the most toxic to P. phosphoreum , whereas dihydrogen arsenate, H 2 AsO 4 – , may be responsible for the toxicity to D. magna and D. rerio . As(III) and As(V) are more toxic to P. phosphoreum in an alkaline environment, while for D. magna in an acidic environment and As(III) is more toxic to D. rerio in an alkaline environment and As(V) in an acidic environment.

Chlorantraniliprole (CAP), a representative bisamide insecticide, is widely used in rice fields around the world, posing potential toxicity risks to aquatic organisms . In this study, we examined the effects of exposure to CAP on growth and metabolic phenotype of zebrafish ( Danio rerio ) and oxidative stress and apoptosis in the liver of zebrafish ( Danio rerio ). First, we identified that CAP had a low bioaccumulation in zebrafish. Subsequently, growth phenotype analysis revealed that CAP could significantly increase liver weight and liver index in zebrafish. In addition, we found that CAP exposure could cause significant changes in indicators of oxidative stress, resulting in a significant increase in the content of malondialdehyde (MDA), causing oxidative stress in the liver of zebrafish. Meanwhile, the expression levels of apoptosis-related genes were also significantly changed and apoptosis was promoted in the liver of zebrafish with CAP exposure. Importantly, the results of metabolomics analysis shown that CAP exposure could significantly disrupt the metabolic phenotype of zebrafish, interfering with multiple metabolic pathways, mainly including valine , leucine and isoleucine biosynthesis and degradation, alanine , aspartate and glutamate metabolism and d -glutamine and D-glutamate metabolism. Last but not least, correlation analysis identified strong links between changes in liver function involving oxidative stress and apoptosis and changes in metabolic phenotype of zebrafish following CAP exposure. In brief, these results indicate that potential environmental risks of CAP to aquatic organisms should receive more attention.

Pefloxacin (PFLX) is a type of fluoroquinolone (FQ) that has been widely used in aquaculture and animal husbandry. It is also a new kind of pseudo-persistent pollutant, which has some toxic effects. In this study, zebrafish (Danio rerio) embryos were used as exposure models to analyze the toxicity of PFLX. The results showed that PFLX was absorbed on the outer membrane surface at the beginning, and then a few PFLX slowly entered the cytoplasm. The adsorption complied with the Langmuir isotherm adsorption model. The surface morphology of the embryonic membrane was changed. PFLX exposure had no effect on fetal autonomous movement, but it significantly inhibited the embryo hatching rate. After PFLX exposure, the embryo and larval survival rates showed a clear dose–effect relationship. Though PFLX exposure did not cause teratogenic toxicity, it still affected cardiovascular function. The potential toxicity mechanism and developmental toxicity of PFLX to zebrafish were revealed. These results could be important for studies concerning FQ toxicology. Keywords:pefloxacin;zebrafish;embryonic membrane;developmental toxicity

The possible detrimental effects of nanomaterials on organisms have become a public concern because of the wide applications of nanomaterials in modern society. This study investigates the effects of silica nanoparticles (nano-SiO 2 ) on zebrafish development. Fluorescent nano-SiO 2 (FITC-nano-SiO 2 ) is synthesized with an emission wavelength of 517 nm. Due to similar hydrodynamic size, FITC-nano-SiO 2 is used to simulate the distribution of nano-SiO 2 in zebrafish. Nano-SiO 2 regulated col2a1a , col9a2 , lect1 , and her12 to delay liver development and regulated prox , wnt2bb , mypt1 , and hhex caused spinal curvature. The hand2 , mef2a , nkx2.5 , and atp1a2a are significantly down-regulated. The whole embryo in situ hybridization revealed amhc and flk1 are also down-regulated. This suggests that nano-SiO 2 affected heart and blood vessels development of zebrafish. Furthermore, it is found that cell apoptosis occurred in the heart. The proapoptotic genes ( bax , bid ) are upregulated, and the antiapoptotic genes ( bcl-2 , mcl-1b ) are down-regulated. Nano-SiO 2 increases the endoplasmic reticulum (ER) stress-related genes ( chop , bip , perk, elF2α ), indicating that the ER stress is involved in cell apoptosis. This study provides a toxicological basis for evaluating the possible hazards of nano-SiO 2 to aquatic organisms.

Chlorpyrifos is a widely used organophosphate insecticide and ubiquitously detected in the environment. However, little attention has been paid to its endocrine disrupting effect to non-target organisms. In the present study, zebrafish was exposed to 13 and 65 μg/L of chlorpyrifos for 7 and 10 days to determine the induced neurotoxicity and the alteration of thyroid metabolism. The 120 h LC 50 and LC 10 of chlorpyrifos was estimated as 1.35 mg/L and 0.62 mg/L based on the acute embryo toxicity assay, respectively. The acetylcholinesterase (AChE) inhibitory was detected by 13 μg/L chlorpyrifos and could be reversed by the co-exposure of 100 and 1000 μg/L anticholinergic agent atropine. For thyroid hormone level, 13 and 65 μg/L of chlorpyrifos induced increased free T 3 levels in 10 dpf (days post-fertilization). The expression of thyroid related genes in 7 and 10 dpf exposed zebrafish were measured by the quantitative Real-Time PCR (qRT-PCR) assay. The mRNA expression of tshba , thrb , crhb , ttr , tpo , ugt1ab and slc5a5 had significant change. However, the alterations of thyroid hormone and mRNA expression could be partly rescued by the addition of atropine. The molecular docking of chlorpyrifos and T 3 to the thyroid receptor β in zebrafish using homology modelling and CDOCKER procedures shown weaker binding ability of chlorpyrifos compared to T 3 . Therefore, we concluded that the disturbance of thyroid signaling in zebrafish might arise from the developmental neurotoxicity induced by chlorpyrifos.

Thimerosal (THI) is the most widely used form of organic mercury in pharmaceutical and personal care products, and has become a major source of ethylmercury pollution in aquatic ecosystems. However, knowledge about its potential risk to aquatic species is limited. In this study, zebrafish were exposed to THI for 7 days, and variations in their behavioral traits, brain monoaminergic neurotransmitter contents, and related gene expression were investigated. After the 7-day exposure, THI reduced locomotor activity and thigmotaxis in males but not females. Exposure to THI increased the social interaction between females but decreased that between males. The THI exposure also significantly reduced the serotonin (5-HT), 5-hydroxyindoleacetic acid, dopamine (DA), and 3,4-dihydroxyphenylacetic acid contents in the brain of males, but only significantly decreased the DA content in females. Correlation analysis revealed that the neurochemical alterations in the brain of zebrafish play critical roles in the behavioral abnormalities induced by THI exposure. Moreover, THI also significantly altered the expression of some genes associated with the synthesis, metabolism, and receptor binding of 5-HT and DA in the brain of zebrafish. The differences in these gene expressions between female and male zebrafish exposed to THI seem to be an important mechanism underlying their sex-specific responses to this chemical. This is the first report on the sex-specific effects of THI on behaviors and brain monoaminergic neurotransmitter contents in zebrafish, which can further improve our understanding of its toxic effects on teleost.

Ochratoxin A (OTA) has been increasingly detected in foodstuffs and human samples. However, the molecular mechanisms of developmental toxicity induced by OTA remain poorly explored. Our results demonstrated that oxidative stress induced by OTA caused mitochondrial dysfunction in larvae, characterized by malformed mitochondria, and decreased mitochondrial membrane potential and complex II, IV and V activities. Central carbon metabolism analysis indicated that exposure to OTA induced energy metabolism disturbance by decreasing ATP contents and altering energy-relevant metabolite contents. Energy metabolism disturbance was further confirmed by transcriptomics analysis, with differentially expressed genes significantly enriching in glycolysis and TCA cycle. Additionally, molecular docking simulations and surface plasmon resonance analysis demonstrated that OTA interacted with mfn1 and mfn2, verifying its interference effects on mitochondrial dynamics. The insufficient energy supply induced by OTA activated AMP-activated protein kinase signal pathways, which accelerated catabolic processes, accompanied by decreases in yolk sac size and yolk lipid. Finally, insufficient energy supply caused developmental retardation of zebrafish larvae. Collectively, this study systematically elucidated the roles of mitochondrial dynamics imbalance and energy dyshomeostasis in the developmental toxicity induced by OTA, providing critical insights for refining food safety standards and therapeutic interventions.

Microplastics (MPs) pollution, together with its consequential effect on aquatic biota, represent a burgeoning environmental concern that has garnered significant scholarly attention. Thiamethoxam (TMX), a prevalently utilized neonicotinoid insecticide, is renowned for its neurotoxic impact and selective action against targeted pests. The aquatic environment serves as a receptacle for numerous pollutants, such as MPs and neonicotinoid insecticides. However, there is currently a lack of comprehensive understanding regarding the toxic effects of co-exposure to aged MPs and neonicotinoid insecticides in aquatic organisms. Therefore, we endeavor to elucidate the deleterious impacts of aged polystyrene (PS) and TMX on zebrafish ( Danio rerio ) larvae when present at environmentally relevant concentrations, and to reveal the underlying molecular mechanisms driving these effects. Our study showed that exposure to aged PS, TMX, or their combination notably inhibited the heart rate and locomotion of zebrafish larvae, with a pronounced effect observed under combined exposure. Aged PS and TMX were found to diminish the activity of antioxidative enzymes (SOD, CAT, and GST), elevate MDA levels, and disrupt neurotransmitter homeostasis (5-HT, GABA and ACh). Notably, the mixtures exhibited synergistic effects. Moreover, gene expression related to oxidative stress (e.g., gstr1, gpx1a, sod1, cat1, p38a, ho-1, and nrf2b ) and neurotransmission (e.g., ache, ChAT, gat1, gabra1, 5ht1b, and 5ht1aa ) was significantly altered upon co-exposure to aged PS and TMX in larval zebrafish. In summary, our findings support the harmful effects of aged MPs and the neonicotinoid insecticides they carry on aquatic organisms. Results from this study enhance our understanding of the biological risks of MPs and insecticides, as well as help fill existing knowledge gaps on neonicotinoid insecticides and MPs coexistence toxicity in aquatic environment.

Metalaxyl is an acylanilide systemic fungicide that is widely applied and can readily enter ecosystems through leaching and soil runoff. This research utilized zebrafish as a model organism to thoroughly investigate the detrimental impacts of environmentally relevant levels of metalaxyl on the development of the notochord in zebrafish embryos and to elucidate the underlying molecular mechanisms through transcriptomics, pharmacological intervention and molecular biological detection. The preliminary results demonstrated that metalaxyl induced significant modifications in the developmental parameters of zebrafish embryos. This study has also assessed the long-term consequences of metalaxyl exposure during the embryonic development of zebrafish. This study have demonstrated that zebrafish exposed to metalaxyl exhibit a range of abnormalities, including defects in notochord vacuole biogenesis, somite segmentation disorders, anomalous notochord curvatures, craniofacial cartilage deformities, and irregular chordacentra mineralisation. Through transcriptomic and bioinformatics analysis, it was found that most of the genes exhibiting differential expression were linked to oxidative stress. Furthermore, the evidence indicated that oxidative stress was present, as demonstrated by increased malondialdehyde (MDA) production and a decrease in antioxidant enzyme activity (CAT, SOD, GSH). Interestingly, the developmental dysfunction induced by metalaxyl was partially rescued by chlorogenic acid. Overall, metalaxyl disrupts notochord and skeletal formation in zebrafish embryos by modulating oxidative stress mediated by reactive oxygen species.

Nanoplastics (NPs) have been commonly detected in aquatic ecosystems, and their negative effects on aquatic organisms have raised concerns in the scientific community and general public. The acute toxicity, neurotoxicity, and metabolic toxicity induced by NPs on fishes have been reported by many studies, although less attention has been focused on how mother exposed to NPs affected their offspring in aquatic organisms. Here, female zebrafish (F0) were exposed to 0, 200 and 2000 μg/L polystyrene nanoplastics (PS-NPs) for 42 d, with their offspring (F1) reared in clear water until sexual maturity. The results showed that PS-NPs were detected in various organs of F0 and F1. PS-NPs exposure significantly decreased gonadal 17-estradiol (E2), while increasing testosterone (T) contents. Lower levels of cyp19a1a, lhr and erα expressions in the 2000 μg/L group were consistent with a reduced number of mature oocytes (MO), but an increase in perinucleolar oocytes (PO). Interestingly, the expression of vtg was only up-regulated by 200 μg/L PS-NPs. After exposure, the egg production was dramatically reduced, but the hatching rate and heartbeat of F1 embryos from treated females were significantly higher than those observed in females from the control group. Maternal PS-NPs exposure significantly decreased the E2 and T levels in F1 adults, while PS-NPs exposure significantly up-regulated the sox9a but down-regulated the foxl2a in F1 larvae of 30 days post fertilization (dpf). This study showed that PS-NPs caused reproductive toxicity by changing the hypothalamic-pituitary-gonadal (HPG) axis-related genes, impairing the reproductive capacity of female zebrafish, affecting the development and disrupting the endocrine function of F1. These results suggested that PS-NPs had adverse effects on fish reproductive system both in the directly exposed generation and in their unexposed offspring.

Carbofuran , a carbamate pesticide , is widely used in developing countries to manage insect pests. Studies have found that carbofuran posed potential risks for the neurotransmitter systems of non-target species, we speculated that these disruptive effects on the neurotransmitter systems could trigger anxiety-like behaviors. In this study, female zebrafish were exposed to environmental levels (5, 50, and 500 μg/L) of carbofuran for 48 h to evaluate the effects of carbofuran on anxiety-like behaviors. Results showed that zebrafish exhibited more anxiety-like behaviors which proved by the observed higher bottom trend and more erratic movements in the novel tank after carbofuran treatment. In order to elucidate the underlying molecular mechanisms of carbofuran-induced anxiety-promoting effects, we measured the levels of neurotransmitters, precursors, and major metabolites, along with the level of gene expression and the enzyme activities involved in neurotransmitter synthesis and metabolism. The results demonstrated that acute carbofuran exposure stimulated the mRNA expression and enzyme activity of tyrosine hydroxylase, which sequentially induced the increased levels of dopamine and norepinephrine . Tyrosine hydroxylase inhibitor relieved the anxiety-related changes induced by carbofuran, confirming the overactive tyrosine hydroxylase-mediated accumulation of dopamine and norepinephrine in the brain was one of the main reasons for carbofuran-induced anxiety-like behaviors in the female zebrafish. Overall, our study indicated the environmental health risks of carbamate pesticide in inducing neurobehavioral disorders and provided novel insights into the investigation of the relevant underlying mechanisms.
